Abstract

An analysis of the mercury loading in Lake Pepin, which is located on the Upper Mississippi River, can help described how the river has been managed over a number of years. In fact, the analysis showed that the implementation of pollution control efforts since about 1970 has had a pronounced effect on the amount of mercury accumulation in the sediment at Lake Pepin.

Abstract

Evaluation of total mercury (Hg) loadings and concentrations in Minnesota, Mississippi and St. Croix rivers reveals that watershed characteristics play an important role in Hg mobility. The study reveals that Hg levels in the Minnesota river are strongly correlated with total suspended solid concentrations while the Mississippi and St. Croix rivers have lower loadings and concentrations of Hg reflecting higher levels of dissolved mercury.

Abstract

A study of pollution in the Lower Minnesota River reveals that efforts in controling mercury loadings should be concentrated on controlling sediment loadings. The study of the total suspended sediment (TSS) and mercury concentrations in the Lower Minnesota River reveals that amounts remained low during winter, peaked during spring and remained relatively high in summer and fall. Mercury measurements were taken from both point and diffuse sources and were correlated with TSS concentrations. Point source mercury loadings account for only 1% and the remaining 99% comes from atmospheric precipitate.

Abstract

The precise developmental expression and molecular composition of gap junctions during myogenesis was investigated. RNA from cultures of rat L6 myoblasts were isolated and examined using Northern blot analysis with cDNA probes for connexin 32 and connexin 43. The results show that the expression of connexin 43 is temporally correlated with myoblast fusion and may play a role in this process.

Abstract

Mercury (Hg) stable isotope analysis is an emerging technique that has contributed to a better understanding of many aspects of the biogeochemical cycling of Hg in the environment. However, no study has yet evaluated its usefulness in elucidating the sources of methylmercury (MeHg) in songbird species, a common organism for biomonitoring of Hg in forested ecosystems. In the present pilot study, we examined stable mercury isotope ratios in blood of 4 species of songbirds and the invertebrates they are likely foraging on in multiple habitats in a small watershed of mixed forest and wetlands in Acadia National Park in Maine (USA). We found distinct isotopic signatures of MeHg in invertebrates (both mass-dependent fractionation [as δ202Hg] and mass-independent fractionation [as Δ199Hg]) among 3 interconnected aquatic habitats. It appears that the Hg isotopic compositions in bird blood cannot be fully accounted for by the isotopic compositions of MeHg in lower trophic levels in each of the habitats examined. Furthermore, the bird blood isotope results cannot be simply explained by an isotopic offset as a result of metabolic fractionation of δ202Hg (e.g., internal demethylation). Our results suggest that many of the birds sampled obtain MeHg from sources outside the habitat they were captured in. Our findings also indicate that mass-independent fractionation is a more reliable and conservative tracer than mass-dependent fractionation for identifying sources of MeHg in bird blood. The results demonstrate the feasibility of Hg isotope studies of songbirds but suggest that larger numbers of samples and an expanded geographic area of study may be required for conclusive interpretation. Abstract

The total concentration of all mercury species in water, [THg]w, is a key water quality measure. This paper proposes an analytical framework for the analysis, interpretation and eventual projection of stream (and river) [THg]w. Mercury (Hg) is conceptualized as being distributed among four carrier groups in water: suspended mineral particles, particulate organic carbon, dissolved organic carbon, and inorganic complexes and ions. The conceptual understanding is used as a statistical analytical framework and turned into a quantitative model for Minnesota and Wisconsin based on some 1000 data cases from 18 streams (watershed area: 9 ha–115500 km2). The resultant model yields a coefficient of determination of 92% for [THg]w of the fitting data subset, or 94% for the evaluation subset. Its coefficients have well defined physical meanings, and are generally consistent with independent field measurements from the same region or elsewhere. The model provides a comprehensive and quantitative view of the Hg contents of the carriers: the contents increase with the proportion of watershed area as wetlands or as forests, rise drastically and then fall precipitously during spring snowmelt, and otherwise peak in the growing season and reach a minimum in the presence of snowpack. The model further reveals that dissolved Hg tends to increase with emission-related wet sulfate deposition across the data region. [ABSTRACT FROM AUTHOR]

Abstract

Poly [(R)‐3‐hydroxybutyric acid] (PHB) is a prokaryote storage material for carbon and energy that accumulates in cells under unbalanced growth conditions. Because this class of biopolymers has plastic‐like properties, it has attracted considerable interest for biomedical applications and as a biodegradable commodity plastic. Current flow cytometric techniques to quantify intracellular PHB are based on Nile red. Here, an improved cytometric technique for cellular PHB quantification utilizing BODIPY 493/503 staining was developed. This technique was then automated using an automated flow cytometry system.Using flow cytometry, the fluorescence of Saccharomyces cerevisiae and Cupriavidus necator with varying PHB content after staining with BODIPY 493/503 and Nile red was compared, and automated staining techniques were developed for both cultures.BODIPY 493/503 staining had less background staining, higher sensitivity and specificity to PHB, and higher saturation values than did Nile red staining. The developed automated staining procedure was capable of analyzing the PHB content of a bioreactor sample every 25 min and measured the average PHB content with accuracy comparable to offline GC analysis.BODIPY 493/503 produced an overall better staining for PHB than did Nile red. When combined with the automated system, this technique provides a new method for the online monitoring and control of bioreactors. © 2005 Wiley‐Liss, Inc.
