1. Dual function of the transcription factor Ftz-f1 on oviposition in the cockroach Blattella germanica

Abstract

The transcription factor Ftz-f1 has multiple functions in insect development in a spatial-temporal line. One of these roles is in the insect ovaries, specifically in the regulation of steroidogenic enzymes production. We studied the function of F in Blattella germanica oogenesis, as it shows two moments of high expression in ovaries: before the imaginal moult, and just before ovulation in the adult. Injecting dsftz-f1 into adult females, either just after the imaginal moult or just prior to choriogenesis, prevented oviposition, with differences between the two approaches. In 3-day-old adult females treated with dsftz-f1 just after the emergence, the expression of ftz-f1 was not modified, but the steroidogenic genes increased their expression. ftz-f1 transcript levels in the ovaries of 5-day-old dsftz-f1-treated females were significantly depleted, and the expression levels of the same steroidogenic genes began to decrease. These results suggest that Ftz-f1 regulates the expression of steroidogenic genes in B. germanica, with phm possibly being a key target. Ftz-f1 has a different temporal function in the cytoskeleton of follicular cells of the basal ovarian follicles. Early in the gonadotrophic cycle, Ftz-f1 promotes the expression of genes related to the cytoskeleton and muscle proteins, while at the end of the cycle it maintains the expression levels of these genes, thus ensuring correct ovulation.

2. In Vitro Cellular Uptake Studies of Self-Assembled Fluorinated Nanoparticles Labelled with Antibodies

Abstract

Nanoparticles (NPs) functionalized with antibodies (Abs) on their surface are used in a wide range of bioapplications. Whereas the attachment of antibodies to single NPs to trigger the internalization in cells via receptor-mediated endocytosis has been widely studied, the conjugation of antibodies to larger NP assemblies has been much less explored. Taking into account that NP assemblies may be advantageous for some specific applications, the possibility of incorporating targeting ligands is quite important. Herein, we performed the effective conjugation of antibodies onto a fluorescent NP assembly, which consisted of fluorinated Quantum Dots (QD) self-assembled through fluorine-fluorine hydrophobic interactions. Cellular uptake studies by confocal microscopy and flow cytometry revealed that the NP assembly underwent the same uptake procedure as individual NPs; that is, the antibodies retained their targeting ability once attached to the nanoassembly, and the NP assembly preserved its intrinsic properties (i.e., fluorescence in the case of QD nanoassembly).

Abstract

A new CMS tracker detector will be installed for operation at the High Luminosity LHC (HL-LHC). This detector comprises modules with two closely spaced parallel sensor plates and front-end ASICs capable of transmitting tracking information to the CMS Level-1 (L1) trigger at the 40 MHz beam crossing rate. The inclusion of tracking information in the L1 trigger decision will be essential for selecting events of interest efficiently at the HL-LHC. The CMS Binary Chip (CBC) has been designed to read out and correlate hits from pairs of tracker sensors, forming so-called track stubs. For the first time, a prototype irradiated module and a full-sized module, both equipped with the version 2 of the CBC, have been operated in test beam facilities. The efficiency of the stub finding logic of the modules for various angles of incidence has been studied. The ability of the modules to reject tracks with transverse momentum less than 2 GeV has been demonstrated. For modules built with irradiated sensors, no significant drop in the stub finding performance has been observed. Results from the beam tests are described in this paper.

Abstract

Environmental stress factors caused by climate change affect plant growth and crop production, and pose a growing threat to sustainable agriculture, especially for tree crops. In this context, we sought to investigate the responses to climate change of two Prunus rootstocks (GF677 and Adesoto) budded with Catherina peach cultivar. Plants were grown in 15 L pots in temperature gradient greenhouses for an 18 days acclimation period after which six treatments were applied: [CO2 levels (400 versus 700 µmol mol-1), temperature (ambient versus ambient + 4°C), and water availability (well irrigated versus drought)]. After 23 days, the effects of stress were evaluated as changes in physiological and biochemical traits, including expression of relevant genes. Stem water potential decreased under drought stress in plants grafted on GF677 and Adesoto rootstocks; however, elevated CO2 and temperature affected plant water content differently in both combinations. The photosynthetic rate of plants grafted on GF677 increased under high CO2, but decreased under high temperature and drought conditions. The photosynthetic rates of plants grafted onto Adesoto were only affected by drought treatment. Furthermore, in GF677–Catherina plants, elevated CO2 alleviated the effect of drought, whereas in those grafted onto Adesoto, the same condition produced acclimation in the rate. Stomatal conductance decreased under high CO2 and drought stress in both grafted rootstocks, and the combination of these conditions improved water-use efficiency. Changes in the sugar content in scion leaves and roots were significantly different under the stress conditions in both combinations. Meanwhile, the expression of most of the assessed genes was significantly affected by treatment. Regarding genotypes, GF677 rootstock showed more changes at the molecular and transcriptomic level than did Adesoto rootstock. 5. Geometrically designed domain wall trap in tri-segmented nickel magnetic nanowires for spintronics devices

Abstract

[EN] “Domain wall traps” have been engineered and well-exploited in nanostrips by creating a geometrical trapping site, e.g. a single notch along a stripe, compared to diameter-modulated (DM) cylindrical magnetic nanowires (NWs) where multi-segmented DM-NWs have been generally studied. Here, we report our systematic study on the magnetization behavior, domain wall structure and its nucleation/propagation in tri-segmented diameter-modulated Ni nanowires, a simple system to investigate the magnetization reversal as function of segment geometry and lay-out order. We find out that the magnetization behavior of single Ni DM-NWs exhibits the significance of positional ordering of thick and thin segments, distinguished by two distinct geometries including: dumbbell-type (type I) and rolling pin-type (type II). Based on experimental and theoretical simulations, it was evidenced that the wide-narrow junctions create trap sites for domain walls where the narrow segment restricts their motion. This type of geometrically engineered nanowires exhibit potential efficiency for future novel spintronic devices in particular when assembled in arrays of DM-NWs as a practical three-dimensional memory device.

6. Kauniolide synthase is a P450 with unusual hydroxylation and cyclization-elimination activity

Abstract

Guaianolides are an important class of sesquiterpene lactones with unique biological and pharmaceutical properties. They have been postulated to be derived from germacranolides, but for years no progress has been made in the elucidation of their biosynthesis that requires an unknown cyclization mechanism. Here we demonstrate the isolation and characterization of a cytochrome P450 from feverfew (Tanacetum parthenium), kauniolide synthase. Kauniolide synthase catalyses the formation of the guaianolide kauniolide from the germacranolide substrate costunolide. Unlike most cytochrome P450s, kauniolide synthase combines stereoselective hydroxylation of costunolide at the C3 position, with water elimination, cyclization and regioselective deprotonation. This unique mechanism of action is supported by in silico modelling and docking experiments. The full kauniolide biosynthesis pathway is reconstructed in the heterologous hosts Nicotiana benthamiana and yeast, paving the way for biotechnological production of guaianolide-type sesquiterpene lactones.

7. Versatile theranostics agents designed by coating ferrite nanoparticles with biocompatible polymers

Abstract

Three biocompatible polymers, polyethylene glycol (PEG), dextran and chitosan, have been used in this work to control the colloidal stability of magnetic nanoparticles (14 ± 5 nm in diameter) and to vary the aggregation state in order to study their effect on relaxometric and heating properties. Two different coating strategies have been deeply developed; one based on the formation of an amide bond between citric acid coated nanoparticles (NPs) and amine groups present on the polymer surface and the other based on the NP encapsulation. Relaxometric properties revealed that proton relaxation rates strongly depend on the coating layer hydrophilicity and the aggregation state of the particles due to the presence of magnetic interactions. Thus, while PEG coating reduces particle aggregation by increasing inter-particle spacing leading to reduction of both T and T relaxation, dextran and chitosan lead to an increase mainly in T values due to the aggregation of particles in bigger clusters where they are in close contact. Dextran and chitosan coated NPs have also shown a remarkable heating effect during the application of an alternating magnetic field. They have proved to be potential candidates as theranostic agents for cancer diagnosis and treatment. Finally, cytotoxicity of PEG conjugated NPs, which seem to be ideal for intravenous administration because of their small hydrodynamic size, was investigated resulting in high cell viability even at 0.2 mg Fe ml after 24 h of incubation. This suspension can be used as drug/biomolecule carrier for in vivo applications.

Abstract

A proton transfer compound, (OPDH)2(dipic), (dipicH2 = 2,6-pyridinedicarboxylic acid and OPD = o-phenylenediamine), was synthesized and characterized by IR, 1H and 13C NMR spectroscopy. The reaction of (OPDH)2(dipic) with CoCl2·6H2O in water results in the formation of the novel complex [OPDH]2[Co(dipic)2]·H2O (1). This complex was characterized by elemental analysis, UV–vis, FTIR and the single-crystal X-ray methods. The crystal system is triclinic with space group P1. The protonation constants of dipic and OPD, the equilibrium constants for the dipic-OPD proton transfer system, the stoichiometry and the stability of complexation of this system with the CoII ion in an aqueous solution were investigated by the potentiometric pH titration method using Hyperquad 2008 program. The stoichiometry of the complex species in solution was the same as that of the metal ion complex found in the solid state.

Stomatal conductance decreased under high CO2 and drought stress in both grafted rootstocks, and the combination of these conditions improved water-use efficiency. Changes in the sugar content in scion leaves and roots were significantly different under the stress conditions in both combinations. Meanwhile, the expression of most of the assessed genes was significantly affected by treatment. Regarding genotypes, GF677 rootstock showed more changes at the molecular and transcriptomic level than did Adesoto rootstock. A coordinated shift was found between the physiological status and the transcriptomic responses. This study revealed adaptive responses to climate change at the physiological, metabolic, and transcriptomic levels in two Prunus rootstocks budded with 'Catherina'. Abstract

Tracker group of the CMS collaboration: et al., A new CMS tracker detector will be installed for operation at the High Luminosity LHC (HL-LHC). This detector comprises modules with two closely spaced parallel sensor plates and front-end ASICs capable of transmitting tracking information to the CMS Level-1 (L1) trigger at the 40 MHz beam crossing rate. The inclusion of tracking information in the L1 trigger decision will be essential for selecting events of interest efficiently at the HL-LHC. The CMS Binary Chip (CBC) has been designed to read out and correlate hits from pairs of tracker sensors, forming so-called track stubs. For the first time, a prototype irradiated module and a full-sized module, both equipped with the version 2 of the CBC, have been operated in test beam facilities. The efficiency of the stub finding logic of the modules for various angles of incidence has been studied. The ability of the modules to reject tracks with transverse momentum less than 2 GeV has been demonstrated. For modules built with irradiated sensors, no significant drop in the stub finding performance has been observed. Abstract

[EN] “Domain wall traps” have been engineered and well-exploited in nanostrips by creating a geometrical trapping site, e.g. a single notch along a stripe, compared to diameter-modulated (DM) cylindrical magnetic nanowires (NWs) where multi-segmented DM-NWs have been generally studied. Here, we report our systematic study on the magnetization behavior, domain wall structure and its nucleation/propagation in tri-segmented diameter-modulated Ni nanowires, a simple system to investigate the magnetization reversal as function of segment geometry and lay-out order. We find out that the magnetization behavior of single Ni DM-NWs exhibits the significance of positional ordering of thick and thin segments, distinguished by two distinct geometries including: dumbbell-type (type I) and rolling pin-type (type II). Based on experimental and theoretical simulations, it was evidenced that the wide-narrow junctions create trap sites for domain walls where the narrow segment restricts their motion. This type of geometrically engineered nanowires exhibit potential efficiency for future novel spintronic devices in particular when assembled in arrays of DM-NWs as a practical three-dimensional memory device., The financial support of Iranian National Science Foundation (INSF) under a grant number of 94010670 is acknowledged. Abstract

Calcineurin is an essential calcium‐activated serine/threonine phosphatase. The six NMR‐observable methionine methyl groups in the catalytic domain of human calcineurin Aα (CNA) were assigned and used as reporters of the presence of potential cis‐trans isomers in solution. Proline 84 is found in the cis conformation in most calcineurin X‐ray structures, and proline 309, which is part of a highly conserved motif in phosphoprotein phosphatases, was modeled with a cis peptide bond in one of the two molecules present in the asymmetric unit of CNA. We mutated each of the two prolines to alanine to force the trans conformation. Abstract

Contact: berville@supagro.inra.fr; The genetic base of sunflower elite lines is very narrow, due to many years of selection and breeding. To broaden the genetic diversity of the cultivated sunflower, in 1995 73 wild sunflower populations were crossed with 3 cultivated lines (Testers), and 219 hybrid offspring's were evaluated in the field. GCA and SCA effects were computed suggesting for all traits a genetic potential for improvement through selection. Study of the hybrids revealed that the wild accessions bear different genetic abilities to combine with the testers for traits of morphological architecture, phenology and yield (seed weight and seed oil). The variance due to GCA and SCA showed that gene action was additive for days to flowering, branching and plant height. Genotypes derived from the same geographic origin may have either good or poor general combing ability. The correlation between GCA and per se genotype performance was positive for all traits except for seed oil content. This was the first attempt to evaluate wild-cultivated hybrids in sunflower on a large scale and will be the starting point for the management of hybrid Helianthus annuus populations for breeding. GCA and SCA estimations will facilitate the definition of strategies to manage and exploit the natural diversity for this crop

Abstract

International audience; The objectives of the present study were to estimate the general combining ability (GCA) and specific combining ability (SCA) for partial resistance to phoma black stem and to identify the most promising combination for the selection of improved breeding lines. The response of five parental genotypes and their F1 hybrids to a phoma black stem isolate (MA6) were evaluated in a diallel programme under controlled growth chamber conditions. Significant GCA and SCA indicate that both additive and non-additive gene effects contributed in the inheritance of partial resistance to phoma black stem, however, the Baker ratio showed that the additive genetic effects were more important than nonadditive ones. It is recommended that the GGEbiplot methodology could be an excellent tool for visualizing entry by tester (diallel) data. By using this technique to analyse black stem severity data, interaction among the sunflower genotypes in providing partial resistance to phoma black stem was clearly identified. Based on GGEbiplot presentation and Griffing's diallel analysis, the mutant line ‘M6-54-1’ showed the largest GCA, indicating contribution towards partial resistance, and the genotype B454/03 presented the smallest GCA, indicating contribution towards susceptibility. Our results show that the F1 hybrids ‘SDR18×B454/03’ and ‘M6-54-1×B454/03’ showing heterosis for partial resistance to phoma black stem come from the crosses between a susceptible genotype ‘B454/03’ and two partially resistant genotypes (SDR18 and M6-54-1), originated from different breeding programmes. We conclude therefore that these genotypes possess at least some different resistance genes, which were expressed in the hybrids and led to the observed effects.
