A traça-da-uva, Lobesia botrana, é praga-chave da vinha na Região Demarcada do Douro (RDD). A sua importância económica, a par da necessidade de reduzir o uso de inseticidas de síntese na vinha, conferem relevância ao desenvolvimento de estratégias de proteção contra a praga, alternativas aos meios químicos. Incluem-se nestas estratégias, a seleção das castas, as medidas culturais e a confusão sexual. O seu adequado uso obriga a um aprofundado conhecimento do comportamento do inseto em relação a diversos parâmetros biológicos. Com o presente trabalho, pretendeu-se, através da análise de dados recolhidos entre 2000 e 2019, estudar a influência, na intensidade do ataque de L. botrana, na RDD, da sub-região, ano, geração e casta. Os resultados mostraram que, a intensidade do ataque da praga: i) foi, em geral, inferior no Douro Superior, comparativamente, quer ao Cima Corgo quer ao Baixo Corgo; ii) diferiu entre anos, na dependência das condições climáticas, mas tendencialmente diminuiu ao longo dos anos; iii) na primeira geração, em geral manteve-se em valores inferiores ao nível económico de ataque, enquanto nas restantes gerações se situou entre os limites estabelecidos para o mesmo (i.e. 1 – 10% cachos atacados); iv) as castas ‘Touriga Franca’, no caso das castas tintas, e ‘Malvasia Fina’, no caso das castas brancas foram, de entre as estudadas, as tendencialmente mais atacadas.

The starting point of this paper is three works of the contemporary Portuguese writer Mário Cláudio – the novels Peregrinação de Barnabé das Índias (1998), Os Naufrágios de Camões (2017) and the play A Ilha de Oriente (1989) –, focusing on how the author rewrites the voyages of Discovery of the 16th century and shapes an image of the East. My aim is to analyse the representation of the so-called Orient and the memory of maritime travels, not only from the point of view of Mário Cláudio’s poetics, but also in the light of a collective discourse that is at the same time aesthetic, historical and mythical.

- Abstract
Background/Aims Retinopathy of prematurity (ROP) is currently diagnosed through repeated eye examinations to find the low percentage of infants that fulfil treatment criteria to reduce vision loss. A prediction model for severe ROP requiring treatment that might sensitively and specifically identify infants that develop severe ROP, DIGIROP-Birth, was developed using birth characteristics. DIGIROP-Screen additionally incorporates first signs of ROP in different models over time. The aim was to validate DIGIROP-Birth, DIGIROP-Screen and their decision support tool on a contemporary Swedish cohort. Methods Data were retrieved from the Swedish national registry for ROP (2018-2019) and two Swedish regions (2020), including 1082 infants born at gestational age (GA) 24 to <31 weeks. The predictors were GA at birth, sex, standardised birth weight and age at the first sign of ROP. The outcome was ROP treatment. Sensitivity, specificity and area under the receiver operating characteristic curve (AUC) with 95% CI were described. Results For DIGIROP-Birth, the AUC was 0.93 (95% CI 0.90 to 0.95); for DIGIROP-Screen, it ranged between 0.93 and 0.97. The specificity was 49.9% (95% CI 46.7 to 53.0) and the sensitivity was 96.5% (95% CI 87.9 to 99.6) for the tool applied at birth. For DIGIROP-Screen, the cumulative specificity ranged between 50.0% and 78.7%. One infant with Beckwith-Wiedemann syndrome who fulfilled criteria for ROP treatment and had no missed/incomplete examinations was incorrectly flagged as not needing screening. Conclusions DIGIROP-Birth and DIGIROP-Screen showed high predictive ability in a contemporary Swedish cohort. At birth, 50% of the infants born at 24 to <31 weeks of gestation were predicted to have low risk of severe ROP and could potentially be released from ROP screening examinations. 26. Development of 3D Cancer Cell Models to Test Metabolic Interventions as an Anticancer Strategy

In recent years, it has become evident that the role of mitochondria in the metabolic rewiring is essential for cancer development and progression. The metabolic profile during tumorigenesis has been performed mainly in traditional 2D cell models, including cell lines of various lineages and phenotypes. Although useful in many ways, their relevance can be often debatable, as they lack the interactions between different cells of the tumour microenvironment and/or interaction with the extracellular matrix 1,2. Improved models are now being developed using 3D cell culture technology, contributing with increased physiological relevance 3,4. In this work, we improved a method for the generation of 3D models from healthy and tumour colon tissue, based on organoid technology, and performed their molecular and biochemical characterization and validation. Further, in-plate cryopreservation was applied to these models, and optimal results were obtained in terms of cell viability and functionality of the cryopreserved models. We also cryopreserved colon fibroblasts with the aim to introduce them in a co-culture cryopreserved model with organoids. This technology allows the conversion of cell models into “plug and play” formats. Therefore, cryopreservation in-plate facilitates the accessibility of specialized cell models to cell-based research and application, in cases where otherwise such specialized models would be out of reach. Finally, we briefly explored the field of bioprinting, by testing a new matrix to support the growth of colon tumour organoids, which revealed promising preliminary results. To facilitate the reader, we organized this thesis into chapters, divided by the main points of work which include development, characterization and validation of the model, commercial output, and associated applications. Each chapter has a brief introduction, followed by results and discussion and a final conclusion. The thesis has also a general discussion and conclusion section in the end, which covers the main results obtained during this work.

Conjunctival concretions are single or clustered lesions frequently found on the palpebral conjunctiva. They are commonly present in older individuals, despite being rarely symptomatic. This case report describes an 83-year-old man with multiple conjunctival concretions, which were surgically treated. The patient was symptomatic on presentation and did not respond to conservative treatment. For this reason, a surgical approach was considered. After wide excision of the conjunctival lesions, a piece of amniotic membrane was fitted using fibrin glue. During follow-up, a markedly improvement in patient's symptoms was observed, along with nearly complete absence of conjunctival concretions. This is the first case report addressing extensive conjunctival concretions with a surgical approach using amniotic membrane. The authors conceived the technique described after noticing the limited clinical options in the literature. This technique was easily performed and achieved satisfactory results. RESUMO As concreções conjuntivais representam lesões amareladas, simples ou múltiplas, frequentemente encontradas na conjuntiva palpebral. São mais prevalentes em idades avançadas e raramente sintomáticas. Este relato de caso descreve o quadro clínico de um paciente de 83 anos com múltiplas concreções conjuntivais, cirurgicamente tratadas. Por se tratar de um paciente sintomático com resposta insuficiente ao tratamento conservador, foi considerada a abordagem cirúrgica. Após remoção das lesões, foi aplicado um enxerto de membrana amniótica, adaptado com cola de fibrina. No acompanhamento pós-operatório, verificou-se melhoria significativa dos sintomas, com desaparecimento quase total das concreções conjuntivais. Este é o primeiro caso que descreve uma abordagem cirúrgica com utilização de membrana amniótica na resolução desse tipo de lesões. A técnica, de fácil execução e com resultados muito favoráveis, foi desenvolvida pelos autores após constatarem a escassez de alternativas na literatura.

Resumo Diversos fármacos têm o potencial de causar danos nos tecidos se ocorrer extravasamento. A gravidade do extravasamento e a dimensão da lesão estão dependentes de inúmeros fatores, tais como a dose do medicamento, a sua concentração, local de administração e tempo de exposição ao mesmo. A prevenção é a forma mais eficaz de gestão do extravasamento. Perante a ocorrência de extravasamento torna-se crucial adotar um conjunto de medidas/protocolo atempados e adequados com o intuito de prevenir sequelas a longo prazo. As lesões por extravasamento, não relacionadas com a administração de quimioterapia (QT), mais frequentemente reportadas são causadas por soluções hiperosmolares e agentes vasopressores. Dissertação de Mestrado Integrado em Engenharia Electrotécnica e de Computadores apresentada à Faculdade de Ciências e Tecnologia Códigos Lidos por Máquinas ( Machine-Readable Codes - MRC) têm sido usados para diversas finalidades ao longo dos anos, e abordagens clássicas como o código de barras ou o código QR podem ser vistas frequentemente no nosso dia-a-dia. Contudo, mais recentemente, um novo MRC foi criado com a capacidade de juntar o poder de comunicação dos métodos clássicos com uma melhoria significativa na estética e na capacidade de codificação. Este método é designado por UniQode.O UniQode possui duas grandes vantagens sobre os MRCs clássicos: estética e maior capacidade de codificação. Consequentemente, abre novas possibilidades para diversas finalidades, como identificação de produtos, rastreamento, marketing e comunicação vendedor-comprador. Esse novo método MRC é composto por vários elementos, sendo um deles o Código Gráfico (Graphic Code - GC). Este elemento pode ser criado de duas formas diferentes, com pixéis ou com ícones, ou por vezes com ambos.O GC baseado em pixéis foi amplamente estudado e os seus resultados serão apresentados neste documento. No entanto, o GC baseado em ícones ainda representa um novo conceito, com um método de criação não estruturado.Nesta dissertação, o trabalho proposto abrange não só a formalização do método de geração de GC baseado em ícones, mas também a criação de uma ferramenta para este processo de codificação, de modo de agilizar esta operação e gerar a respetiva documentação formal.Com a realização do trabalho proposto, a industrialização deste tipo de código torna-se possível e é facilitada, e o trabalho futuro nesta área é ampliado. Esse trabalho engloba, por exemplo, o desenvolvimento de uma ferramenta de decodificação correspondente, o aprimoramento das funções deste projeto, entre outros aspetos. Machine-Readable Codes (MRC) have been used for several purposes over the years and classic approaches like the bar code or the QR Code can be seen everywhere in our day-to-day. However, more recently, a new MRC was created with the ability to combine the communication power of classical methods with a meaningful improvement on aesthetics and data capacity. This method is named UniQode. The UniQode has two major advantages over classical MRCs: aesthetics and larger coding capacity. Consequently, it opens new possibilities for several purposes such as product identification, tracking, marketing and seller-buyer communication. This new MRC method is composed by several elements, one of them being the Graphic Code (GC). This element can be created in two different ways, either with pixels or with icons, or, sometimes, both.The pixel-based GC has been extensively researched and its accomplishments will be presented in this document. However, the icon-based GC still represents a new concept with a mostly unstructured creation method.In this dissertation, the proposed work encompasses not only a formalisation of the generation method for icon-based GC, but also the creation of a tool for this type of encoding process, in order to quicken this operation while also generating its formal documentation.With the achievement of the proposed work, the industrialisation of this type of code is made possible and facilitated, and future work on this area is widened. Such work could comprise the development of a corresponding decoding tool, the enhancement of this project's tools abilities, among other aspects.

32. Development of 3D Cancer Cell Models to Test Metabolic Interventions as an Anticancer Strategy

In recent years, it has become evident that the role of mitochondria in the metabolic rewiring is essential for cancer development and progression. The metabolic profile during tumorigenesis has been performed mainly in traditional 2D cell models, including cell lines of various lineages and phenotypes. Although useful in many ways, their relevance can be often debatable, as they lack the interactions between different cells of the tumour microenvironment and/or interaction with the extracellular matrix 1,2. Improved models are now being developed using 3D cell culture technology, contributing with increased physiological relevance 3,4. In this work, we improved a method for the generation of 3D models from healthy and tumour colon tissue, based on organoid technology, and performed their molecular and biochemical characterization and validation. Further, in-plate cryopreservation was applied to these models, and optimal results were obtained in terms of cell viability and functionality of the cryopreserved models. We also cryopreserved colon fibroblasts with the aim to introduce them in a co-culture cryopreserved model with organoids. This technology allows the conversion of cell models into “plug and play” formats. Therefore, cryopreservation in-plate facilitates the accessibility of specialized cell models to cell-based research and application, in cases where otherwise such specialized models would be out of reach. Finally, we briefly explored the field of bioprinting, by testing a new matrix to support the growth of colon tumour organoids, which revealed promising preliminary results. To facilitate the reader, we organized this thesis into chapters, divided by the main points of work which include development, characterization and validation of the model, commercial output, and associated applications. Each chapter has a brief introduction, followed by results and discussion and a final conclusion. The thesis has also a general discussion and conclusion section

Depending on their tissue of origin, genetic and epigenetic marks and microenvironmental influences, cancer cells cover a broad range of metabolic activities that fluctuate over time and space. At the core of most metabolic pathways, mitochondria are essential organelles that participate in energy and biomass production, act as metabolic sensors, control cancer cell death, and initiate signaling pathways related to cancer cell migration, invasion, metastasis and resistance to treatments. While some mitochondrial modifications provide aggressive advantages to cancer cells, others are detrimental. This comprehensive review summarizes the current knowledge about mitochondrial transfers that can occur between cancer and nonmalignant cells. Among different mechanisms comprising gap junctions and cell-cell fusion, tunneling nanotubes are increasingly recognized as a main intercellular platform for unidirectional and bidirectional mitochondrial exchanges. Understanding their structure and functionality is an important task expected to generate new anticancer approaches aimed at interfering with gains of functions (e.g., cancer cell proliferation, migration, invasion, metastasis and chemoresistance) or damaged mitochondria elimination associated with mitochondrial transfer.

Com as várias mudanças que o ensino vai sofrendo, é crucial que as mesmas sejam analisadas, refletidas e postas em prática. Os avanços e os novos conhecimentos que as neurociências nos concedem e a simbiose estabelecida com a neuroeducação, permite-nos promover práticas pedagógicas e mobilizar estratégias neurodidáticas que respeitam o modo como os alunos aprendem. Além disso, o Programa de Autonomia e Flexibilidade Curricular surge-nos, atualmente, como uma proposta para gerirmos o currículo e proporcionarmos atividades diversificadas, centradas no aluno e nas suas capacidades, cujo foco principal é contribuir para o sucesso da sua aprendizagem e para a diminuição do abandono escolar. O presente relatório tem como base a intervenção educativa decorrente da prática de ensino supervisionada em 1.º Ciclo do Ensino Básico, com uma turma composta por 18 alunos do 4ºano de escolaridade durante o ano letivo de 2018/2019, numa instituição de ensino privada da cidade do Porto. O nosso objetivo é compreender de que forma é que a relação entre as estratégias neurodidáticas e o Programa de Autonomia e Flexibilidade Curricular influenciam a aprendizagem e a motivação dos alunos. Para atingir este objetivo foram desenhadas e planificadas um conjunto de intervenções educativas. Os registos dos resultados obtidos – preenchimento de uma grelha de avaliação para cada uma das estratégias, comentários dos alunos e do professor titular – comprovam que as estratégias utilizadas contribuem positivamente para o rendimento académico dos alunos e potenciam o seu desenvolvimento de uma forma integral e integrada, estimulando a atenção, a motivação e a participação das crianças. With the various changes that teaching is undergoing, it is crucial that they are analyzed, reflected and put into practice. The advances and the new knowledge that the neurosciences grant us and the established symbiosis with neuroeducation allow us to promote pedagogical practices and to mobilize neurodidathic strategies that respect the way students learn. In addition, the Curriculum Autonomy and Flexibility Program, currently, appears as a proposal to manage the curriculum and to provide diversified activities, centered on the student and their abilities, whose main focus is to contribute to the success of their learning and to the reduction of school dropout. This report is based on the educational intervention resulting from supervised teaching practice in the 1st Cycle of Basic Education, with a group consisting of 18 students from the 4th year of schooling during the academic year 2018/2019, in a private educational institution of the city of Porto. Our goal is to understand how the relationship between neurodidactic strategies and the Program of Autonomy and Curriculum Flexibility influences students learning and motivation. To achieve this goal, a set of educational interventions were designed and planned. The recording of the results obtained - completing an evaluation grid for each one of the strategies, the comments of the students and the head professor - show that the strategies used contribute positively to the academic performance of the students and potentiate their development in an integral and integrated way, stimulating the attention, the motivation and the participation of the children. Prosjektleder Inger Lise Nerland Bråte Jordmassene hentet ut med maskinell rydding fra Samuelskilen i Færder nasjonalpark på øya Mellom-Bolæren, inneholdt totalt 5% plast per vektenhet (av størrelse over 5 mm) og 50% per volumenhet. All plasten fra jordmassene fra Samuelskilen var polyetylen (PE; 73%) og polypropylen (PP; 27%). Det ble funnet vekst-inhibering av saltvanns-algen S. pseudocostatum i 100% elutriat av jord og mikroplast fra Samuelskilen og en noe mindre inhibering for ferskvannsalgen R. subcapitata. Det er usikkert om det var en faktisk påvirkning av plast fra jordmassene, eller om det er en pH- eller salinitets-effekt. Det kan også ha blitt påvirket av lagring av jordmassene over tid under suboptimale forhold. Det er en krevende og stor vurdering å finne ut hvorvidt et plast-akkumuleringssted som Samuelskilen bør ryddes maskinelt eller om de plastinfiltrerte jordmassene bør bli liggende. Flere tester bør utføres av arter fra forskjellige trofiskenivå. Det kan også være andre tester som er relevante å utføre, f.eks. knyttet til biologisk mangfold. 40. Quality Evaluation of Requirements Models: The Case of Goal Models and Scenarios

Context: Requirements Engineering approaches provide expressive model techniques for requirements elicitation and analysis. Yet, these approaches struggle to manage the quality of their models, causing difficulties in understanding requirements, and increase development costs. The models’ quality should be a permanent concern. Objectives: We propose a mixed-method process for the quantitative evaluation of the quality of requirements models and their modelling activities. We applied the process to goal-oriented (i* 1.0 and iStar 2.0) and scenario-based (ARNE and ALCO use case templates) models, to evaluate their usability in terms of appropriateness recognisability and learnability. We defined (bio)metrics about the models and the way stakeholders interact with them, with the GQM approach. Methods: The (bio)metrics were evaluated through a family of 16 quasi-experiments with a total of 660 participants. They performed creation, modification, understanding, and review tasks on the models. We measured their accuracy, speed, and ease, using metrics of task success, time, and effort, collected with eye-tracking, electroencephalography and electro-dermal activity, and participants’ opinion, through NASA-TLX. We characterised the participants with GenderMag, a method for evaluating usability with a focus on gender-inclusiveness. Results: For i*, participants had better performance and lower effort when using iStar 2.0, and produced models with lower accidental complexity. For use cases, participants had better performance and lower effort when using ALCO. Participants using a textual representation of requirements had higher performance and lower effort. The results were better for ALCO, followed by ARNE, iStar 2.0, and i* 1.0. Participants with a comprehensive information processing and a conservative attitude towards risk (characteristics that are frequently seen in females) took longer to start the tasks but had a higher accuracy. The visual and mental effort was also higher for these participants. Conclusions: A mixed-method process, with (bio)metric measurements, can provide reliable quantitative information about the success and effort of a stakeholder while working on different requirements models’ tasks.

Na actualidade, impera a necessidade de aplicar novas metodologias de trabalho que valorizem a liberdade dos alunos, a reflexão e o pensamento crítico, de forma a aproximá-los do conhecimento, aumentando a sua autonomia e consequente motivação. A presente investigação insere-se nesta temática e tem como objectivo dar o seu contributo para uma maior dinâmica em contexto de sala de aula, visível através da aplicação de uma metodologia diferenciada, o Flipped Classroom, assente numa maior intervenção dos alunos e na construção dos seus conhecimentos, valorizando algumas das competências definidas no Perfil dos Alunos para o século XXI. O conceito base do Modelo Flipped Classroom ou aula invertida é reverter a ordem pela qual as aulas explicativas precedem as actividades dos alunos, isto é, os alunos obtêm a informação teórica em casa, de acordo com materiais previamente disponibilizados pelo docente, e a componente lectiva é utilizada para a realização de actividades que aprofundem os conhecimentos, sob a supervisão do professor. Dos alunos, durante a componente lectiva, espera-se que continuem o processo de aprendizagem que iniciaram em casa, permitindo ao professor realizar um acompanhamento mais efectivo das aprendizagens, dando resposta às dúvidas que existirem. O aprofundamento dos conteúdos poderá incluir um conjunto de metodologias diversificadas, como a aula-oficina, o trabalho colaborativo ou o trabalho projecto, assim como a exploração de fontes diversas ou a dramatização dos conteúdos. A metodologia Flipped Classroom encontra-se assente num modelo pedagógico e didáctico activo, centrado no aluno, na resolução de problemas e na promoção do trabalho colaborativo. O presente relatório pretende apresentar os resultados da implementação desta metodologia na Escola Secundária da Quinta do Marquês, em Oeiras, no âmbito do Mestrado de Ensino de História no 3º Ciclo do Ensino Básico e no Ensino Secundário da Faculdade de Ciências Sociais e Humanas da Universidade Nova de Lisboa, firmando o debate em torno da premente necessidade em aplicar metodologias diferenciadas em contexto de sala de aula. Currently there is a need to apply new work methodologies that value student’s freedom, reflection and critical thinking, in order to bring them closer to knowledge, increasing their autonomy and consequent motivation. The present research is part of this theme and aims to contribute to a greater dynamic in the classroom context, visible through the application of a differentiated pedagogical model, the Flipped Classroom, which is based on a greater intervention of the students in the construction of their knowledge, valuing some of the Educational competences for the 21st Century. The basic concept of the Flipped Classroom model is to reverse the order, as such the lecture classes precede the students' activities, students obtain the theoretical information at home, according to materials previously made available by the teacher, and the class time component is used to carry out activities that improve their knowledge, under the supervision of the teacher. During class time the students are expected to continue the learning process that they started at home, allowing the teacher to carry out a more effective follow-up, answering any questions that may arise. The exploration of the contents may include a set of diverse methodologies, such as Classroom Workshops, Collaborative Work or Project Based Learning, as well as the exploration of diverse sources or the dramatization of contents. The Flipped Classroom model is based on an active pedagogical and didactic model, centered on the student, on problem solving and promoting collaborative work. This report aims to present the results of the implementation of this methodology at the Secondary School of Quinta do Marquês, in the scope of the Masters course of History Education in the 3rd Cycle of Basic Education and Secondary Education by the Faculty of Human and Social Sciences, New University of Lisbon, centering the debate around the pressing need to apply differentiated methodologies in the classroom context.

O Estágio Pedagógico foi realizado na Escola Básica com Secundária de Arcozelo em Ponte de Lima, tendo como orientador cooperante o Professor Carlos Cunha e como companheiros de Estágio, o José Pita e a Ana Gomes. No âmbito de todo o trabalho realizado durante o estágio pedagógico, o presente relatório encontra-se organizado em dois capítulos. O capítulo I traduz uma reflexão crítica e fundamentada do estágio pedagógico com o propósito de elaborar uma descrição consciente acerca de todo o caminho percorrido. Nele faço uma breve caraterização sumativa do meio e da escola onde realizei o meu estágio, relato a experiência que vivi ao longo deste ano letivo, as expetativas iniciais, as dificuldades que encontrei e as estratégias que tomei para as combater. Ainda neste capítulo, descrevo todas as tarefas de Ensino Aprendizagem desenvolvidas (planeamento anual, unidades didáticas, planos de aula e práticas de ensino supervisionadas) e tarefas de Estágio de relação Escola - Meio (estudo de turma e atividades desenvolvidas pelo núcleo de estágio). Para finalizar apresento as conclusões referentes ao Estágio pedagógico/ Reflexão critica. O Capítulo II é de caráter científico onde é apresentado o estudo científico com o tema estados de ansiedade e níveis de autoconfiança em praticantes de desporto escolar, com o objetivo de analisar a ansiedade (cognitiva e somática) e a autoconfiança no período pré-competitivo em praticantes de desporto escolar, comparar as possíveis diferenças entre modalidades individuais e modalidades coletivas e eventualmente estabelecer uma ponte para o rendimento escolar. This document titled as Traineeship Final Report is included within the discipline of Traineeship I and II, in the curriculum of the second cycle in Physical Education Teaching in Middle and High School, of the University of Trás-os-Montes and Alto Douro. This document is a requirement for obtaining a master's degree according to Article 10, which relates to the Regulatory Standards of the Second Study Cycles in Teaching of UTAD, and was made under the guidance of the Master’s Supervisor Jorge Soares. The traineeship was held at the Middle and High School of Arcozelo (Ponte de Lima), having Professor Carlos Cunha as advisor, and José Pita and Ana Gomes as traineeship colleagues. This report is organized in two chapters. Chapter One is a critical and founded reflection about the traineeship, with the description of all my journey. In this chapter I make a global characterization of the surrounding environment and of the school where my traineeship was held, I describe the experiences I had during this school year, initial expectations, difficulties I have experienced and the strategies I used to overcome them. Yet in this chapter, I describe all Ensino Aprendizagem developed tasks (annual planning, teaching units, lesson plans and supervised teaching practices) and also the trainee tasks of the School – Surrounding Environment relationship (classroom study and activities developed by the trainee group). Finally, I present my conclusions regarding the Traineeship/Critical Reflection. Chapter Two has a scientific character and presents a scientific study under the subject states of anxiety and self-confidence levels in school sports practitioners, in order to analyze the anxiety (cognitive and somatic) and the self-confidence in the period pre-competitive, compare the possible differences between individual and team sports anxiety and self-confidence levels, and eventually relate them to school performance.

Trabalho final de mestrado para obtenção do grau de mestre em Engenharia Química e Biológica No presente trabalho determinaram-se os teores dos carotenoides, α-caroteno, β-caroteno, β-criptoxantina, licopeno, luteína e zeaxantina presentes em amostras, de produtos hortícolas e pratos compostos representativos da alimentação da população portuguesa, recolhidas no âmbito do projeto TDSEXPOSURE (Total Diet Stu dy Exposure – Estudo de dieta total para avaliação da exposição). Os resultados obtidos são um contributo para a avaliação da ingestão de carotenoides em Portugal. As amostras foram sujeitas a um procedimento analítico validado que incluiu etapas de extração, assim como de saponificação, quando o teor de gordura e/ou a presença de ésteres de carotenoide o justificou. A determinação dos analitos foi realizada através do método de cromatografia líquida de alta eficiência (HPLC) de fase reversa comdetetor DAD-UV/Vis e, a sua quantificação foi feita através de calibração externa e recorrendo também a um padrão interno. O desempenho do método analítico foi avaliado com dois tipos de materiais de referência, o material de referência certificado (NIST 2383) e o material de referência interno preparado no laboratório. Os resultados obtidos para z-score permitiram concluir que os valores medidos são exatos. O processo de homogeneização das amostras foi também avaliado estudando a homogeneidade da matriz alimentar tomate com pele e grainhas. A análise estatística dos resultados permitiu confirmar a homogeneidade das amostras. Os carotenoides que se encontraram na maioria das amostras foram o β-caroteno (0,018-14 mg/100 g) e a luteína (0,0014-8,2 mg/100 g). Em 25% das amostras não foi detetado qualquer carotenoide. O estudo sobre o efeito da sazonalidade no teor de carotenoides em produtos hortícolas recolhidos em duas estações do ano (outono e inverno) evidenciou alterações no teor de β -caroteno no feijão-verde, de luteína na couve-flor e de zeaxantina no pimento. A determinação da atividade pró-vitamina A evidenciou um maior valor de equivalentes de retinol (RE) para a cenoura (2710 µg/100 g). Abstract: In the present work, the carotenoids, α-carotene, β-carotene, β-cryptoxanthin, lycopene, lutein and zeaxanthin present in samples, in particular vegetables and composite dishes representative of the Portuguese population diet, were collected as part of theTDSEXPOSURE project (Total Diet Study Exposure). The results are a contribution to the evaluation of carotenoid intake in Portugal. The samples were subjected to a validated analytical procedure that included extraction steps, as well as saponification st eps, when the fat content and/or the presence of carotenoid esters justified it. The determination of the analytes was carried out using the reverse phase high performance liquid chromatography (HPLC) method with DAD-UV / Vis detector and quantification was done through external calibration and also using an internal standard. To evaluate the performance of the analytical method, two types of reference materials were analyzed, the certified reference material (NIST 2383) and the internal reference material prepared in the laboratory. The results obtained for z-score allowed to conclude that the measured values are exact. The homogenization process of the samples was evaluated by studying the homogeneity of the tomato food matrix with skin and seeds. Statistical analysis confirmed that the homogenization process was adequate. The carotenoids found in most of the analyzed samples were β-carotene (0.018-14 mg/100 g) and lutein (0.0014-8.2 mg/100 g). In 25% of the samples no carotenoid was detected. The effect of seasonality on the carotenoid content in vegetables collected in two seasons of the year (autumn and winter) showed changes in β-carotene content in green beans, lutein in cauliflower and zeaxanthin in the pepper. The activity evaluation as pro-vitamin A showed the highest value of retinol equivalent (RE) for carrot (2710 μg/100 g). N/A

50. Multifunctional mussel inspired coatings for orthopaedic applications

Dissertação de mestrado em Engenharia Biomédica, Currently, there is still a significant rate of implant failures in clinical practice. Current solutions would consist of the development of robust, biocompatible, biodegradable coatings with enhanced adhesive and bioactive properties. So, in this work the development of multifunctional coatings inspired by adhesive properties of mussels and the robust nacre structure were proposed. Based on the configuration of the 3,4-dihydroxy-L-phenylalanine (DOPA) amino-acid of the mussel’s adhesive proteins, catechol groups were conjugated to chitosan (CHT) and hyaluronic acid (HA). Layer-by-layer (LbL) assembly was used to mimic the nacre structure, where the organic phase consisted of both polymers and the inorganic phase of bioactive glass nanoparticles (BGNPs). In parallel, polymeric LbL coatings were constructed for the sake of comparison. The modified polymers were characterized by ultraviolet-visible (UV-Vis) spectroscopy. The construction of various LbL configurations was monitored by quartz crystal microbalance and the adhesive properties were evaluated by lap shear adhesive tests. The bioactivity and the in-vitro cell behaviour were analysed for the coatings with and without BGNPs. In-vitro tests were conducted using the cell line L929. Hydroxyapatite deposition was evaluated by scanning electron microscopy (SEM) coupled with energy dispersive X-ray spectroscopy (EDS) and X-ray powder diffraction (XRD). Since the structure and topography play an important role in the functional performance of the films, two LbL assembly methods, dip- and spin-coating, were compared using three different substrates: glass, stainless steel, and titanium. The coatings were characterized by SEM, Fourier transform infrared spectroscopy (FT-IR), atomic force microscopy (AFM) and water contact angle (WCA). Given the enhanced adhesion and bioactivity of the developed films, they could be used as coatings of a variety of implants. In addition, spin-coating was found to be a particularly suitable method for the build-up, since films with smoother and more uniform surfaces were produced., Atualmente, ainda há uma percentagem significativa de falhas dos implantes na prática clínica. Soluções atuais envolveriam o desenvolvimento de revestimentos robustos, biocompatíveis, biodegradáveis, com propriedades adesivas e bioativas melhoradas. Assim, neste trabalho foi proposto o desenvolvimento de revestimentos multifuncionais inspirados nas propriedades adesivas dos mexilhões e na estrutura robusta do nácar. Baseado na configuração do aminoácido 3,4-dihidroxi-L-fenilalanina (DOPA) das proteínas adesivas dos mexilhões, foram conjugados grupos catecóis ao quitosano (CHT) e ao ácido hialurónico (HA). A montagem camada-a-camada (LbL) foi utilizada para mimetizar a estrutura do nácar, onde a fase orgânica consistiu em ambos os polímeros e a fase inorgânica nas nanopartículas de vidro bioativas (BGNPs). Paralelamente, foram construídos revestimentos LbL poliméricos para fins de comparação. Os polímeros modificados foram caracterizados por espectroscopia ultravioleta-visível (UV-Vis). A construção das várias configurações LbL foi monitorizada através da microbalança de cristal de quartzo e as suas propriedades adesivas avaliadas através de testes adesivos sob tensão de corte. A bioatividade e o comportamento celular in-vitro foram analisados para os revestimentos com e sem BGNPs. Os testes in-vitro foram realizados usando a linha celular L929. A deposição de hidroxiapatita foi avaliada por microscopia eletrónica de varrimento (SEM) acoplada com espectroscopia de energia dispersiva de raios-X (EDS) e por difração de raios-X (XRD). Uma vez que a estrutura e topografia apresentam um papel importante no desempenho funcional dos filmes, dois métodos de montagem LbL, revestimento por imersão e por rotação, foram comparados usando três substratos diferentes: vidro, aço inoxidável e titânio. Os revestimentos foram caracterizados por SEM, espectroscopia de infravermelho por transformada de Fourier (FT-IR), microscopia de força atómica (AFM) e ângulo de contacto da água (WCA). Dado à adesão e bioatividade melhoradas dos filmes desenvolvidos, estes poderiam ser utilizados como revestimentos para uma variedade de implantes. Além disso, verificou-se que o revestimento por rotação foi um método particularmente adequado para a construção, uma vez que foram produzidos filmes com superfícies mais lisas e uniformes.
