A survey on "diversity of insect pollinators on Litchi chinensis flower" was carried out in the Horticulture Experimental Farm, Assam Agricultural University, Jorhat during 2015 and 2016. The survey revealed that insect pollinators belonging to seven families viz., Apidae, Vespidae, Papilionidae, Muscidae, Formicidae, Pyralidae and Syrphidae were commonly available out of which, Apis cerena was found to be dominant with 49.50 per cent while the lowest per cent abundance was found to be Pieris rapae with 2.66 per cent. Mola carplet (Amblypharyngodon mola) is one of the most popular small fish species of the Indian subcontinent. There are limited studies on captive breeding of this species, which is important for aquaculture and the conservation prospects of this species. The conventional induced breeding method using an inducing agent (GnRHa and dopamine antagonist) is one of the most effective and prevalent methods of breeding fish. It is a laborious and time-consuming process, particularly in mass fish breeding and in lieu of that, a less time-consuming method - sympathetic induction of the broodstock, is used in some regions of India, particularly in big carp fish. However, this method has not been reported in commercial-scale breeding of small indigenous fish species. Therefore, an experiment was conducted to compare the spawning efficiency of Amblypharyngodon mola bred by sympathetic induction with the conventional complete induced breeding method. The spawning performance in terms of latency period, relative fecundity, fertilization rate, incubation period, and hatching rates of sympathetically induced Amblypharyngodon mola were compared to completely induced Amblypharyngodon mola brooders. Although the latency period (7.8 hrs), relative fecundity (39 nos./g), fertilization rates (81.61%) and spawning efficiency coefficient (0.681) were better in conventionally induced fish, but lower post-spawning mortality (1.29%) and better hatching rates (86.21%) were observed in sympathetically induced fish. The results indicate that quality offspring of Amblypharyngodon mola could be obtained in terms of survivability through sympathetic breeding. Background. Spindle cell/sclerosing rhabdomyosarcoma is a rare neoplasm and has an aggressive clinical course. Because of its rarity, we performed a multi-institutional collaboration to comprehend the overarching clinical, histopathological, and immunohistochemical characteristics of a cohort of spindle cell/sclerosing rhabdomyosarcoma. Materials and Methods. Forty-five patients with spindle cell/sclerosing rhabdomyosarcoma were identified. Demographics, clinical, histopathological, and immunohistochemistry data were reviewed and recorded. Results. The patients' age ranged from 1 to 85 years with a male to female ratio of 1.2:1. There were 15 children/adolescents and 30 adults. Eighteen (40%) tumors were located in the head and neck region. Twenty-four (53%) tumors displayed a bimorphic cellular arrangement with hypercellular areas having short, long, and sweeping fascicular and herringbone pattern, and hypocellular areas with stromal sclerosis and associated hyalinized and/or chondromyxoid matrix. Histomorphological differentials considered were leiomyosarcoma, malignant peripheral nerve sheath tumor, fibrosarcoma, nodular fasciitis, liposarcoma, synovial sarcoma, sarcomatoid carcinoma, solitary fibrous tumor, dermatofibrosarcoma protuberans, and schwannoma. Six tumors exhibited marked stromal sclerosis. The myogenic nature was confirmed by immunohistochemistry. Positivity for at least one skeletal muscle-associated marker (MyoD1 and/or myogenin) was observed. Conclusion . Spindle cell/sclerosing rhabdomyosarcoma diagnosis can be challenging as a number of malignant spindle cell neoplasm mimic this entity. Thus a correct diagnosis requires immunohistochemical work up with a broad panel of antibodies. Deepor Beel, located in the state of Assam in India, is a Wetland of International Importance with a Wildlife Sanctuary and is the only RAMSAR site in the state. Though of invaluable ecological significance, the wetland is facing anthropogenic stressors, leading to rapid degradation of ecological health. In December 2022, surface water was collected from six stations of Deepor Beel to elucidate biological communities using the eDNA approach. At the time of sampling, in-situ environmental parameters were measured in triplicates. The dissolved nutrients and concentrations of metals and metalloids were estimated using UV-Vis Spectrophotometry and ICP-MS approaches respectively. The study revealed a high concentration of dissolved nitrate in the surface water. High-throughput sequencing using Nanopore sequencing chemistry in a MinION platform indicated the overwhelming abundance of Moraxellaceae (Prokaryotes) and Eumetazoa (Eukaryotes). The abundance of Cyprinidae were also encountered in the studied wetland reflecting the biodiversity of fish populations. High nitrate along with elucidated microbial signals are crucial to designate ecological health status of Deeper Beel. We demonstrate fabrication of photodetectors in the UVC and UVA regions, based on surface engineering of Mn 2+ -doped ZnS Qdot. Mn 2+ -doped ZnS Qdot exhibited UVC detection with a responsivity of 0.3 ± 0.02 A·W -1 and detectivity of 1.7 ± 0.2 10 11 Jones. Following this, the Qdot was surface modified with 8-hydroxyquinoline 5-sulfonic acid ligand, which resulted in the formation of a bluish green zinc quinolate complex (Zn(QS) 2 ) at the Qdot surface (defined as the quantum dot complex, QDC) exhibiting overall white photoluminescence. The detector developed with QDC as the photoactive material exhibited a responsivity of 0.2 ± 0.02 A·W -1 and detectivity of 1.2 ± 0.2 10 11 Jones in the UVA band. This shift in the detection band from UVC in Qdot to UVA in QDC, through the surface complexation mechanism, is a new approach for tuning spectral detection featured in this work. Besides, the self-powered response of both the detectors exhibited attractive photoelectric characteristics. The detectors were incorporated in a portable prototype to show their potential application toward selective UVC and UVA spectral detection. Additionally, the dual-mode emission of the QDC was used for data encryption and decryption.

The conventional paper-based system for malaria surveillance is time-consuming, difficult to track and resource-intensive. Few digital platforms are in use but wide-scale deployment and acceptability remain to be seen. To address this issue, we created a malaria surveillance mobile app that offers real-time data to stakeholders and establishes a centralised data repository. The MoSQuIT app was designed to collect data from the field and was integrated with a web-based platform for data integration and analysis. The MoSQuIT app was deployed on mobile phones of accredited social health activists (ASHA) working in international border villages in the northeast (NE) Indian states of Assam, Tripura and Arunachal Pradesh for 20 months in a phased manner. This paper shares the challenges and opportunities associated with the use of MoSQuIT for malaria surveillance. MoSQuIT employs the same data entry formats as the NVBDCP's malaria surveillance programme. Using this app, a total of 8221 fever cases were recorded, which included 1192 (14.5%) cases of P. falciparum malaria, 280 (3.4%) cases of P. vivax malaria and 52 (0.6%) mixed infection cases. Depending on network availability, GPS coordinates of the fever cases were acquired by the app. The present study demonstrated that mobile-phone-based malaria surveillance facilitates the quick transmission of data from the field to decision makers. Geospatial tagging of cases helped with easy visualisation of the case distribution for the identification of malaria-prone areas and potential outbreaks, especially in hilly and remote regions of Northeast India. However, to achieve the full operational potential of the system, operational challenges have to be overcome.

The aetiology of non-malaria vector-borne diseases in malaria-endemic, forested, rural, and tribal-dominated areas of Dhalai, Tripura, in north-east India, was studied for the first time in the samples collected from malaria Rapid Diagnostic Kit negative febrile patients by door-to-door visits in the villages and primary health centres. Two hundred and sixty serum samples were tested for the Dengue NS1 antigen and the IgM antibodies of Dengue, Chikungunya, Scrub Typhus (ST), and Japanese Encephalitis (JE) during April 2019-March 2020. Fifteen Dengue, six JE, twelve Chikungunya, nine ST and three Leptospirosis, and mixed infections of three JE + Chikungunya, four Dengue + Chikungunya, three Dengue + JE + Chikungunya, one Dengue + Chikungunya + ST, and one Dengue + ST were found positive by IgM ELISA tests, and four for the Dengue NS1 antigen, all without any travel history. True prevalence values estimated for infections detected by Dengue IgM were 0.134 (95% CI: 0.08-0.2), Chikungunya were 0.084 (95% CI: 0.05-0.13), Scrub were 0.043 (95% CI: 0.01-0.09), and Japanese Encephalitis were 0.045 (95% CI: 0.02-0.09). Dengue and Chikungunya were associated significantly more with a younger age. There was a lack of a defined set of symptoms for any of the Dengue, Chikungunya, JE or ST infections, as indicated by the k-modes cluster analysis. Interestingly, most of these symptoms have an overlapping set with malaria; thereby, it becomes imperative that malaria and these non-malaria vector-borne disease diagnoses are made in a coordinated manner. Findings from this study call for advances in routine diagnostic procedures and the development of a protocol that can accommodate, currently, in practicing the rapid diagnosis of malaria and other vector-borne diseases, which is doable even in the resource-poor settings of rural hospitals and during community fever surveillance.

Background: A surveillance system is the foundation for disease prevention and control. Malaria surveillance is crucial for tracking regional and temporal patterns in disease incidence, assisting in recorded details, timely reporting, and frequency of analysis., Objective: In this study, we aim to develop an integrated surveillance graphical app called FeverTracker, which has been designed to assist the community and health care workers in digital surveillance and thereby contribute toward malaria control and elimination., Methods: FeverTracker uses a geographic information system and is linked to a web app with automated data digitization, SMS text messaging, and advisory instructions, thereby allowing immediate notification of individual cases to district and state health authorities in real time., Results: The use of FeverTracker for malaria surveillance is evident, given the archaic paper-based surveillance tools used currently. The use of the app in 19 tribal villages of the Dhalai district in Tripura, India, assisted in the surveillance of 1880 suspected malaria patients and confirmed malaria infection in 93.4% (114/122; Plasmodium falciparum), 4.9% (6/122; P vivax), and 1.6% (2/122; P falciparum/P vivax mixed infection) of cases. Digital tools such as FeverTracker will be critical in integrating disease surveillance, and they offer instant data digitization for downstream processing., Conclusions: The use of this technology in health care and research will strengthen the ongoing efforts to eliminate malaria. With India aiming to achieve malaria elimination by 2030, several strategies have been put in place. With that aim, mass surveillance is now being conducted in some malaria-endemic pockets. As dry season mass surveillance has been shown to have its importance in targeting the reservoir, a study was undertaken to assess the parasite load by a sensitive molecular method during one of the mass surveys conducted in the dry winter period. It was executed in two malaria-endemic villages of Dhalai District, Tripura, in northeast India, also reported as P. falciparum predominated area. The present study found an enormous burden of Rapid Diagnostic Test negative malaria cases with P. vivax along with P. vivax and P. falciparum mixed infections during the mass surveillance from febrile and afebrile cases in dry winter months (February 2021-March 2021). Of the total 150 samples tested, 72 (48%) were positive and 78 (52%) negative for malaria by PCR. Out of the 72 positives, 6 (8.33%) were P. falciparum , 40 (55.55%) P. vivax , and 26 (36.11%) mixed infections. Out of 78 malaria negative samples, 6 (7.7%) were with symptoms, while among the total malaria positive, 72 cases 7 (9.8%) were with symptoms, and 65 (90.2%) were asymptomatic. Out of 114 samples tested by both microscopy and PCR, 42 samples turned out to be submicroscopic with 4 P. falciparum , 23 P. vivax , and 15 mixed infections. Although all P. vivax submicroscopic infections were asymptomatic, three P. falciparum cases were found to be febrile. Evidence of malaria transmission was also found in the vectors in the winter month. The study ascertained the use of molecular diagnostic techniques in detecting the actual burden of malaria, especially of P. vivax , in mass surveys. As Jhum cultivators in Tripura are at high risk, screening for the malarial reservoirs in pre-Jhum months can help with malaria control and elimination.

The excited-state reductive elimination (RE) activities of a metal-metal bonded PdIII2 complex, [(2-phenylpyridyl)Pd(μ-acetate)Cl]2, are described. The C-Cl bond-forming RE reaction is accelerated by up to five orders of magnitude upon visible photoexcitation, which induces the Pd-Pd bond dissociation. This ligand field variation reduces the energy cost for intramolecular charge transfer (ICT) from coupling substrate to the metal center involved in the RE reaction. The correlation found between the ligand field and the RE activity indicates the ICT energy as a fundamental descriptor for RE reactions.

Purpose: Age-related macular degeneration (AMD) is one of the leading causes of irreversible central vision loss in the elderly population. The current study aims to find non-invasive prognostic biomarkers in the urine specimens of the AMD patients., Methods: Peripheral blood and urine samples were collected from 23 controls and 61 AMD patients. Genomic DNA was extracted from the buffy coat of peripheral blood. Allele specific PCR was used to assay SNPs in complement factor H (CFH), complement component 3 (C3). Comparative proteomic analysis of urine samples from early AMD, choroidal neovascular membrane (CNVM), geographic atrophy (GA), and healthy controls was performed using isobaric labelling followed by mass spectrometry. Validation was performed using enzyme-linked immunosorbent assay (ELISA)., Results: Comparative proteomic analysis of urine samples identified 751 proteins, of which 383 proteins were found to be differentially expressed in various groups of AMD patients. Gene ontology classification of differentially expressed proteins revealed the majority of them were involved in catalytic functions and binding activities. Pathway analysis showed cell adhesion molecule pathways (CAMs), Complement and coagulation cascades, to be significantly deregulated in AMD. Upon validation by ELISA, SERPINA-1 (Alpha1 antitrypsin), TIMP-1 (Tissue inhibitor of matrix metaloprotease-1), APOA-1 (Apolipoprotein A-1) were significantly over-expressed in AMD (n = 61) patients compared to controls (n = 23). A logistic model of APOA-1 in combination with CFH and C3 polymorphisms predicted the risk of developing AMD with 82% accuracy., Conclusion: This study gives us a preliminary data on non-invasive predictive biomarkers for AMD, which can be further validated in a large cohort and translated for diagnostic use., Competing Interests: None

An unprecedented Ru(ii)-catalyzed Csp2-H bond activation and annulation reaction of phenylindazolones with diaryl-substituted alkynes and dialkyl-substituted alkynes provided efficient routes for the construction of all-carbon quaternary-centered indolo[1,2-a]quinazolinones and quaternary carbon-centered indazolo[1,2-a]indazolones, respectively. The indolo[1,2-a]quinazolinones were fomed via Csp2-H activation, alkyne insertion and a 1,2-phenyl shift. Indazolo[1,2-a]indazolones were formed through a cascade reaction via the formation of exocyclic double bonds containing indolo[1,2-a]quinazolinones.

Asiatic lions (Panthera leo persica) are an icon of conservation success, yet their status is inferred from total counts that cannot account for detection bias and double counts. With an effort of 4,797 km in 725 km2 of western Gir Protected Area, India, we used polygon search based spatially explicit capture recapture framework to estimate lion density. Using vibrissae patterns and permanent body marks we identified 67 lions from 368 lion sightings. We conducted distance sampling on 35 transects with an effort of 101.5 km to estimate spatial prey density using generalized additive modeling (GAM). Subsequently, we modeled lion spatial density with prey, habitat characteristics, anthropogenic factors and distance to baiting sites. Lion density (>1-year-old lions) was estimated at 8.53 (SE 1.05) /100 km2 with lionesses having smaller movement parameter (σ = 2.55 km; SE 0.12) compared to males (σ = 5.32 km; SE 0.33). Detection corrected sex ratio (female:male lions) was 1.14 (SE 0.02). Chital (Axis axis) was the most abundant ungulate with a density of 63.29 (SE 10.14) as determined by conventional distance sampling (CDS) and 58.17 (SE 22.17)/km2 with density surface modeling (DSM), followed by sambar (Rusa unicolor) at 3.84 (SE 1.07) and 4.73 (SE 1.48)/km2 estimated by CDS and DSM respectively. Spatial lion density was best explained by proximity to baiting sites and flat valley habitat but not as much by prey density. We demonstrate a scientifically robust approach to estimate lion abundance, that due to its spatial context, can be useful for management of habitat and human-lion interface. We recommend this method for lion population assessment across their range. High lion densities in western Gir were correlated with baiting. The management practice of attracting lions for tourism can perturb natural lion densities, disrupt behavior, lion social dynamics and have detrimental effects on local prey densities., Competing Interests: The authors have declared that no competing interests exist.

Artemisinin-based combination therapy (ACT) has been used to treat uncomplicated Plasmodium falciparum infections in India since 2004. Since 2008, a decrease in artemisinin effectiveness has been seen throughout the Greater Mekong Subregion. The geographic proximity and ecological similarities of northeastern India to Southeast Asia may differentially affect the long-term management and sustainability of ACT in India. In order to collect baseline data on variations in ACT sensitivity in Indian parasites, 12 P. falciparum isolates from northeast India and 10 isolates from southwest India were studied in vitro Ring-stage survival assay (RSA) showed reduced sensitivity to dihydroartemisinin in 50% of the samples collected in northeast India in 2014 and 2015. Two of the 10 assayed samples from the southwest region of India from as far back as 2012 also showed decreased sensitivity to artemisinin. In both these regions, kelch gene sequences were not predictive of reduced artemisinin sensitivity, as measured by RSA. The present data justify future investments in integrated approaches involving clinical follow-up studies, in vitro survival assays, and molecular markers for tracking potential changes in the effectiveness of artemisinin against P. falciparum throughout India., (Copyright © 2019 Chakrabarti et al.)

Herein, we report the construction of intracellular logic operations using luminescent histidine stabilized gold nanoclusters (His Au NCs). The luminescence intensity of His Au NCs was found to be significantly enhanced following interaction with zinc ions, owing to "Crystallization induced emission enhancement". Further, the luminescence intensity of His Au NCs was found to be effectively quenched in presence of sulphide ions, owing to transformation of emissive His Au NCs to non-emissive gold nanoparticles. Thus, the collective and individual effects of zinc ions and sulphide ions causing significant variation in the luminescence intensity of His Au NCs, were used as input parameters for construction of intracellular logic operations such as Tri state buffer, "on-off" switch and INHIBIT gate within mammalian cells., (© 2019 Wiley-VCH Verlag GmbH & Co. This study aims to examine the hypothesis that circulatory heavy metals may be associated with lung function decline and lower plasma GST activity and GSH level in COPD patients via activating monocytes mediated by impairing the NOX4/Nrf2/GCLC/GST signaling pathway. Results showed that the blood levels of heavy metals (cadmium, lead, mercury, and chromium) were significantly higher in COPD patients of coal mine site compared to the healthy controls. The levels of heavy metals in COPD patients were significantly and negatively correlated with lung function, GST activity, and GSH level. Using flowcytometry, fluorescence spectroscopy, and immunoblotting studies we have further demonstrated that treatment with individual heavy metals dose-dependently increased the NOX4 protein expression, intracellular ROS production, and decreased the Nrf2, GCLC, and GST protein expression, GST activity, and GSH level in THP-1 monocytes. None of the treatment caused any change in cell viability compared to control. In conclusion, this study suggests that circulatory heavy metals in COPD patients of coal mine site weakened the lung function, decreased the plasma GST activity and GSH level via impairing the NOX4/Nrf2/GCLC/GST signaling pathway in monocytes, which may cause monocyte activation and initiate the COPD pathophysiology., (Copyright © 2018 Elsevier Ltd. A cobalt-nitrosyl complex, [(BPI)Co(NO)(OAc)], 1 {BPI = 1,3-bis(2'-pyridylimino)isoindol} was prepared and characterized. Structural characterization revealed that the cobalt center has a distorted square pyramidal geometry with the NO group coordinated from the apical position in a bent fashion. The addition of dioxygen (O 2 ) to the dichloromethane solution of complex 1 resulted in the formation of nitro complex, [(BPI)Co(NO 2 )(OAc)], 2. It was characterized structurally. Kinetic studies suggested the involvement of an associative mechanism. FT-IR spectroscopic studies suggested the formation of the intermediate 1a [(BPI)Co III (NO)(O 2 - )(OAc)] in the reaction. The intermediate 1a decomposed to complex 2 via a presumed peroxynitrite intermediate which was implicated by its characteristic phenol ring nitration reaction.

A Co(II) complex, [Co(L) 2 ]Cl 2 , 1 of the ligand L (L = bis(2-ethyl-4-methylimidazol-5-yl)methane) upon reaction with H 2 O 2 in methanol solution at -40 °C resulted in the formation of the corresponding Co(III)-peroxo complex [Co(L) 2 (O 2 )] + (2). The addition of NO gas to the freshly generated solution of the complex 2 led to the formation of the Co(II)-nitrato complex 3 through the putative formation of a Co(II)-peroxynitrite intermediate, 2a. The intermediate 2a was found to mediate the nitration of the externally added phenol resembling the nitration of tyrosine in biological systems.

The cobalt porphyrin complex [(Cl 4 TPP)Co], 1, {Cl 4 TPP = 5,10,15,20-tetrakis(4'-chlorophenyl)porphyrinate dianion} in dichloromethane solution was subjected to react with nitric oxide (NO) gas and resulted in the formation of the corresponding nitrosyl complex [(Cl 4 TPP)Co(NO)], 2, having {CoNO} 8 description. It was characterized by spectroscopic studies and single-crystal X-ray structure determination. It did not react with dioxygen. However, in CH 2 Cl 2 /CH 3 CN solution, it reacted with H 2 O 2 to result in the Co-nitrito complex [(Cl 4 TPP)Co(NO 2 )], 3, with the simultaneous release of O 2 . It induced ring nitration to the added phenol in an appreciable yield. The reaction presumably proceeds through the formation of corresponding Co-peroxynitrite intermediate.

Dura-based intracranial neoplasms include a wide range of primary and metastatic tumors, varying in their clinical, radiologic, morphologic, and immunophenotypic characteristics. At this anatomic location, sarcomas are rare, however, they exhibit close morphologic resemblances to meningioma. Herein we describe the third case of primary synovial sarcoma of the parafalcine region in a50-years-old female, who presented with left-sided hemiplegia. The radiologic survey revealed a 5.5cm×5.8cm contrast enhancing dura-based mass at the right parafalcine region with meningeal enhancement and edema in the surrounding areas. Morphologic evaluation exhibited a high-grade spindle cell neoplasm, with focal hemangiopericytomatous pattern. The tumor cells were diffusely immunoreactive for CD99, Bcl2, TLE-1, and vimentin. The Ki-67 proliferation index was 40%. Pancytokeratin was focally positive. Epithelial membrane antigen, progesterone receptor, CD34, S-100, and glial fibrillary acidic protein were negative. Fluorescence in situ hybridization confirmed tumor specific translocation t(X;18)(p11.2;q11.2). Hence, final diagnosis of synovial sarcoma was rendered. Primary meningeal synovial sarcoma should be considered in the differential of aggressive and high-grade dura-based tumors in view of their relative chemosensitivity and future prospect of a molecular target-based therapy. The index case highlights the importance of an extensive pathologic analysis of high-grade mesenchymal lesions of the meninges to arrive at a definitive diagnosis and differentiate such tumors from other usual dura-based tumors, which has important therapeutic and prognostic implications., (Copyright © 2017 Elsevier GmbH. In the present study, we are reporting antimalarial potential of silver (AgNPs) and gold (AuNPs) nanoparticles synthesized by leaf and bark extract of Syzygium jambos (L.) Alston (Myrtaceae). AuNPs and AgNPs obtained by both the extracts were characterized using UV-vis spectroscopy, zeta potential, transmission electron microscopy (TEM), X-ray diffraction (XRD), and Fourier Transform Infrared spectroscopy (FTIR). NMR and FTIR spectra indicate that the saccharides and phenolics present in the S. jambos extracts were the major contributors responsible for the synthesis and stabilization of NPs. NPs were also synthesized by chemical methods and were compared for their antiplasmodial potential against chloroquine sensitive (3D7) and resistant (Dd2) strain of Plasmodium falciparum by using 24h schizont maturation assay. AgNPs synthesized by both the extracts showed higher antiplasmodial activity than the rest. Further, NPs synthesized by S. jambos extracts have shown insignificant cytotoxicity against human cervical cancer cell line (HeLa) and rat skeletal muscle cell line (L6), which proved their biocompatibility., (Copyright © 2017 Elsevier Masson SAS. To reduce the dependency on fresh AB + serum in continuous culture of Plasmodium falciparum , a comparative study was undertaken to assess the in vitro adaptability of P. falciparum to media supplemented with fresh AB + serum from whole blood, AB + plasma, serum derived from AB + plasma, AB + human serum from Sigma, Albumax II, fetal bovine serum and new born calf serum, independently and in different combinations. Combinations were used to analyze whether two different substitutes demonstrate any synergistic effect on the growth of the parasites. Our findings exhibited that the combination of fresh human serum and Albumax II showed good growth pattern in comparison to that of fresh serum and can thereby be instrumental in reducing the role of fresh human serum in continuous parasite maintenance. Culture maintained with Albumax II with or without hypoxanthine showed average growth.

A binuclear Cu(II) complex, 1, [Cu 2 (L - ) 2 (OAc)](OAc) of imidazole-based ligand LH {LH = 2-(bis(2-ethyl-5-methyl-1H-imidazol-4-yl)methyl)phenol} was synthesized and characterized spectroscopically and structurally. Addition of an equivalent amount of nitric oxide (NO) by a gastight syringe to the acetonitrile:methanol (5:1, v/v) solution of complex 1 at room temperature resulted in the reduction of Cu(II) center to Cu(I) with concomitant C-nitrosation of the ligand. Spectroscopic characterization of the resulting Cu(I) complex (1a) of the C-nitrosylated ligand, L' {L' = 2-(bis(2-ethyl-5-methyl-1H-imidazol-4-yl)methyl)-4-nitroso-phenol} has been done. The Cu(I) complex, 1a, further reacted with NO to result in the corresponding N-nitrosohydroxylaminato complex, 2, [Cu 2 (L-ONNO) 2 ](OAc) 2 through the formation of a Cu(I)-nitrosyl intermediate. A small fraction of the nitrosyl intermediate decomposed to the corresponding Cu(II) complex 3, [Cu(L') 2 ], and N 2 O in a parallel reaction.

The polyphenolic compound curcumin has been reported for its antimalarial properties in various scientific studies. Plasmodium falciparum ATP6, the parasite orthologue of mammalian sarcoplasmic Ca 2+ ATPase (SERCA) has been identified as a key molecular target of both artemisinin and curcumin. The work was thereby undertaken to study the anti-malarial properties of two different series of curcumin analogues based on their docking interactions with PfATP6 and correlating the results with their anti-malarial activity. The compounds were designed retaining similar functional groups as that of the parent curcumin nucleus while incorporating changes in the carbon chain length, unsaturated groups and the number of ketone groups. The compounds (1E, 4E)-1,5-bis(4-methylphenyl)penta-1,4-dien-3-one (CD-9), (1E, 4E)-1,5-bis(4-methoxyphenyl)penta-1,4-dien-3-one (CD-8) and (E)-1,3-bis(4-hydroxylphenyl)prop-2-en-1-one (CD-1) showed IC 50 values of 1.642 μM, 1.764 μM and 2.59 μM in 3D7 strain and 3.039 μM, 7.40 μM and 11.3 μM in RKL-2 strain respectively. Detailed structure-activity relationship studies of the compounds showed that CD-9 and CD-8 had a common hydrophobic interaction with the residue Leu268 of the PfATP6 protein and has been postulated through our study to be the reason for their antimalarial activity as seen after corroborating the results with the in vitro study. Bioactive macromolecular peptides and oligonucleotides have significant therapeutic potential. However, due to their size, they have no ability to enter the cytoplasm of cells. Peptide/Protein transduction domains (PTDs), also called cell-penetrating peptides (CPPs), can promote uptake of macromolecules via endocytosis. However, overcoming the rate-limiting step of endosomal escape into the cytoplasm remains a major challenge. Hydrophobic amino acid R groups are known to play a vital role in viral escape from endosomes. Here we utilize a real-time, quantitative live cell split-GFP fluorescence complementation phenotypic assay to systematically analyze and optimize a series of synthetic endosomal escape domains (EEDs). By conjugating EEDs to a TAT-PTD/CPP spilt-GFP peptide complementation assay, we were able to quantitatively measure endosomal escape into the cytoplasm of live cells via restoration of GFP fluorescence by intracellular molecular complementation. We found that EEDs containing two aromatic indole rings or one indole ring and two aromatic phenyl groups at a fixed distance of six polyethylene glycol (PEG) units from the TAT-PTD-cargo significantly enhanced cytoplasmic delivery in the absence of cytotoxicity. EEDs address the critical rate-limiting step of endosomal escape in delivery of macromolecular biologic peptide, protein and siRNA therapeutics into cells.

The activation of nitric oxide (NO) by transition metal complexes has attracted a wide range of research activity. To study the role of ligand denticity on the NO reactivity of Co(ii) complexes, three complexes (, and ) were prepared with ligands , and [ = N(1),N(2)-bis(2,4,6-trimethylbenzyl)ethane-1,2-diamine; = N(1)-(2,4,6-trimethylbenzyl)-N(2)-(2-((2,4,6-trimethylbenzyl)amino)ethyl)ethane-1,2-diamine] and = N(1)-(2,4,6-trimethylbenzyl)-N(2),N(2)-bis(2-((2,4,6-trimethylbenzyl)amino)ethyl)ethane-1,2-diamine], respectively. The complexes differ from each other in terms of denticity and flexibility of the ligand frameworks. In degassed methanol solution, they were exposed to NO gas and their reactivity was studied using various spectroscopic techniques. In the case of complex with a bidentate ligand, reductive nitrosylation of the metal ion with concomitant dinitrosation of the ligand framework was observed. Complex with a tridentate ligand did not undergo reductive nitrosylation; rather, the formation of [Co(III)(NO(-))] was observed. The nitrosyl complexes were isolated and structurally characterized. On the other hand, complex with a tetradentate tripodal ligand did not react with NO. This can be attributed to the geometry of the complex as well as due to the accessibility of the corresponding redox potential.

Ni(ii) complex of ligand ( = bis(2-ethyl-4-methylimidazol-5-yl)methane) in methanol solution reacts with an equivalent amount of NO resulting in a corresponding Ni(i) complex. Adding further NO equivalent affords a Ni(i)-nitrosyl intermediate with the {NiNO}(10) configuration. This nitrosyl intermediate upon subsequent reaction with additional NO results in the release of N2O and formation of a Ni(ii)-nitrito complex. Crystallographic characterization of the nitrito complex revealed a symmetric η(2)-O,O-nitrito bonding to the metal ion. This study demonstrates the reductive nitrosylation of a Ni(ii) center followed by N2O release in the presence of excess NO.

A series of novel hybrid 4-aminoquinoline 1,3,5-triazine derivatives was synthesized in a five-steps reaction and evaluated for their in vitro antimalarial activity against chloroquine-sensitive (3D7) and chloroquine-resistant (RKL-2) strains of Plasmodium falciparum. Entire synthetic derivatives showed higher antimalarial activity on the sensitive strain while two compounds, viz., 9a and 9c displayed good activity against both the strains of P. falciparum. The observed activity was further substantiated by docking study on both wild and qradruple mutant type P. falciparum dihydrofolate reductase-thymidylate synthase (pf-DHFR-TS)., (Copyright © 2015 Elsevier Inc. A new series of hybrid 4-aminoquinoline-1,3,5-triazine derivatives was synthesized by a four-step reaction. Target compounds were screened for in vitro antimalarial activity against chloroquine-sensitive (3D-7) and chloroquine-resistant (RKL-2) strains of Plasmodium falciparum. Compounds exhibited, by and large, good antimalarial activity against the resistant strain, while two of them, that is 8g and 8a, displayed higher activity against both the strains of P. falciparum. Additionally, docking study was performed on both wild (1J3I.pdb) and quadruple mutant (N51I, C59R, S108 N, I164L, 3QG2.pdb) type pf-DHFR-TS to highlight the structural features of hybrid molecules., (© 2014 John Wiley & Sons A/S.)
