The current investigation assessed the effectiveness of 30 citrus germplasms in combating citrus canker pathogenesis in Assam, North‐East India. Significant differences were observed among the examined citrus germplasms in terms of per cent disease severity and disease reduction against Xanthomonas citri pv. citri. Among the 30 different germplasms, eight germplasms, namely, Jora tenga (14.67%), Bonjora (15.58%), Holong tenga (15.75%), Latipes (17.34%), Patijora (17.75%), Birajora (18.50%), Cleopatra mandarin (20.00%) and Ada jamir (20.18%), exhibited highest disease resistance potential. The study recorded that the germplasm Latipes, which are resistant, had the highest total phenol content (21.26 mg/g dry weight), while the susceptible one, Grapefruit (7.85 mg/g dry weight), had the lowest total phenol content. Jora tenga exhibited the highest flavonoid content, measuring at 6.48 mg/g dry weight, whereas White pummelo had the lowest flavonoid content, measuring at 1.88 mg/g dry weight. Similarly, the highest alkaloid percentage was recorded in resistant germplasm, Adajamir (10.52%), while the lowest was in susceptible germplasm, Volkameriana (2.30%). The levels of total phenol, alkaloid and flavonoid contents exhibited a significant negative correlation with the per cent disease severity of citrus canker disease in both Pearson correlation and Spearmen correlation analysis. The current research on bioprospecting of citrus germplasms in relation to citrus canker pathogenesis has the potential to offer valuable insights into mitigating the transmission of bacterial pathogens and eliminating sources of infection by employing resistant cultivars and implementing approved doses of plant protection formulations. Phytophthora gummosis, foot rot, and root rot are considered major challenges to the citrus industry worldwide. Little is known about the Phytophthora–microbiome interaction, despite several studies demonstrating changes in the microbial composition of the rhizosphere following challenges by a pathogen. In the present study, we studied the microbial diversity and community structure in healthy rhizospheres and Phytophthora-infected rhizospheres of Khasi mandarin (Citrus reticulata Blanco), a commercial cultivar extensively grown in the northeast of India. An exploratory study was conducted to identify Phytophthora-infected orchards of Khasi mandarin, and the isolated pathogen was confirmed as P. nicotianae based on its morpho-cultural and molecular characteristics coupled with pathogenicity tests. This study on culturable microbes established the dominance of Trichoderma spp. in the healthy rhizosphere, while the diseased rhizosphere showed the presence of Fusarium spp. A metagenomic study further revealed that the rhizospheres of Phytophthora-infected plants were dominated by species such as Bacteroidia spp., Patescibacteria spp., and Pythium spp., while the healthy Khasi mandarin rhizospheres had a more diverse community predominantly represented by Trichoderma, Penicillium, Linnemannia, Mortierella, Talaromyces, Saitozyma, Bacteroidetes, Pseudomonas, Cytophagia, Cyanobacteria, Bacteroidia, Sphingobacteriia, Burkholderia, Bacillus, and Bradyrhizobium. Terrabacteria and FCB (Fibrobacterota, Chlorobiota, and Bacteroidota groups) were found to exist in higher relative abundance in disease-free soils than in Phytophthora-infected soils, while phylum Proteobacteria showed identical relative abundance in all soil types. The phyla represented by Pseudomonas, Flavobacteriia, Candidatus, Mycobacterium, Rhizobium, Mesorhizobium, Sphingomonas, and Cytophagia were the most common bacterial phyla in all soil samples, but healthy soil exhibited a greater abundance of Bacteroidetes, Pseudomonas, Cytophagia, Cyanobacteria, Bacteroidia, Sphingobacteriia, Burkholderia, Bacillus, and Bradyrhizobium. Our study suggests that the presence of Phytophthora spp. in the rhizosphere alters microbial community structure, having potentially strong implications for plant health and productivity. These rhizosphere microbiome-derived citrus responses shed light on exploring effective management strategies for Phytophthora gummosis disease ailing Khasi mandarin. [ABSTRACT FROM AUTHOR]

An innovative tissue culture mediated incorporation of metabolite-based biomolecule (Bio-immune) at in vitro stage itself in banana cv. Grand Naine was developed and validated for the production of Fusarium oxysporum f.sp. cubense TR4 tolerant plantlets. The novel bio-immune formulation developed by us, exhibited a significant antifungal potency against Foc TR4 with a high percent inhibition (100%) at a 2.5% concentration of bio-immune on the 5th, 7th, and 9th DAI. Bio-immune integrated during in vitro shoot proliferation stage in banana cv. Grand Naine recorded significant enhancement in the growth of roots and shoots. Bio-immune (0.5%) fortified media produced 12.67 shoots per clump whereas control registered only 9.67 shoots per clump. Similarly, maximum root numbers (7.67) were observed in bio-immune plants which were significantly higher over control (5.0). The bio-immunized banana transplants recorded a higher survival rate (97.57%) during acclimatization as compared to the control (94.53%). Furthermore, evaluation of the bio-immunized plants in pot experiments revealed that unimmunized plants treated with FocTR4 (TF) exhibited mortality between 60 and 90  days. On the 90th day after planting, a high mean disease severity index (DSI) of 3.45 was observed with unimmunized plantlets while the bio-immunized plants (TFBI) and ICAR-FUSICONT treated plants (TFTR) showed substantially reduced DSI (0.20 and 1.00) compared to FocTR4 treated control (TF). Significant increases in polyphenol oxidase (PPO), peroxidase (POD), β-1,3-glucanase, phenylalanine ammonia-lyase (PAL), chitinase activities, and enhanced phenol contents were recorded in bio-immunized plants compared to unimmunized plants. Field experiments at two different locations in Bihar, India revealed that bunch weight, no. of hands/bunch, and no. of fingers/hand of bioimmune treated plants were significantly higher compared to the control. In-vitro studies were conducted to check the compatibility of microbial bioagent, Trichoderma viride with seven fungicides, eigtht insecticides, two herbicides, three fertilizers, and two antibiotics at different concentrations. The bioagent was compatible with insecticides viz., imidacloprid 27.8% SL, diafenthiuron 50% WP, chlorantraniliprole 18.5% SC, flubendiamide 20% WG, azoxystrobin 23% SC, deltamethrin 2.8% EC, clothianidin 50% WDG, fenpropathrin 10% EC, and tiamethoxam 25% WG at all the four concentrations. However, fungicides hexaconazole 5% EC, propiconazole 25% EC, tebuconazole 25.9 % EC, and tebuconazole 50% + trifloxistrobin 25% WG and copper oxychloride 50% WP showed incompatibility with T. viride. Among the herbicides, glyphosate 41% SL was compatible, while paraquat dichloride 24% SL showed an incompatible reaction. Urea and muriate of potash (MOP) showed varied compatibility at different concentrations, while single super phosphate (SSP) showed a compatible reaction with the microbial bioagent. The results also indicate that antibiotic like streptomycin and gentamycin are compatible with T. viride. Knowledge of the compatibility of the microbes and the agrochemicals will be useful to apply biopesticide formulations in combination with inorganic pesticides, targeting different pests to reduce the cost and time. Arbuscular mycorrhizal fungi (AMF) enhance plant disease resistance, while the underlying mechanisms in the molecular levels are not yet known. In this study, five-leaf-old trifoliate orange seedlings were inoculated with Funneliformis mosseae for 14 weeks and subsequently were infected by a citrus root rot pathogen Phytophthora parasitica by 7 days. The transcriptome results by Illumina HiSeq 4000 revealed that the percentage of Q30 bases reached 92.99% or above, and 29696 unigenes were annotated in a total of 63531 unigenes. 654 and 103 differentially expressed genes (DEGs) were respectively annotated in AMF-inoculated versus non-AMF-inoculated plants under non-infection and infection with P. parasitica, respectively, whilst these DEGs were related to defense mechanisms, signal transduction mechanisms and secondary metabolites biosynthesis. Forty-two genes were functionally annotated as the putative 'defense mechanism', whilst AMF inoculation induced 1 gene down-regulated and 3 genes up-regulated under P. parasitica infection. AMF inoculation stimulated more genes linked to signal transduction mechanism down-regulated than non-AMF plants. Eight genes were involved in secondary metabolites biosynthesis in AMF versus non-AMF seedlings under P. parasitica-infection conditions. Such transcriptome database provided total information in the molecular levels regarding mycorrhizal roles in tolerating Phytophthora parasitica infection. [ABSTRACT FROM AUTHOR]
