Glacial/interglacial transitions in mountainous areas are marked by significant glacier retreat from forelands to inner massifs, resulting in large-scale and ephemeral lake formation that are subsequently filled (or not) by sediment transfer during lateglacial to postglacial times. When valley paleo-infills are preserved, they form precious archives to investigate (1) Alpine erosion dynamics and paleo-environmental conditions during key transition periods from full glacial stages to interglacials, and (2) glacial erosion patterns during susbequent glaciation.In this contribution, we investigate such sedimentary deposits (locally called as "banquettes") in the French western Alps, and more precisely along the Isère valley and Val du Bourget. Previous research have attributed these deposits to the Riss – Würm transition due to their position under a basal compact till and to the MIS 6/5 transition up to early MIS 4 from palynological constraints, although no absolute ages has been available so far. Based on existing mapping of their spatial distribution and stratigraphic reconstructions, we sampled coarse-sand and sandy-gravel layers within these deposits for constraining both sediment deposition time (OSL dating) and provenance (glacial/postglacial origin, using terrestrial cosmogenic nuclide TCN 10Be in quartz). In addition, their spatial distribution provides estimates of maximum glacial erosion during the last glacial cycle, which can be subsequently used as spatial constraints for ice model predictions.Our results confirm deposition times of these sedimentary units at the MIS 6/5 transition, with dating constraints from the late MIS 6 (ca. 145 ka) to the early MIS 5 (Eemien, 115-130 ka) for sandy layers. Upper sandy-gravel layers have younger deposition ages of ca. 80 ka, illustrating sediment fluxes at the transition from late MIS 5 to early MIS 4. We compare this temporal sequence to more recent sediment infills of the Isère valley (14C and OSL dating) during the Lateglacial to Holocene (MIS 2/1) transition. TCN data from sand samples also illustrate the sharp transition from full glacial to interglacial conditions, with a significant increase in 10Be concentrations from Lateglacial to post-glacial sediments. We propose that the observed signal can reflect changes in erosion rates, but also in glacier expansion or in paleo-environmental conditions, with export of stored subglacial sediments as well as the re-establishment of sediment/soil production and transfer along the catchment routing system following glacier retreat.

International audience; This study tests the application of combined detrital apatite fission track (AFT) and U-Pb dating to infer both glacial erosion spatial patterns and long-term rock cooling histories in Alpine mountainous settings. We have dated 716 detrital apatite grains from glacial sediments collected in the Maurienne and Arve valleys (Western European Alps, France) from moraine deposits corresponding to different stages of glacial retreat since the Last Glacial Maximum (LGM, ca. 24-21 ka).The Maurienne valley crosses the internal and external Alps, which exhibit contrasting in-situ AFT and U-Pb ages. Here, we present the measured distribution of both detrital AFT and U-Pb ages at 6 locations along the valley, with catchment elevations ranging from 390 to 1740 m. We show that during glacial retreat, erosion is mainly concentrated in the downstream part of the glacier, near the sampled moraine deposits. This inference suggests that during glacial retreat, glacial erosion is more effective below the ELA (Equilibrium Line Altitude) and specifically close to the glacier front, in areas where ice flow velocity is high and subglacial water is abundant, as predicted by ice-dynamics reconstructions in the European Alps over the last 20 ka.In the Arve valley, previous studies showed that in situ AFT ages are systematically younger than 7 Ma for the Mont-Blanc massif. We compare the thermal history obtained from these literature bedrock-derived data to that derived from the new detrital AFT data collected in the Little Ice Age (LIA) moraine, just at the front of the Bossons glacier (~1300 m elevation). We also compare our results with 5 other samples down the valley at catchment elevations between 460 and 1050 m to evaluate potential changes in the detrital AFT signal as well as the consistency in the retrieved long-term cooling history.Based on these first results, we plan to extend our study to other areas (e.g., Patagonia) to investigate both (1) spatial patterns of glacial erosion for older glacial periods (pre-LGM), and (2) long-term rock cooling histories from moraine deposits where modern bedrock is inaccessible (e.g. under modern glaciers or ice fields).

Paleoglacier reconstructions in the northern and southern forelands of the European Alps indicate a synchronous Late Pleistocene glacial maximum during Marine Isotope Stage (MIS) 2, in phase with global ice volume records. However, strong controversy remains for the western foreland, where scarce and indirect dating as well as modeling studies suggest glacial maxima out of phase with the rest of the Alps. New luminescence dating brings the first direct Late Pleistocene glacial chronology for the western Alpine foreland and reveals two major glacier advances of similar maximum extent, at ca. 75–60 and ca. 40–30 ka, coinciding with MIS 4 and late MIS 3. We propose that asynchrony in glacial maxima between the western and the northern and southern Alpine forelands results from a progressive spatial reorganization of the atmospheric circulation over the North Atlantic in response to Northern Hemisphere ice-sheet fluctuations. While such a feedback mechanism has emerged from general circulation models, our Late Pleistocene paleoglacial reconstruction permits tracking of the spatiotemporal evolution of moisture advection patterns over Western Europe.

The microvasculature facilitates gas exchange, provides nutrients to cells, and regulates blood flow in response to stimuli. Vascular abnormalities are an indicator of pathology for various conditions, such as compromised vessel integrity in small vessel disease and angiogenesis in tumors. Traditional immunohistochemistry enables visualization of tissue cross-sections containing exogenously labeled vasculature. Although this approach can be utilized to quantify vascular changes within small fields-of-view, it is not a practical way to study the vasculature on the scale of whole organs. Three-dimensional (3D) imaging presents a more appropriate method to visualize the vascular architecture in tissue. Here we describe the complete protocol that we use to characterize the vasculature of different organs in mice encompassing the methods to fluorescently label vessels, optically clear tissue, collect 3D vascular images, and quantify these vascular images with a semi-automated approach. To validate the automated segmentation of vascular images, one user manually segmented fifty random regions of interest across different vascular images. The automated segmentation results had an average sensitivity of 80±8% and an average specificity of 90±5% when compared to manual segmentation. Applying this procedure of image analysis presents a method to reliably quantify and characterize vascular networks in a timely fashion. This procedure is also applicable to other methods of tissue clearing and vascular labels that generate 3D images of microvasculature.

Cardiac arrest (CA) afflicts ~ 550,000 people each year in the USA. A small fraction of CA sufferers survive with a majority of these survivors emerging in a comatose state. Many CA survivors suffer devastating global brain injury with some remaining indefinitely in a comatose state. The pathogenesis of global brain injury secondary to CA is complex. Mechanisms of CA-induced brain injury include ischemia, hypoxia, cytotoxicity, inflammation, and ultimately, irreversible neuronal damage. Due to this complexity, it is critical for clinicians to have access as early as possible to quantitative metrics for diagnosing injury severity, accurately predicting outcome, and informing patient care. Current recommendations involve using multiple modalities including clinical exam, electrophysiology, brain imaging, and molecular biomarkers. This multi-faceted approach is designed to improve prognostication to avoid “self-fulfilling” prophecy and early withdrawal of life-sustaining treatments. Incorporation of emerging dynamic monitoring tools such as diffuse optical technologies may provide improved diagnosis and early prognostication to better inform treatment. Currently, targeted temperature management (TTM) is the leading treatment, with the number of patients needed to treat being ~ 6 in order to improve outcome for one patient. Future avenues of treatment, which may potentially be combined with TTM, include pharmacotherapy, perfusion/oxygenation targets, and pre/postconditioning. In this review, we provide a bench to bedside approach to delineate the pathophysiology, prognostication methods, current targeted therapies, and future directions of research surrounding hypoxic–ischemic brain injury (HIBI) secondary to CA. Introduction: Cerebral microhemorrhages (CMH) are the pathologic substrate of MRI-demonstrable cerebral microbleeds, and are thought to occur when integrity of cerebral microvessels is compromised, leading to deposits of hemosiderin/iron accumulating within brain tissue. The presence of CMH is associated with cognitive decline and ischemic and hemorrhagic stroke. Hypertension (HTN) is a major risk factor for cerebral microbleeds and has been linked to a more significant microbleed burden in patients. Despite the clinical significance of HTN and cerebral microbleeds, our understanding of HTN-induced microbleed formation remains limited, and the vascular origin of CMH remains obscure. Methods: We used a perfusion-based vascular label with tissue clearing to enable three-dimensional visualization of CMH with the surrounding microvascular network. Adult (18-month-old) C57BL/6J mice were administered angiotensin II to induce HTN (n=10) or PBS (n=10). Lectin-DyLight-649 was injected retro-orbitally to bind to the endothelial walls of all blood vessels in a mouse. Brains were cut into 1-mm thick sections, stained with Prussian blue to label CMH, cleared using iDISCO, and imaged via confocal microscopy. MATLAB and neuTube were used to quantify vessel diameters. Results: Using this methodology, we were able to collect three-dimensional transmission images of CMH that were colocalized with three-dimensional fluorescence images of the surrounding microvasculature. The average vessel diameter of the 5 nearest vessels to 17 different CMH was found to be 4.22±0.81 μm, which corresponds to capillary-size vessels. Conclusions: These data support a capillary origin for cerebral microhemorrhages in this mouse model. The capillary origin implies a role for the blood-brain barrier in the development of CMH and cerebral microbleeds in this model of HTN and aging. Three-dimensional imaging has substantial utility for improved understanding of CMH origin.

International audience; Submerged fault ruptures generate earthquake-triggered mass flow deposits, which are extensively used as a tool in subaqueous paleoseismology. In tectonically active deep sedimentary basins, such as the Sea of Marmara (SoM), these mass flow deposits are defined as turbidite-homogenite units (THUs), consisting essentially of a coarse basal part and an overlying homogeneous mud (homogenite). Detailed characterization of THUs is crucial in order to establish meaningful criteria to link these units with earthquakes events and to identify their transport routes and depositional mechanisms. Here, we combine μ-X-ray Fluorescence (μ-XRF), Anisotropy of Magnetic Susceptibility (AMS) and additional rock magnetism analyses of a 21-m long piston core from the Kumburgaz Basin of SoM to define the upper stratigraphic boundary of THUs with hemipelagic sediments and investigate the controls of hydrological changes on turbidite frequency and thickness over the last 15 kyrs BP. The sedimentary succession of this period includes a lower lacustrine and an upper marine unit with two Holocene sapropel intervals. The sequence is interrupted by a total of 70 THUs, characterized by a significant magnetic foliation related to the depositional setting rather than the magnetic signature. Magnetic mineralogy of the coarse basal parts of THUs have more ferromagnetic particles than the overlying homogenites and background sediments. While the homogenite parts have a more constant mineralogy than the basal parts, they do not differ the background sediments.Based on an event-free chronostratigraphic model derived from radiocarbon ages and the published age of lacustrine-marine (L-M) transition, the average THU occurrence intervals in the lacustrine (14.8–12.6 kyrs BP), lower sapropel (11.2–5.7 kyrs BP), upper sapropel (5.4–2.7 kyrs BP) and non-sapropelic part of the marine unit (2.7 kyrs BP-present) are 235 yrs., 287 yrs., 114 yrs. and 160 yrs. respectively. The average thickness of the THUs in the same units are 20.8 cm, 15.7 cm, 6.1 cm and 6.1 cm. The variability of average THU occurrence intervals and THU thicknesses are controlled by the sea level rise and salinity increase following the full marine connection of the SoM at 12.6 kyrs BP, which caused changes in slope stability, sediment composition and sediment deposition in different parts of the basin. Geomechanical properties of the lower sapropel appears to have been important in resulting long THU recurrence intervals and relatively high THU thicknesses.

The prevalence of mild cognitive impairment increases with age and is further exacerbated by chronic kidney disease (CKD). CKD is associated with (1) mild cognitive impairment, (2) impaired endothelial function, (3) impaired blood-brain barrier, (4) increased cerebral microhemorrhage burden, (5) increased cerebral blood flow (CBF), (6) impaired cerebral autoregulation, (7) impaired cerebrovascular reactivity, and (8) increased arterial stiffness. We report preliminary findings from our group that demonstrate altered cerebrovascular reactivity in a mouse model of CKD-associated vascular calcification. The CBF of CKD mice increased more quickly in response to hypercapnia (p < 0.05) but then decreased prematurely during hypercapnia challenge (p < 0.05). Together, these results indicate that altered kidney function can lead to alterations in the cerebral microvasculature, and hence brain health.

Background Spreading depolarizations (SDs) are self-propagating waves of neuronal and glial depolarizations often seen in neurological conditions in both humans and animal models. Because SD is thought to worsen neurological injury, the role of SD in a variety of cerebral insults has garnered significant investigation. Anoxic SD is a type of SD that occurs because of anoxia or asphyxia. Although asphyxia leading to a severe drop in blood pressure may affect cerebral hemodynamics and is widely known to cause anoxic SD, the effect of anoxic SD on peripheral blood pressure in the extremities has not been investigated. This relationship is especially important to understand for conditions such as circulatory shock and cardiac arrest that directly affect both peripheral and cerebral perfusion in addition to producing anoxic SD in the brain. Methods In this study, we used a rat model of asphyxial cardiac arrest to investigate the role of anoxic SD on cerebral hemodynamics and metabolism, peripheral blood pressure, and the relationship between these variables in 8- to 12-week-old male rats. We incorporated a multimodal monitoring platform measuring cortical direct current simultaneously with optical imaging. Results We found that during anoxic SD, there is decoupling of peripheral blood pressure from cerebral blood flow and metabolism. We also observed that anoxic SD may modify cerebrovascular resistance. Furthermore, shorter time difference between anoxic SDs measured at different locations in the same rat was associated with better neurological outcome on the basis of the recovery of electrocorticography activity (bursting) immediately post resuscitation and the neurological deficit scale score 24 h post resuscitation. Conclusions To our knowledge, this is the first study to quantify the relationship between peripheral blood pressure, cerebral hemodynamics and metabolism, and neurological outcome in anoxic SD. These results indicate that the characteristics of SD may not be limited to cerebral hemodynamics and metabolism but rather may also encompass changes in peripheral blood flow, possibly through a brain–heart connection, providing new insights into the role of anoxic SD in global ischemia and recovery.

Heart rate variability (HRV) provides insight into cardiovascular health and autonomic function. Electrocardiography (ECG) provides gold standard HRV measurements but is inconvenient for continuous acquisition when monitored from the extremities. Optical techniques such as photoplethysmography (PPG), often found in health and wellness trackers for heart rate measurements, have been used to estimate HRV peripherally but decline in accuracy during increased physical stress. Speckleplethysmography (SPG) is a recently introduced optical technique that provides benefits over PPG, such as increased signal amplitude and reduced susceptibility to temperature-induced vasoconstriction. In this research, we compare SPG and PPG to ECG for estimation of HRV during an orthostatic challenge performed by 17 subjects. We find that SPG estimations of HRV are highly correlated to ECG HRV for both time and frequency domain parameters and provide increased accuracy over PPG estimations of HRV. The results suggest SPG measurements are a viable alternative for HRV estimation when ECG measurements are impractical.

Cerebral microhemorrhages (CMHs) are associated with cerebrovascular disease, cognitive impairment, and normal aging. One method to study CMHs is to analyze histological sections (5-40 μm) stained with Prussian blue. Currently, users manually and subjectively identify and quantify Prussian blue-stained regions of interest, which is prone to inter-individual variability and can lead to significant delays in data analysis. To improve this labor-intensive process, we developed and compared three digital pathology approaches to identify and quantify CMHs from Prussian blue-stained brain sections: 1) ratiometric analysis of RGB pixel values, 2) phasor analysis of RGB images, and 3) deep learning using a mask region-based convolutional neural network. We applied these approaches to a preclinical mouse model of inflammation-induced CMHs. One-hundred CMHs were imaged using a 20x objective and RGB color camera. To determine the ground truth, four users independently annotated Prussian blue-labeled CMHs. The deep learning and ratiometric approaches performed better than the phasor analysis approach compared to the ground truth. The deep learning approach had the most precision of the three methods. The ratiometric approach has the most versatility and maintained accuracy, albeit with less precision. Our data suggest that implementing these methods to analyze CMH images can drastically increase the processing speed while maintaining precision and accuracy.

Significance: To explore brain architecture and pathology, a consistent and reliable methodology to visualize the three-dimensional cerebral microvasculature is beneficial. Perfusion-based vascular labeling is quick and easily deliverable. However, the quality of vascular labeling can vary with perfusion-based labels due to aggregate formation, leakage, rapid photobleaching, and incomplete perfusion. Aim: We describe a simple, two-day protocol combining perfusion-based labeling with a two-day clearing step that facilitates whole-brain, three-dimensional microvascular imaging and characterization. Approach: The combination of retro-orbital injection of Lectin-Dylight-649 to label the vasculature, the clearing process of a modified iDISCO+ protocol, and light-sheet imaging collectively enables a comprehensive view of the cerebrovasculature. Results: We observed ∼threefold increase in contrast-to-background ratio of Lectin-Dylight-649 vascular labeling over endogenous green fluorescent protein fluorescence from a transgenic mouse model. With light-sheet microscopy, we demonstrate sharp visualization of cerebral microvasculature throughout the intact mouse brain. Conclusions: Our tissue preparation protocol requires fairly routine processing steps and is compatible with multiple types of optical microscopy.

We propose a new image sensor based on multi-aperture optics and multi-tap charge modulators for simultaneous multi-band spatial frequency domain imaging (SFDI) and blood flow mapping by multi-exposure laser speckle contrast imaging (ME-LSCI). Wavelength-multiplexed imaging implemented by attaching different bandpass filters to each lens enables to perform multi-band SFDI and ME-LSCI simultaneously. A prototype camera for 4-band SFDI (568, 668, 734, 846nm) and single-band LSCI (780nm) was built based on a commercial CMOS camera with enhanced NIR sensitivity to demonstrate the concept and tested using a rat model of graded burns.

We describe the development of a perturbation-free technique to measure tissue blood flow and metabolic rate of oxygen (MRO2) using LSI and SFDI. Analytical (diffusion) and computational (Monte Carlo) models are employed to characterize the contributions of diffuse and directed flow to the measured speckle contrast. Measured flow data is combined with the deoxygenated hemoglobin concentration and a path-length factor (both obtained from SFDI) to model tissue oxygen consumption in units of M O2/min. The results of this model were comparable with values of MRO2 measured with other imaging methods (PET, MRI/MRS).

Cerebral microbleeds (CMB) are deposits of blood that accumulate within the brain. An increase in CMBs is associated with an increased risk of cognitive impairment and stroke. The types of vessels associated with CMB formation remains unclear. We recently demonstrated the combined use of exogenous labels, vessel painting, and optical clearing to achieve three-dimensional views of blood vessels and CMBs. Here, we aimed to characterize brain vasculature by quantifying key vasculature-related metrics. An automated algorithm was developed to segment blood vessels within fluorescence images. An open-source neuron tracing software, neuTube, was then used to quantify blood vessel diameters.

Seismic hazard calculations are based on the assumption that seismicity rates are stable over time. In a given area, the seismicity recorded through historical archives and seismometers is considered a reliable indicator to model the occurrence of future high magnitude seismic events. But, to discuss this hypothesis regionally, it is essential to reconstruct long term seismicity.The junction between the Jura mountains and the Alps is seismically active, as shown by the occurrence of numerous seismic events and the presence of several active faults (De La Taille, 2015). Since the 15th century, more than twenty earthquakes of epicentral intensity greater than VII have been identified in this area. In addition, sedimentary sequences from Lake Annecy and Lake du Bourget have highlighted the capacity of these "natural archives" to record recurrent seismic activity (Beck 2009), with a potential major seismic event identified around 9900 cal. BP (Arnaud et al., 2012). Such lacustrine archives are key to better understand 1) the occurrence of major seismic events and 2) the evolution of seismicity rates through time, prior to historical and instrumental records.Here, we present two sedimentary sequences of 11 and 16 metres long respectively, sampled in the shallowest and deepest basins of Lake Aiguebelette (altitude: 374 m). We performed sedimentological, geochemical and paleomagnetic analyses combined with seismic profile analyses and radiocarbon dating to study processes of event layer deposition in this lake. Multi-proxy analyses allow a quantitative identification of event layers, contrasting with varved-sedimentation. In the deepest basin sequence, 33 homogenites are identified through variations of the laboratory induced isothermal remanent magnetization of sediments measured with a high-resolution fluxgate scanner (Demory et al., 2019) and high foliation (>2%) of the Anisotropy of Magnetic Susceptibility. These parameters are usually associated with seiche effect induced by seismic activity (Campos et al., 2013). Among these event layers, archived in the deep basin sequence, three of them occured synchronously in the shallow basin (at 3000 ± 100, 6900 ± 100 and 11400 ± 300 cal. BP, respectively).The oldest and thickest event layer recorded in Lake Aiguebelette was deposited at the transition between the Late Glacial and Holocene stages. In the deepest basin, this 1.15-meter-thick deposit is composed of an upward-graded base and a 0.84 meter-thick homogenite, which was also identified as a transparent facies on seismic profiles. In Lake Le Bourget, Lake Annecy, and central Swiss perialpine lakes, several seismic profiles analyses show transparent seismic facies interpreted as mass movement deposits occurring at the same period of time: the Late glacial-Holocene transition.Did this climatic transition influence the seismic activity in the Alps? International audience; Regime shifts are major reorganization of ecological processes, creating new sets of mechanisms that drive the new ecological regime. Such rearrangements can affect how and how much the system responds to pressures other than those that created the shift (interactive carryover). Lake Geneva still exhibits high levels of productivity despite reductions in phosphorus to its reference baseline; the continued high productivity is likely due to the synergistic effects of climate change. We tested whether the contemporary Lake Geneva plankton community response to air temperature, one symptom of climate change, differed from the responses to past changes in air temperature. We used paleoecology to quantify the changes in plankton communities, as a proxy of general ecological changes, over the past 1500 years. Our results show that from 563 AD (beginning of the record) to the twentieth century, the cladoceran assemblage remained stable, despite climate variability of 3 °C in air temperature. The plankton community of Lake Geneva appeared to shift for the first time in the 1500 year record in 1946, and dynamic linear models suggested that 1958–1961 was a critical transition period when the ecosystem changed state. Littoral species were lost, and the assemblage became dominated by pelagic species. The shift took place around the beginning of the current long-term monitoring program, when local perturbations (eutrophication) were escalating. Our results suggest that eutrophication acted as a switch towards a lake more vulnerable to climate change.

Background The regulation of cerebral blood flow is critical for normal brain functioning, and many physiological and pathological conditions can have long-term impacts on cerebral blood flow. However, minimally invasive tools to study chronic changes in animal models are limited. New method We developed a minimally invasive surgical technique (cyanoacrylate skull, CAS) allowing us to image cerebral blood flow longitudinally through the intact mouse skull using laser speckle imaging. Results With CAS we were able to detect acute changes in cerebral blood flow induced by hypercapnic challenge. We were also able to image cerebral blood flow dynamics with laser speckle imaging for over 100 days. Furthermore, the relative cerebral blood flow remained stable in mice from 30 days to greater than 100 days after the surgery. Comparison with existing methods Previously, achieving continuous long-term optical access to measure cerebral blood flow in individual vessels in a mouse model involved invasive surgery. In contrast, the CAS technique presented here is relatively non-invasive, as it allows stable optical access through an intact mouse skull. Conclusions The CAS technique allows researcher to chronically measure cerebral blood flow dynamics for a significant portion of a mouse’s lifespan. This approach may be useful for studying changes in blood flow due to cerebral pathology or for examining the therapeutic effects of modifying cerebral blood flow in mouse models relevant to human disease.

Studying cerebral microhemorrhages (CMHs) is a laborious and time-intensive process. To improve this process, we developed and compared three digital pathology approaches to identify and quantify CMHs from Prussian blue-stained brain sections.

We present a method for visualizing cerebral microvasculature in three-dimensions using an exogenous fluorescent label, tissue clearing, and confocal microscopy. A segmentation algorithm was developed for quantitative analysis of microvasculature.

Sustainably maintaining the densely populated upland plains of Madagascar as operationally safe spaces for the food security of the nation and the urban growth of its capital city, Antananarivo, hinges critically on avoiding crop and infrastructure destruction by their through-flowing rivers. The flood regime, however, is also a function of two ‘slow’ variables hitherto undocumented: tectonic subsidence regime, and floodplain sedimentation rate. From a radiocarbon-dated chronostratigraphy and environmental history of the sediment sequences in three of Madagascar's semi-enclosed upland basins (Antananarivo, Ambohibary, and Alaotra), we quantify and compare how the precarious equilibrium between the two variables entails differentials in accommodation space for sediment and floodwater. Results show that all these plains have been wetlands for at least 40,000 years, but that the Antananarivo Basin is the most vulnerable because the imbalance between sedimentation and subsidence is the largest. Although the tectonic regime and the endemic forms of gully erosion that occur in the catchments are beyond human control, we advocate that flood mitigation strategies should focus on the natural grassland savanna, which makes up most of the contributing areas to surface runoff in the watersheds. Pastoralists are persistently left out of rural development programmes, yet the rangelands could benefit from the introduction of multi-purpose grasses and legumes known to withstand high stocking rates on poor soils while combining the benefits of nutritiousness, fire and drought resistance, with good runoff-arrest and topsoil-retainment abilities. Future-proofing Madagascar's upland grainbaskets and population centres thus calls for joined-up action on the sediment cascade, focusing on soil and water sequestration through integrated watershed management rather than on hard-defence engineering against overflowing rivers on the plains, which has been the costly but ineffectual approach since the 17th century.

International audience; Cosmogenic nuclide dating of glacial landforms may lead to ambiguous results for ice retreat histories. The persistence of significant cosmogenic concentrations inherited from previous exposure may increase the apparent exposure ages for polished bedrocks affected by limited erosion under ice and for erratic boulders transported by glaciers and previously exposed in high‐altitude rock walls. In contrast, transient burying by moraines, sediments and snow decreases the apparent exposure age. We propose a new sampling strategy, applied to four sites distributed in the Arc and Arve valleys in the Western Alps, to better constrain the factors that can bias exposure ages associated with glacial processes. We used the terrestrial cosmogenic nuclide 10Be (TCN) to estimate the exposure time from paired sampling of depth profiles in polished bedrock and on overlying erratic boulders. For a given sampling site, the exposure ages for both the polished bedrock and boulder are expected to be the same. However, in six cases out of seven, boulders had significantly higher 10Be surface concentrations than those of the associated polished surfaces. In present and past glacial processes, the 10Be distribution with depth for boulders and bedrocks implies the presence of an inheritance concentration of 10Be. Our study suggests that 10Be concentrations in erratic boulders and in polished bedrocks provide maximum and minimum exposure ages of the glacial retreat, respectively.

Background Impaired neurological function affects 85% to 90% of cardiac arrest ( CA ) survivors. Pulsatile blood flow may play an important role in neurological recovery after CA . Cerebral blood flow ( CBF ) pulsatility immediately, during, and after CA and resuscitation has not been investigated. We characterized the effects of asphyxial CA on short‐term (CA ) CBF and femoral arterial blood pressure ( ABP ) pulsatility and studied their relationship to cerebrovascular resistance ( CVR ) and short‐term neuroelectrical recovery. Methods and Results Male rats underwent asphyxial CA followed by cardiopulmonary resuscitation. A multimodal platform combining laser speckle imaging, ABP , and electroencephalography to monitor CBF , peripheral blood pressure, and brain electrophysiology, respectively, was used. CBF and ABP pulsatility and CVR were assessed during baseline, CA , and multiple time points after resuscitation. Neuroelectrical recovery, a surrogate for neurological outcome, was assessed using quantitative electroencephalography 90 minutes after resuscitation. We found that CBF pulsatility differs significantly from baseline at all experimental time points with sustained deficits during the 2 hours of postresuscitation monitoring, whereas ABP pulsatility was relatively unaffected. Alterations in CBF pulsatility were inversely correlated with changes in CVR , but ABP pulsatility had no association to CVR . Interestingly, despite small changes in ABP pulsatility, higher ABP pulsatility was associated with worse neuroelectrical recovery, whereas CBF pulsatility had no association. Conclusions Our results reveal, for the first time, that CBF pulsatility and CVR are significantly altered in the short‐term postresuscitation period after CA . Nevertheless, higher ABP pulsatility appears to be inversely associated with neuroelectrical recovery, possibly caused by impaired cerebral autoregulation and/or more severe global cerebral ischemia.

International audience; A set of 24 AMS 14C measurements were performed on different sediment fractions from short gravity cores taken from the Gulf of Cariaco (northeastern Venezuela) in order to construct a detailed sedimentary archive for recent centuries. A local reservoir effect was expected because of i) strong upwelling from the neighboring Cariaco Trough and anoxia and ii) the specific geomorphological setting related to active faults with seeps and diapirism. Measurements were thus performed on different components: sediment bulk organic fraction (SBOF), pteropods shells (surface water), bivalve shells (bottom), and plant fragments. Based on comparisons of the different results, we discuss different age corrections and calibrations, which lead us to consider a negligible local reservoir correction (ΔR) for the bivalve and pteropod shells, which show no reservoir age for carbonate and an absence of water stratification. However, a local 633 ± 64 yr ΔR appears in the organic fraction of the sediment, and its origin is primary related to the upwelling mechanism

International audience; Chronologies of lake-sediment records covering the last centuries to millennia are usually based on both short-lived radionuclides and radiocarbon dating. However, beyond the range of short-lived radionuclides, age model accuracy often suffers from large radiocarbon uncertainties. For high-altitude records, this issue is even more prominent as terrestrial plant fragments for radiocarbon dating are often lacking due to the sparse vegetation in such environments. In this study, we evaluate the potential of the geomagnetic field secular variations as a complementary tool to establish more robust age–depth relationships. Our palaeomagnetic study, applied to five high-altitude lakes from the western European Alps, first shows that recent unconsolidated sediments can carry stable remanent magnetization. The analysis of the magnetic parameters indicates that low-coercivity pseudo-single domain magnetite grains carry the natural magnetization. Nevertheless, the quality of palaeomagnetic secular variation records varies from one lake to another. This quality can be illustrated through the calculation of the declination/inclination maximum angular variations and their comparison to the expected value. Compared with available models, the declination variations are usually too large and the inclination too high. We discuss the validity of palaeosecular variation (PSV) of the Earth’s magnetic field regarding rock magnetism, magnetization processes and possible deformation during coring. From a magnetic point of view, the quality of data is variable, but the characteristic remanent magnetization direction is consistent at site level between neighbouring lakes and with the reference curve, suggesting that geomagnetic field secular variations are approximately recorded. Finally, we attempt to correlate the declination/inclination variations of the characteristic remanent magnetization measured in the five records to the reference geomagnetic model to provide additional chronological markers for age–depth modelling. These stratigraphic chrono-markers appear in systematic agreement with our previous chronological data and enable a reduction of dating uncertainties up to 30% when including these chrono-markers in the age–depth modelling. This agreement supports the interpretation that PSV may have been recorded more or less accurately depending on the studied lake. Therefore, coupled with a comprehensive understanding through other analysis (sedimentology, dating, geochemistry), PSV can be used to improve the age models in the more favourable cases.

Laser speckle imaging (LSI) is a wide-field, noninvasive optical technique that allows researchers and clinicians to quantify blood flow in a variety of applications. However, traditional LSI devices are cart or tripod based mounted systems that are bulky and potentially difficult to maneuver in a clinical setting. We previously showed that the use of a handheld LSI device with the use of a fiducial marker (FM) to account for motion artifact is a viable alternative to mounted systems. Here we incorporated a handheld gimbal stabilizer (HGS) to produce a motion stabilized LSI (msLSI) device to further improve the quality of data acquired in handheld configurations. We evaluated the msLSI device in vitro using flow phantom experiments and in vivo using a dorsal window chamber model. For in vitro experiments, we quantified the speckle contrast of the FM (K(FM)) using the mounted data set and tested 80% and 85% of K(FM) as thresholds for useable images (K(FM,Mounted,80%) and K(FM,Mounted,85%)). Handheld data sets using the msLSI device (stabilized handheld) and handheld data sets without the HGS (handheld) were collected. Using K(FM,Mounted,80%) and K(FM,Mounted,85%) as the threshold, the number of images above the threshold for stabilized handheld (38 ± 7 and 10 ± 2) was significantly greater (p = 0.031) than for handheld operation (16 ± 2 and 4 ± 1). We quantified a region of interest within the flow region (K(FLOW)), which led to a percent difference of 8.5% ± 2.9% and 7.8% ± 3.1% between stabilized handheld and handheld configurations at each threshold. For in vivo experiments, we quantified the speckle contrast of the window chamber (K(WC)) using the mounted data set and tested 80% of K(WC) (K(WC,Mounted,80%)). Stabilized handheld operation provided 53 ± 24 images above K(WC,Mounted,80%), while handheld operation provided only 23 ± 13 images. We quantified the speckle flow index (SFI) of the vessels and the background to calculate a signal-to-background ratio (SBR) of the window chamber. Stabilized handheld operation provided a greater SBR (2.32 ± 0.29) compared to handheld operation (1.83 ± 0.21). Both the number of images above threshold and SBR were statistically significantly greater in the stabilized handheld data sets (p = 0.0312). These results display the improved usability of handheld data acquired with an msLSI device.

Quantitative measures of blood flow and metabolism are essential for improved assessment of brain health and response to ischemic injury. In this report, we demonstrate a multimodal technique for measuring the cerebral metabolic rate of oxygen (CMRO2) in the rodent brain on an absolute scale (μM O2 / min). We use laser speckle imaging (LSI) at 809 nm and spatial frequency domain imaging (SFDI) at 655 nm, 730 nm, and 850 nm to obtain spatiotemporal maps of cerebral blood flow (CBF), tissue absorption (μa), and tissue scattering (μs’). Knowledge of these three values enables calculation of a characteristic blood flow speed, which in turn is input to a mathematical model with a “zero-flow” boundary condition to calculate absolute CMRO2. We apply this method to a rat model of cardiac arrest (CA) and cardiopulmonary resuscitation. With this model, the zero-flow condition occurs during entry into CA. The CMRO2 values calculated with our method are in good agreement with those measured with magnetic resonance (MR) and positron emission tomography (PET) by other groups. Our technique provides a quantitative metric of cerebral metabolism that can potentially be used for comparison between animals and longitudinal monitoring of a single animal over multiple days, to assess differences in baseline metabolism and track recovery of metabolism in survival studies following ischemia and reperfusion.

Spreading depolarization (SD) accompanies numerous neurological conditions, including migraine, stroke, and traumatic brain injury. There is significant interest in understanding the relationship between SD and neuronal injury. However, characteristics underlying SD and repolarization (RP) induced by global cerebral ischemia (e.g., cardiac arrest (CA)) and reperfusion are not well understood. Quantifying features of SD and RP during CA and cardiopulmonary resuscitation (CPR) may provide important metrics for diagnosis and prognosis of neurological injury from hypoxia-ischemia. We characterized SD and RP in a rodent model of asphyxial CA+CPR using a multimodal platform including electrocorticography (ECoG) and optical imaging. Improved quantitative understanding of the dynamic relationship among cerebral blood flow, oxygen consumption, and electrical activity is important to clinicians treating acute brain injury. Such knowledge would elucidate the neurovascular response to ischemia, helping to potentially guide treatment. Using a multimodal optical imaging platform and a clinically-relevant rat model of cardiac arrest (CA) and cardiopulmonary resuscitation (CPR), we continuously measured cerebral blood flow (CBF), brain tissue oxygenation (StO2), cerebral metabolic rate of oxygen (CMRO2), and cerebral electrical activity (electrocorticography; ECoG). Multiple phases of cerebral hemodynamic recovery, with different degrees of mismatch between CBF and CMRO2, were observed following CPR. At 1 min post-resuscitation, we observed that the ratio CBF/CMRO2 is indicative of CA duration/severity and prognostic (with 87% accuracy) of short-term neurological recovery measured by the re-initiation of ECoG activity. These measurements provide the earliest known metrics for assessment of CA severity and prognosis post-CPR. Interestingly, the accuracy of this information is lost beyond 2-3 minutes post-CPR, highlighting a critical, easily overlooked, period immediately post-CPR. These metrics do not require pre-resuscitation data, underscoring translational potential in emergency-response settings when pre-CA information is unavailable. These metrics encourage validation in human studies, potentially offering real-time feedback during CA/CPR to optimize neurological outcome.

Multi-exposure laser speckle contrast imaging (MELSCI) systems based on high frame rate cameras are suitable for wide-field quantitative measurement of blood flow. However, high-speed camera-based MELSCI requires high power consumption, large memory, and high processing capability, which may lead to relatively large and expensive hardware. To realize a compact and cost-efficient MELSCI system, we discuss an application of the multi-tap CMOS image sensor originally designed for time-of-flight range imaging. This image sensor operated in the global shutter mode and every pixel was provided with multiple charge-storage diodes. Multiple images for different exposures were acquired simultaneously because exposure patterns were programmable to implement an arbitrary exposure duration for each tap. The frame rate was close to video frame rates (30 frames per second (fps)) regardless of the exposure pattern. The feasibility of the proposed method was verified by simulations that were performed with real speckle images captured by a high-speed camera at 40 kfps. Experiments with a four-tap CMOS image sensor demonstrated that a flow speed map was obtained at a moderate frame rate such as 35 fps for a moving ground glass plate and 45 fps for flowing Intralipose, which were linearly moved at speeds of 1-5 mm/s.

International audience; PurposeKnowledge of suspended sediment provenance in mesoscale catchments is important for applying erosion control measures and best management practices as well as for understanding the processes controlling sediment transport in the critical zone. As suspended sediment fluxes are highly variable in time, particularly given the variability of soil and rainfall properties in mesoscale catchments, knowledge of sediment provenance at high temporal resolution is crucial.Materials and methodsSuspended sediment fluxes were analyzed at the outlet of a 42-km2 Mediterranean catchment belonging to the French critical zone observatory network (OZCAR). Spatial origins of the suspended sediments were analyzed at high temporal resolution using low-cost analytical approaches (color tracers, X-ray fluorescence, and magnetic susceptibility). As the measurements of magnetic susceptibility provide only one variable, they were used for cross-validation of the results obtained with the two alternative tracing methods. The comparison of the tracer sets and three mixing models (non-negative least squares, Bayesian mixing model SIMMR, and partial least squares regression) allowed us to estimate different sources of errors inherent in sediment fingerprinting studies and to assess the challenges and opportunities of using these fingerprinting methods.Results and discussionAll tracer sets and mixing models could identify marly badlands as the main source of suspended sediments. However, the percentage of source contributions varied between the 11 flood events in the catchment. The mean contribution of the badlands varied between 74 and 84%; the topsoils on sedimentary geology ranged from 12 to 29% and the basaltic topsoils from 1 to 8%. While for some events the contribution remained constant, others showed a high within-event variability of the sediment provenance. Considerable differences in the predicted contributions were observed when different tracer sets (mean RMSE 19.9%) or mixing models (mean RMSE 10.1%) were used. Our result shows that the choice of the tracer set was more important than the choice of the mixing model.ConclusionsThese results highlighted the importance of using multi-tracer multi-model approaches for sediment fingerprinting in order to obtain reliable estimates of source contributions. As a given fingerprinting approach might be more sensitive to one type of error, i.e., source variability, particle size selectivity, multi-tracer ensemble predictions allow to detect and quantify these potential biases. High sampling resolution realized with low-cost methods is important to reveal within- and between-event dynamics of sediment fluxes and to obtain reliable information of main contributing sources.

Soil erosion is strongly linked to both precipitation patterns and land-use. We examine the effects of erosion and its drivers (i.e. human or/and climate) on soil evolution from the study of lacustrine archives in the northern French Alps. Multi-proxy analyses of the Lake Gers sediment sequence combined with the study of soils and rocks of its catchment allowed to reconstruct its evolution over the past 4.6 kyr. This included 14C dating, short-lived radionuclides, geochemistry, loss on ignition, grain-size analyses, as well as plant and mammal DNA analyses. A total of 127 instantaneous deposits were identified among the continuous sedimentation in the lake and 93 were interpreted as flood deposits. Erosion was quantified for the whole catchment considering both the continuous sedimentation and the flood deposit thicknesses. The catchment is mainly formed by andesitic sandstones over which Andosols and Podzols can develop. However, low weathered materials from colluviation constitute the main input to the lake. Four main phases of changes in soil weathering were recorded by increases in K2O/TiO2 ratios, associated with an increase in both erosion and flood-frequency. Two phases were associated with climate cooling in the Western Alps, from 2.65 to 2.5 and from 1.45 to 1.3 cal kyr BP. The two others (1.9–1.75 and 0.9–0.4 cal kyr BP) were triggered with deforestation, with cow grazing during the Roman period, and with grazing of cows, sheep and goats during the Medieval period, amplified by the onset of the Little Ice Age. The increase in erosion and flood frequency after 950 cal yr BP indicates substantial damages to soils of the catchment including increase of their erodibility over the last millennia.

We have previously demonstrated that the use of a commercially-available immersion-based optical clearing agent (OCA) enables, within 3-6 hours, three-dimensional visualization of subsurface exogenous fluorescent and absorbing markers of vascular architecture and neurodegenerative disease in thick (0.5-1.0mm) mouse brain sections. Nonetheless, the relative performance of immersion-based OCAs has remained unknown. Here, we show that immersion of brain sections in specific OCAs (FocusClear, RIMS, sRIMS, or ScaleSQ) affects both their transparency and volume; the optical clearing effect occurs over the entire visible spectrum and is reversible; and that ScaleSQ had the highest optical clearing potential and increase in imaging depth of the four evaluated OCAs, albeit with the largest change in sample volume and a concomitant decrease in apparent microvascular density of the sample. These results suggest a rational, quantitative framework for screening and characterization of the impact of optical clearing, to streamline experimental design and enable a cost-benefit assessment.

The Avignonet landslide in the Trieves area is made of Quaternary paraglacial sediments among which imbricated till, glacio-lacutrine and fluviatil layers. Numerous boreholes (drillings and corings) were conducted during the mid-1980s and the mid-2000s to assess the geological and geotechnical setting of the inhabited, southern part of this slow-moving earthslide. Heterogeneous paramaters available from these different boreholes make interpretation difficult. Gamma-ray logging along with magnetic susceptibility and pocket vane tests on core samples allowed to distinguish between the different lithological and geotechnical units. Mechanical strength obtained from drilling parameter recording acquired during drilling was correlated to corings and their combination, along with outcrop observations, was used to delineate the lithological units and shear surfaces. The joint analysis of the dataset with outcrop observations and chronological data reveals a high geological complexity with the intrication of various sedimentary units. Two ancient landslides, several tens of metres in width, were furthermore identified within the large Avignonet landslide, almost 2 km in width. All the results point toward a high lithological and hydrogeological complexity that impacts the dynamics of the landslide. This work shows the interest of a multi-proxy approach in studies involving Quaternary sedimentary units. The methodology developed here could be applied to other sites with different palaeoenvironmental contexts initially exhibiting sedimentary compaction, such as lacustrine or coastal environments.

International audience; The long-term response of the flood activity to both Atlantic and Mediterranean climatic influences was explored by studying a lake sequence (Lake Foreant) of the Western European Alps. High-resolution sedimentological and geochemical analysis revealed 171 event layers, 168 of which result from past flood events over the last millennium. The layer thickness was used as a proxy of intensity of past floods. Because the Foreant palaeoflood record is in agreement with the documented variability of historical floods resulting from local and mesoscale, summer-to-autumn convective events, it is assumed to highlight changes in flood frequency and intensity related to such events typical of both Atlantic (local events) and Mediterranean (mesoscale events) climatic influences. Comparing the Foreant record with other Atlantic-influenced and Mediterranean-influenced regional flood records highlights a common feature in all flood patterns that is a higher flood frequency during the cold period of the Little Ice Age (LIA, AD 1300-1900). In contrast, high-intensity flood events are apparent during both the cold LIA and the warm Medieval Climate Anomaly (MCA, AD 9501250). However, there is a tendency towards higher frequencies of high-intensity flood events during the warm MCA. The MCA extremes could mean that under the global warming scenario, we might see an increase in intensity (not in frequency). However, the flood frequency and intensity in the course of the 20th century warming trend did not change significantly. Uncertainties in future evolution of flood intensity lie in the interpretation of the lack of 20th century extremes (transition or stable?) and the different climate forcing factors between the two periods (greenhouse gases vs. solar and/or volcanic eruptions).

Seismic hazard assessment is a critical but challenging issue for modern societies. A key parameter to be estimated is the recurrence interval of damaging earthquakes. This requires the establishment of earthquake records long enough to be relevant, i.e., far longer than historical observations. We study how lake sediments can be used for this purpose and explore conditions that enable lake sediments to record earthquakes. This was achieved (i) through the compilation of eight lake‐sediment sequences from the European Alps to reconstruct chronicles of mass movement deposits and (ii) through the comparison of these chronicles with the well‐documented earthquake history. This allowed 24 occurrences of mass movements to be identified, of which 21 were most probably triggered by an earthquake. However, the number of earthquake‐induced deposits varies between lakes of a same region, suggesting variable thresholds of the lake sequences to record earthquake shaking. These thresholds have been quantified by linking the mass movement occurrences in a single lake to both intensity and distance of the triggering earthquakes. This method offers a quantitative approach to estimate locations and intensities of past earthquake epicenters. Finally, we explored which lake characteristics could explain the various sensitivities. Our results suggest that sedimentation rate should be larger than 0.5 mm yr−1 so that a given lake records earthquakes in moderately active seismotectonic regions. We also postulate that an increasing sedimentation rate may imply an increasing sensitivity to earthquake shaking. Hence, further paleoseismological studies should control carefully that no significant change in sedimentation rates occurs within a record, which could falsify the assessment of earthquake recurrence intervals.
