The extraction, isolation, purification, and structural characterization of proso millet bran polysaccharides (MBP), as well as its antibacterial activity were conducted to provide a reference for the development and utilization of nutrients in the processing byproducts of millet. An alkali extraction and alcohol precipitation method was used to extract the polysaccharides. Subsequent purification was achieved through column chromatography utilizing DEAE-50 and Sephadex G-100. Structural analysis of the millet bran polysaccharides was performed using a combination of techniques, including HPLC chromatography, infrared spectroscopy, atomic force microscopy, scanning electron microscopy, and X-ray diffraction. The results showed that the molecular weight of MBP was determined to be 3.479×104 Da. The monosaccharide composition was exhibited a molar ratio of mannose:rhamnose:glucose:galactose:arabinose:xylose=0.11:0.13:5.86:0.62:1.00:0.52. Moreover, MBP was identified as a β-type pyranose polysaccharide and exhibits remarkable thermal stability. In terms of morphology, MBP was laminated, closely connected, and had no crystal structure. The antibacterial assay unveiled the minimum inhibitory concentration of MBP against Staphylococcus aureus was 2 mg/mL. This study showed that the polysaccharides from proso millet bran had good antibacterial ability.

Abstract Objective This study aimed to compare and analyze the expression and significance of the GRP78 protein in cochlear cell injury induced by a high glucose and high-fat diet in obese and diabetic rats. Methods Male SD rats were randomly divided into two groups: normal (NC) and high-fat (HF) groups. The NC group was fed a standard diet for eight weeks, while the HF group received a high-glucose, high-fat diet. The HF group was further categorized into the obesity group (OB group) and the type II diabetes mellitus group (T2DM group). To induce a type II diabetes mellitus (T2DM) model, the T2DM group received an intraperitoneal injection of a small dose of STZ (45 mg/kg). After four weeks on the original diet, body weight, blood glucose, blood lipid levels, and auditory brainstem response (ABR) thresholds were measured. The cochlea was dissected, and its morphology was observed using HE staining. Immunohistochemistry and western blotting were utilized to examine the expression level of the GRP78 protein in the cochlea. Based on the Guangzhou Business Center project, a typical super high-rise building with an asymmetric plan, taking the construction speed, closure time of mega braces and belt trusses as influencing factors, a parametric analysis on its lateral and vertical deformations, as well as the maximum stress of key structural members was conducted. The analysis results indicated that the construction speed had a relatively small impact on the deformation and the maximum stress of key members. However, synchronous closure of belt truss compared with the delayed closure would result in smaller horizontal and vertical deformation differences, as well as the stress of belt truss. Meanwhile, the closure timing of the mega braces had little influence on the vertical deformation difference and the stress of belt truss. And the earlier the closure, the smaller the horizontal drift ratio, the greater the maximum stress of the mega braces. Further, deformation control measurements were brought forward. On the one hand, FEM simulation was carried out according to the above construction suggestions. On the other hand, real-time monitoring was also used. Finally, by comparing both results, proposed construction deformation control measures and simulation methods were verified.

Backgrounds: This study aims to explore the clinical value of P4HA2 (prolyl 4-hydroxylase subunit alpha 2) in Osteosarcoma (OSC), and assess its potential to provide directions and clues for the practice of precision nursing. Methods: The GSE73166 and GSE16088 datasets were used to explore the P4HA2 expression in OSC. We then used the clinical data of patients obtaining from TARGET database to assess the prognostic value of P4HA2 in OSC. We also evaluated the predictive value of prognostic model based on P4HA2-related genes. Further, GSEA analysis was performed to explore related pathways. Results: The P4HA2 mRNA expression was higher in OSC than that in normal tissues and other bone cancer samples. Survival analysis found that P4HA2 high expression caused poor overall survival (OS) of patients with OSC and P4HA2 presented a favorable performance for predicting OS. Specifically, P4HA2 high expression statistically influenced the OS of patients with age≥15 years old and those with or without metastasis. Cox regression analysis indicated the independent prognostic value of P4HA2 in OSC, and nomogram analysis revealed its significant contribution to the survival probability of patients. We further established a prognostic model based on P4HA2-related genes, finding that prognostic model had a good prediction ability on OS. These results supported the clinical significance of P4HA2 in OSC. GSEA analysis suggested that P4HA2 was significantly related to the MAPK signaling pathway. In addition, P4HA2-associated natural killer cell-mediated cytotoxicity and T cell receptor signaling pathway were also predicted. Conclusions: This study revealed that P4HA2 can serve as an important prognostic biomarker for OSC patients, and it may become a promising therapeutic target in OSC treatment.

Human reliability analysis (HRA) is widely used to evaluate the impact of human errors on various complex human–machine systems for enhancing their safety and reliability. Nevertheless, it is hard to estimate the human error probability (HEP) in reality due to the uncertainty of state assessment information and the complex relations among common performance conditions (CPCs). In this paper, we aim to present a new integrated cognitive reliability and error analysis method (CREAM) to solve the HRA problems under probabilistic linguistic environment. First, the probabilistic linguistic term sets (PLTSs) are utilized to handle the uncertain task state assessments provided by experts. Second, the minimum conflict consensus model (MCCM) is employed to deal with conflict task state assessment information to assist experts reach consensus. Third, the entropy weighting method is used to determine the relative objective weights of CPCs. Additionally, the CPC effect indexes are introduced to assess the overall effect of CPCs on performance reliability and obtain the HEP estimation. Finally, the reliability of the proposed CREAM is demonstrated via a healthcare practical case. The result shows that the new integrated CREAM can not only effectively represent experts’ uncertain task state assessments but also determine more reliable HEP estimation in HRA.

Carbon fiber-reinforced polymer (CFRP) components were extensively used and current studies mainly refer to CFRP laminates. The dynamic performance of the complicated CFRP antenna beams is yet to be explored. Therefore, a sensor layout based on fiber Bragg gratings (FBGs) in series was designed to measure the dynamic response of the CFRP antenna beam, and various vibration tests (sweep frequency test, simulated long-life vibration test, shock vibration test, functional vibration test, and constant frequency vibration test) were conducted. The time and frequency domain analysis on FBG sensing signals was performed to check the vibration performance and assess the health condition of this novel CFRP structure. The results indicate that strain values reach a maximum of only 300 µε under different test conditions. The antenna beam exhibited clear vibration patterns, with the first four intrinsic frequencies identified at 44, 94.87, 107.1, and 193.45 Hz. It shows that strain distribution and vibration modes of the antenna beam can be characterized from the sensing data, and the dynamic feature can be much more accurately assessed. The FBG sensors attached on the surface of CFRP antenna beam can accurately and stably measure the dynamic response, which validates that the interfaces between optical fiber sensing elements and CFRP material have excellent interfacial bonding characteristics. The novel CFRP antenna beam exhibits the excellent dynamic performance and stability, offering the replacement of traditional steel antenna beams. The study can finally instruct the development of self-sensing CFRP antenna beams assembled with FBGs in series.

Abstract Objective To investigate the prescription rate of short-term systemic use of glucocorticoids during hospitalization in patients with cardiogenic shock (CS), and outcomes related with glucocorticoid use. Methods We extracted patients' information from the Medical Information Mart for Intensive Care IV version 2.0 (MIMIC-IV v2.0) database. The primary endpoint was 90-day all-cause mortality. Secondary safety endpoints were infection identified by bacterial culture and at least one episode of hyperglycemia after ICU admission. Propensity score matching (PSM) was used to balance baseline characteristics. The difference in cumulative mortality rate between these treated with and without glucocorticoids was assessed by Kaplan–Meier curve with log-rank test. Independent risk factors for endpoints were identified by Cox or Logistic regression analysis. Results A total of 1528 patients were enrolled, and one-sixth of these patients received short-term systemic therapy of glucocorticoids during hospitalization. These conditions, including rapid heart rate, the presence of rheumatic disease, chronic pulmonary disease and septic shock, high lactate level, the requirements of mechanical ventilation and continuous renal replacement therapy, were associated with an increase in glucocorticoid administration (all P ≤ 0.024). Abstract Chronic wounds seriously affect the quality of life of the elderly, obese people, and diabetic patients. The excessive inflammatory response is a key driver of delayed chronic wound healing. Although lavender essential oil (EO [lav]) has been proven to have anti‐inflammatory and accelerate wound curative effects, the specific molecular mechanism involved is still ambiguous. The results showed that the wounds treated with lipopolysaccharide (LPS) not only had delayed healing, but also the expression levels of pro‐inflammatory cytokines, such as tumor necrosis factor‐α (TNF‐α), interleukin‐6 (IL‐6), interleukin‐1β (IL‐1β), and the inflammatory mediator protein, high‐mobility group box 1 protein (HMGB‐1), in the wound tissues were significantly increased. However, treatment of LPS‐induced chronic wounds with EO (lav) accelerated wound healing and decreased IL‐1β and HMGB‐1 expression levels. It was further found that LPS induced macrophage pyroptosis to produce IL‐1β. After treatment with EO (lav), the expression level of macrophage pyroptosis marker Gasdermin D (GSDMD) and pyroptosis‐related cytotoxic effects were significantly reduced. Immunofluorescence results also directly indicate that EO (lav) can protect macrophages from LPS‐induced pyroptosis. Moreover, EO (lav) can down‐regulate expression levels of IL‐1β, GSDMD, and nucleotide‐binding oligomerization domain‐like receptor protein 3 (NLRP3) in the caspase‐11‐related pyroptotic signaling pathway. This study demonstrates that EO (lav) can reduce proinflammatory factor production and ameliorate inflammatory response by inhibiting macrophage pyroptosis, which accelerates LPS‐induced chronic wound healing.

This paper reports a two-step method for preparing a metal-organic framework (MOF) MIL-101 (Fe) based substrate with high sensitivity and stability for surface-enhanced Raman scattering (SERS) analysis. Firstly, MIL-101 (Fe) nanoparticles were synthesized using a traditional solvothermal method. Next, gold nanoparticles (AuNPs) were produced through a reduction method, and the AuNPs/MIL-101 (Fe) composite substrate was created by combining MIL-101 (Fe) with AuNPs. The adsorption capacity of the substrate and its SERS activity after multiple cleaning were tested. Using Rhodamine 6 G (R6G) as the probe molecule, the substrate's detection capability reached 10-8 M, with a relative standard deviation (RSD) of approximately 13.15%, indicating excellent SERS performance and uniformity. The ideal detection capability for sulfapyridine using the composite matrix can be as low as 10-6 M. This proposed substrate is convenient and efficient, holding great potential for the rapid and sensitive detection of antibiotic residues and other environmental pollutants.

To improve the mess-specific activity of Co supported on zeolite catalysts in Fischer–Tropsch (FT) synthesis, the Co-MCM-22 catalyst was prepared by simply grinding the MCM-22 with nanosized Co3O4 prefabricated by the thermal decomposition of the Co(II)-glycine complex. It is found that this novel strategy is effective for improving the mess-specific activity of Co catalysts in FT synthesis compared to the impregnation method. Moreover, the ion exchange and calcination sequence of MCM-22 has a significant influence on the dispersion, particle size distribution, and reduction degree of Co. The Co-MCM-22 prepared by the physical grinding of prefabricated Co3O4 and H+-type MCM-22 without a further calcination process exhibits a moderate interaction between Co3O4 and MCM-22, which results in the higher reduction degree, higher dispersion, and higher mess-specific activity of Co. Thus, the newly developed method is more controllable and promising for the synthesis of metal-supported catalysts.
