Axenfeld-Rieger syndrome (ARS) is a disorder affecting the anterior segment of the eye and causing systemic malformations, and follows an autosomal-dominant inheritance pattern. The aim of the present study was to identify the underlying cause of ARS in a Chinese family. Genomic DNA was extracted from the peripheral blood of the subjects from a family with ARS. The pathogenic variant was identified by targeted next-generation sequencing and confirmed by Sanger sequencing. A novel heterozygous mutation of the forkhead box (FOX)C1 gene (c.1494delG, p.G499Afs*20) was detected in all affected members of the family, while no mutation was identified in the unaffected members or in the 150 normal controls. The affected members exhibited typical ocular and craniofacial anomalies. The results of the present study demonstrated that a novel deletion in exon 1 of the FOXC1 gene caused ARS in this Chinese family.

Understanding the origins of lipid membrane bilayer rearrangement in response to external stimuli is an essential component of cell biology and the bottom-up design of liposomes for biomedical applications. The enzymes phospholipase C and D (PLC and PLD) both cleave the phosphorus–oxygen bonds of phosphate esters in phosphatidylcholine (PC) lipids. The atomic position of this hydrolysis reaction has huge implications for the stability of PC-containing self-assembled structures, such as the cell wall and lipid-based vesicle drug delivery vectors. While PLC converts PC to diacylglycerol (DAG), the interaction of PC with PLD produces phosphatidic acid (PA). Here we present a combination of small-angle scattering data and all-atom molecular dynamics simulations, providing insights into the effects of atomic-scale reorganization on the supramolecular assembly of PC membrane bilayers upon enzyme-mediated incorporation of DAG or PA. We observed that PC liposomes completely disintegrate in the presence of PLC, as conversion of PC to DAG progresses. At lower concentrations, DAG molecules within fluid PC bilayers form hydrogen bonds with backbone carbonyl oxygens in neighboring PC molecules and burrow into the hydrophobic region. This leads initially to membrane thinning followed by a swelling of the lamellar phase with increased DAG. At higher DAG concentrations, localized membrane tension causes a change in lipid phase from lamellar to the hexagonal and micellar cubic phases. Molecular dynamics simulations show that this destabilization is also caused in part by the decreased ability of DAG-containing PC membranes to coordinate sodium ions. Conversely, PLD-treated PC liposomes remain stable up to extremely high conversions to PA. Here, the negatively charged PA headgroup attracts significant amounts of sodium ions from the bulk solution to the membrane surface, leading to a swelling of the coordinated water layer. These findings are a vital step toward a fundamental understanding of the degradation behavior of PC lipid membranes in the presence of these clinically relevant enzymes, and toward the rational design of diagnostic and drug delivery technologies for phospholipase-dysregulation-based diseases., Phospholipase C activity destabilizes PC liposomes, ultimately leading to nonlamellar phases. Phospholipase D activity attracts cations to PC liposome membranes but does not affect their morphology.

DOI: 10.1021/acsnano.5b07966 As more biological activities of ribonucleic acids continue to emerge, the development of efficient analytical tools for RNA identification and characterization is necessary to acquire an in-depth understanding of their functions and chemical properties. Herein, we demonstrate the capacity of label-free direct surface-enhanced Raman scattering (SERS) analysis to access highly specific structural information on RNAs at the ultrasensitive level. This includes the recognition of distinctive vibrational features of RNAs organized into a variety of conformations (micro-, fully complementary duplex-, small interfering- A nd short hairpin-RNAs) or characterized by subtle chemical differences (single-base variances, nucleobase modifications and backbone composition). This method represents a key advance in the ribonucleic acid analysis and will have a direct impact in a wide range of different fields, including medical diagnosis, drug design, and biotechnology, by enabling the rapid, high-throughput, simple, and low-cost identification and classification of structurally similar RNAs.

Recognition of chemical modifications in canonical nucleobases of nucleic acids is of key importance since such modified variants act as different genetic encoders, introducing variability in the biological information contained in DNA. Herein, we demonstrate the feasibility of direct SERS in combination with chemometrics and microfluidics for the identification and relative quantification of 4 different cytosine modifications in both single- and double-stranded DNA. The minute amount of DNA required per measurement, in the sub-nanogram regime, removes the necessity of pre-amplification or enrichment steps (which are also potential sources of artificial DNA damages). These findings show great potentials for the development of fast, low-cost and high-throughput screening analytical devices capable of detecting known and unknown modifications in nucleic acids (DNA and RNA) opening new windows of activity in several fields such as biology, medicine and forensic sciences.

Biomineralizing organisms can grow nanomaterials with unexpected morphologies in an organic matrix where temporal and vectorial gradients of crystal growth precursors are established. Here, concentration gradients for the crystallization of gold nanoparticles are generated and applied on silicon substrates. Gradients of crystal growth precursors are generated by enzymes patterned as lines that are separated by distances ranging from the micro- to the nanoscale. The concentration of crystallization precursors around the lines separated by nanometric distances is not only determined by mass transport and enzyme activity but also by the nanoscale organization of biocatalysts. This nanoscale organization favors non-classical crystal growth conditions that lead to the formation of nanoparticle clusters containing nanocrystals that are highly crystallographically aligned. The combination of bottom-up crystal growth with top-down electron beam lithography enables the fabrication of micrometric patterns containing gold nanoparticles of different size, shape, and surface density. These are all critical parameters that determine the physical properties of these nanomaterials.

Recently, plasmonic-based biosensing has experienced an unprecedented level of attention, with a particular focus on the nucleic acid detection, offering efficient solutions to engineer simple, fast, highly sensitive sensing platforms while overcoming important limitations of PCR and microarray techniques. In the broad field of plasmonics, surface-enhanced Raman scattering (SERS) spectroscopy has arisen as a powerful analytical tool for detection and structural characterization of biomolecules. Today applications of SERS to nucleic acid analysis largely rely on indirect strategies, which have been demonstrated very effective for pure sensing purposes but completely dismiss the exquisite structural information provided by the direct acquisition of the biomolecular vibrational fingerprint. Contrarily, direct label-free SERS of nucleic acid shows an outstanding potential in terms of chemical-specific information which, however, remained largely unexpressed mainly because of the inherent poor spectral reproducibility and/or limited sensitivity. To address these limitations, we developed a fast and affordable high-throughput screening direct SERS method for gaining detailed genomic information on nucleic acids (DNA and RNA) and for the characterization and quantitative recognition of DNA interactions with exogenous agents. The simple strategy relies on the electrostatic adhesion of DNA/RNA onto positively-charged silver colloids that promotes the nanoparticle aggregation into stable clusters yielding intense and reproducible SERS spectra at picogram level (i.e. the analysis can be performed without the necessity of amplification steps thus providing realistic direct information of the nucleic acid in its native state). We anticipate this method to gain a vast impact and set of applications in different fields, including medical diagnostics, genomic screening, drug discovery, forensic science and even molecular electronics.

Hollow gold nanospheres (HGNs) provide a tunable surface plasmon resonance from 550 to 820 nm by controlling their inner diameter and wall thickness. Although they have been used for biological imaging based on their optical properties in the near-infrared region, their surface-enhanced Raman scattering (SERS) performance has not been thoroughly studied. Herein, HGNs with different surface plasmon resonances were synthesized and functionalized with different Raman reporters. HGNs coupled with Raman reporters, on and off resonant with the laser excitation wavelength, were systematically interrogated in isolated and partially aggregated situations. HGNs with thicker shells generated higher SERS responses than thinner shells no matter whether they were isolated, or partially aggregated, or whether their surface plasmon resonances were resonant with the excitation wavelength or not. This study gives insight into the basis of the SERS properties for these kinds of materials.

Surface enhanced Raman scattering (SERS) "hot spots" are the regions where the electromagnetic field is significantly enhanced, resulting in much greater SERS activity than other areas. Therefore, the engineering and characterization of "hot spots" have attracted much attention. Herein, we have synthesized nanoworms formed by connecting hollow gold nanospheres and investigated their composition as well as their SERS properties, including their potential for simultaneous analysis of multiple species. The three-dimensional nanostructures that led to different SERS activities were investigated using correlated optical/SERS and SEM imaging. Gold nanoworms with hollow nanosphere segments and partial tube formation were confirmed. All highly SERS-active structures were found to be three-dimensional arrays consisting of inter-/intrananoworm interactions; no individual nanospheres were found to give significant SERS signal under the experimental conditions. The major SERS contribution was found to be the electromagnetic coupling effect within the three-dimensional arrays and the localized surface plasmon resonance of the nanostructure was found to have a minimal effect. This study gives insight into the basis of enhancement for these nanoworm structures and adds to the growing body of evidence in relation to nanostructures and surface enhancement of Raman scattering. © 2011 American Chemical Society.

A new facile fabrication of Pt, Pd and their alloy films on Si, intended mainly for in situ ATR surface infrared spectroscopy and electrocatalysis study, has been achieved through an alternate electroless deposition approach from very simple acidic baths with hydrazine dihydrochloride as the reducing agent. Compositional analyses reveal that Pt and Pd elemental ratios in the alloy films are close to those in the plating baths, and higher in skin layers compared to those of bulk films. Electrochemical and IR spectroscopic characterizations suggest the Pt-Pd alloy films may exhibit synergetic effects in surface electrochemistry toward oxidation of CO adlayer and formic acid. Of particular interest is the reproducible control of internal reflection absorption responses through adjusting Pt and Pd compositions in the films. More enhanced surface IR absorption with normal band shape and direction was observed for surface species at Pt and Pd electrodes. For the as-deposited Pt-Pd alloys, in addition to less enhanced IR absorption intensity, bipolar and totally inverted bands may appear for different alloy films.

In situ ATR–FTIR spectroscopy has been extended to the study of CO and pyridine adsorptions at crystalline and amorphous Ni–P alloy film electrodes, with an emphasis on the alloying effect of P on their adsorption configurations on Ni sites. The Ni–P films were prepared with initial seeding of a catalytic Pd layer on Si, followed by chemical deposition. Transition from bridge-bonded CO (COB) dominant to linearly bonded CO (COL) dominant adsorption was found with increasing P content, together with a blue-shift in the COL vibrational frequency and an attenuation of the Stark tuning rate of COL. As for Py on Ni–P electrodes, essentially the N-end-on adsorption was revealed, in contrast to the edge-tilted adsorption on Ni electrode. Modification in the adsorption configurations as compared to that on Ni electrode is ascribed mainly to the site-blocking effect with alloying P, rather than to partial electron transfer between Ni and P.

Exceptionally preserved organic remains are known throughout the vertebrate fossil record, and recently, evidence has emerged that such soft tissue might contain original components. We examined samples from eight Cretaceous dinosaur bones using nano-analytical techniques; the bones are not exceptionally preserved and show no external indication of soft tissue. In one sample, we observe structures consistent with endogenous collagen fibre remains displaying ∼67 nm banding, indicating the possible preservation of the original quaternary structure. Using ToF-SIMS, we identify amino-acid fragments typical of collagen fibrils. Furthermore, we observe structures consistent with putative erythrocyte remains that exhibit mass spectra similar to emu whole blood. Using advanced material characterization approaches, we find that these putative biological structures can be well preserved over geological timescales, and their preservation is more common than previously thought. The preservation of protein over geological timescales offers the opportunity to investigate relationships, physiology and behaviour of long extinct animals., Soft tissue from vertebrate fossils has previously been documented, but only in exceptionally preserved specimens. Here, Bertazzo et al. describe structures consistent with collagen fibres and red blood cells from eight Cretaceous dinosaur bones, none of which are exceptionally preserved.

Track it down: A recognized surface-enhanced Raman scattering (SERS) nanotag signal was monitored from a thin, dispersed layer of bisphosphonate-functionalized nanotags on a bone sample, through a 20 mm thick specimen of porcine muscle tissue by surface-enhanced spatial offset Raman spectroscopy (SESORS; see picture). The result demonstrates the great potential for non-invasive in vivo bisphosphonate drug tracking.

The standard protocols for DNA analysis largely involve polymerase chain reaction (PCR). However, DNA structures bound to chemical agents cannot be PCR-amplified, and therefore any sequence changes induced by external agents may be neglected. Thus, the development of analytical tools capable of characterizing the biochemical mechanisms associated with chemically induced DNA damage is demanded for the rational design of more effective chemotherapy drugs, understanding the mode of actions of carcinogenic chemicals, and monitoring the genotypic toxicology of environments. Here we report a fast, high-throughput, low-cost method for the characterization and quantitative recognition of DNA interactions with exogenous agents based on surface-enhanced Raman scattering spectroscopy. As representative chemical agents, we selected a chemotherapeutic drug (cisplatin) which forms covalent adducts with DNA, a duplex intercalating agent (methylene blue), and a cytotoxic metal ion (Hg(II)) which inserts into T:T mismatches. Rich structural information on the DNA complex architecture and properties is provided by the unique changes of their SERS spectra, which also offer an efficient analytical tool to quantify the extent of such binding.

The identification of intracellular distributions of noble metal nanoparticles is of great utility for many biomedical applications. We present an effective method to distinguish intracellular from extracellular nanoparticles by selectively quenching the SERS signals from dye molecules adsorbed onto star-shaped gold nanoparticles that have not been internalized by cells.

The Nrf2-ARE pathway plays a cytoprotective role in many tissues,but its protective function in the optic nerve is unclear. The purpose of the study is to investigate the changes in activation of the Nrf2-ARE pathway following optic nerve injury (ONI) in mice.Using ONI mice models, the expression levels of Nrf2 in optic nerves were determined by real-time PCR at various time points.The expression of Nrf2mRNA was significantly upregulated at 1 d after ONI, peaking at 30 min after ONI.The Nrf2-ARE pathway was activated after ONI, providing evidence for the study of the protection and underlying mechanism of Nrf2-ARE pathway on optic nerves.

A highly SERS-active substrate was fabricated by trapping gold "nanoworms" on commercially available filter membranes providing significant enhancement of the Raman signal as a result of the remarkable electromagnetic couplings induced by the dense packing. The resultant substrate provides a simple and cost-effective porous SERS surface for use and quantitative analytical procedures.

Rapid advances in nanofabrication techniques of reproducibly manufacturing plasmonic substrates with well-defined nanometric scale features and very large electromagnetic enhancements paved the way for the final translation of the analytical potential of surface-enhanced Raman scattering (SERS) to real applications. A vast number of different SERS substrates have been reported in the literature. Among others, discrete particles consisting of an inorganic micrometric or sub-micrometric core homogeneously coated with plasmonic nanoparticles stand out for their ease of fabrication, excellent SERS enhancing properties, long-term optical stability and remarkable experimental flexibility (manipulation, storage etc). In this article, we performed a systematic experimental study of the correlation between the size of quasi-spherical gold and silver nanoparticle and the final optical property of their corresponding assembles onto micrometric polystyrene (PS) beads. The size and composition of nanoparticles play a key role in tuning the SERS efficiency of the hybrid material at a given excitation wavelength. This study provides valuable information for the selection and optimization of the appropriate PS@NPs substrates for the desired applications.

Optical analysis in the near infrared region is of significant biological importance due to better tissue penetration and reduced autofluorescence. In this work, an improved synthesis of hollow gold nanospheres (HGNs), which provides a tunable localized surface plasmon resonance (LSPR) from 610 nm up to 1320 nm, is demonstrated. The scattering properties of these nanoparticles are shown using surface enhanced Raman scattering (SERS) at 1064 nm excitation wavelength and are compared to citrate reduced gold and silver nanoparticles of similar physical sizes and surface properties. After the addition of salts, a strong signal was observed from hollow gold with a LSPR of 650 nm and a weaker, yet observable, signal from HGNs with a LSPR of 775 nm. However, no obvious signals were observed in the case of standard citrate reduced gold, silver or HGNs with a LSPR of 1080 nm. The absorption properties of HGNs were investigated by monitoring their photothermal activity. In this case, different nanoparticle suspensions including citrate reduced gold, silver, and HGNs were illuminated by a continuous laser at 785 nm excitation wavelength and the absorption efficiency of HGNs with a LSPR of 775 nm was calculated to be 0.81% which is more than 5 times higher than the absorption efficiency of citrate reduced gold nanoparticles under similar conditions.
