Wheat is a major cereal crop that possesses a large allopolyploid genome formed through hybridisation of tetraploid and diploid progenitors. During meiosis, crossovers (COs) are constrained in number to 1–3 per chromosome pair that are predominantly located towards the chromosome ends. This reduces the probability of advantageous traits recombining onto the same chromosome, thus limiting breeding. Therefore, understanding the underlying factors controlling meiotic recombination may provide strategies to unlock the genetic potential in wheat. In this mini-review, we will discuss the factors associated with restricted CO formation in wheat, such as timing of meiotic events, chromatin organisation, pre-meiotic DNA replication and dosage of CO genes, as a means to modulate recombination. Among plausible causes for replicability failure, one that has not received sufficient attention is the environment in which the research is conducted. Consisting of the population, equipment, personnel, and various conditions such as location, time, and weather, the research environment can affect treatments and outcomes, and changes in the research environment that occur when an experiment is redone can affect replicability. We examine the extent to which such changes contribute to replicability failure. Our framework is that of an initial experiment that generates the data and a follow-up experiment that is done the same way except for a change in the research environment. We assume that the initial experiment satisfies the assumptions of the two-sample t-statistic and that the follow-up experiment is described by a mixed model which includes environmental parameters. We derive expressions for the effect that the research environment has on power, sample size selection, p-values, and confidence levels. We measure the size of the environmental effect with the environmental effect ratio (EER) which is the ratio of the standard deviations of environment by treatment interaction and error. Aim: Geographical patterns of migrant species arrival have been little studied, despite their relevance to global change responses. Here, we quantify continent‐wide interspecific variation in spatiotemporal patterns of spring arrival of 30 common migrant bird species and relate these to species characteristics and environmental conditions. Methods: Using citizen science data from EuroBirdPortal, we modelled arrival phenology for 30 Afro‐Palaearctic migrant species across Europe to extract start and duration of species arrival at a 400 km square resolution. We related inter and intraspecific variation in arrival and duration to species characteristics and temperature at the start of the growing season (green‐up). Results: Spatial variation in start of arrival times indicates that it took, on average, 1.6 days for the leading migratory front to move northwards by 100 km (range: 0.6–2.5 days). There was a major gradient in arrival phenology, from species which arrived earlier, least synchronously, in colder temperatures and progressed slowly northwards to species which arrived later, most synchronously and in warmer temperatures and advanced quickly through Europe. The slow progress of early arrivers suggests that temperature limits their northward advance; this group included Aerial Insectivores and species wintering north of the Sahel. For the late arrivers, which included species wintering further south, seasonal resource availability in Africa may delay their arrival into Europe. Main Conclusions: We found support for the green‐wave hypothesis applying widely to migratory landbirds. Species arrival phenologies are linked to ecological differences between taxa, such as diet, and wintering location. Understanding these differences informs predictions of species' sensitivity to global change. Publishing these arrival phenologies will facilitate further research and have additional conservation benefits such as informing designation of hunting seasons. Language outcomes after speech and language therapy in post-stroke aphasia are challenging to predict. This study examines behavioral language measures and resting state fMRI (rsfMRI) as predictors of treatment outcome. Fifty-seven patients with chronic aphasia were recruited and treated for one of three aphasia impairments: anomia, agrammatism, or dysgraphia. Treatment effect was measured by performance on a treatment-specific language measure, assessed before and after three months of language therapy. Each patient also underwent an additional 27 language assessments and a rsfMRI scan at baseline. Patient scans were decomposed into 20 components by group independent component analysis, and the fractional amplitude of low-frequency fluctuations (fALFF) was calculated for each component time series. Post-treatment performance was modelled with elastic net regression, using pre-treatment performance and either behavioral language measures or fALFF imaging predictors. Analysis showed strong performance for behavioral measures in anomia (R2 = 0.948, n = 28) and for fALFF predictors in agrammatism (R2 = 0.876, n = 11) and dysgraphia (R2 = 0.822, n = 18). Models of language outcomes after treatment trained using rsfMRI features may outperform models trained using behavioral language measures in some patient populations. Investigating the neurobiology of language impairment and treatment in chronic stroke aphasia using fMRI requires an understanding of measurement variability within and between participants. In this multicenter study, we evaluated the scan-rescan reliability of an auditory and visual (written) story comprehension paradigm in stroke participants with aphasia (N = 65) and healthy controls (N = 22). The multi-modal task was conducted twice (~1 week apart) on separate visits upon study enrolment and twice again at completion three months later. A non-language visuomotor task was studied in the aphasia group only, which was conducted once per time point (3 months apart). While participants were asked to make responses during the comprehension task, these in-scanner responses were not recorded. Reliability was assessed using intraclass correlation coefficient (ICC) at both group and individual participant levels. The visual story comprehension condition had higher reliability than the auditory condition in both groups, with participants with aphasia exhibiting lower reliability than controls in both conditions (stroke ICC = .43, healthy ICC = .81). Differences in reliability within the group of participants with aphasia were found to be partially explained by overall language impairment as well as greater head motion. In the participants with aphasia, the visuomotor paradigm was found to have greater reliability than the story comprehension task at equivalent interscan intervals (visuomotor = 0.50, comprehension = 0.34), and its reliability was not associated with language impairment. This work highlights the importance of considering the reliability of fMRI tasks in aphasia research, provides strategies to improve reliability and has potential implications for the field of clinical neuroimaging in general. The intensity of pastoral management in areas of High Nature Value farming is declining in some regions of Europe but increasing in others. This affects open habitats of conservation concern, such as the British uplands, where bird species that benefit from low‐intensity grazing may be most sensitive to such polarization. While experimental manipulations of livestock grazing intensities have improved our understanding of upland breeding bird responses in the short term, none have examined the long‐term impacts of altered management on reproductive success.Using a replicated landscape‐scale experiment that started in 2003, we investigated the effects of four grazing treatments (intensive sheep; low‐intensity sheep; low‐intensity mixed sheep and cattle; and no grazing) on the breeding productivity of meadow pipits Anthus pratensis, the most common upland passerine. Surveys were carried out systematically during early (2003 and 2004) and late (2015 and 2016) sampling periods of the experiment to compare the short‐ and long‐term effects of grazing treatments on breeding density and productivity of pipits specifically, but also on the overall bird community.Pipit breeding density was lowest under low‐intensity sheep grazing while the highest egg‐stage nest survival was observed in the same treatment, although no significant treatment effects were detected on overall nest survival or fledgling output. There were no significant differences in treatment effects between the sampling periods on any breeding variable, but overall nest survival was lower in the later sampling period across all treatments.Breeding bird species richness differed between treatments in the later sampling period, with highest species richness in the ungrazed treatment.Synthesis and applications. Livestock grazing management can have different outcomes for different upland birds. Our results showed that, with time, meadow pipit breeding productivity tended to be higher when sheep grazing intensity was reduced and/or mixed with cattle, and lower when livestock were removed, but not significantly so. Removal of grazing, however, can significantly increase bird species richness. The long‐term experiment showed an overall decline in fledglings regardless of grazing treatments, potentially a result of increased predator numbers harboured by nearby developing woodland, highlighting the importance of considering wider landscape processes in grazing management decisions. Climate change is an increasing threat to global biodiversity. Whilst there is growing evidence about the potential effectiveness of some aspects of climate change adaptation, the role for site-based management to increase the resilience of vulnerable populations to climate change has been little studied. Here, we test whether such management may reduce the negative effects of unfavourable weather upon a southern range margin Welsh black grouse Tetrao tetrix population that is vulnerable to climate change. The Welsh black grouse recovery programme funded a range of interventions that contributed to a 39% population increase over 10 years. One likely mechanism linking black grouse populations to climate change is the sensitivity of chicks to high June rainfall. We modelled the relationship between June rainfall and management interventions which aimed to increase breeding success (habitat management and lethal predator control) to test whether management could increase the resilience of black grouse populations to such unfavourable weather. Importantly, we found that the negative effect of June rainfall upon productivity was eliminated at sites where predator control occurred, and that productivity was maximised when predator and habitat management were combined. Active management therefore reduced the negative effects of June rainfall upon this vulnerable southern range margin black grouse population, although further work is required to quantify any limits to the success of such management. Active management should be considered as a tool for climate change adaptation in other vulnerable populations, particularly where there is limited potential for species to undergo climate-driven range changes. Public data archiving (PDA) is widely advocated as a means of achieving open data standards, leading to improved data preservation, increased scientific reproducibility, and transparency, as well as additional data use.Public data archiving was primarily conceived to archive data from short‐term, single‐purpose scientific studies. It is now more widely applied, including to large‐scale citizen science biodiversity recording and monitoring schemes which combine the efforts of volunteers with professional scientists.This may affect the financial security of such schemes by reducing income from data and analytical services. Communication between scheme organizers and researchers may be disrupted, reducing scientific quality and impeding scheme development. It may also have an impact on the participation of some volunteers.Synthesis and applications. In response to the challenges of public data archiving for citizen science biodiversity recording and monitoring schemes, the archive function of scheme organizations should be better recognized by those promoting open data principles. Increased financial support from the public sector or from commercial or academic data users may offset financial risk. Those in favour of public data archiving should do more to facilitate communication between nonscheme users and the originating schemes, while a more flexible approach to data archiving may be required to address potential impacts on volunteer participation. Case: We describe a patient with Bain grade-2b Kienböck disease with a coronal fracture of the distal lunate articular surface with proximal structure collapse. To reestablish midcarpal congruity, we used a bone-anchored suture to repair the coronal split in the lunate in preparation for lunate preservation with osteochondral medial femoral trochlea (MFT) reconstruction. Conclusion: In Bain grade-2b Kienböck disease, lunate reconstruction of both the proximal and distal joint surfaces can be performed. Successful reconstruction of the distal articular surface can be achieved using a bone-anchored FiberWire (Arthrex) suture technique. This can be employed in conjunction with an osteochondral MFT reconstruction of the proximal aspect of the lunate. Patients with this commonly encountered coronal fracture of the distal articular surface may be considered candidates for lunate reconstruction via this technique rather than conventional ablative procedures. Abstract: A consequence of climate change has been an advance in the timing of seasonal events. Differences in the rate of advance between trophic levels may result in predators becoming mismatched with prey availability, reducing fitness and potentially driving population declines. Such “trophic asynchrony” is hypothesized to have contributed to recent population declines of long‐distance migratory birds in particular. Using spatially extensive survey data from 1983 to 2010 to estimate variation in spring phenology from 280 plant and insect species and the egg‐laying phenology of 21 British songbird species, we explored the effects of trophic asynchrony on avian population trends and potential underlying demographic mechanisms. Species which advanced their laying dates least over the last three decades, and were therefore at greatest risk of asynchrony, exhibited the most negative population trends. We expressed asynchrony as the annual variation in bird phenology relative to spring phenology, and related asynchrony to annual avian productivity. In warmer springs, birds were more asynchronous, but productivity was only marginally reduced; long‐distance migrants, short‐distance migrants and resident bird species all exhibited effects of similar magnitude. Long‐term population, but not productivity, declines were greatest among those species whose annual productivity was most greatly reduced by asynchrony. This suggests that population change is not mechanistically driven by the negative effects of asynchrony on productivity. The apparent effects of asynchrony on population trends are therefore either more likely to be strongly expressed via other demographic pathways, or alternatively, are a surrogate for species' sensitivity to other environmental pressures which are the ultimate cause of decline. Modelling species distribution and abundance is important for many conservation applications, but it is typically performed using relatively coarse-scale environmental variables such as the area of broad land-cover types. Fine-scale environmental data capturing the most biologically relevant variables have the potential to improve these models. For example, field studies have demonstrated the importance of linear features, such as hedgerows, for multiple taxa, but the absence of large-scale datasets of their extent prevents their inclusion in large-scale modelling studies., We assessed whether a novel spatial dataset mapping linear and woody-linear features across the UK improves the performance of abundance models of 18 bird and 24 butterfly species across 3723 and 1547 UK monitoring sites, respectively., Although improvements in explanatory power were small, the inclusion of linear features data significantly improved model predictive performance for many species. For some species, the importance of linear features depended on landscape context, with greater importance in agricultural areas., Synthesis and applications. This study demonstrates that a national-scale model of the extent and distribution of linear features improves predictions of farmland biodiversity. The ability to model spatial variability in the role of linear features such as hedgerows will be important in targeting agri-environment schemes to maximally deliver biodiversity benefits. Although this study focuses on farmland, data on the extent of different linear features are likely to improve species distribution and abundance models in a wide range of systems and also can potentially be used to assess habitat connectivity. Aim Time-lagged population synchrony, where spatially separated populations show similar fluctuations in abundance lagged over time, is thought to be driven by dispersal among populations. When dispersal is proportional to population density or positively density dependent, and individuals move readily from population A to population B, then as population A increases, the increased number of dispersers from population A to B will cause a subsequent increase in population B. If true, then time-lagged synchrony should be strongest at a species' typical dispersal distance, because the rate of exchange between populations will be greatest at that distance. Methods We estimated the spatial scale of 1-year-lagged population synchrony for 76 U.K. bird species, using 20 years of bird count data collected at 2,415 locations by the British Breeding Bird Survey. We then compared these spatial scales with published mean natal and breeding dispersal distance estimates (ranging from 0.1 to 25.8 km) for the same species based on an independent, large-scale, mark-recapture dataset of 492,272 bird recaptures in Britain and Ireland. Results We found strong, positive cross-species relationships between the spatial scale of time-lagged synchrony and mean natal and breeding dispersal distance estimates from the mark-recapture study. However, average spatial scales of time-lagged synchrony were more than 60 km longer than those from mark-recapture data, with scales ranging from 5 to 185 km. Main conclusions Ours is the first study to show that the spatial scale of time-lagged synchrony increases with species dispersal distance. The scale of synchrony was larger than expected, probably because mark-recapture data underestimated the real dispersal distances, or because dispersal synchronizes populations at a larger spatial scale than that of dispersal (e.g., through formation of travelling waves), or both. It is important for conservationists to be able to assess the risks that climate change poses to species, in order to inform decision making. Using standardised and repeatable methods, we present a national-scale assessment of the risks of range loss and opportunities for range expansion that climate change could pose for over 3000 plants and animals. Species were selected by their occurrence in England, the primary focus of the study, but climate change impacts were assessed across Great Britain, widening their geographical relevance. A basic risk assessment that compared projected future changes in potential range with recently observed changes classified 21% of species as being at high risk and 6% at medium risk of range loss under a B1 climate change scenario. A greater number of species were classified as having a medium (16%) or high (38%) opportunity to potentially expand their distribution. A more comprehensive assessment, incorporating additional ecological information, including potentially confounding and exacerbating factors (e.g. dispersal, habitat availability and other constraints), was applied to 402 species, of which 35% were at risk of range loss and 42% may expand their range extent. This study covers a temperate region with a significant proportion of species at their poleward range limit; the balance of risks and opportunities from climate change may be different elsewhere. The outcome of both risk assessments varied between taxonomic groups, with bryophytes and vascular plants containing the greatest proportion of species at risk from climate change. Upland habitats contained more species at risk than other habitats. Whilst the overall pattern was clear, confidence was generally low for individual assessments, with the exception of well-studied taxa such as birds. Yen1/GEN1 are canonical Holliday junction resolvases that belong to the RAD2/XPG family. In eukaryotes, such as budding yeast, mice, worms, and humans, Yen1/GEN1 work together with Mus81-Mms4/MUS81-EME1 and Slx1-Slx4/SLX1-SLX4 in DNA repair by homologous recombination to maintain genome stability. In plants, the biological function of Yen1/GEN1 remains largely unclear. In this study, we characterized the loss of function mutants of OsGEN1 and OsSEND1, a pair of paralogs of Yen1/GEN1 in rice (Oryza sativa). We first investigated the role of OsGEN1 during meiosis and found a reduction in chiasma frequency by ~6% in osgen1 mutants, compared to the wild type, suggesting a possible involvement of OsGEN1 in the formation of crossovers. Postmeiosis, OsGEN1 foci were detected in wild-type microspore nuclei, but not in the osgen1 mutant concomitant with an increase in double-strand breaks. Persistent double-strand breaks led to programmed cell death of the male gametes and complete male sterility. In contrast, depletion of OsSEND1 had no effects on plant development and did not enhance osgen1 defects. Our results indicate that OsGEN1 is essential for homologous recombinational DNA repair at two stages of microsporogenesis in rice. [ABSTRACT FROM AUTHOR]

During meiosis, centromeres in some species undergo a series of associations, but the processes and progression to homologous pairing is still a matter of debate. Here, we aimed to correlate meiotic centromere dynamics and early telomere behaviour to the progression of synaptonemal complex ( SC) construction in hexaploid wheat (2 n = 42) by triple immunolabelling of CENH3 protein marking functional centromeres, and SC proteins ASY1 (unpaired lateral elements) and ZYP1 (central elements in synapsed chromosomes). We show that single or multiple centromere associations formed in meiotic interphase undergo a progressive polarization (clustering) at the nuclear periphery in early leptotene, leading to formation of the telomere bouquet. Critically, immunolabelling shows the dynamics of these presynaptic centromere associations and a structural reorganization of the centromeric chromatin coinciding with key events of synapsis initiation from the subtelomeric regions. As short stretches of subtelomeric synapsis emerged at early zygotene, centromere clusters lost their strong polarization, gradually resolving as individual centromeres indicated by more than 21 CENH3 foci associated with unpaired lateral elements. Only following this centromere depolarization were homologous chromosome arms connected, as observed by the alignment and fusion of interstitial ZYP1 loci elongating at zygotene so synapsis at centromeres is a continuation of the interstitial synapsis. Our results thus reveal that centromere associations are a component of the timing and progression of chromosome synapsis, and the gradual release of the individual centromeres from the clusters correlates with the elongation of interstitial synapsis between the corresponding homologues. Background Reconstruction of high-risk fascia, tendon, or ligament defects may benefit from vascularized tissue. The iliotibial band (ITB), a thick fibrous tract of connective tissue, serves as a potential donor site for free tissue transfer but its blood supply has not been thoroughly investigated. The aim of this anatomical study was to investigate the vascular supply to the distal ITB and its role as a free fascial flap. Methods We dissected 16 fresh-frozen cadaveric legs and injected latex into the superolateral geniculate artery (SLGA). A distal ITB fascial flap was designed and measurements were taken for flap dimensions, pedicle length and size, and SLGA perfusion territory. Results The SLGA perfused 11.5 ± 2.3 cm of distal ITB (proximal to the lateral femoral epicondyle) and provided 6.4 ± 0.7cm of pedicle length to the ITB flap. Conclusions Chimeric options to include bone (from the lateral femoral condyle), cartilage (from the lateral femoral trochlea), muscle (from vastus lateralis or biceps femoris), and skin are possible. Surgical harvest techniques are proposed, including preservation of ITB insertions to minimize lateral knee instability. Clinical validation is needed to determine the role of the distal ITB free fascial flap in reconstructive microsurgery. [ABSTRACT FROM AUTHOR]

Despite increasing concerns about the vulnerability of species' populations to climate change, there has been little overall synthesis of how individual population responses to variation in climate differ between taxa, with trophic level or geographically. To address this, we extracted data from 132 long-term (greater than or equal to 20 years) studies of population responses to temperature and precipitation covering 236 animal and plant species across terrestrial and freshwater habitats. Our results identify likely geographical differences in the effects of climate change on populations and communities in line with macroecological theory. Temperature tended to have a greater overall impact on populations than precipitation, although the effects of increased precipitation varied strongly with latitude, being most positive at low latitudes. Population responses to increased temperature were generally positive, but did not vary significantly with latitude. Studies reporting significant climatic trends through time tended to show more negative effects of temperature and more positive effects of precipitation upon populations than other studies, indicating climate change has already impacted many populations. Most studies of climate change impacts on biodiversity have focused on temperature and are from middle to high northern latitudes. Yet many of its core proteins show evidence of rapid or adaptive evolution. What drives the evolution of meiosis proteins? How can constrained meiotic processes be modified in response to challenges without compromising their essential functions? In surveying the literature, we found evidence of two especially potent challenges to meiotic chromosome segregation that probably necessitate adaptive evolutionary responses: whole‐genome duplication and abiotic environment, especially temperature. Evolutionary solutions to both kinds of challenge are likely to involve modification of homologous recombination and synapsis, probably via adjustments of core structural components important in meiosis I. Synthesizing these findings with broader patterns of meiosis gene evolution suggests that the structural components of meiosis coevolve as adaptive modules that may change in primary sequence and function while maintaining three‐dimensional structures and protein interactions. The often sharp divergence of these genes among species probably reflects periodic modification of entire multiprotein complexes driven by genomic or environmental changes. We suggest that the pressures that cause meiosis to evolve to maintain fertility may cause pleiotropic alterations of global crossover rates. We highlight several important areas for future research. Climate change is reported to have caused widespread changes to species' populations and ecological communities. Warming has been associated with population declines in long-distance migrants and habitat specialists, and increases in southerly distributed species. However, the specific climatic drivers behind these changes remain undescribed., We analysed annual fluctuations in the abundance of 59 breeding bird species in England over 45 years to test the effect of monthly temperature and precipitation means upon population trends., Strong positive correlations between population growth and both winter and breeding season temperature were identified for resident and short-distance migrants. Lagged correlations between population growth and summer temperature and precipitation identified for the first time a widespread negative impact of hot, dry summer weather. Understanding large-scale drivers of animal breeding densities and demography has a range of important uses, including informing conservation management. Given the threat of climate change, the importance of developing a process-based understanding of variation in animal populations is increased to inform adaptive management. For a climate-change sensitive species, the European Golden Plover Pluvialis apricaria, we use novel field-collected data on large-scale spatial variation in prey abundance and vegetation structure to understand drivers of breeding abundance and breeding success, and inform potential management responses. The abundance of the key prey, crane flies (Tipulidae), increased with altitude (a surrogate for temperature) and peat depth (a surrogate for soil moisture). Golden plover breeding densities were highest where vegetation was shortest, probably reflecting greater prey accessibility. In contrast, breeding success was not strongly related to vegetation height, but positively correlated with both crane fly abundance and daily minimum temperatures. When combined to model the number of likely successful pairs in any 1 year, the magnitude of vegetation height effect far exceeded that of crane fly abundance. Thus, for golden plover and other shorebirds sharing similar habitats, management to optimize breeding habitat (grazing or burning to promote short vegetation) may differ from management to promote breeding success (drain blocking to increase soil moisture and prey abundance). Adaptive management in the face of climate change should therefore include appropriate vegetation management, as well as maximizing prey abundance. More broadly, as the drivers of breeding density and demographic parameters may differ, we advocate that conservation practitioners collect not just information on species' distributions but also underpinning demographic processes when using science to inform management. The formation of at least one chiasma/CO between homologous chromosome pairs is essential for accurate chromosome segregation at the first meiotic division as well as for generating genetic variation. Although DNA double-strand breaks, which initiate recombination, are widely distributed along the chromosomes, this is not necessarily reflected in the chiasma distribution. In many species there is a tendency for chiasmata to be distributed in favored regions along the chromosomes, whereas in others, such as barley and some other grasses, chiasma localization is extremely pronounced. Localization of chiasma to the distal regions of barley chromosomes restricts the genetic variation available to breeders. Studies reviewed herein are beginning to provide an explanation for chiasma localization in barley. Background The medial and lateral femur provide a source of convex osteochondral vascularized bone. The medial trochlea has been demonstrated to have similar contour to the proximal scaphoid, lunate, and capitate. Other sites of osteochondral harvest such as the posteromedial femur and the lateral trochlea are similar in morphology to the humeral capitellum and medial talus, respectively. These analogous structures offer potential solutions to difficult articular problems. Patients and Methods Patients who underwent osteochondral reconstruction of the extremities were reviewed. These included 16 medial femoral trochlea (MFT) scaphoid nonunion reconstructions, 16 MFT Kienböck lunate reconstructions, 5 MFT capitate reconstructions, 2 lateral femoral trochlea osteochondral reconstructions of medial tarsal avascular necrosis, and 5 posteromedial femoral osteochondral reconstructions of the humeral capitellum for posttraumatic arthritis. Results Computed tomography (CT) imaging demonstrated 15 of 16 reconstructed scaphoids achieving union. Follow-up wrist motion averaged 46.0 degrees extension and 43.8 degrees flexion, similar to preoperative measurements. CT imaging confirmed healing in 15 of 16 reconstructed lunates. Lichtman staging remained unchanged in 10 patients, improved in 4 patients (3A-2), and worsened in 2 patients (3A-3B). All but one patient experienced improvement in wrist pain. Wrist range of motion at follow-up averaged 50 degrees extension and 38 degrees flexion, similar to preoperative measurements. Capitate, talar, and capitellar reconstructions all resulted in achievement of osseous healing and restoration of joint alignment. Conclusions Vascularized osteochondral flaps provide a useful tool in the treatment of difficult articular problems in the extremities. Large-scale declines in terrestrial insects have been reported over much of Europe and across the world, however, population change assessments of other key invertebrate groups, such as soil invertebrates, have been largely neglected through a lack of available monitoring data. This study collates historic data from previously published studies to assess whether it is possible to infer previously undocumented long-term changes in soil invertebrate abundance. Earthworm and tipulid data were collated from over 100 studies across the UK, spanning almost 100 years. Analyses suggested long-term declines in earthworm abundance of between 1.6 to 2.1% per annum, equivalent to a 33% to 41% decline over 25 years. These appeared greatest in broadleaved woodlands and farmland habitats, and were greater in pasture than arable farmland. Significant differences in earthworm abundance between habitats varied between models but appeared to be highest in urban greenspaces and agricultural pasture. More limited data were available on tipulid abundance, which showed no significant change over time or variation between enclosed farmland and unenclosed habitats. Declines in earthworm populations could be contributing to overall declines in ecosystem function and biodiversity as they are vital for a range of ecosystem services and are keystone prey for many vertebrate species. If robust, our results identify a previously undetected biodiversity decline that would be a significant conservation and economic issue in the UK, and if replicated elsewhere, internationally. In many cereal crops, meiotic crossovers predominantly occur toward the ends of chromosomes and 30 to 50% of genes rarely recombine. This limits the exploitation of genetic variation by plant breeding. Previous reports demonstrate that chiasma frequency can be manipulated in plants by depletion of the synaptonemal complex protein ZIPPER1 (ZYP1) but conflict as to the direction of change, with fewer chiasmata reported in Arabidopsis thaliana and more crossovers reported for rice (Oryza sativa). Here, we use RNA interference (RNAi) to reduce the amount of ZYP1 in barley (Hordeum vulgare) to only 2 to 17% of normal zygotene levels. In the ZYP1RNAi lines, fewer than half of the chromosome pairs formed bivalents at metaphase and many univalents were observed, leading to chromosome nondisjunction and semisterility. The number of chiasmata per cell was reduced from 14 in control plants to three to four in the ZYP1-depleted lines, although the localization of residual chiasmata was not affected. DNA double-strand break formation appeared normal, but the recombination pathway was defective at later stages. A meiotic time course revealed a 12-h delay in prophase I progression to the first labeled tetrads. Meiosis involves reciprocal exchange of genetic information between homologous chromosomes to generate new allelic combinations. In cereals, the distribution of genetic crossovers, cytologically visible as chiasmata, is skewed toward the distal regions of the chromosomes. However, many genes are known to lie within interstitial/proximal regions of low recombination, creating a limitation for breeders. We investigated the factors underlying the pattern of chiasma formation in barley (Hordeum vulgare) and show that chiasma distribution reflects polarization in the spatiotemporal initiation of recombination, chromosome pairing, and synapsis. Consequently, meiotic progression in distal chromosomal regions occurs in coordination with the chromatin cycles that are a conserved feature of the meiotic program. Recombination initiation in interstitial and proximal regions occurs later than distal events, is not coordinated with the cycles, and rarely progresses to form chiasmata. Early recombination initiation is spatially associated with early replicating, euchromatic DNA, which is predominately found in distal regions. We demonstrate that a modest temperature shift is sufficient to alter meiotic progression in relation to the chromosome cycles. The polarization of the meiotic processes is reduced and is accompanied by a shift in chiasma distribution with an increase in interstitial and proximal chiasmata, suggesting a potential route to modify recombination in cereals. A variable-number tandem repeat (VNTR) protocol targeting 10 loci in the Brucella abortus genome was used to assess genetic diversity among 366 field isolates recovered from cattle, bison, and elk in the Greater Yellowstone Area (GYA) and Texas during 1998 to 2011. Minimum spanning tree (MST) and unweighted-pair group method with arithmetic mean (UPGMA) analyses of VNTR data identified 237 different VNTR types, among which 14 prominent clusters of isolates could be identified. Cattle isolates from Texas segregated into three clusters: one comprised of field isolates from 1998 to 2005, one comprised of vaccination-associated infections, and one associated with an outbreak in Starr County in January 2011. An isolate obtained from a feral sow trapped on property adjacent to the Starr County herd in May 2011 clustered with the cattle isolates, suggesting a role for feral swine as B. abortus reservoirs in Starr County. Isolates from a 2005 cattle outbreak in Wyoming displayed VNTR-10 profiles matching those of strains recovered from Wyoming and Idaho elk. Additionally, isolates associated with cattle outbreaks in Idaho in 2002, Montana in 2008 and 2011, and Wyoming in 2010 primarily clustered with isolates recovered from GYA elk. The human hereditary disease Fanconi anemia leads to severe symptoms, including developmental defects and breakdown of the hematopoietic system. It is caused by single mutations in the FANC genes, one of which encodes the DNA translocase FANCM (for Fanconi anemia complementation group M), which is required for the repair of DNA interstrand cross-links to ensure replication progression. We identified a homolog of FANCM in Arabidopsis thaliana that is not directly involved in the repair of DNA lesions but suppresses spontaneous somatic homologous recombination via a RecQ helicase (At-RECQ4A)–independent pathway. In addition, it is required for double-strand break–induced homologous recombination. The fertility of At- fancm mutant plants is compromised. Evidence suggests that during meiosis At-FANCM acts as antirecombinase to suppress ectopic recombination-dependent chromosome interactions, but this activity is antagonized by the ZMM pathway to enable the formation of interference-sensitive crossovers and chromosome synapsis. Surprisingly, mutation of At-FANCM overcomes the sterility phenotype of an At- MutS homolog4 mutant by apparently rescuing a proportion of crossover-designated recombination intermediates via a route that is likely At-MMS and UV sensitive81 dependent. However, this is insufficient to ensure the formation of an obligate crossover. Summary 1. There has been a rapid increase in the development of renewable energy because of the need to combat climate change. One of the most widely used technologies has been onshore wind farms. These have the potential to affect birds through disturbance or collision, but the extent to which such developments cause general population declines, and therefore are of wider conservation concern, remains largely untested. 2. Monitoring data from wind farms located on unenclosed upland habitats in the UK were collated to test whether breeding densities of upland birds were reduced as a result of wind farm construction or during wind farm operation. 3. Data were available for ten species although none were raptors. Red grouse Lagopus lagopus scoticus, snipe Gallinago gallinago and curlew Numenius arquata densities all declined on wind farms during construction. Red grouse densities recovered after construction, but snipe and curlew densities did not. Post-construction curlew densities on wind farms were also significantly lower than reference sites. Conversely, densities of skylark Alauda arvensis and stonechat Saxicola torquata increased on wind farms during construction. 4. There was little evidence for consistent post-construction population declines in any species, suggesting for the first time that wind farm construction can have greater impacts upon birds than wind farm operation. 5. The impacts of wind farms were largely unaffected by technical specifications (turbine height, number or total generating power) and therefore are widely applicable. 6. Synthesis and applications. This study confirms that regulatory authorities and developers should particularly consider the likely impacts of wind farms on large waders. Greater weight should be given to the effects of construction on wildlife in impact assessments than at present. Mitigation measures during construction, including restricting construction activity to non-breeding periods, should be considered and tested as a means to reduce these negative effects. Threats to biodiversity resulting from habitat destruction and deterioration have been documented for many species, whilst climate change is regarded as increasingly impacting upon species' distribution and abundance. However, few studies have disentangled the relative importance of these two drivers in causing recent population declines. We quantify the relative importance of both processes by modelling annual variation in population growth of 18 farmland bird species in the UK as a function of measures of land-use intensity and weather. Modelled together, both had similar explanatory power in accounting for annual fluctuations in population growth. When these models were used to retrodict population trends for each species as a function of annual variation in land-use intensity and weather combined, and separately, retrodictions incorporating land-use intensity were more closely linked to observed population trends than retrodictions based only on weather, and closely matched the UK farmland bird index from 1970 onwards. Despite more stable land-use intensity in recent years, climate change (inferred from weather trends) has not overtaken land-use intensity as the dominant driver of bird populations. In this study we have analysed AtASY3, a coiled-coil domain protein that is required for normal meiosis in Arabidopsis. Analysis of an Atasy3-1 mutant reveals that loss of the protein compromises chromosome axis formation and results in reduced numbers of meiotic crossovers (COs). Although the frequency of DNA double-strand breaks (DSBs) appears moderately reduced in Atasy3-1, the main recombination defect is a reduction in the formation of COs. Immunolocalization studies in wild-type meiocytes indicate that the HORMA protein AtASY1, which is related to Hop1 in budding yeast, forms hyper-abundant domains along the chromosomes that are spatially associated with DSBs and early recombination pathway proteins. Loss of AtASY3 disrupts the axial organization of AtASY1. Furthermore we show that the AtASY3 and AtASY1 homologs BoASY3 and BoASY1, from the closely related species Brassica oleracea, are co-immunoprecipitated from meiocyte extracts and that AtASY3 interacts with AtASY1 via residues in its predicted coiled-coil domain. Together our results suggest that AtASY3 is a functional homolog of Red1. Since studies in budding yeast indicate that Red1 and Hop1 play a key role in establishing a bias to favor inter-homolog recombination (IHR), we propose that AtASY3 and AtASY1 may have a similar role in Arabidopsis. Loss of AtASY3 also disrupts synaptonemal complex (SC) formation. In Atasy3-1 the transverse filament protein AtZYP1 forms small patches rather than a continuous SC. The few AtMLH1 foci that remain in Atasy3-1 are found in association with the AtZYP1 patches. This is sufficient to prevent the ectopic recombination observed in the absence of AtZYP1, thus emphasizing that in addition to its structural role the protein is important for CO formation. Its size and location enable it to serve as a useful recipient vessel in free flap reconstruction about the knee. It provides the ease and convenience of two-team surgery in the supine position and its distal position permits convenient and distal insetting of the transferred flap of choice. Management initially focused on reestablishment of hand function and provision of soft tissue coverage via local and free tissue transfer. Secondary procedures involved partial flap elevation, targeted debulking, and redefining topographic regions where possible. By designing asymmetric skin excision patterns, the resultant length and width discrepancies can be used to purposefully enhance topographic features while re-creating the appearance of natural crease lines bordering adjacent subterritories. We have analysed the role of RBR (retinoblastoma related), the Arabidopsis homologue of the tumour suppressor Retinoblastoma protein (pRb), during meiosis. We characterise the rbr-2 mutation, which causes a loss of RBR in male meiocytes. The rbr-2 plants exhibit strongly reduced fertility, while vegetative growth is generally unaffected. The reduced fertility is due to a meiotic defect that results in reduced chiasma formation and subsequent errors in chromosome disjunction. Immunolocalisation studies in wild-type meiocytes reveal that RBR is recruited as foci to the chromosomes during early prophase I in a DNA double-strand-break-dependent manner. In the absence of RBR, expression of several meiotic genes is reduced. The localisation of the recombinases AtRAD51 and AtDMC1 is normal. However, localisation of the MutS homologue AtMSH4 is compromised. Additionally, polymerisation of the synaptonemal complex protein AtZYP1 is abnormal. Together, these data indicate that loss of RBR during meiosis results in a reduction of crossover formation and an associated failure in chromosome synapsis. RecQ helicases are a conserved group of proteins with a role in the maintenance of genome integrity. In Saccharomyces cerevisiae (budding yeast), meiotic recombination is increased in the absence of the RecQ helicase Sgs1. Here we investigated the potential meiotic role of the Sgs1 homologue AtRECQ4A and the closely related AtRECQ4B. Both proteins have been shown to function during recombination in somatic cells, but so far their meiotic role has not been investigated. Both AtRECQ4A and AtRECQ4B were expressed in reproductive tissues. Although immunolocalization studies showed that AtRECQ4A associates with recombination intermediates, we found no evidence that its loss or that of AtRECQ4B had a significant effect on meiotic cross-overs, suggesting functional redundancy with other RECQ family members. Nevertheless, pollen viability decreased in Atrecq4A, resulting in a reduction in fertility, although this was not the case in Atrecq4B. Cytological analysis revealed chromatin bridges between the telomeres of non-homologous chromosomes in Atrecq4A at metaphase I, in some instances accompanied by chromosome fragmentation at anaphase I. The bridges required telomeric repeats and were dependent on meiotic recombination. Immunolocalization confirmed the association of AtRECQ4A with the telomeres during prophase I, which we propose enables dissolution of recombination-dependent telomeric associations. Thus, this study has identified a hitherto unknown role for a member of the RECQ helicase family during meiosis that contributes to the maintenance of chromosome integrity. Understanding the mechanisms by which climate change will affect animal populations is vital for adaptive management. Many studies have described changes in the timing of biological events, which can produce phenological mismatch. Direct effects on prey abundance might also be important, but have rarely been studied. We examine the likely importance of variation in prey abundance in driving the demographics of a European golden plover ( Pluvialis apricaria) population at its southern range margin. Previous studies have correlated plover productivity with the abundance of their adult cranefly (Tipulidae) prey, and modelled the phenology of both plover breeding and cranefly emergence in relation to temperature. Our analyses demonstrate that abundance of adult craneflies is correlated with August temperature in the previous year. Correspondingly, changes in the golden plover population are negatively correlated with August temperature 2 years earlier. Predictions of annual productivity, based on temperature-mediated reductions in prey abundance, closely match observed trends. Modelled variation in annual productivity for a future scenario of increasing August temperatures predicts a significant risk of extinction of the golden plover population over the next 100 years, depending upon the magnitude of warming. Direct effects of climate warming upon cranefly populations may therefore cause northward range contractions of golden plovers, as predicted by climate envelope modelling. Craneflies are an important food source for many northern and upland birds, and our results are likely to have wide relevance to these other species. Using data from 12 upland wind farms in the UK, we examine whether there is reduced occurrence of breeding birds close to wind farm infrastructure (turbines, access tracks and overhead transmission lines). To our knowledge, this is the first such multi-site comparison examining wind farm effects on the distribution of breeding birds. 2. Bird distribution was assessed using regular surveys during the breeding season. We took a conservative analytical approach, with bird occurrence modelled as a function of habitat, before examining the additional effects of wind farm proximity. 3. Seven of the 12 species studied exhibited significantly lower frequencies of occurrence close to the turbines, after accounting for habitat variation, with equivocal evidence of turbine avoidance in a further two. No species were more likely to occur close to the turbines. There was no evidence that raptors altered flight height close to turbines. Turbines were avoided more strongly than tracks, whilst there was no evidence for consistent avoidance of overhead transmission lines connecting sites to the national grid. 4. Levels of turbine avoidance suggest breeding bird densities may be reduced within a 500-m buffer of the turbines by 15–53%, with buzzard Buteo buteo, hen harrier Circus cyaneus, golden plover Pluvialis apricaria, snipe Gallinago gallinago, curlew Numenius arquata and wheatear Oenanthe oenanthe most affected. 5. Despite being a correlative study, with potential for Type I error, we failed to detect any systematic bias in our likelihood of detecting significant effects. 6. Synthesis and applications. This provides the first evidence for consistent and significant effects of wind farms on a range of upland bird species, emphasizing the need for a strategic approach to ensure such development avoids areas with high densities of potentially vulnerable species. St Thomas Aquinas enjoys the repute of having given five proofs of God's existence, the famous ‘Five Ways’: Respondeo dicendum quod Deum esse probari potest quinque viis– I reply that you have to say that God's existence can be proved in five ways – and here they come! For students of philosophy they make their deadly, inescapable appearance in any and every manual, trotted out with a granite certainty comparable only to that with which we are assured that English Catholics once nearly succeeded in blowing up the Houses of Parliament. Here, however, our concern is with St Thomas's teaching method, rather than with a readers' consensus about what he said, that of a readership largely content to ignore the question: and this article is an endeavour, not indeed to ‘flog a dead horse’, but simply to explain the pedagogy implicit in the great Summa. That simply means, how to read it with as much ease and understanding as its author intended – and indeed indicated from his opening sentence onwards. The article ends by mentioning some highly regrettable consequences of the neglect, briefly updating (if the paradox be allowed) the post-Tridentine moral tradition, by adding to it from St Thomas's nowadays largely forgotten legacy.
