Abstract Background Approximately 90% of colorectal cancer (CRC) deaths are caused by tumors ability to migrate into the adjacent tissues and metastase into distant organs. More than 40 genes have been causally linked to the development of CRC but no mutations have been associated with metastasis yet. To identify molecular basis of CRC metastasis we performed whole-exome and genome-scale transcriptome sequencing of 7 liver metastases along with their matched primary tumours and normal tissue. Multiple, spatially separated fragments of primary tumours were analyzed in each case. Uniformly malignant tissue specimen were selected with macrodissection, for three samples followed with laser microdissection. Results > 100 sequencing coverage allowed for detection of genetic alterations in subpopulation of tumour cells. Mutations in KRAS, APC, POLE, and PTPRT, previously associated with CRC development, were detected in most patients. Several new associations were identified, including PLXND1, CELSR3, BAHD1 and PNPLA6. Conclusions We confirm the essential role of inflammation in CRC progression but question the mechanism of matrix metalloproteinases activation described in other work. Comprehensive sequencing data made it possible to associate genome-scale mutation distribution with gene expression patterns. To our knowledge, this is the first work to report such link in CRC metastasis context.

Accumulation of allelic variants in genes that regulate cellular proliferation, differentiation, and apoptosis may result in expansion of the aberrant intestinal epithelium, generating adenomas. Herein, we compared the mutation profiles of conventional colorectal adenomas (CNADs) across stages of progression towards early carcinoma. DNA was isolated from 17 invasive adenocarcinomas (ACs) and 58 large CNADs, including 19 with low-grade dysplasia (LGD), 21 with LGD adjacent to areas of high-grade dysplasia and/or carcinoma (LGD-H), and 28 with high-grade dysplasia (HGD). Ion AmpliSeq Comprehensive Cancer Panel libraries were prepared and sequenced on the Ion Proton. We identified 956 unique allelic variants; of these, 499 were considered nonsynonymous variants. Eleven genes (APC, KRAS, SYNE1, NOTCH4, BLNK, FBXW7, GNAS, KMT2D, TAF1L, TCF7L2, and TP53) were mutated in at least 15% of all samples. Out of frequently mutated genes, TP53 and BCL2 had a consistent trend in mutation prevalence towards malignancy, while two other genes (HNF1A and FBXW7) exhibited the opposite trend. HGD adenomas had significantly higher mutation rates than LGD adenomas, while LGD-H adenomas exhibited mutation frequencies similar to those of LGD adenomas. A significant increase in copy number variant frequency was observed from LGD through HGD to malignant samples. The profiling of advanced CNADs demonstrated variations in mutation patterns among colorectal premalignancies. Only limited numbers of genes were repeatedly mutated while the majority were altered in single cases. Most genetic alterations in adenomas can be considered early contributors to colorectal carcinogenesis.

Diclofenac (DCF) degradation in aqueous solution under electron beam (EB) irradiation after nanobubbling treatment was studied and compared with treatments using nanobubbling or EB irradiation alone. It was found that the removal efficiency of DCF increased by increasing the adsorbed dose, and it depended on the initial concentration of DCF in solution, being higher for the lower concentrations. Furthermore, when using the nanobubbling treatment alone, about 16% of the DCF was removed from the aqueous solution due to the OH radicals generated during the process. On the other hand, using EB treatment at 0.5 kGy, the degradation of DCF increased from 36% to 51% when adding a nanobubbling pretreatment before the EB radiation. At higher doses (5 kGy), the degradation of DCF was 96% using EB radiation and 99% using nanobubbling before EB radiation, indicating that the nanobubbling effect was not synergistic. With an increase in the adsorbed doses, EB radiation seemed to play a more important role on the degradation of DCF, probably due to the reactive species generated. Moreover, the solutions treated with nanobubbling and EB radiation presented higher COD values and radiolytic by-products with aromatic rings with chlorine. This work can support the development of innovative strategies to treat municipal wastewaters using ionizing radiation technologies.

Introduction: The Flexor Carpi Ulnaris (FCU) is a part of the palmar the forearm muscle group and one of the most important muscles for upper limb functioning - is responsible for flexion and adduc­tion of the hand at the radio-carpal joint. There are clinically significant but rare anatomical variations of FCU. The variability of the FCU has not been described up to now, and no typology of the muscle based on its more variable terminal attachment has been created. Aim of the study: Determination of FCU muscle typology based on available fetal material. Material and methods: A total of 114 human fetuses (53 female, 61 male) between 117 and 197 days of fetal life were eligible for the study. Preparations were carried out using classical anatomical techniques based on a previously published procedure. Thanks to that significant anthropometric landmarks were vis­ible for the gathering of metric measurements. Metric measurements were taken and statistically analysed using R-Project software. Results: A new typology was created based on variable muscle insertions. Additionally, the presence of an atypically located, additional, separated muscle belly was described. A comparison of measurements of the left upper limb in relation to the right upper limb showed significant differences for forearm length to the anthropometric point of the stylion radiale, limb length, total FCU length and FCU length which means that the left limb is longer than the right limb. A comparison of FCU insertion types between left and right upper limb showed there’s no significant difference between counts of each type. Conclusion: The FCU is a muscle that is easy to palpate and may therefore act as a topographical marker for healthcare professionals. Knowledge of its variability is not only of theoretical importance but also has clinical sig­nificance. The current publication demonstrates presence of variability in FCU terminal attachment. Certainly, this topic requires further research and continued work on a detailed understanding of forearm anatomy in the fetal period.

Rectal cancer constitutes over one-third of all colorectal cancers (CRCs) and is one of the leading causes of cancer-related deaths in developed countries. In order to identify high-risk patients and better adjust therapies, new markers are needed. Systemic inflammatory response (SIR) markers such as LMR, NLR, and PLR have proven to be highly prognostic in many malignancies, including CRC; however, their roles in locally advanced rectal cancer (LARC) are conflicting and lack proper validation. Sixty well-selected patients with LARC treated at the Maria Sklodowska-Curie National Research Institute of Oncology in Warsaw, Poland, between August 2017 and December 2020 were prospectively enrolled in this study. The reproducibility of the pre-treatment levels of the SIR markers, their correlations with clinicopathological characteristics, and their prognostic value were evaluated. There was a significant positive correlation between LMR and cancer-related inflammatory infiltrate (r = 0.38, p = 0.044) and PD-L1 expression in tumor cells, lymphocytes, and macrophages (combined positive score (CPS)) (r = 0.45, p = 0.016). The PLR level was correlated with nodal involvement (p = 0.033). The SIR markers proved to be only moderately reproducible and had no significant prognostic value. In conclusion, the LMR was associated with local cancer-related inflammation and PD-L1 expression in tumor microenvironments. The validity of SIR indices as biomarkers in LARC requires further investigation.

This study presents the results of research on energy self-sufficiency, safety and production reliability in a salmonids fish farm. For the needs of the breeding facility under construction in the Recirculating Aquaculture System, energy requirement analyses were carried out and guidelines collected for the implementation of innovative solutions, which ensure the security and continuity of energy supplies and the plant’s energy self-sufficiency. This research included the development of solutions for the reliability of power supply and ensuring the continuity of operation of devices that are key to the fish farming process, i.e., lighting and pumps. In terms of ensuring the necessary amounts of energy, the possibility to diversify electricity sources was analysed, taking into account the possibility of self-production, including primarily from a photovoltaic power plant and from average power wind farms. The profitability of using energy storage was also examined. Recommendations and a list of energy balances for selected cases of potential power sources are presented.

The article harks back to the publication entitled “The Motif of the Angel(s) of Death in Islamic Foundational Sources” (VV 38/2 [2020]), which was devoted to the analysis of the eponymous theme in the foundational sources of Islam: the Quran and the sunna of the Prophet Muhammad. The purpose of this paper is to examine whether the motif of angel(s) may have been borrowed from two monotheistic traditions that came before. The verification of the thesis that the motif of the angel(s) of death underwent diffusion was carried out in several steps. First, the motif was identified in the textual traditions of Judaism and early Christianity (i.e. sets of texts that were known and, in all likelihood, widespread in the Middle East during the formative period of Islam). As a result of the analysis, most of the themes recognised in the foundational texts of Islam were found. The next step was to identify possible routes of their transmission and percolation into the Islamic tradition and to determine the “ideological demand” for the motif of the angel(s) of death in the burgeoning Islam. Although Jewish and Christian imagery and beliefs about angels are an important (if not the primary) source of influence on Muslim angelology, there was most likely a two-way interaction between the monotheistic traditions, albeit to a limited extent.

Colorectal cancer (CRC) is one of the leading causes of cancer-related mortality worldwide. Novel markers are required in order to select high-risk patients and better adjust the treatment. Both peripheral and local markers of cancer-related inflammation (CRI) such as lymphocyte-to-monocyte ratio (LMR), neutrophil-to-lymphocyte ratio (NLR) or platelet-to-lymphocyte ratio (PLR) and tumor-infiltrating lymphocytes (TILs) have been thoroughly investigated in recent years and deemed to be highly prognostic. We hypothesized that there is an association between local and peripheral CRI indices and that blood-based biomarkers may serve as a surrogate of TILs. We retrospectively analyzed 87 patients with locally advanced left-sided CRC treated with radical-intent surgery in the Maria Skłodowska-Curie National Research Institute of Oncology in Warsaw, Poland, between January 2014 and December 2015. Fifty patients were found eligible for the study. The patients were divided in terms of pre-treatment values of systemic inflammatory response (SIR) markers into LMR/NLR/PLR-high and low groups. We evaluated the resected specimens by immunohistochemistry in order to assess the densities of CD3+ and CD8+ lymphocytes in the center of the tumor and in the invasive margin. We found that the level of CD3+ lymphocytes in the center of the tumor was statistically significantly higher in patients with low pre-treatment NLR (p = 0.044); however, no correlation between any of the SIR markers and CD3+ or CD8+ TILs was observed. Five-year overall survival (OS) was longer in patients with high LMR (p < 0.001), low NLR (p = 0.001) and low PLR (p = 0.095). No correlation between the density of TILs and OS was demonstrated. In conclusion, based on our study, peripheral blood-based markers and CD3+ and CD8+ TILs are not interrelated.

The topic of the Pope’s authority has been known for nearly two millennia. To what extent, however, does this authority rely on a prophetic element? The question of prophecy in the Church – ekklesia – was tackled at the Second Vatican Council. This was primarily a response to the problems that had arisen in the relations between ekklesia and the world. These relations began to change from at least the end of the Middle Ages, and were almost lost during the Enlightenment. Papal authority of a prophetic character can be interpreted precisely in such ecclesiological context because the prophetic nature of Christianity is at the foundation of the community as the Church. The prophetic authority of Pope Francis is based precisely on this prophetism of the Church. The terminology of the description of the prophetic authority of the Pope has its origins in biblical prophecy. Its basis is in a tripartite Christian mission, one of which is referred to as prophetic. This prophetic aspect of the mission is closely related to teaching and the Teacher Office which bases in the prophetic mission discussed. Thus the teaching authority of the Pope is also apparent from his participation in the prophetic mission of Christ. The papal authority of the Pope is an element of the fulfillment of the tasks of the ekklesia sent to the world with a mission. It means discovering the problems of this world and, above all, providing their solutions. In view of a world which is simultaneously given and created, the figure of the Pope, and today Pope Francis, expresses the authority of the biblical prophet, discovering the challenges, expressing admonitions, but at the same time giving hope resulting from the realization of the basilea or the Kingdom.

The management of the power transformer population is a complex process, as the grid companies operate thousands of devices. For this issue, the health index method can be applied to facilitate asset management. The algorithm can be used not only in the technical assessment of the individual units, but also to determine the relationships within the whole population. In this paper, the presented health index method consists of periodic oil diagnostics, including the physicochemical properties, dissolved gas analysis, and furfural content, and further assessment in terms of the criticality of the device to determine the technical condition. The algorithm was specifically designed to reflect even the smallest changes of the input parameters in the final score. The performance of the health index was tested on 620 oil analyses from 220 transformers divided into four subpopulations based on the service conditions. The results have proven to be largely dependent on the criticality level and the operating conditions of the device. The analysis of the study group has shown the influence of corrective maintenance on the mean value of the health index score.

This paper presents the dielectric response of the insulation of bushings as an effect of the simulated long-term aging process. The experiment was conducted under a condition of a high temperature difference between the current circuit and the cover. The dielectric response was measured with the FDS (Frequency Dielectric Spectroscopy) and the SVM (Step Voltage Measurement) methods. The research has shown the correlation between the aging time and some parameters obtained with the FDS and SVM analysis.

The babylonian judaism, that has formed the babylonian gemara to the mishnah, came into contact mainly with the syriac christianity. The syriac language, which were used there belongs with mandaic and aramaic of the babylonian talmud to the same group of the eastern aramaic dialects. undoubtedly this situation has facilitated contact and exchange of ideas between jewish and local christian communities. both the jewish synagogues and the syriac churches were using the same text of tanakh, which was translated into syriac probably in the judeo-christian circles. Thus has the jewish community known the syriac text of the new testament? If so, then in which version: Diatessaron, Peshitta or Vetus Syra of the new testament? an important source of information about the relationship between jewish and christian communities are apocrypha – some developed in the common literary circles. In addition, the syriac patristic literature, on the one hand, shows the influence of judaic exegetical techniques or at least its literary motifs on the christian literature. on the other hand, it reflects the growing conflict between religions. some features of this literature could have some influence on the development of the babylonian gemara topics related to jesus, mary and christians. Thus, the syrian christianity was an eyewitness to the gradual detachment of christianity from judaism, even in the third century and the disappearance of judeo-christianity.
