The Arabidopsis (Arabidopsis thaliana) root epidermis is a simple model for investigating cell fate specification and pattern formation. In addition to regulatory networks consisting of transcription factors, histone deacetylases are also involved in the formation of cellular patterns. Here, we report thatHistone Deacetylase19 (HDA19) affects the root epidermal cellular pattern through regulation of cortical cell fate by interacting with SCARECROW (SCR). HDA19 binds to the DNA sequence upstream of SCR, as well as to those of several of SCR’s target genes, and regulates their expression. Mutant lines of several SCR target genes show impaired patterns of epidermal differentiation and cortical cell division, similar to that of hda19. This work presents HDA19 and SCR as two further players in the regulation of cortical and epidermal cell specification and describes an additional function for SCR.

China Spallation Neutron Source (CSNS) consists of 80 MeV H⁻ LINAC, 1.6 GeV RCS, RTBT line and one target. Many wire scanners and beam loss monitors (BLM) distributed along the LINAC and the RTBT for the profile and beam loss measurement. For the wire scanner, signal on the wire induced by the secondary electron is used for the profile measurement. Signal lost may happen when the wire or the signal chain shorted, thus a backup readout chain is required for the accident condition. As for the BLM, it is difficult to do the online calibration to see how sensitive the monitor is. Based on the two requests above, a crosscheck study was carried out recently, one wire scanner and the BLM next to it were chose at LINAC and RTBT. Both wire signal and BLM signal were recorded while the wire scanner crossing the beam. We found these two type of signals have the same accuracy for the profile measurement, and ~ 1 μA beam loss induced by the wire disturbance can be detected. Also thermal electron emission suspected happening during the measurement. More detailed experiment will be carried out in December. To monitor the size and position of 1.6 Gev proton beam in front of proton beam window(PBW) of China spallation neutron source (CSNS), one multi-wire profile monitor (MWPM) is designed and installed with PBW. It can bear the heat caused by beam and generate signal to electronic in local station. Lung adenocarcinoma (LUAD) is the most common histologic subtype of lung cancer. Previous research has shown heterogeneity in lung cancer, with the parallel existence of multiple subclones characterized by their own specific mutational landscape. The aim of our study was to gain insight into the evolutionary pattern of lung cancer by investigating the genomic heterogeneity between a nodule and its distant tumor. Luckily, we obtained nodule and tumor samples derived from surgery and a blood sample from a single patient. The samples are very unique, for tissues with the same genetic background from nodules to malignant tumors are rarely available and require precise micro-cutting. In this study, we performed whole-genome sequencing of these two samples, to identify novel candidate driver genes associated with LUAD. The nodule and tumor were found to have common significant ubiquitin-specific protease 40 (USP40) mutations, indicating an important driver role for the gene. Moreover, we also observed the two novel candidate driver genes ASCL5 and CAPNS1 in the LUAD sample. In summary, we pinpoint the predominant mutations in LUAD by WES, highlighting the substantial genetic alterations contributing to LUAD tumorigenesis. This may provide a better understanding of the clonal evolution during tumor development.

In order to inject the H⁻ beam to the Rapid Cycling Synchrotron (RCS) with high precision and high transport efficiency, the injection beam parameters need to be measured and then corrected while its eccentric position or direction angle is too large. In this paper, firstly, a method to measure the injection beam parameters by using two of the four multi-wire scanners (MWSs) is presented. The injection commissioning results confirmed that this method works well. Secondly, a method to measure the signals of injection beam and circular beam by the INMWS02 is presented and the method work well during the beam commissioning., Proceedings of the 9th Int. China Spallation Neutron Source(CSNS), the biggest platform for neutron scattering research in China, will be finished built and run in the end of 2017. It mainly consists of a 80MeV H⁻ linac and a 80MeV to 1.6GeV Rapid Cycling Synchrotron, two beam transport lines, one target station and relative ancillary facilities. The Linac beam commissioning with beam loss monitors, current transformers, BPMs, beam profile monitors and beam emission measurement has been the main task since last year. This study aimed at elucidating regulatory components behind floral organ identity determination and tissue development. It remains unclear how organ identity proteins facilitate development of organ primordia into tissues with a determined identity, even though it has long been accepted that floral organ identity is genetically determined by interaction of identity genes according to the ABC model. The first section DTL commissioning of China Spallation Neutron Source (CSNS) project has been successful finished in January, 2016. The H⁻ beam can be accelerated to 21.6 MeV at peak current 18 mA, achieved the design point. Different elements of the beam instrumentation system have been tested during the commissioning, including BPM, CT, FCT, WS, EM, BLM, and corresponding electronics and control systems. High accuracy phase measurement (precision @ ±1°) system has been started into operation. Beam loss monitor (BLM) for low energy, 3 MeV to 21.6 MeV, has been tested too, and got very positive results. For the LRBT, RCS and RTBT, different type wire scanner, BPM, WCM, CT were designed. The monitors fit for the high-radiation environments were considered. All the physical design work has been finished, and being manufactured. Increasing evidence shows that some cells from peripheral blood fibroblast-like mononuclear cells have the capacity to differentiate into mesenchymal lineages. However, the insufficiency of these cells in the circulation challenges the cell isolation and subsequently limits the clinical application of these cells. In the present study, the peripheral blood mononuclear cells (pbMNCs) were isolated from wound animals and treated with the supernatant of bone marrow mesenchymal stromal cells (bmMSCs). Results showed these pbMNCs were fibroblast-like, had stromal morphology, were negative for CD34 and CD45, but positive for Vimentin and Collagen I, and had the multipotency to differentiate into adipocytes and osteoblasts. We named these induced peripheral blood-derived mesenchymal stromal cells (ipbMSCs). Skin grafts in combination with ipbMSCs and collagen I were applied for wound healing, and results revealed ipbMSC exhibited similar potency and effectiveness in the promotion of wound healing to the bmMSCs. Hereafter, we speculate that the mixture of growth factors and chemokines secreted by bmMSCs may play an important roles in the induction of the proliferation and mesenchymal differentiation of mononuclear cells. Our results are clinically relevant because it provide a new method for the acquisition of MSCs which can be used as a candidate for the wound repair.
