Oxygen deprivation and excess are both toxic. Thus, the bodys ability to adapt to varying oxygen tensions is critical for survival. While the hypoxia transcriptional response has been well studied, the post-translational effects of oxygen have been underexplored. In this study, we systematically investigate protein turnover rates in mouse heart, lung, and brain under different inhaled oxygen tensions. We find that the lung proteome is the most responsive to varying oxygen tensions. In particular, several extracellular matrix (ECM) proteins are stabilized in the lung under both hypoxia and hyperoxia. Furthermore, we show that complex 1 of the electron transport chain is destabilized in hyperoxia, in accordance with the exacerbation of associated disease models by hyperoxia and rescue by hypoxia. Moreover, we nominate MYBBP1A as a hyperoxia transcriptional regulator, particularly in the context of rRNA homeostasis. Overall, our study highlights the importance of varying oxygen tensions on protein turnover rates and identifies tissue-specific mediators of oxygen-dependent responses.

Aging is the single largest risk factor for most chronic diseases, and thus possesses large socioeconomic interest to continuously aging societies. Consequently, the field of aging research is expanding alongside a growing focus from the industry and investors in aging research. This year's 8th Annual Aging Research and Drug Discovery (ARDD) meeting was organized as a hybrid meeting from August 30th to September 3rd 2021 with more than 130 attendees participating on-site at the Ceremonial Hall at University of Copenhagen, Denmark, and 1800 engaging online. The conference comprised of presentations from 75 speakers focusing on new research in topics including mechanisms of aging and how these can be modulated as well as the use of AI and new standards of practices within aging research. This year, a longevity workshop was included to build stronger connections with the clinical community.

Ribosomal DNA (rDNA) genes encode the structural RNAs of the ribosome and are present in hundreds of copies in mammalian genomes. Age-linked DNA hypermethylation throughout the rDNA constitutes a robust "methylation clock" that accurately reports age, yet the consequences of hypermethylation on rDNA function are unknown. We confirmed that pervasive hypermethylation of rDNA occurs during mammalian aging and senescence while rDNA copy number remains stable. We found that DNA methylation is exclusively found on the promoters and gene bodies of inactive rDNA. To model the effects of age-linked methylation on rDNA function, we directed de novo DNA methylation to the rDNA promoter or gene body with a nuclease-dead Cas9 (dCas9)-DNA methyltransferase fusion enzyme in human cells. Hypermethylation at each target site had no detectable effect on rRNA transcription, nucleolar morphology, or cellular growth rate. Instead, human UBF and Pol I remain bound to rDNA promoters in the presence of increased DNA methylation. These data suggest that promoter methylation is not sufficient to impair transcription of the human rDNA and imply that the human rDNA transcription machinery may be resilient to age-linked rDNA hypermethylation.

Abstract The lifespans of proteins range from minutes to years within mammalian tissues. Protein lifespan is relevant to organismal aging, as long‐lived proteins accrue damage over time. It is unclear how protein lifetime is shaped by tissue context, where both cell turnover and proteolytic degradation contribute to protein turnover. We develop turnover and replication analysis by 15N isotope labeling (TRAIL) to quantify protein and cell lifetimes with high precision and demonstrate that cell turnover, sequence‐encoded features, and environmental factors modulate protein lifespan across tissues. Cell and protein turnover flux are comparable in proliferative tissues, while protein turnover outpaces cell turnover in slowly proliferative tissues. Physicochemical features such as hydrophobicity, charge, and disorder influence protein turnover in slowly proliferative tissues, but protein turnover is much less sequence‐selective in highly proliferative tissues. Protein lifetimes vary nonrandomly across tissues after correcting for cell turnover. Multiprotein complexes such as the ribosome have consistent lifetimes across tissues, while mitochondria, peroxisomes, and lipid droplets have variable lifetimes. TRAIL can be used to explore how environment, aging, and disease affect tissue homeostasis.

Freshwater salinity varies in natural systems and plays a role in species distribution. Anthropogenic alterations to freshwater salinity regimes include sea level rise and subsequent intrusion of saline waters to inland habitats. While mayflies are generalized to be sensitive to increasing salinity, we still know remarkably little about the physiological processes (and their plasticity) that determine the performance of species in a changing world. Here, we explored life-history outcomes and physiological plasticity in a population of Callibaetis floridanus (Ephemeroptera: Baetidae) from a coastal pond that routinely experiences saltwater intrusion. We reared naiads from egg hatch to adulthood across a gradient of increasing salinities (113, 5,020, 9,921 μS/cm). Radiotracer flux studies (22Na, 35SO4, and 45Ca) were conducted in naiads reared at each salinity, revealing a positive association between ionic concentration and uptake rates. However, the influence of rearing history on ionic influx rates was apparent when naiads were transferred from their respective rearing water to the other experimental conditions. For example, we observed that naiads reared in the low salinity treatment (113 μS/cm) had 10.8-fold higher Na uptake rates than naiads reared at 9,921 μS/cm and transferred to 113 μS/cm. Additionally, naiads acclimated to the higher salinity water exhibited reduced uptake in ion-rich water relative to those reared in more dilute conditions (e.g., in 9,921 μS/cm water, 113 and 5,020 μS/cm acclimated naiads had 1.5- and 1.1-fold higher Na uptake rates than 9,921 μS/cm acclimated naiads, respectively). We found no significant changes in survival (80 ± 4.4%, mean ± s.e.m.) or naiad development time (24 ± 0.3 days, mean ± s.e.m.) across these treatments but did observe a 27% decrease in subimago female body weight in the most dilute condition. This reduction in female weight was associated with higher oxygen consumption rates in naiads relative to the other rearing conditions. Collectively, these data suggests that saline adapted C. floridanus may be more energetically challenged in dilute conditions, which differs from previous observations in other mayfly species.

Aquatic insects typically dominate the ecology of freshwater ecosystems while relatively few species are found in saline waters. As human activities are changing the salinity regimes of freshwater ecosystems--typically by making them more saline, ecologists are reporting declines in insect diversity. These observations imply that insect physiological systems play a major role in how species respond to salinity change. Abstract Background Our previous studies showed that the expression of the monocyte-chemoattractant protein (MCP)-1, a chemokine, which triggers the infiltration and activation of cells of the monocyte-macrophage lineage, is abrogated in human papillomavirus (HPV)-positive premalignant and malignant cells. In silico analysis of the MCP-1 upstream region proposed a putative p53 binding side about 2.5 kb upstream of the transcriptional start. The aim of this study is to monitor a physiological role of p53 in this process. Results The proposed p53 binding side could be confirmed in vitro by electrophoretic-mobility-shift assays and in vivo by chromatin immunoprecipitation. Moreover, the availability of p53 is apparently important for chemokine regulation, since TNF-α can induce MCP-1 only in human keratinocytes expressing the viral oncoprotein E7, but not in HPV16 E6 positive cells, where p53 becomes degraded. A general physiological role of p53 in MCP-1 regulation was further substantiated in HPV-negative cells harboring a temperature-sensitive mutant of p53 and in Li-Fraumeni cells, carrying a germ-line mutation of p53. In both cases, non-functional p53 leads to diminished MCP-1 transcription upon TNF-α treatment. In addition, siRNA directed against p53 decreased MCP-1 transcription after TNF-α addition, directly confirming a crosstalk between p53 and MCP-1. Conclusion These data support the concept that p53 inactivation during carcinogenesis also affects immune surveillance by interfering with chemokine expression and in turn communication with cells of the immunological compartment.

The syntheses and crystal structures of 2,3-bis[3-(trifluoromethyl)phenyl]-2,3-dihydro-4H-1,3-benzothiazin-4-one (di-m-CF3; C22H13F6NOS) (1) and 2,3-bis(4-methylphenyl)-2,3-dihydro-4H-1,3-benzothiazin-4-one (di-p-CH3; C22H19NOS) (2) are reported. Each structure is racemic: the asymmetric unit of 2 consists of two molecules. In both 1 and 2, the six-membered 1,3-thiazine ring is close to an envelope conformation with the chiral C atom forming the flap. In 1, the 2-aryl group is pseudo-equatorial, while in 2 it is pseudo-axial. In 1, the pendant aryl rings form a V shape with an inter-centroid distance of 3.938 (3) Å and an acute dihedral angle of 48.3 (2)° between them. Both CF3 groups are disordered over two orientations in 0.687 (19):0.313 (19) and 0.667 (16):0.33 (16) ratios. In each of the independent molecules of 2, the aryl rings are almost orthogonal to each other [dihedral angles = 85.50 (12) and 86.07 (11)°]. In both structures, the chiral C atom and the O atoms participate in C—H...O-type hydrogen bonding between symmetry-related molecules of 1 or the independent enantiomers in 2, forming chains along the c-axis direction in 1 and the b-axis direction in 2. Additionally, in 1, π–π contacts of both face-to-face and edge-to-face type, as well as π-H...O and π-H...F interactions are observed. In 2, a racemic mixture of molecules forms layers in the ac plane linked by weak π–π and C—H...π interactions.
