We report the demonstration of optical compression of an electron beam and the production of controllable trains of femtosecond, soft x-ray pulses with the Linac Coherent Light Source (LCLS) free-electron laser (FEL). This is achieved by enhanced self-amplified spontaneous emission with a 2 μm laser and a dechirper device. Optical compression was achieved by modulating the energy of an electron beam with the laser and then compressing with a chicane, resulting in high current spikes on the beam which we observe to lase. A dechirper was then used to selectively control the lasing region of the electron beam. Field autocorrelation measurements indicate a train of pulses, and we find that the number of pulses within the train can be controlled (from 1 to 5 pulses) by varying the dechirper position and undulator taper. These results are a step toward attosecond spectroscopy with x-ray FELs as well as future FEL schemes relying on optical compression of an electron beam.

We present the new drive laser system for the photo-injector of the LCLS-II XFEL at SLAC, including the first commissioning results and challenges encountered due to high power, high repetition rate ultraviolet laser operation.

New high-repetition-rate x-ray free electron lasers (XFELs) require for their operation highly reliable ultrafast laser systems with high pulse energy, high repetition rate, and high average power. In this Letter, we present high-average-power scaling of near-infrared optical parametric chirped pulse amplification (OPCPA) in potassium titanyl arsenate (KTA) with tunable center wavelengths from 1.5 to 2.0 μm. Using a three-stage amplification scheme and a kW-level InnoSlab Yb:YAG pump amplifier for the final non-collinear KTA stage, we demonstrate an amplified output power of 106.2 W at a center wavelength of 1.75 μm and 100 kHz. Description of the Coherent X-ray Imaging (CXI) instrument at the Linac Coherent Light Source. Recent scientific highlights illustrate the femtosecond crystallography, high power density and extreme matter capabilities of the CXI instrument., The Coherent X-ray Imaging (CXI) instrument specializes in hard X-ray, in-vacuum, high power density experiments in all areas of science. Two main sample chambers, one containing a 100 nm focus and one a 1 µm focus, are available, each with multiple diagnostics, sample injection, pump–probe and detector capabilities. The flexibility of CXI has enabled it to host a diverse range of experiments, from biological to extreme matter.

The LCLS X-Ray FEL at SLAC typically delivers one bunch at the time. Different schemes of two pulses have been developed: Two bucket, Twin bunch, split undulator, and fresh slice. The photocathode rf gun is one of the most critical components in x-ray free electron lasers. The drive laser strikes the photocathode surface, which emits electrons with properties that depend on the shape of the drive laser. Most free electron lasers use photocathodes with work function in the ultraviolet, a wavelength where direct laser manipulation becomes challenging. In this paper, we present a novel application of a digital micromirror device (DMD) for the 253 nm drive laser at the Linear Coherent Light Source. Laser profile shaping is accomplished through an iterative algorithm that takes into account shaping error and efficiency. Next, we use laser shaping to control the X-ray laser output via an online optimizer, which shows improvement in FEL pulse energy. Lastly, as a preparation for electron beam shaping, we use the DMD to measure the photocathode quantum efficiency across cathode surface with an averaged laser rms spot size of $59\text{ }\text{ }\ensuremath{\mu}\mathrm{m}$. Our experiments demonstrate promising outlook of using DMD to shape ultraviolet lasers for photocathode rf guns with various applications.

We present a 100 kHz, sub-20 fs optical parametric chirped-pulse amplifier (OPCPA) system delivering 88.6 W average power at a center wavelength of 800 nm. The seed pulses are derived from the pump laser via white-light continuum generation and are amplified in three non-collinear OPCPA stages. The final two high-power stages are pumped with a 661 W Yb:YAG InnoSlab amplifier. A simple and robust design is used for the OPCPA system to avoid thermal effects and enhance long-term stability, resulting in excellent beam quality and high conversion efficiency. To the best of our knowledge, this is the highest average power OPCPA system reported to date.

In the Linear Coherent Light Source (LCLS) at SLAC, the injector laser plays an important role as the source of the electron beam for the Free Electron Laser (FEL). The beam emittance and FEL performance are highly related to the transverse shape of the injector laser. When the injector laser has hot spots and non-uniformities that can carry over to the electron beam and degrade electron emittance and FEL performance, it requires long hours of manual adjustment by laser engineers and strenuous machine tuneup. The injector laser shaping project at LCLS aims to have precise control of the driver laser transverse profile in order to produce arbitrary electron beam profiles, which will enable us to study effects of laser shape on beam emittance and FEL performances. We use a digital micromirror device (DMD) to manipulate the drive laser profile. In this paper, we briefly discuss the implementations of laser shaping at LCLS. We demonstrate two applications of laser shaping. We present results of using laser shaping to control the X-ray laser output via an online optimizer. In the Linear Coherent Light Source (LCLS) at SLAC, the injector laser plays an important role as the source of the electron beam for the Free Electron Laser (FEL). The emittance of the beam is highly related to the transverse profile of the injector laser. Currently the LCLS injector laser has undesired features, such as hot spots, which carry over to the electron beam. These undesired features increase electron emittance, degrade the FEL performance, and complicate operations. The injector laser shaping project at LCLS aims to produce arbitrary electron beam profiles, such as cut-Gaussian, uniform, and parabolic, and to study the effect of spatial profiles on beam emittance and FEL performance. Effectively it also allows easy transition between the two spare lasers, where the operators can use the spatial shaper to achieve identical profiles for the two lasers. An electrospun liquid microjet has been developed that delivers protein microcrystal suspensions at flow rates of 0.14–3.1 µl min−1to perform serial femtosecond crystallography (SFX) studies with X-ray lasers. Thermolysin microcrystals flowed at 0.17 µl min−1and diffracted to beyond 4 Å resolution, producing 14 000 indexable diffraction patterns, or four per second, from 140 µg of protein. Nanoflow electrospinning extends SFX to biological samples that necessitate minimal sample consumption.

The LCLS hard X-ray Free Electron Laser at SLAC reported first lasing in April 2009. Since then two successful user runs have been completed at the two soft X-ray stations. The first hard X-ray station has started commissioning in July 2010. Beam diagnostics play an essential role for tuning the machine and delivering the requested beam properties to the users. An overview of the LCLS photon diagnostics will be presented including some selected commissioning results. Plans for future improvements and upgrades will be briefly discussed.

The Linac Coherent Light Source free-electron laser has now achieved coherent X-ray generation down to a wavelength of 1.2 A and at a brightness that is nearly ten orders of magnitude higher than conventional synchrotrons. Researchers detail the first operation and beam characteristics of the system, which give hope for imaging at atomic spatial and temporal scales.

The X-ray free-electron laser has opened a new era for photon science, improving the X-ray brightness by ten orders of magnitude over previously available sources. Similar to an optical laser, the spectral and temporal structure of the radiation pulses can be tailored to the specific needs of many experiments by accurately manipulating the lasing medium, that is, the electron beam. Here we report the generation of mJ-level two-colour hard X-ray pulses of few femtoseconds duration with an XFEL driven by twin electron bunches at the Linac Coherent Light Source. This performance represents an improvement of over an order of magnitude in peak power over state-of-the-art two-colour XFELs. The unprecedented intensity and temporal coherence of this new two-colour X-ray free-electron laser enable an entirely new set of scientific applications, ranging from X-ray pump/X-ray probe experiments to the imaging of complex biological samples with multiple wavelength anomalous dispersion., Two-colour X-ray pulses from free-electron lasers can be used to probe ultrafast dynamics, but the total power is a fraction of the saturation power. Here, Marinelli et al. use twin electron bunches to reach full saturation power and increase the two-colour intensity by an order of magnitude at hard-X-ray energies.

This manuscript serves as a reference to describe the optical laser sources and capabilities at the Linac Coherent Light Source., Ultrafast optical lasers play an essential role in exploiting the unique capabilities of recently commissioned X-ray free-electron laser facilities such as the Linac Coherent Light Source (LCLS). Pump–probe experimental techniques reveal ultrafast dynamics in atomic and molecular processes and reveal new insights in chemistry, biology, material science and high-energy-density physics. This manuscript describes the laser systems and experimental methods that enable cutting-edge optical laser/X-ray pump–probe experiments to be performed at LCLS.

X-ray free-electron laser (XFEL) sources enable the use of crystallography to solve three-dimensional macromolecular structures under native conditions and free from radiation damage. Results to date, however, have been limited by the challenge of deriving accurate Bragg intensities from a heterogeneous population of microcrystals, while at the same time modeling the X-ray spectrum and detector geometry. Here we present a computational approach designed to extract statistically significant high-resolution signals from fewer diffraction measurements.

One Protein, Two Probes A central challenge in the use of x-ray diffraction to characterize macromolecular structure is the propensity of the high-energy radiation to damage the sample during data collection. Recently, a powerful accelerator-based, ultrafast x-ray laser source has been used to determine the geometric structures of small protein crystals too fragile for conventional diffraction techniques. Kern et al. (p. 491 , published online 14 February) now pair this method with concurrent x-ray emission spectroscopy to probe electronic structure, as well as geometry, and were able to characterize the metal oxidation states in the oxygen-evolving complex within photosystem II crystals, while simultaneously verifying the surrounding protein structure.

The ultrabright femtosecond X-ray pulses provided by X-ray free-electron lasers open capabilities for studying the structure and dynamics of a wide variety of systems beyond what is possible with synchrotron sources. Recently, this “probe-before-destroy” approach has been demonstrated for atomic structure determination by serial X-ray diffraction of microcrystals. There has been the question whether a similar approach can be extended to probe the local electronic structure by X-ray spectroscopy. To address this, we have carried out femtosecond X-ray emission spectroscopy (XES) at the Linac Coherent Light Source using redox-active Mn complexes. XES probes the charge and spin states as well as the ligand environment, critical for understanding the functional role of redox-active metal sites. Kβ 1,3 XES spectra of Mn II and Mn 2 III,IV complexes at room temperature were collected using a wavelength dispersive spectrometer and femtosecond X-ray pulses with an individual dose of up to >100 MGy. The spectra were found in agreement with undamaged spectra collected at low dose using synchrotron radiation. Our results demonstrate that the intact electronic structure of redox active transition metal compounds in different oxidation states can be characterized with this shot-by-shot method. This opens the door for studying the chemical dynamics of metal catalytic sites by following reactions under functional conditions. The technique can be combined with X-ray diffraction to simultaneously obtain the geometric structure of the overall protein and the local chemistry of active metal sites and is expected to prove valuable for understanding the mechanism of important metalloproteins, such as photosystem II.

Most of the dioxygen on earth is generated by the oxidation of water by photosystem II (PS II) using light from the sun. This light-driven, four-photon reaction is catalyzed by the Mn 4 CaO 5 cluster located at the lumenal side of PS II. Various X-ray studies have been carried out at cryogenic temperatures to understand the intermediate steps involved in the water oxidation mechanism. However, the necessity for collecting data at room temperature, especially for studying the transient steps during the O–O bond formation, requires the development of new methodologies. In this paper we report room temperature X-ray diffraction data of PS II microcrystals obtained using ultrashort (4 CaO 5 cluster in PS II at room temperature. We show that these data are comparable to those obtained in synchrotron radiation studies as seen by the similarities in the overall structure of the helices, the protein subunits and the location of the various cofactors. This work is, therefore, an important step toward future studies for resolving the structure of the Mn 4 CaO 5 cluster without any damage at room temperature, and of the reaction intermediates of PS II during O–O bond formation.

The Linac Coherent Light Source (LCLS) is an x-ray Free-Electron Laser (FEL) project that has just achieved its first lasing at 1.5 {angstrom} radiation wavelength. The very bright electron beam required to drive this FEL is susceptible to a microbunching instability in the magnetic bunch compressors that may increase the slice energy spread beyond the FEL tolerance. To control the slice energy spread and to suppress the microbunching instability, a laser heater (LH) system is installed in the LCLS injector area at 135 MeV, right before the RF deflector that is used for the time-resolved electron diagnostics. This unique component is used to add a small level of intrinsic energy spread to the electron beam in order to Landau damp the microbunching instability before it potentially breaks up the high brightness electron beam. The system was fully installed and tested in the fall of 2008, and effects of heating on the electron beam and the x-ray FEL were studied during the 2009 commissioning period. The laser heater system is composed of a 4-dipole chicane; a 9-period, planar, permanent-magnet, adjustable-gap undulator at the center of the chicane; one OTR screen on each side of the undulator for electron/laser spatial alignment; and more » an IR laser (up to 15-MW power) which co-propagates with the electron beam inside the undulator generating a 758-nm energy modulation along the bunch. The final two dipoles of the 4-dipole chicane time-smear this modulation leaving only a thermal-like intrinsic energy spread within the bunch. Table 1 lists the main parameters for this system. The last half of the chicane time-smears the energy modulation leaving an effective thermal energy spread increase. We present the first commissioning results of this system, its operational issues, its impact on the microbunching instability, and finally its effect on the FEL performance. « less

The start-up of the Linac Coherent Light Source (LCLS), the new femtosecond hard X-ray laser facility in Stanford, California, has brought high expectations of a new era for biological imaging. The intense, ultrashort X-ray pulses allow diffraction imaging of small structures before radiation damage occurs. Two papers in this issue of Nature present proof-of-concept experiments showing the LCLS in action. Chapman et al. tackle structure determination from nanocrystals of macromolecules that cannot be grown in large crystals. They obtain more than three million diffraction patterns from a stream of nanocrystals of the membrane protein photosystem I, and assemble a three-dimensional data set for this protein. Seibert et al. obtain images of a non-crystalline biological sample, mimivirus, by injecting a beam of cooled mimivirus particles into the X-ray beam. The start-up of the new femtosecond hard X-ray laser facility in Stanford, the Linac Coherent Light Source, has brought high expectations for a new era for biological imaging. The intense, ultrashort X-ray pulses allow diffraction imaging of small structures before radiation damage occurs. This new capability is tested for the problem of imaging a non-crystalline biological sample. Images of mimivirus are obtained, the largest known virus with a total diameter of about 0.75 micrometres, by injecting a beam of cooled mimivirus particles into the X-ray beam. The measurements indicate no damage during imaging and prove the concept of this imaging technique. X-ray lasers offer new capabilities in understanding the structure of biological systems, complex materials and matter under extreme conditions1,2,3,4. Very short and extremely bright, coherent X-ray pulses can be used to outrun key damage processes and obtain a single diffraction pattern from a large macromolecule, a virus or a cell before the sample explodes and turns into plasma1. The continuous diffraction pattern of non-crystalline objects permits oversampling and direct phase retrieval2. Here we show that high-quality diffraction data can be obtained with a single X-ray pulse from a non-crystalline biological sample, a single mimivirus particle, which was injected into the pulsed beam of a hard-X-ray free-electron laser, the Linac Coherent Light Source5. Calculations indicate that the energy deposited into the virus by the pulse heated the particle to over 100,000 K after the pulse had left the sample. The reconstructed exit wavefront (image) yielded 32-nm full-period resolution in a single exposure and showed no measurable damage. The reconstruction indicates inhomogeneous arrangement of dense material inside the virion. We expect that significantly higher resolutions will be achieved in such experiments with shorter and brighter photon pulses focused to a smaller area. The resolution in such experiments can be further extended for samples available in multiple identical copies.

In this paper, we report detailed measurements of laser-heater-induced energy spread, including the unexpected self-heating phenomenon when the laser energy is very low. We discuss the suppression of the microbunching instability with the laser heater and its impact on the x-ray FEL performance. We also present the analysis of these experimental results and develop a three-dimensional longitudinal space charge model to explain the self-heating effect.
