1. Defining Activity Thresholds Triggering a 'Stand Hour' for Apple Watch Users: Cross-Sectional Study

Abstract: BackgroundSedentary behavior (SB) is one of the largest contributing factors increasing the risk of developing noncommunicable diseases, including cardiovascular disease and type 2 diabetes. Guidelines from the World Health Organization for physical activity suggest the substitution of SB with light physical activity. The Apple Watch contains a health metric known as the stand hour (SH). The SH is intended to record standing with movement for at least 1 minute per hour; however, the activity measured during the determination of the SH is unclear. ObjectiveIn this cross-sectional study, we analyzed the algorithm used to determine time spent standing per hour. To do this, we investigated activity measurements also recorded on Apple Watches that influence the recording of an SH. We also aimed to estimate the values of any significant SH predictors in the recording of a SH. MethodsThe cross-sectional study used anonymized data obtained in August 2022 from 20 healthy individuals gathered via convenience sampling. Apple Watch data were extracted from the Apple Health app through the use of a third-party app. Appropriate statistical models were fitted to analyze SH predictors. ResultsOur findings show that active energy (AE) and step count (SC) measurements influence the recording of an SH. Comparing when an SH is recorded with when an SH is not recorded, we found a significant difference in the mean and median AE and SC. Above a threshold of 97.5 steps or 100 kJ of energy, it became much more likely that an SH would be recorded when each predictor was analyzed as a separate entity. ConclusionsThe findings of this study reveal the pivotal role of AE and SC measurements in the algorithm underlying the SH recording; however, our findings also suggest that a recording of an SH is influenced by more than one factor. Irrespective of the internal validity of the SH metric, it is representative of light physical activity and might, therefore, have use in encouraging individuals through various means, for example, notifications, to reduce their levels of SB.

2. Tropical vs. temperate sea turtle population resilience to extreme temperatures

Abstract: Rising temperatures due to global warming over the last decades pose threats to marine biodiversity. Sea turtles are ectothermic species, and their embryonic development depends on nest conditions, particularly temperature. Here, we explore how increasing and extreme temperatures within the 558 nests can impact the hatching success of two sea turtle species, the green turtle (Chelonia mydas) and the loggerhead turtle (Caretta caretta), at two tropical locations, Cape Verde and Australia, and at temperate nesting sites, Turkey and Greece. Mean nest temperatures were higher for green turtles (30.84 °C) than for loggerhead turtles (29.42 °C) and significantly increased in all locations as the air temperature increased. However, nests from temperate locations, such as green turtles in Turkey and loggerhead turtles in Cape Verde, experienced higher temperatures than the nests in Australia and Greece, reaching lethal temperatures at extreme temperature peaks. Thus, temperate populations might be at risk as global warming continues because it is uncertain whether these higher latitudes can provide beach habitats cool enough for successful sea turtle nesting given the projected more frequent extreme temperatures in the future.

3. The effect of hyperkalemia and long inter-dialytic interval on morbidity and mortality in patients receiving hemodialysis: a systematic review

Abstract: Background Patients with chronic kidney disease, especially those receiving hemodialysis (HD), are at risk of hyperkalemia (HK). This systematic review aimed to evaluate the prevalence of HK in patients with renal disease receiving HD and collate evidence on the effect of HK and differing HD patterns (i.e., long vs. short inter-dialytic intervals [LIDI and SIDI, respectively] in a thrice weekly schedule) on mortality. Methods Comprehensive searches were conducted across six databases and selected conference proceedings by two independent reviewers up to September 2020. A hundred and two studies reporting frequency of HK, mortality, or cardiovascular (CV) outcomes in adult patients with acute, chronic or end-stage renal disease in receipt of HD were included. Narrative synthesis of results was undertaken with key findings presented in tables and figures. Results Median prevalence of HK in patients with renal disease receiving HD was 21.6% and increased in patients receiving concomitant medications – mainly renin–angiotensin–aldosterone system inhibitors and potassium-sparing diuretics. Associations between elevated potassium levels and increased risk of both all-cause and CV mortality in the HD population were consistent across the included studies. In addition, there was a rise in all-cause and CV mortality on the day following LIDI compared with the day after the two SIDIs in patients on HD. Conclusions Evidence identified in this systematic review indicates a relationship between HK and LIDI with mortality in patients with renal disease receiving HD, emphasizing the need for effective monitoring and management to control potassium levels both in emergency and chronic HD settings.

4. Deep Metric Learning for Scalable Gait-Based Person Re-Identification Using Force Platform Data

Abstract: Walking gait data acquired with force platforms may be used for person re-identification (re-ID) in various authentication, surveillance, and forensics applications. Current force platform-based re-ID systems classify a fixed set of identities (IDs), which presents a problem when IDs are added or removed from the database. We formulated force platform-based re-ID as a deep metric learning (DML) task, whereby a deep neural network learns a feature representation that can be compared between inputs using a distance metric. The force platform dataset used in this study is one of the largest and the most comprehensive of its kind, containing 193 IDs with significant variations in clothing, footwear, walking speed, and time between trials. Several DML model architectures were evaluated in a challenging setting where none of the IDs were seen during training (i.e., zero-shot re-ID) and there was only one prior sample per ID to compare with each query sample. The best architecture was 85% accurate in this setting, though an analysis of changes in walking speed and footwear between measurement instances revealed that accuracy was 28% higher on same-speed, same-footwear comparisons, compared to cross-speed, cross-footwear comparisons. These results demonstrate the potential of DML algorithms for zero-shot re-ID using force platform data, and highlight challenging cases.

Abstract: Summary: The etiology of peripartum cardiomyopathy remains unknown. One hypothesis is that an increase in the 16-kDa form of prolactin is pathogenic and suggests that breastfeeding may worsen peripartum cardiomyopathy by increasing prolactin, while bromocriptine, which blocks prolactin release, may be therapeutic. An autoimmune etiology has also been proposed. The authors investigated the impact of breastfeeding on cellular immunity and myocardial recovery for women with peripartum cardiomyopathy in the IPAC (Investigations in Pregnancy Associated Cardiomyopathy) study. Women who breastfed had elevated prolactin, and prolactin levels correlated with elevations in CD8+ T cells. Abstract: Deep learning algorithms have shown remarkable performance in classification tasks, however, they typically perform poorly with small training datasets due to overfitting. Overfitting occurs for all data types, although for the purposes of this study we are interested in time-based signals. This study introduces a novel technique to simulate time series signals from a dataset of categorically labeled data which can be used to train a deep neural network. The objective is to improve the predictive accuracy of a deep neural network on a separate validation dataset. To demonstrate the simulation methodology and improvements to the model's performance, a small dataset of ground reaction forces was used with the goal of identifying a person based on the raw signal. Our results show that the simulation method presented improves validation accuracy and reduces model training time for each of the three signal types.

Abstract: Sepsis is associated with a dysregulated inflammatory response to infection. Despite the activation of inflammation, an immune suppression is often observed, predisposing patients to secondary infections. Therapies directed at restoration of immunity may be considered but should be guided by the immune status of the patients. In this paper, we described the use of a high-dimensional flow cytometry (HDCyto) panel to assess the immunophenotype of patients with sepsis. We then isolated peripheral blood mononuclear cells (PBMCs) from patients with septic shock and mimicked a secondary infection by stimulating PBMCs for 4 h in vitro with lipopolysaccharide (LPS) with or without prior exposure to either IFN-γ, or LAG-3Ig. We evaluated the response by means of flow cytometry and high-resolution clustering cum differential analysis and compared the results to PBMCs from healthy donors. We observed a heterogeneous immune response in septic patients and identified two major subgroups: one characterized by hypo-responsiveness (Hypo) and another one by hyper-responsiveness (Hyper). Hypo and Hyper groups showed significant differences in the production of cytokines/chemokine and surface human leukocyte antigen-DR (HLA-DR) expression in response to LPS stimulation, which were observed across all cell types. When pre-treated with either interferon gamma (IFN-γ) or lymphocyte-activation gene 3 (LAG)-3 recombinant fusion protein (LAG-3Ig) prior to LPS stimulation, cells from the Hypo group were shown to be more responsive to both immunostimulants than cells from the Hyper group. Our results demonstrate the importance of patient stratification based on their immune status prior to any immune therapies. Once sufficiently scaled, this approach may be useful for prescribing the right immune therapy for the right patient at the right time, the key to the success of any therapy.

10. Green and Screen: Does Mobile Photography Enhance or Hinder Our Connection to Nature?

Abstract: Connection to nature, which describes the affective, experiential and cognitive aspects of our relationship with the natural world, has been positively associated with increased wellbeing, as well as pro-environmental beliefs and behaviours. It has also been identified as a worthwhile and distinct goal of education, both in terms of environmental education assessment, as well as in broader terms. This short article aims to explore the effect of using mobile technology to enhance our connection to nature, through a short-term intervention. A total of 57 undergraduates (age range 20 - 31) were randomised into two groups and undertook a short walk in urban nature, with the instruction to notice the beauty in nature and note three beautiful things. One of the groups was asked to record these beautiful things using mobile technology (photography, audio or video recording). Pre- and post-activity measures of the nature connection, using the Connection to Nature Scale (CNS) State were taken and analysed. Results showed that although the intervention had an overall positive effect on participants’ connection to nature, technology neither enhanced nor hindered it. Qualitative analysis of participants’ description of the activity shows a largely positive experience. Limitations of this small trial are presented, and the potential to build an application to engage young people with nature, for increased wellbeing and sustainability, is discussed as a future direction.

11. Increasing Nature Connection in Children: A Mini Review of Interventions

Abstract: Half of the world’s population live in the urban environment. Lifestyle changes in the 20th century have led to spending more time indoors and less in nature. Due to safety concerns, longer hours in formal education, as well as lack of suitable outdoor environments, children in particular have been found to spend very little time outdoors. We have an opportunity, both timely and unique to have our children (re)connect with nature. Nature connection is a subjective state and trait that encompasses affective, cognitive, and experiential aspects in addition to being positively associated with wellbeing, and strong predictor of pro-environmental attitudes and behaviors. This mini-review brings together recent studies that report on interventions to increase nature connection in children. Fourteen studies were identified through electronic searches of Web of Science, Scopus, PsychInfo, ERIC, and Google Scholar. The review aims to offer an overview of the interventions identified, provide a snapshot of the current state of the literature, briefly present themes and trends in the studies identified in relation to nature connection in young people, and propose potential guidelines for future work.

Abstract: Lambda interferons (IFN-λs) are a major component of the innate immune defense to viruses, bacteria, and fungi. In human liver, IFN-λ not only drives antiviral responses, but also promotes inflammation and fibrosis in viral and non-viral diseases. Here we demonstrate that macrophages are primary responders to IFN-λ, uniquely positioned to bridge the gap between IFN-λ producing cells and lymphocyte populations that are not intrinsically responsive to IFN-λ. While CD14+ monocytes do not express the IFN-λ receptor, IFNLR1, sensitivity is quickly gained upon differentiation to macrophages in vitro. IFN-λ stimulates macrophage cytotoxicity and phagocytosis as well as the secretion of pro-inflammatory cytokines and interferon stimulated genes that mediate immune cell chemotaxis and effector functions. In particular, IFN-λ induced CCR5 and CXCR3 chemokines, stimulating T and NK cell migration, as well as subsequent NK cell cytotoxicity. Using immunofluorescence and cell sorting techniques, we confirmed that human liver macrophages expressing CD14 and CD68 are highly responsive to IFN-λ ex vivo. Together, these data highlight a novel role for macrophages in shaping IFN-λ dependent immune responses both directly through pro-inflammatory activity and indirectly by recruiting and activating IFN-λ unresponsive lymphocytes.

Abstract: Podjęte badania w projekcie Skills Training for Effective Practice miały na celu określenie „aktywnych form” skutecznej pomocy osobom długotrwale bezrobotnym w poszukiwaniu zatrudnienia. Przeanalizowano ponad 200 badań i opracowań badawczych, które miały pierwszeństwo przed randomizowanymi badaniami kontrolowanymi (RCT) i badaniami niezależnymi. Wzięto także pod uwagę wnioski z badań w dziedzinie psychologii, doradztwa interpersonalnego i motywacji. Najważniejszym czynnikiem pomyślnego wsparcia okazała się jakość relacji między doradcą a bezrobotnym, a także szczegółowe opisy odpowiednich umiejętności i zachowań doradcy. Metody budowania własnej skuteczności zwiększają wybór stanowiska, wyszukiwania, konkurencji i umiejętności utrzymywania stanowiska wśród klientów. Bezpośredni kontakt z pracodawcami dostarcza użytecznych informacji na temat istniejących wakatów i zadań związanych z pracą oraz może pomóc w przezwyciężeniu uprzedzeń. Schemat programu może wpływać pozytywnie lub negatywnie na wyniki. Badania zostały opracowane jako program nauczania umiejętności doradców ds. zatrudnienia.

Abstract: Human genetic and animal studies have implicated the costimulatory molecule CD40 in the development of multiple sclerosis (MS). We investigated the cell specific gene and protein expression variation controlled by the CD40 genetic variant(s) associated with MS, i.e. the T-allele at rs1883832. Previously we had shown that the risk allele is expressed at a lower level in whole blood, especially in people with MS. Here, we have defined the immune cell subsets responsible for genotype and disease effects on CD40 expression at the mRNA and protein level. In cell subsets in which CD40 is most highly expressed, B lymphocytes and dendritic cells, the MS-associated risk variant is associated with reduced CD40 cell-surface protein expression. In monocytes and dendritic cells, the risk allele additionally reduces the ratio of expression of full-length versus truncated CD40 mRNA, the latter encoding secreted CD40. We additionally show that MS patients, regardless of genotype, express significantly lower levels of CD40 cell-surface protein compared to unaffected controls in B lymphocytes. Thus, both genotype-dependent and independent down-regulation of cell-surface CD40 is a feature of MS. Lower expression of a co-stimulator of T cell activation, CD40, is therefore associated with increased MS risk despite the same CD40 variant being associated with reduced risk of other inflammatory autoimmune diseases. Our results highlight the complexity and likely individuality of autoimmune pathogenesis, and could be consistent with antiviral and/or immunoregulatory functions of CD40 playing an important role in protection from MS.

Abstract: Influenza A infection is a global disease that has been responsible for four pandemics over the last one hundred years. However, it remains poorly understood as to why some infected individuals succumb to life threatening complications whilst others recover and are relatively unaffected. Using gene-expression analysis of circulating leukocytes, here we show that the progression towards severe influenza A infection is characterised by an abnormal transcriptional reprogramming of cell cycle and apoptosis pathways. In severely infected humans, leukocyte gene-expression profiles display opposing cell cycle activities; an increased aberrant DNA replication in the G(1)/S phase yet delayed progression in the G(2)/M phase. In mild infection, cell cycle perturbations are fewer and are integrated with an efficient apoptotic program. Importantly, the loss of integration between cell cycle perturbations and apoptosis marks the transition from a mild viral illness to a severe, life threatening infection. Our findings suggest that circulating immune cells may play a significant role in the evolution of the host response. Further study may reveal alternative host response factors previously unrecognized in the current disease model of influenza.

Abstract: BACKGROUND: Among patients with stable coronary disease and moderate or severe ischemia, whether clinical outcomes are better in those who receive an invasive intervention plus medical therapy than in those who receive medical therapy alone is uncertain. METHODS: We randomly assigned 5179 patients with moderate or severe ischemia to an initial invasive strategy (angiography and revascularization when feasible) and medical therapy or to an initial conservative strategy of medical therapy alone and angiography if medical therapy failed. The primary outcome was a composite of death from cardiovascular causes, myocardial infarction, or hospitalization for unstable angina, heart failure, or resuscitated cardiac arrest. A key secondary outcome was death from cardiovascular causes or myocardial infarction. RESULTS: Over a median of 3.2 years, 318 primary outcome events occurred in the invasive-strategy group and 352 occurred in the conservative-strategy group. At 6 months, the cumulative event rate was 5.3% in the invasive-strategy group and 3.4% in the conservative-strategy group (difference, 1.9 percentage points; 95% confidence interval [CI], 0.8 to 3.0); at 5 years, the cumulative event rate was 16.4% and 18.2%, respectively (difference, -1.8 percentage points; 95% CI, -4.7 to 1.0). Results were similar with respect to the key secondary outcome. The incidence of the primary outcome was sensitive to the definition of myocardial infarction; a secondary analysis yielded more procedural myocardial infarctions of uncertain clinical importance. There were 145 deaths in the invasive-strategy group and 144 deaths in the conservative-strategy group (hazard ratio, 1.05; 95% CI, 0.83 to 1.32). CONCLUSIONS: Among patients with stable coronary disease and moderate or severe ischemia, we did not find evidence that an initial invasive strategy, as compared with an initial conservative strategy, reduced the risk of ischemic cardiovascular events or death from any cause over a median of 3.2 years. The trial findings were sensitive to the definition of myocardial infarction that was used. Abstract: While nature connection, which describes a positive relationship between humans and the rest of the natural world, has been a focus of numerous research studies in the last few decades, relatively little attention has been paid to nature disconnection. While the majority of the populations reported in most studies tend to be highly connected, there is a small percentage of those who feel they have no connection to the natural world. In this paper, we examine this novel construct of nature disconnection through secondary analysis of existing data from the Monitor of Engagement with the Natural Environment survey (MENE) by Natural England. From our analysis of this disconnected population, we can see that they are more likely to be young (16–24 years old), male, not employed and living in rented accommodation. We also observe that they have lower levels of life satisfaction and pro-environmental behaviours. We go on to present an initial theoretical discussion as to the origins of disconnection and propose further research directions to tackle the under-theorisation of this construct.

Abstract: Excessive sea turtle nest predation is a problem for conservation management of sea turtle populations. For green turtles (Chelonia mydas) nesting at Chagar Hutang beach, Redang Island in Malaysia, Asian water monitor lizards (Varanus salvator) are nest predator. To help deter water monitors from attacking nest, plastic mesh is placed on top of sea turtle nests, although this is not always successful in preventing predation. Due to human food waste is frequently dumped near sea turtle nesting area, no studies have documented the if Asian water monitor lizards are attracted by human food waste at Chagar Hutang beach. This study assessed the prevalence of Asian water monitors on the sea turtle nesting beach using passive track-count plots (2×1m) every 50 m along the beach 5-10 m above the high tide mark in the area used by sea turtles to construct their nests. Results indicated Asian water monitors were the only vertebrate nest predator at Chagar Hutang and were widely distributed along the beach and had a Passive Activity Index (PAI) of 1.25. The stomach contents of 20 Asian water monitors were examined by stomach flushing, and 76.8% of stomach contents were human food wastes, 21.2% were turtle egg and hatchling and only 1% were from other natural food sources. This study suggests human food waste attract Asian water monitor to Chagar Hutang beach and that this may result in water monitors visiting sea turtle nests at a higher rate compared to a situation if human food waste was not available.

47. Ocean warming and acidification degrade shoaling performance and lateralization of novel tropical-temperate fish shoals

Abstract: Gregarious behaviours are common in animals and provide various benefits such as food acquisition and protection against predators. Many gregarious tropical species are shifting poleward under current ocean warming, creating novel species and social interactions with local temperate taxa. However, how the dynamics of these novel shoals might be altered by future ocean warming and acidification remains untested. Here we evaluate how novel species interactions, ocean acidification and warming affect shoaling dynamics, motor lateralization and boldness of range-extending tropical and co-shoaling temperate fishes under controlled laboratory conditions. Fishes were exposed to 1 of 12 treatments (combinations of three temperature levels, two pCO2 levels and two shoal type levels: mixed species or temperate only) for 38 days. Lateralization (a measure of asymmetric expression of cognitive function in group coordination and predator escape) of tropical and temperate species was right-side biased under present-day conditions, but side bias significantly diminished in tropical and temperate fishes under ocean acidification. Ocean acidification also decreased shoal cohesion irrespective of shoaling type, with mixed-species shoals showing significantly lower cohesion than temperate-only shoals irrespective of climate stressors. Tropical fish became bolder under ocean acidification (after 4 weeks), and temperate fish became bolder with increasing temperature, while ocean acidification dampened temperate fish boldness. Our findings highlight the direct effect of climate stressors on fish behaviour and the interplay with the indirect effects of novel species interactions. Because strong shoal cohesion and lateralization are key determinants of species fitness, their degradation under ocean warming and acidification could adversely affect species performance in novel assemblages in a future ocean, and might slow down tropical species range extensions.
