Background: Freshwater fish in Bangladesh are adversely affected by the washed off pesticides, used in agriculture. The aim of this study was to evaluate the impacts of a commonly used organophosphate pesticide on freshwater stinging catfish, Heteropneustes fossilis, which envisioned that the possible threats might occur by this organophosphate group to other species in the wild. Methodology: To study the potential hazards of Envoy 50 SC on H. fossilis, fry of the fish were exposed to the acute toxicity tests. Changes in the hematological parameters, organ-specific histomorphologies, and brain acetylcholinesterase (AChE) activities were determined by treating the fish with agricultural recommended dose and below that dose of 0.015 and 0.0075 ppm, respectively. Results: LC50 of Envoy 50 SC for the fish was determined as 0.151 (0.014–0.198) ppm. Pesticide abruptly altered the normal tissue structures of the gill, liver, and kidney. The major alterations included were the gill lamellae missing, gill clubbing, hyperplasia, nuclear hypertrophy, vacuolation, glomerular expansion, increasing diameter of the renal tubules, hemorrhage, necrosis, and pyknosis. In blood cells, changes observed in the peripheral nuclear erythrocyte were large lymphocyte, dead cell, fusion of the cells, binucleated cells, tear-shaped cells, ghost cells, senile cells, and abnormal structures of the cells. Significantly lower (P < 0.05) red blood cell (RBC) count and AChE activities in fish brain due to the pesticide exposure suggested the reasons of abrupt behavior, increased oxygen consumption, and fish mortality at higher concentration of this organophosphate pesticide. Conclusion: The presence of pesticides, even at low concentrations, caused deleterious effects on the earlier life stages of a comparatively harder and robust fish, suggesting a wider range effect on the more sensitive wild life, in particular decrease in survival in their native environment. Cutaneous leishmaniasis (CL) is caused by the bite of the infected sand fly, which inoculates parasites of Leishmania spp and triggers an immune response. An exacerbated cutaneous inflammatory response is crucial for controlling parasite burden but can also promote tissue damage. This study aimed to characterize the populations of natural killer (NK), CD57+, CD4+, and CD8+ T cells, CD20+ B cells, as well as CD68+ macrophages, in biopsies of ulcerated CL lesions, and quantify the production of perforin+, grazyme B+, interleukin 1 beta (IL-1β+) and Tumor Necrosis Factor (TNF-α+ cells). We then correlated these parameters with necrosis, inflammation and the number of amastigotes. CD4+ T cells were positively correlated to the extent of inflammation, B cells and IL-1β+ were associated with the extent of necrosis, CD68+ macrophages and perforin were correlated with the number of amastigotes, and CD57+ NK cells was correlated to CD68+ macrophages and amastigotes. In sum, the finding suggests that the production of cytotoxic granules and cytokines by inflammatory cells contributes to tissue damage in CL lesions. [ABSTRACT FROM AUTHOR]

Purpose: The use of electrocautery during arthroscopy may heat intra-articular saline and subsequently damage intra- and extra-articular tissue. Newer electrocautery devices have the ability to measure the outflow fluid temperature and switch off before reaching a certain threshold; however, the scientific evidence establishing these temperature thresholds' potential for inadvertent damage is lacking. The aim of this study was to analyse current available literature on temperature thresholds for tissue damage after exposure to heated fluid and provide a recommendation for the maximum temperature of intra-articular fluid to prevent tissue damage.Methods: In February 2018, a systematic literature review was performed using the MEDLINE/PubMed and Embase databases. Inclusion was limited to studies investigating temperature thresholds for thermal damage to at least one of the tissues of interest: skin, bone, cartilage, soft tissues, and nerves. Studies not reporting specific temperature thresholds for thermal damage were excluded.Results: Twenty articles were selected for the final evaluation and data extraction. Varying temperature thresholds, based on the lowest reported temperature causing tissue damage, were found for the different tissues of interest: 44 °C for dermal tissues, between 47 and 50 °C for bony tissues, 50 °C for cartilage, between 43 and 55 °C for soft tissues, and 43 °C for nerves.Conclusion: Based on the current literature, a temperature threshold for intra-articular fluid of 43 °C during an arthroscopic procedure is recommended to prevent tissue damage. Higher temperatures may cause damage to surrounding intra- and extra-articular tissues. The threshold for irreversible damage is likely to be higher. Extravasation is an unintended release of intravenous fluids into the surrounding tissues, which may lead to inflammation, irritation and necrosis of the tissues. This happens primarily in intensive care patients due to a large number of regularly prescribed medications, multiple attempts and regular cancellations. Multiple puncture and vein gap holes and deterred venous structures should be avoided. If a lack of blood return from the cannula is generally referred to as a indication that extravasation has occurred. It is the most misleading of all symptoms and has been implicated in a variety of serious incidents. When using an infusion pump, close monitoring is required to test the flow rate, where potential changes in the location of the body, e.g. bending of the wrist or elbow, cannula support or bandaging, have been omitted as possible triggers, suggesting the risk of extravagance. Geriatrics and paediatrics are the primary casualties of extravagance. The understanding of how to treat these injuries differs depending on medications. Omadacycline is known to induce extravasation of the infusion site and has minimal treatment options. It is the first drug in the amino methylcycline antibiotic that belongs to the tetracycline subclass. Extravasation accidents remain rare, with a reported occurrence of between 0.1% and 6% recorded in the literature on inpatients receiving chemotherapy. Elimination of human error can be seen as training nurses and helping patients reduce the occurrence of extravasation and prevent local injuries. High content of mineral salts reduces crop yields. Agricultural areas are being reduced due to the secondary salinization of land. Wheat is one of the main crops, however, wheat is a type of plant that is sensitive to the toxic effects of mineral salts. Therefore, the identification of cytological and biochemical markers for determining resistant and sensitive wheat varieties to abiotic stress caused by the toxic effects of mineral salts is one of the important tasks of agriculture. Methods of fluorescence, cytophotometry, electrophoresis, and light-optical microscopy produced suitable cytological markers for creating a test system for the early diagnosis of wheat sensitivity to the tested salts. Different wheat genotypes of Triticum aestivum Host. (varieties Agata, Uchitel', and Zhemchuzhina Povolzh'ya) underwent treatment to investigate their reaction to the toxic effects of sodium chloride and sodium sulfate. The varieties responded differently to the presence of the salts; these were reflected in changes in biometric data, the passage of the cell cycle, features of ROS accumulation, and a quantitative assessment of the viability of seedling cells. In the genotypes sensitive to sodium chloride and sodium sulfate (varieties Zhemchuzhina Povolzh'ya and Uchitel'), tissue damage was observed in root cells under toxic effects, while in the resistant genotype (variety Agata) cell damage was minimal. The developed complex of intravital markers of various wheat genotypes in the early development stages can be effectively used to characterize wheat with different tolerance to chloride and sulfate salinization. Background/aim: Sepsis is a condition caused by infection followed by unregulated inflammatory response which may lead to organ dysfunction. The aim of this study is to be the first in the literature and it has been designed to show the thiol/disulphide changes in patients with sepsis and septic shock and their correlation with acute phase reactants. Material and methods: A total of 113 patients (septic shock 53 and sepsis 60) and 60 healthy control subjects have been enrolled in this study from the period February 2018 to 2019. The patients were divided in 2 groups: nonsurvivors (74) and survivors (39). The investigation includes measurements of native thiol, total thiol, dynamic disulphide bond, oxidized thiol ratio, reduced thiol ratio and thiol oxidation reduction ratio, erythrocyte sedimentation rate, C-reactive protein, and procalcitonin. Results: The findings of this study suggest that changes in thiol levels play a role in the pathogenesis of patients with sepsis and septic shock. Conclusions: Thiol/disulphide homeostasis is impaired in patients with sepsis and septic shock. Understanding the role of thiol/ disulphide homeostasis in sepsis and septic shock may provide different therapeutic intervention strategies for patients. [ABSTRACT FROM AUTHOR]

Duck tembusu virus (DTMUV) is newly emerged in poultry and causes great losses to the breeding industry in China and neighboring countries. Effective antiviral strategies are still being studied. Autophagy is a cellular degradative pathway, and our lab's previous data show that autophagy promotes DTMUV replication in vitro. To study the role of autophagy further in vivo , we utilized ducks as the animal model to investigate the autophagy responses in DTMUV-targeted tissues. And also, we utilized autophagy regulators, including Rapamycin (Rapa) as the autophagy enhancer, 3-Methyladenine (3-MA) and Chloroquine (CQ) as the autophagy inhibitors, to adjust the host autophagic levels and then study the effects of autophagy on tissue damages and virus replication. As a result, we first found DTMUV infection trigged autophagy and autophagy regulator treatments regulated autophagy levels successfully in duck spleens and brains. Next, we found that autophagy inhibitors inhibited DTMUV replication and alleviated DTMUV-induced pathological symptoms, whereas the autophagy inducer treatment led to the opposite effects. And we also found that autophagic regulation was correlated with the expression of innate immune genes, including pattern recognition receptors, type I interferons, and cytokines, and caused different effects in different tissues. In summary, we demonstrated that autophagy facilitated DTMUV replication, aggravated the developments of pathological symptoms and possibly counteracts the host's innate immunity response in vivo. Ovarian torsion is agynecologic emergency that affects females of all ages. Early diagnosis is important to preserve ovarian function. The false positive rate for sonographic diagnosis of ovarian torsion is 50%; therefore, a new real-time approach is required to improve diagnostic accuracy. We investigated diffuse reflectance spectroscopy for diagnosing ovarian torsion. Spectroscopic measurements were performed in vivo prior to, during and after detorsion. After bilateral oophorectomy, hemoxygenase and myeloperoxidase enzyme activity in ovarian tissue was evaluated and the tissues were examined for pathology. Spectroscopic data were compared to histopathological and biochemical data to assess the diagnostic value of the spectroscopic method for differentiating healthy and damaged ovarian tissue. We found a good correlation between spectroscopy and histopathology. We also found a correlation between the spectroscopic data and heme oxygenase enzyme activity. We found no correlation between the histopathological tissue damage score and myeloperoxidase enzyme activity. Objective: The aim of our study was to perform comparative investigation of the tissue safety of three different endoscopic lithotripter devices including a new single-probe/dual-energy lithotripter in an in vivo animal model. The Swiss LithoClast Trilogy was compared to the Storz Calcuson and the Swiss LithoClast Vario. The safety test simulated the accidental direct contact between lithotripter probes and the urothelium, which can occur when sliding off a stone or drilling through a calculus during lithotripsy. The safety test included a smallest (1.5 mm) and largest (3.3/3.4 mm) probe diameter per device. Methods: Testing was performed in nine pigs (three animals per device). The bladder tissue was exposed to direct lithotripter probe contact at maximum power for 10 s to produce visible tissue lesions. Acute tissue trauma was evaluated using a simplified scoring model describing the expected bladder wall injuries for histological examination. After 7 days, all animals were killed, necropsied and examined post mortem. For between-group comparisons regarding microscopic histopathologic features, a Chi-square test was used. A p value < 0.05 was considered to be statistically significant. Results: Irrespective of the lithotripter used, no systemic signs of toxicity were observed. Histologically, signs of normal ongoing healing were observed on the bladder mucosa. There were no significant differences in histological findings taking changes of the epithelium (p = 0.360), the leucocyte infiltration (p = 0.123), the vascular congestion (p = 0.929) and the edema (p = 1.0) between the groups into account. • A protocol was established to investigate tissue response to prosthesis loading. • The protocol was evaluated on individuals without amputation, who displayed distinct responses. • Ischaemic and inflammatory precursors were observed under low loads in all limbs, including one example residual limb. • Results show potential measurements for early detection of skin and sub-dermal damage risk during prosthesis use. In the early stages of rehabilitation after primary amputation, residual limb soft tissues have not been mechanically conditioned to support load and are vulnerable to damage from prosthetic use. There is limited quantitative knowledge of skin and soft tissue response to prosthetic loading. An in-vivo protocol was developed to establish suitable measures to assess tissue tolerance during loading representative of early prosthesis use. Ten participants without amputation one participant with trans-tibial amputation were recruited, and pressure applied to their calf in increments from 20 to 60 mmHg. Measurements were recorded at relevant skin sites including interface pressures, transcutaneous oxygen (T C PO 2) and carbon dioxide (T C PCO 2) tensions and inflammatory biomarkers. At the maximum cuff pressure, mean interface pressures were between 66 and 74 mmHg, associated with decreased T C PO 2 values. On the release of pressure, the ischaemic response was reversed. Significant upregulation (p < 0.05) in inflammatory biomarker IL-1α and its antagonist IL-1RA were observed at all sites immediately following loading. The protocol was successful in applying representative prosthetic loads to lower limb tissues and monitoring the physiological response, both in terms of tissue ischemia and skin inflammation. Results indicated that the measurement approaches were sensitive to changes in interface conditions, offering a promising approach to monitor tissue status for people with amputation. Targeting virulence factors represents a promising alternative approach to antimicrobial therapy, through the inhibition of pathogenic pathways that result in host tissue damage. Yet, virulence inhibition remains an understudied area in parasitology. Several medically important protozoan parasites such as Plasmodium , Entamoeba , Toxoplasma , and Leishmania secrete an inflammatory macrophage migration inhibitory factor (MIF) cytokine homolog, a virulence factor linked to severe disease. The aim of this study was to investigate the effectiveness of targeting parasite-produced MIF as combination therapy with standard antibiotics to reduce disease severity. Here, we used Entamoeba histolytica as the model MIF-secreting protozoan, and a mouse model that mirrors severe human infection. We found that intestinal inflammation and tissue damage were significantly reduced in mice treated with metronidazole when combined with anti– E. histolytica MIF antibodies, compared to metronidazole alone. Thus, this preclinical study provides proof-of-concept that combining antiparasite MIF-blocking antibodies with current standard-of-care antibiotics might improve outcomes in severe protozoan infections. BACKGROUND: Mesenchymal stem cells have been used in the treatment of various diseases due to their advantages, and mitochondrial transfer is an important mechanism of mesenchymal stem cell therapy. OBJECTIVE: To review the role of mesenchymal stem cells via mitochondrial transfer in various tissue injuries. METHODS: The key words were “stem cells, mitochondrial transfer, cell communication, rescue and repair, tissue injury” in English and Chinese, respectively. PubMed, CNKI and WanFang databases were retrieved for the articles published from 2008 to 2019. The articles concerning the mitochondrial transfer function and mechanism of mesenchymal stem cells and the treatment of tissue injury were collected. After removal of the repetitive and irrelevant articles, 64 eligible articles were further analyzed. RESULTS AND CONCLUSION: Mesenchymal stem cells have been used in stroke, myocardial infarction, and acute renal failure. Mitochondrial transfer has been widely recognized as a new mechanism of stem cell therapy and has been considered as a potential therapy for tissue injuries. Mitochondrial transfer has been found to be mediated by different modes, including tunnel tube formation, gap junctions, microvesicles, cell fusion and transfer of isolated mitochondria. However, specific mechanisms of mitochondrial transfer in tissue injury have not been fully elucidated and its specific role in tissue repair also needs further investigation. [ABSTRACT FROM AUTHOR]

Electrosurgical electrode has been widely used as surgery instrument, because it can cut tissues quickly with the blood being coagulated. There are two important issues, namely thermal damage and toxic fumes, which need to be solved urgently. This paper proposes a heat-assisted (H-A) electrosurgical electrode, that is, the active electrode is heated quickly at the moment when the cutting tissue starts. The advantage of this technology is that the electrode can smoothly cut the tissue with lower power supply. The experimental result shows that if the temperature of electrode was heated to 300 ℃ immediately before tissue cutting, the power output was decreased by approximately 22 %, the thickness of thermal damage was decreased by approximately 20 % and the concentration of toxic fume was decreased by approximately 43 %. Systemic inflammatory response and generation of oxidative stress are known to contribute to scorpion venom-induced tissue damage. TLR receptors might represent a link between oxidative stress and inflammation; we therefore investigated whether or not TLR4 is involved in venom-induced immunopathology. The obtained results showed that pharmacological targeting of TLR4 with the selective inhibitor TAK-242 (Resatorvid) prevents the inflammatory response induced by subcutaneous administration of Androctonus australis hector (Aah) venom, as revealed by a significant decrease of neutrophil cell count in peripheral blood associated with significant decline of neutrophil degranulation and sequestration to the lung, liver, and kidney tissues. Moreover, TAK-242 administration inhibited nitrite levels increase in serum, malondialdehyde (MDA), and protein carbonyl tissue contents concomitantly with a significant increase of catalase activity and reduced glutathione (GSH) level in tissue homogenates. Furthermore, venom-induced increases in serum levels of organ dysfunction markers (lactate deshydrogenase, aminotransferase ALT and AST, creatinine and urea) were also significantly suppressed by pre-treatment with TLR4 inhibitor, concordantly with a remarkable improvement in the histological features in lung and liver tissues. The results of the present study indicate the potential role of TLR4 in venom-induced immunopathology and show the in vivo requirement of TLR4 signaling in mediating venom-induced tissue damage. It was found recently that acrolein (CH2=CH–CHO), mainly produced from spermine, is more toxic than ROS (reactive oxygen species, O2−·, H2O2, and ·OH). In this review, we describe how the seriousness of brain infarction, dementia, renal failure, and Sjӧgren's syndrome is correlated with acrolein. In brain infarction and dementia, it was possible to identify incipient patients with high sensitivity and specificity by measuring protein-conjugated acrolein (PC-Acro) in plasma together with IL-6 and CRP in brain infarction and Aβ40/42 in dementia. The level of PC-Acro in plasma and saliva correlated with the seriousness of renal failure and Sjӧgren's syndrome, respectively. Thus, development of acrolein scavenger medicines containing SH-group such as N-acetylcysteine derivatives is important to maintain QOL (quality of life) of the elderly. [ABSTRACT FROM AUTHOR]

Rhein is one of active anthraquinone components in traditional Chinese herbal medicine Rheum palmatum L. , possessing anti-inflammatory, antioxidant, antitumor, antiviral, and hepatoprotective activities. Human respiratory syncytial virus (RSV), a common virus, is able to result in pneumonia and bronchitis, which usually can be seen in infants. However, so far the effects of Rhein on RSV-induced pneumonia are still unknown. As the NLRP3 inflammasome is activated excessively, it is able to lead to inflammatory response and tissue injury in most viral infection process (including RSV infection) of respiratory tract. Therefore, we designed experiments to reveal whether Rhein can treat RSV-induced pneumonia by inhibiting NLRP3 inflammasome activation. In present research, we established the pneumonia model of BALB/C mice caused by RSV. First of all, the pathology of lung tissue and the weight of mice were evaluated, and the corresponding lung index was calculated. Additionally, the expression of pro-inflammatory mediators in serum and lung tissues, and related proteins (NLRP3, ASC and Caspase-1) of NLRP3 inflammasome and NF-κB pathway were detected by Enzyme-linked immunosorbent assay (ELISA), Real-time PCR (RT-PCR), Immunohistochemistry (IHC), and Western blot (WB), respectively. The determination of lung index and lung tissue pathological evaluation revealed that Rhein was able to alleviate lung infection and injury caused by RSV. The results of ELISA showed that Rhein was able to reduce the release of pro-inflammatory cytokines in the serum and lung tissues of RSV-induced BALB/c mice, including IL-1β, IL-6, TNF-α, IL-18, and IL-33. Additionally, it was revealed that Rhein inhibited the immune inflammatory response of RSV-infected mice, which was likely to be associated with the inhibition the NLRP3 inflammasome activation via NF-κB pathway. To sum up, our results indicated that Rhein may inhibit RSV-induced pulmonary inflammatory response effectively; meanwhile, it is emphasized that Rhein therapy is likely to be a promising treatment on the RSV-infected lung inflammation and avoidance of lung tissue damage. Introduction: Commercially available junctional tourniquets (JTQs) have several drawbacks. We developed a low-cost, compact, easy to apply JTQ. The aim of this study was to assess the tourniquets' safety and efficacy in a swine model of controlled hemorrhage.Materials and Methods: Five pigs were subjected to controlled bleeding of 35% of their blood volume. Subsequently, the JTQ was applied to the inguinal area for 180 minutes. Afterwards, the tourniquet was removed for additional 60 minutes of follow up. During the study, blood flow to both hind limbs and blood samples for tissue damage markers were repeatedly assessed. Following sacrifice, injury to both inguinal areas was evaluated microscopically and macroscopically.Results: Angiography demonstrated complete occlusion of femoral artery flow, which was restored following removal of the tourniquet. No gross signs of tissue damage were noticed. Histological analysis revealed mild necrosis and infiltration of inflammatory cells. Blood tests showed a mild increase in potassium and lactic acid levels throughout the protocol.Conclusions: The tourniquet achieved effective arterial occlusion with minimal tissue damage, similar to reports of other JTQs. Subjected to further human trials, the tourniquet might be a suitable candidate for widespread frontline deployment because of its versatility, compactness, and affordable design. Background: To assess the toxicity of environmental pollutants in fish, there are different physiological parameters including histology. This study aimed to investigate the effect of prebiotic dietary supplement on fish immune response after exposure to toxicant. Methods: The study was conducted in the Gorgan University of Agricultural and Natural Resources aquaculture Laboratory in 2017. Common carp species were obtained and exposed to 2 ppm, 3 ppm, 6 ppm Abamectin as a toxicant. Prebiotic isomalto-oligosaccharides added to diet with spray method as 1 g/kg and histopathological examinations were done after 60 d experiment. There were no significant differences between aquariums in water quality during the test and no mortality and injuries were observed during accumulation. Results: The Abamectin caused some lesions such as vacuolization latest, bleeding, necrosis, degeneration of the epithelium, the destruction of the villi in the intestine, destruction of liver cells, ascites, hemorrhage, necrosis and nuclear karyolysis in the liver and lymphocytes penetration and degradation of intestinal epithelium in intestine. Maximum lesions observed in 6-ppm toxicant concentration. Conclusion: The isomalto-oligosaccharides probiotics was not successful in stimulating the immune system and reducing adverse effect of toxicant in common carp, significantly. However, usage of this prebiotic could be useful in some cases.

Plasma Medicine tools exploit the therapeutic effects of the exposure of living matter to plasma produced at atmospheric pressure. Since these plasmas are usually characterized by a non-thermal equilibrium (highly energetic electrons, low temperature ions), thermal effects on the substrate are usually considered negligible. Conversely, reactive oxygen and nitrogen species (RONS), UV radiation and metastables are thought to play a major role. In this contribution, we compare the presence of thermal effects in different operational regimes (corresponding to different power levels) of the Plasma Coagulation Controller (PCC), a plasma source specifically designed for accelerating blood coagulation. In particular, we analyze the application of PCC on human blood samples (in vitro) and male Wistar rats tissues (in vivo). Histological analysis points out, for the highest applied power regime, the onset of detrimental thermal effects such as red cell lysis in blood samples and tissues damages in in-vivo experiments. Calorimetric bench tests performed on metallic targets show that the current coupled by the plasma on the substrate induces most of measured thermal loads through a resistive coupling. Frostbite causes tissue damage through five major mechanisms, out of which two are amenable to treatment. The first-line treatment is rapid rewarming therapy using water at 40°C to 42°C, which addresses the formation of ice crystals in the intra and extra cellular compartments. The second mechanism is progressive tissue ischemia after rewarming and is only accessible to a second-line therapy represented by thrombolysis. This study aimed to determine the efficacy of thrombolysis. This is a single-center retrospective cohort study, where it was aimed to evaluate two groups of patients. A total of 18 patients were included in this study. Mean times between injury to thrombolytic therapy and admission to thrombolytic therapy was 26.04 hours (SD 13.6) and 9.65 hours (SD 9.89), respectively. All patients suffered injuries ranging from second-degree deep to third degree. The rate of patients having complete, partial, and no angiographic responses were 55.6%, 11.1%, and 33.3%, respectively. The main outcome of interest showed that 11 (61.1%) patients in total had amputations at different levels. Results showed that in the intervention group, five (55.6%) of the patients had amputations compared with six (66.7%) from the control group (P = .6) at comparable levels of amputation. The literature supports that the use of intra-arterial tissue plasminogen activator might be beneficial for severe cases of frostbites; however, it lacks of studies of major significance and results are often controversial. Our study has not shown statistically significant results on amputation levels and cannot support the hypothesis of efficacy of thrombolytic therapy. [ABSTRACT FROM AUTHOR]

Focused ultrasound (FUS)-induced cavitation-mediated brain therapies have become emerging therapeutic modalities for neurologic diseases. Cavitation monitoring is essential to ensure the safety of all cavitation-mediated therapeutic techniques as inertial cavitation can be associated with tissue damage. The objective of this study was to reveal the correlation between the inertial cavitation dose, quantified by passive cavitation imaging (PCI), and brain tissue histologic-level damage induced by FUS in combination with microbubbles. An ultrasound image-guided FUS system consisting of a single-element FUS transducer (1.5 MHz) and a co-axially aligned 128-element linear ultrasound imaging array was used to perform FUS treatment of mice. Mice were sonicated by FUS with different peak negative pressures (0.5 MPa, 1.1 MPa, 4.0 MPa and 6.5 MPa) in the presence of systemically injected microbubbles. The acoustic emissions from the FUS-activated microbubbles were passively detected by the imaging array. The pre-beamformed channel data were acquired and processed offline using the frequency-domain delay, sum and integration algorithm to generate inertial cavitation maps. All the mice were sacrificed after the FUS treatment, and their brains were harvested and processed for hematoxylin and eosin staining. The obtained inertial cavitation maps revealed the dynamic changes of microbubble behaviors during FUS treatment at different pressure levels. It was found that the inertial cavitation dose quantified based on PCI had a linear correlation with the scale of histologic-level tissue damage. • Electrolysis products have a major role in electrochemical treatment of Tumors(EChT). • This work evidences that the extreme pH fronts affect the permeabilization of the tissue and consequently its destruction. • Modeled ions demonstrate the diffusive regime of ionic transport and show the low influence of the electric field on EChT. Electrochemical treatment is used in the local control of solid tumors in preclinical and clinical studies. We study pH fronts, permeabilization, electrical field and concentration of four chemical species in a tissue under electrochemical treatment by means of in vitro and in silico modeling. The in vitro model uses a piece of potato (solanum tuberosum L.) as analogy of tumor tissue, due to the characteristics of the potato tissue and the way in which it reacts against pH and potential. The in silico model solves the two-dimensional Nernst-Planck equations for ionic transport in a four-ion electrolyte. Modeled ions demonstrate the diffusive regime of ionic transport and show the little influence of the low electric field applied on this phenomenon. This work evidences that extreme pH fronts affect the permeabilization of the tissue and consequently its destruction. We also evaluate different shapes of electrode arrays by means of simulated and in vitro models. Envenomations by venomous snakes have major public health implications on a global scale. Despite its medical importance, snakebite has long been a neglected tropical disease by both governments and medical science. Many aspects of the resulting pathophysiology have been largely under-investigated. Most research on snake venom has focused on the neurological effects, with coagulotoxicity being relatively neglected, especially for venoms in the Elapidae snake family. In order to fill the knowledge gap regarding the coagulotoxic effects of elapid snake venoms, we performed functional activity tests to determine the fibrinogenolytic activity of 29 African and Asian elapid venoms across eight genera. The results of this study revealed that destructive (non-clotting) fibrinogenolytic activity is widespread across the African and Asian elapids. This trait evolved independently twice: once in the Hemachatus/Naja last common ancestor and again in Ophiophagus. Further, within Naja this trait was amplified on several independent occasions and possibly explains some of the clinical symptoms produced by these species. Species within the Hemachatus/Naja with fibrinogenolytic activity only cleaved the Aα-chain of fibrinogen, whereas Ophiophagus venoms degraded both the Aα- and the Bβ-chain of fibrinogen. All other lineages tested in this study lacked significant fibrinogenolytic effects. Artificial tissues made of fiber-reinforced elastomers typically have mechanical properties similar to biological tissues. Such a similarity motivates artificial tissue usage in building human-like soft robots aiming primarily to perform a wide range of human-like motions and effectively interact with the human environment. Soft robots can be ripped or damaged for unknown reasons in a variety of applications. The literature has no unified stress-based criterion for predicting the critical zones of artificial tissues undergoing deformations. Such a unified stress-based criterion typically entails detecting internal damage and establishing the stress limit criterion based on the maximum stretching threshold. The current study establishes an experimentally validated rupture criterion for a novel class of smart artificial tissues made of electro-active polymers motivated by biological tissue damages in several robust conditions. A thermodynamically consistent mechanoelectrical deformation model is formulated, demonstrating that such tissues have inherent anisotropy that must be taken into account when modeling such materials. Later, a continuum physics-based rupture model is derived as a unified stress-based criterion to investigate the effect of physical parameters associated with artificial tissue failure. Experimental burn models are essential tools for simulating human burn injuries and exploring the consequences of burns or new treatment strategies. Unlike clinical studies, experimental models allow a direct comparison of different aspects of burns under controlled conditions and thereby provide relevant information on the molecular mechanisms of tissue damage and wound healing, as well as potential therapeutic targets. While most comparative burn studies are performed in animal models, a few human or humanized models have been successfully employed to study local events at the injury site. However, the consensus between animal and human studies regarding the cellular and molecular nature of systemic inflammatory response syndrome (SIRS), scarring, and neovascularization is limited. The many interspecies differences prohibit the outcomes of animal model studies from being fully translated into the human system. Thus, the development of more targeted, individualized treatments for burn injuries remains a major challenge in this field. This review focuses on the latest progress in experimental burn models achieved since 2016, and summarizes the outcomes regarding potential methodological improvements, assessments of molecular responses to injury, and therapeutic advances.

The human respiratory syncytial virus (hRSV) remains one of the leading pathogens causing acute respiratory tract infections (ARTIs) in children younger than 2 years old, worldwide. Hospitalizations during the winter season due to hRSV-induced bronchiolitis and pneumonia increase every year. Despite this, there are no available vaccines to mitigate the health and economic burden caused by hRSV infection. The pathology caused by hRSV induces significant damage to the pulmonary epithelium, due to an excessive inflammatory response at the airways. Cytokines are considered essential players for the establishment and modulation of the immune and inflammatory responses, which can either be beneficial or harmful for the host. The deleterious effect observed upon hRSV infection is mainly due to tissue damage caused by immune cells recruited to the site of infection. This cellular recruitment takes place due to an altered profile of cytokines secreted by epithelial cells. As a result of inflammatory cell recruitment, the amounts of cytokines, such as IL-1, IL-6, IL-10, and CCL5 are further increased, while IL-10 and IFN-γ are decreased. However, additional studies are required to elicit the mediators directly associated with hRSV damage entirely. In addition to the detrimental induction of inflammatory mediators in the respiratory tract caused by hRSV, reports indicating alterations in the central nervous system (CNS) have been published. Indeed, elevated levels of IL-6, IL-8 (CXCL8), CCL2, and CCL4 have been reported in cerebrospinal fluid from patients with severe bronchiolitis and hRSV-associated encephalopathy. In this review article, we provide an in-depth analysis of the role of cytokines secreted upon hRSV infection and their potentially harmful contribution to tissue damage of the respiratory tract and the CNS.

The restoration of homeostasis after tissue damage relies on proper spatial-temporal control of damage-induced apoptosis and compensatory proliferation. In Drosophila imaginal discs these processes are coordinated by the stress response pathway JNK. We demonstrate that JNK signaling induces a dose-dependent extension of G2 in tissue damage and tumors, resulting in either transient stalling or a prolonged but reversible cell cycle arrest. G2-stalling is mediated by downregulation of the G2/M-specific phosphatase String(Stg)/Cdc25. Ectopic expression of stg is sufficient to suppress G2-stalling and reveals roles for stalling in survival, proliferation and paracrine signaling. G2-stalling protects cells from JNK-induced apoptosis, but under chronic conditions, reduces proliferative potential of JNK-signaling cells while promoting non-autonomous proliferation. Thus, transient cell cycle stalling in G2 has key roles in wound healing but becomes detrimental upon chronic JNK overstimulation, with important implications for chronic wound healing pathologies or tumorigenic transformation.

Swelling in the vocal folds is caused by the local accumulation of fluid, and has been implicated as a phase in the development of phonotraumatic vocal hyperfunction and related structural pathologies, such as vocal fold nodules. It has been posited that small degrees of swelling may be protective, but large amounts may lead to a vicious cycle wherein the engorged folds lead to conditions that promote further swelling, leading to pathologies. As a first effort to explore the mechanics of vocal fold swelling and its potential role in the etiology of voice disorders, this study employs a finite-element model with swelling confined to the superficial lamina propria, which changes the volume, mass, and stiffness of the cover layer. The impacts of swelling on a number of vocal fold kinematic and damage measures, including von Mises stress, internal viscous dissipation, and collision pressure, are presented. Swelling has small but consistent effects on voice outputs, including a reduction in fundamental frequency with increasing swelling (10 Hz at 30 % swelling). Average von Mises stress decreases slightly for small degrees of swelling but increases at large magnitudes, consistent with expectations for a vicious cycle. Both viscous dissipation and collision pressure consistently increase with the magnitude of swelling. This first effort at modeling the impact of swelling on vocal fold kinematics, kinetics, and damage measures highlights the complexity with which phonotrauma can influence performance metrics. Further identification and exploration of salient candidate measures of damage and refined studies coupling swelling with local phonotrauma are expected to shed further light on the etiological pathways of phonotraumatic vocal hyperfunction., (© 2023. Severe infections induce immune defense mechanisms and initial tissue damage, which produce an inflammatory neutrophil response. Upon dysregulation of these responses, inflammation, further tissue damage, and systemic spread of the pathogen may occur. Subsequent vascular inflammation and activation of coagulation processes may cause microvascular obstruction at sites distal to the primary site of infection. Low immunoglobulin (Ig) M and IgG levels have been detected in patients with severe infections like sCAP and sepsis, associated with increased severity and mortality. Based on Ig's modes of action, supplementation with polyvalent intravenous Ig preparations (standard IVIg or IgM/IgA-enriched Ig preparations) has long been discussed as a treatment option for severe infections. A prerequisite seems to be the timely administration of Ig preparations before excessive tissue damage has occurred and coagulopathy has developed. This review focuses on nonclinical and clinical studies that evaluated tissue-protective activities resulting from interactions of Igs with neutrophils, complement, and the coagulation system. The data indicate that coagulopathy, organ failure, and even death of patients can possibly be prevented by the timely combined interactions of (natural) IgM, IgA, and IgG with neutrophils and complement.

Collagen, the most abundant protein in our bodies, plays a crucial role in maintaining the structural integrity of various tissues and organs. Beyond its involvement in skin elasticity and joint health, emerging research suggests that collagen may significantly impact the treatment of complex diseases, particularly those associated with tissue damage and inflammation. The versatile functions of collagen, including skin regeneration, improving joint health, and increasing bone strength, make it potentially useful in treating different diseases. To the best of my knowledge, the strategy of using collagen to treat comorbid diseases has not been widely studied. This paper aims to explore the potential of collagen in treating comorbid diseases, including rheumatoid arthritis, osteoarthritis, osteoporosis, psoriatic arthritis, sarcopenia, gastroesophageal reflux, periodontitis, skin aging, and diabetes mellitus. Collagen-based therapies have shown promise in managing comorbidities due to their versatile properties. The multifaceted nature of collagen positions it as a promising candidate for treating complex diseases and addressing comorbid conditions. Its roles in wound healing, musculoskeletal disorders, cardiovascular health, and gastrointestinal conditions highlight the diverse therapeutic applications of collagen in the context of comorbidity management.

Viral infections trigger robust secretion of interferons and other antiviral cytokines by infected and bystander cells, which in turn can tune the immune response and may lead to viral clearance or immune suppression. However, aberrant or unrestricted cytokine responses can damage host tissues, leading to organ dysfunction, and even death. To understand the cytokine milieu and immune responses in infected host tissues, non-human primate (NHP) models have emerged as important tools. NHP have been used for decades to study human infections and have played significant roles in the development of vaccines, drug therapies and other immune treatment modalities, aided by an ability to control disease parameters, and unrestricted tissue access. In addition to the genetic and physiological similarities with humans, NHP have conserved immunologic properties with over 90% amino acid similarity for most cytokines. For example, human-like symptomology and acute respiratory syndrome is found in cynomolgus macaques infected with highly pathogenic avian influenza virus, antibody enhanced dengue disease is common in neotropical primates, and in NHP models of viral hepatitis cytokine-induced inflammation induces severe liver damage, fibrosis, and hepatocellular carcinoma recapitulates human disease. To regulate inflammation, anti-cytokine therapy studies in NHP are underway and will provide important insights for future human interventions. This review will provide a comprehensive outline of the cytokine-mediated exacerbation of disease and tissue damage in NHP models of viral infections and therapeutic strategies that can aid in prevention/treatment of the disease syndromes.
