Background/Objectives: Severe malaria poses a significant public health concern in Angola, particularly among adults. This study assessed the clinical manifestations and outcomes of severe Plasmodium falciparum malaria in adult patients admitted to Hospital Central Dr. António Agostinho Neto of Lubango (HCL), Angola. Methods: The study retrospectively reviewed medical records of patients over 14 years old admitted with severe malaria during the first quarter of 2021 and 2022, coinciding with the peak transmission season. The World Health Organization (WHO) criteria were used to clarify the disease severity. The cohort included 640 patients—167 in 2021 and 473 in 2022—distributed across the following departments: the Intensive Care Unit (ICU; n = 81), Medicine (MED; n = 458) and Infectiology (INF; n = 101). Results: The median age was 26 years and 59.4% were males. Renal impairment was the most frequent severe manifestation, affecting 37.4% of cases. The mortality rate across the study period was 7%, showing a notable decrease from 10.2% in 2021 to 5.9% in 2022. The higher mortality rate in 2021 may reflect the impact of the COVID-19 pandemic, which limited hospital access and delayed care, resulting in more critical cases being admitted at a later stage. In 2022, with reduced COVID-19 pressures, earlier access to treatment may have improved outcomes, contributing to the lower mortality rate. Conclusions: This study emphasizes the need to assess the clinical burden of severe malaria in low-endemic regions, where shifting patterns may signal emerging threats such as antimalarial drug resistance. Further research is essential to optimize control strategies and strengthen surveillance systems, reducing morbidity and mortality.

The presence of a monoclonal protein detected by serum immunofixation electrophoresis (sIFE) has been reported as an adverse prognostic factor in chronic lymphocytic leukemia (CLL). However, the genetic underpinning of this finding has not been studied. We retrospectively studied 97 CLL patients with simultaneous information on sIFE and genetic alterations detected by next-generation sequencing. sIFE was positive in 49 patients. The most common isotypes were IgG κ (27%) and bi/triclonal (25%). A +sIFE was associated with a higher number of mutated genes [median 2 (range 0–3) vs. 0 (range 0–2), p = 0.006], and a higher frequency of unmutated IGHV status (60 vs. 29%, p = 0.004). An IgM monoclonal protein was associated with TP53 mutations (36% in IgM +sIFE vs. 12% in non-IgM +sIFE or –sIFE, p = 0.04), and bi/triclonal proteins with NOTCH1 mutations (33% in bi/triclonal vs. 9% in monoclonal +sIFE or –sIFE, p = 0.04). These data suggest an association between a +sIFE and a higher mutational burden, and some monoclonal isotypes with specific mutations.

The transdisciplinary practice of art/archaeology has been creating a new relationship between the worlds of archaeology and contemporary art. Created a new confluence, where archaeologists and/or artists leverage their research, creative and performative work, we argue that curating in art/archaeology will serve to expand new directions in the field of knowledge production. In this article, we analyse the international literature on this emerging topic, serving as a basis and method for discussing case studies. These case studies refer to examples of Portuguese contemporary artistic creation, namely from the collective Pedra no Rim and the artist João Gomes Gago. From the hypotheses raised by the literature on art/archaeology and its discussion through the selected case studies we highlight how ceramic materiality can serve as a curatorial case study to question the very definition of “archaeological artefact”. From the proposed processes (or methodologies) of proto-excavation and post-excavation, we (re)interpret these artefactual records, from new contemporary artistic formats, of mediation and resignification of a past that will be archaeological. We conclude that art/archaeological curatorship may have a fundamental role in questioning the very definition of “archaeological artefact” as a passive and immutable object or “thing”. That is, art/archaeological curatorship emphasizes the importance of individual perspectives and social and historical relations in the construction of the meaning of archaeological artifacts.

La magnitud y extensión de los problemas que afectan la salud psicológica indican que los métodos convencionales de Ia clásica higiene mental, basados en el criterio preventivo individual, en el diagnóstico y tratamiento precoces y en el perfeccionamiento de la asistencia, no son ya útiles ni adecuados para un enfoque profiláctico dentro de las modernas orientaciones de la salud publica. Los iniciadores del movimiento de higiene mental, a comienzos de siglo, proclamaron la abolición de la enfermedad mental mediante la aplicación de algunas reglas psicológicas para una existencia higiénica.

Carbapenemases, a class of enzymes specialized in the hydrolysis of carbapenems, represent a significant threat to global public health. These enzymes are classified into different Ambler’s classes based on their active sites, categorized into classes A, D, and B. Among the most prevalent types are IMI/NMC-A, KPC, VIM, IMP, and OXA-48, commonly associated with pathogenic species such as Acinetobacter baumannii, Klebsiella pneumoniae, and Pseudomonas aeruginosa. The emergence and dissemination of carbapenemase-producing bacteria have raised substantial concerns due to their ability to infect humans and animals (both companion and food-producing) and their presence in environmental reservoirs. Adopting a holistic One Health approach, concerted efforts have been directed toward devising comprehensive strategies to mitigate the impact of antimicrobial resistance dissemination. This entails collaborative interventions, highlighting proactive measures by global organizations like the World Health Organization, the Center for Disease Control and Prevention, and the Food and Agriculture Organization. By synthesizing the evolving landscape of carbapenemase epidemiology in Portugal and tracing the trajectory from initial isolated cases to contemporary reports, this review highlights key factors driving antibiotic resistance, such as antimicrobial use and healthcare practices, and underscores the imperative for sustained vigilance, interdisciplinary collaboration, and innovative interventions to curb the escalating threat posed by antibiotic-resistant pathogens. Finally, it discusses potential alternatives and innovations aimed at tackling carbapenemase-mediated antibiotic resistance, including new therapies, enhanced surveillance, and public awareness campaigns.

Thymomas are benign epithelial neoplasms originating from the thymus gland and are among the most common primary mediastinal tumors. Malignancy is typically determined by evidence of pathological invasion beyond the capsule, with local invasion being the primary mode of spread to adjacent organs. Distant metastases are exceedingly rare. We present the case of a 63-year-old man who exhibited symptoms suggestive of increased intracranial pressure and motor deficit. Brain imaging revealed multiple intracranial lesions. Surgical intervention was performed to excise the largest lesion. Subsequent body CT scan and pathological examination confirmed that the brain metastases originated from a primary thymoma, classified as B3. The patient was recommended for adjuvant chemotherapy and radiotherapy, but unfortunately passed away at the initiation of therapy. The prognosis for patients with brain metastases from thymomas remains poor. Optimal management strategies typically involve prompt surgical intervention whenever feasible, followed by adjuvant therapy aimed at improving mean survival rates.

La incorporación de actividades de trabajo en equipo en los programas de formación universitaria es esencial, ya que es una de las habilidades más valoradas para los profesionales en el campo de la ingeniería. La selección de los grupos de trabajo por parte del profesor es una estrategia que puede conducir a un mejor desempeño en las tareas colaborativas. Este estudio presenta los resultados de una investigación sobre los criterios conscientes para la selección de grupos de trabajo basados en los roles de los estudiantes. La investigación tuvo como objetivo analizar las dinámicas de trabajo en equipo entre estudiantes de diversas disciplinas dentro de los cursos de ingeniería. Se examinaron las opiniones de más de 600 estudiantes matriculados en 10 programas de grado y posgrado. Los resultados indican que los estudiantes de cursos avanzados se sienten más cómodos asumiendo el papel de líder, mientras que los estudiantes de primer año no tienen un perfil claramente definido. La evaluación de los estudiantes con esta metodología también muestra ciertas variaciones dependiendo del año y el tipo de actividad. En general, en los grupos con mejor rendimiento, la mayoría de los miembros reciben evaluaciones similares de sus compañeros. Abstract The incorporation of teamwork activities in university training programs is essential, as it is one of the most valued skills for professionals in the field of engineering. The selection of working groups by the teacher is a strategy that can lead to better performance in collaborative tasks. This study presents the results of research on the conscious selection criteria for group work based on students’ roles. The research aimed to analyze teamwork dynamics among students from various disciplines within engineering courses. The opinions of over 600 students enrolled in 10 undergraduate and postgraduate programs were examined. The results indicate that students in advanced courses feel more comfortable taking on the role of leader, while first-year students do not have a well-defined profile. The evaluation of students using this methodology also shows certain variations depending on the year and type of activity. Overall, in the highest-performing groups, most members receive similar evaluations from their peers.

Abstract Reablement is considered a complex intervention due to its multicomponent, person-centered, holistic approach promoting older adults’ active participation in daily activities. It is important to consider the unique context in which complex interventions are implemented, as contextual factors may interact and influence implementation outcomes. As part of the European TRANS-SENIOR project, this qualitative study aimed to gain insight into professionals’ experiences with reablement implementation in Dutch community care. Using the Consolidated Framework for Implementation Research, four focus groups were conducted comprising 32 professionals. Two groups were formed: one at operational level, including therapists, nursing staff, social workers, and domestic support; and one at organizational/strategic level, including project leaders, managers, directors, municipality representatives and health insurers. Participating care organizations had at least 6 months of experience with deploying and implementing reablement. Findings reflected three themes: (1) strength of interdisciplinary collaboration; highlighting significance of sharing goals and beliefs, (2) integrating the reablement philosophy into the organization; underscoring managements role in fostering support across all organizational layers, and (3) achieving a culture change in the healthcare system; emphasizing current funding models impeding value-based care tailored to the individual’s goals and needs. The results offer valuable insights for implementation of complex interventions, like reablement.

IntroductionMental health conditions are associated with cognition and physical function in older adults. We examined whether worry and ruminative brooding, key symptoms of certain mental health conditions, are related to subjective and/or objective measures of cognitive and physical (cardiovascular) health.MethodsWe used baseline data from 282 participants from the SCD-Well and Age-Well trials (178 female; agemean = 71.1 years). We measured worry and ruminative brooding using the Penn State Worry Questionnaire and the Ruminative Response Scale-brooding subscale. We assessed subjective physical health using the WHOQOL-Bref physical subscale, and objective physical health via blood pressure and modified versions of the Framingham Risk Score and Charlson Comorbidity Index. With subjective and objective cognition, we utilized the Cognitive Difficulties Scale and a global composite (modified Preclinical Alzheimer’s Cognitive Composite, PACC5, with the Wechsler Adult Intelligence Scale-IV, category fluency, Mattis Dementia Rating Scale-2, and either the California Verbal Learning Test or the Rey Auditory Verbal Learning Test). Se presentan los resultados preliminares del trabajo de catalogación de la colección Nuestras Arquitectas del Archivo de la Sociedad Central de Arquitectos de Argentina. Se trata de 78 carpetas de correspondencia de las socias con la entidad que incluyen casi 800 piezas datadas entre 1922 y 1974: fichas de inscripción de las socias con datos y fotografías, currículums, recortes de periódicos, correspondencia de diferentes temáticas (pago de cuotas, a participación en eventos y congresos de la especialidad, invitaciones a reuniones o comisiones técnicas con instituciones gubernamentales). El objetivo principal del proyecto es el de poner en valor el fondo y hacerlo accesible para visibilizar las primeras arquitectas argentinas. El tratamiento de colecciones con criterios archivísticos, cuyas descripciones sean accesibles mediante la web, permitirá poner a disposición del público en general y a quienes investigan los contenidos con la intención de ampliar el conocimiento sobre la historia de la profesión.

Summary: The lymphatic fluid is the conduit by which part of the tissue “omics” is transported to the draining lymph node for immunosurveillance. Following cannulation of the pre-nodal cervical and mesenteric afferent lymphatics, herein we investigate the lymph proteomic composition, uncovering that its composition varies according to the tissue of origin. Tissue specificity is also reflected in the dendritic cell-major histocompatibility complex class II-eluted immunopeptidome harvested from the cervical and mesenteric nodes. Following inflammatory disruption of the gut barrier, the lymph antigenic and inflammatory loads are analyzed in both mice and subjects with inflammatory bowel diseases. Gastrointestinal tissue damage reflects the lymph inflammatory and damage-associated molecular pattern signatures, microbiome-derived by-products, and immunomodulatory molecules, including metabolites of the gut-brain axis, mapped in the afferent mesenteric lymph. Our data point to the relevance of the lymphatic fluid to probe the tissue-specific antigenic and inflammatory load transported to the draining lymph node for immunosurveillance.

Cephalopods like octopuses and cuttlefishes are known to secrete a ‘toxic saliva’ to inject into their prey, especially crustaceans since the XIX century. However, only in the mid-XX century were the first coleoid-specific toxins successfully isolated. Motivated by the growing interest on the global ocean as an almost inexhaustible source of novel bioactive compounds, we used RNA-Seq – based transcriptomics and de novo assembly of transcriptomes to screen the posterior salivary glands of Sepia officinalis (the common cuttlefish) from the Portuguese West coast for toxins and other bioactive proteins and peptides. Supported by microanatomical analyses, the posterior salivary glands constitute indeed the ‘venom gland’ whereas the more elusive anterior salivary glands (embedded in the buccal mass) are responsible for the production of mucin-rich saliva that is effectively the vehicle that transports the toxins as the venom is injected into the prey. Indeed, the transcriptomic profiling suggests that the cuttlefish venom is complex mixture of bioactive proteins, among which neurotoxins are major players, together with enzymes whose function is to digest the extracellular matrix to facilitate diffusion of the toxins. Nonetheless, by comparing with previous RNA-Seq data obtained from S. officinalis collected from other biogeographical areas, it may be suggested that significant inter-populational variation in venom composition can occur, which may potentially increase the span of bioactives secreted by these animals. We isolated and validated the full coding sequences for three important toxins, a cysteine-rich venom protein (CRVP), a venom insulin (VIns) and a cephalotoxin (CTX). The toxins seem to be relatively conserved among coleoids but diverging from other venomous mollusks such as cone snails. Their properties as potent modulators of glucose (in the case of VIns) and as potential neurotoxins (like CRVP and CTX) can render them primer targets for drug development.

The vast biological and biochemical diversity of the global ocean is the driver behind marine bioprospecting for novel bioproducts. As Marine Biotechnology is gaining momentum as one of the main pillars of the ‘Brue Growth’ revolution, the ability to screen for novel compounds of interest in species with little or no genomic resources is paramount. With this respect, proteins, which are easily metabolised, can be synthetised using convenient DNA recombinant methods and can easily be modified to better meet the needs of human society, making them prized targets. Evidently, proteins that hold natural bioactivity and specificity such as toxins and other venom components, have long captured the focus of biotechnologists, leading to the merger between environmental omics and toxinology termed as ‘venomics’. Indeed, bioactive proteins such as conopeptides, conotoxins, turripeptides and others are long deemed important subjects of research. Even though current mainstream paradigms set the focus on secondary metabolites from marine organisms, transcriptomics and proteomics approaches and their combination are rising strategies for screening for thousands of proteins and peptides in non-conventional biological models, emphasising, but not limited to, marine invertebrate animals due to their abundance, biodiversity and uncanny biochemical strategies to cope with selective pressure in literally every known marine habitat. Untargeted approaches, such as RNA-Seq – based transcriptomics and tandem mass spectrometry – based proteomics, can circumvent limitations related with absent or reduced genomic annotation. The present review will outline the main contributions of ‘omics’ and computational approaches for bioprospecting for proteinaceous marine bioactives. Despite the relatively low number of ‘omics’ studies with the main purpose of discover novel compounds, there is already important literature showcasing pipelines and approaches for revolutionising the exploration of the ocean.

The endomembrane system in plant cells enables the cell to manage and coordinate a variety of membranous compartments so that they and their contents arrive at the right location. The secretory pathway is an essential part of this complex network and has its gateway at the Endoplasmic Reticulum. Therefore, alterations at the ER can affect how protein trafficking takes place and how cargo leaves this organelle. With this work, we assessed how abnormalities at the Endoplasmic Reticulum would interfere with protein sorting and trafficking. We used an Arabidopsis mutant—leb-2 GFP-h, presenting abnormal ER morphology, and evaluated the expression of aspartic proteinases and genes related to vacuolar transport along with the localization of a specific vacuolar sorting signal—plant-specific insert (PSI). Our results show that alterations in the leb-2 GFP-h mutant did not disrupt the transport of PSI–mCherry to the vacuole but influenced the expression of endogenous aspartic proteinases. Furthermore, the study of key endomembrane genes expression revealed an upregulation of the SNARE proteins AtVAMP722 and AtVAMP723. The leb-2 mutant seems not to interfere with vacuolar routes but may be implicated in secretion events.

Abstract This research assessed the effect of decoupling hydraulic retention time (HRT) and solid retention time (SRT) on the production of volatile fatty acids (VFAs) via anaerobic fermentation of beet molasses. The performance of a continuous stirred tank reactor (CSTR, STR = HTR = 30 days) and two anaerobic sequencing batch reactors (AnSBR) with decoupled STR (30 days) and HRT (20 and 10 days) was compared. Previously, a temperature study in batch reactors (25, 35, and 55 °C) revealed 25 °C as the optimal temperature to maximize the VFAs yield and the long-chain VFAs (> C4) production, being selected for the continuous reactors operation. An HRT of 20 days in AnSBR led to an enhancement in bioconversion efficiency into VFAs (55.5% chemical oxygen demand basis) compared to the CSTR (34.9%). In contrast, the CSTR allowed the production of valuable caproic acid (25.4% vs 4.1% w/w of total VFAs in AnSBR). Decreasing further the HRT to 10 days in AnSBR was detrimental in terms of bioconversion efficiency (21.7%) due to primary intermediates (lactate) accumulation. By decoupling HRT and SRT, VFAs were maximized, revealing HRT as an effective tool to drive specific conversion routes (butyrate- or lactate-fermentation).

(1) Background: The aim was to analyze the reliability and validity of the Spanish version of the Mini Sarcopenia Risk Assessment (MSRA) in older adults. (2) Methods: A total of 136 participants (72.24 ± 5.21 years, 68.38% women) took part in the study. The MSRA includes two questionnaires with seven (MSRA-7) and five items (MSRA-5). First, reliability (inter-rater and test–retest) of the Spanish MSRA was studied, and then the total scores were compared with the presence of sarcopenia according to three different diagnostic criteria and with other parameters related to sarcopenia (clinical validation). (3) Results: The analysis showed excellent inter-rater and test–retest reliability. As for the clinical validation, and regardless of the criteria, both questionnaires had a high sensitivity (81.82–88.89% for the MSRA-5 and 90.91–94.44% for the MSRA-7), while the MSRA-5 showed a better specificity (32.00–33.90%) than the MSRA-7 (20.80–22.88%). Predictive positive values ranged from 9.57–17.02% (MSRA-5) and 9.17–15.54% (MSRA-7), while predictive negative values were high for both the MSRA-5 (95.24%) and the MSRA-7 (96.30–96.43%). The accuracy was better for the MSRA-5 (36.03–41.18%) than the MSRA-7 (26.47–32.35%), as well as the area under the curve (0.67–0.76 vs. 0.65–0.73, respectively). Higher MSRA-5 and MSRA-7 total scores significantly correlated with greater muscle strength, quantity and gait speed. (4) Conclusions: The adaptation of the Spanish MSRA questionnaires was successfully performed, and they are reliable and clinically valid tools for assessing sarcopenia.

This technical note aims to characterize and contextualize the creation of a new collection of identified skeletons stored in the University Institute of Health Sciences—CESPU. This project presents unique and innovative pedagogical opportunities, with students actively involved in the cleaning and processing of the bones and antemortem/postmortem information gathering, as well as in anthropology research projects. This XXI Century Identified Skeletal Collection is composed of 98 complete adult skeletons of both sexes and is still expanding, being the 10th contemporary collection available in Portugal. The skeletons consist exclusively of Portuguese nationals who died between 1946 and 2007, and the 42% that have been processed so far are in good preservation. Comparative learning outcomes after implementing this active learning strategy are not yet available. Improvements are expected since more participation and enthusiasm in classes and research were perceived.

Introduction: Gastric cancer (GC), as a multifactorial disease, exhibits a complex pathogenesis, having intra and inter-tumor heterogeneity, challenging the efficacy of current treatments. GC is a major global health problem, and better diagnostic and therapeutic strategies are needed, leading to further biomarkers research associated with innovative targeted therapies. Our aim was to study human epidermal growth factor receptor 2 (HER2), Epstein Barr virus (EBV), and CD34 expression in GC samples to understand the relationship between these markers and relate them with the clinicopathological characteristics. Methods: Thirty samples of primary carcinoma gastrectomy cases performed between February 2017 and December 2021 at the Hospital Distrital da Figueira da Foz, EPE, were studied after approval by the Ethics Committee. Immunohistochemistry assays were performed in sections of the selected tumor representative samples. Results: All samples were negative for EBV and only two GC samples were positive for HER2. A significant statistical relationship was found between gender and CD34+ stroma cells. Microvascular density and stroma CD34+ cells presented relations with clinicopathological features and a positive tendency between them. Our study was able to identify a relationship between CD34+ stroma cells and females. Conclusion: With this study, we mainly explored the potential role of CD34 as a biomarker in GC and projected possible associations of prognostic and therapeutic value toward other HER2 and EBV markers.

Abstract Muscle stem cells, the engine of muscle repair, are affected in myotonic dystrophy type 1 (DM1); however, the underlying molecular mechanism and the impact on the disease severity are still elusive. Here, we show using patients’ samples that muscle stem cells/myoblasts exhibit signs of cellular senescence in vitro and in situ. Single cell RNAseq uncovers a subset of senescent myoblasts expressing high levels of genes related to the senescence-associated secretory phenotype (SASP). We show that the levels of interleukin-6, a prominent SASP cytokine, in the serum of DM1 patients correlate with muscle weakness and functional capacity limitations. Drug screening revealed that the senolytic BCL-XL inhibitor (A1155463) can specifically remove senescent DM1 myoblasts by inducing their apoptosis. Clearance of senescent cells reduced the expression of SASP, which rescued the proliferation and differentiation capacity of DM1 myoblasts in vitro and enhanced their engraftment following transplantation in vivo. Altogether, this study identifies the pathogenic mechanism associated with muscle stem cell defects in DM1 and opens a therapeutic avenue that targets these defective cells to restore myogenesis.

Objetivos: Caracterizar o impacto da atual pandemia de COVID-19 na ansiedade, medo e stress nos profissionais de saúde oral. Métodos: Utilizou-se um questionário autoaplicado online que foi enviado, via email, para três associações na área da Medicina Dentária. As variáveis categóricas foram descritas através de frequências absolutas e relativas. As variáveis contínuas foram descritas utilizando a média e o desvio-padrão. Utilizou-se um modelo de regressão linear múltipla para selecionar preditores da ansiedade avaliados no questionário. Resultados: Obteve-se uma taxa de participação de 21,1%. A maioria dos participantes era do sexo feminino (67,2%). Através da escala GAD-7 apurámos que 18,3% dos profissionais apresentaram uma perturbação de ansiedade moderada e grave. A maioria dos participantes (86,7%) referiu ter medo de infetar os familiares e amigos. Verificámos que ser do sexo feminino, o medo de infetar a família e amigos, a possibilidade de a pandemia afetar negativamente a profissão, a perda de rendimentos e as novas condições de trabalho eram preditores estaticamente significativos para o aumento da ansiedade (p < 0,05). Pelo contrário, à medida que o número de anos de prática clínica aumenta, a ansiedade diminui significativamente (p=0,006). Conclusões: A pandemia de COVID-19 afetou negativamente os profissionais de saúde oral. As medidas de prevenção e os protocolos de controlo de infeção devem ser rigorosamente cumpridos, para que haja diminuição da transmissão do vírus e, consequentemente, diminua a ansiedade, o stress e o medo sentidos por estes profissionais.

We address the problem of systematizing the authoring of digital dictionaries for craft education from ethnographic studies and recordings. First, we present guidelines for the collection of ethnographic data using digital audio and video and identify terms that are central in the description of crafting actions, products, tools, and materials. Second, we present a classification scheme for craft terms and a way to semantically annotate them, using a multilingual and hierarchical thesaurus, which provides term definitions and a semantic hierarchy of these terms. Third, we link ethnographic resources and open-access data to the identified terms using an online platform for the representation of traditional crafts, associating their definition with illustrations, examples of use, and 3D models. We validate the efficacy of the approach by creating multimedia vocabularies for an online eLearning platform for introductory courses to nine traditional crafts.

Advances in tissue preservation techniques have allowed reproductive medicine and assisted reproductive technologies (ARTs) to flourish in recent years. Because radio- and chemotherapy procedures are often gonadotoxic, irreversible damage can preclude future gamete production and endocrine support. Accordingly, in recent years, the freezing and storage of gonadal tissue fragments prior to the first oncological treatment appointment and autologous transplantation post-recovery have been considered improved solutions for fertility recovery in cancer survivors. Nevertheless, the cryopreservation and transplantation of thawed tissues is still very limited, and positive outcomes are relatively low. This review aims to discuss the limitations of oncofertility protocols with a focus on the impacts of mitochondrial dysfunction, oxidative stress, and the loss of antioxidant defense in graft integrity.

Introdução: A afasia pode interferir com a capacidade de comunicar da pessoa, impactando negativamente na sua qualidade de vida (Patrício, 2015). Interlocutores preparados para comunicar com pessoas com dificuldades comunicativas podem ser um facilitador, nomeadamente profissionais de saúde que intervêm no processo de reabilitação (Hallowell, 2017). Existem poucos estudos que abordam o conhecimento que estes profissionais e os estudantes da área da saúde apresentam sobre afasia. Objetivos: Caraterizar o conhecimento sobre afasia; identificar as fontes de conhecimento, as estratégias comunicativas utilizadas e consideradas importante utilizar no contacto com a pessoa com afasia; descrever a relação entre o género e a área de formação no conhecimento sobre afasia e nas estratégias comunicativas utilizadas pelos estudantes finalistas dos cursos de enfermagem, fisioterapia, medicina e terapia ocupacional. Materiais e Métodos: Estudo descritivo-correlacional e quantitativo, de metodologia observacional e transversal. Integra uma amostra de 209 estudantes finalistas dos cursos de enfermagem, fisioterapia, medicina e terapia ocupacional. A recolha de dados efetuou-se através de um questionário de autoadministração, divulgado online. Posteriormente, analisaram-se as frequências, médias das respostas e, adicionalmente, os resultados do teste qui-quadrado de Pearson e do teste de Fisher. Resultados: A maioria dos participantes (98,1%) ouviu falar sobre afasia. Em geral, apresentam bom conhecimento sobre as características, a causa e as estratégias a utilizar perante esta perturbação, mas têm menor conhecimento acerca dos tipos de afasia existentes. Os alunos de medicina são os que têm mais conhecimento sobre as pessoas com afasia apresentarem dificuldades de compreensão e sobre o facto de não se tratar de uma perturbação da deglutição. Os estudantes de terapia ocupacional são os que têm mais conhecimento sobre a etiologia da afasia e os de medicina sobre os tipos de afasia existentes. Relativamente às estratégias, os estudantes de enfermagem são os que apresentam mais conhecimento sobre quais utilizar. Conclusão: Os estudantes finalistas revelam ter conhecimento sobre afasia, parecendo ser ainda pertinente reforçar o conhecimento referente aos tipos de afasia existentes. Apesar dos bons resultados, para que o processo comunicativo seja eficaz e os estudantes consigam prestar serviços de maior qualidade, poderá ser benéfico treinar o uso de estratégias comunicativas.

The article presents the RuralYU university entrepreneurship project, developed by the Youth University Design (YUDesign) Spontaneous Generation (GE) group of the School of Design Engineering (ETSID), at the Polytechnic University of Valencia (UPV). RuralYU aspires to reduce rural depopulation in collaboration with university students, who, by carrying out academic work, cover specific needs with the aim of strengthening rural development. The idea of ​​an inter-university collaboration programme with the rural world arose after having won the Vodafone Foundation’s Project Lab 2021 national contest within the framework of GLOBAL CHANGE with the proposal “RuralLife4Good. Accommodation grant programme for the empowerment of rural environments”. The winning proposal, which was implemented as a pilot project in Ràfol de Salem during the 2021-2022 academic year, values ​​the importance of the university at the service of society, in this case as an axis of urgent and necessary activation to avoid rural abandonment.

Forensic sciences aggregate a series of scientific and technological domains that aid the judiciary and judicial system. Despite the highly specialized and qualified professionals taking a role in forensic practice, this paradigmatic integration of law, science, and technology often leads them to face significant ethical challenges. Indeed, forensic sciences hold a unique position due to the social and legal implications inherent to forensic expertise. The potential interference with personal rights and freedoms requires the professional practice of the forensic specialist to be guided by alignment with scientific-technical competence under the seal of the highest ethical principles. However, the absence of a specific statutory regulation blurs the performance of the forensic professional and may harm the professional credibility but also the scientific domain as a whole. The existence of ethical pillars is essential to support professional practice, following international recommendations in this regard. Herein, we propose a framework for a code of ethics and conduct that is based on the professional particularities characterizing the forensic practice but also considers the ethical issues that are mandatory to ensure high levels of reliability and credibility of forensic specialists.

Abstract Microglia are brain-resident myeloid cells and play a major role in the innate immune responses of the CNS and the pathogenesis of Alzheimer's disease (AD). However, the contribution of nonparenchymal or brain-infiltrated myeloid cells to disease progression remains to be demonstrated. Here, we show that monocyte-derived cells (MDC) invade brain parenchyma in advanced stages of AD continuum using transcriptional analysis and immunohistochemical characterization in post-mortem human hippocampus. Our findings demonstrated that a high proportion (60%) of demented Braak V–VI individuals was associated with up-regulation of genes rarely expressed by microglial cells and abundant in monocytes, among which stands the membrane-bound scavenger receptor for haptoglobin/hemoglobin complexes or Cd163. These Cd163-positive MDC invaded the hippocampal parenchyma, acquired a microglial-like morphology, and were located in close proximity to blood vessels. Moreover, and most interesting, these invading monocytes infiltrated the nearby amyloid plaques contributing to plaque-associated myeloid cell heterogeneity. However, in aged-matched control individuals with hippocampal amyloid pathology, no signs of MDC brain infiltration or plaque invasion were found. The previously reported microglial degeneration/dysfunction in AD hippocampus could be a key pathological factor inducing MDC recruitment. Our data suggest a clear association between MDC infiltration and endothelial activation which in turn may contribute to damage of the blood brain barrier integrity. The recruitment of monocytes could be a consequence rather than the cause of the severity of the disease. Whether monocyte infiltration is beneficial or detrimental to AD pathology remains to be fully elucidated. These findings open the opportunity to design targeted therapies, not only for microglia but also for the peripheral immune cell population to modulate amyloid pathology and provide a better understanding of the immunological mechanisms underlying the progression of AD.

Abstract Peer victimization have a negative impact on Health-Related Quality of Life (HRQoL) during adolescence, however some personal skills such a person's resilience could play a significant role in this relationship. In this context, this study aims to analyse if resilience is a moderator of the relation between peer victimization and HRQoL’s psychological dimensions. Sociodemographic data, peer victimization, psychological domains of HRQoL and resilience were measures in a sample of 1428 secondary school students using the following scales: “Adolescent Peer Relations Instrument-Bullying”, “KIDSCREEN-52” and “Brief Resilient Coping Scale. Different multivariate analyses were carried out using linear regression. PROCESS tool was used to examine the moderating role of resilience, with John-Neyman post-hoc approach to quantify moderation. Results suggest that resilience could moderate the association between physical (β = − 0.0021; p = 0.025) and verbal victimization (β = − 0.0018; p = 0.024) and the “Mood and Emotions” dimension of adolescents’ HRQoL. Nevertheless, this regulating influence appears to be faint (∆R2 0.004). Resilience showed no moderating effect between social victimization and psychological dimensions of HRQoL. We concluded that resilience could function as a protective factor that weakly regulates the negative association between physical and verbal victimization and the psychological sphere of adolescents’ HRQoL.

Objectives As age increases, people generally start experiencing problems related to independent living, resulting in an increased need for long-term care services. Investing in sustainable solutions to promote independent living is therefore essential. Subsequently, reablement is a concept attracting growing interest. Reablement is a person-centred, holistic approach promoting older adults’ active participation through daily, social, leisure and physical activities. The aim of this paper is to describe the development and content of I-MANAGE, a model for a reablement programme for community-dwelling older adults.Design The development of the programme was performed according to the Medical Research Council framework as part of the TRANS-SENIOR international training and research network. A co-creation design was used, including literature research, observations, interviews, and working group sessions with stakeholders.Setting and participants The interviews and working group sessions took place in the Dutch long-term home care context. Stakeholders invited to the individual interviews and working group sessions included care professionals, policymakers, client representatives, informal caregiver representatives, informal caregivers, and scientific experts.Results The co-creation process resulted in a 5-phase interdisciplinary primary care programme, called I-MANAGE. The programme focuses on improving the self-management and well-being of older adults by working towards their meaningful goals. During the programme, the person’s physical and social environment will be put to optimal use, and sufficient support will be provided to informal caregivers to reduce their burden. Lastly, the programme aims for continuity of care and better communication and coordination.Conclusion The I-MANAGE programme can be tailored to the local practices and resources and is therefore suitable for the use in different settings, nationally and internationally. If the programme is implemented as described, it is important to closely monitor the process and results.

IntroductionThe engagement of the SARS-CoV-2 spike protein with ACE2 is a critical step for viral entry to human cells, and, therefore, blocking this interaction is a major determinant of the efficacy of monoclonal antibody therapeutics and vaccine elicited serum antibodies. The emergence of SARS-CoV-2 variants has necessitated the development of adaptable assays that can be applied to assess the effectiveness of antibody-based therapeutics.MethodsThrough the testing of a range of recombinant spike proteins, we have developed a cell-based, ACE2/spike protein interaction assay that characterises monoclonal anti-spike protein antibodies and neutralising antibodies in donor serum. The assay uses high-content imaging to quantify cell-bound spike protein fluorescence.ResultsUsing spike proteins from the original “Wuhan” SARS-CoV-2 strain and the Delta and Omicron variants, we identified differential blocking activity of three monoclonal antibodies directed against the spike receptor-binding domain. Importantly, biological activity in the spike interaction assay translated to efficacy in a SARS-CoV-2 infection assay.DiscussionThe spike protein interaction assay can be used to monitor anti-spike antibodies against the major known SARS-CoV-2 variants and is readily adaptable for quantification of the impact of antibodies against new and emerging SARS-CoV-2 variants.

Tacrolimus (Tac) is pivotal in preventing acute graft-versus-host disease (GVHD) after allogeneic hematopoietic stem cell transplantation (alloHSCT). It has been reported that genetic factors, including CYP3A5*3 and CYP3A4*22 polymorphisms, have an impact on Tac metabolism, dose requirement, and response to Tac. There is limited information regarding this topic in alloHSCT. The CYP3A5 genotype and a low Tac trough concentration/dose ratio (Tac C0/D ratio) can be used to identify fast metabolizers and predict the required Tac dose to achieve target concentrations earlier. We examined 62 Caucasian alloHSCT recipients with a fast metabolizer phenotype (C0/dose ratio ≤ 1.5 ng/mL/mg), assessing CYP3A5 genotypes and acute GVHD incidence. Forty-nine patients (79%) were poor metabolizers (2 copies of the variant *3 allele) and 13 (21%) were CYP3A5 expressers (CYP3A5*1/*1 or CYP3A5*1/*3 genotypes). CYP3A5 expressers had lower C0 at 48 h (3.7 vs. 6.2 ng/mL, p = 0.03) and at 7 days (8.6 vs. 11.4 ng/mL, p = 0.04) after Tac initiation, tended to take longer to reach Tac therapeutic range (11.8 vs. 8.9 days, p = 0.16), and had higher incidence of both global (92.3% vs. 38.8%, p < 0.001) and grade II-IV acute GVHD (61.5% vs. 24.5%, p = 0.008). These results support the adoption of preemptive pharmacogenetic testing to better predict individual Tac initial dose, helping to achieve the therapeutic range and reducing the risk of acute GVHD earlier.

The current study investigates the venom-delivery system of green and red morphotypes of the sea anemone Actinia equina to disclose its potential as a source of bioactive compounds. We compared the two morphotypes using electron and optical microscopy, proteomics, and toxicity assessment on zebrafish embryos. Specialized venom-injecting cells (nematocysts) are equally distributed and found in the tentacles of both varieties. Proteomics revealed proteins of interest in both red and green Actinia, yielding the three most abundant Gene Ontology (GO) terms related to the biological processes “proteolysis”, “hemolysis in another organism” and “lipid catabolic process”. Neurotoxins and cytolytic toxins similar to known cnidarian toxins like PsTX-60A and AvTX-60A, for instance, were identified in both types. Extracts from green and red anemones were toxic to zebrafish embryos, with green anemone venom appearing to be more potent. The findings highlight the presence of proteinaceous toxins in A. equina and the potential for different varieties to possess distinct bioactive compounds. Notably, pore-forming toxins are suggested for molecular probes and immunotoxins, making them valuable assets for potential biotechnological and biomedical purposes.

Between 2016 and 2023, a cross-sectional study was conducted in the central region of Portugal in order to better understand the epidemiology and public health risks resulting from the handling and consumption of game animals infected with Brucella spp. The seroprevalence and risk factors for Brucella spp. seropositivity were evaluated. Antibodies against Brucella spp. were determined using a commercial enzyme-linked immunosorbent assay (ELISA) according to the manufacturer’s instructions. Results showed that in the 650 serum samples collected from red deer (n = 298) and wild boars (n = 352) in Portugal, 21.7% (n = 141; 95% CI: 18.6–25.1%) tested positive. Wild boar had a significantly higher prevalence (35.5%; 95% CI: 30.5–40.8%) than red deer (5.4%, 95% CI: 3.1–8.6%; p ≤ 0.001). Risk factors for seropositivity were investigated using multivariable logistic regression models. The odds of being seropositive was 8.39 (95% CI: 4.75–14.84; p ≤ 0.001) times higher in wild boar than in red deer. Correlations between sex, age, body condition, and seropositivity could not be observed. The higher seroprevalence in wild boar suggests that this species may primarily contribute to the Brucella spp. ecology in central Portugal.

Proteinaceous toxins are peptides or proteins that hold great biotechnological value, evidenced by their ecological role, whether as defense or predation mechanisms. Bioprospecting using bioinformatics and omics may render screening for novel bioactives more expeditious, especially considering the immense diversity of toxin-secreting marine organisms. Eulalia sp. (Annelida: Phyllodocidae), a toxin bearing marine annelid, was recently shown to secrete cysteine-rich protein (Crisp) toxins (hitherto referred to as ‘phyllotoxins’) that can immobilize its prey. By analyzing and validating transcriptomic data, we narrowed the list of isolated full coding sequences of transcripts of the most abundant toxins or accompanying bioactives secreted by the species (the phyllotoxin Crisp, hyaluronidase, serine protease, and peptidases M12A, M13, and M12B). Through homology matching with human proteins, the biotechnological potential of the marine annelid’s toxins and related proteins was tentatively associated with coagulative and anti-inflammatory responses for the peptidases PepM12A, SePr, PepM12B, and PepM13, and with the neurotoxic activity of Crisp, and finally, hyaluronidase was inferred to bear properties of an permeabilizing agent. The in silico analysis succeeded by validation by PCR and Sanger sequencing enabled us to retrieve cDNAs can may be used for the heterologous expression of these toxins.

Este artículo examina la experiencia del boletín barcelonés Quienes no han tenido jamás el “derecho” a la(s) palabra(s), la(s) toma(n) ya, que publicó seis números entre 1976 y 1978 en diálogo permanente con la Coordinadora de Presos en Lucha (COPEL). En un primer momento, se aborda el ejercicio de toma de palabra de los presos desarrollado en esta publicación, a través del análisis de los mensajes y comunicados contenidos en ella, pero también de los “desmontajes” o détournements a los que somete a la prensa generalista. Después, se tratan de abordar las prácticas de producción y distribución del boletín, y su función en la construcción de solidaridad desde el exterior. En último lugar, se localizarán algunas de las supervivencias de esta práctica comunicativa desde el interior de las prisiones en la actualidad, apuntando los rastros de una memoria viva en la lucha anticarcelaria, e identificando una vez más la centralidad que guarda el testimonio carcelario en ella.

Organismal aging is impacted by the deterioration of tissue turnover mechanisms due, in part, to the decline in stem cell function. This decline can be related to mitochondrial dysfunction and underlying energetic defects that, in concert, help drive biological aging. Thus, mitochondria have been described as a potential interventional target to hinder the loss of stem cell robustness, and subsequently, decrease tissue turnover decline and age-associated pathologies. In this review, we focused our analysis on the most recent literature on mitochondria and stem cell aging and discuss the potential benefits of targeting mitochondria in preventing stem cell dysfunction and thus influencing aging.

Violence in adolescent intimate relationships poses a real threat to adolescent well-being and is a risk factor for recurrent violent patterns in adult marital relationships. The present study aimed to understand the relationship between different dimensions of childhood trauma and dating violence perpetration and the mediating role of temperament. The sample was composed of 3497 adolescents (n = 1549 boys, n = 1948 girls; M = 1.56, SD = 0.497) aged between 10 and 22 years (M = 15.15, SD = 1.83). Instruments used in this study included the Social Desirability Scale, the Childhood Trauma Questionnaire (CTQ), and the Early Adolescent Temperament Questionnaire (EATQ). The results show that temperament plays a mediating role in the relationship between dating violence perpetration and emotional, physical, and sexual abuse. Furthermore, the results suggest that adolescents who have been abused in childhood tend to demonstrate more temperamental problems and a greater susceptibility to the perpetration of teen dating violence, but only in the more severe dimensions of childhood trauma.

Cytomegalovirus (CMV) infection is a relevant cause of morbimortality in patients receiving allogeneic stem cell transplantation (allo-HCT). Foscarnet (FCN) is an effective drug against CMV administered intravenously and usually on an inpatient basis. The Home Care Unit (HCU) for hematologic patients at our hospital designed an at-home FCN administration model to avoid the hospitalization of patients requiring FCN treatment. This study analyzes whether the at-home administration of FCN is as safe and effective as its hospital administration. We collected and compared demographic, clinical, analytical, and economic data of patients with CMV infection post-allo-HCT who received FCN in the hospital (n = 16, 17 episodes) vs. at-home (n = 67, 88 episodes). The proportions of patients with cured CMV infections were comparable between the two groups (65.9% vs. 76.5%, p = 0.395). The median duration of FCN treatment was 15 (interquartile range [IQR] 9–23) and 14 (IQR 11–19) days in the HCU and inpatient cohorts, respectively (p = 0.692). There were no significant differences in the FCN toxicities between groups except for hypocalcemia (26.1% vs. 58.8%, p = 0.007), which was more prevalent in the inpatient cohort. A significant cost-effectiveness was found in the HCU cohort, with a median savings per episode of EUR 5270. It may be concluded that home administration of FCN is a safe, effective, and cost-efficient therapeutic option for patients with CMV infection and disease.

The immense biodiversity of marine invertebrates makes them high-value targets for the prospecting of novel bioactives. The present study investigated proteinaceous toxins secreted by the skin and proboscis of Glycera alba (Annelida: Polychaeta), whose congenerics G. tridactyla and G. dibranchiata are known to be venomous. Proteomics and bioinformatics enabled the detection of bioactive proteins that hold potential for biotechnological applications, including toxins like glycerotoxins (GLTx), which can interfere with neuromuscular calcium channels and therefore have value for the development of painkillers, for instance. We also identified proteins involved in the biosynthesis of toxins. Other proteins of interest include venom and toxin-related bioactives like cysteine-rich venom proteins, many of which are known to interfere with the nervous system. Ex vivo toxicity assays with mussel gills exposed to fractionated protein extracts from the skin and proboscis revealed that fractions potentially containing higher-molecular-mass venom proteins can exert negative effects on invertebrate prey. Histopathology, DNA damage and caspase-3 activity suggest significant cytotoxic effects that can be coadjuvated by permeabilizing enzymes such as venom metalloproteinases M12B. Altogether, these encouraging findings show that venomous annelids are important sources of novel bioactives, albeit illustrating the challenges of surveying organisms whose genomes and metabolisms are poorly understood.

Abstract Alzheimer’s disease (AD) is a major adult-onset neurodegenerative condition with no available treatment. Compelling reports point amyloid-β (Aβ) as the main etiologic agent that triggers AD. Although there is extensive evidence of detrimental crosstalk between Aβ and microglia that contributes to neuroinflammation in AD, the exact mechanism leading to neuron death remains unknown. Using postmortem human AD brain tissue, we show that Aβ pathology is associated with the necroptosis effector pMLKL. Moreover, we found that the burden of Aβ oligomers (Aβo) correlates with the expression of key markers of necroptosis activation. Additionally, inhibition of necroptosis by pharmacological or genetic means, reduce neurodegeneration and memory impairment triggered by Aβo in mice. Since microglial activation is emerging as a central driver for AD pathogenesis, we then tested the contribution of microglia to the mechanism of Aβo-mediated necroptosis activation in neurons. Using an in vitro model, we show that conditioned medium from Aβo-stimulated microglia elicited necroptosis in neurons through activation of TNF-α signaling, triggering extensive neurodegeneration. Notably, necroptosis inhibition provided significant neuronal protection. Together, these findings suggest that Aβo-mediated microglia stimulation in AD contributes to necroptosis activation in neurons and neurodegeneration. As necroptosis is a druggable degenerative mechanism, our findings might have important therapeutic implications to prevent the progression of AD.
