Uncontrolled inflammation is a crucial factor in the development of many diseases. Anti-inflammatory molecules based on natural sources are being actively studied, among which Aristida depressa Retz (Ar.dp) has been traditionally used as a paste to heal inflammation. The present study aimed to evaluate the anti-inflammatory, analgesic, and antipyretic potential of an ethanolic extract of A. depressa through a battery of in vivo and in vitro models. The ethanolic extract of A. depressa was prepared by maceration and chemically characterized using high-performance liquid chromatography, which revealed the presence of quercetin, vanillic acid, chlorogenic acid, p-coumaric acid, m-coumaric acid, ferulic acid, cinnamic acid, and sinapic acid; its antioxidant capacity was then screened with the DPPH in vitro assay, which indicated moderate scavenging capacity. A protein denaturation assay was next performed to evaluate the in vitro anti-inflammatory potential of Ar.dp, which showed significant inhibition (44.44%) compared to the standard drug (diclofenac sodium), with 89.19% inhibition at a concentration of 1 mg/mL. The in vivo safety profile of Ar.dp was evaluated in accordance with the OECD-425 acute toxicity guidelines and found to be safe up to 5 g/kg. The in vivo anti-inflammatory potentials of Ar.dp were evaluated at three different doses (125, 250, and 500 mg/kg) in acute (carrageenan-induced edema: 84.60%, histamineinduced paw edema: 84%), sub-chronic (cotton-pellet-induced granuloma: 57.54%), and chronic (complete-Freund's-adjuvant-induced arthritis: 82.2%) models. Our results showed that Ar.dp had significant (p < 0.05) antiinflammatory effects over diclofenac sodium in the acute and chronic models. Histopathology studies indicated reduced infiltration of paw tissues with inflammatory cells in Ar.dp-treated animals. Similarly, Ar.dp showed significant (p < 0.05) analgesic (yeast-induced-pyrexia model: 23.53%) and antipyretic (acetic-acid-induced writhing model: 51%) effects in a time-dependent manner. In silico studies on the interactions of COX-1 and COX-2 with the eight ligands mentioned earlier confirmed the inhibition of enzymes responsible for inflammation and fever. Based on the findings of the present study, it is concluded that Ar.dp has anti-inflammatory, analgesic, and antipyretic properties that are likely linked to its pharmacologically active phenolic bioactive molecules. Background: Fever remains a common clinical indicator of disease, accounting alone for over 25% of paediatric emergency rooms consultations. Perception of this important sign is a crucial prelude and determinant of outcome in febrile children. The aim was to determine knowledge and attitudes of parents regarding fever in their children. Methods: The study is a crosssectional descriptive study carried out at the Emergency Paediatric Unit of the University of Ilorin Teaching Hospital (UITH). Fourhundred under-five children presenting with fever were recruited along with their caregivers. A semi-structured questionnaire was administered to collect information on sociodemographics, and caregiver’s knowledge and attitudes regarding fever in their wards. Results: The mean age of the caregivers was 29.5 ± 4.46 years (Range 21 – 41 years). Only 30.3% of caregivers could correctly describe what fever was. The most frequently utilised fever detection method was tactile assessment. Only 3.2% of the caregivers used a thermometer to detect fever. Social class, maternal age and religion significantly influenced the decision to use thermometers. The most common remedies caregivers would use for fever in their children were to give paracetamol (96.3%), sponging (73.2%) and a warm bath (63.8%). About 61% of caregivers had a wrong perception of the possible complications of fever. Conclusions: Caregivers in this study had a poor knowledge of fever, and parental educational and socioeconomic status impacted on their responses to fever. There is a need for education of caregivers at all contacts with the healthcare system on fever, and its management.

Background and purpose: Inflammation, fever, and pain can be associated with several diseases, and the synthetic drugs used in the treatment of these conditions often have severe side effects. As a result, there is a need for effective, economical, and safe alternative drugs, such as those derived from medicinal plants. Therefore, this study aimed to evaluate the anti-inflammatory, antipyretic, analgesic, and antioxidant activities of Castanopsis costata leaf fractions (CcLF), as well as its acute toxicity. Experimental approach: For anti-inflammatory, antipyretic, and analgesic tests, rats were given CcLF (WFCC, EAFcC, and n-HFCC) at 50 and 100 mg/kg, diclofenac sodium (10 mg/kg), paracetamol (150 mg/kg), aspirin (100 mg/kg), and tramadol (20 mg/kg). For the antioxidant activity test, various concentrations of CcLF were used ranging from 25 to 200 μg/mL. This study also looked into whether there could be any acute toxicity and histopathology of the liver, stomach, and kidneys in experimental animals. Findings/Results: The administration of CcLF significantly inhibited the increase in foot edema volume, and CcLF (EAFCC at 100 mg/kg) considerably decreased rectal temperature and was proportional to the standard drug paracetamol, and significantly inhibited pain sensation in various models. Additionally, CcLF showed strong antioxidant activity, and its administration at a dose limit of 5000 mg/kg/day did not show any toxic effects or death in test animals. Conclusions and implications: The results of the current confirmed that CcLF has demonstrated anti-inflammatory, antipyretic, analgesic, and antioxidant properties in experimental models, and is practically non-toxic.

The green synthesis of zinc oxide nanoparticles (ZnO NPs) using plants has grown in significance in recent years. ZnO NPs were synthesized in this work via a chemical precipitation method with Jasminum sambac (JS) leaf extract serving as a capping agent. These NPs were characterized using UV-vis spectroscopy, FT-IR, XRD, SEM, TEM, TGA, and DTA. The results from UV-vis and FT-IR confirmed the band gap energies (3.37 eV and 3.50 eV) and the presence of the following functional groups: CN, OH, C=O, and NH. A spherical structure and an average grain size of 26 nm were confirmed via XRD. The size and surface morphology of the ZnO NPs were confirmed through the use of SEM analysis. According to the TEM images, the ZnO NPs had an average mean size of 26 nm and were spherical in shape. The TGA curve indicated that the weight loss starts at 100 °C, rising to 900 °C, as a result of the evaporation of water molecules. An exothermic peak was seen during the DTA analysis at 480 °C. Effective antibacterial activity was found at 7.32 ± 0.44 mm in Gram-positive bacteria (S. aureus) and at 15.54 ± 0.031 mm in Gram-negative (E. coli) bacteria against the ZnO NPs. Antispasmodic activity: the 0.3 mL/mL sample solution demonstrated significant reductions in stimulant effects induced by histamine (at a concentration of 1 µg/mL) by (78.19%), acetylcholine (at a concentration of 1 µM) by (67.57%), and nicotine (at a concentration of 2 µg/mL) by (84.35%). The antipyretic activity was identified using the specific Shodhan vidhi method, and their anti-inflammatory properties were effectively evaluated with a denaturation test. A 0.3 mL/mL sample solution demonstrated significant reductions in stimulant effects induced by histamine (at a concentration of 1 µg/mL) by 78.19%, acetylcholine (at a concentration of 1 µM) by 67.57%, and nicotine (at a concentration of 2 µg/mL) by 84.35%. These results underscore the sample solution's potential as an effective therapeutic agent, showcasing its notable antispasmodic activity. Among the administered doses, the 150 mg/kg sample dose exhibited the most potent antipyretic effects. The anti-inflammatory activity of the synthesized NPs showed a remarkable inhibition percentage of (97.14 ± 0.005) at higher concentrations (250 µg/mL). Fever is a common complaint in the Emergency Department. Despite being frequently benign, it's still a cause for anxiety for caregivers. This study aimed to characterize parents' knowledge and attitudes towards fever. A cross sectional and descriptive survey was conducted by applying a questionnaire to pediatric patients' caregivers that attended consultation from April to August 2021. It included questions regarding sociodemographic data, knowledge and myths about fever and interventions used to treat it. Descriptive analysis was performed. A total of 1014 questionnaires were considered valid. The thermometer was used by 97.7% caregivers to measure fever, with armpit being the preferred location (80.9%). Around 87% wrongly defined fever. About 11.8% didn't offer antipyretics before seeking health care, mostly because of fear of masking the disease (47.7%). Approximately 91% undressed the child in thermal ascent and 49% used sponging with cold water to decrease temperature. Caregivers sought help mainly in primary care (66.2%). The physician was the preferred source of information (75.8%). Seizures (83.4%) were named as the main complication, 10.3% thought death was a possible effect. Around 78% answered dental eruption was a cause of fever, 54.8% considered it to be harmful for children's health, 67% believed temperature will go up indefinitely if not treated and 85.2% answered that temperature should always reach normal values after taking antipyretics. Fever phobia is still a reality, with wrong beliefs and misconceptions about its consequences and treatment. The current study was aimed to investigate the ethno-pharmacology, diversity, biological potentials of different mushrooms species from District Swabi, Pakistan. Ethno-pharmacological data was collected from 73 respondents based on interview, group discussion and field visits. Quantitative analysis such as informant consent factor (ICF), Use value (UV), Relative Frequency of Citation (RFC) were done. Morphological identification of mushrooms, extract preparation, phytochemical screening and statistical analysis of data was conducted. Phytochemical analysis of six mushroom species revealed that saponins, alkaloids, glycosides, flavonoids and phenols were present in six selected mushrooms, while tannin and steroids (−) present in Agaricus arvensis. In Morchella conica only steroids (−) was absent, Qunion (−) in Polyporus tuberaster, Pleurotus ostreatus and Omphalotus olearius were found absent. Moisture contents (10.17 ± 1.27) and crude fats (5.26 ± 1.50) present in Omphalotus olearius. Highest content of crude fibers (26.57 ± 0.86) and carbohydrates (62.18 ± 0.73) were found in Pleurotus ostreatus. Highest ash contents (12.62 ± 0.46) were reported in Polyporus tuberaster. The respondents’ consensus factor values ranged from 1.66 to 61.74 in which highest ICF value was 61.74 while the lowest was 1.66. The high RFC was recorded for Agaricus (0.67) while lowest RFC was observed in Clitocybe frondasa (0.05). The results of analgesic activity were compared to the diclofenac (Standard drug). The extracts of aqueous and methanolic extract were most active at the level of (400 mg/kg). As antipyretic agents, methanolic and aqueous extracts of Agaricus species were analyzed for anti-inflammatory activity compare to diclofenac (Standard drug) where the aqueous (200 mg/kg) and methanolic (400 mg/kg) were found to be very active. Finally, mushrooms with high (ICF and UV) were further investigated for phytochemicals and pharmacological analysis which revealed significant biological potentials. This provides basic ground information which can be used to explore novel mushroom-based medicines for the treatment of various diseases. The commercial cultivation of mushrooms represents a promising opportunity to leverage their diverse health benefits and nutritional properties. By prioritizing research and cultivation methods while educating the public, the potential of mushrooms as vital components of both nutrition and medicine can be significantly realized, impacting health outcomes and contributing to sustainable agricultural practices.

Argyreia nervosa bojar, a traditional and aesthetic medicinal plant is native to the Indian subcontinent. It has a large variety of traditional uses and therapeutically proven activities such as analgesic & anti-inflammatory, anti-cancer, anti-convulsant, anti-diarrheal, anti-fungal, antimicrobial, anti-obesity, antipyretic, anti-stress, anti-ulcer, anti-viral, aphrodisiac, CNS effect, hepatoprotective, hypoglycaemic, immunomodulatory, nootropic, wound healing activity, skin disorders, gonorrhoea, diabetes and sexual disorders. We aimed to systematically analyse pharmacological importance of A. nervosa to treat respiratory tract infection. As respiratory tract infection is accompanied with fever, pain, and inflammation. We collected literature from various online databases on antimicrobial, antipyretics, analgesic and anti-inflammatory activity of A. nervosa. We analysed the data based on eligibility criteria of the study and noted that, A. nervosa has high potential to treat many diseases, but it is ignored by researchers to find its pharmacological effect on respiratory tract infection. Our review found only four appropriate literatures available on various databases. This plant parts were noted effective against microbial activity of Streptococcus pneumoniae and Mycobacterium tuberculosis, responsible for causing respiratory tract infections. The chitin and chitosan biopolymers are extremely valuable because of their numerous industrial and pharmacological uses. Chitin and chitosan were extracted from the exoskeleton of Periplaneta americana (cockroaches) and termites using various acid and alkali techniques. The extraction process involves an initial demineralization step, during which integument dry powder was subjected to 500 mL (2.07 mol/L) of concentrated HCl at 100 degrees Celsius for 30 min, followed by meticulous rinsing with distilled water to restore the pH to its baseline. Deproteinization was conducted at 80 degrees Celsius using 500 mL (1 mol/L) of NaOH solution, which was repeated for 24 h. A total of 250 mL (0.06 mol/L) of NaOH was added at 100 degrees Celsius for 4 h to obtain chitosan, followed by extensive washing and subsequent drying. FTIR analysis was used to identify the functional groups in Periplaneta americana and termites. The crystallinity of these biopolymers, which have a face-centered cubic structure, was determined by X-ray diffraction analysis. This study assessed the analgesic properties of chitin and chitosan via an acetic-acid-induced writhing test in mice, revealing a significant reduction in writhing behavior following the chitin and chitosan extract. Notably, chitin exhibits the highest degree of analgesic activity compared to chitosan. Both chitin and chitosan show anti-inflammatory effects, with chitosan absorbing proton ions at sites of inflammation, while chitin effectively inhibits ear edema and elicits an analgesic response in mice. Furthermore, the present study revealed antipyretic activity, with termite chitin demonstrating the most significant effect at a concentration of 500 µL/mL, followed by chitosan and chitin at 100 µL/mL. These findings indicate the potential of using chitin and chitosan derived from termites and Periplaneta americana as natural anti-inflammatory compounds, implying prospective uses in anti-inflammatory, antipyretic, and analgesic capabilities. The aim of this study was to explore the antipyretic role and mechanism of Saposhnikoviae Radix (SR). To achieve the objectives TCM-ingredient-target network and PPI network were constructed by TCMSP, TTD, DisGenet, OMIM databases and Cytoscape 3.7.2 software, and topology analysis was performed on the PPI network. KEGG signaling pathways were analyzed using DAVID database, and molecular docking verification was performed on the core targets after topology analysis. Subsequently, 24 rats were randomly divided into control group, model group, wild high-dose group and wild low-dose group, 6 rats in each group. Results of in vivo experiments showed that high-dose group (P<0.01) and low-dose group (P<0.01) significantly decreased body temperature in rats; high-dose group significantly decreased TNF-α, IL-1β and IL-6 levels (P<0.01), and low-dose group significantly decreased TNF-α and IL-6 levels (P<0.01); high-dose group significantly decreased BAX, caspase3, COX2 and TNF-α protein expression levels in hypothalamus (P<0.01 or P<0.05), and low-dose group significantly increased BCL2 protein expression levels (P<0.01). It was found that SR can significantly inhibit lipopolysaccharide-induced fever in rats with elevated body temperature, reduce serum TNF-α, IL-1β and IL-6 levels, reduce the hypothalamus COX2, caspase3 and Bax protein expression levels and improve BCL2 protein expression levels, play an antipyretic effect. The objective of the current study was to evaluate the anti-inflammatory, analgesic, and antipyretic potential of Oxystelma esculentum using different animal models. The phytochemical profile was determined by assessing its total phenolic content (TPC) and total flavonoid content (TFC), followed by the high-performance liquid chromatography (HPLC) technique. The in vitro anti-inflammatory potential of O. esculentum ethanolic extract (OEE) was evaluated by lipoxygenase enzyme inhibition activity and a human red blood cell (HRBC) membrane stability assay. The in vivo anti-inflammatory potential of the plant was determined by the carrageenan-induced paw edema test, and the analgesic potential by the hot plate test, tail-flick test, formalin-induced analgesia, acetic acid-induced writhing activities, and yeast-induced elevation of body temperature. The values of total phenolic content (212.6 ± 3.18 µg GAE/g) and total flavonoid content (37.6 ± 1.76 µg QE/g) were observed. The results showed that OEE exhibited significant antioxidant capacity in DPPH (2,2- diphenyl-1-picrylhydrazyl) (266.3 ± 7.35 μmol TE/g), ABTS (2,2'-azino-bis(3- ethylbenzothiazoline-6-sulfonic acid) (1,066.3 ± 7.53 μmol TE/g), and FRAP (ferric reducing antioxidant power) (483.6 ± 3.84 μmol TE/g) assays. The HPLC analysis demonstrated phytocompounds with anti-inflammatory potential, such as chlorogenic acid, gallic acid, 4-hydroxybenzoic acid, caffeic acid, ferulic acid, and coumarin. The plant showed in vitro anti-inflammatory activity through the inhibition of lipoxygenase enzyme with a high percentage (56.66%) and HRBC membrane stability (67.29%). In in vivo studies, OEE exhibited significant (p < 0.05) anti-inflammatory (carrageenan-induced paw edema model), analgesic (hot plate test, tail-flick test, formalin-induced analgesia, and acetic acid-induced writhing), and antipyretic (rectal temperature reduction) responses at different doses (100, 300, and 500 mg/kg). Molecular docking studies showed significant binding affinities of phytocompounds compared to indomethacin and predicted various binding interactions for stable conformations. Cordia dichotoma Forst. has been utilized in the administration of diabetes in conventional medication. An analgesic-antipyretic drug widely used is paracetamol, which has various health benefits and several adverse effects. Therefore, various natural products have been extensively studied as alternative analgesic-antipyretics, one of which is sweet orange peel. This study aimed to investigate sweet orange peel's analgesic and antipyretic activity by in vivo methods. This experimental study evaluated the analgesic and antipyretic effects of sweet orange peel extract extracted by the maceration method. The analgesic effect was evaluated by tail immersion (Maximum Possible Analgesia) and acetic acid-induced writhing method (total abdominal writhing). Meanwhile, the antipyretic effect was evaluated by the brewer yeast-induced hyperpyrexia (body temperature) method. This study showed that sweet orange peel methanol extract significantly increased the maximum possible analgesia value (132.79%) and reduced the number of abdominal writhing (44.05%) at the highest dose of 750 mg/kg BW. It indicated analgesic activity from sweet orange peels. Meanwhile, the antipyretic effect of sweet orange peel methanol extract was observed from 1-4 hours after administration, and the highest percentage inhibition of body temperature 4 hours after administration was found in a moderate dose, that was 5.98% (P value: 0.042). Therefore, it can be concluded that sweet orange peel methanol extract has analgesic and antipyretic effects with an optimal dose range of 500-750 mg/kg BW.

This study aims to determine the effect of neurogenic, inflammatory, and infective fevers on acutely injured human spinal cord. In 86 patients with acute, severe traumatic spinal cord injuries (TSCIs; American Spinal Injury Association Impairment Scale (AIS), grades A–C) we monitored (starting within 72 h of injury, for up to 1 week) axillary temperature as well as injury site cord pressure, microdialysis (MD), and oxygen. High fever (temperature ≥38°C) was classified as neurogenic, infective, or inflammatory. The effect of these three fever types on injury-site physiology, metabolism, and inflammation was studied by analyzing 2864 h of intraspinal pressure (ISP), 1887 h of MD, and 840 h of tissue oxygen data. High fever occurred in 76.7% of the patients. The data show that temperature was higher in neurogenic than non-neurogenic fever. Neurogenic fever only occurred with injuries rostral to vertebral level T4. Compared with normothermia, fever was associated with reduced tissue glucose (all fevers), increased tissue lactate to pyruvate ratio (all fevers), reduced tissue oxygen (neurogenic + infective fevers), and elevated levels of pro-inflammatory cytokines/chemokines (infective fever). Spinal cord metabolic derangement preceded the onset of infective but not neurogenic or inflammatory fever. By considering five clinical characteristics (level of injury, axillary temperature, leukocyte count, C-reactive protein [CRP], and serum procalcitonin [PCT]), it was possible to confidently distinguish neurogenic from non-neurogenic high fever in 59.3% of cases. We conclude that neurogenic, infective, and inflammatory fevers occur commonly after acute, severe TSCI and are detrimental to the injured spinal cord with infective fever being the most injurious. Further studies are required to determine whether treating fever improves outcome. Inflammation, fever, and pain are common symptoms that can adversely affect overall health and quality of life. Hedyotis capitellata Wall. ex G. Don (Rubiaceae) is a traditional herbal medicine that alleviates these symptoms. However, scientific evidence supporting its effectiveness remains limited. Therefore, this study aimed to evaluate the potential of H. capitellata in controlling pain, fever, and inflammation. The analgesic properties of H. capitellata, were assessed using acetic acid and formalin tests. Paw edema induced by 1% carrageenan was used to evaluate anti-inflammatory activity, while the yeast-induced fever model was employed to investigate the antipyretic activity of H. capitellata. Ethanol leaf extract of H. capitellata (EHCW) was administered at different doses of 200, 250, and 300 mg/kg, with positive control groups receiving aspirin, tramadol, indomethacin, and paracetamol, and a negative control group receiving normal saline. The results showed that ethanol leaf extract of H. capitellata exhibited the highest analgesic activity (p < 0.05) at 300 mg/kg. It also demonstrated significant inhibition of paw edema (p < 0.05), at 300 mg/kg. Ethanol leaf extract of H. capitellata demonstrated a significant reduction (p < 0.05) in rectal temperature in the yeast-induced fever model, with the maximum effect at 300 mg/kg. Scutellaria sibthorpii is used in treatment of bacterial infections, pains and inflammations. The leaf was extracted by maceration and then partitioned with hexane, ethyl acetate and n-butanol. The extract was screened for phytochemicals. The antioxidant activity was assayed using DPPH, H2O2 and ß-carotene. The total phenolic and flavonoid contents were estimated using Folin-Ciocalteu's and AlCl3. The crude extract/fractions were tested on E. coli, S. typhi, B. subtilis and S. aureus using agar disk diffusion. Lorke's method was used to determine the LD50. The analgesic activity was determined using ethanoic acid-induced writhing, hotplate and formalin-induced nociception. Anti-inflammatory activity was investigated by means of formalin-induced acute inflammation. The antipyretic activity was studied using Brewer's yeast. The ethyl acetate fraction (EtFSs) had IC50 of 0.4352mg/mL in the H2O2 and IC50 of 0.00014mg/mL. The MIC of EtFSs against S. aureus was 1.25mg/mL and 2.5mg/mL on S. typhi. LD50 of the CrESs/fractions were greater than 5000mg/Kg. The analgesic and anti-inflammatory activities were higher in EtFSs when compared to piroxicam. The fractions decrease the rectal temperature of the rats in the same way as paracetamol at 200mg/Kg. Lamiaceae is a large plant family with valuable curative properties and consequently present a great interest in pharmacological development. Thus, the present study aims to investigate the anti-pyretic and anti-inflammatory effects of Thymus vulgaris L. leaf extracts in rats and rabbits. Essential oils and aqueous leaf extract were prepared by using hydrodistillation in Clevenger apparatus and infusion methods respectively. The anti-pyretic activity was in vitro tested using Limulus amebocyte lysate (LAL) test, and the in vivo study was performed on rabbits. The evolution of local acute inflammation induced by caraghenin injection was followed in rats treated with different concentrations of essential oil and aqueous extract. The latter have caused a non-dose dependent pyrogenic effect. Acetaminophen (APAP) is a widely used drug known for its analgesic and antipyretic properties. However, its exact mechanism of action is still unclear and has been primarily associated with its effects on COX enzymes and serotonergic pathways. Till date, the immunological mechanisms affected by APAP has gained only inadequate attention. A distinct group of immune cells called Group 2 Innate Lymphoid Cells (ILC2s) are known to play an important role in type 2 cytokine-mediated immunity and their regulatory dysfunction is associated with numerous type 2 pathologies, such as allergy. Conversely, another class of recently characterized Innate Lymphoid Cells called Innate Regulatory Cells (ILCregs) suppress the activation of ILC2s. Maintaining a balance between ILC2s and ILCregs is vital for achieving a well-controlled immune response and tissue homeostasis. Recent studies have highlighted the critical role of prostaglandin D2 (PGD2) in the maturation and development of ILC2 functions. Since APAP also suppresses PGD2 synthesis, it was speculated that APAP treatment could reduce the number of ILC2s while potentially increasing the number of ILCregs. To investigate this hypothesis, here, we administered therapeutic doses of APAP to OVA-sensitized mice, a well-established model of type 2 pathological inflammation. The mice received oral doses of 200 mg/kg body wt. of APAP twice weekly, along with weekly OVA sensitizations for six weeks. The mice were sacrificed at different time points (days 14, 28 and 42) to assess the kinetics of ILC2s, ILCregs and immunoglobulins (IgE and IgG1). The results demonstrated that APAP treatment effectively suppressed OVA-induced ILC2s while significantly increasing the number of IL-10+ ILCregs. APAP exposure also led to decreased levels of serum PGD2, OVA-specific IgE and IgG1, and enhanced the level of IL-10 in OVA sensitized mice. Moreover, OVA sensitized mice treated with APAP did not develop pathological changes in spleen, when compared to OVA sensitized mice. Additionally, APAP treatment did not cause any adverse effects on mice liver in treatment groups. This study investigates the analgesic and antipyretic effects of Gardenia jasminoides Ellis fruit in mice. In vivo experiments were conducted using ethanol extract from Gardenia jasminoides fruit (EEGJ) at 75, 100, and 125 mg/kg. The positive control group was treated with standard medications, including aspirin (10 mg/kg) and fentanyl (10 mL/kg) for pain experiments, and acetaminophen (10 mg/kg) for fever experiments. In contrast, the negative control group received normal saline (10 mL/kg). The analgesic effectiveness of EEGJ was assessed by measuring response latency and the reduction in writhing responses. Fever was induced by injecting a 20% yeast suspension (10 mL/kg), and rectal temperature was measured both before and after the intervention. The results demonstrated that EEGJ significantly reduced acetic acid-induced writhing responses by 25.8%, 34.78%, and 48.27%, respectively. Furthermore, this extract exhibited a pronounced dose-dependent inhibition of formalin-induced pain during the second phase, with inhibition rates of 15.31%, 25.23%, and 33.57%, respectively. The highest pain inhibition due to mechanical stimulus in the Haffner tail clip test at 15 minutes was observed at a dose of 125 mg/kg, achieving an inhibition rate of 43.08%. Additionally, after 3 hours, rectal temperature significantly decreased in the EEGJ 125 mg/kg group (37.29 ± 0.12oC) (p < 0.05). In conclusion, our study unequivocally demonstrates that ethanol extract from Gardenia jasminoides fruit exhibits potent analgesic and antipyretic effects, with the highest activity observed at the dose of 125 mg/kg. The objective of the current study was to evaluate the anti-inflammatory, analgesic, and antipyretic potential of Oxystelma esculentum using different animal models. The phytochemical profile was determined by assessing its total phenolic content (TPC) and total flavonoid content (TFC), followed by the high-performance liquid chromatography (HPLC) technique. The in vitro anti-inflammatory potential of O. esculentum ethanolic extract (OEE) was evaluated by lipoxygenase enzyme inhibition activity and a human red blood cell (HRBC) membrane stability assay. The in vivo anti-inflammatory potential of the plant was determined by the carrageenan-induced paw edema test, and the analgesic potential by the hot plate test, tail-flick test, formalin-induced analgesia, acetic acid-induced writhing activities, and yeast-induced elevation of body temperature. The values of total phenolic content (212.6 ± 3.18 µg GAE/g) and total flavonoid content (37.6 ± 1.76 µg QE/g) were observed. The results showed that OEE exhibited significant antioxidant capacity in DPPH (2,2-diphenyl-1-picrylhydrazyl) (266.3 ± 7.35 μmol TE/g), ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (1,066.3 ± 7.53 μmol TE/g), and FRAP (ferric reducing antioxidant power) (483.6 ± 3.84 μmol TE/g) assays. The HPLC analysis demonstrated phytocompounds with anti-inflammatory potential, such as chlorogenic acid, gallic acid, 4-hydroxybenzoic acid, caffeic acid, ferulic acid, and coumarin. The plant showed in vitro anti-inflammatory activity through the inhibition of lipoxygenase enzyme with a high percentage (56.66%) and HRBC membrane stability (67.29%). In in vivo studies, OEE exhibited significant (p < 0.05) anti-inflammatory (carrageenan-induced paw edema model), analgesic (hot plate test, tail-flick test, formalin-induced analgesia, and acetic acid-induced writhing), and antipyretic (rectal temperature reduction) responses at different doses (100, 300, and 500 mg/kg). Molecular docking studies showed significant binding affinities of phytocompounds compared to indomethacin and predicted various binding interactions for stable conformations. The results of in vitro, in vivo, and in silico studies supported the anti-inflammatory, analgesic, and antipyretic potential of O. esculentum.

OBJECTIVE: To determine the pharmacokinetics and clinical safety of acetaminophen after oral administration of 40 mg/kg q 12 hours or 60 mg/kg q 24 hours for 14 days. ANIMALS. 12 healthy light-breed neonatal foals. PROCEDURES: 6 foals received acetaminophen at 40 mg/kg q 12 hours and 6 foals received 60 mg/kg q 24 hours for 14 days. The study dates were January 31 to April 15, 2023. Physical examinations were performed daily. Plasma disposition of acetaminophen was determined after the first, mid-point drug administration. Hematology and biochemistry analysis was performed before the study, day 7, and the last day of drug administration. Plasma acetaminophen concentrations were determined by high-performance liquid chromatography. Plasma pharmacokinetic parameters were estimated using noncompartmental analysis. RESULTS: No statistically significant changes occurred on hematology or biochemistry profiles. Elevations in γ-glutamyl transferase (GGT) and sorbitol dehydrogenase (SDH) were noted in 4 foals at various time points. The maximum plasma concentration (Cmax) occurred within 2 hours for both doses. The 60 mg/kg dose resulted in a larger median Cmax (range) at 28 µg/mL (22-32) than the 40 mg/kg dose at 23 µg/mL (19-27). The median area under the concentration-vs-time curve from 0 to 8 hours (AUC0-8 hour [range]) was 100 h•µg/mL (82-100) at 40 mg/kg and 128 h•µg/mL (120-168) for 60 mg/kg. Trough concentrations decreased over time for both regimens. CLINICAL RELEVANCE: Foals tolerate oral acetaminophen at 40 mg/kg q 12 hours or 60 mg/kg q 24 hours. By using Proton Nuclear Magnetic Resonance spectroscopy, Carbon -13 NMR spectroscopy, and mass spectroscopy, a novel series of pyrazolyl indoline-3-one (5a-5j) hybrids, were created. At a dose of ten mg, each final product's antiinflammatory, analgesic, and antipyretic qualities were evaluated. The weight of each Wistar rat in kilograms were taken. The outcomes were compared with a reference standard, Indomethacin. It came to the conclusion that the final compounds had positive anti-inflammatory, analgesic, and antipyretic properties. Abstract Background Paracetamol (acetaminophen) is recommended as a first‐line drug in the management of pain and pyrexia in humans due to its minor gastrointestinal, renal and vascular side effects. Objectives To explore the perceptions and use of paracetamol in dogs by UK veterinarians. Methods Veterinarians were invited to complete an online survey. Questions addressed career history, previous use, and personal perceptions of the use of paracetamol in dogs. Results A total of 450 veterinarians were included in the survey; 96% (n = 431) of whom worked in small animal practice only. The majority reported a frequency of use of 1–3 times a week (n = 197), with oral dosing being the most common route of administration (n = 440). Only 8 had never prescribed paracetamol. Paracetamol was more commonly used to provide analgesia (n = 431) than for the treatment of pyrexia (n = 292) and was predominantly used as an alternative to NSAIDs (n = 408) or as part of multimodal analgesia (n = 406). Hepatotoxic (n = 311) and gastrointestinal (n = 120) side effects were a common concern, although the majority perceived a low risk of side effects and felt they were likely to be mild if they did occur (n = 279). Only 5% (n = 24) of respondents reported having experienced side effects, with gastrointestinal signs (n = 14) and hepatotoxicity (n = 6) being the most common. Conclusions Paracetamol is widely used by veterinarians working in the UK. Most veterinarians have changed their perception on the use of paracetamol over the last decade and consider it to be associated with fewer side effects and with more effective analgesic and antipyretic effects than they had previously believed.

The green synthesis of zinc oxide nanoparticles (ZnO NPs) using plants has grown in significance in recent years. ZnO NPs were synthesized in this work via a chemical precipitation method with Jasminum sambac (JS) leaf extract serving as a capping agent. These NPs were characterized using UV-vis spectroscopy, FT-IR, XRD, SEM, TEM, TGA, and DTA. The results from UV-vis and FT-IR confirmed the band gap energies (3.37 eV and 3.50 eV) and the presence of the following functional groups: CN, OH, C=O, and NH. A spherical structure and an average grain size of 26 nm were confirmed via XRD. The size and surface morphology of the ZnO NPs were confirmed through the use of SEM analysis. According to the TEM images, the ZnO NPs had an average mean size of 26 nm and were spherical in shape. The TGA curve indicated that the weight loss starts at 100 °C, rising to 900 °C, as a result of the evaporation of water molecules. An exothermic peak was seen during the DTA analysis at 480 °C. Effective antibacterial activity was found at 7.32 ± 0.44 mm in Gram-positive bacteria (S. aureus) and at 15.54 ± 0.031 mm in Gram-negative (E. coli) bacteria against the ZnO NPs. Antispasmodic activity: the 0.3 mL/mL sample solution demonstrated significant reductions in stimulant effects induced by histamine (at a concentration of 1 µg/mL) by (78.19%), acetylcholine (at a concentration of 1 µM) by (67.57%), and nicotine (at a concentration of 2 µg/mL) by (84.35%). The antipyretic activity was identified using the specific Shodhan vidhi method, and their anti-inflammatory properties were effectively evaluated with a denaturation test. A 0.3 mL/mL sample solution demonstrated significant reductions in stimulant effects induced by histamine (at a concentration of 1 µg/mL) by 78.19%, acetylcholine (at a concentration of 1 µM) by 67.57%, and nicotine (at a concentration of 2 µg/mL) by 84.35%. These results underscore the sample solution’s potential as an effective therapeutic agent, showcasing its notable antispasmodic activity. Among the administered doses, the 150 mg/kg sample dose exhibited the most potent antipyretic effects. The anti-inflammatory activity of the synthesized NPs showed a remarkable inhibition percentage of (97.14 ± 0.005) at higher concentrations (250 µg/mL). Furthermore, a cytotoxic effect was noted when the biologically synthesized ZnO NPs were introduced to treated cells.

The chitin and chitosan biopolymers are extremely valuable because of their numerous industrial and pharmacological uses. Chitin and chitosan were extracted from the exoskeleton of Periplaneta americana (cockroaches) and termites using various acid and alkali techniques. The extraction process involves an initial demineralization step, during which integument dry powder was subjected to 500 mL (2.07 mol/L) of concentrated HCl at 100 degrees Celsius for 30 min, followed by meticulous rinsing with distilled water to restore the pH to its baseline. Deproteinization was conducted at 80 degrees Celsius using 500 mL (1 mol/L) of NaOH solution, which was repeated for 24 h. A total of 250 mL (0.06 mol/L) of NaOH was added at 100 degrees Celsius for 4 h to obtain chitosan, followed by extensive washing and subsequent drying. FTIR analysis was used to identify the functional groups in Periplaneta americana and termites. The crystallinity of these biopolymers, which have a face-centered cubic structure, was determined by X-ray diffraction analysis. This study assessed the analgesic properties of chitin and chitosan via an acetic-acid-induced writhing test in mice, revealing a significant reduction in writhing behavior following the chitin and chitosan extract. Notably, chitin exhibits the highest degree of analgesic activity compared to chitosan. Both chitin and chitosan show anti-inflammatory effects, with chitosan absorbing proton ions at sites of inflammation, while chitin effectively inhibits ear edema and elicits an analgesic response in mice. Furthermore, the present study revealed antipyretic activity, with termite chitin demonstrating the most significant effect at a concentration of 500 µL/mL, followed by chitosan and chitin at 100 µL/mL. These findings indicate the potential of using chitin and chitosan derived from termites and Periplaneta americana as natural anti-inflammatory compounds, implying prospective uses in anti-inflammatory, antipyretic, and analgesic capabilities.

Background: Artabotrys hexapetalus is a rare plant in the Indian subcontinent. This plant is used for different therapeutic purposes in folk medicine and many bioactive compounds are reported in this plant. Considering its traditional uses and the presence of medicinal compounds, we aimed to evaluate some pharmacological properties of its leaves along with in silico analysis in scientific ways. Methods: Antioxidant tests were conducted in both qualitative and quantitative ways. 2,2-Diphenyl-1-picryl hydrazyl (DPPH), hydrogen peroxide, and superoxide radicals scavenging with FeCl3 reducing properties of A. hexapetalus extract were carried out. Acetic acid-induced writhing, tail immersion, formalin-induced paw edema, and Brewer's yeast-induced pyrexia methods were followed to conduct peripheral analgesic, central analgesic, anti-inflammatory, and antipyretic potentialities, respectively. Finally, molecular docking and absorption, distribution, metabolism, excretion, and toxicity (ADMET) analysis of reported compounds were conducted. Results: The A. hexapetalus extract was found to possess a quite good amount of phenolics, flavonoids, and tannins. This extract also revealed good radical scavenging effects along with ferric chloride-reducing capabilities. In the analgesic tests, the extract showed excellent responses in writhing inhibition and elongation in tail withdrawal time. The extract also significantly lessened paw edema over time at both doses. A notable decrease in body temperature was also observed. Those responses revealed nice pharmacological potentialities. Along with these findings and conducting in silico analysis, we finally proposed seven molecules as lead compounds. Conclusion: Our analysis revealed that A. hexapetalus leaves are highly enriched with antioxidants and the extract showed good pharmacological characteristics.

Polystichum lonchitis L. is a fern belonging to the family Dryopteridaceae. The present study was conducted to evaluate its pharmacological, antioxidant, and phytochemical properties, and to conduct GC–MS screening of P. lonchitis. The acetic acid-induced writhing test, yeast-induced hyperpyrexia method, carrageenan-induced rat paw edema model, and charcoal meal test model were carried out to assess analgesic, antipyretic, anti-inflammatory, and antispasmodic activity, respectively. DPPH was used as an antioxidant, while the phytochemical screening was conducted using standard scientific methods. Among the pharmacological activities, the most significant effects were observed in the analgesic and anti-inflammatory activities, followed by the antipyretic and antispasmodic activities, at a dose of 450 mg/kg after the 4th hour, compared with 150 mg/kg and 300 mg/kg. For the evaluation of antioxidant activities, the most significant results were detected in the methanolic and aqueous extracts. The detection of flavonoids and phenol occurred most significantly in the methanolic extract, and then in the ethanolic and aqueous extracts. The main compounds detected using GC–MS analysis with a high metabolic rate was -D-Galactopyranoside, which had a metabolic rate of 0.851, and methyl and n-hexadecanoic, which had a metabolic rate of 0.972. Overall, the results suggested that P. lonchitis had a strong potential for pharmacological activities. The suggested assessment provided a way to isolate the bioactive constituents and will help to provide new medicines with fewer side effects. Red dragon fruit (Hylocereus polyrhizus) is a tropical fruit that is currently cultivated in all tropical parts of the world. It is popular to consume its fruit flesh, while the peel is often thrown away. This red dragon fruit peel (RDFPE) is known to possess lots of phytochemical compounds with multitudes of usage, amongst them as an analgesic and antipyretic. Therefore, the active compounds of RDFPE play an important role in the natural product. This study aims to observe and analyze the analgetic and antipyretic activity possessed by red dragon fruit peel. This study is an in-vivo experiment on 25 white male rats divided into five groups, in which each group receive Na CMC, acetaminophen, and the other three groups will receive three different doses of RDFPE (500 mg/kg, 750 mg/kg, and 1000 mg/kg), respectively. Acetic acid writhing and tail immersion methods were performed to induce inflammation and Brewer’s yeast injection performed induced pyrexia. In the investigation of the acetic acid writhing test, the intervention was administered before induction, and for the tail immersion test, induction was given before and after the intervention was administered. Meanwhile, in brewer’s yeast-induced pyrexia, the rectal temperature was measured before induction, 24 hours after induction, and each hour for five four after the intervention; intervention was administered 24 hours after induction. Abstract Background Chukrasia velutina is an enthnomedicinally used plant reported to have significant medicinal values. The present study aimed to explore the pharmacological activities of bark methanol extract using in vitro, in vivo and in silico models. Methods The study was designed to investigate the pharmacological effects of methanol extract of Chukrasia velutina bark (MECVB) through in vitro, in vivo and in silico assays. Analgesic activity was tested using formalin‐induced nociception and acetic acid‐induced writhing assays while the antipyretic effect was tested using yeast‐induced hyperthermia in mice model. The antioxidant effect was tested using the DPPH and reducing power assay and the cytotoxic screening was tested using the brine shrimp lethality bioassay. In addition, in silico studies were conducted using computer aided methods. Antipyretic drugs such as suspensions of mefenamic acid (ponstan), Ibuprofen and paracetamol (acetaminophen) are the most common drugs that wildly used in children to decrease the fever, pain and inflammation. From clinical observations of children using these drugs, they cause gastrointestinal complications and from this, the idea of this research was to find the effect of these drugs on the mucous membrane of the gastrointestinal tract in Swiss albino mice. In the present study, we used 30 mice classified into five groups including G1 as control group, G2 received 15 mg/kg/day panadol, G3, 30 mg/kg/day ibuprofen, G4, 5 mg/kg/day ponstan and G5, a combination of panadol and ibuprofen in the same previously- mentioned doses for 7 days respectively. The gastric histological sections of G2 exhibited normal mucosal; G4 displayed mild mucosal glandular hyperplasia; while G3 and G5 e appeared flat mucosal surface with submucosal hyperplasia of gland mild atypical cells; and G2 revealed mucosal glandular hyperplasia. The intestine histological sections of G2 appeared normal intestinal villi with mild inflammatory cells infiltration; G3 showed dispersed slight shortening of intestinal villi with mild inflammatory cells infiltration; finally, G4 and G5 indicated villi hyperplasia with a slight widening of villi with mild inflammatory cells infiltration. NSAIDs are available over-the-counter drugs for adult and in paediatric population and are considered as a safe medicine if used in properly dose in the short-term. Background: Paracetamol (acetaminophen) is recommended as a first‐line drug in the management of pain and pyrexia in humans due to its minor gastrointestinal, renal and vascular side effects. Objectives: To explore the perceptions and use of paracetamol in dogs by UK veterinarians. Methods: Veterinarians were invited to complete an online survey. Questions addressed career history, previous use, and personal perceptions of the use of paracetamol in dogs. Results: A total of 450 veterinarians were included in the survey; 96% (n = 431) of whom worked in small animal practice only. The majority reported a frequency of use of 1–3 times a week (n = 197), with oral dosing being the most common route of administration (n = 440). Only 8 had never prescribed paracetamol. Paracetamol was more commonly used to provide analgesia (n = 431) than for the treatment of pyrexia (n = 292) and was predominantly used as an alternative to NSAIDs (n = 408) or as part of multimodal analgesia (n = 406). Hepatotoxic (n = 311) and gastrointestinal (n = 120) side effects were a common concern, although the majority perceived a low risk of side effects and felt they were likely to be mild if they did occur (n = 279). Only 5% (n = 24) of respondents reported having experienced side effects, with gastrointestinal signs (n = 14) and hepatotoxicity (n = 6) being the most common. Conclusions: Paracetamol is widely used by veterinarians working in the UK. Most veterinarians have changed their perception on the use of paracetamol over the last decade and consider it to be associated with fewer side effects and with more effective analgesic and antipyretic effects than they had previously believed. Objective: To compare the clinical effectiveness of Paracetamol with Ibuprofen as an antipyretic, fever relief and linked discomfort in children. Study Design: Comparative cross-sectional study. Place and Duration of Study: Combined Military Hospital, Abbottabad Pakistan from Apr to Nov 2019. Methodology: Children aged six months to five years, who reported fever between 38-41°C were included. Two temperature groups were developed, i.e., 37.8--38.9ºC and 39-41ºC with baseline temperatures of 38.60±0.56 for Paracetamol-Group and 38.58± 0.56ºC for Ibuprofen-Group. The prescriptions were given over to the parents with clarification about the dose of the medication. The medication had active and placebo forms. Information was gathered until saturation was done. Results: A total of 104 children were taken in the study. Of these, 77(74.08%) were females, and 27(24.96) were males. Comparing Ibuprofen-treated children with Paracetamol-Group, the final result showed that the extent of children's temperature management was greater in the Ibuprofen-Group 37(71%) than in the Paracetamol-Group 34(65%). Conclusion: Doctors, attendants and guardians wishing to use medications to treat children suffering from fever should be encouraged to use Ibuprofen first and to consider the relative risks and advantages of using Ibuprofen in addition to Paracetamol for fever. The leaves of Ficus lyrata warb (Moraceae) is used to treat GI disorder such as diarrhea in folklore use. The aim of present research is to explore the antidiarrheal and antipyretic activity of methanolic extract of leaves of F. lyrata. The diarrhea was induced by using castor oil. The anti-pyretic effects were also explored on yeast induced pyrexia. The physicochemical analysis of powder such as (total ash, water soluble ash, acid insoluble ash, moisture content, foaming index and swelling index) along with organoleptic evaluation of leaf of F. lyrata in order to access the quality Purity of the substance was also done. The phytochemical analysis of methanolic extract of F. lyrata leaves confirmed the presence of polyphenols, tannins, terpenoids, cardiac glycosides, flavonoids and reducing sugar. The antidiarrheal activity of MEFL at 100 and 200 mg/kg body weight showed 62% and 76% reduction in defecation respectively. The decrease in intestinal fluid accumulation at same dose revealed 67.2% and 75% reduction in fluid accumulation in intestine of Wister albino rats. The methanolic extract of F. lyrata showed reduction in distance travelled by charcoal meal such as 54% and 72% at dose 100 and 200 mg/kg respectively. The antipyretic effect at a dose of 100 and 200 mg/kg showed significant reduction in temperature. The anti-diarrheal activity of the F. lyrata might be due to the presence of flavonoids. Context: The anti-nociceptive and anti-pyretic properties of the crude aqueous extract of Anogeissus leiocarpa (DC). Guill. & Perr. leaves have been reported. Aims: To investigate the anti-inflammatory, anti-nociceptive, and anti-pyretic properties of aqueous extract of A. leiocarpa leaves (AEAL) and its fractions to identify the bioactive compound(s). Methods: AEAL was fractionated successively and eluted in gradients of solvent mixture (n-hexane, ethylacetate, and methanol). The eco-friendly green bio-synthesis of Ephedra-AuNPs was constructed from Ephedra plant extract. The synthesis process was optimized using different volumes of Ephedra-extract, 10-3M (HAuCl4·3H2O) solution, reaction temperature, time, and pH of the solution. Where, 4 mL of Ephedra-extract, 6 mL of 10-3M (HAuCl4·3H2O) solution at 25 °C for 3 h at a pH of 4 are showed the optimum reaction parameters respectively. The characterization of AuNPs was verified by UV–vis. spectrophotometer, FTIR, microscopic techniques (TEM, HRTEM), Zeta potential, and X-ray techniques (XRD, EDX, XPS). The nanoparticles are extremely important materials in emerging the future sustainable technologies for the living beings. The AuNPs were employed in biological assays where ABTS assay used to investigate the antioxidant activity. The in-vivo study of AuNPs was executed to study the anti-pyretic and anti-asthma activities of Ephedra-AuNPs. Ephedra-AuNPs of 1.3 nm to 15.6 nm size were achieved which have as importance as proficient to bind with the biological systems. The Ephedra-AuNPs prepared exploiting Ephedra-extract which is considered to be influential anti-asthma agent and antipyretic agent reduced the fever by 83.3 % at 32μg/g/day. Further the Ephedra-AuNPs used to scrutinize the anti-bacterial activities for Staphylococcus aureus, Listeria moncytogenes, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, and Salmonella typhimurium, and anti-fungal activities on Candida albicans, Aspergillus nigra and Aspergillus flavus. The synthesized Ephedra-AuNPs exhibited great potential activities comparable to the standard drugs in case of Staphylococcus aureus, Escherichia coli, Candida albicans with the zone of inhibition of 25 ± 0.5 nm, 26 ± 0.6 nm and 24 ± 0.7 nm respectively. It is believed that the synthesized Ephedra-AuNPs to be a hopeful option intended for diverse biological applications.

This study evaluates the pharmacological activity of the methanolic extract of Cissus elongata Roxb. leaves. Dried leaves of C. elongata were extracted with methanol. The crude extract was then tested to identify the presence of different phytoconstituents. To evaluate the pharmacological effects of this plant-analgesic, anti-diarrheal, antiinflammatory and antipyretic, tests were conducted using animal models. Analgesic activity was evaluated by an acetic acid-induced writhing test, formalin-induced paw-licking test paw-licking immersion test. Castor oilinduced anti-diarrheal test was performed to evaluate anti-diarrheal activity. Xylene-induced ear edema and brewer's yeast-induced pyrexia test were performed to investigate anti-inflammatory and antipyretic activity respectively. Phytochemical screening revealed the presence of carbohydrates, tannins, fat, and fixed oil. In writhing test, the extract showed 45.68±3.99 % and 52.28±1.67 % inhibition of writhing at a dose of 200 mg/kg and 400 mg/kg respectively whereas it was 73.09±1.01 % for the standard drug compared to the control group. In the formalin-induced paw-licking test, the percentage inhibition of licking by both doses of the extract was more in the delayed phase than in the acute phase. In the tail immersion test, 200 mg/kg dose didn't show a significant effect but 40 0mg/kg showed significant (*P<0.05 vs. control) analgesic activity at 30 min, 60 min and 120 min time periods. In an anti-diarrheal study, 400 mg/kg dose showed (68.42±0.87 % inhibition of diarrhea) almost similar anti-diarrheal effect as the standard drug loperamide HCl (71.05±0.58 %). In xylene induced ear edema test, both doses of the extract showed little anti-inflammatory effect compared to the standard drug but both doses showed significant (*P<0.05 vs. control) reduction in body temperature in the antipyretic study. The aim of this study is to evaluate the anti-obesity, anti-hyperglycaemic, analgesic and antipyretic activities of Globularia alypum (GA). GA methanol and water extracts (GAME, GAWE) were administered to high-fat-high-glucose diet (HFFD) rats. This study showed that GAME exhibited the highest antioxidant, anti-α-amylase and anti-lipase activities, with half inhibitory concentration (IC50) values 0.067, 1.05 and 2.97 mg/ml respectively. In HFFD rats, the administration of GAME inhibited lipase activity by 36, 37 and 30% in the intestine, pancreas and serum, respectively, reduced body weight by 17.7% and modulated lipid profile. In addition, administration of GAME to HFFD-rats decreased α-amylase activity, improved glucose level and protected liver function. Furthermore, the administration of GA extracts to rats revealed antipyretic (reduction in writhing by 64%) and analgesic (decrease of temperature by 1.11 °C) activities. Aim: To explore factors influencing fever management practices and antipyretic use among New Zealand Emergency Department (ED) doctors and nurses using the Theoretical Domains Framework (TDF).Methods: Cross-sectional survey of doctors and nurses across 11 New Zealand EDs. The questionnaire examined eight of 12 TDF domains, based on a generic questionnaire validated to assess TDF-based determinants of health-care professional behaviour. Relevant domains were identified by the frequency of beliefs; the presence of conflicting beliefs within a domain; and the likely strength of impact of a belief on paediatric fever management in the ED.Results: About 602 participants (243 doctors, 353 nurses and 6 unknown) completed the survey (response rate 47.5%). Over half (351/591, 59.6%, 95% confidence interval (CI) 55.5-63.5%) knew the content of clinical practice guidelines regarding antipyretic use in febrile children (TDF Domain Knowledge), or had been trained to ensure antipyretics are given to febrile children only if they appear distressed (347/592, 58.6%, 95% CI 54.5-62.6%) (Skills). Over 40% (246/590, 95% CI 37.7-45.8%) aim to reduce the fever before discharge (Goals). Most (444/591, 75.1%, 95% CI 71.4-78.6%) participants felt capable of explaining appropriate antipyretic use to parents/care givers (Beliefs about Capabilities). Only a minority (155/584, 26.5%, 95% CI 23.0-30.3%) thought that they can ensure antipyretics are given to febrile children only if they appear distressed when the ED is busy (Environmental Context and Resources).Conclusions: Using the TDF, we identified factors influencing fever management practices and antipyretic use in the ED. Khamira Banafsha (KB) is a semi-solid blend of three dried flowers, Viola odorata L., Borago officinalis L., and Rosa damascena Mill and the distillate of Rosa damascena, and sugar. The components in this formulation are well-known and frequently utilized in the treatment of respiratory problems. The formulation has been used to treat a wide range of illnesses for decades. This review will discuss the pharmacology, ethnopharmacology, and repurposing of KB as an adjuvant or symptomatic treatment for Covid-19 illness. Background A severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) mRNA vaccine booster elicits sufficient antibody responses that protect against coronavirus disease 2019, whereas adverse reactions such as fever have been commonly reported. Associations between adverse reactions and antibody responses have not been fully characterized, nor has the influence of antipyretic use. Methods This is a prospective observational cohort study in Japan, following our prior investigation of BNT162b2 2-dose primary series. Spike-specific immunoglobulin G (IgG) titers were measured for SARS-CoV-2–naive hospital healthcare workers who received a BNT162b2 booster. The severity of solicited adverse reactions, including the highest body temperature, and self-medicated antipyretics were reported daily for 7 days following vaccination through a web-based self-reporting diary. Results The data of 281 healthcare workers were available. Multivariate analysis extracted fever after the booster dose (β =.305, P <.001) as being significantly correlated with the specific IgG titers. The analysis of 164 participants with data from the primary series showed that fever after the second dose was associated with the emergence of fever after the booster dose (relative risk, 3.97 [95% confidence interval, 2.48–6.35]); however, the IgG titers after the booster dose were not associated with the presence or degree of fever after the second dose. There were no significant differences in the IgG titers by the use, type, or dosage of antipyretic medication. Conclusions These results suggest an independent correlation between mRNA vaccine–induced specific IgG levels and post–booster vaccination fever, without any significant influence of fever after the primary series. Dimethyl dimethoxy biphenyl (DDB) dicarboxylate has been applied as a therapeutic modality for curing liver diseases, particularly hepatitis virus. The objective of this study was to assess the protective potential against Triton X-100 induced abnormal fat metabolism in addition to anti-inflammatory, analgesic, and antipyretic effects of DDB. The anti-inflammatory, antinociceptive, and antipyretic of DDB were investigated through induction of paw edema, pain, and fever in experimental rats. DDB decreased cholesterol and triglyceride contents. DDB resulted in inhibition of inflammation, nociception, and fever in the experimental models. DDB improved lipid profile, as evidence of hypolipidemic potential. It also showed anti-inflammatory, analgesic, and antipyretic properties.
