A variety of experimental and epidemiological studies lend support to the Developmental Origin of Health and Disease (DOHaD) concept. Yet, the actual mechanisms accounting for mid- and long-term effects of early-life exposures remain unclear. Epigenetic alterations such as changes in DNA methylation, histone modifications and the expression of certain RNAs have been suggested as possible mediators of long-term health effects of environmental stressors. This report captures discussions and conclusions debated during the last Prenatal Programming and Toxicity meeting held in Japan. Its first aim is to propose a number of criteria that are critical to support the primary contribution of epigenetics in DOHaD and intergenerational transmission of environmental stressors effects. The main criteria are the full characterization of the stressors, the actual window of exposure, the target tissue and function, the specificity of the epigenetic changes and the biological plausibility of the linkage between those changes and health outcomes. The second aim is to discuss long-term effects of a number of stressors such as smoking, air pollution and endocrine disruptors in order to identify the arguments supporting the involvement of an epigenetic mechanism. Based on the developed criteria, missing evidence and suggestions for future research will be identified. The third aim is to critically analyze the evidence supporting the involvement of epigenetic mechanisms in intergenerational and transgenerational effects of environmental exposure and to particularly discuss the role of placenta and sperm. While the article is not a systematic review and is not meant to be exhaustive, it critically assesses the contribution of epigenetics in the long-term effects of environmental exposures as well as provides insight for future research. Abstract Bisphenol A (BPA) is a high-production-volume chemical with endocrine disrupting properties commonly used as color developer in thermal paper. Concerns about the potential hazards of human BPA exposure have led to the increasing utilization of alternatives such as bisphenol S (BPS) and bisphenol F (BPF). This study was designed to assess: (i) BPA, BPS, and BPF concentrations in 112 thermal paper receipts from Brazil, France, and Spain by liquid chromatography coupled to mass spectrometry (LC-MS); and (ii) hormone-like activities of these receipts using two receptor-specific bioassays, the E-Screen for (anti-)estrogenicity and PALM luciferase assay for (anti-)androgenicity. BPA was present in 95.3% of receipts from Spain, 90.9% of those from Brazil, and 51.1% of those from France at concentrations up to 20.27 mg/g of paper. Only two samples from Brazil, two from Spain, and ten from France had a BPS concentration ranging from 6.46 to 13.29 mg/g; no BPA or BPS was detected in 27.7% of French samples. No BPF was detected in any receipt. Estrogenic activity was observed in all samples from Brazil and Spain and in 74.5% of those from France. Anti-androgenic activity was observed in > 90% of samples from Brazil and Spain and in 53.2% of those from France. Only 25.5% of French samples were negative for both estrogenic and anti-androgenic activity. Estrogenic and anti-androgenic activities per gram of paper were up to 1.411 µM estradiol (E 2) equivalent units (E 2 eq) and up to 359.5 mM procymidone equivalent units (Proceq), respectively. BPA but not BPS concentrations were positively correlated with both estrogenic and anti-androgenic activities. BPA still dominates the thermal paper market in Brazil and Spain, and BPS appears to be one of the main alternatives in France. There is an urgent need to evaluate the safety of alternatives proposed to replace BPA as developer in thermal printing. The large proportion of samples with hormonal activity calls for the adoption of preventive measures. The aryl hydrocarbon receptor (AhR) is commonly described as a transcription factor, which regulates xenobiotic-metabolizing enzymes. Recent studies have suggested that the binding of ligands to the AhR also activates the Src kinase. In this manuscript, we show that the AhR, through the activation of Src, activates focal adhesion kinase (FAK) and promotes integrin clustering. These effects contribute to cell migration. Further, we show that the activation of the AhR increases the interaction of FAK with the metastatic marker, HEF1/NEDD9/CAS-L, and the expression of several integrins. Aryl hydrocarbon receptor (AhR), or dioxin receptor, is a transcription factor that induces adaptive metabolic pathways in response to environmental pollutants. Recently, other pathways were found to be altered by AhR and its ligands. Indeed, developmental defects elicited by AhR ligands suggest that additional cellular functions may be targeted by this receptor, including cell migration and plasticity. Here, we show that dioxin-mediated activation of Ahr induces Nedd9/Hef1/Cas-L, a member of the Cas protein family recently identified as a metastasis marker. The Hef1 gene induction is mediated by two xenobiotic responsive elements present in this gene promoter. Moreover, using RNA interference, we show that Nedd9/Hef1/Cas-L mediates the dioxin-elicited changes related to cell plasticity, including alterations of cellular adhesion and shape, cytoskeleton reorganization, and increased cell migration. Furthermore, we show that both E-cadherin repression and Jun N-terminal kinases activation by dioxin and AhR also depend on the expression of Nedd9/Hef1/Cas-L. Our study unveils, for the first time, a link between pollutants exposure and the induced expression of a metastasis marker and shows that cellular migration and plasticity markers are regulated by AhR and its toxic ligands. Environmental chemicals such as dioxin adversely affect immune, neurological and reproductive functions and have been implicated in cancer development. However, the mechanisms responsible for dioxin toxicity are still poorly understood. Here, we show that dioxin and related pollutants trigger a marked morphological change in epithelial cells that remodel their cytoskeleton to increase interaction with extra cellular matrix while loosening cell–cell contacts. Furthermore, dioxin-treated cells show increased motility. These dioxin-mediated effects are mimicked by constitutive expression and activation of the intracellular dioxin receptor (aryl hydrocarbon receptor (AhR)). They correlate with activation of the Jun NH2-terminal kinase (JNK) and are reverted by treatment with a JNK inhibitor. Dioxin-induced effects occur 48 h post-treatment initiation, a time scale, which argues for a genomic effect of the AhR, linked to induction of target genes. Introduction et but de l’étude Une exposition à la citrulline (CIT) pendant 24 h d’explants de tissu adipeux blanc intra-abdominal (WAT) issus de rats en surpoids par un régime hyperlipidique (HFD) provoque la libération des acides gras et stimule la bêta oxydation [1] . Des mécanismes de « brunisation » du WAT, augmentant la thermogenèse sans frisson et la dépense énergétique ont été décrits. Celle-ci implique l’induction de la protéine découplante (UCP1), longtemps considérée comme spécifique du tissu adipeux brun. Nous avons étudié l’effet potentiel de la CIT sur l’induction de l’UCP1 dans des explants de WAT rétropéritonéal (RET) et péri-épididymaire (EPI). Matériel et méthodes Les explants de WAT issus de rats Sprague-Dawley de quatre mois, ayant été maintenus pendant deux mois soit sous régime standard (CD, n = 7), soit HFD ( n = 7), ont été exposés à 2,5 mmoles/L de citrulline pendant 8 h ou 24 h. Les ARN totaux et les protéines ont été préparés. L’expression d’UCP1 et celle de son ARNm ont été étudiées par western blot et par RT-qPCR. Les niveaux d’expression des ARNm des deux principaux régulateurs de l’expression du gène UCP1 , le récepteur activé par les proliférateurs de péroxysomes alpha (PPARα) et le PPARγ-coactivateur-1-α (PGC-1α), ont aussi été analysés par RT-qPCR. Résultats et analyse statistique Dans les explants des tissus RET et EPI de rats HFD, l’expression de l’ARNm UCP1 et celle de sa protéine sont diminuées respectivement de 45–55 % et de 40–50 % ( p < 0,01) par rapport à celle des explants CD [2] . À la suite de l’exposition des explants des deux tissus à la CIT pendant 24 h, l’expression de l’ARNm UCP1 et celle de la protéine UCP1 sont augmentées respectivement de 3,5–6 fois ( p < 0,01) et de 1,5–2 fois ( p < 0,05), quel que soit le régime initial [2] . L’expression des ARNm PPARα et PGC-1α est aussi induite par la CIT dans les explants des deux tissus respectivement de 3,2–6 fois et 2,2–5,5 fois ( p < 0,01), quel que soit le régime initial. Conclusion La CIT induit très rapidement l’expression d’UCP1 et de deux des principaux régulateurs de l’expression de son gène, dans des explants de WAT. La stimulation de l’expression d’UCP1 sous l’effet de CIT, conduirait à la « brunisation » du WAT comme médiatrice de dépense énergétique permettant de lutter contre le surpoids induit par un régime riche en lipides. Introduction et but de l’étude Les amines aromatiques (AA) sont une classe importante de cancérigènes, caractérisées comme contaminants alimentaires. Ils sont notamment produits involontairement au cours de combustions (cuissons…) en parallèle d’autres composés toxiques comme le benzo(a)pyrène (BaP). Leurs effets biologiques (cancérogenèse) dépendent de leur transformation en métabolites réactifs. Ils peuvent toutefois être biotransformés en dérivés N-acétylés par les N-acétyl transférases. Ces derniers métabolites sont généralement considérés comme moins toxiques. Quelques études montrent que certaines AA semblent activer la voie du récepteur Ah (ou AhR, un récepteur de polluants et un activateur transcriptionnel d’enzymes du métabolisme des polluants, permettant l’élimination de ceux-ci). Le but de notre étude est de caractériser l’effet biologique de plusieurs AA et de leurs dérivés acétylés sur la voie du AhR, en conjugaison avec un ligand classique de ce récepteur, le BaP, un co-contaminant des AA. Matériel et méthodes Trois lignées cellulaires sont utilisées au cours de cette étude. La lignée d’hépatocarcinome humaine HepG2, la lignée HepG2 modifiée de manière stable (CYP1A1-XRE-Luc) exprimant une luciférase secrétée sous la dépendance d’un promoteur activé par le AhR et la lignée d’épithélium pulmonaire humaine NCI-H292. Trois couples d’amines sont utilisés : 2-Aminofluorène acétylé ou non (2-AF/2-AFA), 2-Naphtylamine acétylée ou non (2-NA/2-NAA) et 4-aminobiphenyl acétylé ou non (4-ABP/4-ABPA). L’ensemble des cellules ont été traitées à différents temps (0–48 h) et avec différentes doses d’AA (0–25 μ M) et/ou de BaP (5 μ M). Les paramètres suivants ont été mesurés : viabilité cellulaire (test WST-1), activité de la voie AhR (mesure de l’activité lucif érase de la lignée HepG2 CYP1A1-XRE-Luc), expression des gènes cibles du AhR (mesure de l’expression de l’ARN messager du CYP1A1), activité de l’enzyme CYP1A1 (mesure de l’activité EROD pour Ethoxyre-sorufin-O-Deethylase). Résultats et Analyse statistique Nous mettons en évidence une activation de la voie AhR par certaines AA mais de manière surprenante, une modulation de cette activation par la N-acétylation. En effet, alors que le couple 4-ABP/4-ABPA est inactif, le 2-NA et le 2-AFA sont des activateurs puissants de la voie alors que le 2-NAA et le 2-AF sont beaucoup moins actifs voire inactifs. En présence du BaP, la voie AhR est activée; les co-traitements avec les AA et leurs métabolites, conduisent à des synergies ou des antagonismes, également en fonction du statut d’acétylation de l’AA 1 . Conclusion Nos travaux montrent pour la première fois que la métabolisation des AA conduit à la production ou à l’inactivation de ligands du AhR. L’activation de la voie AhR conduit à l’expression de nombreuses enzymes du métabolisme des xénobiotiques (dont le CYP1A1) qui impactent le métabolisme des AA, conduisant dans certains cas à la production de dérivés plus mutagènes (AA O-acé-tylés). Nos travaux ont donc un intérêt en santé publique à la fois par l’identification de nouveaux ligands du AhR et par la perspective de travaux permettant de mieux comprendre en cancérogenèse, l’effet des AA et des cocktails les contenant (par exemple avec le BaP également m&#2 [ABSTRACT FROM AUTHOR]
