Abstract: Introduction: Connected landscapes can increase the effectiveness of protected areas by facilitating individual movement and gene flow between populations, thereby increasing the persistence of species even in fragmented habitats. Connectivity planning is often based on modeling connectivity for a limited number of species, i.e., “connectivity umbrellas”, which serve as surrogates for co-occurring species. Connectivity umbrellas are usually selected a priori, based on a few life history traits and often without evaluating other species. Methods: We developed a quantitative method to identify connectivity umbrellas at multiple scales. We demonstrate the approach on the terrestrial large mammal community (24 species) in continental Europe at two scales: 13 geographic biomes and 36 ecoregions, and evaluate the interaction of landscape characteristics on the selection of connectivity umbrellas. Results: We show that the number, identity, and attributes of connectivity umbrellas are sensitive to spatial scale and human influence on the landscape. Multiple species were selected as connectivity umbrellas in 92% of the geographic biomes (average of 4.15 species) and 83% of the ecoregions (average of 3.16 species). None of the 24 species evaluated is by itself an effective connectivity umbrella across its entire range. We identified significant interactions between species and landscape attributes. Species selected as connectivity umbrellas in regions with low human influence have higher mean body mass, larger home ranges, longer dispersal distances, smaller geographic ranges, occur at lower population densities, and are of higher conservation concern than connectivity umbrellas in more human-influenced regions. Abstract: The problem of data-driven control is addressed here in the context of switched affine systems. This class of nonlinear systems is of particular importance when controlling many types of applications in electronic, biology, medicine and so forth. Still in the view of practical applications, providing an accurate model for this class of systems can be a hard task, and it might be more relevant to work on data issued from some trajectories obtained from experiments and to deploy a new branch of tools to stabilize the systems that are compatible with the processed data. Following the recent concept of data-driven control design, this paper first presents a generic equivalence lemma that shows a matrix constraint based on data, instead of the system parameter. Then, following the concept of robust hybrid limit cycles for uncertain switched affine systems, robust model-based and then data-driven control laws are designed based on a Lyapunov approach. The proposed results are then illustrated and evaluated on an academic example.

Abstract: Dilatancy plays a key role in mixtures of grains and fluid but is poorly investigated in dry granular flows. These flows may however dilate by more than 10% in granular column collapses. We investigate here dilatancy effects in dry flows with a shallow depth-averaged model designed to be further applied to simulate natural landslides. We use a compressible , rheology with a dilatancy law, where is the volume fraction at the equilibrium (i.e. critical) state and I the so-called inertial number. This law is obtained by simply removing the fluid phase in the solid/fluid model of our previous work (Bouchut et al., 2016) [7] and derived from critical state theory. A numerical method is proposed to solve the equations, that have however singularities that are rather difficult to handle. Simulations of granular collapses on horizontal and sloping beds show that the maximum height of the deposits changes as a function of the initial volume fraction with higher (lower) deposits for initially denser (looser) granular masses, as observed with Discrete Element simulations. The front position and the deposit shape behind it are on the contrary poorly affected by the initial volume fraction, as if the flow had almost forgotten its initial state. However subtle effects can be observed with the occurrence of low velocity regimes on steep slopes that strongly depend on the initial volume fraction. Simulations show complex compression/dilation effects during the flow, in particular with front dilation (compression) during the acceleration (deceleration) phases. These effects may dramatically change the effective friction that is observed to decrease at the front in some situations, while the rheology without dilatancy would have predicted an increasing friction. The model predicts an increasing dilation of the mass for increasing slopes by up to 10% in the studied configurations, in agreement with laboratory experiments. Abstract: This paper focuses on designing a robust control law to manage the demand response of islanded microgrids composed of shifting and adjusting loads. On one side, Hybrid Dynamical System theory is suited here, because the microgrid model is composed of continuous-time dynamics (the energy stored in the battery and the adjustable loads), and discrete-time dynamics (the shifting loads). On the other side, Multi Agent System theory is used to control the adjusting loads in order to guarantee a consensus between them. Hence, non-uniform convergence of the State of Charge of the battery to a given reference is ensured. Robustness with respect to plug and play of any load and parameter variations is also ensured. Experimental results from a laboratory-scale microgrid validate the approach.

Abstract: There exists in the literature a huge range of geophysical applications that have been modeled trough two-layer or two-phase models. In this work first some averaged two-layer and two-phase models are presented. We focus on applications to submarine avalanches, debris flows and sediment transport in rivers. Secondly, their numerical approximation by a finite volume method is discussed and a numerical test is presented.

Abstract: International audience; Objectives:Broadly neutralizing antibodies have been proposed as key actors for HIV vaccine development. However, they display features of highly matured antibodies, hampering their induction by vaccination. As protective broadly neutralizing antibodies should be induced rapidly after vaccination and should neutralize the early-transmitted founder (T/F) viruses, we searched whether such antibodies may be induced following HIV infection.Design:Sera were collected during acute infection (Day 0) and at viral set point (Month 6/12) and the neutralizing activity against T/F strains was investigated. Neutralizing activity in sera collected from chronic progressor was analyzed in parallel.Methods:We compared neutralizing activity against T/F strains with neutralizing activity against non-T/F strains using the conventional TZM-bL neutralizing assay.Results:We found neutralizing antibodies (nAbs) preferentially directed against T/F viruses in sera collected shortly after infection. This humoral response evolved by shifting to nAbs directed against non-T/F strains.Conclusion:Although features associated with nAbs directed against T/F viruses need further investigations, these early-induced nAbs may display lesser maturation characteristics; therefore, this might increase their interest for future vaccine designs.

Abstract: The stability and accuracy of atomic devices can be degraded by the evolution of their cell inner atmosphere. Hence, the undesired entrance or leakage of background or buffer gas, respectively, that can permeate through the cell walls, should be slowed down. In this work, we investigate helium permeation in microfabricated alkali vapor cells filled with He and whose windows are made of borosilicate glass (BSG) or aluminosilicate glass (ASG). The permeation is then derived from routine measurements of the pressure-shifted hyperfine transition frequency of an atomic clock. We first confirm that ASG reduces He permeation rate by more than two orders of magnitude, in comparison with BSG. In addition, we demonstrate that Al$_2$O$_3$ thin-film coatings, known to avoid alkali consumption in vapor cells, can also significantly reduce He permeation. The permeation through BSG is thereby reduced by a factor 110 whereas the one through ASG is decreased by a factor up to 5.8 compared to uncoated substrates. These results may contribute to the development of miniaturized atomic clocks and sensors with improved long-term stability or sensitivity., Comment: 7 pages, 5 figures

All scripts and additional tables as well as all cluster phylogeny figures are available under the github repository : (BenjaminGuinet/Viral-domestication-among-Hymenoptera).; International audience; The accidental endogenization of viral elements within eukaryotic genomes can occasionally provide significant evolutionary benefits, giving rise to their long-term retention, that is, to viral domestication. For instance, in some endoparasitoid wasps (whose immature stages develop inside their hosts), the membrane-fusion property of viruses has been repeatedly domesticated following the ancestral endogenizations of double-stranded DNA viruses. The endogenized genes provide female wasps with a delivery tool to inject virulence factors that are essential to the developmental success of their offspring. Because all known cases of viral domestication involve endoparasitic wasps, we hypothesized that this lifestyle, relying on a close interaction between individuals, may have promoted the endogenization and domestication of viruses. By analyzing the composition of 124 Hymenoptera genomes, spread over the diversity of this clade and including free-living, ecto- and endoparasitoid species, we tested this hypothesis. Our analysis first revealed that double-stranded DNA viruses, in comparisons with other viral genomic structures (ssDNA, dsRNA, ssRNA), are more often integrated (that is, endogenized) and domesticated (that is, retained by selection) than expected from their estimated abundance in insect viral communities. Secondly, our analysis indicates that the rate at which dsDNA viruses are endogenized is higher in endoparasitoids than in ectoparasitoids or free-living hymenopterans, which also translates into more frequent events of domestication. Hence, these results are consistent with the hypothesis that the endoparasitoid lifestyle has facilitated the endogenization of dsDNA viruses, in turn increasing the opportunities of domestications that now play a central role in the biology of many parasitoid lineages.

Abstract: Type II secretion systems (T2SS) allow diderm bacteria to secrete hydrolytic enzymes, adhesins or toxins important for growth and virulence. In T2SS, secretion of folded proteins from the periplasm to the cell surface requires assembly of periplasmic filaments called pseudopili. Like the related type IV pili, pseudopili are polymerized in the inner membrane through addition of subunits at the filament base, mediated by the essential assembly platform (AP). To understand the structure and molecular role of the AP, we focused on its components PulL and PulM from the Klebsiella oxytoca T2SS. By combining biophysical methods, NMR and X-ray crystallography we studied the structure and associations of their periplasmic domains. We describe the first structure of the heterodimer complex formed by the PulL and PulM ferredoxin-like domains and show how their structural complementarity and plasticity favor their association during the secretion process. Cysteine scanning and cross-linking of transmembrane segments provided additional constraints to build a structural model of the PulL–PulM complex and assembly in the cellular context. Together with the relative abundance of PulL, PulM and their partners our findings suggest a model of the AP as a dynamic hub that orchestrates pseudopilus polymerization.

Abstract: Introduction In the general population, urinary sodium-to-potassium (uNa/K) ratio associates more strongly with high blood pressure (BP) than either urinary sodium or potassium alone. Whether this is also the case among patients with chronic kidney disease (CKD) is unknown. Methods We studied the associations of spot urine sodium-to-creatinine (uNa/Cr), potassium-to-creatinine (uK/Cr), and uNa/K ratios with a single office BP reading in 1660 patients with moderate to severe CKD at inclusion in the CKD-REIN cohort. Results Patients' median age was 68 (interquartile range [IQR], 59–76) years; most were men (65%), had moderate CKD (57%), and albuminuria (72%). Mean systolic and diastolic BP was 142/78 mm Hg. Spot uNa/Cr and uNa/K ratios were positively associated with systolic, mean arterial, and pulse pressures. The mean adjusted difference in systolic BP between the highest and the lowest quartile (Q4 vs. Q1) was 4.24 (95% confidence interval [CI], 1.53–6.96) mm Hg for uNa/Cr and 4.79 (95% CI, 2.18–7.39) mm Hg for uNa/K. Quartiles of spot uK/Cr were not associated with any BP index. The higher the quartile of uNa/K, the higher the prevalence ratio of uncontrolled (Q4 vs. Q1, 1.43; 95% CI, 1.19–1.72) or apparently treatment-resistant hypertension (Q4 vs. Q1, 1.35; 95% CI, 1.14–1.60). Findings were consistent in a subset of 803 individuals with 2 BP readings. Conclusion In patients with CKD, higher urinary sodium excretion is associated with higher BP, but unlike in general population, lower potassium excretion is not. Urinary Na/K does not add significant value in assessing high BP risk, except perhaps for hypertension control assessment., Graphical abstract

Abstract: T cell receptor (TCR) activation is modulated by mechanisms such as TCR endocytosis, which is thought to terminate TCR signalling. Here we show that, upon internalization, TCR continues to signal from a set of specialized endosomes that are crucial for T cell functions. Mechanistically, TCR ligation leads to clathrin-mediated internalization of the TCR-CD3ζ complex, while maintaining CD3ζ signalling, in endosomal vesicles that contain the insulin responsive aminopeptidase (IRAP) and the SNARE protein Syntaxin 6. Destabilization of this compartment through IRAP deletion enhances plasma membrane expression of the TCR-CD3ζ complex, yet compromises overall CD3ζ signalling; moreover, the integrity of this compartment is also crucial for T cell activation and survival after suboptimal TCR activation, as mice engineered with a T cell-specific deletion of IRAP fail to develop efficient polyclonal anti-tumour responses. Our results thus reveal a previously unappreciated function of IRAP-dependent endosomal TCR signalling in T cell activation., T cell receptors (TCR) are internalized when activated by their ligands. Here the authors show that the internalized TCRs are localized to endosomes expressing IRAP and Syntaxin 6 to maintain intracellular signalling capacity, whose importance is shown by the absence of efficient polyclonal anti-tumour response in mice with T-specific conditional deletion of IRAP.

Abstract: International audience; The large tolerance of hybrid perovksites to the trapping of electrons by defects is a key asset in photovoltaic applications. Here, the ionic surface terminations of CH3NH3PbI3 are employed as a testbed to study the effect of electrostatic fields on the dynamics of excited carriers. We characterize the transition across the tetragonal to orthorhombic phase. The observed type II band offset and drift of the excited electrons highlight the important role that organic cations have on the screening of local electrostatic fields. When the orientation of organic cations is frozen in the orthorhombic phase, the positively charged termination induces a massive accumulation of excited electrons at the surface of the sample. Conversely, no electron accumulation is observed in the tetragonal phase. We conclude that the local fields cannot penetrate in the sample when the polarizability of freely moving cations boosts the dielectric constant up to ϵ = 120.

Abstract: Objectives To assess the efficacy and tolerance of the conventional first-line treatment by MTX and CS in patients with JDM regardless of severity. Methods We conducted a monocentric retrospective study of patients with newly diagnosed JDM treated with MTX and CS from 2012 to 2020. The proportion of clinically inactive disease (CID) within 6 months of MTX initiation was evaluated using both Paediatric Rheumatology International Trials Organisation (PRINTO) criteria (evaluating muscle inactive disease) and DAS (evaluating skin inactive disease). We compared responders and non-responders using univariate analyses. Results Forty-five patients with JDM, out of which 30 (67%) severe JDM, were included. After 6 months of treatment with MTX and CS, complete CID, muscle CID and skin CID were achieved in 14/45 (31%), 19/45 (42%) and 15/45 (33%) patients, respectively. The absence of myositis-specific (MSA) or myositis-associated autoantibodies (MAA) at diagnosis was associated with a better overall, cutaneous and muscular therapeutic response, compared with antibody-positive forms (P Conclusions Conventional first-line treatment with MTX was not efficient in a large subset of JDM patients, especially in patients with MSA-positive forms, and in patients with severe JDM. Larger, multicentre cohorts are required to confirm these data and to identify new predictive biomarkers of MTX response, in order to treat patients with JDM as early as possible with appropriate targeted drugs.

Structure property relationships have been established between the thermal and optical properties of the resulting glass thanks to Raman and Infrared vibrational spectroscopies. The germano-gallate glass networks are mainly made of three-dimensional interconnected corner shared GeO4 and GaO4 tetrahedra forming Ge – O – Ge and Ga – O− – Ge bridges in which gallium tetrahedral GaO4 units are charged-compensated by barium and lanthanum cations. In GeO2-rich compositions, the excess of positive charges produces mainly non-bridging oxygens on germanate moieties, leading to a depolymerization of the glass network. In the case of gallium rich compositions, the infrared cut-off is red-shifted as compared to the germanate glasses, in accordance with the disappearance of germanium associated entities such as germanium and gallium bridges or germanium sites with non-bridging oxygen both vibrating at higher wavenumber. Abstract: For many intracellular bacterial pathogens manipulating host cell survival is essential for maintaining their replicative niche, and is a common strategy used to promote infection. The bacterial pathogen Listeria monocytogenes is well known to hijack host machinery for its own benefit, such as targeting the host histone H3 for modification by SIRT2. However, by what means this modification benefits infection, as well as the molecular players involved, were unknown. Here we show that SIRT2 activity supports Listeria intracellular survival by maintaining genome integrity and host cell viability. This protective effect is dependent on H3K18 deacetylation, which safeguards the host genome by counteracting infection-induced DNA damage. Mechanistically, infection causes SIRT2 to interact with the nucleic acid binding protein TDP-43 and localise to genomic R-loops, where H3K18 deacetylation occurs. This work highlights novel functions of TDP-43 and R-loops during bacterial infection and identifies the mechanism through which L. monocytogenes co-opts SIRT2 to allow efficient infection., Author summary To cause systemic disease Listeria monocytogenes assumes an intracellular lifestyle which supports its growth and dissemination during infection. In order to maintain the intracellular niche L. monocytogenes manipulates various host cell processes thereby promoting its own survival and infection. One such example is the hijacking of a host deacetylase called SIRT2 which upon infection localises to chromatin, specifically modifies lysine 18 of histone H3 and promotes intracellular bacterial growth. Here we identify how SIRT2 promotes infection. We show that SIRT2-mediated H3K18 deacetylation counteracts infection-induced DNA damage and identify the molecular complex at play. Such SIRT2 activity has a crucial role in promoting host cell viability during infection, allowing for better survival upon heavy intracellular bacterial burden, and resulting in enhanced infection by L. monocytogenes.

Abstract: International audience; The rabbit has been widely used in immunology and infectiology. Rabbit immunoglobulins have been extensively studied, leading to the discovery of their idiotypes, allotypic diversity, and of the diversification of the primary repertoire by hyperconversion. Much less is known about rabbit T cell receptors (TR), especially TRA. This isotype is particularly important for innate-like T cells, which typically express invariant TRA (iTRA). The presence of such cells in the rabbit remains an enigma. Rabbit NKT cells seem to be very rare, and lagomorphs lack MAIT cells. TRAV1, the variable gene expressed in the iTRA of these cells across most mammals, and MR1, the MH1-like receptor that present riboflavin derivatives to MAIT cells, are missing in rabbit. An alternative iTRA has been identified, that may be expressed by new innate-like T cells. To facilitate TRA repertoire analyses in rabbit, we report here a full description of TRA and TRD loci and a subgroup definition based on IMGT® classification. Rabbit TRA rearrangements follow the same temporal pattern that is observed in mouse and human. Rare transcripts expressing TRDV/TRDD/TRDJ rearrangements spliced to TRAC were detected. TRA and TRD genes have been made available in IMGT and IMGT/HighV-QUEST, allowing easy analysis of TRA/TRD RepSeq.

Abstract: Epithelial ovarian cancer (EOC) is the most common gynecologic disorder. Even with the recent progresses made towards the use of new therapeutics, it still represents the most lethal gynecologic malignancy in women from developed countries.The discovery of the anterior gradient proteins AGR2 and AGR3, which are highly related members belonging to the protein disulfide isomerase (PDI) family, attracted researchers’ attention due to their putative involvement in adenocarcinoma development. This review compiles the current knowledge on the role of the AGR family and the expression of its members in EOC and discusses the potential clinical relevance of AGR2 and AGR3 for EOC diagnosis, prognosis, and therapeutics.A better understanding of the role of the AGR family may thus provide new handling avenues for EOC patients.

Abstract: International audience; The structure and crystallization of silver-doped germano-gallate glasses for substitution of potassium for barium ions have been investigated. By means of Raman spectroscopy, a structural study has been carried out revealing a glass network depolymerization with the formation of non-bridging oxygens likely localized on germanium tetrahedra when the barium concentration increases. These structural modifications do not appear to significatively change the silver ion environments or ionic states, as reported on luminescence and optical transmission spectroscopies. A combined differential scanning calorimetry, X-ray diffraction characterization, scanning electron microscopy and micro-luminescence spectroscopy demonstrate that the barium ions introduction tends to stabilize the silver ions and to influence drastically the devitrification and the crystal growth kinetics. Correlations have been established between ionic conductivity, silver distribution and crystal growth mechanisms.

Abstract: We introduce the hemicubic codes, a family of quantum codes obtained by associating qubits with the $p$-faces of the $n$-cube (for $n>p$) and stabilizer constraints with faces of dimension $(p\pm1)$. The quantum code obtained by identifying antipodal faces of the resulting complex encodes one logical qubit into $N = 2^{n-p-1} \tbinom{n}{p}$ physical qubits and displays local testability with a soundness of $\Omega(1/\log(N))$ beating the current state-of-the-art of $1/\log^{2}(N)$ due to Hastings. We exploit this local testability to devise an efficient decoding algorithm that corrects arbitrary errors of size less than the minimum distance, up to polylog factors. We then extend this code family by considering the quotient of the $n$-cube by arbitrary linear classical codes of length $n$. We establish the parameters of these generalized hemicubic codes. Abstract: Background Results regarding the association between hormonal exposure and risk of Parkinson's disease (PD) are heterogeneous. Objectives To investigate the association of reproductive life characteristics with PD among postmenopausal women. Methods The PARTAGE case-control included 130 female cases and 255 age-matched female controls. Information on gynecological history was obtained from a standardized questionnaire and PD was validated by neurological examination. Odds ratios (ORs) and 95% confidence intervals (CIs) were computed using conditional logistic regression. Results After adjustment for education level, smoking status, professional exposure to pesticides, and coffee and alcohol drinking, bilateral oophorectomy (OR = 3.55, 95%CI = 1.75-7.20), but neither menopause before age 50 years (OR = 1.24, 95%CI = 0.74-2.09) nor hormone therapy (HT; OR = 1.07, 95%CI = 0.62-1.86), was associated with PD. Conclusion Our findings suggest that bilateral oophorectomy is associated with increased risk of PD. © 2021 International Parkinson and Movement Disorder Society.

Abstract: Decision making in immuno‐oncology is pivotal to adapt therapy to the tumor microenvironment (TME) of the patient among the numerous options of monoclonal antibodies or small molecules. Predicting the best combinatorial regimen remains an unmet medical need. Here, we report a multiplex functional and dynamic immuno‐assay based on the capacity of the TME to respond to ex vivo stimulation with twelve immunomodulators including immune checkpoint inhibitors (ICI) in 43 human primary tumors. This "in sitro" (in situ/in vitro) assay has the potential to predict unresponsiveness to anti‐PD‐1 mAbs, and to detect the most appropriate and personalized combinatorial regimen. Prospective clinical trials are awaited to validate this in sitro assay., To predict cancer resistance to PD‐1 blockade and design suitable combinations of immunomodulators, a 60‐h functional in sitro assay was set up in 43 tumors that allowed calculation of the “Immune Reactivity Score (IRS)” based on 17 TCR‐dependent‐ cytokines/chemokines.

Abstract: International audience; The severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) is responsible for the current COVID-19 pandemic. An unbalanced immune response, characterized by a weak production of type I interferons (IFN-Is) and an exacerbated release of proinflammatory cytokines, contributes to the severe forms of the disease. SARS-CoV-2 is genetically related to SARS-CoV and Middle East respiratory syndrome-related coronavirus (MERS-CoV), which caused outbreaks in 2003 and 2013, respectively. Although IFN treatment gave some encouraging results against SARS-CoV and MERS-CoV in animal models, its potential as a therapeutic against COVID-19 awaits validation. Here, we describe our current knowledge of the complex interplay between SARS-CoV-2 infection and the IFN system, highlighting some of the gaps that need to be filled for a better understanding of the underlying molecular mechanisms. In addition to the conserved IFN evasion strategies that are likely shared with SARS-CoV and MERS-CoV, novel counteraction mechanisms are being discovered in SARS-CoV-2-infected cells. Since the last coronavirus epidemic, we have made considerable progress in understanding the IFN-I response, including its spatiotemporal regulation and the prominent role of plasmacytoid dendritic cells (pDCs), which are the main IFN-I-producing cells. While awaiting the results of the many clinical trials that are evaluating the efficacy of IFN-I alone or in combination with antiviral molecules, we discuss the potential benefits of a well-timed IFN-I treatment and propose strategies to boost pDC-mediated IFN responses during the early stages of viral infection.

Abstract: International audience; Since their discovery, single-domain antigen-binding fragments of camelid-derived heavy-chainonly antibodies, also known as nanobodies (Nbs), have proven to be of outstanding interest as therapeutics against human diseases and pathogens including viruses, but their use against phytopathogens remains limited. Many plant viruses including Grapevine fanleaf virus (GFLV), a nematode-transmitted icosahedral virus and causal agent of fanleaf degenerative disease, have worldwide distribution and huge burden on crop yields representing billions of US dollars of losses annually, yet solutions to combat these viruses are often limited or inefficient. Here, we identified a Nb specific to GFLV that confers strong resistance to GFLV upon stable expression in the model plant Nicotiana benthamiana and also in grapevine rootstock, the natural host of the virus. We showed that resistance was effective against a broad range of GFLV isolates independently of the inoculation method including upon nematode transmission but not againstits close relative, Arabis mosaic virus. We also demonstrated that virus neutralization occurs at an early step of the virus life cycle, prior to cell-to-cell movement. Our findings will not only be instrumental to confer resistance to GFLV in grapevine, but more generally they pave the way for the generation of novel antiviral strategies in plants based on Nbs

Abstract: Countering TB prodrug resistance The arsenal of antibiotics for treating tuberculosis (TB) contains many prodrugs, such as ethionamide, which need activation by normal metabolism to release their toxic effects. Ethionamide is potentiated by small molecules. Blondiaux et al. screened for more potent analogs and identified a lead compound called SMARt-420. This small molecule inactivates a TetR-like repressor, EthR2, and boosts ethionamide activation. SMARt-420 successfully promoted clearance of multidrug-resistant strains of Mycobacterium tuberculosis from the lungs of mice. Science , this issue p. 1206

Abstract: International audience; L’étude proposée porte sur la conception et la mise en place d’une séquence de formation au sein d’une filière professionnelle secondaire agricole. L’analyse est menée à l’aide du concept de transposition didactique interne. La méthodologie mobilisée est de type ethnographique, avec une enquête déployée sur le temps long (3 ans) et une combinaison de différents modes de production de données (observations, entretiens, enregistrements audio-vidéo, analyse de documents). Les analyses mettent en particulier en évidence la diversité des références à l’origine de ce processus transpositif et des opérations de transposition complexes qui dépendent pour une part importante de l’organisation socio- technique d’un établissement, elle-même fruit d’une réflexion et de choix en partie didactiques, effectués parfois plusieurs années plus tôt.

Abstract: Background: Although it is rarely fatal in developed countries, acute gastroenteritis (AGE) still induces significant morbidity and economic costs. The objective of this study was to identify factors associated with AGE in winter in the general population.Methods: A prospective study was performed during winter seasons from 2014-2015 to 2016-2017. Participants filled an inclusion survey and reported weekly data on acute symptoms. Factors associated with having at least one AGE episode per winter season were analyzed using the generalized estimating equations (GEE) approach. Results: They were 13,974 participants included in the study over the three seasons. On average, 8.1% of participants declared at least one AGE episode during a winter season. People over 60 declared fewer AGE episodes (adjusted OR (aOR)=0.76, 95% CI [0.64; 0.89]) compared to individuals between 15 and 60 years old, as well as children between 10 and 15 (aOR=0.60 [0.37; 0.98]). Overweight (aOR=1.25 [1.07; 1.45]) and obese (aOR=1.47 [1.19; 1.81]) individuals, those having frequent cold (aOR=1.63 [1.37; 1.94]) and those with at least one chronic condition (aOR=1.35 [1.16; 1.58]) had more AGE episodes. Living alone was associated with a higher AGE episode rate (aOR=1.31 [1.09; 1.59]), as well as having pets at home (aOR=1.23 [1.08; 1.41]). Conclusions: Having a better knowledge of AGE determinants will be useful to adapt public health prevention messages.

Abstract: International audience; The field of small air quality sensors is of growing interest within the scientific community, especially because this new technology is liable to improve air pollutant monitoring as well as be used for personal exposure quantification. Amongst the myriad existing devices, the performances are highly variable; this is why the sensors must be rigorously assessed before deployment, according to the intended use. This study is included in the Polluscope project; its purpose is to quantify personal exposure to air pollutants by using portable sensors. This paper designs and applies a methodology for the evaluation of portable air quality sensors to eight devices measuring PM, BC, NO2 and O3. The dedicated testing protocol includes static ambient air measurements compared with reference instruments, controlled chamber and mobility tests, as well as reproducibility evaluation. Three sensors (AE51, Cairclip and Canarin) were retained to be used for the field campaigns. The reliability of their performances were robustly quantified by using several metrics. These three devices (for a total of 36 units) were deployed to be worn by volunteers for a week. The results show the ability of sensors to discriminate between different environments (i.e., cooking, commuting or in an office). This work demonstrates, first, the ability of the three selected sensors to deliver data reliable enough to enable personal exposure estimations, and second, the robustness of this testing methodology.

Abstract: Naphthalene is certainly not a common arene. In contrast to benzene, the bicyclic feature of naphthalene offers multiple differentiable positions and thus a broad diversity of substitution patterns. Naphthalene is a central building block for the construction of elaborated polycyclic architectures with applications in broad domains such as life and materials sciences. As a result, C–H functionalization strategies specially designed for naphthalene substrates have become essential to install valuable substituents on one or both rings towards polysubstituted naphthalenes. Abstract: Banerdt, William B. et al., NASA’s InSight (Interior exploration using Seismic Investigations, Geodesy and Heat Transport) mission landed in Elysium Planitia on Mars on 26 November 2018. It aims to determine the interior structure, composition and thermal state of Mars, as well as constrain present-day seismicity and impact cratering rates. Such information is key to understanding the differentiation and subsequent thermal evolution of Mars, and thus the forces that shape the planet’s surface geology and volatile processes. Here we report an overview of the first ten months of geophysical observations by InSight. As of 30 September 2019, 174 seismic events have been recorded by the lander’s seismometer, including over 20 events of moment magnitude M = 3–4. The detections thus far are consistent with tectonic origins, with no impact-induced seismicity yet observed, and indicate a seismically active planet. An assessment of these detections suggests that the frequency of global seismic events below approximately M = 3 is similar to that of terrestrial intraplate seismic activity, but there are fewer larger quakes; no quakes exceeding M = 4 have been observed. The lander’s other instruments—two cameras, atmospheric pressure, temperature and wind sensors, a magnetometer and a radiometer—have yielded much more than the intended supporting data for seismometer noise characterization: magnetic field measurements indicate a local magnetic field that is ten-times stronger than orbital estimates and meteorological measurements reveal a more dynamic atmosphere than expected, hosting baroclinic and gravity waves and convective vortices. With the mission due to last for an entire Martian year or longer, these results will be built on by further measurements by the InSight lander., With funding from the Spanish government through the "María de Maeztu Unit of Excellence" accreditation (MDM-2017-0737)

Abstract: Publisher's version (útgefin grein), Background: Emerging evidence suggests that androgens and estrogens have a role in respiratory health, but it is largely unknown whether levels of these hormones can affect lung function in adults from the general population. This study investigated whether serum dehydroepiandrosterone sulfate (DHEA-S), a key precursor of both androgens and estrogens in peripheral tissues, was related to lung function in adult women participating in the European Community Respiratory Health Survey (ECRHS). Methods: Lung function and serum DHEA-S concentrations were measured in n = 2,045 and n = 1,725 women in 1999–2002 and in 2010–2013, respectively. Cross-sectional associations of DHEA-S levels (expressed as age-adjusted z-score) with spirometric outcomes were investigated, adjusting for smoking habits, body mass index, menopausal status, and use of corticosteroids. Longitudinal associations of DHEA-S levels in 1999–2002 with incidence of restrictive pattern and airflow limitation in 2010–2013 were also assessed. Abstract: Innate lymphoid cells (ILC) are important players of early immune defenses in situations like lymphoid organogenesis or in case of immune response to inflammation, infection and cancer. Th1 and Th2 antagonism is crucial for the regulation of immune responses, however mechanisms are still unclear for ILC functions. ILC2 and NK cells were reported to be both involved in allergic airway diseases and were shown to be able to interplay in the regulation of the immune response. CXCR6 is a common chemokine receptor expressed by all ILC, and its deficiency affects ILC2 and ILC1/NK cell numbers and functions in lungs in both steady-state and inflammatory conditions. We determined that the absence of a specific ILC2 KLRG1+ST2&ndash, subset in CXCR6-deficient mice is probably dependent on CXCR6 for its recruitment to the lung under inflammation. We show that despite their decreased numbers, lung CXCR6-deficient ILC2 are even more activated cells producing large amount of type 2 cytokines that could drive eosinophilia. This is strongly associated to the decrease of the lung Th1 response in CXCR6-deficient mice.

Abstract: Notre travail s’inscrit dans les ancrages théoriques de l’ethnométhodologie et de l’analyse conversationnelle. Notre intérêt est avant tout d’analyser les pratiques interactionnelles entre joueurs en situation de jeux vidéo (Colón de Carvajal, 2013, 2016 ; Mondada, 2011, 2012 ; Piirainen–Marsh & Tainio, 2009a et b) ; et d’autre part, de comparer différents jeux sur le plan de leur organisation interactionnelle (les cadres de participation) ainsi que leur structure séquentielle et multimodale (l’organisation de l’espace et la gestion des objets). Á partir des pratiques de joueurs, nous questionnons les problèmes et contraintes rencontrés, d’une part lors des analyses de ces données en situation de jeux vidéo, et d’autre part lors de présentations académiques de nos recherches. Une étude en cours, sur les séquences d’encouragement et d’instruction dans les interactions entre joueurs, nous permet de montrer qu’il est indispensable de contextualiser le type de jeu joué, ses règles, ses options spécifiques parfois, les relations entre les joueurs, les rôles de chacun à l’instant de l’extrait, et aussi les actions en cours dans l’univers du jeu lui-même (un objet qui tombe, un avatar touché, etc.) qui donneraient du sens à l’analyse. Ces contextualisations et prises en compte des ressources multimodales permettent d’expliquer et de décrire plus clairement et objectivement le phénomène linguistique, l’action ou encore la problématique analysée. Our research is embedded in the theoretical issues of ethnomethodology and conversational analysis. Our interest is above all to analyze the interactional practices between players in a video game situation (Colón de Carvajal, 2013, 2016, Mondada, 2011, 2012, Piirainen-Marsh & Tainio, 2009a and b); and on the other hand, to compare different games in terms of their interactional organization (participation frameworks) as well as their sequential and multimodal structure (the organization of space and the management of objects). From the practices of players, we question the problems and constraints encountered, on the one hand during the analysis of these data in a video game situation, and on the other hand during academic presentations of our research. A work in progress on encouragement’s and instruction’s sequences in interactions between players, allows to show that it is essential to contextualize the type of game played, its rules, its specific options, the relationships between players, the roles of each player at the moment of the extract, and also the current actions in the universe of the game itself (a falling object, a touched avatar, etc.) that would give meaning to the analysis. These contextualizations and these considerations of multimodal resources make it possible to explain and describe more clearly and objectively the linguistic phenomenon, the action or the problem analyzed.

TET 1, 2 and 3 proteins are oxygenases that can hydroxylate 5-methylcytosine (5-mC) into 5-hydroxymethylcytosine (5-hmC) and further oxidize 5-hmC into 5-formylcytosine (5-fC) and 5-carboxylcytosine (5-caC). The base excision repair (BER) pathway removes the resulting 5-fC and 5-caC bases paired with a guanine and replaces them with regular cytosine. The question arises whether active modification of 5-mC residues and their subsequent elimination could affect the genomic DNA stability. Here, we generated two inducible cell lines (Ba/F3-EPOR, and UT7) over-expressing wild-type or catalytically inactive human TET2 proteins. Wild-type TET2 induction resulted in an increased level of 5-hmC and a cell cycle defect in S phase associated with higher level of phospho-rylated P53, chromosomal and centrosomal abnormalities. Furthermore, in a thymine-DNA glycosylase (Tdg) deficient context, the TET2-mediated increase of 5-hmC induces mutagenesis characterized by GC > AT transitions in CpG context suggesting a mutagenic potential of 5-hmC metabolites. Altogether, these data suggest that TET2 activity and the levels of 5-hmC and its derivatives should be tightly controlled to avoid genetic and chromosomal instabilities. Moreover, TET2-mediated active demethylation might be a very dangerous process if used to entirely demethylate the genome and might rather be used only at specific loci.

Abstract: International audience; We report the observation of a pressure-induced non-monotonic behavior of the long-wavelength asymptote of total charge density fluctuations right in the region of molecular-to-atomic fluid transformation from ab-initio molecular dynamics (AIMD) simulations of fluid hydrogen. Screening in the total charge density composed of point protons and distribution of electronic density is studied within density functional theory (DFT) via total charge-charge static structure factor SQQ DFT(k). We show from theory and AIMD that the long-wavelength behavior of SQQ DFT(k → 0) ~ kη with η = 4 in neutral atomic and in fully metallic liquids, while η = 2 is in an ionic mixture with long-range Coulomb interaction. At high temperature (2500 K) we find a change in η with density around the region of molecular dissociation. At lower temperature (1000 K, below the critical point), we observe an η value different from 4 only in the pressure vs. density plateau region. This observation is consistent with a smooth molecular to atomic transformation, and implies existence of a semiconducting state with some unscreened protons responsible for long-range interaction.

Abstract: International audience; Mid-infrared supercontinuum (mid-IR SC) sources in the 2-20 mu m molecular fingerprint region are in high demand for a wide range of applications including optical coherence tomography, remote sensing, molecular spectroscopy, and hyperspectral imaging. Herein, mid-IR SC generation is investigated in a cascaded silica-ZBLAN-chalcogenide fiber system directly pumped with a commercially available pulsed fiber laser operating in the telecommunications window at 1.55 mu m. This fiber-based system is shown to generate a flat broadband mid-IR SC covering the entire range from 2 to 10 mu m with several tens of mW of output power. This technique paves the way for low cost, practical, and robust broadband SC sources in the mid-IR without the requirement of mid-infrared pump sources or Thulium-doped fiber amplifiers. A fully realistic numerical model used to simulate the nonlinear pulse propagation through the cascaded fiber system is also described and the numerical results are used to discuss the physical processes underlying the spectral broadening in the cascaded system. Finally, recommendations are provided for optimizing the current cascaded system based on the simulation results.

Abstract: International audience; Background: Vaccination against seasonal influenza (SIV) is recommended for patients with diabetes, but their vaccination coverage is unsatisfactory in France and elsewhere. This qualitative survey of people with diabetes sought to explore 1) the extent to which SIV-related behaviour is more or less automatic; 2) reasons they choose/reject SIV; 3) their trust/distrust in authorities, science, and medicine. Methods: We conducted semi-structured in-depth interviews of 19 adults with diabetes in 2014. We recruited them through physicians or patient associations and implemented an analysis of thematic content. Results: Eight patients were vaccinated against flu in the preceding flu season and 11 were not. SIV uptake and refusal were stable over time and justified by multiple arguments. Coupons for free vaccines and regular doctor visits contributed to the habit of vaccination. Vaccination decisions were frequently anchored in past experiences of influenza and its vaccine. Patients often justified non-vaccination with attitudes of trivialisation/relativisation of influenza-associated risks and the perception that these can be controlled by means other than vaccination (e.g., through the avoidance of exposure). Some misbeliefs (e.g., SIV causes influenza) and doubts about SIV effectiveness and safety also existed. Several patients reported increased mistrust of SIV since the A/H1N1 pandemic in 2009. Patients trusted their doctors strongly regardless of their SIV behaviour, but unvaccinated patients had little trust in the government and pharmaceutical companies. Some discordances were found between perceptions and behaviour (e.g., remaining vaccinated despite doubts about SIV effectiveness or remaining unvaccinated despite feelings of vulnerability towards influenza complication), suggesting the existence of some vaccine hesitancy among patients. Conclusion: This study among patients with diabetes suggest that SIV uptake is stable, thanks to a favourable environment. Nonetheless, SIV refusal is also stable over time. Unvaccinated patients used multiple arguments to justify SIV refusal, including compensatory health beliefs. Physicians should take every opportunity to recommend SIV. The necessary individualised patient education regarding SIV requires better physician training in patients priorities. While almost all patients strongly trust their doctors, unvaccinated patients distrust distal stakeholders: it is absolutely essential to restore trust in them and to develop new more effective influenza vaccines.

Abstract: Background Ticks and tick-borne pathogens significantly impact both human and animal health and therefore are of major concern to the scientific community. Knowledge of tick-borne pathogens is crucial for prescription of mitigation measures. In Africa, much research on ticks has focused on domestic animals. Little is known about ticks and their pathogens in wild habitats and wild animals like the endangered chimpanzee, our closest relative. Methods In this study, we collected ticks in the forested habitat of a community of 100 chimpanzees living in Kibale National Park, Western Uganda, and assessed how their presence and abundance are influenced by environmental factors. We used non-invasive methods of flagging the vegetation and visual search of ticks both on human team members and in chimpanzee nests. We identified adult and nymph ticks through morphological features. Molecular techniques were used to detect and identify tick-borne piroplasmids and bacterial pathogens. Results A total of 470 ticks were collected, which led to the identification of seven tick species: Haemaphysalis parmata (68.77%), Amblyomma tholloni (20.70%), Ixodes rasus sensu lato (7.37%), Rhipicephalus dux (1.40%), Haemaphysalis punctaleachi (0.70%), Ixodes muniensis (0.70%) and Amblyomma paulopunctatum (0.35%). The presence of ticks, irrespective of species, was influenced by temperature and type of vegetation but not by relative humidity. Molecular detection revealed the presence of at least six genera of tick-borne pathogens (Babesia, Theileria, Borrelia, Cryptoplasma, Ehrlichia and Rickettsia). The Afrotopical tick Amblyomma tholloni found in one chimpanzee nest was infected by Rickettsia sp. Conclusions In conclusion, this study presented ticks and tick-borne pathogens in a Ugandan wildlife habitat whose potential effects on animal health remain to be elucidated. Graphical Abstract

Abstract: International audience; Potassium vanadium oxyfluoride phosphate of composition KVPO4F0.5O0.5 was modified by a carbon coating to enhance its electrochemical performance. Two distinct methods were used, first, chemical vapor deposition (CVD) using acetylene gas as a carbon precursor and second, an aqueous route using an abundant, cheap, and green precursor (chitosan) followed by a pyrolysis step. The formation of a 5 to 7 nm-thick carbon coating was confirmed by transmission electron microscopy and it was found to be more homogeneous in the case of CVD using acetylene gas. Indeed, an increase of the specific surface area of one order of magnitude, low content of C sp2, and residual oxygen surface functionalities were observed when the coating was obtained using chitosan. Pristine and carbon-coated materials were compared as positive electrode materials in potassium half-cells cycled at a C/5 (C = 26.5 mA g–1) rate within a potential window of 3 to 5 V vs K+/K. The formation by CVD of a uniform carbon coating with the limited presence of surface functions was shown to improve the initial coulombic efficiency up to 87% for KVPFO4F0.5O0.5-C2H2 and to mitigate electrolyte decomposition. Thus, performance at high C-rates such as 10 C was significantly improved, with ∼50% of the initial capacity maintained after 10 cycles, whereas a fast capacity loss is observed for the pristine material.

Abstract: International audience; The parasite Leishmania~ donovani causes a fatal disease termed visceral leishmaniasis. The process through which the parasite adapts to environmental change remains largely unknown. Here we show that aneuploidy is integral for parasite adaptation and that karyotypic fluctuations allow for selection of beneficial haplotypes, which impact transcriptomic output and correlate with phenotypic variations in proliferation and infectivity. To avoid loss of diversity following karyotype and haplotype selection, L. donovani utilizes two mechanisms: polyclonal selection of beneficial haplotypes to create coexisting subpopulations that preserve the original diversity, and generation of new diversity as aneuploidy-prone chromosomes tolerate higher mutation rates. Our results reveal high aneuploidy turnover and haplotype selection as a unique evolutionary adaptation mechanism that L. donovani uses to preserve genetic diversity under strong selection. This unexplored process may function in other human diseases, including fungal infection and cancer, and stimulate innovative treatment options.

Abstract: This review is an in-depth survey of the reported synthetic approaches for the preparation of racemic α-amino esters via the reduction of α-nitro or α-oxime ester intermediates. Accordingly, it describes the many pathways that have been designed to prepare such intermediates. This includes synthesis starting with α-nitroacetates, dialkyl malonates, acetoacetates, diethyl oxalates as well as [2+3] or [2+4] cycloadditions using, respectively, alkyl carbonocyanidate N-oxides or alkyl 2-nitrosoacrylates. Abstract: International audience; Peptide-YY (PYY) and Glucagon-Like Peptide-1 (GLP-1) play important roles in the regulation of food intake and insulin secretion, and are of translational interest in the field of obesity and diabetes. PYY production is highest in enteroendocrine cells located in the distal intestine, mirroring the sites where high concentrations of short chain fatty acids (SCFAs) are produced by gut microbiota. We show here that propionate and butyrate strongly increased expression of PYY but not GCG in human cell line and intestinal primary culture models. The effect was predominantly attributable to the histone deacetylase inhibitory activity of SCFA and minor, but significant contributions of FFA2 (GPR43). Consistent with the SCFA-dependent elevation of PYY gene expression, we also observed increased basal and stimulated PYY hormone secretion. Interestingly, the transcriptional stimulation of PYY was specific to human-derived cell models and not reproduced in murine primary cultures. This is likely due to substantial differences in PYY gene structure between mouse and human. In summary, this study revealed a strong regulation of PYY production by SCFA that was evident in humans but not mice, and suggests that high fibre diets elevate plasma concentrations of the anorexigenic hormone PYY, both by targeting gene expression and hormone secretion.

Abstract: International audience; The goal of this paper is to observe revision during handwritten text production of French students with and without dyslexia. Subjects with typical language development automate spelling during childhood and adolescence, progressively with experience, this enables them – according to capacity theory applied to written text production (McCutchen, 1996) to allocate more cognitive resources to higher-level processes (Bereiter & Scardamalia, 1987). A lack of automation in spelling could result in poor compositional performance (Fayol & Miret, 2005). Moreover, Morken & Helland (2013) have shown that young children with dyslexia, engaged in a sentence dictation task, revise their text as much as control children, however their revisions are of lower quality. If students with dyslexia have not totally automated spelling (Author, 2016), what is the impact on higher-level processes, such as revision? Our goal is to examine if students with dyslexia and control students proofread their texts in the same way. Results show that they display some similarity but students with dyslexia seem to have a deficit in the error detection mechanism (Horowitz & Breznitz, 2011) and revisions are less efficient. We discuss these results by considering previous studies we conducted on spelling, speech and neuropsychological assessments of our dyslexic participants.

Abstract: International audience; Last-mile collection and delivery services often rely on multi-echelon logistic systems with many types of practical spatial, temporal, and resource constraints. We consider three extensions of the basic 2-echelon vehicle routing problem that are of practical interest: First, second-echelon vehicles need to simultaneously deliver and collect goods at customers within their specified time window. Second, first-echelon vehicles are allowed to perform multiple trips during the planning horizon. Third, the intermediate facilities, called satellites, allow temporary storage of goods, but the quantity that can be stored at a time is limited. This paper integrates these complicating features in a single mathematical model. To solve the problem, we design a decomposition-based matheuristic. It employs a reduced and refined mixed-integer programming formulation and two echelon-specific large neighborhood searches (LNS) to produce improving routes for the respective echelon. The most important algorithmic component is the feasibility check of LNS that relies on a sequence of constant-time and low-complexity tests. The final test allows rescheduling of the operations taking place at a satellite. It adapts the double-justification algorithm known from the scheduling literature. Extensive computational experiments systematically evaluate the new components of the matheuristic and benchmark it against a recent exact method for a related problem. Moreover, the impact of the main problem features such as the number of satellites and their capacity as well as the integration of forward and reverse flows is analyzed.

Abstract: International audience; M-ion batteries (M = Li, Na, K ...) positive electrode materials most often operate through the reversible oxidation of transition-metal ions. In complex materials involving many transition metals or many redox centers, understanding the sequence in which they participate to the reaction is not trivial but is often necessary to explain the electrochemical properties. Mixed anion vanadium phosphates, such as KVPO4F0.5O0.5, are known to contain two different redox entities that are V3+O4F2 and V3+O5F “ionic” entities on the one hand and {V4+═O}O5 and {V4+═O}O4F “covalent” vanadyl-type units on the other hand. However, their participation to the redox mechanism occurring during the charge of this material has never been studied. Here, we use V K-edge X-ray absorption spectroscopy to unveil the redox mechanism of KVPO4F1–yOy (y = 0, 0.5, 1), performing data analysis via a chemometric approach. With XAS being very sensitive to the oxidation state and bond length, it was found that the ionic V3+–F units oxidize at a lower potential than the covalent {V4+═O} ones, which is surprising considering the high electronegativity of fluoride anions but is consistent with the redox potential observed for KVPO4F and KVOPO4. Further, ab initio calculations and ex situ X-ray diffraction analyses allowed an atomistic description of the redox mechanism with the sequential oxidation of the cis V site before the trans V site in KVPO4F upon charge. Finally, the complete atomically resolved redox mechanism of KVPO4F0.5O0.5 is proposed.
