National audience; Non-targeted analysis is currently experiencing significant developments in the field of environmental analysis since it opens up investigations of environmental quality to a very large number of compounds. This article consists of feedback from 12 French laboratories on the practices and cautions to be considered when carrying out non-targeted analyses, in particular for the detection of suspect compounds in samples from aquatic environments. Although this analytical approach provides new and very promising information, it is still a complex technique that must be used with many precautions to ensure high-quality results. The joint discussions carried out within the group of laboratories make it possible to synthesise technical information in order to present the challenges of this methodology of major interest in environmental analysis. These issues are presented in a simple and accessible way to laboratories, to those requesting analysis or anyone interested in this new analytical approach. We also address locks related to the specificity of non-targeted analyses, the development of methods and the reporting of results, in order to provide keys of understanding for the laboratories and the end-users of the results. This work is thus intended to be a technical guide to discover the benefits and opportunities of non-targeted analysis, and also a set of good practices for those who collect or analyse data obtained with such analytical technologies.; L’analyse non ciblée connaît une forte période de croissance dans le domaine de l’analyse environnementale, car elle ouvre le champ des investigations de la qualité des milieux à un très grand nombre de composés. Cet article consiste en un retour d’expérience de 12 laboratoires français sur les pratiques et les précautions à considérer lors de la réalisation d’analyses non ciblées, en particulier pour la recherche de composés suspects dans les échantillons issus des milieux aquatiques. En effet, si cette technique apporte des informations nouvelles et très prometteuses, elle n’en reste pas moins une technique complexe qu’il convient d’utiliser avec de nombreuses précautions pour assurer des résultats de qualité. La réflexion commune entreprise dans le groupe de laboratoires, a permis de synthétiser des informations techniques afin de présenter les enjeux de cette méthodologie d’intérêt majeur en analyse environnementale de façon simple et accessible aux laboratoires, demandeurs d’analyse ou toute personne intéressée par ce type d’approche. Les verrous liés à la spécificité des analyses non ciblées, au développement de méthodes et au rendu de résultats ont été développés pour fournir des clés de compréhension pour les laboratoires et les utilisateurs de résultats. Ce travail s’attache ainsi à être un guide de découverte de la technique pour les personnes désireuses de voir ce que peut apporter l’analyse non ciblée, mais également un guide de bonnes pratiques pour celles et ceux qui acquièrent ou qui analysent des données issues de ces techniques.

De l'évaluation des risques à l'action publique, Toulouse, , 04-/07/2018 - 06/07/2018; International audience; L'évolution de la production qualitative viticole, s'est faite durant plusieurs décennies par l'usage souvent excessif de produits phytosanitaires. L'objectif était la lutte contre les cryptogames, les ravageurs des plantes et la gestion de la concurrence des adventices concernant les éléments minéraux, l'eau, l'espace de développement. Ces pesticides, faciles à utiliser et très efficaces, ont été appliqués durant cette période de façon systématique suivant des recommandations, généralement à l'encontre de considérations agronomiques et écologiques. Mais l'usage bénéfique de ces molécules pour les agrosystèmes trouve aussi ses conséquences dans la contamination des écosystèmes et particulièrement les hydrosystèmes, ainsi que sur la santé humaine. Le projet pluridisciplinaire PhytoCOTE (agronomie, hydrobiologie, chimie environnementale, écotoxicologie, économie) étudie l'usage des produits phytosanitaires dans les agrosystèmes viticoles, leurs transferts dans les écosystèmes connexes (eau, air, sol), leur bioaccumulation et leur potentiel toxique chez les organismes aquatiques. Le site d'étude est localisé dans le Blayais, région viticole située sur la rive droite de l'estuaire de la Gironde. Les recherches ont d'abord permis d'évaluer la contribution de parcelles viticoles aux risques de contamination des eaux de surface par les produits phytosanitaires utilisés en viticulture, en tenant compte de critères descriptifs du milieu (sols, relief, connectivité des parcelles par rapport aux cours d'eau) et des pratiques viticoles (pression phytosanitaire, degré d'enherbement des parcelles, zones tampons). Des cartes de ces risques au niveau du parcellaire ont pu être établies. Parallèlement, un suivi annuel de la variabilité spatio-temporelle des concentrations en pesticides a été mis en place dans les différents compartiments environnementaux. Le suivi des cours d'eau permanents repose notamment sur le déploiement de capteurs passifs POCIS (Polar Organic Chemical Integrative Samplers) classiques à une fréquence mensuelle et de prélèvements d'eau ponctuels (bi-mensuels). L'analyse de la cartographie des risques, de la concentration en pesticides, de la modélisation des modes de conduite viticole (conventionnel, agrobiologie, agroécologie,...) et du comportement décisionnel des agriculteurs face aux choix des méthodes de protection phytosanitaire, devrait permettre d'établir des scenarii de changements de ces pratiques. Le but in fine est de proposer aux viticulteurs des mesures concrètes de nature agro-écologiques pour une réduction des usages de ces produits phytosanitaires et d'analyser comment ces mesures pourraient être influencées par les politiques publiques.

Wastewaters represent a major pathway of introduction of EDCs into the aquatic environment. Considering the large diversity of compounds and their possible transformation products, many EDCs are currently unknown. Therefore, characterization of the presence and identification of EDCs in wastewater are major issues in order to assess their occurrence in natural waters and the associated risks for wildlife. To date, in vitro assays based on luciferase reporter gene expression, are available to assess the biological activities of samples in a quantitative, sensitive, specific and fast way. The comparison of concentrations derived from bioassays and from chemical analyzes allows assessing the contribution of micropollutants to the overall biological or toxic activity of a sample. In this study, a systematic approach combining effect-directed analysis (EDA) and high resolution spectrometry was applied to several urban WWTPs to establish an overall contamination diagnostic and to identify major contaminant that could be released in environment. For this purpose, crude extract from influent and effluent of an urban WWTP were analyzed by LC-QTOF and tested on estrogens, androgens and glucocorticoids receptors. The non-target screening allowed detecting more than 7000 and 4000 signals in influent and effluent extracts of WWTP, respectively. We observed that 70% of compounds detected in effluent were produced by the treatment process suggesting the formation of transformation products. Concerning the strategy implemented to identify compounds of interest, it was decided to use effect-directed analyses methodology to have a tool to target active compounds in relation with selected biological activity. To this end, the crude extracts were fractionated by HPLC and biologically active fractions were isolated for further chemical identification. Biological fractionation profile of samples indicated the presence of estrogenic and glucocorticoid activities at all studied sites with very similar fractionation patterns between sites, highlighting major and recurrent individual fractions. The identification of active compounds was performed using LC-QTOF and several drugs and their transformation products (e.g. O-desmethyl venlafaxine, O-desmethyl tramadol) were recurrent found. This approach combined to LC-QTOF has allowed establishing a list of systematic detected non-target compounds in several wastewaters.

Aquatic ecosystems are widely contaminated by trace micropollutants that may represent a risk for human health and wildlife. Even if some compounds are well removed by wastewater treatments, the discharge of effluents remains one of the main entry of several micropollutants in the environment. It is particularly worrying for the compounds that are endocrine disruptors and/or that can induce genotoxicity. New approach using quantitative biological activities has to be applied along the water line, especially to monitor the pollution of wastewater. The objective of this study was to apply a panel of in vitro bioassays to determine water contamination profiles from the wastewater network to the outlet of the WWTP. A panel of bioassays were applied on various samples collected in six sites on the network, along the WWTP in order to evaluate the impact of biological treatment and tertiary ozonation. Selected in vitro bioassays, applied on water SPE extract, enabled detection of active chemicals responsible of endocrine disruption (estrogen, androgen, progestogen, mineralocorticoid, glucocorticoid, pregnane X and dioxin receptors) and the genotoxicity was evaluated by using the SOS Chromotest. In all sites along the wastewater network, ER, AR, MR activities and genotoxicity were detected. GR activity was measured at only three points of the network and was particularly important in the WWTP influent. Endocrine disruptor activities as well as genotoxicity were detected in raw wastewater (WWTP influent). Biological treatment highly reduced ED activities and genotoxicity. Nevertheless residual activities were still detected in the tertiary effluent. Results of the 1st sampling campaign allowed the identification of three notable points of interest on the network and at the WWTP influent. Results of two next sampling campaigns will help to confirm these “hot-spot” sites in order to apply effect-directed analysis to perform chemical identification of the compounds responsible of endocrine disruption activities and/or genotoxicity.
