Oxidative stress is a leading cause of different diseases. Genistein is a valuable bioflavonoid possessing antioxidant and anti-inflammatory activity but unfortunately, it suffers from low aqueous solubility, extremely poor bioavailability and first pass effect when used in its pure state. The aim of this work was to formulate and characterize genistein-loaded highly phospholipid-containing lipid nanocarriers to improve oral bioavailability and pharmacodynamic performance. Lipid nanocarriers were prepared by the emulsification/sonication technique. The influence of phospholipid percentage (1%-10%) on physicochemical properties, drug release and stability was investigated. The particle size, zeta potential and EE% were in ranges from 211.9 ± 21.6 to 342.3 ± 7.9 nm, -11.6 ± 1.7 to -19.4 ± 3.1 mV and 78.5 ± 4.7% to 92.2 ± 1.9%, respectively. Drug release was less predominant in the case of SLN formulations when compared to corresponding NLC formulations. High phospholipid percentage produced less stable formulations in terms of particle size growth, gelation and heterogeneous particle distributions. DSC, FT-IR and XRD tools revealed that genistein has existed in an amorphous form in NLC4. The bioavailability of NLC4 was approximately 2.6-fold greater than that of conventional suspension. Additionally, lipid peroxidation in liver homogenate and histopathological alterations in liver and kidney sections were particularly improved, providing a promising strategy for oral administration of genistein.

This work aims to prepare a novel phosphate-embedded silica nanoparticles (P@SiO2) nanocomposite as an effective adsorbent through a hydrothermal route. Firstly, a mixed solution of sodium silicate and sodium phosphate was passed through a strong acidic resin to convert it into hydrogen form. After that, the resultant solution was hydrothermally treated to yield P@SiO2 nanocomposite. Using kinetic studies, methylene blue (MB) dye was selected to study the removal behavior of the P@SiO2 nanocomposite. The obtained composite was characterized using several advanced techniques. The experimental results showed rapid kinetic adsorption where the equilibrium was reached within 100 s, and the pseudo-second-order fitted well with experimental data. Moreover, according to Langmuir, one gram of P@SiO2 nanocomposite can remove 76.92 mg of the methylene blue dye. The thermodynamic studies showed that the adsorption process was spontaneous, exothermic, and ordered at the solid/solution interface. Finally, the results indicated that the presence of NaCl did not impact the adsorption behavior of MB dye. Due to the significant efficiency and promising properties of the prepared P@SiO2 nanocomposite, it could be used as an effective adsorbent material to remove various cationic forms of pollutants from aqueous solutions in future works.

Background Biosorption method for Cr(VI) removal from domestic and industrial wastewater is widely desirable technicality due to its high efficiency, natural affinity, selectivity, easily operation, low costs and eco-friendly procedures. The presented work demonstrates the sequestration of hexavalent chromium ion, Cr(VI) from contaminated water by the biosorbent Artemisia monosperma (AM) powder modified by trimethyloctadecylammonium bromide (TOAB). Methods Analytical tools of XRD, FTIR, SEM, EDS, TGA and XPS were used from one side to characterize the developed modified biosorbent TOAB@AM and from the other side to investigate the Cr(VI) adsorption. Operational parameters of pH, adsorbent dose, contact time, coexisting ions and temperature that affect adsorption process were optimized. Significant Findings Adsorption kinetics emerges the best linearity when applying the pseudo-second-order model referring to the chemisorption assumption in controlling sorption process of TOAB@AM biosorbent and Cr(VI) ions. Adsorption isothermal results are distinctly well fitted with Langmuir correlation. Furthermore, adsorption thermodynamics reveals the spontaneity and exothermic modality in adsorbing Cr(VI) onto TOAB@AM biosorbent with less randomness behavior. The optimal operational conditions for Cr (VI) removal are pH = 3; 0.1g dose of TOAB@AM; and 40 min contact time that result in adsorption capacity of Cr (VI) about 36.9 mg/g. Merits of low cost, high selectivity, and adsorptive potency of the novel TOAB@AM biosorbent towards Cr(VI) ions makes it a worthy alternative for remediating water sources from Cr(VI) contaminates within a facile and fast avenue.

In this study, the preparation of graphene oxide@chitosan (GO@CS) composite beads was investigated via continuous dropping techniques to remove methylene blue (MB)-dye from an aqueous media. The prepared beads were characterized using various techniques before and after the adsorption of MB. The experimental results showed that the adsorption processes fit the kinetic pseudo-second-order and Langmuir isotherm models. Moreover, the GO@CS beads achieve maximum adsorption capacities of 23.26 mg g−1, which was comparable with other adsorbents in the literature. An important advantage of our adsorbent is that the GO@CS can remove 82.1% of the real sample color within 135 min.

An appropriate and efficient Q-tube-assisted ammonium acetate-mediated protocol for the assembly of the hitherto unreported 5-arylazopyrazolo[3,4-b]pyridines was demonstrated. This methodology comprises the cyclocondensation reaction of 5-amino-2-phenyl-4H-pyrazol-3-one with an assortment of arylhydrazonals in an NH4OAc/AcOH buffer solution operating a Q-tube reactor. This versatile protocol exhibited several outstanding merits: easy work-up, mild conditions, scalability, broad substrate scope, safety (the Q-tube kit is simply for pressing and sealing), and a high atom economy. Consequently, performing such reactions under elevated pressures and utilizing the Q-tube reactor seemed preferable for achieving the required products in comparison to the conventional conditions. Diverse spectroscopic methods and X-ray single-crystal techniques were applied to confirm the proposed structure of the targeted compounds.

The risk of human exposure to phenylxanthene dyes is widely documented. Preliminary analysis of bottled chili sauce samples revealed the presence of two banned phenyl xanthene dyes (rhodamine B and eosin Y). Therefore, a simple, specific, and sensitive approach was implemented for the simultaneous estimation of rhodamine B (RhB) and eosin Y (EsY) in bottled chili sauce. The first high performance liquid chromatography-fluorescence detector (HPLC-FLD) was used for the simultaneous estimation of RhB and EsY, based on their native fluorescence. In addition, to overcome the difference in the excitation and emission wavelengths between the two dyes and maintain their sensitivity, a zero-order emission mode was selected in the fluorescence detector (FLD) unit. The excitation wavelength was adapted at 530 nm in mobile phase composed of 0.1% formic acid in ultrapure water (A) and methanol (B) at ratio of 30:70 v/v). Furthermore, using the Sep-Pak® C18 cartridge for solid-phase extraction method, interference from the chili sauce matrix was eliminated. Under the optimum condition, a linear calibration graphs in the range of 0.5- 300 and 0.5- 300 of RhB and EsY with r = 0.9994 and 0.9991, respectively were acquired. The limits of detection for RhB and EsY were 0.344 ng/mL (0.355 ng/g) and 0.215 ng/mL (0.222 ng/g), respectively. The recovery percentage and relative standard deviation of two analytes were obtained in the range of 98.38-101.86% and 0.83-1.66 % respectively. According to the results, the presented method offers specificity, convenience and sufficient sensitivity for the simultaneous estimation of RhB and EsY in wide variety of food matrices.

The hydrodistilled volatile constituents of Cistanche tubulosa (commonly known as Desert Ginseng) have been chemically and biologically investigated. Based on the retention times and mass fragmentation of the obtained GC-MS chromatogram, 106 individual components which representing ≈ 99.29 % of the total volatile constituents have been identified. The major compounds (66.57% of the total composition) were identified as hexanal (15.98%), trans-sabinyl acetate (12.22%), allo-aromadendrene (9.30%), nonanoic acid (6.66%), 3Z- hexeny-2-methyl butanoate (6.09%), valeranone (5.25%), (E, E)-α-Farnesene (3.18%), a-pinene (3.06%), linalool isovalerate (3.03%) and a-humulene (1.8%). Estimation of the antioxidant activity of EO showed promising effect at 80 mg/mL concentration, it exerted 62.40, 863.29 and 62.72 % inhibition compared to TBHQ that showed 78.62, 77.56 and 79.23 % inhibition using DPPH, ABTS and β-carotene/Linoleic acid, respectively. The current study aimed to screen the preliminary phytochemicals in the leaf extract of the medicinal plant Simarouba glauca and to analyze its potential antimicrobial, antioxidant and anticancer properties. The phytochemical profile of the methanol extract was analyzed, and bioactive compounds were identified using chromatography, FTIR and GCMS. Antimicrobial activity and Minimum Inhibitory Concentration (MIC) were determined against 14 bacterial and 6 fungal strains. Moreover, the synergistic effect of a plant extract with commercially available antibiotics was also evaluated using the checkerboard method. The ethanolic and methanolic extracts showed exclusive activity against S. aureus and profound activity against E. coli and S. marcescens. Upon comparing breakpoints, methanolic extract demonstrated higher antimicrobial activity with a MIC value of 3.2 mg/mL against the test pathogens. Furthermore, the extracts demonstrated potential antioxidant activity; methanol extract had higher antioxidant potential compared to the ethanol extract. The major proactive bioactive compound with maximum antioxidant capacity was observed to be terpenoids. The methanol extract of S. glauca showed significant cytotoxicity against the MCF-7 breast cancer cell line with an IC50 value of 16.12 µg/mL. The overall results of our work provide significant evidence for the usage of methanolic extract of S. glauca as an efficient ethnomedicinal agent and a potential candidate for relieving many human ailments.

Graphical abstract, Remdesivir (REM) is considered the first therapeutic option approved by US Food and Drug Administration (FDA) for clinical care in case of hospitalized patients suffering in COVID-19 epidemic. In the presented multilateral comparative search, four eco friendlessness approaches —National Environmental Methods Index (NEMI), Eco-Scale Assessment (ESA), Green Analytical Procedure Index (GAPI), and Analytical Greenness metric (AGREE) are tested to assess 16 analytical chromatographic procedures reported for the analysis of the commonly used antiviral drug; Remdesivir (REM). The values of testing more than one approach when estimating the eco-friendly characters for analytical methods are illustrated in this study. On the light of the outcomes, ESA and AGREE approaches are recommended as they are easily applied and digitally presented. Furthermore, GAPI is also a reliable tool in terms of comprehensiveness for the whole analytical procedures, from sampling till the final assessment. NEMI is the easiest and fastest greenness evaluation tool; however, the information it provides is particularly of limited scope and sometimes inaccurate. To ensure greenness of chromatographic analytical methods, there must be clear planning beforehand, to reduce chemical hazards sent to environment. Additionally, it is highly recommended in method validation protocols to consider the greenness of a given analytical procedure before releasing to routine use. The LC-MS/MS analysis for the active metabolite of REM (Nuc) reported by Avataneo et al. and Du et al. proved to be the best bio-analytical methods regarding the environmental aspects depending on the GAPI and AGREE tools. The objective of the current work was to develop and evaluate suppository dosage form in order to improve ranitidine bioavailability as a substitute to the oral administration. Suppocire (different grades), Witepsol W25 and polyethylene glycol (PEG) were used as suppository bases and prepared by molding method. The prepared formulations were examined for hardness, disintegration time, melting point, content uniformity, drug release, stability and bioavailability. The hardness ranged from 3.82 to 12.53 kg and disintegration time from 13.32 to 28.22 min. The melting points of fatty bases had values from 33.94 to 36.82±0.36ºC while PEG based suppositories melting points were directly proportional chain length. Higher content uniformity was observed in PEG based suppositories due to easy incorporation of RT into water soluble base. Release was affected by hydroxyl value and molecular weight (in cases of fatty and PEG bases respectively). All formulations were relatively stable after 12 months. In vivo studies of all formulations exhibited double peak phenomena. PEG based formula (S8) showed significant higher Cmax (10.05±1 μg/ml) and AUC0-12 (58.313±3.9 µg.h/mL) than fatty bases and oral solution. In conclusion, rectal administration of S8 could be prepared as an alternative to the oral dosage form to improve bioavailability and overcome the first-pass metabolism.

A new highly sensitive high-performance liquid chromatographic method with fluorescence detection (HPLC-FLD) in zero-order emission mode was developed for the first time for the simultaneous determination of piroxicam (PRX) and norfloxacin (NRF) in biological fluids. The fluorescence detector wavelengths were set at 278 nm for excitation and zero-order mode for emission. The zero-order emission mode produced greater sensitivity for the measurement of both drugs than a fixed emission wavelength (446 nm). The new developed method was validated according to International Conference of Harmonization (ICH) guidelines. Linearity was found to be over concentration ranges 0.001-20 μg/ml and 0.00003-0.035 μg/ml for PRX and NRF, respectively. The limits of detection were 4.87 × 10-4 and 1.32 × 10-5 μg/ml for PRX and NRF, and the limits of quantitation were 1.47 × 10-3 and 4.01 × 10-5 μg/ml, respectively. The current fluorescence method was found to be more sensitive than most commonly used analytical methods and was successfully applied for simultaneous determination of PRX and NRF in biological fluids (serum and urine) with recoveries ranging from 91.67% to 100.36% for PRX and from 96.00% to 101.43% for NRF.

Levetiracetam (LEV) has been used for the treatment of epilepsy, especially partial seizures, owing to its pharmacokinetic profile, but the LEV impurity C (IMP C), pyridine-2-ol, is regarded as a main toxic substance. The existing methods for the analysis of LEV and its impurity are not fully effective. This study aimed to develop a simple gas chromatography-mass spectrometric (GC–MS) technique, for the first time, to analyze LEV and its toxic IMP-C without derivatization process, and to compare the technique with the previously reported UPLC method. Two different dosage forms of LEV, i.e. the tablet and syrup, plus IMP-C were determined by the same procedure. The analysis, without any derivatization processes, was performed using the gas chromatography coupled with Single Quadrupole Mass Spectrometer in the selected ion monitoring (SIM) mode that used electron ionization with 70 eV energy from 4 to 400 Amu. The helium carrier gas was run through a TG-5MS column with the flow rate of 1 mL/min via temperature programming. USP validation parameters were employed for the method validation regarding linearity and accuracy. The proposed method was successfully utilized to determine LEV in the tablet and syrup dosage forms. The statistical analysis showed no significant difference (p > 0.05) between this novel technique and the available UPLC method. Overall, the newly developed chromatographic method is characterized as simple and very sensitive for the simultaneous analysis of LEV and its impurity.

Isothiazolinones methylisothiazolinone (MI) and methylchloroisothiazolinone (MCI), and parabens methylparaben (MP), ethylparaben (EP), propylparaben (PP) and butylparaben (BP) are the most common synthetic preservatives. They are all known to be potential skin allergens that lead to contact dermatitis. Thus, the identification of these unsafe chemicals in cosmetic products is of high importance. In the present study, solid-phase extraction (SPE) based on HyperSep reversed-phase C8/benzene sulfonic acid ion exchanger (HyperSep C8/BSAIE) and Sep-Pak C18 sorbents, and ultra-high performance liquid chromatography/diode array detector (UHPLC/DAD) were optimized for the simultaneous determination of MI, MCI, MP, EP, PP and BP in cosmetic products. HyperSep C8/BSAIE and UHPLC/DAD with the eluting solvent mixture (acetonitrile/methanol, 2:1, v/v) and detection wavelength (255 nm) were found to be the optimal conditions, respectively. The method illustrates the excellent linearity range (0.008&ndash, 20 &mu, g/mL) with coefficient of determination (R2, 0.997&ndash, 0.999), limits of detection (LOD, 0.001&ndash, 0.002 &mu, g/mL), precision in terms of relative standard deviation (RSD &lt, 3%, intra-day and &lt, 6%, inter-day) when examining a standard mixture at low (0.07 &micro, g/mL), medium (3 &micro, g/mL) and high (15 &micro, g/mL) concentrations. A total of 31 cosmetic samples were studied, achieving concentrations (MI, not detected (nd)-0.89 &micro, g/g), (MCI, nd-0.62 &micro, g/g), (MP, nd-6.53 &micro, g/g), (EP, nd-0.90 &micro, g/g), (PP, nd-9.69 &micro, g/g) and (BP, nd-17.80 &micro, g/g). Recovery values ranged from 92.33 to 101.43% depending on the types of sample. To our knowledge, this is the first specific method which covers the theme and describes background amounts of such preservatives in cosmetics.

In the present work, a method based on solvent extraction and gas chromatography–mass spectrometry (GC-MS) has been validated for the determination of 1,4-dioxane in cosmetics. Various solvents including ethyl acetate, hexane, methanol, dichloromethane and acetone have been used for the extraction of 1,4-dioxane, among them the ethyl acetate was found to be the most efficient extracting solvent. This method has offered excellent quality parameters for instance linearity (R2 > 0.9991), limit of detection (LOD, 0.00065-0.00091 µg/mL), limit of quantification (LOQ, 0.00217–0.00304 µg/mL) and, precision intra-day (1.65–2.60%, n = 5) and inter-day (0.16–0.32%, n = 5) in terms of relative standard deviation (RSD%). A total of thirty-nine cosmetic samples of different brands and origin have been studied. Among them, the 1,4-dioxane was found in twenty-three samples (FB1-FB7, MC1-MC4, MC6-MC8, HS3, HS5, BL1-BL3, BL5 and PLD1-PLD3) at the levels between 0.15 µg/mL and 9.92 µg/mL, whereas in sixteen samples (MC5, HS1, HS2, SG1-SG5, BL4 and HP1- HP6) was found to be not detected. The recovery values were achieved between 93% and 99% in both low and high level of spiked samples. In comparison to the traditional analytical techniques, the proposed method was found to be very sensitive and cost-effective for the routine analysis of 1,4-dioxane at low concentration in cosmetics.
