Abstract

We read with interest the letter by Giammarile et al. [1] addressing our editorial in which we proposed that the European Medicines Agency should allow the option of a dosimetry-based approach to the treatment of cancer with radionuclide therapy [2]. Our editorial was intended to draw attention to the potential legal issues of recommending an approach to treatment that could contravene the European Council Directive 2013/59 [3] and national legislation, as the directive (article 56) states that “For all medical exposure of patients for radiotherapeutic1 purposes, exposures of target volumes shall be individually planned and their delivery appropriately verified...”. This directive is intended to “lay down basic safety standards for the protection of dangers arising from exposure to ionizing radiation” and emphasizes the need for ‘justification’ and ‘optimization’ of intentional radiation exposures of patients. Abstract

International audience; In this study, coriander straw fiber was effectively incorporated as a reinforcing filler in polypropylene and biobased low-density polyethylene composite materials through twin-screw extrusion compounding and injection molding. Maleic anhydride-grafted copolymers were added as a coupling agent and effectively provided fiber/matrix compatibilization. With a significant reinforcing effect, resulting in a 50% increase in the flexural and tensile strength (from 19 to 28 MPa and from 12 to 17 MPa, respectively, for polypropylene composites) as compared to the native polymer, coriander straw allowed the production of 40% filled thermoplastic biocomposites with adequate mechanical properties comparable to those of commercial wood fibers, excellent durability in terms of UV and hygrothermal weathering and high potential for recycling. At the same time, such coriander biocomposites show a favorable cost structure, with 28% reduction of the granule cost as compared to wood fiber composites.

Abstract

New synthetic pathways are proposed for the synthesis of a new set of bolaamphiphilic derivatives starting from microbiologically produced sophorolipids. A total set of 43 new derivatives was synthesized Via reductive amination of a previously synthesized sophorolipid aldehyde with diamines and primary amines. The new derivatives were evaluated for their antimicrobial activity against Gram-negative and Gram-positive bacteria. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values were determined for the active compounds. Transfection efficiencies were also evaluated for some of the deprotected derivatives via the assessment of their capacity to transfect three different eukaryotic cell lines in vitro. Finally, the self-assembly properties were evaluated for the deprotected derivatives. Antimicrobial activities were mostly observed for the peracetylated mono- or dicationic bolaamphiphiles and only the deprotected monocationic bolaamphiphile with an octadecyl chain on the nitrogen atom was eligible for the evaluation of its transfection properties. Evaluation of the self-assembly properties indicated that the presence of an octadecyl chain was necessary for micelle formation. Both micelle formation and the net charge of the compounds seem to have an influence on the antimicrobial activity and transfection efficiency. These results are promising for use of bolaamphiphilic sophorolipids in medical and self-assembly applications., Dans ce travail de nouveaux composés amphiphiles cationiques sont préparés à partir de sophoro lipides. Abstract

We present in our work numerical results on the performance of a $\mathbf {4 \times 4}$ swirl-topology photonic reservoir integrated on a silicon chip. Nonlinear microring resonators are used as nodes. We analyze the performance of such a reservoir on a classical nonlinear Boolean task (the delayed XOR task) for: various designs of the reservoir in terms of lengths of the waveguides between consecutive nodes, and various injection parameters (injected power and optical detuning). From this analysis, we find that this kind of reservoir can perform–for a large variety of parameters–the delayed XOR task at 20 Gb/s with bit error rates lower than $\mathbf {10^{-3}}$ and an averaged injection power lower than 2.5 mW.

Abstract

International audience; The “intended purpose” in all therapeutic exposures is treatment efficacy against malignant disease. The optimization principle (as low as reasonably achievable, ALARA) of article 56, when applied in a therapy situation, states that absorbed doses to nontarget tissues should be kept reasonably low, but not so low as to lose efficacy. We think that this applies above all to the fight against life-threatening cancer. As a consequence, we believe that to adhere to the optimization principle in oncological patients, nuclear medicine therapy should be based on individualized dosimetry.

Abstract

International audience; The vascular network is a major target of ischemia-reperfusion, but has been poorly investigated in renal transplantation. The aim of this study was to characterize the remodeling of the renal vascular network that follows ischemia-reperfusion along with the most highly affected cortex section in a preclinical renal transplantation model. There were two experimental groups. The first was a grafted kidney group consisting of large white pigs for which the left kidney was harvested, cold flushed, preserved for 24 h in the University of Wisconsin’s preservation solution, and then auto-transplanted (n = 5); the right kidney was removed to mimic the situation of human kidney transplantation. The second group (uni-nephrectomized kidney group) consisted of animals that underwent only right nephrectomy, but not left renal transplantation (n = 5). Three months after autotransplantation, the kidneys were studied by X-ray microcomputed tomography. Vessel morphology and density and tortuosity of the network were analyzed using a 3D image analysis method. Cortical blood flow was determined by laser doppler analysis and renal function and tissue injury assessed by plasma creatinine levels and histological analysis. Renal ischemia-reperfusion led to decreased vascular segment volume associated with fewer vessels of less than 30 μm, particularly in the inner cortex:0.79 ± 0.54% in grafted kidneys vs. 7.06 ± 1.44% in uni-nephrectomized kidneys, p < 0.05. Vessels showed higher connectivity throughout the cortex (the arborescence factor of the whole cortex was less in grafted than uni-nephrectomized kidneys 0.90 ± 0.04 vs. 1.07 ± 0.05, p < 0.05, with an increase in the number of bifurcations). Furthermore, cortical blood flow decreased early in kidney grafts and remained low three months after auto-transplantation. The decrease in microvasculature correlated with a deterioration of renal function, proteinuria, and tubular dysfunction, and was associated with the development of fibrous tissue. This work provides new evidence concerning the impact of ischemia-reperfusion injuries on the spectrum of renal vascular diseases and could potentially guide future therapy to preserve microvessels in transplantation ischemia-reperfusion injury.

Période 2000-2012. http://prodinra.inra.fr/record/256699; International audience; BACKGROUND: Saponins are a class of secondary plant metabolites consisting of a sugar moiety glycosidically linked to a hydrophobic aglycone (sapogenin) that often possess insecticidal activities. Four saponins were selected: two triterpene saponins, Q. saponaria saponins and aescin, and two steroidal saponins, digitonin and diosgenin. Their effects were investigated on an important pest species and a model piercing-sucking insect, the pea aphid Acyrthosiphon pisum. The triterpene Q. saponaria saponins bark saponin received special attention because of its high activity. Aphids were challenged by oral and contact exposure to demonstrate aphicidal activities, and in choice experiments to support use as a natural deterrent. RESULTS: When aphids were exposed to supplemented artificial diet for 3 days, a strong aphicidal activity was recorded for three of the four saponins, with an LC(50) of 0.55 mg mL(-1) for Q. saponaria saponins, 0.62 mg mL(-1) for aescin and 0.45 mg mL(-1) for digitonin. The LT(50) values ranged between 1 and 4 days, depending on the dose. For diosgenin, only low toxicity (14%) was scored for concentrations up to 5 mg mL(-1) . In choice experiments with treated diet, a deterrence index of 0.97 was scored for Q. saponaria saponins at 1 mg mL(-1) . In contrast, direct contact showed no repellent effect. Spraying of faba bean plants with Q. saponaria saponins resulted in an LC(50) of 8.2 mg mL(-1) . Finally, histological analysis in aphids fed with Q. saponaria saponins demonstrated strong aberrations of the aphid gut epithelium, and exposure of midgut CF-203 cell lines to Q. saponaria saponins in vitro confirmed the cytotoxic effect. CONCLUSIONS: The present insect experiments provide strong evidence that saponins, as tested here with triterpene Q. saponaria saponins, can be useful as natural aphicides and deterrents. Furthermore, the insect midgut epithelium is suggested to be a primary target of saponin activity.Copyright © 2011 Society of Chemical Industry.

Abstract

International audience; The Arabidopsis MAP65s are a protein family with similarity to the microtubule-associated proteins PRC1/Ase1p that accumulate in the spindle midzone during late anaphase in mammals and yeast, respectively. Here we investigate the molecular and functional properties of AtMAP65-5 and improve our understanding of AtMAP65-1 properties. We demonstrate that, in vitro, both proteins promote the formation of a planar network of antiparallel microtubules. In vivo, we show that AtMAP65-5 selectively binds the preprophase band and the prophase spindle microtubule during prophase, whereas AtMAP65-1-GFP selectively binds the preprophase band but does not accumulate at the prophase spindle microtubules that coexists within the same cell. At later stages of mitosis, AtMAP65-1 and AtMAP65-5 differentially label the late spindle and phragmoplast. We present evidence for a mode of action for both proteins that involves the binding of monomeric units to microtubules that "zipper up" antiparallel arranged microtubules through the homodimerization of the N-terminal halves when adjacent microtubules encounter.
