This study aims to assess the chemical composition of the aqueous extract of Cistus albidus L. leaves, as well as the potential of aqueous and hydroethanol extracts of the leaves and seeds as analgesic, anti--inflammatory, and antioxidant agents. The contents of phenolics and inorganic constituents were determined in C. albidus seeds and leaves; antioxidant capacity was assessed by 3 complementary and diverse tests. The carrageenan-induced paw edema technique was used to investigate the anti-inflammatory effect in vivo, and albumin denaturation to evaluate the anti-inflammatory effect in vitro. The acetic acid-induced contortion test, the tail-flick test, and the plantar test were used to assess the analgesic effi cacy in vivo.

Cytisus villosus Pourr. (C. villosus) is a medicinal plant belonging to the Fabaceae family, which grows in the Mediterranean area. It is used in traditional medicine against diseases related to inflammation. The objective of the present study was to identify the mineral and polyphenolic composition as well as to evaluate some biological properties including antioxidant, anti-inflammatory, and analgesic activities of C. villosus leaf aqueous extract. The chemical constituents were identified and quantified using ultra performance liquid chromatography-electrospray ionization tandem mass spectrometry (UPLC-ESI-MS/MS) methods. The antioxidant properties of C. villosus leaves were tested using reducing power (RP), 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS), and 2,2′-diphenyl-1-picrylhydrazyl (DPPH) assays. The anti-inflammatory potency was evaluated in vitro and in vivo using the albumin denaturation test and the carrageenan test, respectively. Furthermore, the analgesic effect was performed in vivo using tail flick, acetic acid-induced contortion, and plantar tests. Mineralogical analysis revealed that potassium and calcium were the most abundant minerals. The analysis and quantification of the phytochemical composition using UPLC-ESI-MS/MS showed that quinic acid (57.478 ± 1.72 mg/kg) was the major compound of the aqueous extract, followed by salicylic acid (17.38 ± 0.2 mg/kg), isoquercetin (16.895 ± 1.01 mg/kg), and gallic acid (15.914 ± 1.51 mg/kg). The extracts showed potent antioxidant activity for all tests used. The highest antioxidant activity was recorded for the DPPH, ABTS and RP methods, with an IC50 of 3.94 ± 0.09, 2.88 ± 0.07, and 1.94 ± 0.10 μg/mL, respectively. Additionally, using the most frequent analgesic assays, the aqueous extract at a dose of 500 mg/kg exhibited a potent analgesic activity. Notably, an interesting inhibition of albumin denaturation was recorded with an IC50 of 383.94 μg/mL, corroborating the in vivo test. Overall, the results presented here may represent a scientific basis for the traditional use of C. villosus in the treatment of inflammation-related diseases.

Inhibin is a dimeric glycoprotein comprised of two subunits, α and β. Immunization against dimeric inhibin is mainly used in assisted reproductive technology to induce superovulation. However, the specific function of immunoreactive-free inhibin α subunit remains unclear. In this study, two main investigations were conducted (first on ovulation and the other on fertilization) using a novel monoclonal antibody targeting free inhibin α subunit (Pro-αN-αC). The ovulation study was conducted in 6 replicates, involving a total of 48 female CD1 mice aged 4-6 weeks. In each replicate, 4 control mice received PMSG/hCG treatment, and 4 treated mice received PMSG/hCG with mAb-Free α subunit. The fertilization study was conducted in 3 replicates, involving a total of 22 female CD1 mice. In each replicate, there were 4, 3, and 4 mice respectively for both control and treatment groups. In both investigations, female mice were injected intraperitoneally with 50 units/ml of Pregnant Mare Serum Gonadotropin (PMSG), alone or combined with 400ug of mAb-Free α subunit, followed by an injection of 50 units/ml of Human Chorionic Gonadotropin (hCG) 48 hours later. Seventeen hours post-injection, the females from all groups were sacrificed, and the ovulated oocytes were collected from the oviducts. For the fertilization study, in vitro fertilization was performed using fresh sperm from male CD1 mice. The results revealed that neutralization of the free inhibin α subunit significantly decreased the ovulation rate by 47.29% compared to the control group, while immunoneutralization significantly increased the fertilization rate by 55.68% and the blastocyst development by 43.85% compared to the control group. This study suggests that immunization against free inhibin α subunit decreases ovulation, in contrast to the effect of immunoneutralization of dimeric inhibin. Abstract Background Animal vaccination is an important way to stop the spread of diseases causing immense damage to livestock and economic losses and the potential transmission to humans. Therefore effective method for vaccine production using simple and inexpensive bioprocessing solutions is very essential. Conventional culture systems currently in use, tend to be uneconomic in terms of labor and time involved. Besides, they offer a limited surface area for growth of cells. In this study, the CelCradle™-500A was evaluated as an alternative to replace conventional culture systems in use such as Cell factories for the production of viral vaccines against small ruminant morbillivirus (PPR), rift valley fever virus (RVF) and lumpy skin disease virus (LSD). Results Two types of cells Vero and primary Lamb Testis cells were used to produce these viruses. The study was done in 2 phases as a) optimization of cell growth and b) virus cultivation. Vero cells could be grown to significantly higher cell densities of 3.04 × 109 using the CelCradle™-500A with a shorter doubling time as compared to 9.45 × 108 cells in Cell factories. This represents a 19 fold increase in cell numbers as compared to seeding vs only 3.7 fold in Cell factories. LT cells achieved modestly higher cell densities of 6.7 × 108 as compared to 6.3 × 108 in Cell factories. The fold change in densities for these cells was 3 fold in the CelCradle™-500A vs 2.5 fold in Cell factories. The titers in the conventional system and the bioreactor were not significantly different. However, the Cell-specific virus yield for rift valley fever virus and lumpy skin disease virus are higher (25 virions/cell for rift valley fever virus, and 21.9 virions/cell for lumpy skin disease virus versus 19.9 virions/cell for rift valley fever virus and 10 virions/cell for lumpy skin disease virus). Conclusions This work represents a novel study for primary lamb testis cell culture in CellCradle™-500A bioreactors. In addition, on account of the high cell densities obtained and the linear scalability the titers could be further optimized using other culture process such us perfusion.

Understanding the role of humans in the dispersal of predominantly animal pathogens is essential for their control. We used newly developed Bayesian phylogeographic methods to unravel the dynamics and determinants of the spread of dog rabies virus (RABV) in North Africa. Each of the countries studied exhibited largely disconnected spatial dynamics with major geopolitical boundaries acting as barriers to gene flow. Road distances proved to be better predictors of the movement of dog RABV than accessibility or raw geographical distance, with occasional long distance and rapid spread within each of these countries. Using simulations that bridge phylodynamics and spatial epidemiology, we demonstrate that the contemporary viral distribution extends beyond that expected for RABV transmission in African dog populations. These results are strongly supportive of human-mediated dispersal, and demonstrate how an integrated phylogeographic approach will turn viral genetic data into a powerful asset for characterizing, predicting, and potentially controlling the spatial spread of pathogens.

Mycoplasma capricolum subsp. capricolum (Mcc) and Mycoplasma capricolum subsp. capripneumoniae (Mccp) are pathogens that affect large and small ruminants. Indeed, Mcc affects both sheep and goats, causing contagious agalactia (CA). Mccp affects only goats, causing contagious caprine pleuropneumonia (CCPP). CA and CCPP are mainly controlled using inactivated Mcc and Mccp vaccines. However, producing the vaccine with the Mccp strain is complex, fastidious, and costly due to the slow growth of the bacterium. In this study, we present new oil-adjuvanted and inactivated Mcc and Mccp vaccines for sheep and goats against CA and CCPP. The vaccines were evaluated for safety and efficacy using experimental infection. A serological response was observed one week after of the first vaccination of sheep and goats with Mcc and goats with Mccp. The vaccinated animals were subsequently challenged with the virulent Mcc MOR20 strain. The Mcc vaccine was demonstrated to provide robust protection when the animals were challenged with Mcc MOR20. Cross-protection against the Mcc MOR20 challenge was also obtained with the Mccp vaccine. This finding revealed, for the first time, the safety and efficacy of an inactivated Mcc vaccine against contagious agalactia and cross-protection between Mcc and Mccp strains. Abstract Background Peste des petits ruminants (PPR) is a highly contagious viral disease of small ruminants in Asia and Africa. In 2008, a PPR outbreak was reported for the first time in Morocco and a mass vaccination campaign allowed control of the disease. In this study, the susceptibility of four Moroccan local breeds of small ruminants to PPR virus was investigated by experimental infections. The objective was to make recommendations for improved epidemiological surveillance in Morocco by evaluating the susceptibility of the dominant Moroccan small ruminant breeds. Three parameters were studied: hyperthermia, clinical scoring and virus excretion. The outcome was compared to Alpine goats, which are considered one of the most sensitive breeds. Results The study showed that the local goat breed was the most sensitive breed with a susceptibility rate of 67%, followed by Timahdit, Beni Guil and Sardi sheep with 48, 29 and 26%, respectively. Serological testing including enzyme-linked immunosorbent assay and viral neutralization showed that the Timahdit breed developed a stronger antibody response compared to the other breeds. Although the clinical signs observed in the sheep were mild, evidence of viral excretion was detected by means of a polymerase chain reaction assay. Conclusions It is recommended that effective surveillance should focus on susceptible breeds complemented with serological surveillance of the sheep population.
