Abstract

The International Fusion Materials Irradiation Facility - Demo Oriented NEutron Source (IFMIF-DONES) is a single-sited novel Research Infrastructure for testing, validation and qualification of the materials to be used in a fusion reactor. The main purpose of IFMIF-DONES is related to fusion technology and neutron irradiation of the materials to be used in the future fusion power plants. However, there is an important effort to take advantage of the outstanding characteristics of the facility in terms of neutrons and deuterons. One of the applications could be radioisotopes production with deuterons and neutrons. We discuss here the possible production of radioisotopes with deuterons at DONES. In this work, we have focused on the production of 177Lu with deuterons. The study has been carried out through the design and simulation of a device as cooling system for the sample producing the radioisotope. The results show the viability of using DONES for such production. In addition, the study suggests that new nuclear energy data above 20 MeV for deuterons is mandatory for an accurate study in this field., Comment: 7 pages, 8 figures

Abstract

Research Question: Could a modification in the ovarian tissue freezing protocol improve follicle survival after cryopreservation and xenotransplantation?, Design: Ovarian tissue was used from 13 adult patients, frozen either with our original protocol, or a modified version involving a higher concentration of dimethyl sulphoxide (DMSO), larger volume of cryopreservation solution and lower seeding temperature. After thawing, the ovarian fragments were xenotransplanted to six mice with severe combined immunodeficiency (SCID) for 3 weeks., Results: The proportion of primordial follicles decreased, and the proportion of growing follicles increased significantly (all P < 0.01) after cryopreservation and xenografting compared with fresh controls for both protocols. Follicle density, development, ultrastructure and function were similar between treatments., Conclusions: This study showed that, although the higher DMSO concentration did not improve survival of preantral follicles, it did not seem to induce any major toxicity in the follicle population either., (Copyright © 2018 Reproductive Healthcare Ltd. Abstract

Cerebral damage secondary to the vasospasm due to subarachnoid hemorrhage (SAH) is an important cause of morbid-mortality. We propose the use of the PET tracer [ 18 F]Fluoromisonidazole to visualize the hypoxia due to the vasospasm. On the other hand [ 18 F]Fluoromisonidazole synthesis process was optimized, avoiding HPLC purification using SPE cartridges instead, and reducing some synthesis steps. [ 18 F]Fluoromisonidazole in vitro stability was tested for ten hours, and in vivo PET/CT images showed higher cerebral uptake in hemorrhagic animals than in control rats., (Copyright © 2017 Elsevier Ltd. Abstract

Acromegaly is a disorder resulting from excessive production of growth hormone (GH) and consequent increase of insulin-like growth factor 1 (IGF-I), most frequently caused by pituitary adenomas. Elevated GH and IGF-I levels results in wide range of somatic, cardiovascular, endocrine, metabolic, and gastrointestinal morbidities. Subcutaneous implantation of the GH-secreting GC cell line in rats leads to the formation of tumors. GC tumor-bearing rats develop characteristics that resemble human acromegaly including gigantism and visceromegaly. However, GC tumors remain poorly characterized at a molecular level. In the present work, we report a detailed histological and molecular characterization of GC tumors using immunohistochemistry, molecular biology and imaging techniques. GC tumors display histopathological and molecular features of human GH-producing tumors, including hormone production, cell architecture, senescence activation and alterations in cell cycle gene expression. Furthermore, GC tumors cells displayed sensitivity to somatostatin analogues, drugs that are currently used in the treatment of human GH-producing adenomas, thus supporting the GC tumor model as a translational tool to evaluate therapeutic agents. The information obtained would help to maximize the usefulness of the GC rat model for research and preclinical studies in GH-secreting tumors.
