The assessment and improvement of animal welfare is not only an ethical requirement, but also a guarantee of efficient and high quality translational research. In recent years, sophisticated animal models were created to study human diseases. However, the assessment of distress in these animal models is scarce. In order to provide a solid basis for optimizing animal welfare in future studies, body weight, burrowing, nesting activity, faecal corticosterone metabolites (FCMs), and distress scores were analyzed in several animal models.

In dieser Feldstudie wurde nach dem Design-Based-Research das Gesundheitspotenzial von naturwissenschaftlichem Unterricht im Schulgarten untersucht. Zur Erfassung des aktuellen Wohlbefindens wurden die Emotionen, die Befindlichkeit und die prosozialen Interaktionen der SchülerInnen eines 6. Jahrgangs durch systematische Befragungen und Beobachtungen dokumentiert. Die Ergebnisse zeigen eine positive Wirkung des Schulgartenunterrichts - positive Gefühle, gute Stimmung und prosoziale Interaktionen sind im Vergleich häufiger als im Klassenraumunterricht.

The utility of chemotaxonomic identification with compatible solute expression in the Stichococcaceae was investigated. Strains were put under abiotic stress to connect their taxonomy to their ecophysiology. Innate resistance to salinity and desiccation stress were independent of one another. Halotolerance varied more between strains than drought tolerance. Pseudostichococcus MOEWUS, 1951had higher stress tolerance than strains of the other genera. This work examined the effects of these two abiotic stress factors on the Stichococcaceae and connects their ecophysiology to their taxonomy.

Adipositas geht mit verschiedenen Komorbiditäten einher wie einer erhöhten Prävalenz für das metabolische Syndrom, der nichtalkoholischen Fettlebererkrankung und zentralnervösen Erkrankungen. Ein prominenter Kandidat in der hepatisch-neuronalen Kommunikation ist der Fibroblast Growth Factor 21 (FGF21), der den Stoffwechsel bei gesunden und adipösen Individuen moduliert. Mit der vorgelegten Arbeit ist es gelungen, neurodegenerative und Adipositas-induzierte (neuro-)inflammatorische Prozesse durch verschiedene „Lifestyle“-Faktoren zu modulieren und mit FGF21 zu assoziieren.

Currently, the Eastern Baltic cod (EBC) is in continuing decline. Supporting management efforts to assist in its recovery will require a functional understanding of the dynamics of the EBC and the Baltic ecosystem. However, aquatic environments are challenging to research as they are elusive, encompass many scientific disciplines and are complex adaptive systems. This thesis explores how modelling and simulation methods can be applied and adapted to meet the specific needs of fisheries biologies’ current challenges regarding the EBC and potentially those of other stocks in similar situations.

Die Spleen Tyrosin Kinase (SYK) und die Proteine der Bromodomänen und extraterminale Domänen (BET)-Familie stellen therapeutische Zielstrukturen zielgerichteter Therapien dar. Die Kombinationsinhibition von SYK und BET Proteinen bei humanen und caninen B-Zell Neoplasien induzierte synergistische anti-proliferative Effekte. Gesamt-Transkriptomanalysen zeigten eine gesteigerte Genexpressionsänderung und eine kombinationsspezifische Gensignatur. Toxicity of nanoparticles have raised concerns about their potential effects on coastal ecosystems, yet the mechanisms of the nanoparticle toxicity and their interactions with common abiotic stressors such as temperature and salinity are not well understood. The aim of our present study was to investigate the combined effects of seasonal warming, acclimation at elevated temperatures and salinity stress on bioenergetics, immune and general cellular stress responses to nZnO nanoparticles pollution in the blue mussels M. edulis.

The impact of salinity on coastal peatland vegetation was investigated. Phragmites australis is the most dominant emergent macrophyte in freshwater to mesohaline peatlands. Other common species include Typha latifolia, Carex spp. and Schoenoplectus tabernaemontani. P. australis and S. tabernaemontani are resilient to salinity and frequent salinity pulses while Carex sp. and T. latifolia are not. Litter decomposition rates of these macrophytes do not vary in freshwater and oligohaline peatlands., Es wurde untersucht, wie sich der Salzgehalt auf die Moorvegetation der Küstengebiete auswirkt. Die Schwimmkrabbe als Morphotyp unterscheidet sich von vielen anderen Vertretern der Echten Krabben (Brachyura) durch Eigenschaften, die sie dazu befähigen, auf einzigartige Art und Weise zu schwimmen. In dieser Arbeit wird gezeigt, dass der Morphotyp sich von Nicht-Schwimmkrabben auch bezogen auf die innere Anatomie unterscheidet. Eine Rekonstruktion der Gestalt von Vorfahren kommt zu dem Ergebnis, dass der Morphotyp nur einmal während der Evolution entstanden ist, aber in mehreren Fällen wieder zu sich ausschließlich laufend fortbewegenden Formen umgekehrt wurde.

Ziel der Arbeit war es, die Interaktion zwischen T-Zellen und autologen kolorektalen Tumorzellen detailliert zu untersuchen. In Kokulturversuche beider Zellpopulationen wurde eine immunsuppressive Wirkung der Tumorzellen gegenüber den T-Zellen nachgewiesen, die auch nicht durch den Einsatz von Immuncheckpoint-Inhibitoren aufgehoben werden konnte. Eine verbesserte T-Zell-Aktivierung sowie Tumorzellerkennung wurde jedoch durch eine Peptidstimulation der T-Zellen induziert. Dabei handelte es sich neben Neoantigenen auch um tumorspezifische kryptische Peptide.

Ocean circulation distributes heat, salt, and nutrients through the different basins. Thus, changes in circulation will ultimately impact the biogeochemistry of the ocean. In this work, the variability of currents and water column properties were studied in the Northeast Subtropical Atlantic, together with changes in the biogeochemistry in the upper 300 m over the North Atlantic Subtropical gyre. It was found that ocean circulation and biogeochemistry in the North Atlantic underwent drastic changes since the 1960s, leading to a reduction in the CO2 uptake by the oceans in the last 100 years.

Phosphorus (P) is indispensable for organisms but also contributes to eutrophication in aquatic ecosystems. The objective of the work was to characterize P speciation and transformation at boundaries between land and sea. Samples were analyzed by sequential P fractionation, P X-ray absoprtion near edge structure spectroscopy and 31P nuclear magnetic resonance spectroscopy. Results confirmed a transition from labile Fe- and Al P in terrestrial soils towards stable Ca-P in aquatic sediments. To protect aquatic ecosystems from further P inputs, sinks for P have to be preserved., Phosphor (P) ist ein unverzichtbares Nährelement, aber trägt gleichzeitig erheblich zur Eutrophierung aquatischer Ökosysteme bei. Proben wurden mithilfe der sequentiellen P-Fraktionierung, P-Röntgenabsorptionsspektroskopie und 31P Kernmagnetresonanz Spektroskopie analysiert. Die Ergebnisse bestätigten Übergänge von labilem Fe- und Al-P in terrestrischen Böden zu stabilerem Ca-P in aquatischen Sedimenten. Um aquatische Ökosysteme vor P-Einträgen zu schützen, müssen P-Senken erhalten bleiben.

New serodiagnostic formats for the detection of influenza virus and NDV infections were established and validated. Here, several recombinant proteins of influenza A virus and ND virus were expressed via bacterial expression systems. Authentic folding of these proteins was achieved in vitro. These recombinant proteins served as antigens in indirect ELISA assays or in Luminex bead-based liquid arrays. It is demonstrated that serological assays based on these recombinant proteins are potential alternative methods to classical yet difficult to standardize hemagglutination inhibition assays.

In terms of traits and components, the innate immune system of teleost fish is similar to that of mammals. Despite the fact that gene or protein orthology and conservation allow for function similarity, the Whole Genome Duplication (WGD) that occurred early in the life of ray-finned fishes permitted the formation of paralog genes with novel functions in fish. Therefore, the aim of this thesis was focused towards the characterisation of selected factors able to regulate the innate immune response of rainbow trout and in salmonid cell models.

In addition, the results suggest that psychosocial stress may increase the susceptibility of piglets to disease in the long term and that sex already plays a crucial role in the adaptive mechanisms of young animals. However, it was also shown that the negative effects of stress can be mitigated by the presence of littermates.

Zusammenfassend belegen die Ergebnisse dieser Promotionsarbeit die hohe Wirksamkeit der Cyclin-abhängigen Kinaseinhibitoren allein und in Kombination. Besonders das gezeigte breite Wirkspektrum deutet auf ein großes therapeutisches Potential beim Glioblastoma multiforme (GBM) und Kopf-Hals-Tumoren (HNSCC) hin. Hierbei wurden neuartige Wirkmechanismen identifiziert, die eine ideale Ausgangsbasis für die künftige Nutzung dieser Substanzen als innovative Therapiestrategie für GBM- und HNSCC-PatientInnen sowohl in der Primär- als auch in der Rezidivsituation darstellen.

The presented work contributes to the overall understanding of the bubble-mediated transport process, by defining the parameters controlling the transport efficiency and identifying the methanotrophic bacteria transported into the water column via the gas bubbles. Further, it highlights the importance of bentho-pelagic transport processes at seep sites and their positive feedback on the pelagic methane sink.

This dissertation focused on the ecophysiological responses of unialgal diatom isolates from the microphytobenthos in the shallow southern Baltic Sea and adjacent coastal peatlands to the fundamental variables salinity, temperature, and light availability in their respective habitats, as well as the influence of potential mixing of waterbodies of the two ecosystems on growth rates and photosynthetic performance of the benthic diatoms. Results within three publications showed eurythermal and euryhaline growth, a high photo-physiological plasticity and stimulated growth by peatland water.

Passive electroreception is the ability to perceive electric fields in water. It is found mainly in different fish species as well as in the monotremes or in the Guiana dolphin. In this dissertation, it was investigated whether bottlenose dolphins also possesses this sensory ability. In a complex behavioral experiment, four dolphins learned to respond reliably to weak electric fields in water. For two of the animals, a detection threshold for direct current and low-frequency alternating current electric fields was determined, establishing passive electroreception as a sensory modality.

This dissertation investigates the effects of different dietary phosphorus (P) intake on the endogenous mechanism of P homeostasis in a wide range of organs and tissues of pigs. The results suggests that the gene expression of transcellular P transporters is tissue-specific and that dietary interventions changes the gene expression levels of P transporters in relevant tissues to maintain P homeostasis in the animal. The results of this study also suggest that the variable P-containing diet may influence vitamin D metabolism in animals to maintain bone and mineral homeostasis., In dieser Dissertation werden die Auswirkungen einer unterschiedlichen Phosphor (P)-Zufuhr mit der Nahrung auf den endogenen Mechanismus der P-Homöostase in einer Vielzahl von Organen und Geweben von Schweinen untersucht. Die Ergebnisse deuten darauf hin, dass die Genexpression von transzellulären P-Transportern gewebespezifisch ist und dass Ernährungsmaßnahmen die Genexpression von P-Transportern in relevanten Geweben verändern, um die P-Homöostase im Tier aufrechtzuerhalten.

Die Indikatorfunktion beruht darauf, dass viele Phytoplankter unter niedrigen Phosphatkonzentrationen die Synthese von alkalinen Phosphatasen induzieren. In dieser Dissertation wurde die Indikatorfunktion anhand von drei, ökologisch sehr unterschiedlichen, Gewässern neu bewertet. Es werden Empfehlungstipps gegeben wie die AlkPA-Messungen durchgeführt werden sollten, um Missinterpretationen der Ergebnisse zu minimieren.

The sandy eulittoral of the west coast of Europe is the habitat of the green worm Symsagittifera roscoffensis. It’s green colour results from an endosymbiosis with the micro algae Tetraselmis convolutae. The availability of light and nutrients plays a fundamental role for both partners. This theses elucidates the behaviour of the host S. roscoffensis under different light conditions and its ability to react to nutrients. S. roscoffensis chooses optimal light conditions for its symbiont and buries itself into the sand at high light intensities and protects its symbiont against photo damage.

The indicator function is based on the fact that many phytoplankton species induce the synthesis of alkaline phosphatases under low phosphate concentrations. In this dissertation, the indicator function was reassessed on the basis of three ecologically very different bodies of water. Recommendations are given on how the AlkPA measurements should be done to minimize misinterpretations of the results.

This dissertation deals with the identification of suitable indicators for monitoring the health status of intensively farmed pikeperch, a fish species of great importance for the diversification of European aquaculture. Suitable candidate genes were identified to investigate potentially inadequate husbandry conditions as well as ontogenetic processes at the transcriptional level. Furthermore, it could be shown that reduced oxygen saturation (40%) and increased water temperatures (25 °C) do not lead to severe stress responses in juvenile fish., Diese Dissertation befasst sich mit der Identifizierung geeigneter Indikatoren zur Überwachung des Gesundheitszustands des intensiv gezüchteten Zanders, welcher von großer Bedeutung für die Diversifizierung der europäischen Aquakultur ist. Außerdem konnte gezeigt werden, dass eine verringerte Sauerstoffsättigung (40 %) und erhöhte Wassertemperaturen (25 °C) bei Jungfischen zu keinen schweren Stressreaktionen führen.

In the past, absolute concentrations of individual hormones were mainly used to describe the IGF system. According to current understanding, however, the analysis of the biological activity comes to the foreground for the description of the IGF system. In this work, the interaction of several factors is given greater consideration for the description of growth regulation in mouse lines selected for growth and in transgenic mouse lines. Furthermore, a bio-assay for in vitro analysis is presented.

The aim of this study was to investigate the effect of therapeutically used immunoglobulin G (IgG) on colon cancer in the presence of oxaliplatin. We observed that IgG failed to significantly influence tumor cell viability and tumor growth; however, it significantly inhibited the cytotoxic activity of oxaliplatin in vitro. In addition, we observed that oxaliplatin-induced the activation of ERK; however, IgG inhibits ERK activity. This implies that the application of therapeutical IgG to treating patients, which are receiving oxaliplatin, might have harmful side effects.

The present study was conducted to investigate the parasite fauna of four freshwater fish species (perch, Perca fluviatilis bream, Abramis brama; roach, Rutilus rutilus and rudd, Scardinius erythrophthalmus) from Lake Tollense, Mecklenburg-Western Pomerania (MV) during 2018-2019. In addition, parasitological raw data of similar freshwater fish species from Lake Malchin and Lake Hohen Sprenz were obtained from Pikalov (2017) for analysis and comparison. Finally, we studied the direct transfer of parasites from Lake Tollense into an open raceway flow-through aquaculture system., Die vorliegende Dissertation befasst sich mit der Untersuchung der Parasitenfauna von vier Süßwasserfischarten (Flussbarsch, Perca fluviatilis; Brasse, Abramis brama; Rotauge, Rutilis rutilus und Rotfeder, Scardinius erythrophthalmus) aus dem Tollensesee 2018-2019, Mecklenburg-Vorpommern (MV). Die erzielten Ergebnisse wurden mit parasitologische Rohdaten von Pikalov (2017) von ähnlichen Süßwasserfischarten aus dem Malchiner See und dem Hohen Sprenzer See verglichen. Zusätzlich wurde der direkte Transfer von Parasiten aus dem Tollensesee in eine offene Durchfluss-Aquakulturanlage untersucht.

Ziel dieser tierexperimentellen Studie an Thy1-YFP Mäusen war es, die morphologischen Veränderungen von kornealen und intraepidermalen Nervenfasern nach Induktion eines Diabetes mellitus zu untersuchen. In vivo wurden die kornealen Nervenfasern mittels corneal confocal Laser-Scanning Mikroskopie (CLSM) untersucht als auch ex vivo mittels Fluoreszenzmikroskopie. Korneale Nervenfasern der diabetischen Tiere degenerierten und regenerierten sich frühzeitig, während intraepidermale Nervenfasern morphologisch nach vierwöchiger Diabetes-Dauer und 8-wöchiger Insulintherapie unbeeinträchtigt blieben.

This study aimed to optimize the use of plant-based culture media for exploration of the plant-associated microbiota and extend the culturability borders to isolate novel bacterial taxa and meticulously characterize the culture-dependent and culture-independent microbiomes in view of the developed plant-based culture media. Throughout the study, we could prove that plant-based culture media are able to support the recovery of previously uncultured plant-associated bacteria, and also support the enrichment of bacterial lineages that belong to the candidate phyla radiation (CPR).

In den hier zusammengefassten Arbeiten wurden die Mechanismen der interzellulären Koordinierung für die Steuerung der Kraft- bzw. Druckentwicklung glattmuskulärer Organe sowie ihre Relevanz im Kontext von Erkrankungen des Nervensystems untersucht. Die erhobenen Befunde belegen, dass Veränderungen der Synchronisation innerhalb des Gewebes drastische Änderungen der Kraftentwicklung auslösen. Die experimentell dargestellte Fragilität der interzellulären Kopplung legt nahe, dass Funktionsstörungen des Nervensystems auch im klinischen Kontext gastrointestinale Funktionen beeinträchtigen können.

Die Salzstressantwort von Clostridium acetobutylicum wurde untersucht, um Schutzsubstanzen und Schlüsselgene zur Erhöhung der Butanolbildung in salzhaltigem Medium zu identifizieren. Arginin zeigte einen positiven Effekt auf das Wachstum unter Salzstress. Die Argininbiosynthese von C. acetobutylicum wurde durch Inaktivierung des entsprechenden Repressors gesteigert und die Butanolbildung signifikant erhöht. Transkriptomdaten zeigten eine Vernetzung der Aminosäurebiosynthesen und Stressantworten in C. acetobutylicum. Dies bietet Ansatzpunkte für die weitere Optimierung der Butanolfermentation., The salt stress response of Clostridium acetobutylicum was analyzed to identify protective substances and key genes to increase solvent formation in saline medium. Arginine showed a positive effect on growth under salt stress. The arginine biosynthesis of C. acetobutylicum was enhanced by inactivating the corresponding repressor, significantly increasing butanol production. The transcriptome revealed interlinked amino acid biosyntheses and a connection between different stress responses in C. acetobutylicum, offering starting points for the further optimization of biobutanol fermentation.

Microbial communities react rapidly and specifically to changing environments, indicating distinct microbial fingerprints for a given environmental state. Machine learning with community data predicted the Baltic Sea-detected pollutants glyphosate and 2,4,6-trinitrotoluene, using the developed R package “phyloseq2ML”. Predictions by Random Forest and Artificial Neural Network were accurate. Relevant taxa were identified. The interpretability of machine learning models was found of particular importance. Microbial communities predicted even minor influencing factors in complex environments.

The ever-increasing amount and diversity of biological and medical data is a major challenge in computational analyses. Computational methods have to be combined into analysis workflows for seamless, swift, and transparent computation. In this work, numerous workflows were developed for the general processing of bulk RNA sequencing (RNA-Seq), single-cell sequencing experiments, and non-coding RNA identification. Mathematical concepts of machine learning and univariate meta-analyses were successfully implemented to independently investigate the role of cell therapies in cardiac regeneration.

Ovarian follicles are exposed to stressors such as higher levels of non-esterified free fatty acids (NEFAs) or hypoxia. High follicular levels of NEFA severely affect granulosa cells (GCs) by targeting key genes of estradiol biosynthesis. Cultured GCs exposed to hypoxia showed specific regulation of genes that initiate luteinization. These effects were partly mediated by hypoxia inducible factor1 (HIF1). Thus, ovarian follicles are susceptible to elevated NEFA levels, but cope up with severe hypoxia prevailing during late folliculogenesis through specific transcription factors such as HIF1.

This study evaluates the repression activities of two DUF3669-containing KRAB-ZNF proteins; ZNF746 and ZNF777 in addition to the oligomerizing ability of their domain of unknown function (DUF3669). A new domain model for DUF3669 has been established in this study. DUF3669 as an oligomerizing domain was confirmed in our study. Our data revealed the adding functionalities for the repression activities of ZNF746; DUF3669 as an oligomerizing domain might recruit other DUF3669-KRAB-ZNF proteins and the SUMOylation might stabilize the repression complex and directly recruit chromatin modifiers.

Es konnte gezeigt werden, dass das Zusammenwirken beider mtDNA-Punktmutationen im mtBPL-Mausstamm den Leberstoffwechsel signifikant beeinflusst. Das untersuchte Modell liefert damit für den Menschen relevantes Wissen zu mitochondrial getriggerten Alterungsprozessen.

Mit Hilfe eines Kartoffel-Assoziationspanels konnten SSR-Marker mit Trockentoleranzbezug identifiziert werden. Diese und weitere publizierte SSR- sowie AFLP-Primerpaare wurden für die Kartierung zweier F1-Kartoffelpopulationen eingesetzt, um Genkarten zu erstellen. Mittels der Genkarten sowie Ertragsdaten (Stärkeertrag und -gehalt, Knollenfrischgewicht), ausgewählter Transkript- und Metabolit-Daten sowie Daten für einen Trockentoleranzindex DRYM wurden QTL-Analysen durchgeführt. Dabei konnten eine Vielzahl an QTL generiert und trockentoleranzrelevante Bereiche identifiziert werden.

The study of laterality (i.e. asymmetries of brain and behaviour) is a potential non-invasive approach to gain insights into the common neural mechanisms underpinning both personality and emotions in animals. The hypotheses underlying this thesis state that the left (respectively the right) hemisphere regulates approach or positive (respectively avoidance or negative) emotions. The goal of this thesis was to study lateralized motor functions (Study 1) and their associations with personality indices (Study 2), and the effect of monocular viewing on emotional reactions (Study 3) in pigs.

W��hrend einer Tumortherapie mit Protonenstrahlen kommt es zur Exposition gesunden Gewebes au��erhalb des Zielgebietes mit sekund��ren Neutronen (Streustrahlung). Diese Arbeit besch��ftigt sich mit dem Risiko der Entstehung neuer Tumoren als Sp��tfolge dieser Neutronenexposition. Zur Untersuchung der radiobiologischen Effekte der Streustrahlung wurden gesunde humane Mammaepithelzellen mit Neutronen verschiedener Energiespektren bestrahlt. Diese Ergebnisse sind f��r weitere Untersuchungen strahleninduzierter Karzinogenese und f��r eine bessere Risikobewertung von hoher Relevanz.

Within breast tissue, adipose-derived stem cells (ADSCs) are responsible for maintaining cellular homeostasis. However, this potential can also contributed to the undesirable effect of tumor promotion. Therefore, this work deals with the effect of ionizing radiation on these interactions. In summary, the promoting effects of ADSCs on MCF-7 breast cancer cells were neutralized or reversed by a radiation treatment. Furthermore, ADSCs exhibit a medium radiation sensitivity associated with a functional DNA repair system., Adipose-derived stem cells (ADSCs) des Brustgewebes sind für die Aufrechterhaltung der zellulären Homöostase verantwortlich. Dieses Potenzial kann jedoch auch zum unerwünschten Effekt der Tumorförderung beitragen. Diese Arbeit befasst sich daher mit der Wirkung ionisierender Strahlung auf diese Wechselwirkungen. Hierbei wurden die fördernden Effekte von ADSCs auf MCF-7-Brustkrebszellen durch eine Bestrahlung neutralisiert oder umgekehrt. Im Weiteren weisen ADSCs eine mittlere Strahlungsempfindlichkeit auf, verbunden mit einem funktionierenden DNS Reparatursystem.

Die Rolle der Klasse der langen nichtkodierenden RNA (lncRNA) bei der Merkmalsausprägung von Nutztieren ist bisher wenig erforscht. In dieser Dissertation wurden in einem integrativen Ansatz das Transkriptom, Phänom und Metabolom von Bullen (N = 24) und Kühen (N = 24) einer Kreuzungspopulation (Fleischrind x Milchvieh) der zweiten Generation analysiert, um neue, unbekannte lncRNAs und ihre möglichen biologischen Funktionen und ihren Einfluss auf die Nährstoffverteilung zu ermitteln. Die Ergebnisse haben zur weiterten Annotation des Rindergenoms beigetragen.

Nitrogen fixation (diazotrophy) is an important input pathway of nitrogen to surface oceans. Diazotrophy can occur in physicochemically complex waters, requiring new tools for assessing diazotroph biogeography and influence. I present a simple method for delineating habitats and pair it with a multi-scale ecosystem assessment of diazotroph impacts in the South China Sea (SCS). This strategy identified clear habitat-specific trends in diazotroph influence across the plankton food web, indicating that diazotrophy is more prevalent in the SCS than previously thought.

Das entz��ndliche Mikroklima im Tumorgewebe und dem umgebenden Gewebe beeinflusst unter anderem die Entstehung, das Wachstum und die Metastasierung von Tumoren. Dieses Mikroklima kann durch ionisierende Strahlung beeinflusst werden, welche sich je nach Bestrahlungsdosis anti- oder proinflammatorisch auswirkt. Zudem ist die Interaktion zwischen Tumor- und Endothelzellen in Hinblick auf die Metastasierung des Tumors entscheidend. Ziel dieser Arbeit war es, in verschiedenen In-vitro-Untersuchungen die Auswirkungen der ionisierenden Strahlung auf diese Zellinteraktionen hin zu untersuchen.

Aberrant behaviour of resident cells causes several illnesses. Among them, cardiovascular diseases are the leading cause of death in the western world. The human heart possesses only negligible myocardial regeneration capacity after a myocardial infarction, thus, it cannot compensate the massive cell loss self-reliantly. The thesis addresses the optimization of different cell biological approaches for enrichment of cardiac subtypes. Moreover, the thesis demonstrates different genetic manipulation strategies (DNA-, mRNA- and microRNA-based) for an efficient modulation of cell fate.

Die Gene TAC1 und LAZY1 beeinflussen die Pflanzenarchitektur im gesamten Pflanzenreich maßgeblich. Bei der Weinrebe ist eine aufrechte Triebhaltung für die Rebenerziehung von großem Vorteil. Über einen Kandidatengenansatz wurde bei Referenzsorten und S1-Populationen der Weinrebe der Einfluss von VviTAC1 und VviLAZY1 auf die Ausbildung der Triebhaltung mittels der Bonitur des Pflanzenmaterials sowie einer Charakterisierung der Kandidatengene auf DNA-, RNA und Proteinebene untersucht. Dies ermöglichte eine erstmalige und umfassende Charakterisierung von VviTAC1 und VviLAZY1 bei der Weinrebe.

Das Glycine Cleavage System (GCS) ist ein aus 4 Proteinen bestehendes und für alle Eukaryoten essentielles Multienzymsystem. Es wandelt Glycin in Serin um und ist in der Lage, in Erbsen-Pflanzen unter hohen Konzentrationen einen Multienzymkomplex zu bilden. Untersuchungen am Modell-Cyanobakterium Synechocystis sp. PCC 6803 haben für Cyanobakterien erstmals gezeigt, dass diese 4 Proteine auch hier in der Lage sind, einen relativ stabilen Komplex zu bilden. Somit kann das cyanobakterielle GCS als Modell für zukünftige Struktur- und Funktionsanalysen des GCS genutzt werden., The glycine cleavage system (GCS) is essential for all eukaryotes. It consists of four proteins and converts glycine to serine. Early research from plants showed complex formation of these proteins under high concentrations in vitro but not in vivo. Using the very similar proteins of the GCS from the model cyanobacteria Synechocystis sp. PCC 6803 we analysed if complex formation was possible for organisms other than plants. The analysis showed complex formation of the Synechocystis proteins. Therefore, this GCS can be used as a model for further structure and function analysis.

Horses have a low reproductive performance compared to other species. The aim of this study was to evaluate different sperm quality parameters in groups of fertile and subfertile stallions during the breeding and non-breeding season. Furthermore, molecular processes in response to different seasonal conditions as well as those are associated with male fertility were investigated. The comparison of transcript abundance between fertile and subfertile stallions and between breeding and non-breeding seasons provided a glimpse of the various functional pathways involved in stallion fertility.

Morbus Niemann-Pick Typ C1 (NPC1) ist eine seltene autosomal rezessive, lysosomale Lipidose, die sich durch eine Akkumulation von Cholesterol in den Lysosomen auszeichnet. Ziel der Arbeiten war es, ein humanes neuronales in vitro Zellmodellsystem für NPC1 zu entwickeln. Hierzu wurden NPC1 patientenspezifische Fibroblasten in patientenspezifische IPS-Zellen reprogrammiert. Diese wurden in Neurone und Gliazellen differenziert und hinsichtlich der Ausprägung von pathophysiologischen Merkmalen, als Beleg für die Verwendbarkeit der Zellen als NPC1 in vitro Modellsystem, untersucht.

Riechstörungen treten häufig bei neurodegenerativen Erkrankungen auf. In dieser Arbeit konnten morphologische und immunhistochemische Auswirkungen von Morbus Niemann Pick Typ C1 (NPC1) auf das olfaktorische System in einem NPC1-Mausmodell nachgewiesen werden, insbesondere auf die Regenerationsfähigkeit des olfaktorischen Epithels und des Bulbus olfactorius. Die Kombination von Miglustat, HPßCD und Allopregnanolon sowie die Monotherapie mit HPßCD zeigten eine signifikante Reduktion der Morphopathologie und eine Normalisierung der Riechfähigkeit in einem olfaktorischen Verhaltenstest.
