Eclipta alba (L.) is a valuable medicinal plant. However, its medicinal efficacy can be affected by geminivirus infection. Therefore, identification of healthy specimen is essential before to use as medicine. The present study provided the taxonomic characterization of geminivirus infected and healthy E. alba plant by studying apparent morphology and microscopic features through light and scanning electron microscopy. Before taxonomic characterization infected and healthy specimens were separated through molecular detection of geminivirus. Results of morphological studies reported that geminivirus infected E. alba plant showed systematic symptoms of infection like stunted growth, distortion and chlorosis of leaves, decrease in size of root, shoot and fruit, and so forth in comparison to healthy specimen. Anatomical findings reported that in both plants anomocytic and anisocytic types of stomata with multicellular warty trichomes were present. However, variations were observed in quantitative measures such as size of trichomes, epidermal, subsidiary and guard cells. Palynological observations identifies that both plants possessed tricolporate type of pollen but variation was mainly observed in size and shape of pollen, thickness of exine and intine, P/E ratio, pore size, interspecific difference, size of colpi, and pollen ornamentation. Overall this study concluded that both healthy and infected E. alba do not reported much variations in qualitative taxonomic features, but can be differentiated in terms of quantitative taxonomic evidences. Future studies are recommended for pharmacological analysis of both healthy and virus infected plants. RESEARCH HIGHLIGHTS: Eclipta alba has incredible therapeutic worth, but due to geminivirus infection the plant is affecting badly. Hence, the present studies give a comprehensive taxonomic report on the geminivirus infected and healthy plant species.

The objective of this study was to investigate the nephroprotective effects of Daucus carota and Eclipta prostrata extracts on cisplatin-induced nephrotoxicity in Wistar albino rats. The study involved male Wistar albino rats of 8 weeks weighing 220–270 g. A single injection of 5 mg/kg was injected into the rats for nephrotoxicity. Environmental exposure of N-nitroso compounds (NOCs) from various sources like tobacco smoke, pesticides, smoked meat, and rubber manufacturing industries has been an alarming cause of carcinogenesis. Neonatal exposure to the carcinogenic N-N'ethylnitrosourea (ENU), a NOC has been established to cause leukemogenesis. Our world is constantly battling against cancer with consistent investigations of new anti-cancer therapeutics. Plant derived compounds have grasped worldwide attention of researchers for their promising anti-cancer potentials. Eclipta prostrata is one such ayurvedic herb, renowned for its anti-inflammatory properties. Currently, it has been explored in various cancer cell lines to establish its anti-cancer effect, but rarely in in-vivo cancer models. Wedelolactone (WDL), the major coumestan of E. prostrata is recognized as an inhibitor of IKK, a master regulator of the NF-kB inflammatory pathway. As persistent inflammation and activated inflammasome contribute to leukemogenesis, we tried to observe anti-leukemogenic efficacy of E. prostrata and its active compound WDL on the marrow cells of ENU induced experimental leukemic mice. Treatment groups were administered an oral gavage at a dose of 1200 mg/kg and 50 mg/kg b.w of crude extract and WDL respectively for 4 weeks. Various parameters like hemogram, survivability, cytological and histological investigations, migration assay, cell culture, flowcytometry and confocal microscopy were taken into consideration pre- and post-treatment. Interestingly, the plant concoction portrayed maximum effects in comparison to WDL alone. The study suggests E. prostrata and WDL as vital complementary adjuncts for anti-inflammasome mechanism in ENU-induced leukemia.

Objective: This research focused on the design of fast dissolving herbal film of Eclipta Prostrate leaves extract for mouth ulcers. Methods: The extract of Eclipta Prostrata leaves was formulated as films by solvent casting method using various polymers viz., HPMC E5, HPMC E15, sodium alginate and PVA. The films were designed by using propylene glycol as a plasticizer, SSG as super disintegrate and honey as a sweetener. Furthermore, the films were evaluated for thickness, folding endurance, weight variation, % elongation, surface pH, % moisture uptake, % moisture loss, disintegration and in vitro drug release study. Results: The revealed that all the films were good in appearance and had a smooth texture. Out of all ten formulations, F3 and F5 disintegrated rapidly with a disintegration time of 27 and 32 seconds. The drug release studies revealed that all the formulations had a good release profile, but the F3 formulation showed rapid release i.e. 83.57% in 4 min. The stability studies revealed that the formulations F3 and F5 were found good with non-tackiness, easily separable and disintegrated at 29 and 33 sec respectively with no appearance and drug release. Conclusion: The research revealed that Eclipta prostrate leaves extract can be formulated into oral films for the treatment of mouth ulcers with improved bioavailability and expected patient compliance.

The UV-B-induced signals play a crucial role in improving the analeptic values of medicinally important plants. Eclipta alba L. (Hassak), commonly known as False Daisy, holds supreme stature with its pharmaceutical association in treating various ailments, particularly in Ayurvedic medicine. The present study aimed to evaluate the response of E. alba plants exposed to ambient (AT) and two different supplemental UV-B doses (eUV-B, ambient ±7.2 kJ m-2 day-1), i.e., intermittent (IT) and continuous (CT) UV-B treatment for 130 and 240 h respectively. Antioxidative activities and medicinally important compounds (wedelolactone) were measured in different plants' parts at three growth stages. Under both the eUV-B treatments, the photosynthetic pigments were adversely affected (along with reductions in protein content) with a concomitant increase in secondary metabolites. Substantial variations in enzymatic antioxidants and non-enzymatic compounds showed the adaptive resilience strategies of plants against eUV-B. The wedelolactone content increased in leaves but compromised in stem and roots under IT. The results concluded that IT UV-B exposure led to the improvement of plant growth and the yield of wedelolactone compared to CT, suggesting its ameliorative role in improving the test plant's medicinal value.

The aim of this study was to evaluate of the density for four weeds on growth inhibition against soybean (Glycine max) (Chiang Mai 2 variety). The experiment were interfered with four weeds namely wild poinsettia (Euphorbia heterophylla), eclipta (Eclipta prostrata), slender amaranth (Amaranthus viridis) and spiny amaranth (Amaranthus spinosus) against soybean (G. max). The number of four weeds interfered with soybean planting divided into five treatments; 0 (control treatment) 4 8 12 and 16 plants/m2. Stem height, leaf area, fresh weight of aboveground part and dry weight of aboveground part of soybean were measured at 28 days after planting. The results showed that the increase of number of four weeds resulted in the increase of inhibition of all growth performance in soybean. The planting of soybean interfered with the weeds for 16 plants/m2 exhibited that the maximum of inhibitory degree (52.61%) followed by the planting of soybean interfered with the weeds for 12 8 and 4 plants/m2 by 40.04, 25.94 and 13.08%, respectively. The planting of soybean interfered with spiny amaranth (A. spinosus) exhibited that the highest of inhibitory degree (35.41%), followed by the planting of soybean interfered with eclipta (E. prostrata), wild poinsettia (E. heterophylla) and slender amaranth (A. viridis) showed the inhibitory degree for 34.35, 31.65 and 30.27%, respectively. This result presented the effects of density of weeds against soybean on growth parameters (stem height, leaf area, fresh weight of aboveground part and dry weight of aboveground part) as well as suggested that the planting of soybean should be the planting of soybean were not interfered with different weed density in order to the minimization of weed competition as well as the optimum method for weed arrangement.

Pathogenesis of Parkinson's disease (PD) specifically involves the degeneration of dopaminergic neurons in the substantia nigra region, which mainly begun with the overwhelmed oxidative stress and neuroinflammation. Considering the antioxidant and other pharmacological properties, Eclipta alba needs to be exploited for its possible neuroprotective efficacy against PD and other neurological disorders. Therefore, the current study was conducted to exemplify the remedial effects of hydro-alcoholic extract of E. alba (EA-MEx) against MPP+-elicited in vitro and in vivo PD models. SH-SY5Y, a neuroblastoma cell culture and male Wistar rats were used to impersonate the hallmarks of PD. Qualitative and quantitative analyses of EA-MEx revealed the presence of quercetin, ellagic acid, catechin, kaempferol, and epicatechin at varying concentrations. EA-MEx was found to deliver considerable protection against MPP+-induced oxidative damages in SH-SY5Y cells. Furthermore, in vivo study also supported the neuroprotective efficacy of EA-MEx, with significant mitigation of behavioral deficits induced by intrastriatal injection of MPP+. Furthermore, the disturbed levels of cellular antioxidant machinery have been significantly improved with the pre-treatment of EA-MEx. Mechanistically, the expression of α-synuclein, tyrosine hydroxylase, and mortalin were also found to be improved with the prior treatment of EA-MEx. Hence, the study suggests Eclipta alba as a suitable candidate for the development of better neuropathological therapeutics.

Ichthyophthiriasis, commonly known as white spot disease, occurs in both wild and cultured fish and is responsible for heavy economic losses to the aquaculture industry. In past decade, several chemical therapeutants were used to treat ichthyophthiriasis, but the effective drugs, such as malachite green, have been banned for use in food fish due to its genotoxic and carcinogenic properties. To find efficacious drugs to control Ichthyophthirius multifiliis (Ich), whole Eclipta alba plants and dried root of Arctium lappa were evaluated for their antiprotozoal activity. E. alba and A. lappa extracts significantly reduced the survival of Ich trophonts and theronts. In vitro, the E. alba and A. lappa methanol extracts killed all trophonts at 3200 mg l-1. All trophonts were killed after exposure to E. alba aqueous extract at 3200 mg l-1. The methanol extracts of E. alba and A. lappa killed 100% of I. multifiliis theronts at 400 mg l-1 and 800 mg l-1, respectively. The aqueous extract of E. alba and A. lappa killed 100% of I. mulitifiliis theronts at 1600 mg l-1 and 3200 mg l-1, respectively. E. alba and A. lappa extracts may be new and efficacious drugs for the control of ichthyophthiriasis.

Introduction The diversity and complexity of components are important reasons for the unstable efficacy and safety of Chinese materia medica (CMM) in quality control. The good and stable quality control may be related to the quality markers with structural composition of multi-components. Objective In the present study, we take Eclipta prostrata L. as a representative example. The 11 samples were collected from the different areas in China, and the discrepancy in bioactivity and chemical composition of these samples were compared. DOX (doxorubicin hydrochloride)-induced ICR mice were established for in vivo nephrotic syndrome experiments. The biochemical indicators including 24-h urine protein, triglyceride (TG), etc. were measured and the pathological change of kidney tissue was observed. MPC-5 cells damage model was induced to compare the difference of these samples in bio-activity. High-performance liquid chromatography (HPLC) method for 11 EEP (extract of Eclipta prostrata L.) samples were performed to analyse the content of the quality markers. Results In vivo experiments, EEP could mitigate the content or activity of urine protein, TG, etc. compared with the positive group (TG content was 2.53 ± 0.39 mmol/L, urinary protein quantification on 35th day was 16.79 ± 2.32 mg). In vitro experiments, CCE (coumestans component of Ecliptae) and EEP had equivalent effects on biochemical indicators such as cell viability, etc. According to the HPLC analysis, the content of wedelolactone was 45.88% and demethylwedelolactone was 23.74% in CCE. Conclusion The CCE with a ratio of 2:1 can be considered as a quality marker of Eclipta prostrata L.. This research may provide a perspective for quality control of CMM.

The aim of this study is to evaluate the antioxidant activity, α-amylase inhibition activity, estimation of total phenolic and flavonoid content and the toxicity in ten medicinal plants Woodfordia fructicosa, Tectaria coadunate, Prunus cerasoides, Abrus precatorius, Eclipta prostrate, Poranopsis paniculata, Chenopodium album, Oroxylum indicum, Curcuma caesia, and Butea monosperma collected from Gorkha District of Nepal. Methanolic extracts of all the plants showed the presence of different phytoconstituents such as alkaloids, polyphenols, flavonoids, terpenoids, saponins, glycosides, and quinones. The highest radical scavenging was observed in methanol extract of P. cerasoides with IC50 = 7.54±0.223 µg/ml. The potency of the radical scavenging effect of P. cerasoides was about six times greater than standard ascorbic acid (39.85±0.025 µg/ml) taken. P. cerasoides showed high phenol content (805.48±0.024 mg GAE/g extract) whereas total flavonoid content varied from O. indicum (16.96±0.015 mg QE/g extract) to W. fructicosa (722.76±0.108 mg QE/g extract). The methanol extract of E. prostrate was found to be toxic against brine shrimp as shown in the LC50 value of 6.3 µg/ml. T. coadunate and A. precatorius showed effective results with an IC50 value of 80.89 and 70.29 µg/ml respectively in α-amylase inhibition test. This study provides some scientific support for traditional uses of plants for diabetes management and other ailments. Since extracts of W. fructicosa and P. cerasoides are rich sources of bioactive chemical constituents, further in-vitro and in-vivo bioactivity of these extracts need to be studied for their exact mechanism of action. Eclipta prostrata has been extensively studied for its promoting effect on human health. Previous study proved that E. prostrata possessed anti-osteoporosis function in SAMP6 mice via gut microbiota (GM). Endophytes are widespread in plants, especially in Chinese herbal medicines. However, little is known regarding the endophytes of E. prostrata. In this study, we focus on screening and identifying the endophytes from plant E. prostrata and exploring their roles in modulating GM. According to biochemical, physiological tests and 16S rDNA sequence analysis, ten endophytes were characterized in different locations of plant E. prostrata belonging to Lactococcus, Bacillus, Enterococcus, Exiguobacterium and Pantoea. The antimicrobial activity of endophytes EP01-10 was investigated via the oxford cup method. Furthermore, the acid and bile salt resistant ability of EP01-10 was detected to explore their survival ability in gastrointestinal tract. Results indicated that strains of Lactococcus, Bacillus and Exiguobacterium (EP01, 03 and 05) showed strong antimicrobial activity against Escherichia coli and Staphylococcus aureus and strains of Lactococcus, Bacillus and Enterococcus (EP01, 02, 04, 08 and 10) have the ability to survive in the gastrointestinal tract. 16S rRNA sequencing of GM in C57BL/6J mice was performed for diversity and structure analysis responded to the administration of E. prostrata extract. E. prostrata extract acted on improving the microflora diversity, particularly in increasing the abundance of Lactococcus significantly. Thus, as an endophyte, Lactococcus plays an important role in E. prostrata modulating intestinal tract. Our study suggested that endophytes of Chinese herbal medicines might be a novel target for the treatment of diseases by regulating the GM.

Atopic dermatitis (AD) is a kind of inflammation on the skin following with swollen, itchy, dryness and cracked skin. Though the exact cause of AD is unknown, there are evidence that people with AD have a compromised skin barrier along with inflammation. Eclipta prostrata Linné is a traditional herbal medicinal plant, has been used for the diabetes, obesity, jaundice, and inflammation. We supposed E. prostrata L. has an anti-inflammatory effect on the skin.We aimed to assess the effect of E. prostrata L. EtOH extract (EP) and elucidate the associated molecular mechanisms.The effect of EP and the molecular mechanisms were eluciated in house dust mite (HDM)-induced AD mice model and TNF-α/IFN-γ-stimulated HaCaT keratinocytes by histological analysis, enzyme-linked immunosorbent assay, quantitative real time polymerase chain reaction, and Western blot.The results revealed that EP improved the progression of AD symptoms, decreasing epidermis/dermis thickness, infiltrated immune cells, and restored the skin barrier dysfunction and imbalanced immune response. EP suppressed the expressions of T helper (Th)1, Th2, Th17 cytokines, phosphorylation of extracellular signal-regulated kinase/signal transducer and activator of transcription 1 in skin of HDM-induced AD mice as well as inhibition the translocation of nuclear factor-κB in HaCaT keratinocytes.Collectively, EP improved the allergic inflammation of the skin through recovery the skin barrier, and regulation the immune balance. These results suggest EP may have therapeutic potential as an anti-atopic agent.

Benign prostatic hyperplasia (BPH) is a urinary system disease with high prevalence among the middle and elder men. In BPH, proliferation of prostate cells and the imbanlance between androgen and estrogen are both important inducers. Previous studies have demonstrated that compounds from Ligustri Lucidi Fructus (LLF) and Ecliptae Herba (EH) are of phytoestrogenic or phytoandrogenic activities. The combination of LLF with EH at the ratio of 1:1 on crude drugs quantity is called Erzhi formula (EZF), which is used for in vivo research of our study.This study aimed to investigate potential mechanisms of EZF and its active pharmaceutical ingredients on BPH in vitro and in vivo.Therapeutic effects of EZF was evaluated in EEZF treatment significantly attenuated rat prostate enlargement, alleviated BPH pathological features, and decreased the expression of PCNA. The up-regulation of AR, ERα, SRD5A1/2 expressions, and down-regulation of ERβ expression at prostate of rat BPH model were significantly blocked by EZF administration. The expression levels of testosterone, DHT, EEZF, ligustrosidic acid from LLF and echinocystic acid from EH showed inhibitive effects on BPH via down-regulating prostatic AR, ERα, SRD5A1/2 expressions and up-regulating ERβ expression.

Ecliptae prostrata (L.) L. has been widely used in East Asia with reported biological activities, including anti-cancer properties.We aimed to investigate the effect of ethyl acetate extract of Ecliptae prostrata (L.) L. (EAE) and its component wedelolactone on the proliferation and migration of head and neck squamous cancer cells.The proliferation of human SCC-4 and mouse CU110-1 tongue squamous carcinoma cells was assessed using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide (MTT) method. Scratch wound assays were performed to assess cell migration rates. The levels of Ecadherin and vimentin were used as markers of the epithelial-to-mesenchymal transition (EMT). AhR, CYP1A1, and CYP1B1 levels were examined to uncover the mechanism of inhibition of cell migration by wedelolactone.We found that EAE and wedelolactone decreased the proliferation of human SCC-4 cells and mouse CU110-1 cells at doses of EAE at25 μg/ml and wedelolactone at6.25 μg/ml. Similarly, both EAE and wedelolactone produced inhibitory effects against migration, but the effective doses that significantly inhibited migration were lower than those affecting proliferation. Wedelolactone below 12.5 μg/ml inhibited the epithelial-to-mesenchymal transition (EMT) with increased expression of E-cadherin and decreased expression of vimentin in SCC-4 and CU110-1 cells. Further analysis showed wedelolactone inhibited the expression of AhR and its downstream target molecules CYP1A1 and CYP1B1 in both squamous carcinoma cells at the same doses inhibiting cell migration. The addition of benzo (a)pyrene [B(a)P], an agonist of AhR, stimulated migration, especially in the CU110-1 cells with reported cancer stem cell-like characteristics. Instructively, B(a)P reversed the inhibitory effects of wedelolactone on AhR expression and cell migration, suggesting that wedelolactone antagonizes cell migration through the AhR pathway. Moreover, the higher activity of EAE and wedelolactone against the migration of cancer stem-like CU110-1 cells relative to SCC-4 cells suggests selective activity against cancer stem cells.Our study identifies wedelolactone as a major active component of Ecliptae prostrata (L.) L. with promising anti-cancer properties against head and neck squamous cancer cells.

Two field experiments were conducted at the Experimental Farm of Sakha Agricultural Research Station, Kafrelsheikh, Egypt during 2016 and 2017 summer seasons to study the effect of seedling age and weed management on growth and yield of transplanted rice (Sakha 106 cv). A split plot experimental design arranged in randomized complete block with four replications was used in both seasons. Three seedling ages viz., 21, 28 and 35 days were allocated to the main plots, while five weed control treatments i.e., thiobencarb (50% EC) at 2.39 kg ai ha-1, oxadiazon (25% EC) at 0.045 kg ai ha-1, penoxsulam (24% SC) at 0.029 kg.ai ha-1, hand weeding two times (20 and 40 DAT) and untreated (weedy check) were assigned to the sub-plots. The results showed transplanting rice seedlings at 21 days old recorded the lowest dry weight of Eclipta sp C. difformis E crus-galli and total weeds. Also, the increasing in rice dry weight, number of panicles m-2, panicle weight, number of filled grains panicle-1, 1000-grain weight and rice grain yield as compared to rest seedling ages (28 and 35 days). The application of penoxsulam at 0.029 kg ai ha-1 applied at 15 DAT achieved the lowest dry weight of weed species and total weeds as well as the highest rice yield as compared to weedy check. The interaction between seedling ages and weed control treatments had significant effect on dry weights of weed species, total weeds dry, rice yield and its components. Where, transplanting rice seedlings treatment at 21 days old treated with herbicide of penoxsulam at 0.029 kg ai ha-1 at 15 DAT are the best among the studied treatments to achieve the best weed control and the highest rice dry mater, grain yield and its components for Sakha 106 cv. under this study conditions.

Weed management in container crops is primarily accomplished through frequent PRE herbicide applications and supplemental hand weeding. However, many ornamental species are sensitive to herbicides, and a significant number of tropical plants, ornamental grasses, and foliage crops have not been screened for herbicide tolerance. As nursery crops are produced in inert substrates that are largely composed of bark or peat, strategic fertilizer placement has the potential to significantly reduce weed growth in container-grown ornamentals. Growth and reproduction of three common container nursery weed species, eclipta [Eclipta prostrata(L.) L.], large crabgrass [Digitaria sanguinalis(L.) Scop.], and spotted spurge (Euphorbia maculataL.), were evaluated following fertilization via alternative methods, including subdressing or dibbling in comparison with industry standard practices of topdressing or incorporating a controlled-release fertilizer (17-5-11 [8 to 9 mo.]) to each 3.8-L container at 36.5 g per container. Fertilizer placement had little to no effect on germination ofEclipta prostrataorD. sanguinalis, but incorporation increasedE. maculatagermination by 77% to 183% compared with other placements or a nonfertilized control. Subdressing reduced seed production by 94%, 63%, and 92% forEclipta prostrata,D. sanguinalis, andE. maculata, respectively, compared with the average number of seeds produced in the conventional placement methods (average of incorporation and topdressing). Dibbling fertilizer resulted in similar decreases in the case ofD. sanguinalisandE. maculata, whileEclipta prostrataproduced no seeds when fertilizer was dibbled. Similar to reductions observed in reproduction, subdressing fertilizer resulted in biomass decreases of 90%, 81%, and 85% compared with the average biomass of the incorporation and topdressed placements. Results suggest alternative fertilizer placements could be implemented as part of an integrated weed management program in container production to reduce weed growth.

Row-middle weed control in Florida vegetable production is challenging and often necessitates several PRE and POST herbicide applications. Coordinating POST spray applications with emergence timings should increase herbicide efficacy by targeting susceptible growth stages. Most published emergence models were developed in temperate climates, and adapting them to subtropical climates can be complex and requires reductionist insights into seed ecology, particularly germination and dormancy. The study objective was to examine the influence of temperature and osmotic potential on seed germination of carpetweed (Mollugo verticillata L.), Carolina geranium (Geranium carolinianum L.), eclipta [Eclipta prostrata (L.) L.], and goosegrass [Eleusine indica (L.) Gaertn.]. Mollugo verticillata seed germination was positively photoblastic, with increased germination at high temperatures (≥35 C), more so with high fluctuating temperatures (35/20 and 35/25 C), and occurred at osmotic potentials as low as −0.5 MPa. Geranium carolinianum seed germinated between 10 and 20 C in light or darkness and at osmotic potentials as low as −0.4 MPa. Eclipta prostrata seed germination was entirely positively photoblastic, occurring optimally between 15 and 25 C and at osmotic potentials as low as −1 MPa. Eleusine indica seed germination demonstrated some degree of positive photoblasticity, with greater germination in the light, peak germination at 35 C, and germination occurring at osmotic potentials as low as −0.5 MPa. Described germination ecology for selected species will provide insights for building ecology-based growing degree-day accounting restrictions for empirically derived emergence models.

Lead (Pb) contamination of soil is a serious environmental problem, adversely affecting ecosystems, globally. Phytoremediation is an alternative to conventional methods of soil remediation. The success of phytoremediation depends on the identification of suitable native plant species with high biomass to deal with metal contamination. In the present experiment, response of Eclipta prostrata (L.) L., Scoparia dulcis L. and Phyllanthus niruri L. to increase in concentrations of PbNO3·5H2O in the soil for a period of 30 days was tested to assess their suitability in phytoremediation. Pb accumulation in all the three plants was in a concentration-dependent manner. Although S. dulcis survived the soil metal concentrations, it exhibited a stunted growth; P. niruri was found susceptible to Pb toxicity; E. prostrata recorded a maximum uptake of 12484 µg/g dry weight in its root and 7229 µg/g dry weight in its shoot, without any adverse impact on growth traits. Bioconcentration factor and translocation factor of the three plants were also calculated, which revealed that E. prostrata has Pb accumulation potential. Therefore, enzymatic antioxidant activities and transmission electron microscopic analysis were carried out to determine the physiological adaptation and tolerance of E. prostrata to Pb stress. Overall, E. prostrata is identified as a tolerant plant showing Pb hyperaccumulation tendencies with essential features for phytoextraction.

Purpose. Traditional Chinese medicine (TCM) has been extensively studied for its preventive and treatment properties toward osteoporosis (OP). Pharmacological studies have shown that TCM Eclipta prostrata induce anti-OP effects. Considering the growing evidence demonstrating that gut microbiota (GM) is related to OP, we aimed to study the GM-dependent function and mechanism of E. prostrata for preventing OP in mice. Methodology. Bone micro-structure was obtained using micro-computed tomography (micro-CT) and bone-relating factors were detected by molecular biological test. High-throughput sequencing of the 16S rRNA V4 region was performed for GM diversity analysis. Growth effects of E. prostrata on potential targeted strains Lactobacillus and Lactococcus were investigated by in vitro bacterial assay. By feeding Lactobacillus and Lactococcus in mice, GM and bone condition were analysed. Results. Bone micro-structure was significantly improved by E. prostrata with a potential mechanism of inhibiting osteoclast, increasing the number of osteoblasts and regulating the dynamic balance of bone absorption and formation. Sequencing results indicated that E. prostrata altered the bacterial community. The abundance of bacteria genera Lactobacillus and Lactococcus was markedly decreased in individuals with OP and positively correlated with high dose of E. prostrata. GM of the low-dose E. prostrata-fed group did not significantly differ from that of the chow-fed OP group, which was consistent with bone structure test results. Moreover, E. prostrata could promote Lactobacillus and Lactococcus growth in vitro. GM was altered and bone condition was improved via bacterial feeding in vivo. Conclusion. Our findings suggested that E. prostrata might be a novel therapy for OP prevention by targeting GM.

Eclipta prostrata has long been used as a medicinal herb in China. EAP20-1, a homogeneous polysaccharide with anti-complementary activity had been obtained from E. prostrate by using anion-exchange and size-exclusion chromatography. In this study, we found that EAP20-1 could inhibit in vitro lymphocyte proliferation stimulated by concanavalin–A or anti-CD3/anti-CD28 antibodies. Furthermore, in experimental autoimmune encephalomyelitis (EAE) mice, EAP20-1 treatment relieved the clinical symptoms, accompanied by reduced neuroinflammation and demyelination in spinal cords. Mechanistically, EAP20-1 reduced the mRNA expression of interleukin (IL)-17, IL-22, and RAR-related orphan receptor gamma t (RORγt) in the spleen; inhibited auto-reactive T cell proliferation and decreased the percentage of Th17 cells in response to myelin oligodendrocyte glycoprotein (MOG35-55) ex vivo. Moreover, EAP20-1 directly inhibited naive CD4 + T cells differentiate into Th17 cells in vitro. These results indicating EAP20-1 could benefit EAE through inhibiting Th17 cell differentiation and suggesting a therapeutic potential of EAP20-1 in MS.

Eclipta prostata L. commonly known as Bhringaraj is one of the most valuable and important medicinal herb. The whole plants are used in medicinal and cosmetic industries. But still it is considered as a wild weed due to lack of propagation technique. Vegetative propagation plays an important role for its commercial cultivation. The experiment on “Rooting behavior of stem cuttings of Eclipta prostata L. with varying concentrations of different plant hormones” was laid out under CRD design with 13 treatments and 3 replications at demonstration unit of KVK Tirap, Deomali, Arunachal Pradesh. From this experiment it could be concluded that IBA treated cuttings irrespective of concentration was the best for rooting in Eclipta prostata cuttings under climatic condition of Tirap . The highest percentage of rooting, number of roots, survivability percentage and minimum days to rooting were recorded in IBA 400 ppm, IBA 300 ppm, IBA 100 ppm and IBA 400 ppm treated cuttings respectively.

Erzhi formula (EZF) consists of Ecliptae herba (EH) and Fructus Ligustri Lucidi (FLL) at a ratio 1:1, and constitutes a well-known formula in China that is commonly used for treating menopausal diseases.In this study, we explored the pharmacologic actions and potential molecular mechanisms underlying EZF's action in preventing and treating osteoporosis.The active components and related targets of EZF's anti-osteoporotic effects were predicted by network pharmacology, and functional enrichment analysis was also performed. We then used an osteoporosis model of ovariectomized (OVX) mice to detect the effects of EZF on osteoporosis.The results from network pharmacology identified a total of 10 active ingredients from EH and 13 active ingredients from FLL that might affect 65 potential therapeutic targets. GO enrichment analysis revealed that EZF affected bone tissue primarily via hormone (particularly estradiol)-related pathways and bone resorption by osteoclast differentiation. KEGG analysis demonstrated that bone-related factors such as Runt-related transcription factor 2 (Runx2), Ca2, estrogen receptor1 (ESR1), androgen receptors (AR), and TNFα served as the primary targets during osteoclastic differentiation. In vivo experiments showed that the formula significantly improved the diminution in estrogen and the subsequent uterine atrophy induced by ovariectomy (P 0.01 or 0.05), implying that the EZF exerted its actions via regulation of estradiol and the nourishing effects of the uterus in OVX mice. Dual-energy X-ray absorptiometry and micro-CT showed that EZF significantly inhibited bone loss and improved bone micro-architecture by statistically increasing the number of bone trabeculae and decreasing the separation of bone trabeculae in OVX mice (P 0.01 or 0.05); EZF also inhibited bone loss and enhanced bone-fracture load. Furthermore, we confirmed that EZF reduced the calcium concentrations, augmented protein and mRNA levels for Runx2 in the bone marrow, and reduced PPARγ levels. RANKL-a key downstream regulatory protein of many targets that was referred to in our results of network pharmacology as being involved in the regulation of osteoclastogenesis-was significantly diminished by EZF; it also elevated OPG content. In addition, we used monocytes of bone-marrow origin to detect the effects of the potential components of EZF on osteoclast differentiation and found that wedelolactone, oleanolic acid, echinocystic acid, luteolin, and luteolin-7-o-glucoside significantly inhibited osteoclast differentiation from monocytes induced by 25 ng/mL MCSF and 50 ng/mL RANKL (P 0.01 or 0.05).Our present study indicated that EZF significantly inhibited the bone loss induced by OVX in mice by its regulation of estradiol combined with the nourishing effect of the uterus, and that it also attenuated bone resorption by decreasing the RANKL/OPG ratio so as to inhibit osteoclast maturation.

BackgroundsColon cancer is the third most deadly and one of the most diagnosed diseases in the world. Although routine screening and early detection during last decades has improved the survival, colon cancer still claims hundreds of thousands lives each year worldwide. Surgery and chemotherapy is mainstay of current treatment, nevertheless toxicity associated with this treatment underscores the urgency of demand of a better therapeutics. Close to 50% of current chemotherapeutic drugs are direct or indirect descendants compounds isolated from medicinal plants, which indicate plants are great potential sources of novel therapeutics. In our literature review we foundEclipta albato posses many pharmacological activities, including those with anticancer potentials. However, no study on anticancer activity of this kind has been reported.MethodsPhytochemicals were extracted by maceration method from shade dried whole plant ofEclipta albausing methanol as a solvent. The anticancer effect of extract was investigated on various cancer cell lines like human colorectal carcinoma (HCT-116), human prostate cancer (PC-3), Michigan cancer foundation-breast cancer (MCF-7) and renal cell carcinoma (RCC-45). A feeding trial that lasted for 90 days was carried out to determine the effect of dietary supplementation of Eclipa alba and Polyalthia longifolia foliages on digestibility, feeding behaviour, blood profile and carcass characteristics of goats. A total of thirty two West African dwarf goats of about 7–8 months old were randomly assigned to four treatment diets. Each treatment was replicated four times with two goats per replicate in a completely randomized design. The four formulated treatment diets were, D1 (90% guinea grass with 10% maize straw), D2 (50% guinea grass with 10% maize straw and 40% Eclipta alba foliage), D3 (50% guinea grass with 10% maize straw 20% Eclipta alba with 20% Polyalthia longifolia foliages) and D4 (50% guinea grass with 10% maize straw and 40% Polyalthia longifolia foliage). The results indicated that goats on D1 were significantly (P < 0.05) highest in eating rate, feed conversion ratio, urinary-N-output, urea and creatinine while cholesterol was greater in goats on D2. Crude fiber and ether extract digestibility with white blood cell was significantly (P < 0.05) higher for goats on D3 than other treatment diets. Feed intake, ruminating rate, live weight gain, dry matter with crude protein digestibility, nitrogen intake with retention, red blood cell, total protein, dressing weight and stomach were significantly higher (P < 0.05) in goats on D4 compared with those on D1, D2 and D3. Eating with ruminating time, initial weight, faecal-N-output and most of the carcass cuts with relative organs showed no significant different (P > 0.05) among treatment diets. It could therefore be concluded that supplementation with Eclipta alba and Polyalthia longifolia foliages or their mixture in diets enhance digestibility, feeding behaviour, blood profile and carcass characteristics without any deleterious effect in goats.

To investigate the role of Eclipta prostrata (E. prostrata) extract in improving spatial learning and memory deficits in D-galactose-induced aging in rats.Rats were divided into five groups, with 10 animals in each group. Aging rats were produced by treatment with 100 mg·kg-1·d-1 of D-galactose for 6 weeks. Rats in the E. prostrata treatment groups received an aqueous extract of E. prostrata orally at a concentration of 50, 100, or 200 mg·kg-1·d-1 for 3 weeks. Animals in both the normal and model groups were treated with similar volumes of saline. Spatial memory performance was measured using the Morris water maze. The mRNA levels and enzyme activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) were analyzed using real-time quantitative PCR and spectrophotometry, respectively. The levels of induced nitric oxide synthase (iNOS), nitric oxide (NO), dopamine (DA), norepinephrine (NE), and serotonin (5-HT) were determined using enzyme-linked immunosorbent assay and spectrophotometry.Compared with the normal group, rats in the D-galactose-treated model group exhibited significant memory loss. There was severe damage to the hippocampal CA1 area, and expression levels of SOD, CAT, GPx, and GR were significantly decreased in the model group compared with the normal group. In the model group, levels of iNOS and NO were significantly increased compared with the normal group. However, treatment with E. prostrata extract reversed the conditions caused by D-galactose-induced aging, especially in the groups with higher treatment concentrations. Compared with the normal group, the levels of DA, NE, and 5-HT were significantly lower in the D-galactose-treated model group. In the E. prostrata extract-treated groups, however, there was a dose-dependent upregulation of DA, NE, and 5-HT expression.Our results suggest that administration of E. prostrata extract can result in an improvement in the learning and memory impairments that are induced by D-galactose treatment in rats. This improvement may be the result of enhanced antioxidative ability, decreased iNOS and NO levels, and the induction of DA, NE, and 5-HT expression in the brain.

Eclipta alba L., also known as false daisy, is well known and commercially attractive plant with excellent hepatotoxic and antidiabetic activities. Light is considered a key modulator in plant morphogenesis and survival by regulating important physiological cascades. Current study was carried out to investigate growth and developmental aspects of E. alba under differential effect of multispectral lights. In vitro derived callus culture of E. alba was exposed to multispectral monochromatic lights under controlled aseptic conditions. Maximum dry weight was recorded in culture grown under red light (11.2 g/L) whereas negative effect was observed under exposure of yellow light on callus growth (4.87 g/L). Furthermore, red light significantly enhanced phenolics and flavonoids content (TPC: 57.8 mg/g, TFC: 11.1 mg/g) in callus cultures compared to rest of lights. HPLC analysis further confirmed highest accumulation of four major compounds i.e. coumarin (1.26 mg/g), eclalbatin (5.00 mg/g), wedelolactone (32.54 mg/g) and demethylwedelolactone (23.67 mg/g) and two minor compounds (β-amyrin: 0.38 mg/g, luteolin: 0.39 mg/g) in red light treated culture whereas stigmasterol was found optimum (0.22 mg/g) under blue light. In vitro based biological activities including antioxidant, antidiabetic and lipase inhibitory assays showed optimum values in cultures exposed to red light, suggesting crucial role of these phytochemicals in the enhancement of the therapeutic potential of E. alba. These results clearly revealed that the use of multispectral lights in in vitro cultures could be an effective strategy for enhanced production of phytochemicals.
