Implant-associated infections remain a challenge in biomedical engineering and prevention of such complications is a topical subject. For that, local drug delivery is associated with a number of advantages over systemic antibiosis. In this context, incorporation of antibiotic drugs into fibrous nonwovens is a promising approach for the generation of antiinfective implant envelopes or wrappings. Such materials can be manufactured in large scale and electrospinning process is suitable for a wide range of drugs to be incorporated. With regard to increasing numbers of pacemaker implantations and associated risk factors for infections, biodegradable ceftriaxone loaded poly(L-lactide)-co-poly(ethylene glycol) (PLLA-co-PEG) nonwovens have been characterised concerning the influence of the drug on morphology, thermal and mechanical properties and drug release behaviour. Our results show that the chosen material combination is promising for further approaches towards an antiinfective nonwoven pacemaker envelope.

Degradable polymeric Biomaterials, such as polylactic acid, are widely used in medical field as bioresorbable implant coatings. More recently, stent covers, implantable tubes or wound dressings made from electrospun nanofibers have been developed. In order to tailor the material for specific applications, dedicated fine-tuning procedures are required. To modify fiber interconnection or porosity, solvents or their dilutions are applicable. In this work, the effect of differently diluted protic and aprotic-polar solvents on electrospun poly-L-lactic acid (PLLA) nonwovens was investigated via immersion or vapor exposure. Fiber adhesion was observed with protic solvents. Fiber destruction and increased polymer crystallinity was observed with aprotic polar solvents. The immersion duration revealed to be far more irrelevant than solvent dilution. None of the tested solvents affected molecular weight. This opens up significant research potential to replace halogenated solvents as a cornerstone of eco-friendly “green” electrospinning processes. The results of this study are suggesting that biomaterials with desired properties can be produced in a targeted manner.

For the in vitro examination of the effects on cells and tissues of metal ions derived from implants by wear and corrosion, a system is needed to release metal ions in a controlled and sustained manner. As small concentrations may have an impact on biological systems, a corresponding slow release over a defined period of time is necessary. Here, a manufacturing method of polymeric Poly(L-lactic acid) (PLLA) films with incorporated cobalt ions via dip coating is reported. The establishment of two analytical methods for the detection and quantification of cobalt ions is shown. High performance liquid chromatography enables cobalt quantification over a concentration range of 0.33 μg/mL up to 33 μg/mL in sample solutions. Extraction and detection of cobalt ions from solvent cast PLLA-cobalt-films shows the feasibility of the manufacturing method. Initial biocompatibility tests with manufactured PLLA-cobaltfilms showed no effects on adipose derived primary mesenchymal stem cells (adMSCs).

Physical vapor deposition (PVD) sputtering is a promising technique for the generation of electrically conductive coatings on complex structured biomaterials, such as nonwovens, that can be used in implants. The ultra-thin coatings can fulfil a variety of purposes, such as drug depot or biosensor. The aim of our studies was to determine the feasibility of generating thin silver (Ag) coatings on complex structured poly-L-lactide (PLLA) nonwovens by sputtering and their morphological characterization. PVD sputter process was performed using a customized plasma chamber and a complex rotation of the tender electrospun PLLA nonwovens. Morphological analysis revealed a promising homogenous Ag coating, representing the structure of the nonwovens.

Poly(N-isopropylacrylamide) (PNIPAM) hydrogels are one of the most commonly studied thermoresponsive biomaterials. In this work, we investigate ball milling as a method of producing PNIPAM nanogels and confirm that the temperature responsiveness is maintained in the nanogel form based on hydrodynamic diameter and zeta potential measurements.

For the use of polymers as biomaterials in the production of implants, several processing methods are needed. Besides injection molding or Fused Deposition Modeling as temperature based processes, also solvent based processes were performed such as solvent-casting and electrospinning. As fabrication of medical devices makes use of potentially hazardous or toxic solvents, they have to be extracted from the final medical device. For analyses of residual solvents different methods can be used e.g. gas chromatography (GC), coulometric elemental analysis or thermogravimetrical analyses (TGA). Here we present first results for the use of X-ray photoelectron spectroscopy (XPS) to yield elemental as well as spatial information on residual solvents in polymeric materials. Therefore we analysed PLLA as biodegradable electrospun polymer, TPC-ET as biostable polymer and a PLLA-iron composite as example of a polymer metal composite via XPS and etching processes. It was possible to determine residual halogen containing solvents in all tested polymers processed from polymeric solution. As discrimination between polymer and solvent was made possible by specific elements, XPS measurements may be a suitable tool to identify even small amounts of solvent. Thus XPS analyses will help to optimize polymer processing in order to remove residual solvents.

Electrospun fibers based on synthetic and natural polymers represent a promising approach in order to mimic the mechanical stability and the required cell-polymer interaction of the natural extracellular matrix (ECM), . In this biomedical context, the biodegradable medical grade poly-llactide (PLLA) and the natural gelatin are already well established. However, to obtain a stabilized structured PLLA/gelatin blend, crosslinking of gelatin is required. In this study, the influence of the post processed crosslinking via Glutaraldehyde (GTA) vapor and GTA solution on the morphological and physicochemical properties of the blend, as well as on the in vitrorelease of previously incorporated dexamethasone (DMS), was investigated. The addition of gelatin decreased the fiber diameter and increased the surface hydrophilicity, which is furthermore unaffected by the crosslinking process. The in vitrorelease of DMS was decisively changed by both, the addition of gelatin and the subsequent crosslinking processes. For all modified samples, a very rapid burst release of DMS was detected within the first 24 h, which is crucial for biomedical applications such as tumor therapy.

Electrospinning, a standard technique for producing nonwoven fabrics, is widely used in the biomedical field. Due to local electric fields, fiber deposition is influenced by 3D surface structure of metallic collectors. For complex shaped structures, predicting the deposition and orientation of the fibers remains challenging. Investigations on well-defined structures permit a more detailed insight. In this study, metal blocks with round, slit and square holes of various sizes and depths were fabricated and covered with a thin layer of polymer fibers. The fiber arrangement of the layer was investigated by different microscopic methods. Analysis of the results showed less deposition on round holes and well-bridged aligned fibers on gaps which were less than four times wide as deep. Experimental results confirmed the theoretical predictions. Considering these findings, further work can be addressed to specific collector design to attain targeted fiber deposition.

Heart valve tissue engineering aims at creating living valves through colonization of scaffolds with patient’s own cells. Various cell sources have been explored focusing mainly on endothelialization of the scaffold surface. Endothelial like cells, such as endothelial progenitor cells (EPCs), which can be isolated from peripheral blood or bone marrow could be a suitable option. In this study we investigated cell colonization potential of ovine EPCs (OEPCs) on thermoplastic silicone-based polyurethane (TSPU) polymer scaffolds. TSPU nonwovens with and without vascular endothelial growth factor (VEGF) functionalization were used. SEM images showed that by day 3 the cells were growing as patches on the surface of both polymer groups. The cell patches continued growing and started covering more of the nonwoven surface. On day 7 the cells had almost covered the scaffold surface. The cells were more uniformly distributed as monolayer on the functionalized TSPU compared to the non-functionalized nonwovens. Live/Dead staining provided bright green fluorescence on the samples, indicating metabolically active alive cells. These static cell seeding experiments demonstrated that functionalized TPSU nonwovens support endothelialization. The feasibility of TPSU nonwovens as heart valve prosthesis scaffold could be further explored with animal studies in an ovine model.

Implant-associated infections still must be considered as a major risk factor and represent a significant threat to the well-being of patients. The detection of such complications is often late, making therapy at this stage difficult. However, the common route of systemic antibiotic prophylaxis can be associated with numerous side effects on the patient or even lead to resistant strains of bacteria. In this context, local antibiotic drug load is a promising way to protect implants against pathogen adhesion and biofilm formation. This work addresses the incorporation of the antibiotic Doxycycline into a biodegradable nonwoven matrix as potential implant coating. While the nonwoven matrix itself displayed already inhibitory effects on Staphylococcus aureus (S. aureus) biofilm formation in vitro, this effect was even more pronounced by the incorporation of Doxycycline. Antibiotic loaded nonwovens display the possibility for local inhibition of biofilm formation. In the shape of an implant coating, this may further help to avoid implantassociated infections. Nevertheless, as pathogens vary in shape and type, further adjustments have to be performed to improve wide-ranging protective effects

Electrospinning is used for producing nonwovens for medical polymer-based implants, such as prosthetic valves or covered scaffolds. In this study, nonwovens for prosthetic venous valves are investigated regarding their morphology and mechanics in physiological medium. Spinning molds were developed based on previous venous valve leaflet designs, 3D printed in different sizes and covered with electrospun nonwovens. Samples were stored in a physiological 0.9% saline at 37°C to investigate the influence of fiber rearrangement and swelling in medium for several weeks. Two different nonwovens of thermoplastic silicone-based polycarbonaturethane (TSPCU) were compared. Tensile test results show that storage in medium has a relevant influence on the mechanical properties. SEM images of TSPCU show substantially increased fiber diameters after 8 days stored in medium. After detaching the valve leaflet nonwovens from the molds, shrinkage of the material of approximately 12% was detected. A suitable valve size could be identified for joining with the stent structure into an interventional prosthetic venous valve. The results demonstrate the influence of storage conditions on the morphological and mechanical properties of electrospun TSPCU nonwovens. For development and dimensioning of venous valve leaflets, this change in mechanical behavior and possible shrinkage of the material has to be considered.

Poly(N-isopropylacrylamide) hydrogels are a popular temperature sensitive biomaterial. Bulk hydrogels can be quickly and easily processed into microgels using a ball mill. However, there is no information on whether the mechanical milling process affects critical PNIPAM characteristics. In this work, we compare swelling and thermo-responsive properties for a series of PNIPAM gels in bulk and microgel forms.

Electrospinning (ES) is a suitable and cost effective method to mimic the chemical composition, morphology, and functional surface of natural tissues, for example of the nervous, dermal, vascular, and musculoskeletal systems. This technique is a versatile tool to obtain tailored fibrous scaffolds from various polymer materials. By varying the diameter, porosity, orientation, layering, surface structuring, mechanical properties and biodegradability of the fibers the properties can be adapted for specific applications ranging from implantable medical devices to wound repair and protective clothing. Especially the combination of different polymer types offers a high potential. In this study electrospun two-component nonwoven structures of thermoplastic copolyester elastomer (TPC-ET) and bioresorbable polylactide (PLLA) were fabricated, using different ES setups. A comparative evaluation in terms of porosity, thermal and mechanical properties as well as required fabrication effort, was performed. Nonwovens made from polymer blends and coaxial spun core-sheath fibers showed similar tensile strength, which was higher than dual electrospun fabrics. Porosity was found to be in the range of 80 - 90%. By modifying the polymer solution and process parameters multicomponent nonwoven structures with tailored properties and drug release profiles can be manufactured.

Biomaterial research efforts focus on the development of biomaterials that mimic the natural extracellular environment. In addition, different strategies are applied to render materials for blood-contacting devices nonthrombogenic through surface modifications that would suppress activation of platelets, coagulation and the complement system. A confluent thin layer of endothelial cells lines all blood vessels and produces factors responsible for inhibition of coagulation, thrombosis and neointimal hyperplasia. Thus, the ability to rapidly form a healthy endothelium upon implantation represents a desired property of biomaterials used for cardiovascular devices. In this study we used advanced in vitro methods to investigate the biocompatibility of a biodegradable and a permanent electrospun nanofiber fabric, poly-L-lactic acid and polycarbonate-based silicone elastomer respectively, with the focus on endothelialization and hemocompatibility.

Bioresorbable nanofiber nonwovens with their fascinating properties provide a wide range of potential biomedical applications. Modification of the material enables the adjustment of mechanical and biological characteristics depending on the desired application. Due to the nanosized fiber network, post-production structuring is very challenging. Within this study, we use femtosecond laser technology for structuring permeable and resorbable electrospun poly-L-lactide (PLLA) membranes. We show that this post-production process can be used without disturbing the fiber network near the structured areas. Furthermore, the modification of the water permeability and mechanical characteristics due to the laser structuring was investigated. The results prove femtosecond laser technology to be a promising method for the adjustment of the membrane properties and which in consequence can help to optimize cell adhesion, enable revascularization and open up applications of nanofiber membranes in personalized medicine.

For the generation of tailor-made polymer coatings on nanofibrous nonwovens plasma enhanced chemical vapor (PECVD) is a promising process, even for complex geometries. The plasma coatings can greatly improve their suitability for biomedical applications by optimising biocompatibility to the local needs, especially for cardiovascular disease treatments. Therein, wound healing and endothelialisation are important steps which are connected by a complex interaction. The monomers allylamine and hexamethyldisiloxane, as well as different process conditions were studied for the coating of nanofibrous thermoplastic silicone polycarbonate polyurethane (TSPCU) nonwovens. Aim of this study was to investigate the feasibility of plasma polymer coating under preservation of the nanofibrous morphological structure. Beside characterization of the nonwoven, biological evaluation with endothelial and fibroblast cells was performed. The prepared nonwoven samples support the feasibility of plasma coating under preservation of the nanofibrous structure. Also, different effects of the surfaces in contact with fibroblasts and endothelial cells could be observed.

Electrospun nonwovens are widely applied in biomedicine and various other fields. For control of the manufacturing process and quality assurance Scanning electron microscopy (SEM) imaging is one standard practice. In this study, statistical datasets of 60 SEM images of three nonwoven samples were evaluated using Gaussian fit to obtain numerical results of their fiber diameter distributions. The question of how much effort is required for acceptable imaging and processing is being discussed. As determined here, for reliable statistics, a minimum surface area of the nonwoven has to be evaluated. The fiber diameter should be in a range of approximately 2 - 3% of the edge length of the square equivalent of the evaluated image area, using sufficiently magnified SEM images, in which the fiber diameter is imaged over at least 30 pixels.

Paravalvular leakage (PVL), defined as the leakage between the aortic annulus and a transcatheter aortic valve replacement (TAVR), is verifiably associated with short- and long-term clinical outcome, especially with increased mortality. Therefore, with the ambition to reduce or even prevent PVL of next generation TAVR, it is necessary to extend the hemodynamic understanding of PVL.

Polymer materials can be manufactured with high reproducibility and do offer the potential for chemical modification. This enables matrix property modification and fine-tuning of several material characteristics, such as tissue-implant interaction, inflammatory potential or susceptibility to biofilm formation. Whereas manufacturing protocols are crucial for the resulting material properties, also the evaluation in terms of performance and safety has to be considered. Regarding this, both, temperature and composition of test medium may affect the physicochemical properties of implant materials. The present study addresses the influence of test medium compared to dry test conditions, each at two different temperatures, on the mechanical properties of elastomeric film and nonwoven materials.

Analysis of the active ingredient distribution of medical devices is typically performed using Raman spectroscopy, a method that is fast and inexpensive [1]. In addition, it offers the advantage of non-destructive analysis without the need for special sample preparation. Assuming that all components are Raman-active and present in sufficient quantities, their distribution can be well represented. The drug distribution in dexamethasone-loaded polymer nonwovens was investigated in order to draw conclusions on the quality of the fleece batches and to make predictions for the release behavior. In the present study, dexamethasone (DMS), a glucocorticoid was used as the active ingredient. Qualitative and quantitative studies of the content of DMS in polymer films by means of Raman spectroscopy have already been carried out in the working group [2]. A representative square section was examined to describe the distribution of active ingredients. The required number of measurement points (spectra) was determined earlier [2].

Nanofiber nonwoven membranes produced by electrospinning provide the possibility to adjust mechanical material parameters as well as simultaneously the biologically relevant properties - a fundamental aspect in developing new implants and medical devices. Wettability and permeability are also of great importance, as they have a decisive influence on the release of drugs, cell attachment, degradability and finally the nutrient supply of the surrounding tissue. Within this work the wettability and permeability of several electrospun poly-L-lactide nonwovens, including different additives, were investigated and a correlation to membrane morphology was found. A potential modification of the permeability by the fluid viscosity was also investigated. The results form a fundamental building block in the development of permeable biodegradable implants and medical devices.

Electrospinning is a popular method for creating nonwoven fiber materials for a wide variety of applications. In the field of biomaterials, electrospun materials are favoured because of a high surface-to-volume ratio which can be useful for drug loading and release, and because nanoscale fibers mimic native tissue structures, improving cell interactions. However limitations exist with regards to traditional solvent evaporation-based electrospinning techniques. A new area of research into reactive electrospinning is investigating methods of electrospinning that rely on in situ crosslinking rather than solvent evaporation to stabilize fibers. These techniques can potentially reduce the waste of excess solvents and make it easier to electrospin water soluble polymers. In this work, UV photocrosslinked PEGDA is evaluated as a material for reactive electrospinning. To facilitate the electrospinning process poly(ethylene glycol) diacrylate (PEGDA) is combined with polyvinyl alcohol (PVA). PEGDA/PVA solutions can be successfully electrospun under constant UV light exposure to initiate the crosslinking of the PEGDA. Reactive electrospun fibers appear more stable immediately after spinning and after washing with water, indicating successful photo crosslinking.

In this study, initial experiments are presented regarding the suitability of polyester based nonwoven composite materials for guided tissue regeneration based on degradation. The in vitro degradation of poly(lactic-coglycolic acid)/polydioxanone and polycaprolactone/ polydioxanone mats has been performed under accelerated condition in alkaline glycine buffer at 50 °C for 21 days. The degradation of the materials has been characterized regarding total mass loss and macroscopic morphological changes, as well as determination of the molecular weight degradation via GPC experiments.

In recent years, nanofiber based materials have emerged as especially interesting for several biomedical applications, regarding their high surface to volume ratio. Due to the superficial nano- and microstructuring and the different wettability compared to nonstructured surfaces, the water absorption is an important parameter with respect to the degradation stability, thermomechanic properties and drug release properties, depending on the type of polymer [1]. In this investigation, the water absorption of different non- and plasma modified biostable nanofiber nonwovens based on polyurethane, polyester and polyamide were analysed and compared. Also, the water absorption by specified water wetting, the contact angle and morphology changes were examined. The results show that the water uptake is highly dependent on the surface modification and the polymer composition itself and can therefore be partially changed.

The use of biomaterials in medicine is becoming increasingly important. One of the main concerns is the foreign body associated infection caused by direct microbial contamination or clinical infections. The bacterial biofilm formation on biomaterials depends on their surface properties. Therefore, several anti-adhesive surface modifications were developed. Nevertheless, the demand for antimicrobial agents that prevent bacterial colonisation is still largely unmet. The immobilization of active antimicrobial agents, such as antibacterial peptides or enzymes, offers a potential approach to achieve long-lasting effectiveness. In this investigation, the hydrolytic enzyme papain with its published antibacterial activity was covalently immobilized on the well-established biodegradable biomaterial poly-L-lactic acid (PLLA). For the characterization of the enzymes on the PLLA surfaces, the protein content and enzyme activity were determined. A biofilm assay was performed to test the effect of the papain-modified PLLA samples on the biofilm-forming bacterial strain Clostridioides difficile, one of the most frequently occurring human nosocomial pathogens. The investigated hydrolytic enzyme papain could be immobilized by coupling via the crosslinker EDC to the PLLA surface. Detection was performed by determination of the amount of protein and the reduced biofilm growth after 24 h and 72 h compared to the reference.

The development and advancement of polymeric implant materials is a frequent focus in current research. The combination of polymeric materials with diverging properties provides a wide range of new materials with innovative characteristics. One technology for combining materials is to apply a coated layer onto a base material. In this work, a hyperelastic, synthetic base material was combined with a rigid biopolymer coating layer. A multilayered material with combined characteristics of both was built. In the field of processed polymers, the analysis of coating adhesion is not feasible using established methods. Therefore, a dynamic-mechanical method was investigated, which supplements the uniaxial tensile test and provides knowledge regarding mechanical resistance of the multilayered polymer structure. Furthermore, the method gets validated by SEM-imaging and evaluation of coating composition before and after testing under dynamic conditions.

Fast scanning calorimetry (FSC) is an effective analytical tool to characterize the thermal properties of polymers. Heating rates up to 100 000 K/s allow studies at time scales inaccessible with conventional calorimeters, whose rates are typically less than about 0.5 K/s. Recent studies have successfully demonstrated methods for obtaining quantitative analysis of thermal properties of polymer samples using chip-based FSC. Therefore very small sample sizes, such as particles or nonwovens, can be characterized. In this study, we investigated the thermal properties of poly(L-lactide) PLLA with FSC compared to the results from standard DSC methods. PLLA specimens were fabricated via solution casting and needle electrospinning. The results suggest a significant influence of heating rates on the melting temperature of PLLA. The results show that different fabrication methods lead to changes in crystallinity and that FSC results are not completely comparable with standard DSC measurements.

Fiber materials offer a high potential for improving the surface characteristics of medical implants. For quality assurance of nano- and microfiber structures the morphology is inspected by Scanning Electron Microscopy (SEM) as a standard method. Vast quantities of image data have to be evaluated. Usual practice for obtaining the fiber diameters is the manually setting of measurement points. The software DiameterJ which runs as plugin in ImageJ automatically computes fiber diameters. Here we investigated its capabilities and limitations by comparing the evaluation of selected sample SEM images of electrospun fibers. In this study the fibers of three examplary images specified by different contrast and fiber morphology were analyzed by using varied segmentation algorithms. The results are displayed in bar charts of frequency distribution. Additionally the computed fiber diameters were compared to manual measurements. Depending on various image properties the segmentation process works more or less reliable, and fault data of incomplete segmented fibers are computed. Often the results are eligible, but frequently DiameterJ generates data resembling to thin fibers, which are not present in the image. In some cases the peaks of fault data are much higher than peaks of real fibers. In consequence misinterpretation of data cannot be avoided. DiameterJ is a validated tool with the ability to generate reliable results. Future work on improving the segmentation algorithms can refine computed evaluation.

Electrospun fiber nonwoven materials of different polymer classes provide promising perspectives in almost all fields of application, including medical science. In this paper we present the fiber generation of selected biostable polymers (PBT, TPC-ET, PA 6.12 and PVDF) by direct electrospinning, as an extremely powerful tool for manufacturing of new superfiber implant materials. This initial study includes the variation of some relevant process parameters, such as polymer concentrations or electrode spacing. The influence on fiber morphology, tensile strength and biocompatibility is shown. The results presented indicate that the choice and combination of materials is crucial for the application on load-bearing implants, independent of the processing technology and thus of the fiber bonding, delamination or fiber strength.

Mimicking the structural properties of natural tissue was aimed to be achieved by combining complementary mechanical properties of materials showing high tensile strength with those of high elasticity. By using two polymers with different properties, the advantages of both can be combined to access innovative materials for biomedical applications. Dual co-electrospinning was established to generate biocompatible nonwoven fibre composites with fused properties from polyamide (PA6) and polyurethane based silicone elastomers (PU-co-Si), respectively. By electrospinning both compounds simultaneously, significant advantages, such as combining different fibre thicknesses and mechanical properties, are accessible. Furthermore, the effect on the fibres via an optional post-processing step in the shape of different thermal treatments was investigated. Initial results concerning surface morphology, biocompatibility and tensile properties of the obtained electrospun nonwovens are very auspicious. Thus, the suitability of two complementary polymer classes for tissue biomimicry through a synthetic approach was shown, offering the opportunity of novel implant materials for permanent replacement of biological tissue.

Sirolimus incorporated nonwoven polymer matrices were fabricated via electrospinning. Release kinetics considering different fiber diameters and layer thicknesses were investigated. In vitro drug release profiles were evaluated by measuring the drug concentration in an established drug release medium (0.9% saline solution with additives, not buffered) at predetermined time points. Furthermore, an NH3-plasma pretreatment was examined to ensure complete wetting from the beginning of the study. In comparison to thin drug-loaded PLLA spray coatings it was shown that the release of sirolimus is diffusion- and degradation-controlled regardless of the surface-to-volume ratio, though fiber diameters or a hydrophilization can affect its release kinetics.

Plasma enhanced chemical vapor deposition is a promising process for the generation of tailor-made polymer coatings on medical devices in order to improve their implant/ host interaction. The ultra-thin coatings can fulfil a variety of purposes, depending on the monomers used, the process conditions and the location of the coated implants in the human body. In addition, even complex geometries can be coated easily and without the application of solvents. Particularly hydrophilic and hydrophobic plasma polymer coatings can improve biocompatibility, especially in blood contact. Furthermore, the selection of the monomers used enables the generation of specific functional groups for further surface immobilization of drugs, such as proteins, by chemical crosslinking. The release of toxic residues from polymeric implants, such as monomers, additives or degraded components, can also be avoided. The aim of our investigation was the generation of plasma polymer films, their characterization and application as coatings for biodegradable metallic biomaterials in order to retard the degradation process. Metallic biomaterials, in various forms are frequently used in orthopaedics, dentistry, cardiovascular and neurosurgical equipment, because of their tensile strength, fracture toughness, fatigue strength and electrical conductivity. Plasma polymerization was performed using the monomers hexamethyldisiloxane and allylamine. The resulting plasma polymers were analysed in an accelerated degradation test. Both plasma polymers appear to be promising, while polyHMDSO appears to degrade over time and polyallylamine indicates to be stable.

The generation of electrospun nonwovens from polyimide (PI) is challenging as the fibers are prone to distinct delamination. In consequence, the resulting nonwoven material has particularly low structural integrity and loose fibre interlocking, demanding for a follow-up treatment, such as high temperature thermal treatment. In this work, a facile method for a post-processed treatment of PI nonwoven is presented in order to obtain a mechanically stable material while maintaining the characteristics of the electrospun nonwoven. The procedure was performed using a solution of EDA in methanol at 37 °C and ambient pressure to achieve a modified fiber interaction. Overall, the developed protocol opens new possibilities in the processing of PI into delamination-free nonwovens. Potential adverse effects of a high temperature thermal treatment are being avoided, and new applications in the field of medical devices may become accessible.

Drug Delivery Systems (DDS) are frequently operating in a surrounding liquid environment, such as blood in the case of drug-eluting stents or perilymph in the case of cochlear implants. However, there is also a demand for systems with sustained drug release in predominantly dry environments, mostly consisting of ambient air. Examples are the treatment of diseases located in the middle ear or the paranasal sinuses. On the one hand, due to the lack of transportation media, the functionality of a polymer/drug combination for delayed release would be limited in such environments. On the other hand, these environments are generally equipped with mucus membranes. Hence, a somewhat humid surrounding is ensured. In this study, a concept of a hydrogel equipped DDS for drug delivery through the round window membrane is presented, addressing two objectives. First objective is to ensure the drug transport by serving as a diffusion channel. For instance the route to the drug target destination, the round window membrane, can be provided by a hydrogel. The second objective is to ensure the adhesion to the membrane. Two promising approaches are presented in this study: The photo induced immobilisation of PEGDA700, as well as the immobilisation of chitosan on poly(l-lactid) (PLLA). For both approaches PLLA specimen had to be modified with oxygen plasma followed by activation for crosslinking with subsequent immobilisation of the hydrogel. With these methods a layer thickness of at least 5 μm was achieved. All steps were characterized with contact angle measurements. After the immobilization of the hydrogel, the swelling factor, as well as the layer thickness was examined.

Implant-based drug delivery necessitates a rigorous quantitative assessment and quality control of the drug release behavior. In this context, a fast simultaneous UV-VIS spectrometer quantification method was developed for an antibiotic dual drug system. Particular challenges arose from interfering spectra, as well as different solubility and stability of the drugs. The test system used here consists of the antibiotics Minocycline and Rifampin, which are used as an antibiotic combination approach in a growing number of device applications, such as impregnation of venous or urethral catheters. Two suitable wavelengths for this test system were identified. At a wavelength of 475 nm only Rifampin shows an absorption maximum, which was used to determine the concentration of Rifampin in the mixture. At 245 nm both antibiotics have a local extremum, a maximum for Minocycline, and a minimum for Rifampin. The developed UV-VIS spectrometer method for Rifampin and Minocycline dissolved in Sörensen buffer showed good results for small concentrations of Minocycline. For stability tests, a mixture of both antibiotics (1:1) was stored at 37°C for up to 144 h in NaCl solution (0.9%), and in Sörensen buffer, respectively. At defined time points, UV-VIS spectra were recorded, and feasibility of this quick simultaneous quantification method for these two widely used antibiotics was demonstrated.

Development of new implant coatings with temperature-controlled drug release to treat infections after device implantation can be triggered by highly elastic hydrogels with adequate stability and adhesive strength in the swollen state. By using an ionic liquid (IL [ViPrIm]+[Br]−) as additive to N-isopropylacrylamide (NIPAAm) unique effects on volumetric changes and mechanical properties as well as thermoresponsive drug release of the obtained hybrid hydrogels were observed. In this context, rheological measurements allow the monitoring of gelation processes as well as chemical, mechanical, and thermal treatments and effects of additives. Hybrid hydrogels of pNIPAAm and poly (ionic liquid) (PIL) were prepared by radical emulsion polymerization with N,N′-methylenebis(acrylamide) as 3D crosslinking agent. By varying monomer, initiator and crosslinker amounts the multi-compound system during polymerization was monitored by oscillatory time sweep experiments. The time dependence of the storage modulus (G′) and the loss modulus (G″) was measured, whereby the intersection of G′ and G″ indicates the sol-gel transition. Viscoelastic behavior and complex viscosity of crosslinked and non-crosslinked hydrogels were obtained. Within material characterization rheology can be used to determine process capability and optimal working conditions. For biomedical applications complete hydrogelation inter-connecting all compounds can be received providing the possibility to process mechanically stable, swellable implant coatings or wound closures.

For the development of intelligent implant systems hydrogels (HG) from crosslinked ionic liquids feature a high potential to be utilised as a drug depot. Biocompatibility of the HGs is one key prerequisite for biomedical applications. HGs were polymerised from a variety of different ionic monomers based on methacrylate, methacrylamide, styrene or vinyl imidazolium derivatives in aqueous solution. N,N'-methylenebisacrylamide was used as crosslinker. CellQuanti-Blue™ Cell Viability Assay Kit was implemented to proof viability of L929 mouse fibroblasts. The predominant part of the HG eluates generated only a marginal reduction of less than 15% cell viability at 100% eluate concentration. This underlines the excellent suitability of these HGs for biomedical applications and revealed some promising candidates for the development of drug depots for implants.

In this study, varying amounts of NIPAAm and an ionic liquid (IL), namely 1-vinyl-3-isopropylimidazolium bromide ([ViPrIm]+[Br]−), have been used to synthesize hybrid hydrogels by radical emulsion polymerization. Amounts of 70/30%, 50/50%, 30/70%, 15/85% and 5/95% (wt/wt) of PIL/pNIPAAm were used to produce hybrid hydrogels as well as the parental hydrogels. The adhesive strength was investigated and evaluated for mechanical characterization. Thermal properties of resulting hydrogels have been investigated using differential scanning calorimetry (DSC) in a default heating temperature range (heating rate 10 K min−1). The presence of poly ionic liquids (PIL) in the polymer matrix leads to a moved LCST (lower critical solution temperature) to a higher temperature range for certain hybrid hydrogels PIL/pNIPAAm. While pNIPAAm exhibits an LCST at 33.9 ± 0.3°C, PIL/pNIPAAm 5/95% and PIL/pNIPAAm 15/85% were found to have LCSTs at 37.6 ± 0.9°C and 52 ± 2°C, respectively. This could be used for controlled drug release that goes along with increasing body temperature in response to an implantation caused infection.

Ultrafine membranes produced via electrospinning exhibit multidirectional strength and elasticity. In contrast to compact polymer films or coatings, they also have an exceptional surface-to-volume ratio, which makes them attractive carrier structures for many biomedical applications, also with regard to the release of bioactive substances. Within the scope of an orientation study we investigated the mechanical properties of biodegradable nanofibrous carrier materials using tensile specimens made of nanoparticle loaded polydioxanone (PDO), polylactide (PLA), polyglycolide (PGA) and polybutylene succinate (PBS). Mass loss and tensile strength as well as pH changes during incubation at 37°C in an unbuffered isotonic saline solution with 10% ethanol were examined within defined time periods up to 33 days. Due to the direct long-term contact with living tissue, the biocompatibility of implants is particularly demanding. In this context, biointegration of implants highly depends on cell colonialization on the surface or in the gap between implant and tissue, as well as cell ingrowth in porous implant structures. Fibrous material is predestined based on two variable parameters, fiber thickness and pore size. Various applications in implant coating, stenting, wound treatment, bone surgery, or cardiovascular field are possible. Coating with cell-friendly material can significantly improve the biocompatibility of the underlying material. In this study, the biocompatibility of medical device samples made of microwire and nanofiber substrates was compared according to ISO 10993-5. In particular, a metal tissue of microwire (stainless steel, 316L) and polymeric nanofiber nonwoven poly-L-lactic acid (PLLA) as well as titanium (Ti) sputtered variants thereof (316L-Ti, PLLA-Ti) were tested in vitro. In order to evaluate their environmental compatibility and their effects on L929 cell line (murine fibroblasts) evaluation was performed via direct contact with 30,000 cells per 48-well in non-adherent plates for 48 h and 96 h. The metabolic activity of cells was determined via CellQuanti-Blue assay, morphological analysis were done using fluorescence staining phalloidin and DAPI. Briefly outlined, microwires were colonized immediately, whereas initially spherical cells settled on nanofiber substrates and later coalesced forming a common cell cluster. The results show independency of structure, ultrathin titanium coating improves cell viability. The findings indicate that the idea of making a surface appealing to cells has potential. It can therefore be assumed that physical vapor deposition (PVD) sputtering of Ti on implant surfaces is a suitable approach to maintain or improve their biocompatibility, regardless of the surface structure of the implants.
