A group of 5-(2-chlorophenyl)-10-(substituted)-7H-pyrido[4,3-f][1,2,4]triazolo[4,3-a][1,4]diazepines 7a-c were synthesized by the acid catalyzed reaction of 5-(2-chlorophenyl)-2-hydrazino-3H-pyrido[3,4-e]-[1,4]diazepine (6) with either trimethyl orthoformate, triethyl orthoacetate or triethyl orthobenzoate, respectively. 5-(2-Chlorophenyl)-7H-pyrido[4,3-f][1,2,4]triazolo[4,3-a][1,4]diazepine (7a) and 5-(2-chlo-rophenyl)-10-methyl-7H-pyrido[4,3-f][1,2,4]triazolo[4,3-a][1,4]diazepine (7b) exhibited good anticonvulsant activity in the subcutaneous metrazol anticonvulsant screen which serves as a model for absence (petit mal) epilepsy.

The homolytic benzoylation (benzoyl radical) of 5-tert-butylcarbonylaminopyrimidine (6, 1 equiv.) in the presence of benzaldehyde (3 equiv.), water, sulfuric acid, and acetic acid, upon treatment with FeSO4·7H2O (3 equiv.) and 70% t-BuOOH (3 equiv.) at 5-10°C for 10 min afforded a mixture of 4-benzoyl-5-tert-butylcarbonylaminopyrimidine (7, 23%), 4,6-dibenzoyl-5-tert-butylcarbonylaminopyrimidine (8, 44%), and 4-benzoyl-5-tert-butylcarbonylamino-6- methyl pyrimidine (9, 10%). When a similar reaction was performed using 1 equiv. each of PhCHO, FeSO4·7H2O, and t-BuOOH, to prevent formation of the 4,6-dibenzoyl product 8, the monobenzoyl (7, 38%) and 4-benzoyl-6-methyl (9, 6%) products were obtained. A similar homolytic benzoylation of 5-bromopyrimidine (10) using 1.5 equiv. of reagents to generate the benzoyl radical afforded 4-benzoyl-5-bromopyrimidine (11, 61%) as the predominant product. Elaboration of 11 via a six-step reaction sequence afforded 2-hydrazino-5-phenyl-3H-pyrimido[5,4-e][1,4]diazepine (17) in 5.2% overall yield. The acid-catalyzed reaction of 17 with triethyl orthoacetate gave the title compound 5-phenyl-10-methyl-7H-pyrimido[4,5-f][1,2,4]triazolo[4,3-a][1,4]diazepine (18, 62%). The triazolo compound 18 was more potent than valproic acid (Depakene(r)) in both the subcutaneous metrazol (scMet) and maximal electroshock (MES) anticonvulsant screens, and more potent than clonazepam in the MES anticonvulsant screen but less potent than clonazepam in the scMet anticonvulsant screen.Key words: homolytic benzoylation, pyrimidines, pyrimidodiazepines, anticonvulsants.

A series of 7α-methoxy-2-[(substituted)methylene]cephem sulfones was synthesized. The compounds with an acetoxy group at the C-3′ position as a leaving group were found to be potent inhibitors of HLE. Selected compounds are also found to be antithrombin agents.

A new series of 2-spiro(2′,2′-diphenylcyclopropane) cephalosporin sulfones was synthesizes as potent human leukocyte elastase inhibitors.

A series of 2 beta-[(4-substituted)-1,2,3-triazol-1-yl] methyl penicillanic acid sulfones was synthesized as beta-lactamase inhibitors. Many of these compounds showed good in vitro inhibitory activity against penicillinase, cefotaximase and plasmid-mediated class III TEM enzymes, but exhibited weaker cephalosporinase inhibition. One member in this series--2 beta-[(4-pyridiniummethyl)-1,2,3-triazol- 1-yl]-6,6-dihydropenicillanate 1,1-dioxide (12a), when tested in combination with piperacillin, showed excellent synergistic activity against microorganisms producing plasmid-mediated enzymes, but had insufficient activity against microorganisms producing chromosomally mediated class I enzymes.

A series of 2α-alkoxymethyl cephem sulfones were prepared by nucleophilic addition of a variety of alcohols to exo-2-methylene cephem sulfones. The 2α-alkoxymethyl group was introduced with the aim of improving the inhibitory activity against human leukocyte elastase (HLE) over the unsubstituted compounds. However, against HLE the in vitro activity was still inferior to that shown by the C-2 unsubstituted cephem sulfones.

The regiospecific reaction of 3-benzyloxycarbonylaminomethylcarbonylamino-4-benzoylpyridine (6a), or 3-t-butoxycarbonylaminomethylcarbonylamino-4-benzoylpyridine (6b), with either acetyl chloride or ethyl chloroformate, and either n-butylmagnesium chloride or phenylmagnesium bromide afforded the respective 1-acetyl (or ethoxycarbonyl)-2-n-butyl (or phenyl)-3-benzyloxy (or t-butoxy) carbonylaminomethylcarbonylami-no-4-benzoyl-1,2-dihydropyridines 7 in 60-75% yield. Reaction of 1-acetyl (or ethoxycarbonyl)-2-n-butyl (or phenyl)-3-t-butoxycarbonylaminomethylcarbonyl-4-benzoyl-1,2-dihydropyridines 7b, 7f, 7d, 7h with trifluoroacetic acid gave the corresponding 5-phenyl-8-acetyl (or ethoxycarbonyl)-9-n-butyl (or phenyl)-1,3,8,9-tetrahydro-2H-pyrido[3,4-e]-1,4-diazepin-2-ones 8a, 8b, 8c, 8d respectively in 45–63% yield. N1-Methylation of 5-phenyl-8-acetyl-9-n-butyl (or phenyl)-1,3,8,9-tetrahydro-2H-pyrido[3,4-e]-1,4-diazepin-2-ones 8a, 8b using sodium hydride and iodomethane yielded the corresponding N1-methyl derivatives 9a (48%) and 9b (54%). Oxidation of 5,9-diphenyl-8-ethoxycarbonyl-1,3,8,9-tetrahydro-2H-pyrido[3,4-e]-1,4-diazepin-2-one (8d) using p-chloranil afforded 1,3-dihydro-5,9-diphenyl-2H-pyrido[3,4-e]-1,4-diazepin-2-one (10). 5-Phenyl-8-acetyl-9-n-butyl-1,3,8,9-tetrahydro-2H-pyrido[3,4-e]-1,4-diazepin-2-one (8a) and the corresponding 8-ethoxycarbonyl analog 8c exhibited weak anticonvulsant activity indicating that 8a and 8c may be acting at the same site as the 7-halo-1,4-benzodiazepin-2-one class of compounds.

The ortho-directed lithiation of 3-tert-butyl- or 3-tert-butoxycarbonylaminopyridines (3) with alkyllithiums and reaction with N,N-diethylbenzamide followed by acid hydrolysis gave 3-amino-4-benzoylpyridine (6) in good yield. Reaction of BTBO with the glycine derivatives 7a, b and then reaction with 6 afforded 3-alkoxycarbonylaminomethylcarbonylamino-4-benzoylpyridines 8a, b. Acid-catalyzed hydrolysis and cyclization of 8a, b yielded 9, which on methylation gave 1,3-dihydro-1-methyl-5-phenyl-2H-pyrido[3,4-e]-1,4-diazepin-2-one (10) in 36% overall yield from 3a.
