To extend the preservation of donor hearts beyond the current 4-6 h, this paper explores heart cryopreservation by vitrification-cryogenic storage in a glass-like state. While organ vitrification is made possible by using cryoprotective agents (CPA) that inhibit ice during cooling, failure occurs during convective rewarming due to slow and non-uniform rewarming which causes ice crystallization and/or cracking. Here an alternative, "nanowarming", which uses silica-coated iron oxide nanoparticles (sIONPs) perfusion loaded through the vasculature is explored, that allows a radiofrequency coil to rewarm the organ quickly and uniformly to avoid convective failures. Nanowarming has been applied to cells and tissues, and a proof of principle study suggests it is possible in the heart, but proper physical and biological characterization especially in organs is still lacking. Here, using a rat heart model, controlled machine perfusion loading and unloading of CPA and sIONPs, cooling to a vitrified state, and fast and uniform nanowarming without crystallization or cracking is demonstrated. Further, nanowarmed hearts maintain histologic appearance and endothelial integrity superior to convective rewarming and indistinguishable from CPA load/unload control hearts while showing some promising organ-level (electrical) functional activity. This work demonstrates physically successful heart vitrification and nanowarming and that biological outcomes can be expected to improve by reducing or eliminating CPA toxicity during loading and unloading.

Vitrification can dramatically increase the storage of viable biomaterials in the cryogenic state for years. Unfortunately, vitrified systems ≥3 mL like large tissues and organs, cannot currently be rewarmed sufficiently rapidly or uniformly by convective approaches to avoid ice crystallization or cracking failures. A new volumetric rewarming technology entitled “nanowarming” addresses this problem by using radiofrequency excited iron oxide nanoparticles to rewarm vitrified systems rapidly and uniformly. Here, for the first time, successful recovery of a rat kidney from the vitrified state using nanowarming, is shown. First, kidneys are perfused via the renal artery with a cryoprotective cocktail (CPA) and silica‐coated iron oxide nanoparticles (sIONPs). After cooling at −40 °C min−1 in a controlled rate freezer, microcomputed tomography (µCT) imaging is used to verify the distribution of the sIONPs and the vitrified state of the kidneys. By applying a radiofrequency field to excite the distributed sIONPs, the vitrified kidneys are nanowarmed at a mean rate of 63.7 °C min−1. Experiments and modeling show the avoidance of both ice crystallization and cracking during these processes. Histology and confocal imaging show that nanowarmed kidneys are dramatically better than convective rewarming controls. This work suggests that kidney nanowarming holds tremendous promise for transplantation., Vitrification dramatically increases organ storage times in the cryogenic state beyond current storage limits. Purpose Herein, we evaluate the use of MRI as a tool for assessing iron oxide nanoparticle (IONP) distribution within IONP perfused organs and vascularized composite allografts (VCAs) (i.e., hindlimbs) prepared for cryopreservation. Methods Magnetic resonance imaging was performed on room-temperature organs and VCAs perfused with IONPs and were assessed at 9.4 T. Quantitative T1 mapping and T 2 ∗ -weighted images were acquired using sweep imaging with Fourier transformation and gradient-echo sequences, respectively. Verification of IONP localization was performed through histological assessment and microcomputer tomography. Results Quantitative imaging was achieved for organs and VCAs perfused with up to 642 mMFe (36 mgFe /mL), which is above previous demonstrations of upper limit detection in agarose (35.7mMFe [2 mgFe /mL]). The stability of IONPs in the perfusate had an effect on the quality of distribution and imaging within organs or VCA. Finally, MRI provided more accurate IONP localization than Prussian blue histological staining in this system, wherein IONPs remain primarily in the vasculature. Conclusion Using MRI, we were able to assess the distribution of IONPs throughout organs and VCAs varying in complexity. Additional studies are necessary to better understand this system and validate the calibration between T1 measurements and IONP concentration.

Background Magnetic fluid heating has great potential in the fields of thermal medicine and cryopreservation. However, variations among experimental parameters, analysis methods and experimental uncertainty make quantitative comparisons of results among laboratories difficult. Herein, we focus on the impact of calculating the specific absorption rate (SAR) using Time-Rise and Box-Lucas fitting. Time-Rise assumes adiabatic conditions, which is experimentally unachievable, but can be reasonably assumed (quasi-adiabatic) only for specific and limited evaluation times when heat loss is negligible compared to measured heating rate. Box-Lucas, on the other hand, accounts for heat losses but requires longer heating. Methods Through retrospective analysis of data obtained from two laboratories, we demonstrate measurement time is a critical parameter to consider when calculating SAR. Volumetric SAR were calculated using the two methods and compared across multiple iron-oxide nanoparticles. Results We observed the lowest volumetric SAR variation from both fitting methods between 1–10 W/mL, indicating an ideal SAR range for heating measurements. Furthermore, our analysis demonstrates that poorly chosen fitting method can generate reproducible but inaccurate SAR. Conclusion We provide recommendations to select measurement time for data analysis with either Modified Time-Rise or Box-Lucas method, and suggestions to enhance experimental precision and accuracy when conducting heating experiments.

Purpose Iron-oxide nanoparticles (IONPs) have shown tremendous utility for enhancing image contrast and delivering targeted therapies. Quantification of IONPs has been demonstrated at low concentrations with gradient echo (GRE) and spin echo (SE), and at high concentrations with echoless sequences such as swept imaging with Fourier transform (SWIFT). This work examines the overlap of IONP quantification with GRE, SE, and SWIFT. Methods The limit of quantification of GRE, SE, inversion-recovery GRE, and SWIFT sequences was assessed using IONPs at a concentration range of 0.02 to 89.29 mM suspended in 1% agarose. Empirically derived limits of quantification were compared with International Union of Pure and Applied Chemistry definitions. Both commercial and experimental IONPs were used. Results All three IONPs assessed demonstrated an overlap of concentration quantification with GRE, SE, and SWIFT sequences. The largest dynamic range observed was 0.004 to 35.7 mM with Feraheme. Conclusions The metrics established allow upper and lower quantitative limitations to be estimated given the relaxivity characteristics of the IONP and the concentration range of the material to be assessed. Oxygen homeostasis is important in the regulation of biological function. Disease progression can be monitored by measuring oxygen levels, thus producing information for the design of therapeutic treatments. Non-invasive measurements of tissue oxygenation require the development of tools with minimal adverse effects and facile detection of features of interest. Fluorine magnetic resonance imaging (19F-MRI) exploits the intrinsic properties of perfluorocarbon (PFC) liquids for anatomical imaging, cell tracking, and oxygen sensing. However, the highly hydrophobic and lipophobic properties of perfluorocarbons require the formation of emulsions for biological studies. Though, stabilizing these emulsions has been challenging. To enhance the stability and biological loading of perfluorocarbons, one option is to incorporate perfluorocarbon liquids into the internal space of biocompatible mesoporous silica nanoparticles. Here, we developed perfluorocarbon-loaded ultraporous mesostructured silica nanoparticles (PERFUMNs) as 19F-MRI detectable oxygen sensing probes. Ultraporous mesostructured nanoparticles (UMNs) have large internal cavities (average = 1.76 cm3 g−1), facilitating an average 17% loading efficiency of PFCs, meeting the threshold fluorine concentrations needed for imaging studies. Perfluoro-15-crown-5-ether PERFUMNs have the highest equivalent nuclei per PFC molecule, and a spin-lattice (T1) relaxation-based oxygen sensitivity of 0.0032 mmHg−1 s−1 at 16.4 T (657 MHz). The option of loading PFCs after synthesizing UMNs, rather than the more traditional in situ core-shell syntheses, allows for use of a broad range of PFC liquids from a single material. The biocompatible and tunable chemistry of UMNs combined with the intrinsic properties of PFCs makes PERFUMNs a MRI sensor with potential for anatomical imaging, cell tracking, and metabolic spectroscopy with improved stability.

Cryopreservation technology allows long‐term banking of biological systems. However, a major challenge to cryopreserving organs remains in the rewarming of large volumes (>3 mL), where mechanical stress and ice formation during convective warming cause severe damage. Nanowarming technology presents a promising solution to rewarm organs rapidly and uniformly via inductive heating of magnetic nanoparticles (IONPs) preloaded by perfusion into the organ vasculature. This use requires the IONPs to be produced at scale, heat quickly, be nontoxic, remain stable in cryoprotective agents (CPAs), and be washed out easily after nanowarming. Nanowarming of cells and blood vessels using a mesoporous silica‐coated iron oxide nanoparticle (msIONP) in VS55, a common CPA, has been previously demonstrated. However, production of msIONPs is a lengthy, multistep process and provides only mg Fe per batch. Here, a new microporous silica‐coated iron oxide nanoparticle (sIONP) that can be produced in as little as 1 d while scaling up to 1.4 g Fe per batch is presented. sIONP high heating, biocompatibility, and stability in VS55 is also verified, and the ability to perfusion load and washout sIONPs from a rat kidney as evidenced by advanced imaging and ICP‐OES is demonstrated., Nanowarming is a new technology that could solve the technical challenge of volumetric rewarming of cryopreserved biological samples. With the aim of scaling up nanowarming technology from cell and simple tissues to a whole organ, a scalable silica‐coated iron oxide nanoparticle is synthesized and tested for cell nanowarming and organ perfusional loading and removal.

This work evaluates MRI relaxation and the specific absorption rate properties of iron oxide nanoparticles (IONPs) as a function of diameter (6-32 nm). We conclude that the ideal IONP diameter for image guided heating applications is dependent on the magnetic field strength of the MRI for the intended application.

Purpose To use contrast based on longitudinal relaxation times (T1) or rates (R1) to quantify the biodistribution of iron oxide nanoparticles (IONPs), which are of interest for hyperthermia therapy, cell targeting, and drug delivery, within primary clearance organs. Methods Mesoporous silica-coated IONPs (msIONPs) were intravenously injected into 15 naive mice. Imaging and mapping of the longitudinal relaxation rate constant at 24 h or 1 week postinjection were performed with an echoless pulse sequence (SWIFT). Alternating magnetic field heating measurements were also performed on ex vivo tissues. Results Signal enhancement from positive T1 contrast caused by IONPs was observed and quantified in vivo in liver, spleen, and kidney at concentrations up to 3.2 mg Fe/(g tissue wt.) (61 mM Fe). In most cases, each organ had a linear correlation between the R1 and the tissue iron concentration despite variations in intra-organ distribution, degradation, and IONP surface charge. Linear correlation between R1 and volumetric SAR in hyperthermia therapy was observed. Conclusion The linear dependence between R1 and tissue iron concentration in major organs allows quantitative monitoring of IONP biodistribution in a dosage range relevant to magnetic hyperthermia applications, which falls into the concentration gap between CT and conventional MRI techniques. Iron oxide nanoparticles have great potential as diagnostic and therapeutic agents in cancer and other diseases; however, biological aggregation severely limits their function in vivo. Aggregates can cause poor biodistribution, reduced heating capability, and can confound their visualization and quantification by magnetic resonance imaging (MRI). Herein, we demonstrate that the incorporation of a functionalized mesoporous silica shell can prevent aggregation and enable the practical use of high-heating, high-contrast iron oxide nanoparticles in vitro and in vivo. Unmodified and mesoporous silica-coated iron oxide nanoparticles were characterized in biologically relevant environments including phosphate buffered saline, simulated body fluid, whole mouse blood, lymph node carcinoma of prostate (LNCaP) cells, and after direct injection into LNCaP prostate cancer tumors in nude mice. Once coated, iron oxide nanoparticles maintained colloidal stability along with high heating and relaxivity behaviors (SARFe = 204 W/g Fe at 190 kHz and 20 kA/m and r1 = 6.9 mM(-1) s(-1) at 1.4 T). Colloidal stability and minimal nonspecific cell uptake allowed for effective heating in salt and agarose suspensions and strong signal enhancement in MR imaging in vivo. These results show that (1) aggregation can lower the heating and imaging performance of magnetic nanoparticles and (2) a coating of functionalized mesoporous silica can mitigate this issue, potentially improving clinical planning and practical use.

Noble metal nanoparticles have been extensively studied to understand and apply their plasmonic responses, upon coupling with electromagnetic radiation, to research areas such as sensing, photocatalysis, electronics, and biomedicine. The plasmonic properties of metal nanoparticles can change significantly with changes in particle size, shape, composition, and arrangement. Thus, stabilization of the fabricated nanoparticles is crucial for preservation of the desired plasmonic behavior. Because plasmonic nanoparticles find application in diverse fields, a variety of different stabilization strategies have been developed. Often, stabilizers also function to enhance or improve the plasmonic properties of the nanoparticles. This review provides a representative overview of how gold and silver nanoparticles, the most frequently used materials in current plasmonic applications, are stabilized in different application platforms and how the stabilizing agents improve their plasmonic properties at the same time. Specifically, this review focuses on the roles and effects of stabilizing agents such as surfactants, silica, biomolecules, polymers, and metal shells in colloidal nanoparticle suspensions. Stability strategies for other types of plasmonic nanomaterials, lithographic plasmonic nanoparticle arrays, are discussed as well.

This Feature describes several methods for the characterization of magnetic nanoparticles in biological matrices such as cells and tissues. The Feature focuses on sample preparation and includes several case studies where multiple techniques were used in conjunction.

Low thermal-equilibrium nuclear spin polarizations and the need for sophisticated instrumentation render conventional nuclear magnetic resonance (NMR) spectroscopy and imaging (MRI) incompatible with small-scale microfluidic devices. Hyperpolarized 129Xe gas has found use in the study of many materials but has required very large and expensive instrumentation. Recently a microfabricated device with modest instrumentation demonstrated all-optical hyperpolarization and detection of 129Xe gas. This device was limited by 129Xe polarizations less than 1%, 129Xe NMR signals smaller than 20 nT, and transport of hyperpolarized 129Xe over millimeter lengths. Higher polarizations, versatile detection schemes, and flow of 129Xe over larger distances are desirable for wider applications. Here we demonstrate an ultra-sensitive microfabricated platform that achieves 129Xe polarizations reaching 7%, NMR signals exceeding 1 μT, lifetimes up to 6 s, and simultaneous two-mode detection, consisting of a high-sensitivity in situ channel with signal-to-noise of 105 and a lower-sensitivity ex situ detection channel which may be useful in a wider variety of conditions. 129Xe is hyperpolarized and detected in locations more than 1 cm apart. Our versatile device is an optimal platform for microfluidic magnetic resonance in particular, but equally attractive for wider nuclear spin applications benefitting from ultra-sensitive detection, long coherences, and simple instrumentation.

Vitrification, a kinetic process of liquid solidification into glass, poses many potential benefits for tissue cryopreservation including indefinite storage, banking, and facilitation of tissue matching for transplantation. To date, however, successful rewarming of tissues vitrified in VS55, a cryoprotectant solution, can only be achieved by convective warming of small volumes on the order of 1 ml. Successful rewarming requires both uniform and fast rates to reduce thermal mechanical stress and cracks, and to prevent rewarming phase crystallization. We present a scalable nanowarming technology for 1- to 80-ml samples using radiofrequency-excited mesoporous silica-coated iron oxide nanoparticles in VS55. Advanced imaging including sweep imaging with Fourier transform and microcomputed tomography was used to verify loading and unloading of VS55 and nanoparticles and successful vitrification of porcine arteries. Nanowarming was then used to demonstrate uniform and rapid rewarming at >130°C/min in both physical (1 to 80 ml) and biological systems including human dermal fibroblast cells, porcine arteries and porcine aortic heart valve leaflet tissues (1 to 50 ml). Nanowarming yielded viability that matched control and/or exceeded gold standard convective warming in 1- to 50-ml systems, and improved viability compared to slow-warmed (crystallized) samples. Last, biomechanical testing displayed no significant biomechanical property changes in blood vessel length or elastic modulus after nanowarming compared to untreated fresh control porcine arteries. In aggregate, these results demonstrate new physical and biological evidence that nanowarming can improve the outcome of vitrified cryogenic storage of tissues in larger sample volumes.

Aggregation is a known consequence of nanoparticle use in biology and medicine; however, nanoparticle characterization is typically performed under the pretext of well-dispersed, aqueous conditions. Here, we systematically characterize the effects of aggregation on the alternating magnetic field induced heating and magnetic resonance (MR) imaging performance of iron oxide nanoparticles (IONPs) in non-ideal biological systems. Specifically, the behavior of IONP aggregates composed of ~ 10 nm primary particles, but with aggregate hydrodynamic sizes ranging from 50 nm to 700 nm, was characterized in phosphate buffered saline and fetal bovine serum suspensions, as well as in gels and cells. We demonstrate up to a 50% reduction in heating, linked to the extent of aggregation. To quantify aggregate morphology, we used a combination of hydrodynamic radii distribution, intrinsic viscosity, and electron microscopy measurements to describe the aggregates as quasifractal entities with fractal dimensions in the 1.8–2.0 range. Importantly, we are able to correlate the observed decrease in magnetic field induced heating with a corresponding decrease in longitudinal relaxation rate (R1) in MR imaging, irrespective of the extent of aggregation. Finally, we show in vivo proof-of-principle use of this powerful new imaging method, providing a critical tool for predicting heating in clinical cancer hyperthermia.

Although preliminary clinical trials are ongoing, successful the use of iron-oxide magnetic nanoparticles (IONP) for heatbased cancer treatments will depend on advancements in: 1) nanoparticle platforms, 2) delivery of a safe and effective alternating magnetic field (AMF) to the tumor, and 3) development of non-invasive, spatially accurate IONP imaging and quantification technique. This imaging technique must be able to assess tumor and normal tissue anatomy as well as IONP levels and biodistribution. Conventional CT imaging is capable of detecting and quantifying IONPs at tissue levels above 10 mg/gram; unfortunately this level is not clinically achievable in most situations. Conventional MRI is capable of imaging IONPs at tissue levels of 0.05 mg/gm or less, however this level is considered to be below the therapeutic threshold. We present here preliminary in vivo data demonstrating the ability of a novel MRI technique, Sweep Imaging with Fourier Transformation (SWIFT), to accurately image and quantify IONPs in tumor tissue in the therapeutic concentration range (0.1-1.0 mg/gm tissue). This ultra–short, T2 MRI method provides a positive Fe contrast enhancement with a reduced signal to noise ratio. Additional IONP signal enhancement techniques such as inversion recovery spectroscopy and variable flip angle (VFA) are also being studied for potential optimization of SWIFT IONP imaging. Our study demonstrates the use of SWIFT to assess IONP levels and biodistribution, in murine flank tumors, following intra-tumoral and systemic IONP administration. ICP-MS and quantitative histological techniques are used to validate the accuracy and sensitivity of SWIFT-based IONP imaging and quantification.

Optically hyperpolarized $^{129}$Xe gas has become a powerful contrast agent in nuclear magnetic resonance (NMR) spectroscopy and imaging, with applications ranging from studies of the human lung to the targeted detection of biomolecules. Equally attractive is its potential use to enhance the sensitivity of microfluidic NMR experiments, in which small sample volumes yield poor sensitivity. Unfortunately, most $^{129}$Xe polarization systems are large and non-portable. Here we present a microfabricated chip that optically polarizes $^{129}$Xe gas. We have achieved $^{129}$Xe polarizations greater than 0.5$\%$ at flow rates of several microliters per second, compatible with typical microfluidic applications. We employ in situ optical magnetometry to sensitively detect and characterize the $^{129}$Xe polarization at magnetic fields of 1 $\mu$T. We construct the device using standard microfabrication techniques, which will facilitate its integration with existing microfluidic platforms. This device may enable the implementation of highly sensitive $^{129}$Xe NMR in compact, low-cost, portable devices.

We investigate nuclear magnetic resonance (NMR) in near-zero-field, where the Zeeman interaction can be treated as a perturbation to the electron mediated scalar interaction (J-coupling). This is in stark contrast to the high field case, where heteronuclear J-couplings are normally treated as a small perturbation. We show that the presence of very small magnetic fields results in splitting of the zero-field NMR lines, imparting considerable additional information to the pure zero-field spectra. Experimental results are in good agreement with first-order perturbation theory and with full numerical simulation when perturbation theory breaks down. We present simple rules for understanding the splitting patterns in near-zero-field NMR, which can be applied to molecules with non-trivial spectra., 5 pages, 5 figures

We present magnetic detection of a single, 2 {\mu}m diameter cobalt microparticle using an atomic magnetometer based on a microfabricated vapor cell. These results represent an improvement by a factor of 105 in terms of the detected magnetic moment over previous work using atomic magnetometers to detect magnetic microparticles.
