Matematikos ir gamtos fakulteto ištakomis Kaune laikomi 1920-ieji metai, kai buvo įsteigti Lietuvos Aukštieji Kursai, į kurių sudėtį įėjo ir Matematikos-fizikos skyrius. 1922 m. vasario 16 d., įkūrus Lietuvos universitetą Kaune, buvo įsteigtas Matematikosgamtos fakultetas, kuris vykdė veiklą iki 1940 m. rudens. 1940 m. rugsėjo 1 d. Matematikosgamtos fakulteto veikla Kaune sustabdyta, jis perkeltas į Vilnių. Visą, nuo Fakulteto įkūrimo iki sustabdymo, veiklos laiką Kaune Fakultetui vadovavo dekanas prof. Zigmas Žemaitis. 1993 m. rugsėjo 1 d. Kauno technologijos universitete Aukštosios matematikos (pakeičiant pavadinimą į Taikomosios matematikos), Bendrosios matematikos ir Fizikos katedrų pagrindu įsteigtas Fundamentaliųjų mokslų fakultetas, 2014 m. sausio 1 d. (at)gavęs istorinį Matematikos ir gamtos mokslų fakulteto pavadinimą ir skaičiuojantis jau 25-uosius savo veiklos metus. The origins of the Faculty of Mathematics and Natural Sciences in Kaunas are the early 1920, when the Lithuanian Higher Courses were established and included the Mathematical-Physical Department. On the 16th of February 1922, the University of Lithuania was created in Kaunas and the Faculty of Mathematics and Nature was founded as well. It acted up until autumn in 1940. All the time from the very beginning until the suspension, the Dean of the faculty was Professor Zigmas Žemaitis. Later on the 1st of September 1993, the Faculty of Fundamental Sciences was established on the basis of two departments – Kaunas University of Technology Department of Higher Mathematics (later re-named to Department of Applied Mathematics) and Department of General Mathematics and Physics. On the 1st of January, 2014 the faculty received its historical title – the Faculty of Mathematics and Natural Sciences and now is celebrating 25 years of activity.

The stratosphere is attracting more and more attention for its potential to improve northern hemisphere (NH) seasonal weather forecasts, by modulating the North Atlantic Oscillation, or for its importance to reproduce observed southern hemisphere (SH) tropospheric jet trends, shaped by stratospheric ozone depletion. However, open questions remain regarding, for example, the stratospheric impact onto the ocean or the extent to which the complexity of stratospheric chemistry is important for surface climate. Within this thesis these two questions shall be answered. The importance of including a full representation of the stratosphere for North Atlantic and North Pacific Ocean variability is investigated for the first time. Furthermore, this thesis investigates the impact of interactive chemistry onto surface climate variability in an unprecedented model study, systematically reducing the complexity of stratospheric chemistry from an interactive chemistry scheme to a specified one prescribing zonal mean as well as zonally asymmetric ozone concentrations. [...] This thesis advances the understanding of the impact of stratospheric dynamics and feedbacks between chemistry and dynamics on surface variability. It concludes that the dynamical representation of the stratosphere is crucial for a more realistic representation of surface variability in the North Atlantic and Pacific Ocean. It underlines the importance of feedbacks betweenchemistry and dynamics for the characteristics of the stratospheric mean state, variability and stratosphere-troposphere-coupling. Especially under a strong ozone forcing, such as the observed Antarctic ozone depletion, incorporating interactive chemistry or at least a zonally asymmetric ozone forcing in a climate model is necessary for a proper representation of surface climate variability.

Radiogenic lead (Pb) and neodymium (Nd) isotopes are sensitive paleoceanographic proxies for the reconstruction of ocean circulation changes in the past. The goal of this dissertation is to develop improved approaches to recover past seawater Pb and Nd isotope signals from marine sediments and ferromanganese (Fe-Mn) crust, and apply it for tracing water mass sourcing changes in the Southern Ocean at (sub-)millennial resolution.

Die akute lymphatische Leukämie (ALL) ist die häufigste maligne Erkrankung im Kindesalter. Durch Anwendung moderner risikoadaptierter Behandlungsstrategien können heute nahezu 80% der Kinder dauerhaft geheilt werden. Dennoch erleidet immer noch jedes fünfte Kind ein Rezidiv. Ein Ziel ist daher die Identifizierung neuer prognostischer Marker, um die Therapie weiter zu optimieren. Eine Subgruppe der B-Vorläufer-ALL (pB-ALL) ist durch eine hohe CRLF2-Genexpression charakterisiert, welche durch verschiedene genetische Aberrationen verursacht werden kann. Die häufigste Aberration, die P2RY8/CRLF2-Fusion, ist in ALL-BFM-basierten Studien mit einer schlechten Prognose aufgrund einer hohen Rezidivinzidenz assoziiert; dessen zugrundeliegender Mechanismus ist allerdings noch weitestgehend unklar. Darüber hinaus fehlt bislang eine systematische Untersuchung der anderen Aberrationen. Im Fokus dieser Arbeit stand deshalb eine umfassende prospektive Erfassung der CRLF2-Genexpression und der assoziierten genetischen Aberrationen in 1105 Patienten der AIEOP-BFM ALL 2009 Studie. Zusätzlich wurden alle Fälle der pB-ALL mit hoher CRLF2-Genexpression auf Veränderungen der Kopienzahl bestimmter Gene der B-Zell-Differenzierung sowie auf Mutationen von CRLF2 und JAK2 hin untersucht. Außerdem wurde eine vergleichende NGS-basierte Transkriptom-Analyse mit CRLF2-hochexprimierenden Fällen mit und ohne P2RY8/CRLF2-Fusion durchgeführt sowie ein methodischer Vergleich von verschiedenen Techniken zur Detektion der P2RY8/CRLF2-Fusion für eine künftige Implementierung als prognostischer Marker.

In this thesis, we will provide new dimension-free L^p-bounds for the centered maximal function for products of Euclidean balls and for the r-variations of the underlying averaging operators. We also provide similar new results for products of spheres. In dieser Arbeit liefern wir neue dimensionsunabhängige L^p-Schranken für die zentrierte Maximalfunktion bzgl. Produkten euklidischer Kugeln sowie für die r-Variationen der zugrundeliegenden Mittelungsoperatoren. Wir liefern zudem ähnliche neue Resultate für Produkte von Sphären.

Energy is an important concept across the sciences and for physics in particular. However, only few students develop an understanding of energy that includes the critical aspect of conservation and is well-organized. How- ever, having well-organized ideas about energy is a prerequisite for applying that knowledge to make sense of phenomena. While the difficulty of energy for students is well established, there is vigorous debate about how to teach energy. The conceptual foundation of traditional forms-based instruction has been challenged time and again. As an alternative to forms, a transfer-only perspective that does not require forms and emphasizes the connections between energy, systems, and fields has been proposed. This position has also been adopted by the Framework for K- 12 Science Education (NRC, 2012). However, neither the Framework for K-12 Science Education nor the scholars that have argued for transfer-only ap- proaches have provided any guidance for how the instructional challenge of introducing energy, systems, and fields – all difficult ideas – in introductory energy instruction, that is, in middle school, can be approached. As part of the ELeVATE project (Exploring Potential Trajectories for the Energy Concept in Middle School), a transfer-only unit was developed, following the princi- ples of project-based learning. Drawing on data from the ELeVATE project, this dissertation investigates how students build ideas about energy in the transfer-only unit and how they can be supported in doing so. The findings indicate that students successfully build a well-organized understanding of energy, including a qualitative notion of energy conserva- tion, that allows them to use this knowledge to make sense of various phe- nomena. This suggests reconsidering the paradigm of energy learning progres- sions having to go through forms, highlights the power of coherent curriculum that project-based learning facilitates, and underlines the importance of hav- ing well-organized knowledge for being able to apply that knowledge.

Glioblastoma Multiforme is a malignant, primary brain tumor. Its incurability relies on the ability of the tumor to evade therapy and develop recurrences. Due to the impeded transport of molecules across the blood-brain barrier (BBB), subletal doses of agents within the tumor result in chemotherapeutically triggered therapy escape like tumor cell dormancy. This study aimed at both a more detailed understanding of molecular pathways as well as environmental influences on the entry into and exit from dormancy and, based on this knowledge, the improvement of therapy by introducing alternative compounds and strategies. Upon treatment with temozolomide (TMZ), the standard therapeutic, glioma cells entered an unproliferative state, which was not only described by dormant but also stem-cell features, indicated by an upregulation of dormancy and stem-cell markers and the ability of dormant cells to form spheroids. Moreover, dormancy entry was characterized by an increased expression of CCL2 and SAA2, while in dormancy exit, THSD4, FSTL3 and VEGFC were upregulated. Interestingly, co-stimulation with chemokines, mimicking an inflammatory environment, delayed the entry into and exit from dormancy. The use of alternative drugs and stimulation schemes appeared to be promising in overcoming therapy escape including dormancy. Especially, sequential application of TMZ+AT101, the R-(-)-enantiomer of the cottonseed-derived polyphenol gossypol, followed by AT101 alone increased glioma cell death. However, GBM cells were protected by astrocytes and microglia in an incomplete tumor-resection co-culture model, while glioma cells enhanced astrocytic cell death. Considering sequential application of TMZ and AT101, AT101 was encapsulated into cubosomes to improve passaging across the BBB. Further, a PDMS implant based on ZnO tetrapods was developed showing a high drug loading capacity and a sustained, long-term release of AT101 in artificial liquor for an improved therapy for patients.

Atherosklerose ist eine chronisch entzündliche Erkrankung der Gefäße, welche aufgrund der weltweit steigenden Inzidenz ihrer Folgeerkrankungen, wie Herzinfarkt und Schlaganfall, zunehmend an Bedeutung gewinnt. Die bisher verfügbaren Therapien beschränken sich überwiegend auf die symptomatische Behandlung der Atherosklerose, greifen jedoch nicht in die entzündlichen Mechanismen ein, welche die Gefäßveränderungen vorantreiben. Der neu entdeckte G-Protein-gekoppelte „orphan“ Rezeptor GPRC5B ist in den Zucker- und Lipidstoffwechsel involviert, reguliert inflammatorische Prozesse im Fettgewebe und wird auch in den Zellen des vaskulären Systems sowie in Makrophagen endogen exprimiert. In der vorliegenden Arbeit wurde ein möglicher Einfluss von GPRC5B auf die Entzündung der Gefäßwand in vitro sowie auf die atherosklerotische Plaquebildung in vivo untersucht. Die Untersuchungen zeigten, dass ein proatherogenes, inflammatorisches Milieu zu einer verstärkten Expression von GPRC5B in den Zellen des vaskulären Systems führt und dass der Rezeptor selbst über die Aktivierung von ERK 1/2 sowie des NFκB-Signalwegs die inflammatorische Antwort der Zelle im Sinne einer positiven Rückkopplung verstärkt. In vivo verstärkte GPRC5B die Infiltration inflammatorischer Makrophagen in die entzündete Gefäßwand und förderte das Voranschreiten atherosklerotischer Läsionen. Die in dieser Arbeit gewonnenen Erkenntnisse decken neue Funktionen des noch wenig untersuchten „orphan“ Rezeptors im Rahmen der Gefäßentzündung und Atherogenese auf und weisen auf ein therapeutisches Potential einer GPRC5B-Hemmung für die Behandlung dieser Erkrankung hin. Atherosclerosis is a chronic vascular inflammatory disease, which is becoming more and more important due to the increasing incidence of its secondary diseases, such as myocardial infarction and stroke. The therapies available so far are mainly limited to the symptomatic treatment of atherosclerosis, but do not interfere with the inflammatory mechanisms that drive vascular lesions. The newly discovered G-protein coupled orphan receptor GPRC5B is involved in sugar and lipid metabolism, regulates inflammatory processes in adipose tissue and is also expressed endogenously in vascular cells and macrophages. The present study investigated the regulation of GPRC5B in the cells of the vascular wall and the impact of GPRC5B on vascular inflammation in vitro and on atherosclerotic plaque formation in vivo. The investigations in the current study showed that a pro-atherogenic inflammatory signaling leads to an increased expression of GPRC5B in the cells of the vascular wall, with the receptor sequentially enhancing the inflammatory response of the cell via activation of ERK1/2 and NFκB signaling pathways pointing to a positive feedback loop. In vivo, GPRC5B enhanced the infiltration of inflammatory macrophages into the inflamed vessel wall and promoted the progression of atherosclerotic lesions. The findings obtained in this study reveal novel functions of the orphan receptor GPRC5B in vascular inflammation and atherogenesis, might leading to a specific therapeutic target in the treatment of atherosclerosis.

In the context of inverse design of molecules with desired optimal properties, the long-term goal of this Thesis is to develop a general framework which tackles the design of molecular systems for an optimal catalytic effect onto arbitrary chemical reactions. For any given reaction, an arrangement of an additional molecular framework around this reaction center is sought such that the energetic reaction barrier is lowered as much as possible. As necessary abstraction layer, the so-called globally optimal catalyst (GOCAT) model is introduced, and, furthermore, evolutionary algorithms (EAs) are harnessed as implemented in our global optimization suite for chemical problems, ogolem, which was highly extended to allow for these catalysis optimizations. Starting with a maximally reductionistic approach for studying the non-bonding interactions, electrostatic GOCATs are introduced that consist of arbitrary numbers, distributions and strengths of partial point charges around reacting molecules, mostly surrounding these on a common exposed surface. In the end, two reactions are studied in detail within the general topic of electrostatic catalysis. Some of the initially present model approximations are already sufficiently lifted, still-existing ones are critically assessed and further future extensions to the framework are discussed. Moreover, many method development matters are addressed: They range from optimal shared-memory parallelization, exemplified for global parameter optimization of the reactive force field, ReaxFF, via diversity control parameters for the EAs, applied to a cluster structure optimization problem, to EA operator benchmarks and optimizations of abstract electrostatics. Im Kontext von inversem Design von Molekülen mit optimalen Eigenschaften versucht die vorliegende Arbeit als Langzeitziel eine passende Plattform zu entwickeln, welche das generelle Design molekularer Systeme für einen optimalen Katalyseeffekt auf beliebige chemische Reaktionen projektiert. Für eine gegeben Reaktion soll eine hinzukommende chemische Umgebung komponiert werden, welche die Reaktionsenergiebarriere so weit wie möglich vermindert. Als notwendige Abstraktionsschicht wird das sogenannte Modell des globally optimal catalyst (GOCAT) eingeführt und außerdem kommen Evolutionäre Algorithmen (EAs) zur Anwendung, wie sie bereits in unserem Programmpaket zur Lösung allgemeiner globaler Optimierungsprobleme der Chemie, ogolem, bereitgestellt werden, welches jedoch deutlich für diese Katalyseoptimierungen ergänzt wurde. Angefangen in einem maximal-reduktionistischen Ansatz werden elektrostatische GOCATs erarbeitet, die aus einer beliebigen Anzahl, Verteilung und Stärke von Partialladungen bestehen und rund um die reagierenden Moleküle drapiert werden, meist auf einer gemeinsamen exponierten Oberfläche. Insgesamt werden zwei Reaktionen detailliert untersucht im generellen Kontext von elektrostatischer Katalyse. Einige eingangs vorhandene Modellannahmen werden bereits systematisch verbessert, noch vorhandene kritisch beleuchtet und künftige Erweiterungen auseinandergesetzt. Weiterhin werden unterschiedliche Methodenentwicklungsaspekte angesprochen: Diese reichen von verbesserter Parallelisierung in Mehrprozessorarchitekturen, beispielhaft gezeigt anhand einer globalen Parameteroptimierung des reaktiven Kraftfeldes ReaxFF, über Diversitätskontrollparameter des EAs, illustriert mittels eines Clusterstrukturoptimierungsproblems, bis hin zu EA-Operator-Testevaluationen und allgemeinen abstrakten Elektrostatikoptimierungen.

In der Dissertation werden Ergebnisse zur Beschreibung der Realstruktur und der physikalischen Eigenschaften ausgewählter Übergangsmetalldichalkogenide vorgestellt. Die Schichtstrukturen der Verbindungen MoS2 und WS2 zeigen häufig ausgeprägte Stapelfehlordnung. Um diese zu beschreiben wurde der Ansatz der Superzelle gewählt, in den Messdaten aus Röntgenbeugung und Totalstreuung eingeflossen sind. Das Halbmetall HfTe2 wurde im Hinblick auf seine Magnetotransporteigenschaften untersucht, wobei ein ausgeprägter positiver magnetoresistiver Effekt nachgewiesen werden konnte. Die magnetischen wie auch die Magnetotransporteigenschaften des ferromagnetischen Metalls Fe1/3TaS2 wurden umfassend untersucht und im Kontext von struktureller Ordnung und Stöchiometrie der Probe diskutiert. This thesis contains results on the description of the real structure as well as physical properties of selected transition metal dichalcogenides. The layered structures of the compounds MoS2 and WS2 are known to be prone to stacking disorder. For modelling thereof the so called supercell approach was chosen and data of both X-ray diffraction and total scattering were included. The semimetal HfTe2 was studied with respect to its magnetotransport properties, revealing a pronounced positive magnetoresistance effect. The magnetic as well as magnetotransport properties of the ferromagnetic metal Fe1/3TaS2 were studied in detail and discussed in the context of its structural order as well as sample stoichiometry.

The question of how industry develops differently in different regions is an interesting topic in economic geography. Scholars have contributed enormous knowledge concerning various relatedness, the role of multiple actors and their agencies, and regional industrial preconditions, in contributing to local industrial path development. However, our knowledge on the agency-structure relations present in local industrial development is limited, if not totally lacking, not mentioning how such relations influence the industrial development trajectories and ultimately the overall competitiveness of the local industry. Based on such reflections, this thesis aims to fill such an ‘agency-structure’ gap in local industrial development by looking at the developmental trajectories of one creative industry—online games industry, in two regions that belong to different countries (Shanghai in China and Hamburg in Germany). In both cases, all sorts of misalignments between the focal industry and the institutional frameworks (including both formal and informal institutional elements) could be observed during the industrial development processes. It is under such industry-institution misalignments that the agency and activities of multiple actors have been analyzed. By comparing the two trajectories in different institutional contexts, the research contributes to the literature in the following ways. First, this research gives due attention to the structural forces, such as the formal and informal institutional structures, that affect local industrial development. Secondly, it contributes to a nuanced analysis of agency in local industrial development, including innovative entrepreneurship, institutional entrepreneurship and place leadership. Finally, it reveals the dynamic agency-structure relations, which serve as the causal mechanisms leading to different developmental outcomes in the games industry in the two selected cities.

Push-pull conjugated polymers, consisting of electron donating and withdrawing units gained increased attention because of their narrow band gap and their response to the light absorption and emission. Accordingly, four monomers with an intrinsic push-pull effect were synthesized via a dually selective cross-coupling reaction under Stille conditions between two types of heterocycles. The dinucleophile type beared both a trimethyl stannyl group and either a pinacol boronate (3-n-hexylthiophene) or a magnesium chloride group (N-derivate pyrrole). The dielectrophilic type consisted of iodo-bromo heterocycle derivates (phenyl, pyridine and thienyl). For these monomers, the presence of a remaining metal (pinacol boronate or magnesium chloride group) and bromine allowed further polymerizations under Suzuki-Miyaura or Kumada conditions. In the case of the thienyl-phenyl and thienyl-pyridine type monomer, oligomers were formed. Their electroluminescence was successfully demonstrated by the implementation into OLED devices. Additionally, stannoles, which are tin analogues of cyclopentadienes, were studied as possible electron deficient systems in combination with thiophene derivates in organometallic push-pull monomers. The synthesis of stannoles is generally performed using the conditions of the well-known Fagan Nugent reaction of alkynes, where in the first step zirconacyclopentadienes are formed and secondly transmetalated with dichloro stannanes. This thesis focusses on the formation of zirconacyclopentadienes by comparing the synthetic reactions between Negishi’s and Rosenthal’s reagent with several alkynes. The Rosenthal’s reagent proved to give higher reaction rates as well as higher yields of zirconacyclopentadienes in those cases, in which functional groups as aryl-halides, trimethylstannyl and trimethylsilyl were present. Lastly, first attempts of cross-coupling reactions between stannoles with nucleophilic substituents and thiophene electrophilic derivates were done.

Increasing pressures from both natural disturbance and anthropogenic activities challenge river ecosystem resilience. A better understanding of the ecological balancing of structure under multiple stressors is thus critical for sustainable future as well as human wellbeing. Phytoplankton are one of the important autotrophs in the river ecosystem. They are sensitively reflecting multiple filters. However, studies on how flow regime, land-use pattern, physicochemical condition and spatial factors overall affect the lotic phytoplankton community are still scarce. This study aims to examine riverine phytoplankton community structure in relation to combined abiotic gradients, and to consider the potential responses under changing status. Study area is located in a lowland catchment Treene, Germany. We establish an integrated modelling framework in combination of hydrological model with ecological models. The investigations show high spatial and temporal variations of phytoplankton community. Benthic diatom is dominant in most of the study area. Euglenophyta show higher existence percentage in the headwaters and exhibit a strong positive correlation to the share of agricultural land-use. Microcystis has found with high abundance downstream of a lake. Higher phosphorous concentration directly causes the increase of Microcystis population. Both species and traits composition more relate to hydrological and local physicochemical heterogeneity than to species dispersal, which confirm the suitability of lowland phytoplankton-based bioassessment in the study area. Due to the importance of flow regime in shaping pelagic algal community, it is necessary to include hydrological variables in biodiversity conservation. The shares of forest land-use area have outstanding explanation to the variation of species richness. Our findings emphasize the significance of preservation of forest area in protecting the aquatic algal biodiversity for maintaining the river ecosystem functioning.

This dissertation sought to implement an intervention that was meant to support high-achieving young women’s physics engagement. The studies were implemented in the context of the PhysicsOlympiad in order to tap amongst the highest-achieving female physics students and probe effects of specifically designed learning environments. Four empirical studies revealed that even high-achieving young women at times face barriers in their physics engagement that eventually result from constraining expectations by their peer-groups or gender stereotypes. However, the intervention studies also showed that it was possible to support the physics engagement of the high-achieving young women. For example, important resources for physics engagement, such as competence beliefs, could be positively affected for young women while not depressing young men’s physics engagement at the same time. Furthermore, the long-term development of resources was also positively affected through the intervention. Namely, students who experienced a high environmental fit with the intervention environment developed a more positive sense of belonging compared to students who reported a lower environmental fit. In summary, these findings buttress the assumption that considerately designed learning environments that incorporate the motivations, goals, and values of high-achieving young women are potentially effective in order to challenge the overproportional dropout of high-achieving young women from physics learning environments.

The goal of this Thesis was the investigation of ultrafast electronic deactivation pathways in both intra- and intermolecularly H-bonded systems in a time-resolved and structure-sensitive manner by a combination of spectroscopic pump-probe techniques and quantum chemical calculations. Towards these ends, a transient vibrational absorption spectrometer was built, which allows for the investigation of the temporal evolution of characteristic vibrational marker bands in the mid-IR spectral region after UV excitation. Using this setup, 2-aminopurine (2AP) was investigated in its monomeric form and upon H-bonding with thymine (T) in Watson-Crick (WC) and Hoogsteen (HS) conformations as a model system for intermolecular H-bonding. An exploratory study on free 2AP revealed the involvement of an intermediate state in the deactivation dynamics towards concurrent efficient ground-state recovery and triplet state formation within ~740 ps. Upon H-bonding of 2AP with T in the WC conformation, internal conversion towards the intermediate state is hindered by an increased potential energy barrier. Thus, the excited-state lifetime of the 2AP chromophore in the H-bonded 2AP-T network is increased to ~1.6 ns. In the HS form, however, the 2AP moiety features a distorted excited-state minimum structure. Induced by H-bonds with T, this bent structure of 2AP opens up novel deactivation pathways for efficient ground-state recovery with ~68 ps. The observed kinetics are discussed in terms of excited-state interbase double-proton transfer. Moreover, in another case study, the influence of excited-state intramolecular proton transfer (ESIPT) on the electronic deactivation and reaction dynamics of the photoswitch N-(3-pyridinyl)-2-pyridinecarboxamide (NPPCA) was investigated. Unambiguous indications for ESIPT-induced switching dynamics on the ultrafast timescale were found in the reaction pathways towards transient formation of a novel photochromic proton-transferred species in ~20 ps. Es wurden die ultraschnellen elektronischen Desaktivierungswege von intramolekular sowie intermolekular über H-Brücken gebundenen Systemen zeitaufgelöst und struktur-spezifisch durch eine Kombination aus spektroskopischen Anregung-Abfrage Methoden und quantenchemischen Rechnungen untersucht. Zu diesem Zweck wurde ein transientes Schwingungs-Absorptionsspektrometer aufgebaut, welches die Untersuchung der zeitlichen Entwicklung charakteristischer molekularer Schwingungs-Moden im mittleren Infrarot-Bereich nach UV-Anregung ermöglicht. Mithilfe dieses Aufbaus wurde 2-Aminopurin (2AP) sowohl in seiner freien Form als auch nach Paarung mit Thymin (T) in Watson-Crick (WC) und Hoogsteen (HS) Motiven als Modellsysteme für intermolekular H-Brücken gebundene Systeme untersucht. Eine Studie zu freiem 2AP zeigte, dass ein transienter Zustand die Dynamik zum Wiederauffüllen des Grundzustandes und zur gleichzeitigen Bildung eines Triplet-Zustandes aus dem initial bevölkerten Zustand mit ~740 ps dirigiert. Durch Ausbildung von H-Brücken mit T in der WC-Form wird innere Umwandlung erschwert. Folglich wurde eine verlängerte Lebenszeit des angeregten Zustands von ~1.6 ns ermittelt. Im Gegensatz zur WC-Konformation ermöglicht die Ausbildung von H-Brücken in der HS-Form des 2AP-T Basenpaares neue Wege für das Wiederauffüllen des Grundzustandes in ~68 ps aufgrund einer verzerrten Struktur im angeregten Zustand. Die verkürzte Lebenszeit des HS-Dimers wurde im Hinblick auf einen intermolekularen Protonentransfer im angeregten Zustand diskutiert. Außerdem wurde der Einfluss von intramolekularem Protonentransfer in elektronisch angeregten Zuständen (ESIPT) auf die Desaktivierungs- und Reaktions-Dynamik am Beispiel des Photoschalters N-(3-Pyridinyl)-2-Pyridincarboxamid (NPPCA) untersucht. Es konnten eindeutige Beweise für die ESIPT-induzierte Isomerisierungsdynamik auf der ultraschnellen Zeitskala zur Bildung einer neuen H-transferrierten Spezies innerhalb von ~20 ps gefunden werden.

This study on the interaction of Holocene climate, human impact and environment in the northern Central Alps uses geochemical, pollen and radiocarbon analyses in peat samples. Mires were cored and analysed in the Kleinwalser Valley (Vorarlberg, Austria), Piller Mire (Tyrol, Austria) and upper Fimba Valley (Grisons, Switzerland). For different reasons, using portable X-Ray-Fluorescence analysis (pXRF) for peat is uncommon. The calibration of pXRF with Inductively Coupled Plasma – Mass Spectrometry (ICP-MS) shows that e.g. Ti, Pb, Sr can be used as palaeoenvironmental proxies in peat, allowing the following conclusions: At c. 8200 and in the late 7th millennium BP, wet and cold climate conditions prevail in the region. The earliest land use is recorded in the Kleinwalser Valley around 5500 cal BP, with fire clearings, pastoralism and previously undetected regional metallurgy. Centuries before mining boomed in the Eastern Alps, metallurgy around the Piller Mire is detected at c. 4300 cal BP. The strongest prehistoric land use affects all sites in the Bronze Age (3500-3000 cal BP). Potential metal enrichments are masked by mineral input. This period is followed by decreasing land use until the late Roman period. However, a strong Pb enrichment factor (Pb EF) in the Kleinwalser Valley is recorded around 2700 cal BP. Human impact increases in northwestern Tyrol around 2400 cal BP. Metallurgy is indicated by increased Pb EF during the Roman Empire and right after its collapse. Human activities rise again with fading Roman power after 250 cal CE in the Kleinwalser Valley but are interrupted by a climate deterioration after 500 cal CE (Late Antique Little Ice Age). At high elevations, land use intensifies not before the High Middle Ages, when also regional mining operations arise. While Pb EF rises continuously until 1980 cal CE, fluctuating human impact can be linked to climatic, cultural or demographic factors. Recently, growing tourism and construction increase erosion. Diese Arbeit untersucht mit Geochemie, Pollen und Radiokohlenstoffanalysen in Torfproben die Beziehungen von Klima, Mensch und Umwelt im Holozän in den nördlichen Zentralalpen. Dazu wurden Bergmoore in drei Untersuchungsgebieten der NZA ausgewählt: Kleinwalsertal (Vorarlberg, Österreich), Piller Moor (Tirol, Österreich) und oberes Fimbatal (Graubünden, Schweiz). Der Einsatz der tragbaren Röntgenfluoreszenzanalyse (pXRF) für Torfproben ist jedoch selten. Nach dessen Kalibrierung mit Induktiv gekoppelter Plasma-Massenspektrometrie (ICP-MS) lassen sich u.a. Ti, Pb, Sr und andere als Paläoökologieproxys verwenden. Die Ergebnisse lassen folgende Schlüsse zu: Um 8200 und ca. 6200 cal BP herrscht regional feuchtkaltes Klima. Frühe Landnutzung zeigt sich im Kleinwalsertal um 5500 cal BP (Brandrodung, Beweidung und Hinweisen auf regionale Metallurgie). Jahrhunderte vor dem Boom der Bergbaugebiete in den Ostalpen, sind metallurgische Spuren um das Piller Moor für c. 4300 cal BP nachweisbar. Die stärkste prähistorische menschliche Landnutzung betrifft alle Standorte in der Bronzezeit (3500-3000 cal BP). Darauf folgt eine Phase geringerer Landnutzung, die bis weit in die römische Zeit reicht. Im Kleinwalsertal wird jedoch um 2700 cal BP eine erhöhte Bleianreicherung (Pb EF) sichtbar, während östlich die Landnutzung um 2400 cal BP zunimmt. Der römische Bergbau wird durch erhöhte Pb EFs angezeigt. Eine Episode regionalen Bergbaus zeigt sich unmittelbar nach dem Zerfall des Römischen Reiches. Intensivere Landnutzung beginnt ab ca. 250 cal CE im Kleinwalsertal, wird aber durch eine Klimaverschlechterung (500 cal CE) unterbrochen. An den höher gelegenen Standorten findet die Intensivierung erst im Hochmittelalter statt. Während der Pb EF bis 1980 cal CE stark ansteigt, können Landnutzungsspuren mit klimatischen und demografischen Faktoren verbunden werden. Im 20. Jahrhundert verstärken wachsender Tourismus- und Infrastrukturbau die Erosion.

Polyoxovanadate (POVs) stellen eine Untergruppe der Polyoxometallate dar und sind durch eine große Struktur- und Zusammensetzungsvielfalt gekennzeichnet. Durch das erste wasserlösliche antimonsubstituierte POV, {Ni(en)3}3[V15Sb6O42(H2O)x]·≈15 H2O (Ni-{V15Sb6}) (en = ethylendiamin) ergab sich eine völlig neue synthetische Chemie für Postfunktionalisierungsreaktionen, welche vorher nicht möglich war. Die vorliegende Dissertation fokussiert sich auf die Synthese und Charakterisierung neuer Heterometall-POVs (Sb-POVs und Ge/Sb-POVs), welche durch Verwendung der Precursoren Ni-{V15Sb6} und {Zn(en)3}3[V15Sb6O42(H2O)]·3 en·10 H2O (Zn-{V15Sb6}) hergestellt werden konnten. Die Vorläuferverbindung Zn-{V15Sb6} wurde im Laufe der Arbeit synthetisiert und charakterisiert und ist im Vergleich zu Ni-{V15Sb6} besser löslich sowie deutlich stabiler in wässriger Lösung. Die umfassende Charakterisierung der gelösten Spezies gelang mit Elektrosprayionisations-Massenspektrometrie (ESI-MS) Untersuchungen in Zusammenarbeit mit ULRIKE WARZOK und CHRISTOPH A. SCHALLEY (FU Berlin). Die Ergebnisse belegen, dass sich beide Precursoren langsam in den {V14Sb8O42}-Cluster umwandeln, wobei diese Umwandlung bei Zn-{V15Sb6} sehr viel langsamer verläuft. Die Umwandlung kann beschleunigt werden, wenn der pH-Wert erniedrigt wird. Diese Befunde eröffneten völlig neue Perspektiven für die gezielte Synthese neuer Verbindungen. Besonders spektakulär ist die Verbindung {Ni(cyclen)(en)}2[V14Sb8O42(H2O)]·ca 10 H2O, diese enthält ein neues Konfigurationsisomer des {V14Sb8O42}-Clusters als herausragendes Strukturmotiv. Während normalerweise die Spitzen der {VO5}-Pyramiden der Cluster nach außen weisen, zeigt eine {VO5}-Pyramide des neuen Isomers nach innen. Diese Ausrichtung des {VO5}-Polyeders wird durch die kurzen Sb-O-Abstände zwischen zwei benachbarten Clustern verursacht, so dass ein Dimer ausgebildet wird. ESI-MS-Untersuchungen beweisen, dass das Dimer auch in Lösung vorhanden ist. Polyoxovanadates (POVs) are a subgroup of polyoxometallates characterized by a fascinating structural variety and flexible compositions. The discovery of the first water-soluble antimony-substituted POV (Sb-POV), {Ni(en)3}3[V15Sb6O42(H2O)x]∙≈15 H2O (Ni-{V15Sb6}) (en = ethylenediamine) resulted in a completely new synthetic chemistry of post-functionalization reactions, which was previously not possible. The present dissertation focuses on the synthesis and characterization of new hetero-POVs (Sb-POVs and Ge/Sb-POVs) by using the precursors Ni-{V15Sb6} and {Zn(en)3}3[V15Sb6O42(H2O)]∙ 3 en∙10 H2O (Zn-{V15Sb6}). The precursor Zn-{V15Sb6} was synthesized and characterized during this work and the main characteristics are the better solubility compared to Ni-{V15Sb6} and a significantly enhanced stability in aqueous solution. The extensive characterization of the dissolved species was achieved by electrospray ionization mass spectrometry (ESI-MS) in collaboration with ULRIKE WARZOK and CHRISTOPH A. SCHALLEY (FU Berlin). The results demonstrated that both precursors slowly transform into the {V14Sb8O42}-cluster, and this process is much slower for Zn-{V15Sb6}. The conversion can be accelerated by lowering the pH value. These investigations opened up completely unexpected perspectives for the targeted synthesis of new compounds. Particularly noteworthy is {Ni(cyclen)(en)}2[V14Sb8O42(H2O)]·ca 10 H2O, which contains a new configurational isomer of the {V14Sb8O42}-cluster as prominent structural motif. While normally the vanadyl group of the {VO5} pyramids point outward, one {VO5} pyramid of the new isomer faces inward. This arrangement of the {VO5} polyhedron is caused by the short Sb-O distances between two neighboring clusters, forming a dimer. ESI-MS investigations proved that the dimer is also present in solution.

ADAM17 ist eine membranständige, ubiquitär exprimierte Metalloprotease, die in einer Vielzahl von physiologischen und pathophysiologischen Vorgängen eine Rolle spielt. Es sind über 80 Substrate bekannt, welche von ADAM17 proteolytisch prozessiert werden können. Dadurch werden eine Vielzahl biologischer Prozesse, wie z. B. die Immunantwort, Entzündungsreaktionen, sowie entwicklungsbiologische und regenerative Zellantworten, beeinflusst. Darüber hinaus spielt ADAM17 bei der Tumorentwicklung, Autoimmunerkrankung und chronischen Entzündungen eine entscheidende Rolle. Durch die Vielzahl an Prozessen, an denen ADAM17 beteiligt ist, wird die bedeutsame Rolle der Protease im Organismus deutlich und erfordert eine strikte Regulation ihrer Aktivität. Die membranständigen Substrate von ADAM17 werden nach Zellstimulation nahe der Zellmembran prozessiert, wodurch es zur Freisetzung ihrer Ektodomäne kommt. In dieser Arbeit wurden die strukturellen Voraussetzungen untersucht, die zur Freisetzung des sIL-6R erforderlich sind. Dabei stand der Abstand der ADAM17-Schnittstelle im IL-6R zur Zellmembran im Vordergrund. Mit diesem Beitrag sollte das Verständnis des Aktivierungsmechanismus von ADAM17 vertieft werden. Neues Wissen über Grundlagen der Aktivierung bzw. Inaktivierung von ADAM17 ist essentiell um zukünftig die Aktivität der Protease in pathologischen Situationen gezielt zu modifizieren. So wird ADAM17 durch die ER-residente PDIA6 inaktiviert. Dabei interagiert die PDIA6 auf der Zelloberfläche mit der MPD von ADAM17. Die resultierende Isomerisierung zweier Disulfidbrücken führt zu einer Konformationsänderung innerhalb der MPD. Dieser Vorgang resultiert in einer Strukturänderung, die es ADAM17 nicht mehr erlaubt die Schnittstellen der membranständigen Substrate zu erreichen. In dem zweiten Abschnitt der Arbeit wurde untersucht, wie die ER-residenten PDIs an die Zelloberfläche gelangen und auf der Zelloberfläche verbleiben. Damit sollte ein Beitrag zum Verständnis des Inaktivierungsmechanismus von ADAM17 geleistet werden.

Nitrogen deficiency is known as one of the major limitations in crop productivity and the yield losses caused by inadequate N supply are far greater than those caused by a deficiency in other macronutrients. In the case of potato, due to its low capability of N retraction from the soil, there is a higher demand for fertilization and therefore a higher negative impact on the environment. The aim of the presented thesis was to elucidate metabolic pathways, and candidate proteins determining the contrasting responses of the potato cultivars ‘Lambada’ and ‘Topas’ to N deficiency. The potato proteome was analyzed using mass spectrometry-based approaches, in respect to the crude root proteome, with an additional focus on the proteins of the plasma membrane and on the changes in the phosphorylation pattern. The most promising candidate proteins were further validated by using enzymatic assays, Western blotting and A. thaliana mutant plants. In response to N deficiency the tolerant cultivar ‘Topas’ showed higher accumulation of proteins participating in N assimilation, C metabolism as well as in protein synthesis and degradation, together with a stable growth and higher N content. The study of the plasma membrane composition revealed a higher abundance of the nitrate transporter NRT2.1 as well as two isoforms of aquaporins. The relevance of aquaporins in the response to N deficiency was confirmed by experiments with Arabidopsis mutants, which showed significantly reduced N content in the leaves. The main conclusion of the study was, that under non-limiting conditions, the sensitive cultivar used all the applied N to produce biomass, whereas the tolerant cultivar produced N-containing compounds and more proteins. When N is limited, the sensitive cultivar’s ability to grow rapidly decreases. In contrast, the tolerant cultivar uses the NRT2.1 as well as aquaporins for more efficient N uptake but it also consumes the accumulated resources to adapt to the lower N conditions.

Inspiriert von natürlichen Synthesemaschinen wie der ATP-Synthase sollte in dieser Arbeit ein Modell einer künstlichen Kondensationsmaschine für die Oxoanionen Phosphat und Vanadat entwickelt werden. Basierend auf einem Porphyringrundgerüst und unterschiedlichen Bindestellen wurden verschiedene Strukturen identifiziert und synthetisiert. NMR-Titrationsexperimente zeigen Wechselwirkungen mit Vanadaten und Phosphaten. Bei erhöhter Temperatur findet außerdem eine Kondensation von Monophosphat zu Diphosphat statt. Inspired by natural assemblers like ATP synthase, the development of an artifical condensation machine for phosphate and vanadate was the objective of this thesis. Based on a porphyrin scaffold and different binding sites, several structures were identified and synthesized. NMR titration experiments demonstrate an interaction between the receptors and different oxoanions. At elevated temperature, a condensation from monophosphate to diphosphate is observed.

RNase7 (R7) spielt eine große Rolle in der antimikrobiellen Abwehr der Haut gegen Pathogene und verfügt über potente Ribonukleaseaktivität. Das konstitutiv exprimierte antimikrobielle Peptid (R7) lässt sich durch proinflammatorische Zytokine und Bakterien induzieren. R7 befindet sich hauptsächlich in der äußersten Schicht der Epidermis, dem Stratum corneum (S.c.). Ein starker Inhibitor von R7, der Ribonuklease-Inhibitor (RI), befindet sich hauptsächlich in der darunterliegenden Hautschicht, dem Stratum spinsoum, wo er überwiegend im Zytosol der Keratinozyten exprimiert wird. In dieser Arbeit wurde die Interaktion des RI mit R7 untersucht. Es zeigte sich, dass es R7 möglich ist, in vitro an den RI zu binden, der dadurch sowohl die antimikrobielle als auch die Ribonukleaseaktivität von R7 hemmt. Im S.c. vorkommende Kallikrein-Proteasen können den RI degradieren, allerdings nicht, wenn der RI schon einen Komplex mit R7 eingegangen ist. So ist es eher unwahrscheinlich, dass Proteasen im S.c. R7 aus einem RI-R7-Komplex freisetzen können. Die Bindung des RI an R7 führte zur Frage, ob der RI die Expression und insbesondere die Sekretion von R7 aus den Keratinozyten beeinflusst. Transfektionsstudien mit RI-Überexpressionsplasmiden sowie RI-siRNA konnten keinen Einfluss herausstellen. Auch eine potentielle Schutzwirkung des RI vor einer zytotoxischen Aktivität von R7 in den Keratinozyten konnte nicht gefunden werden. Da R7 aus dem Keratinozyten sezerniert wird und der RI nur in den Keratinozyten nachweisbar ist, nicht aber im Zellkultur-Überstand, ist es fraglich, ob der RI außerhalb der Zelle überhaupt einen regulativen Einfluss auf R7 haben könnte. Eine Freisetzung des RI im Rahmen von Zelltod wäre allerdings denkbar. In vivo konnten erhöhte RI-Genexpressionslevel in der läsionalen Haut von Patienten mit den Hauterkrankungen atopischer Dermatitis und Psoriasis nachgewiesen werden. Dies lässt vermuten, dass der RI in diesen Erkrankungen eine Bedeutung haben könnte. RNase 7 plays a major role in the antimicrobial defence of the skin against pathogens and exhibits a potent ribonuclease activity. The constitutively expressed peptide RNase 7 is inducible by proinflammatory cytokines and bacteria. In vivo, RNase 7 is mainly located in the outermost layer of the epidermis, in the Stratum corneum. In contrast, a strong inhibitor of RNase 7, the Ribonuclease inhibitor (RI), is mainly found in the layer below the Stratum corneum, in the Stratum spinosum, where it is mainly expressed in keratinocyte cytosol. In the present work the interaction of RI and RNase 7 was investigated intensively. It was shown, that RNase 7 is able to bind to the RI in vitro. Through the interaction the RI blocks the antimicrobial activity as well as the ribonuclease activity of RNase 7. Kallikrein-proteases in the Stratum corneum may degrade the RI, but not if the RI is already bound to RNase 7. Hence, it is rather unlikely, that those proteases may release RNase 7 from a RI RNase 7 complex in the Stratum corneum. The binding of the RI to RNase 7 led to the question, if the RI influences the expression and especially the secretion of RNase 7 from keratinocytes. Transfection experiments with RI-overexpression plasmids as well as experiments with RI-siRNA here did not show any influence on RNase 7. Furthermore, the RI did not show any potential protective influence on a possible cytotoxic activity of RNase 7 in the keratinocytes. As RNase 7 is predominantly secreted by keratinocytes and the RI is only detectable in the keratinocytes but not in the keratinocyte supernatant it is not clear if the RI has a regulatory influence on RNase 7 outside of the keratinocytes at all. However, a release of the RI within the scope of cell death might be supposable. Increased levels of RI-gene expression in lesional skin of patients with the chronic inflammatory skin disease atopic dermatitis and psoriasis has been shown in vivo. This leads to the conclusion that the RI may play a role for those skin diseases.

Properties of seawater in the ocean are not uniformly distributed. Characteristics in different regions and depths are significantly different. Understanding of the distribution and variation of seawater plays an important role in investigating the thermohaline circulation of the world ocean or predicting climate changes. A body of seawater that originates in a particular area and shares a common formation history always has similar properties and such a water body is defined as a water mass. Source water type defines the properties of the original water mass in the formation area. The properties of water masses do not stay constant but change along the flow path due to biogeochemical changes and also due to the mixing with surrounding water masses so that the OMP method is required to investigate the distribution of water masses. The OMP method is to calculate the best components and fractions of more water masses by analyzing water properties (potential temperature, salinity, oxygen, phosphate nitrate and silicate in this study) and solving the equations of linear mixing without assumptions. With the applications of transient tracers (such as: CFC-12 and SF6), water masses can be labeled and their mean ages (consuming time during the pathway from formation area) are calculated. Combining the distributions and mean ages, the transport time and velocities of water masses can be estimated. Die Eigenschaften des Meerwassers im Ozean sind nicht gleichmäßig verteilt. Merkmale in verschiedenen Regionen und Tiefen unterscheiden sich erheblich. Das Verständnis der Verteilung und Variation von Meerwasser spielt eine wichtige Rolle bei der Untersuchung der thermohalinen Zirkulation des Weltmeers oder der Vorhersage von Klimaänderungen. Ein Meerwasserkörper, der aus einem bestimmten Gebiet stammt und eine gemeinsame Formationsgeschichte aufweist, hat immer ähnliche Eigenschaften, und ein solcher Wasserkörper wird als Wassermasse definiert. Der Quellwassertyp definiert die Eigenschaften der ursprünglichen Wassermasse im Formationsbereich. Die Eigenschaften der Wassermassen bleiben nicht konstant, sondern verändern sich entlang des Fließweges aufgrund biogeochemischer Änderungen und auch aufgrund der Vermischung mit umgebenden Wassermassen, so dass die Optimal Multiparameter (OMP)-Methode zur Untersuchung der Verteilung der Wassermassen erforderlich ist. Bei der OMP-Methode werden die besten Komponenten und Anteile von mehr Wassermassen berechnet, indem die Wassereigenschaften (potentielle Temperatur, Salzgehalt, Sauerstoff, Phosphatnitrat und Silicat in dieser Studie) analysiert und die Gleichungen des linearen Mischens ohne Annahmen gelöst werden. Mit den Anwendungen von transienten Tracern (wie: CFC-12 und SF6) können Wassermassen markiert und ihr mittleres Alter (Zeitaufwand während des Pfads vom Formationsbereich) berechnet werden. Durch die Kombination der Verteilungen und des Durchschnittsalters können die Transportzeit und die Geschwindigkeiten der Wassermassen geschätzt werden.

We model the variations of the crustal and lithospheric thicknesses as well as the variation in temperature, composition, S-wave seismic velocity and density of the lithosphere beneath the Saharan Metacraton (SMC) using three-dimensional (3D) modelling. The SMC was once an intact Archean-Paleoproterozoic craton, but was remobilized (metacratonized) during the Neoproterozoic due to partial loss of its sub-continental lithospheric mantle (SCLM) during collisions along its margin. The partial loss of the SCLM might have permitted the preservation of the cratonic remnants within the metacraton. We discuss the possible preserved cratonic remnants within the SMC exploiting integrated modelling using geophysical, petrological, and geological information for the crust and the lithospheric mantle. The cratonic remnants are overlain by Paleozoic-Mesozoic sedimentary basins and they are surrounded and separated by topographic swells with known Cenozoic volcanism including the Hoggar Swell (HS), the Tibesti Massif (TM), and the Darfur Dome (DD). We find that the three cratonic remnants are underlain by a relatively thicker lithosphere (the thickest being beneath Al-Kufra reaching ~ 200 km) compared to the surrounding SMC, which has an average lithospheric thickness of ~ 150 km. We also find that the SCLM of the cratonic remnants to be relatively colder and denser, as seen by faster S-wave velocities compared to the surrounding SMC. Our preferred models are approached considering different lithospheric thicknesses beneath the SMC with a uniform moderately depleted composition (Proterozoic SCLM composition). We suggest that the presence of a relatively thinner lithosphere beneath the SMC, outside the cratonic remnants, was due to a combination of Precambrian events (partial SCLM delamination during the Neoproterozoic) and Phanerozoic events (Mesozoic-Cenozoic rifting and Cenozoic mantle volcanism).

Rare coding variants in the PLD3 gene coding for Phospholipase D3 were recently identified to increase the risk of developing late onset Alzheimer´s disease (LOAD). In order to address in more detail a possible influence of PLD3 on AD development, it was important to perform a fundamental biochemical characterization of PLD3 that provides a better understanding of its potential function. In this project evidence was provided that the type II transmembrane protein PLD3 is transported through the secretory and endocytic route to reach lysosomes. Using a newly described pathway, PLD3 is delivered to lysosomes via the endosomal sorting complex required for transport (ESCRT) machinery. An ubiquitination of PLD3 is required for its sorting into intraluminal vesicles of multivesicular bodies (MVBs). After fusion of MVBs with lysosomes, PLD3 is proteolytically processed into a stable luminal domain, to exert its main enzymatic function in this compartment. In the course of this project, PLD3 was identified as a lysosomal 5’ single-stranded acid exonuclease in immune cells. We could reveal that PLD3 is predominantly expressed in the brain, with highest expression in the hippocampus and the cerebral cortex. It could be further shown that PLD3 is the main 5’-exonuclease in the mouse brain. In Pld3-deficient mice, microglia alterations in the dentate gyrus of the hippocampus and the subventricular zone, both sites of adult neurogenesis, and a depression-like behavior were revealed. This suggests an important role of PLD3 in neuronal homeostasis, possibly affecting the onset and course of LOAD. Pld3 KO mice were bred with the 5xFAD Alzheimer´s disease (AD) mouse model. Although Pld3 was enriched in amyloid plaques (a hallmark of AD), no obvious changes on APP protein levels or cleavage products were observed in 5xFAD Pld3-/- mice, suggesting a more complex role of PLD3 in the etiology of AD.

By proteolysis, the enzymatic cleavage of peptide bonds, proteins undergo an irreversible posttranslational modification which is used for protein maturation, signaling and cellular adhesion. The work of this thesis approaches the role of the extracellular metalloproteases meprin α and meprin β in physiological and pathological conditions, especially in skin and inflammation related diseases. The first part of this thesis addresses the meprin expression in murine skin and their resulting role in wound healing and deposition of extracellular matrix in this organ. Based on previous findings that meprins are over-expressed in fibrotic and hyperkeratotic skin diseases, we analyzed mouse models over-expressing meprin β in skin in order to study its contribution to the onset of disease. Fra2 transgenic mice develop skin fibrosis and over-express meprin β in skin. Since Fra2 activates several other genes besides the meprin β gene, mouse models directly over-expressing meprin β in skin were generated. Epidermal over-expression of meprin β led to a pigmentary phenotype and hyperproliferation of keratinocytes. On the other hand, a dermal over-expression of meprin β did not show an obvious phenotype or development of fibrosis. The second part focused on proteolytic processing of the adhesion molecule CD99. Cleavage of CD99 by meprin β had an effect on transendothelial migration of cancer cells and inhibition of meprin β delayed infiltration of immune cells in an acute inflammation model. Finally, altered localization of meprin α was investigated during pathological conditions. Under normal conditions, soluble meprin α was observed to bind to epithelial cells via heparan sulphate, which had impact on infiltration of immune cells and epithelial barrier integrity. Moreover, covalent binding of meprin α to meprin β on the secretory pathway could also tether soluble meprin α to the plasma membrane. The altered localization of meprin α contributed to inflammatory disease progression.

In der vorliegenden Arbeit wurden zwei Fragestellungen zum Zuckerstoffwechseln in Archaeen untersucht. Zum einen wurde die allosterische Regulation von archaeellen Pyruvat Kinasen (PKs) sowie deren Evolution analysiert. Zum anderen wurde der Abbau des Desoxy-Zuckers L-Rhamnose in halophilen und hyperthermophilen Archaeen untersucht: PKs aus Bacteria und Eukarya werden durch Fructose-1,6-Bisphosphat oder AMP allosterisch aktiviert. In dieser Arbeit wurde gezeigt, dass 3-Phosphoglycerat (3PG) ein neuartiger allosterischer Aktivator der PKs von hyperthermophilen Archaeen der Ordnung Thermoproteales ist. Außerdem wurde ein 3PG-Bindemotiv identifiziert, mit dem es möglich ist Vorhersagen über die allosterische Aktivierbarkeit von PKs mit 3PG zu treffen. Für die PKs aus hyperthermophilen methanogenen Archaeen wurde eine allosterische Aktivierung durch den klassischen Aktivator AMP nachgewiesen. Im zweiten Teil dieser Arbeit wurde gezeigt, dass das halophile Euryarchaeon Haloferax volcanii Rhamnose über einen ABC-Transporter aufnimmt und oxidativ über einen Diketo-Hydrolase Weg zu Pyruvat und L-Lactat abbaut. Die Transkription der Gene der Rhamnose-Aufnahme und des –Abbaus werden durch den Transkriptionsregulator RhcR aktiviert. Ein Intermediat des Rhamnose-Abbaus wurde als Signalmolekül von RhcR identifiziert. Die Untersuchungen umfassen die Charakterisierung der entsprechenden Enyzme, Transkriptionsanalysen der beteiligten Gene sowie Wachstumsexperimente mit Knockout Mutanten. Weiterhin wurde gezeigt, dass der Diketo-Hydrolase Weg auch von den hyperthermophilen Crenarchaeota Vulcanisaeta distributa und Sulfolobus solfataricus für den Abbau von Rhamnose verwendet wird. In this thesis two questions concerning the sugar metabolism in archaea have been analysed. First, the allosteric regulation of archaeal pyruvate kinases (PKs) and their evolution were studied. Second, the degradation of the deoxy sugar L-rhamnose in halophilic and hyperthermophilic archaea was analysed: PKs from bacteria and eukarya are allosteric enzymes activated by fructose-1,6-bisphosphate or AMP. In this work, it is shown that PKs from hyperthermophilic archaea of the order Thermoproteales are allosterically activated by the novel effector 3-phosphoglycerate (3PG). A 3PG-binding motif could be identified, which allows to predict the allosteric regulation of PKs by 3PG. PKs from hyperthermophilic methanoarchaea were found to be activated by AMP, the classical activator from bacteria. In the second part of this work, it is shown that the halophilic euryarchaeon Haloferax volcanii takes up rhamnose by an ABC transporter and degrades rhamnose oxidatively to pyruvate and L-lactate via the diketo-hydrolase pathway. The genes involved in rhamnose uptake and degradation form a gene cluster, whose transcription is regulated by the transcriptional activator RhcR. An intermediate of rhamnose degradation was identified as the inducer molecule of RhcR. The study includes characterization of the respective enzymes, transcriptional analyses of involved genes and growth experiments with knockout mutants. Further, it is shown that the hyperthermophilic crenarchaeota Vulcanisaeta distributa and Sulfolobus solfataricus also degrades rhamnose via the diketo-hydrolase pathway.

Gelatinous zooplankton (GZ) are comprised of diverse taxonomic groups. The population outbreaks of some GZ species occurring in marine ecosystems have drawn increasing attention because of their eco-economic consequences in recent decades. In this context, many ecological questions about GZ have not been well documented. This dissertation aims to investigate essential biomolecules transfer and consequences on life history traits of Aurelia aurita under the combined effects of food quality, food quantity, and temperature. Besides, large-scale stable isotope data were assembled in order to discover the general community structure of GZ in the oceanic food web. The dissertation structures in the form of five chapters. The first chapter gives a general introduction to GZ in food webs and basic theory of bio-tracers. Chapter 2 displays and discusses the role of food quality combined with food quantity and temperature affecting essential biomolecules transfer in a three-trophic-level food chain encompassing A. aurita polyps. Chapter 3 is a follow-up study, which stresses these environmental factors influencing on life history traits of the moon jelly, A. aurita, polyps. In Chapter 4, the trophic ecology of oceanic GZ is presented. By applying stable isotope analysis in the eastern tropical Atlantic, the general jelly web structure and isotopic niche differentiation of different GZ taxonomic groups in the overall pelagic food web was assessed. Finally, Chapter 5 summarized the findings from the experimental and field studies on GZ and put forwarded outlooks for further GZ ecology research in both coastal and oceanic food webs. In summary, this dissertation stresses food quality as an important factor playing critical roles on essential biomolecules transfer in food webs and explaining polyps’ life history strategies under multiple stressors. Furthermore, as GZ play diverse and critical roles in aquatic food webs, the trophic “dead-end” concept of GZ should be reassessed.

The interaction of an organism with its environment is a hallmark of life and prerequisite for natural selection. Among the strongest evolutionary processes is the interaction between hosts and parasites that are engaged in a constant arms race of parasite exploitation and host defence. This antagonistic co-evolution is shaped through host and parasite genotypes, their local environmental conditions, and their potential for plastic responses. However, the relative contribution of these effects is often unclear. Here, I aimed to find answers to the questions how and why epidemiological traits vary among populations by using hosts and parasites from geographically distinct and ecologically divergent populations. I used three-spined sticklebacks (Gasterosteus aculeatus) as vertebrate model organisms to study defence mechanisms against helminth parasites. Helminth parasites are of exceptional interest because they can have complex immune modulatory effects on their hosts. This phenomenon is already applied in clinical settings, where helminths, their ova, or their products are used to treat autoimmune or inflammatory disorders (helminth therapy). Nevertheless, many questions on the specificity of the host-helminth interaction have yet to be answered. For instance: Are there differences between host genotypes or parasite species? What are the effects over time? Are effects localized or systemic? Using evolutionary and ecological perspectives, I specifically asked: What are the effects of host and parasite genotypes and their interaction? Does the potential for interaction effects differ with geographical scale? Does immune modulation differ over the time course of infection, and if so, is the temporal component dependent on parasite strain and/or host type? Indeed, my colleagues and I found that different strains of the same cestode species (Schistocephalus solidus) had profoundly different effects on divergent G. aculeatus types. This effect was linked to the co-evolutionary history and ecology of G. aculeatus and S. solidus. My results demonstrate that the infection outcome was largely determined by effects of host and parasite genotypes, while interaction effects were generally weak and only evident over the scale of continents. Gene expression profiles that differed between uninfected fish from different populations mostly converged upon infection. Thus, the parasite-induced phenotypic plasticity transcended host genetic differences. This thesis also reveals that S. solidus immune modulation is time-, host- and parasite strain-dependent. Sticklebacks that assumingly co-evolved with a highly virulent S. solidus strain were more resistant against S. solidus and had a well-orchstrated immune response (potentially diminishing immunopathological side effects) compared to hosts without this co-evolutionary background. Late stages of infection with a highly virulent S. solidus strain had a systemic effect by increasing the susceptibility towards another helminth species (Diplostomum pseudospathaceum). My data present a snapshot in time and space that provides insights into potential (co-) evolutionary backgrounds. Whether the epidemiological traits of Gasterosteus aculeatus and Schistocephalus solidus are indeed shaped through co-evolution is one of the challenges for future investigations. However, by revealing the dominant effect of the parasite and the relative importance of induced plasticity, this thesis advances our understanding about the role of each partner in a host-parasite interaction. My results are of significant importance for the investigation of the premises and consequences of helminth therapy. I propose to incorporate evolutionary and ecological perspectives in future research.

Next-Generation Sequencing (NGS) was initially used as an experimental method in exploratory research and in the search for novel scientific insights. In precision medicine additional critera apply, for example the testing method must answer specific clinical questions, while being subject to stringent standards of economy, liability and reproducibility. In accredited clinical NGS testing, laboratories must now increasingly document their standard procedures for computational validation (in silico validation) of NGS results. In this dissertation I describe possible in silico validation procedures based on my publications on the Backmapping method and on the pibase and Vy-PER software tools. These procedures take into account potential sources of errors from the biology itself, the lab methods, and bioinformatic methods. My pibase publication A deals with the validation of individual single nucleotide substitutions and the systematic elimination of error sources. My Backmapping publication B looks at potential artifacts that are based on biological sequence similarity between different regions of the human genome. These similarities may or may not lead to bioinformatic ambiguities when sequences are aligned to a reference genome. In consequence, potential artifacts may crop up which may only be exposed with the help of alternative genomic references and alternative alignment methods. My Vy-PER publication C presents an improved method to detect patient-individual virus genome particle integrations into the patient’s genome and to eliminate artifacts.

Mit immer weiter voranschreitenden Genomsequenzierungen werden auch in immer mehr Arten unterschiedlichste Vertreter der Familie der Aquaporine identifiziert. Die meisten sind bis heute jedoch kaum untersucht. Dennoch konnten bis heute neben den ursprünglichen Aufgaben als Kanalproteine weitere physiologisch wichtige Funktionen identifiziert werden, welche die Bedeutung dieser Proteine weiter unterstreicht. Das Plasmodium falciparum Aquaporin 2 (PfAQP2) vereint dabei in seiner Primärstruktur viele Charakteristika von orthodoxen und unorthodoxen Aquaporinen und bringt darüber hinaus eine ungewöhnliche und interessante Topologie mit. Darüber hinaus verfügt P. falciparum über ein gut untersuchtes Aquaglyceroporin, welches über ein breites, physiologisch sehr relevantes Solutspektrum verfügt. Auch wenn nachgewiesen wurde, dass PfAQP1 nicht essentiell ist, kommt die Frage auf, warum der Parasit ein ähnliches Protein exprimieren sollte. In dieser Arbeit konnte gezeigt werden, dass in einem Hefebasierten Schwellungsassay, der durch Messung der Lichtstreuung durchgeführt wurde, keine Leitfähigkeit für Wasser oder Glycerol, die beiden Hauptsolute der MIP, von dem PfAQP2 ausgeht. Es konnten auch bei einer Veränderung des pH-Wertes keine Leitfähigkeit für Monocarboxylate oder Wasser festgestellt werden. An Proteoliposomen aus polaren E. coli Phospholipiden, in deren Membran zellfrei hergestelltes PfAQP2 rekonstituiert wurde, konnte ebenfalls keine Permeabilität für Wasser beobachtet werden. Dies deutet darauf hin, dass PfAQP2 mit der fehlenden Wasserleitfähigkeit eine der Kerneigenschaften aller Aquaporine, trotz ähnlicher Strukturmerkmale, nicht besitzt. Dies wäre im Einklang mit dem breiten Solutspektrum von PfAQP1. Weitere Untersuchungen hinsichtlich der Leitfähigkeit im zellfreien System und der Strukturaufklärung sind notwendig, um die Funktion dieses Proteins zu entschlüsseln.

Im Rahmen dieser Arbeit wurde die austrocknungsinduzierte Chlorophyllfluoreszenzlöschung (qdes) bei aeroterrestrischen Grünalgen untersucht. Es wurden Verfahren entwickelt um qdes vergleichbar zu messen. Es wurde gezeigt, dass qdes eine Schutzfunktion gegen Photoinhibition zukommt und dass es sich dabei um einen bei aeroterrestrischen Algen weit verbreitetes Phänomen handelt. qdes kann durch die Lichtbedingungen während Kultur und Austrocknung, Arabitol, Jod und Zink sowie den Einsatz von Nigericin, einem Protonophor, und DDT, einem Inhibitor für die Violaxanthindeepoxidase, manipuliert werden. Die Ergebnisse deuten darauf hin, dass qdes mehrere verschiedene Mechanismen zugrunde liegen, von denen einige vom Xanthophyll-Zyklus unabhängig sind. Auch wenn es Ähnlichkeiten zwischen osmotischem Stress und Austrocknung an der Luft gibt, so unterscheidet sich das Verhalten der Chlorophyllfluoreszenz bei den untersuchten Algen deutlich zwischen diesen beiden abiotischen Stressfaktoren. Objective of this doctoral thesis was to examine the desiccation-induced quenching of chlorophyll fluorescence (qdes) of aero-terrestrial green algae. Methods have been developed to comparably measure qdes . It has been demonstrated that qdes is attached to a protective function against photo inhibition and this being a widespread phenomenon among aero-terrestrial algae. qdes can be manipulated by the light conditions during culture and desiccation, by arbitol, iodine and zinc as well as by the use of nigericin, a protonophor, and DTT, an inhibitor for violaxanthin deepoxidase. The results suggest that qdes is based on various different mechanisms some of them being independent from the xanthophyll cycle. Although similarities existing between osmotic stress and desiccation in open air the behaviour of chorophyll fluorescence of the examined algae is significantly differing between these two abiotic stress factors.

This thesis focuses on the nature of oceanic Eddy Kinetic Energy (EKE), its generation and temporal variability. An Ocean General Circulation Model (OGCM) based on the NEMO code builds the foundation for these investigations. For a first case study, several simulations of a 1/4° configuration are used to investigate the temporal variability of EKE in the South Pacific Subtropical Countercurrent (STCC). Decadal changes in wind stress curl associated with the Interdecadal Pacific Oscillation (IPO) lead to up- and downwelling in the STCC, influencing the meridional density gradient and thereby STCC strength, baroclinic instability and the resulting EKE. An additional 30 to 40% of the local density anomalies can be explained by long baroclinic Rossby waves propagating into the region, modulating the decadal signal of the IPO’s influence in the STCC on interannual time scales. In a second case study, the model’s horizontal resolution is regionally increased to 1/20° in the North Atlantic to investigate different types of mesoscale eddies in the Labrador Sea. On decadal time scales, the temporal variability of EKE in the LS is driven by the large-scale atmospheric circulation. In the case of Convective Eddies (CE), local winter heat loss leads to deep convection, a baroclinically unstable rim-current is established along the edge of the convection area and generates EKE at mid-depth. The variations of EKE associated with the surface intensified Irminger Rings (IR) and Boundary Current Eddies are driven by the large-scale changes of the currents of the subpolar gyre. While IR play a vital role in stratifying large parts of the LS and thus suppressing deep convection, CE are the major driver of rapid restratification during and after deep convection. Diese Arbeit untersucht die kinetische Energie von mesoskaligen, ozeanischen Strömungen (EKE), ihre Entstehung, und zeitliche Variabilität. Ein Ozeanzirkulationsmodell basierend auf dem NEMO-Code bildet den Grundstein für die Studie. Für eine erste Fallstudie werden verschiedene Simulationen einer 1/4°-Konfiguration genutzt, um die zeitliche Variabilität der EKE im südpazifischen subtropischen Gegenstrom (STCC) zu untersuchen. Dekadische Schwankungen der Rotation der Windschubspannung, hervorgerufen durch die Interdekadischen Pazifischen Oszillation (IPO), führen zu Auftrieb und Absinken im STCC und beeinflussen den meridionalen Dichtegradienten sowie die Stärke des STCC, barokline Instabilität, und die resultierende EKE. Zusätzlich können 30 bis 40% der lokalen Dichteanomalien durch lange, barokline Rossbywellen erklärt werden, die in der Region eintreffen und das dekadische Signal des Einflusses der IPO auf interannualen Zeitskalen modulieren. In einer zweiten Fallstudie wird die horizontale Auflösung des Modells im Nordatlantik auf 1/20° erhöht um die verschiedenen Arten von mesoskaligen Strömungswirbeln in der Labradorsee zu untersuchen. Auf dekadischen Zeitskalen wird die zeitliche Variabilität der EKE in der LS von der großskaligen Zirkulation der Atmosphäre angetrieben. Im Falle der Konvektiven Wirbel (CE) führt lokaler Wärmeverlust im Winter zu Tiefenkonvektion, eine baroklin instabile Strömung entlang des Randes der Konvektionsregion entsteht und produziert EKE in mittleren Tiefen. Die Veränderungen der EKE, die den oberflächenintensivierten Irminger Ringen (IR) und Randstromwirbeln zuzuordnen ist, werden durch großskalige Schwankungen der Strömungen im subpolaren Wirbel verursacht. Während die IR eine essentielle Rolle in der Stärkung der Schichtung von großen Teilen der LS spielen und somit die Tiefenkonvektion erschweren, sind die CE der hauptsächliche Grund für die schnelle Wiederherstellung der Schichtung während und nach der Tiefenkonvektion.

With the rapid growth of energy production from intermittent renewable sources like wind and solar power plants, large-scale energy storage options are required to compensate for fluctuating power generation on different time scales. Compressed air energy storage (CAES) in porous geological formations is seen as a promising underground storage option for balancing short-term diurnal fluctuations. CAES is a power-to-power energy storage, which converts electricity to mechanical energy, i.e. highly pressurized air, and stores it in the subsurface. This thesis aims at investigating the feasibility, dimensioning the storage capacity, and assessing the induced hydraulic, thermal and geochemical impacts of a large-scale CAES operation in porous formations. For this, scenario based numerical simulations are used.

The constant race to outcompete other organisms, either from the same or another species drives the continuous emergence of new and, better-adapted individuals. As these phenotypes become increasingly specialized to the specific environmental conditions they are exposed too, they grow progressively divergent from both their ancestors as well as their conspecifics in other environmental contexts. In freshwater three-spined sticklebacks (Gasterosteus aculeatus), this has resulted in a number of specific phenotypes, which have adapted different reproductive strategies or have evolved habitat specific growth and parasite resistance. Each of which is characterized by specific adaptations to the environmental context they are exposed to and maintained through environmental pressures and sexual selection. A key difference between rivers and lakes are the different parasite communities they harbor. Parasites impose strong selection pressures on their stickleback host by diverting resources, restricting reproduction and promoting their host’s death. Hence, it is crucial for sticklebacks to minimize their parasite load, resulting in a co-evolutionary arms race between parasite and host. As the parasite community varies between habitat types they demand different adaptations, resulting in increasing divergence between sticklebacks in their respective habitats. The globally reoccurring distinction of river and lake habitats implies an equally reoccurring habitat dependent differentiation into specific river respectively lake ecotypes. Specializing to their environment bestows an ecological advantage onto the resident individuals. This is reinforced by sexual selection which privileges well-adapted phenotypes by actively selecting for such individuals with low parasite burdens and optimal immune gene composition. Incidentally, this selects against potential migrants from other habitats, thus generating genetic isolation, a prerequisite for speciation. Im konstanten Wettstreit mit anderen Organismen, sowohl der eigenen als auch anderer Arten, entstehen kontinuierlich neue und potenziell besser angepasste Individuen. Mit zunehmender Spezialisierung dieser Phänotypen für ihre spezifische Umwelt, differenzieren sie sich zusehends sowohl von ihren Vorfahren als auch von Artgenossen in anderen Habitaten. In Dreistachligen Stichlingen (Gasterosteus aculeatus) hat dies weltweit zu zahlreichen Phänotypen geführt welche unterschiedliche Anpassungen an die Nahrungsaufnahme, Fortpflanzungsstrategien oder Parasitenresistenzen etabliert haben. Jeder dieser Differenzierungen wird durch spezifische Anpassungen an das Habitat charakterisiert und durch Umwelteinflüsse und sexuelle Selektion beibehalten. Ein Hauptunterschied zwischen Flüssen und Seen sind die vorherrschende Parasitengemeinschaften. Diese üben einen starken Selektionsdruck auf ihre Wirte aus. Demzufolge ist für Stichlinge die Minimierung der parasitären Belastung ein Grundanliegen, welches in einem evolutionären Wettrüsten zwischen Parasiten und Wirt mündet. Im selben Maβe wie die Parasitengemeinschaften der verschiedenen Habitate variieren, stellen diese unterschiedlichen Anforderungen an die Stichlinge und fördern damit die Divergenz zwischen den Populationen der entsprechenden Habitate. Der weltweit widerkehrenden Unterschiede zwischen Fluss- und Seehabitat impliziert eine ebenso globale Differenzierung von spezifischen Ökotypen. Habitat spezifische Anpassungen verleihen den ortsansässigen Fischen einen ökologischen Vorteil und erlaubt es ihnen potentielle Migranten und suboptimal angepasste Individuen zu übertrumpfen. Dies wird durch sexuelle Selektion verstärkt welche gut angepassten Phänotypen privilegiert indem diese Individuen mit niedrigem Parasitenbefall und optimalen Immungenen bevorzugt. Ein Nebeneffekt dieser Selektion ist die Benachteiligung von Habitat fremden Tieren was wiederum zu genetisch Isolation führt, eine Grundvoraussetzung der Artbildung.

Seismic tomography is an imaging tool which allows to construct 3-D models of the Earth’s internal structure from observables of seismic waves. Surface wave tomography can be performed using earthquakes and ambient noise data and is sensitive to isotropic as well as anisotropic 3-D shear-wave velocity structure in broad depth ranges sampling the crust and the lithosphere-asthenosphere structure. In this study, surface wave tomography is performed to characterize the structure of the lithosphere-asthenosphere underneath the Mediterranean and the adjacent regions. We utilize a large database consists of 3800 teleseismic earthquakes recorded by 4500 broadband stations provided by IRIS and EIDA in a combination, for the first time, with waveform data from the Egyptian National Seismological Network (ENSN). An automated algorithm for inter-station phase velocities is applied to obtain fundamental mode phase velocities from this database (3.5 millions of waveforms). Path average dispersion curves are obtained by averaging the smooth parts of single-event dispersion curves. We calculated new high resolution Rayleigh and Love wave phase velocity maps using an unprecedentedly large number (200.000) of measurements in the period range from 8s-350s. In order to relate the local dispersion curves to 1-D velocity models as function of depth, the Particle Swarm Optimization (PSO) algorithm has been developed and implemented. The 3-D model has been constructed based on the obtained 1-D shear velocity model.

Die professionelle Kompetenz von Lehrkräften spielt für die Qualität von Unterricht und damit für den Lernerfolg von Schülerinnen und Schülern eine wichtige Rolle. Das Professionswissen stellt dabei einen Kernbereich der professionellen Kompetenz dar. Ein zentrales Ziel der Ausbildung angehender Lehrkräfte besteht daher in der Vermittlung eines umfangreichen und praxisrelevanten Professionswissens. Im Rahmen der vorliegenden Arbeit wurde ein Messinstrument für das Professionswissen angehender Physiklehrkräfte entwickelt und erprobt. Die damit verbundenen Forschungsziele sind die Untersuchung der Struktur des Professionswissens, die Messung der Ausprägung des Professionswissens in verschiedenen Studienjahren sowie die Untersuchung des Zusammenhangs zwischen Personenmerkmalen und der Ausprägung des Professionswissens. Das in der vorliegenden Arbeit schrittweise entwickelte und überprüfte Instrument zur Messung des Professionswissens angehender Physiklehrkräfte wurde in einer bundesweiten Studie an zwölf Universitären eingesetzt. Die Daten dieser Studie wurden sowohl klassisch als auch mit Hilfe einer Raschanalyse ausgewertet. Die Ergebnisse der vorliegenden Studie zeigen, dass beim Professionswissen im Fach Physik die Bereiche Fachwissen, fachdidaktisches Wissen und pädagogisches Wissen voneinander abgrenzbar sind und dass ihre Ausprägungen unabhängig voneinander gemessen werden können. Dabei zeigen sich starke Zusammenhänge zwischen Fachwissen und fachdidaktischem Wissen sowie zwischen fachdidaktischem Wissen und pädagogischem Wissen. In den Bereichen Fachwissen und fachdidaktisches Wissen konnte für die ersten vier Studienjahre eine positive Entwicklung nachgewiesen werden. Personenmerkmale, die diese positive Entwicklung begünstigen können sind zum einen schulpraktische Erfahrungen und zum anderen eine positive Einstellung und Motivation der angehenden Lehrkräfte im Hinblick auf den Studienerfolg und das Unterrichten. Teachers professional competence plays an important role for the quality of teaching and therefore for learning success in school. On this occasion, teachers’ professional knowledge is a core area of professional competence. Accordingly, one central goal of teachers’ education is to impart comprehensive and practice-oriented professional knowledge. Within the present thesis project, a measuring instrument for the professional knowledge of pre-service physics teachers was developed and tested. The related research objectives are to investigate the structure of professional knowledge, to measure the degree of professional knowledge in different academic years and to investigate relations between individual personal characteristics and the degree of professional knowledge. The measuring instrument for pre-service teachers’ professional knowledge that was developed within this thesis project was applied at twelve German universities. The collected data was evaluated using classical analysis and Rasch modelling. The results of the present thesis imply for the subject physics that the areas of content knowledge, pedagogical content knowledge and pedagogical knowledge are distinct areas and their degree can be measured independently. High correlations between content knowledge and pedagogical content knowledge as well as between pedagogical knowledge and pedagogical content knowledge have been found. For the areas of content knowledge and pedagogical content knowledge a positive development during the first four academic years could be observed. Individual personal characteristics that favour the evolvement of pre-service physics teachers’ professional knowledge are on the one hand school experiences and on the other hand a positive attitude towards academic success and teaching physics.

The ubiquity of plasmids in all prokaryotic phyla and habitats and their ability to transfer between cells marks them as prominent constituents of prokaryotic genomes. Many plasmids are found in their host cell in multiple copies. The multi-copy state of plasmids leads to an increased mutational supply of plasmid-encoded genes and genetically heterogeneous plasmid genomes. Nonetheless, the segregation of plasmid copies into daughter cells during cell division is considered to occur in the absence of selection on the plasmid alleles. Consequently, I hypothesize that genetic drift of plasmid alleles during cell division has implication for the evolutionary rates of plasmids by lowering the number of accumulating mutations that are expected from the mutational supply. To test this hypothesis, I performed an experimental evolution experiment of low- and high-copy non-mobile plasmids in an Escherichia coli host. The results of the experiment revealed that the evolutionary rate of multicopy plasmids does not reflect the increased mutational supply expected according to their copy number. The results further suggest that many plasmid mutations are quickly lost due to genetic drift during cell division. Here I term this special type of allele dynamics in the population as ‘segregational drift’. Thus, segregational drift of multicopy plasmids interferes with the retention and fixation of novel plasmid variants. Furthermore, an examination of the experimentally evolved hosts reveals a significant impact of the plasmid type on the host chromosome evolution. In conclusion, depending on the selection pressure on newly emerging variants, plasmid genomes may evolve slower than haploid chromosomes, regardless of their higher mutational supply. Plasmid copy number is thus an important determinant of plasmid evolvability due to the manifestation of segregational drift.

Die molekulare Spinschaltung ist für eine Vielzahl von Anwendungen interessant, wie beispielsweise für die Realisierung eines intelligenten schaltbaren Kontrastmittels für die Magnetresonanztomographie (MRT). Diesbezüglich ist der Light-Driven Coordination-Induced Spin State Switch (LD-CISSS) einer der erfolgreichsten Ansätze für die reversible Spinschaltung in homogener Lösung bei Raumtemperatur. Basierend auf diesem Konzept wird in dieser Dissertation die erste lichtgesteuerte molekulare Spinschaltung auf Fe(III)-Basis vorgestellt. Hierbei wird der Spin-Switching-Prozess durch einen photoschaltbaren Azopyridinliganden induziert, welcher zu einer reversiblen Schaltung zwischen High-Spin (S = 5/2) und Low-Spin (S = 1/2) führt. Um weitere Untersuchungen an diesem System vorzunehmen und es zu optimieren, erfolgten zahlreiche Synthesen zur Herstellung von überbrückten Porphyrinen und Azopyridinen. Zusätzlich stand die Funktionalisierung von Porphyrinen im Fokus, um das Schalten im bio-optischen Fenster zu ermöglichen. Hierbei wurden die Porphyrine mittels einer Diels-Alder-Reaktion in ihre reduzierten Derivate, Chlorine, Isobakteriochlorine und Bakteriochlorine überführt. Durch die neu entwickelte, sehr milde sowie effiziente Methode, Nickel unter Verwendung von Bis(cycloocta-1,5-dien)nickel in Porphyrine einzulagern, konnte erstmals die gesamte Reihe von Hydroporphyrinen mit Ni(II) im Tetrapyrrolkern hergestellt und charakterisiert werden. Desweiteren wurde das metallfreie Bakteriochlorin auf die Anwendbarkeit als Photosensibilisator untersucht und aufgrund der guten Eigenschaften weiterentwickelt. Das neu synthetisierte wasserlösliche Bakteriochlorin zeigt eine starke Absorption im Nahinfrarot-Bereich bei 733 nm und eine gute Quantenausbeute. Damit ist die Verbindung hervorragend für die Anwendung in der Photodynamischen Therapie geeignet. Molecular spin switching is of interest for a number of different applications, such as switchable contrast agents for magnetic resonance imaging (MRI). The Light-Driven Coordination-Induced Spin State Switch (LD-CISSS) concept is the most successful approach for reversible spin switching in homogeneous solution at room temperature. Based on this concept, this thesis presents the first light-controlled molecular spin switch based on Fe(III). This spin-switching process is induced by a photoswitchable azopyridine ligand. In this process the spin state can be reversibly switched between low-spin (S = 1/2) and high-spin (S = 5/2). In order to investigate and optimize this system, a large number of bridged porphyrins and azopyridines were synthesized. In addition, the functionalization of porphyrins has been studied to shift the switching frequencies into the bio-optical window. The porphyrins react as dienophiles with the highly reactive diene isobenzofuran in very high yields under mild conditions to the reduced derivatives, namely chlorins, isobacteriochlorins and bacteriochlorins. The new, very mild and efficient method to insert nickel into porphyrins at room temperature using Ni(COD)2 can be used to prepare the first whole series of hydroporphyrins with Ni(II) in the tetrapyrrolic core. Furthermore, the metal-free bacteriochlorins were investigated as potential photosensitizers. Synthetic modification improved the properties of those compounds. The novel water-soluble bacteriochlorin shows strong near-infrared absorption at 733 nm and good quantum efficiency. This chromophore is a promising candidate for photodynamic therapy.

The focus of this thesis lies on carbohydrates as mediators of bacterial adhesion. The process of bacterial adhesion was addressed from two perspectives regarding the bacteria and their adhesive organelles on the one hand and considering the adhesive glycosylated surfaces on the other hand. The glycosylated surfaces were investigated both on the molecular level and in a broader context as glycosylated lipid layers. Hence, this thesis presents the synthesis and analysis of new functional glycomimetics to explore bacterial adhesion and membrane behaviour.

This thesis investigates the crosstalk between the UPR and autophagy. Both pathways and their synergistic interactions within the intestinal epithelium are essential for cellular homeostasis. Insights provided in this study may contribute to the development of new therapeutic approaches in IBD.

Der Spinzustand von Nickel(II)porphyrinen steht in engem Zusammenhang mit der Porphyrinkonformation und der Koordination axialer Liganden. Ein intramolekularer kooperativer Effekt zur Kontrolle der axialen Koordination soll durch die Kombination dieser Eigenschaften erzeugt und damit ein neuartiger molekularer Schalter erhalten werden. Die Kontrolle der axialen Koordination erfolgt durch eine starre Brücke, die eine Pyridineinheit enthält. Die Komplexe werden als Hair-Clip-Porphyrine bezeichnet, da die Bewegung bei der axialen Koordination an das Schließen einer Haarspange (engl. hair clip) erinnert. Anhand von Dichtefunktionaltheorie-Rechnungen wurde aus über 20 Strukturen ein überbrücktes Nickel(II)porphyrin ausgewählt und vier Derivate der Leitstruktur synthetisiert. Das Schwefelderivat ist das erste vollständig fünffach koordinierte Nickel(II)porphyrin in Lösung und im Kristallgitter. Die Sauerstoffderivate koordinieren in Lösung zu unterschiedlichen Anteilen im dynamischen Gleichgewicht. Mittels NMR- und UV-Vis-Experimenten wurden die intramolekulare und intermolekulare Koordination der Komplexe untersucht und der Einfluss der Brückenstruktur nachgewiesen. Durch Sublimation wurden auf einer Silber(111)-Oberfläche bei 4 K geordnete Inseln der Porphyrinkomplexe erhalten und mittels Rastertunnelmikroskopie charakterisiert. Die Anregung mit einem Tunnelstrom erlaubt die selektive, reversible Schaltung der Komplexe auf der Oberfläche zwischen dem diamagnetischen low-spin- und dem paramagnetischen high-spin-Zustand. Diese Schalter sind vielversprechende Modellsysteme für die Entwicklung molekularer spintronischer Einheiten auf Oberflächen dar. Spin state, conformation and axial coordination are interconnected properties of nickel(II) porphyrins. Coupling the axial coordination to the conformation of the porphyrin yielding intramolecular cooperativity offers a novel approach towards molecular spin switches. Towards this end, the axial coordination of a pyridine moiety is controlled by the introduction of a rigid strap across a porphyrin backbone. The movement necessary to achieve axial coordination in these complexes is similar to closing a hair clip and hence, these molecules were coined hair clip porphyrins. Based on density functional theory calculations, one lead structure out of over 20 candidates was chosen and four derivatives were synthesized. The thioether compound was identified as the first exclusively five-coordinate nickel(II) porphyrin in solution and in the crystal lattice. Two oxygen derivatives exhibit partial axial coordination in a dynamic equilibrium. The axial coordination of the porphyrins was quantified by NMR and UV-vis experiments, indicating the geometry of the bridge has major impact on the dynamic equilibrium. Sublimation of the complexes resulted in ordered islands on a silver(111) surface at 4 K. Scanning tunnelling microscopy has proven selective, reversible switching between the diamagnetic low-spin and the paramagnetic high-spin state of single complexes on the surface by excitation with a tunnelling current. These new molecular spin switches therefore are promising model systems for the development of surface-mounted spintronic devices.

This thesis focussed on the detection and modelling of soil compaction at different spatial scales. At field scale, fieldwork aimed to analyse and describe the effects of different tillage practices and traffic intensities on the spatial distribution of soil compaction. A one-time inversion tillage was conducted to analyse this measure as a management option to reduce topsoil compaction in conservation tillage. At regional scale, the aim was to develop and apply a new model, which enables a daily assessment of soil compaction risk by considering soil management effects and the dynamic changes of soil properties. The field scale analyses revealed clear patterns of soil compaction depending on the kind of primary tillage practice and traffic intensity as indicated by penetration resistance measurements. Measurements of soil physical properties showed that soil density itself does not allow a functional assessment of soils; i.e. assessment of soil compaction always necessitates the combination of soil density and functionality measurements. The one-time inversion tillage removed the detected dense soil layer in the reduced tilled areas and led to approximately the same soil density as in the continuously conventionally tilled area. The improved soil functionality, which developed during long-term reduced tillage, remained. The SaSCiA-model ("spatially explicit soil compaction risk assessment") was developed to calculate daily soil compaction risk at regional scale by considering soil, weather, crop and machinery information. Spatial crop type information was derived from satellite data and daily soil moisture was calculated by integrating a soil moisture model. SaSCiA was applied to calculate the soil compaction risk on a daily basis for entire years for the two study areas. By considering the dynamic changes of soil moisture, the developed SaSCiA-model enables a detailed spatio-temporal assessment of soil compaction risk, which exceeds all currently available models. Die vorliegende Dissertation zielte auf die Identifikation und Modellierung der Bodenverdichtung unter Berücksichtigung verschiedener räumlicher Skalen ab. Auf der Feldskala sollten die Auswirkungen verschiedener Grundbodenbearbeitungen und Verkehrsintensitäten auf die räumliche Verteilung der Bodenverdichtung analysiert werden. Anschließend wurde eine einmalige wendende Bodenbearbeitung mit dem Pflug durchgeführt. Auf regionaler Skala war die Entwicklung und Anwendung eines Modells zur täglichen Bewertung des Bodenverdichtungsrisikos unter Berücksichtigung der dynamischen Änderungen der Bodeneigenschaften das Ziel. Die Analysen auf der Feldskala zeigten in Abhängigkeit von der Bodenbearbeitung und der Verkehrsintensität deutliche Muster der Bodenverdichtung, die durch Messungen des Eindringwiderstandes ermittelt wurden. Ergänzende Messungen der bodenphysikalischen Eigenschaften zeigten jedoch, dass der Eindringwiderstand keine funktionale Beurteilung ermöglicht. Die einmalige wendende Bodenbearbeitung mit dem Pflug entfernte die erkannte dichtere Bodenschicht in den reduziert bearbeiteten Flächen. Die verbesserte Bodenfunktionalität blieb jedoch nach dem einmaligen Pflugeinsatz erhalten. Das SaSCiA-Modell ("Spatially explicit Soil Compaction risk Assessment") wurde entwickelt, um das tägliche Bodenverdichtungsrisiko auf regionaler Skala zu berechnen. Dazu wurden Boden-, Wetter-, Pflanzen- und Maschineninformationen berücksichtigt. Räumliche Informationen über die Fruchtarten wurden aus Satellitendaten abgeleitet, die tägliche Bodenfeuchte wurde durch die Integration eines Bodenfeuchtemodells berechnet. Für zwei Untersuchungsgebiete wurde mit SaSCiA das Bodenverdichtungsrisiko für ganze Jahre in täglicher Auflösung berechnet. Durch die Berücksichtigung der dynamischen Veränderungen der Bodenfeuchte ermöglicht das entwickelte SaSCiA-Modell eine detaillierte räumlich-zeitliche Beurteilung des Bodenverdichtungsrisikos, die über alle derzeit verfügbaren Modelle hinausgeht.

In dieser Arbeit wurde gezeigt, dass die mitochondriale Amidoxim reduzierende Komponente in der Lage ist Hydroxamsäuren zu reduzieren. Hierbei wird die Reaktion mithilfe der Proteinpartner Cytochrom b5-Reduktase und Cytochrom b5 katalysiert. Hydroxamsäuren stellen ein wichtiges Pharmakophor in einer Vielzahl von Arzneistoffen (z.B. antibakteriell, zytostatisch, antiinflammatorisch) dar, sodass ihr Metabolismus für die Entwicklung neuer und bestehender Arzneistoffe von großem Interesse ist. Über die Hydroxamsäurefunktion können leicht mehrwertige Metallkationen chelatiert werden. Die reduzierte Form der Hydroxamsäuren, das Amid, ist hierzu nicht in der Lage. Somit kommt es bei der Reduktion durch mARC von Hydroxamsäuren zur Inaktivierung des Arzneistoffes. Zur Untersuchung wurden folgende Proteinquellen verwendet: subzelluläre Fraktionen, rekombinante Proteine, Zellkulturversuche und Organhomogenate von mARC2-Knockout-Mäusen. Somit sollte die Reduktion durch mARC bei der Entwicklung von neuen Arzneistoffen, welche eine Hydroxamsäurefunktion besitzen, stets bedacht werden. In this thesis it was demonstrated that the mitochondrial amidoxime reducing component is able to reduce hydroxamic acids. The reaction is catalyzed by the protein partners cytochrome b5 reductase and cytochrome b5. Hydroxamic acids display an important pharmacophore in a variety of drugs (e.g. antibacterial, cytostatic, antiinflammatory), so their metabolism is of great interest for the development of new and existing drugs. Via the hydroxamic acid function, multivalent metal cations can easily be chelated. The reduced form of hydroxamic acids, the amide, is not able to do this. Thus, the reduction of hydroxamic acids by mARC leads to inactivation of the drug substance. The following protein sources were used for the investigation: subcellular fractions, recombinant proteins, cell culture experiments and organ homogenates of mARC2 knockout mice. Therefore, reduction by mARC should always be considered in the development of new drugs with hydroxamic acid function.

Our motivation to study such kind of equations is the process equation associatied with the Quicksort algorithm, i.e. $$(Q(t))_{t\in[0,1]}\d \Big(U Q_1\big(1\wedge \tfrac{t}{U}\big) + (1-U) Q_2\big(0 \vee \tfrac{t-U}{1-U}\big) + C(U,t)\Big)_{t\in [0,1]}.$$ Given a sequence $(C, (T_1,\tau_1), (T_2,\tau_2),\ldots)$ we characterize all $D$-valued solutions of the generalized equation and show that every solution can be represented as the convolution of a solution to the inhomogeneous equation and a general solution of the homogeneous equation.

Das sulfatierte Peptidhormon Phytosulfokin (PSK) fördert das Spross- und Wurzelwachstum von Arabidopsis. In der Pflanze kodieren fünf Gene, PSK1 - PSK5, für PSK-Vorläuferproteine. Der Wachstumsfaktor wird durch zwei Rezeptoren, PSKR1 und PSKR2, perzipiert. Es konnte gezeigt werden, dass das Sprosswachstum bei osmotischem Stress durch die Signalvermittlung über PSKR1 unterstützt wird. Osmotischer Stress führt zur Induktion der Genexpression von PSKR1 und des PSK-Vorläufergens PSK3. Diese Art von Stress wirkt zum Teil über Abscisinsäure (ABA), die die Expression von PSKR1 und PSK2 - PSK4 induziert. Analysen mit PSKR-knockout-Mutanten zeigten, dass der PSK-Signalweg für die Akkumulation von ABA im Spross erforderlich ist. Bei osmotischem Stress unterdrückt er die Glykosylierung von ABA durch Inhibierung des UDP-Glucosyltransferasegens UGT71B6 und dadurch die ABA-Inaktivierung. Zusätzlich wird die Freisetzung von glucosegebundenem ABA über Hydrolyse durch Induktion des β-Glucosidasegens BG1 induziert. Dies führt zu einem Anstieg von freiem ABA bei osmotischem Stress. Die Regulation von Entwicklungsprozessen bei osmotischem Stress und in Antwort auf ABA durch den PSK-Signalweg zeigt sich auch in der frühen Keimlingsentwicklung beim Ergrünen der Kotyledonen in einem späteren Entwicklungsstadium der Pflanze bei der Penetration und Elongation von Lateralwurzeln. Microarraydaten zeigten, dass in Antwort auf PSK die Expression von 16 Transkriptionsfaktorgenen (TF-Genen) in der Wurzel reguliert wird. Die Genaktivität dieser TFs wurde überwiegend durch den PSK-Signalweg inhibiert. Es handelt sich um Mitglieder von sieben verschiedenen TF-Familien, die sowohl Entwicklungsprozesse als auch, in den meisten Fällen, Stressantworten regulieren. Wachstumsanalysen an Überexpressions- und T-DNA-Insertionslinien von 14 dieser Gene zeigten, dass das Wurzelwachstum durch sechs dieser TFs induziert wird, durch zwei reprimiert und dass sechs keinen Einfluss auf das Wachstum der Wurzel haben. The sulfated peptide hormone phytosulfokine (PSK) enhances shoot and root growth of Arabidopsis. PSK-precursor proteins are encoded by five genes, PSK1 - PSK5. The growth factor is perceived by the two receptors PSKR1 and PSKR2. It could be shown here that the shoot growth is maintained by PSKR1-signaling during osmotic stress. Osmotic stress caused an induction of the gene expression of PSKR1 and the PSK-precursor PSK3. Osmotic stress is partially transmitted by abscisic acid (ABA) which induced the expression of PSKR1 and PSK2 - PSK4. Analyses of PSKR-knockout mutants showed that PSK-signaling is needed for the accumulation of ABA in the shoot. During osmotic stress, it represses the glycosylation, and hence activation of ABA by inhibiting the UDP-glucosyltransferase UGT71B6. Additionally the hydrolysis-driven release of glucose-bound ABA is induced by the induction of the β-glucosidase gene BG1. This leads to an increase of free ABA during osmotic stress. The regulation of developmental processes during osmotic stress and in response to ABA through the PSK-signaling pathway was observed in the early seedling development in cotyledon greening and at a later developmental stage in the outgrowth and elongation of lateral roots. Microarray data showed a regulation of expression of 16 transcription factor (TF) genes in response to PSK in the root. The gene activity of these TFs was predominantly repressed by the PSK-signaling pathway. The identified TF proteins belong to seven different TF-families and regulate developmental processes as well as, in most cases, stress responses. Growth analyses of overexpression and T-DNA insertion lines of 14 of these genes showed a root growth induction by six TFs, a repression by two TFs and six did not affect the root elongation.

The availability of protein and DNA sequences in the second half of the 20th century revolutionised evolutionary biology. For the first time, it was possible to quantify genetic variation among individuals and populations. Using molecular data to understand past demography and natural selection became an attainable goal. In the era of whole-genome sequences, application of early theoretical results proved to be challenging. The stochastic nature of evolutionary processes acting on DNA sequences makes it hard to distinguish signal from noise. Although progress has been made, models of molecular evolution are still lagging behind the availability of sequence data. In this thesis I contribute to bridging this gap, even if slightly. My main result is the development of the integrated sequentially Markovian coalescent (iSMC) – a novel framework that jointly models the effects of ancestral demography, recombination heterogeneity (Chapter 1) and mutation heterogeneity (Chapter 2) in shaping genetic diversity along the genome. This principled approach represents a step towards more realistic models of Population Genetics. The consequences of intracellular stochasticity extend beyond DNA sequences, however. Due to randomness in the diffusion of key molecules, isogenic cells differ in their gene expression patterns – hence in their phenotypes – even in homogeneous environments. To avoid chaos, intracellular stochasticity must be tamed by natural selection. In Chapter 3, I leverage single-cell transcriptomics data to disentangle the factors that constrain gene expression noise. Although selection against elevated noise acts at different levels of organisation, I show that it responds primarily to the architecture of molecular networks. Trace elements (TEs) play crucial roles in regulating ocean processes including marine biogeochemical cycles, and are therefore vital to support marine life. Understanding the biogeochemical cycling of TEs requires knowledge of their sources/ sinks and transport in the oceans. Radium isotopes are suitable tools to study inputs of elements from the continental margins, as they are produced by the decay of thorium isotopes in sediments, and are soluble in seawater. Therefore, in this Ph.D. thesis, I used Ra isotopes to determine boundary TE fluxes from two diverse environments that constrain the major TE sources, including shelf sediments in an Arctic shelf region and in an eastern boundary system off the western African coast, as well as rivers such as the River Congo. First, the distributions of the dissolved and total dissolvable TEs (Cd, Fe, Ni, Cu, Zn, Pb, Mn, and Co) were investigated in the Chukchi Sea and 228Ra isotope was applied as tracer of benthic TE inputs. The results show that elevated benthic TE inputs on the Chukchi shelf provided suitable conditions for phytoplankton blooms. The 228Ra activities in spring appear to be 2-fold higher than in summer, which was a surprising observation and requires further investigation. Next, 228Ra was used to investigate the influence of the Congo River on surface ocean TE concentrations in the South Atlantic Ocean. The results show that the Congo River plume constitutes a large and unexpected input of TEs (Fe, Mn, and Co) into the South Atlantic. Finally, Ra isotopes were used to investigate a number of key ocean boundary processes in the Southeast Atlantic Ocean. Upwelling in the Benguela region is visible in the Ra distributions, and elevated Ra isotopes, Fe (II) and Si concentrations were observed in the Benguela oxygen minimum zone, possibly due to inputs from the reducing shelf sediments, or submarine groundwater discharge along the Namibian shelf. Spurenelemente spielen eine zentrale Rolle in der Regulation ozeanischer Prozesse einschließlich des marinen biogeochemischen Kreislaufs und sind dadurch eine zentrale Voraussetzung für das Leben im Meer. Das Verständnis der biogeochemischen Kreisläufe der Spurenelemente basiert auf dem Wissen um deren Quellen, Senken und Transportwege innerhalb des Ozeans. Die Radiumisotope, welche als Zerfallsprodukt von Thorium in Sedimenten entstehen, sind wasserlöslich und haben sich als nützliche Werkzeuge herausgestellt, die den Input von solchen Elementen aus den kontinentalen Hängen nachvollziehen lassen. Diese verwende ich in der vorliegenden Dissertation, um die bedeutendsten Spurenelementquellen zu beleuchten, die den Austausch zwischen Schelfsedimenten und dem angrenzenden Ozean vornehmen. Dies geschieht anhand zweier kontrastierender Umgebungen, dem arktischen Schelf und des östlichen „Grenzsystems“ vor der Küste Westafrikas, das auch Flusseinträge wie zum Beispiel aus dem Kongo beinhaltet. Zunächst wurde die Verteilung der gelösten und säurelöslichen Spurenelemente (Cd, Fe, Ni, Cu, Zn, Pb, Mn und Co) in der Tschuktschensee untersucht. Als Tracer zur Bestimmung benthischen Eintrags wurde dort 228Ra verwendet. Des Weiteren wurde 228Ra verwendet, um den Einfluss des Kongo, auf die im oberen Ozean befindlichen Spurenelemente im Südatlantik zu untersuchen. Die Ergebnisse zeigen, dass die Flusswasserfahne des Kongo einen unerwartet großen Eintrag bioessentieller Spurenelemente in den Südatlantik ausmacht. Mit Hilfe der Radiumisotope wurden abschließend eine Reihe von Schlüsselprozessen im Grenzflächenaustausch des Südost-Atlantiks untersucht. Der Auftrieb der Benguela-Region ist ebenfalls in der Radiumverteilung ersichtlich. Gleichzeitig weisen erhöhte Konzentrationen der Radiumisotope, Fe (II) und Si in der Benguela-Sauerstoffminimumzone auf Zufuhr aus reduzierenden Schelfsedimenten oder aus unterseeischem Grundwasseraustritt entlang des Namibianischen Schelfs hin.

Die Dissertation behandelt Untersuchungen zur photokatalytischen Wasserstofferzeugung mit Hilfe von CuCo2S4 geträgert auf TiO2 und Untersuchungen der sensitisierten photokatalytischen Wasserstofferzeugung mit zwei verschiedenen Oxothiostannaten. Enthalten ist auch die Untersuchung der photokatalytische Degradation des Farbstoffs y-Eosin mit Hilfe von ZnCr2O4. The thesis deals with photocatalytic hydrogen generation studies using CuCo2S4 supported on TiO2 and sensitized photocatalytic hydrogen generation studies with two different oxothiostannates. Also included is the investigation of the photocatalytic degradation of the dye y-Eosin using ZnCr2O4.
