Phytohormones play an important role at all stages of plant growth, influencing plant growth and development and regulating plant secondary metabolism, such as the synthesis of flavone, flavonol, anthocyanin, and other flavonoids. Flavonoids, a group of important secondary metabolites ubiquitous in plants, have antioxidative, anti-microbial, and anti-inflammatory activities and thus have a wide range of potential applications in Chinese medicine and food nutrition. With the development of biotechnology, phytohormones' regulation on flavonoids has become a research focus in recent years. This study reviewed the research progress on the mechanism of common phytohormones, such as abscisic acid, gibberellin, methyl jasmonate, and salicylic acid, in regulating flavonoid metabolism, and discussed the molecular mechanism of the synthesis and accumulation of flavonoids, aiming at clarifying the key role of phytohormones in modulating flavonoid metabolism. The result is of guiding significance for improving the content of flavonoids in plants through rational use of phytohormones and of reference value for exploring the mechanism of hormones in regulating flavonoid metabolism.

In this study, citrate synthase gene(CIT2), and malate synthase gene(MLS1) were successfully knocked out in β-amyrin-producing yeast cells by using CRISPR/CAS9. The promoter of phosphoglucose isomerase gene(PGI1) was replaced by that of cytochrome c oxidase subunit Ⅶa(Cox9)to weaken its expression, aiming to channel more carbon flux into the NADPH-producing pathway. The fermentation results showed that CIT2 deletion had no effect on the β-amyrin production. Compared with the control strain, the production of β-amyrin was increased by 1.85 times after deleting MLS1, reaching into 3.3 mg·L~(-1). By replacing the promoter of PGI1, the β-amyrin yield was 3.75 times higher than that of the control strain, reaching up to 6.7 mg·L~(-1). This study successfully knocked out the CITT2 and MLS1 genes and weakened the PGI1 gene by using CRISPR/CAS9, which directly influenced the production of β-amyrin and provided some reference for the the metabolic engineering of triterpernoid producing strain.

Pinellia ternata is a medicinal herb of Araceae, and its tubers are used as medicines. It is a common Chinese herbal medicine in China and has a large market demand. When exposing to strong light intensity and high temperature during the growth process, P. ternata withers in a phenomenon known asquot;sprout tumblequot;, which largely limits tuber production. Shade can effectively delay sprout tumble formation and increase its yield, however the relevant regulation mechanism is unclear. DNA methylation, as a self-modifying response to environmental changes, is often involved in the regulation of plant growth and development. In this study, P. ternata grown under natural light and 90% shading were selected as the control group and the experimental group for genomic DNA methylation analysis by using methylate sensitive amplification polymorphism(MSAP). The results showed that a total of 617 loci were detected with 20 pairs of primers, of which 311 were in the natural light group and 306 in the shading group. The methylation sites in the light and shading groups accounted for 58.2% and 71.57%, respectively, and the methylation ratios in the methylation sites were 27.65% and 29.41%, respectively, indicating that shading significantly induced the genome DNA methylation of P. ternata. Compared to the natural light group, shading promoted 32.51% of the genes methylation, while inducing 16.25% gene demethylation. This study reveals the DNA methylation variation of P. ternata under shading conditions, which lays a preliminary theoretical foundation for further analysis of the mechanism of shading regulation of P. ternata growth from epigenetic level.

Pinellia ternata belongs to the Araceae family and is a medicinal herb. The tuber is the medicinal organ with antitussive, antiemetic and anti-tumor activities. It is easy to encounter high temperature environment during the growth periods, leading to decrease of tuber production. At present, the mechanism of response to high temperature stress in P. ternata is still unknown. DNA methylation plays a vital role in plant protection against adversity stress as a way of epigenetic regulation. In this study, P. ternata was used as material for treatment of high temperature stress at 0 h, 6 h and 80 h, and methylation sensitive amplification polymorphism(MSAP) analysis was conducted on the changes of DNA methylation in its genome. The results showed that 20 pairs of MSAP primers were selected from 100 MSAP primers with multiple clear and uniform bands, and 353, 355 and 342 loci were amplified from materials of P. ternata treated in the high temperature stress 0 h, 6 h and 80 h, respectively. Cytosine methylation levels of CCGG context in the above materials were characterized as 60.91%, 44.79% and 44.74%, respectively. And the full methylation ratios were 16.71%, 22.25% and 29.24, respectively. It demonstrated that high temperature stress significantly induced the down-regulation of DNA methylation level and up-regulation of the full methylation rate in P. ternata genome. This study provides a preliminary theoretical reference for analyzing the mechanism of P. ternata responding to high temperature stress from the epigenetic perspective.

In this study, the synthetic pathway of β-amyrin was constructed in the pre-constructed Saccharomyces cerevisiae chassis strain Y0 by introducing β-amyrin synthase from Glycyrrhiza uralensis, resulting strain Y1-C20-6, which successfully produced β-amyrin up to 5.97 mg·L~(-1). Then, the mevalonate pyrophosphate decarboxylase gene(ERG19), mevalonate kinase gene(ERG12), 3-hydroxy-3-methylglutaryl-CoA synthase gene(ERG13), phosphomevalonate kinase gene(ERG8) and IPP isomerase gene(IDI1)were overexpressed to promoted the metabolic fluxto the direction of β-amyrin synthesis for further improving β-amyrin production, resulting the strain Y2-C2-4 which produced β-amyrin of 10.3 mg·L~(-1)under the shake flask fermentation condition. This is 100% higher than that of strain Y1-C20-6, illustrating the positive effect of the metabolic engineering strategy applied in this study. The titer of β-amyrin was further improved up to 157.4 mg·L~(-1) in the fed-batch fermentation, which was almost 26 fold of that produced by strain Y1-C20-6. This study not only laid the foundation for the biosynthesis of β-amyrin but also provided a favorable chassis strain for elucidation of cytochrome oxidases and glycosyltransferases of β-amyrin-based triterpenoids.

Gekko gecko (Tokay Gecko) is a valuable traditional Chinese medicine. In this study, the loop-mediated isothermal amplification (LAMP) technique was introduced for visual rapid identification of G. gecko from adulterants. A total of sixty-five 12S rRNA sequences of fourteen species of G. gecko and its adulterants were obtained. The results showed that G. gecko could be identified from its adulterants through BLAST analysis based on 12S rRNA regions. The 12S rRNA sequences of ten batches of G. gecko were conserved. There were only two haplotypes and three variation sites in the available regions for primers design. Six specific LAMP primers were successfully designed online based on 12S rRNA sequences. The visual rapid detection of G. gecko could be achieved with the optimized conditions (64 °C for 1 h and 80 °C for 5 min). And the required minimal template concentration was 5 μg·L⁻¹ while conventional PCR with 0.5 mg·L⁻¹. Consequently, the LAMP method established from this study was rapid, specific, highly sensitive, and simple. It could be applied to detect G. gecko from its adulterants efficiently.

To identify the precious bile powder and its adulterants by DNA barcoding, and establish its standard experimental process to ensure the safe and effective utilization. Total twelve sequences from samples of bear bile powder which come from Ursus thibetanus for DNA extraction, PCR(polymerase chain reaction) and sequence, then using CodonCode Aligner V 7.0.1 shear primer region to obtain COI sequence. The COI sequences of U. arctos and their adulterants were obtained from GenBank. MEGA7.0 software was applied for analyzing mutation, calculating intraspecific and interspecific K2P(Kimura 2-Parameter) genetic distance and constructing the Neighbor-joining tree(NJ). The results showed that the maximum K2P genetic distance of bear bile powder of U. thibetanus and U. arctos are far less than minimum K2P genetic distance within its adulterants species, and the results of NJ tree demonstrated that each species could be distinguished from the counterfeits obviously. DNA barcoding is a safe, convenient and reliable technique for species identification, and it is important to establish the standard sequence of COI sequences for animal medicines.

We reported on the generation of Q-switched pulse in an Yb-doped fiber laser by using a filmy polyvinyl alcohol (PVA)-based gold nanorods (GNRs) saturable absorber (SA). The GNRs are synthesized through seed-mediated method whose longitudinal surface plasmon resonance (SPR) absorption peak is located at 1038 nm. The modulation depth of the GNRs SA is ∼4.06%. By gradually increasing the pump power from 62 mW to 128 mW, the repetition rate of Q-switched pulse increases from 8.78 kHz to 20.78 kHz and the pulse duration decreases from 9.43 μs to 3.65 μs. In addition, the dual-wavelength switchable Q-switched operation was also observed. The obtained results further expand the applications of GNRs SA to the field of Q-switched pulsed fiber lasers at 1.0 μm waveband.

We report on the generation of Q-switched and Q-switched mode-locked (QML) pulses in an erbium-doped fiber ring laser by using a polyvinyl alcohol (PVA) -based gold nanorod (GNR) saturable absorber (SA). The PVA-based GNR SA has a modulation depth of ∼4.8% and a non-saturable loss of ∼26.9% at 1.5 μm. A Q-switched pulse train with a repetition rate varying from 18.70 to 39.85 kHz and a QML pulse train with an envelope repetition rate tuning from 20.31 to 31.50 kHz are obtained. Moreover, the lasing wavelengths of the Q-switched pulses can be flexibly tuned by introducing a narrow bandwidth, tunable filter into the laser cavity. The results demonstrate that the GNRs exhibit good optical performance and can find a wide range of applications in the field of laser technology.

Tartary buckwheat Fagopyrum tataricum is an important medicinal and functional herb due to its rich content of flavonoids in the seeds. F.tataricum exhibited good functions for free radicals scavenging， anti-oxidation， anti-aging activities. Although much genetic knowledge of the synthesis， regulation， accumulation of rutin， the genetic basis of proanthocyanidins(PAs) in tartary buckwheat and their related gene expression changes under different lights(blue， red， far red， ultraviolet light) remain largely unexplored. In this study， we cloned one anthocyanidin reductase gene(ANR) and two leucocyanidin reductase gene(LAR) named FtANR，FtLAR1，FtLAR3 involved in formation of(+)-catechin and(-)-epicatechin precusor proanthocyanidin by digging out F. tataricum seed transcriptome data. The expression data showed that the opposite influence of red light on these gene transcript level compared to others lights. The expression levels of FtANR and FtLAR1 decreased and FtLAR3 appeared increment after exposed in the red light， while the expression levels of those genes appeared opposite result after exposed in the blue and far red light.

Residue of Mori Cortex was studied to optimize its enzymatic hydrolysis process, and explore its potential as a carbon source for biochemistry and biofuel production. The cellulose content of diluted acid pretreated (DAP) and non-pretreated from Mori Cortex were measured in this study, and the results showed that the cellulose content of DAP and non-pretreated from Mori Cortex were 52.5% and 47%, respectively. This higher cellulose content indicated that residue of Mori Cortex had the potential to act as a carbon source for biochemistry and biofuel production. Enzymatic hydrolysis of pretreated and non-pretreated from Mori Cortex was conducted under different enzyme loading amount. 40 FPU·(g DW）⁻¹ enzyme loading was determined as the optimal amount by comparing the yield of sugar and the rate of enzymolysis. Under this condition, the concentrations of glucose, xylose, arabinose sugar were 23.82, 4.84, 3.6 g·L⁻¹, and the corresponding enzymatic hydrolysis rate was 45.33% which was 2.3 times higher than that of non-pretreated from Morus alba residues. Fed-batch enzymatic hydrolysis was conducted finally to get higher sugar yield, and the final glucose concentration reached up to 38 g·L⁻¹ with the enzymatic hydrolysis rate of 36.19%. The results indicated that Mori Cortex residue had higher cellulose and hemicellulose contents, so it had the potential to become a carbon source to produce the bio-chemicals and biofuels. Through enzymatic hydrolysis, it can be converted into microbial available monosaccharides; and through fermentation, it can be converted into high value-added chemicals, biofuels, etc., to solve the problem of residue pollution, and achieve the sustainable development and greening of Chinese pharmaceutical production process.

A dumbbell-shaped fiber laser with dual-functional pulsed outputs is demonstrated using a filmy polyvinyl alcohol-based gold nanorods (GNRs) saturable absorber (SA), which can deliver mode-locked soliton and multiwavelength Q-switched pulse. The GNRs-SA is fabricated by seed-mediated method with a modulation depth of 4.1% and nonsaturable loss of 32.9%. When the fiber laser operates in mode-locked regime, stable fundamental solitons with a temporal width of 910 fs and repetition rate of 2.88 MHz are directly generated from the laser cavity. The corresponding spectrum is located at 1560.1 nm with a 3-dB bandwidth of 3.08 nm. When the laser operates in Q-switched regime, more than 5-wavelength oscillation is achieved. The repetition rate of Q-switched monotonically increases from 13.4 to 31.7 kHz and the pulse duration can be significantly narrowed from 6.07 to 3.12 μs by varying the pump power from 128 to 154 mW. The results further expand the applications of GNRs-SA to obtain versatile pulse sources in ultrafast laser field.

The ability ofNocardiasp. 108 to hydrate indole-3-acetonitrile to indole-3-acetamidewas identified. Cell culture showed thatNocardiasp. 108 acrylonitrile-hydrating activity reached its maximum, 1209.8 U/g DCW, after cultivated at 28 8C for 45 h. Under the biotransformation conditions of 28 8C and pH 7.5, the maximal indole-3-acetonitrile conversion ratio and indole-3-acetamide yield were 34.34 and 32.68% (w/w), respectively. Moreover, the Km and Vmax values for the indole-3-acetonitrile biotransformation catalyzed by Nocardia sp. 108 were 1.18 � 10 � 2 mol/l and 1.16 � 10 � 3 mol/l min. Ethanol supplemented to the reaction system dramatically enhanced indole-3-acetonitrile aqueous solubility. Furthermore, 4% (v/v) ethyl acetate raised the indole-3-acetonitrile bioconversion capacity 78%. # 2006 Elsevier Ltd. All rights reserved.

Nocardia sp. 108 exhibited strong acrylonitrile-hydrating activity and its nitrile hydratase was Co2+-dependent. Nocardia sp. 108 was active within a broad pH range from 6.0 to 10.0 at 30°C and thermostable at temperatures below 35°C, but became unstable at temperatures above 45°C. Furthermore, it was found that Nocardia sp. 108 can hydrate indole-3-acetonitrile, p-chlorobenzonitrile, p-hydroxybenzylcyanide, 3,4,5-trimethoxybenzonitrile, p-aminobenzonitrile, 3-cyanopyridine, o-chlorobenzonitrile to the corresponding amides and hence displayed a broad substrate specificity. The temperature and pH optima for these hydrations were 28°C and pH 7.0–7.5, respectively. At the observed concentrations, acrylonitrile was completely converted within 5 min, while 3,4,5-trimethoxybenzonitrile, p-aminobenzonitrile, indole-3-acetonitrile, p-chlorobenzonitrile were approximately 21.71, 8.98, 34.44, 93.10% hydrated. p-Chlorobenzonitrile appeared to be the preferred aromatic nitrile for Nocardia sp. 108.

We presented a bound-state operation in a fiber laser with near-zero anomalous dispersion based on a silica-coated gold nanorods (GNRs@SiO2) saturable absorber (SA). Using a balanced twin detector measurement technique, the modulation depth and nonsaturable loss of the GNRs@SiO2 SA were measured to be approximately 3.5% and 39.3%, respectively. By virtue of the highly nonlinear effect of the GNRs@SiO2 SA, the bound-state pulses could be easily observed. Besides the lower-order bound-state pulses with two, three, and four solitons, the higher-order bound states with up to 12 solitons were also obtained in the laser cavity. The pulse profiles of the higher-order bound states were further reconstructed theoretically. The experimental results would give further insight towards understanding the complex nonlinear dynamics of bound-state pulses in fiber lasers.

To study the effect of sucrose and plant growth substances of different concentrations on the induction of test-tube tuberous roots of Rehmannia glutinosa, in order to establish an efficient system for the induction of test-tube tuberous roots from leaves of R. glutinosa.Leaves from test-tube seedlings of 85-5 R. glutinosa were used as explants. After rooting induction, they were transferred to medium with orthogonal design for inducing test-tube tuberous roots of R. glutinosa.NAA played a significant role in induction of test-tube tuberous roots of R. glutinosa, followed by sucrose and 6-BA. With leaves from test-tube seedlings as the explants, the optimal medium for inducing test-tube tuberous roots of R. glutinosa was MS + BA 3.0 mg x L(-1) + NAA 0.1 mg x L(-1) + sucrose 7%. The study provides an efficient induction system for studies on artificial seeds and secondary metabolism with test-tube tuberous roots of R. glutinosa.

To establish an efficient genetic transformation system of Pinellia ternata.With petioles from test-tube seedlings of P. ternata as explants, Agrobacterium tumefaciens mediation method was adopted to explore the effect of phenolic substances, A. tumefaciens's concentration, infection time, pre-incubation time and co-cultivation time on genetic transformation efficiency of P. ternata.The genetic transformation efficiency could be effectively enhanced by infecting in A. tumefaciens culture containing AS 40 mg x L(-1) for 15 min for three days. The petioles were put into the differentiation medium containing 150 mg x L(-1) Kan and 350 mg x L(-1) Carb to screening and cultivation. After around 30 days, microtubers could be observed at both sides of the petioles. Gus staining and PCR verification on the regenerated plants showed that the exogenous gene sHSP had been integrated into genome of P. ternata.

To investigate the optimization system of SRAP-PCR molecular marker technology in the analysis on Pinellia ternata.SRAP-PCR reaction system for P. ternata was optimized by L16 (5(4)) orthogonal design with five elements (dNTPs, Mg2+, the template DNA, primers, Taq enzyme) and four standards.The most suitable forward primer for SRAP for Pinellia ternata was 5'-TGAGTCCAAACCGGAAG-3', while the reverse primer was 5'-GACTGCGTACGAATTACG-3'. The optimized reaction system contained 70 ng DNA template, 0.9 micromol x L(-1) primer, 0.20 mmol x L(-1) dNTP s, 1.5 - 2.0 mmol x L(-1) Mg2+, and 2 U Taq enzyme.SRAP-PCR system for P. ternata is established to lay a foundation for future construction of SRAP genetic map of P. ternata.

To study the effects of shading on photosynthetic physiology and chlorophyll fluorescence of Pinellia ternata.Plant growth, chlorophyll content, net photosynthetic rate (P(n)) and chlorophyll fluorescence in P. ternata were investigated under different shading treatments (0%, 70% and 90%) when it grew about 15 cm high.The results showed that fresh weight of a tuber, height, leaf length, width, leaf area, specific leaf area (SLA) and contents of chlorophyll content were enhanced after shaded, and chlorophyll a/b rate declined. Compared with control, net photosynthetic rate, light compensation point (LCP) and light saturation point (LSP) of P. ternata decreased after shading, but apparent quantum yield (AQY) increased; quantum yield of PS II (PhiPS II), minimal fluorescence (F(o)), maximal fluorescence (F(m)), intrinsic photochemical efficiency of PS II (F(v)/F(m)) and photochemical quenching coefficient (qP) were enhanced.Compared with control, all data indicated that there were distinctive differences between the height, SLA, chlorophyll content, P(n) and chlorophyll fluorescence characteristics under the shading treatments (70% and 90%), the fresh weight of a tuber increased after 70% shading, and provided better environmental conditions for the growth of P. ternata.

To study the optimal condition for bulb induction of Fritillaria anhuiensis in vitro.Effects of sucrose, salicylic acid, active carbon and 5 degrees C pre-incubation on bulb formation in vitro were investigated by adopting the number of bulb and fresh weight as indexes.The number of bulb and fresh weight in medium added with 50 g x L(-1) sucrose were significantly higher than those with other treatments. The medium with different concentrations of salicylic acid showed no significant promotion on callus differentiation. However, 5 g x L(-1) active carbon (AC) treatment was better than other treatments. Meanwhile, a large number of plantlets formed after 5 degrees C pre-incubation for 30-40 days was suitable for bulb formation and growth.Bulblet formation was optimal in MS + KT 2 mg x L(-1) + NAA 2 mg x L(-1) + 50 g x L(-1) sucrose +5 g x L(-1) AC after callus pre-incucation at 5 degrees 1 for 30-40 days.

In this study, orthogonal design was used to optinize DDRT-PCR amplification system on Pinellia ternata microtubers in vitro in five factors four levels respectively.P. ternata stems and microtubers in vitro were selected as explants. The effects of five kinds of factors were studied by orthogonal design method including emplate cDNA, Mg2+, dNTPs, primers and Taq DNA polymerase, and in order to establish the optimum DDRT-PCR system of P. ternata microtubers in vitro.A satisfactory DDRT-PCR technique system for P. ternata microtubers in vitro with desirable repeatability and polymorphic bands was established. In a total volume of 20 microL DDRT-PCR system, it contained 10 x buffer, 150 micromol L(-1) dNTPs, 2 micromol L(-1) anchor primer, 1 micromol L(-1) arbitrary primer, 2.5 mmol L(-1) Mg2+, 0.6 U Taq DNA polymerase and 2.5 microg template cDNA. The effect of the five factors was in sequence of Taq DNA polymerasetemplate cDNAdNTPsMg2+Primers. The optimum DDRT-PCR system will provide scientific reference basis for studying effecting character of P. ternata microtubers associated with genes expression.

To study the relationship between the condition of cell suspension culture and the polysaccharides content in Achyranthes bidentata.The methods of orthogonal test and alone factor test were used to study the effect of culture time, inoculation concentration, carbon source and pH.The results showed that the optimum medium to induce polysaccharides in A. bidentata was 1/4MS + 6-BA 0.5 + 2,4-D 1.0 mg x L(-1) + sucrose 2% + glucose 2% + fructose 1% cultured after 50 d. The suitable pH was 6.0, and the optimum inoculation concentration was 7g x L(-1).

To study the effects of different factors on buds and microtuber. These factors included plant growth substances and sucrose.stems were selected as explants. The effects of three kinds of factors were studied by orthogonal design method including sucrose, 6-BA, NAA on the buds and microtuber induction. The data were analyzed with range analysis and vadance analysis. RESULT AND CONDUSION: The optimal media to induce many buds from stems were MS + 6-BA 1 mg x L(-1) + NAA 1 mg x L(-1) + sucrose 3%, the effect of the three factors was in sequence of sucrose6-BANAA. The optimal media to induce microtuber from stems were MS+6-BA 1.5 mg x V1 +NAA 1.5 mg x L(-1) + sucrose 5%, the effect of the three factors was in sequence of sucrose6-BANAA.

To extract RNA from Pinellia ternata and lay a foundation for studying the formation mechanism of P. ternata.By modifying the method recommended by Guanidinium for extracting total RNA from plant tissues rich in phenolic and polysaccharidic compounds, a simple and convenient method for extraction of total RNA from the tubers, stems and leaves of P. ternate containing abundant polyphenols and polysaccharides was established. High concentrated p-mercaptoethanol was added in the RNA extracted buffer to remove polyphenols, phenol and chloroform were used to eliminate proteins, and isopropanol and sodium acetate were used to precipitate polysaccharides.The A260/A230 value of RNA extracted with improved method were all over 2.0 and the values of A260/A280 were between 1.7 and 2.0. The electrophoresis bands were cleared on agarosegel and integrity of RNA was good.The results showed that RNA obtained from the tubers, stems and leaves of P. ternate with this method had high purity and quality and could be used in molecular biological research, as DDRT-PCR and reverse Northern blotting analysis directly. This method is simple, economic, stable performance, and has a good repeatability as well as is suitable for extracting total RNA of medicinal plants with high concentrations of phenolics and polysaccharides.

To study the change of endogenous hormone (ABA, IAA, JA, GA3, ZR) in the leaves, petioles, tubers of Pinellia ternate around sprout tumble. It also provided some valuable information to prevent sprout tumble and increase production.Tubers of P. ternata were cultured firstly at (23 +/- 1) degree C for certain days, and then they were coerced under (30 +/- 1 ) degree C stress in the same artificial climate boxes. The endogenous hormones in leaves, petioles and tubers during different stages of high temperature stress were determined with Enzyme-linked Immunosorbent Assays (ELISA).After under high temperature stress, ABA content in leaves, petioles and tubers increased obviously. Similarly, JA content rose all in the leaves, petioles and tubers. But in the same conditions IAA content declined significantly in the leaves and petioles. In the tubers, IAA content also decreased, but not quickly. With the extension of high temperature coercion, the leaves, petioles, tubers, ZR content were gradually falling off. In the leaves of GA3 content rose markedly at the third day, fell down at the sixth day, but remained higher than before treatment. With the extension of the processing time, GA3 content fell off in the petioles and tubers.ABA, JA, ZT and GA3 played an important role in controlling sprout tumble of P. ternata.

To study the effect of different factors on induction of microtubers in Zingiber officinale. These factors included NAA, PP333, 6-BA and sucrose.Orthogonal design and plant tissue culture technique were used.Sucrose was the most important factor on the induction of microtubers, followed by PP333 and NAA. 6-BA was the factor which restrained the formation of microtubers. The optimal media to induce microtubers was MS + NAA 1.0 mg x L(-1) + PP333 0.2 mg x L(-1) + sucrose 8%.

To study the effects of salicylic acid of different concentration (SA) on the growth of Pinellia ternate.SA of different concentration was sprayed to the plant as it grew about 15 cm high. Plant height, total chlorophyll content, activity of SOD, MAD content, photosynthesis speed, intercellular CO2 concentration, the transpiration speed and the leaf temperature were measured after 3 times spray.When 0.5 mmol x L(-1) SA was sprayed, the intercellular CO2 concentration, activity of SOD and photosynthesis speed were increased, while leaf temperature and MDA content were decreased.SA at the concentration of 0.5 mmol x L (-1) was suitable for the growth of P. ternate. It obviously decreased sprout tumble and increased the fresh weight of tuber and the alkaloid content.

Ultrafast fiber lasers play a significant role in our society with many aspects ranging from fundamental physics to industrial purposes. Searching for high-performance saturable absorbers (SAs) is vital to the developments of ultrafast fiber lasers. Gold nanorods (GNRs) have been discovered to possess saturable absorption effect. However, a major obstacle to make the GNRs as high-performance and practical SA is the low optical damage threshold. To overcome this drawback, herein we proposed the nanocomposites with gold nanorods/silica core-shell structure (GNRs@SiO(2)) as a high-performance SA for ultrashort pulse generation in a fiber laser. The GNRs@SiO(2) SA presents a modulation depth of 4.2% and nonsaturable loss of 45.6%. With the proposed GNRs@SiO(2) SA, 379 fs pulse was directly obtained from the fiber laser. The achieved results demonstrated that the GNR@SiO(2) could be indeed a good candidate of high-performance SA towards practical applications in the field of ultrafast photonics.

To study the effects of potassium salts on direct induction of microtubers from different explants in Pinellia ternata.Leaves, petioles and tubers were cut and cultured on the medium with different kinds of potassium salts and plant growth regulators.Low concentration of potassium salts, which were lower than 4.02 mmol x L(-1), could promote the microtuber formation in vitro. While high concentration of potassium salts, which were more than 12.06 mmol x L(-1), inhibited the formation of microtubers.

To study the storage technique on artificial seeds of Pinellia ternata.Microtubers were used as experiment materials. By using orthogonal experiment, the sodium alginate, added with GA3 badistan, chitosan and sodium benzoate, was used as seed vessel. The artificial seeds were stored respectively under 25 degrees C and 4 degrees C for 30 days, then, calculated the germination rate.The sodium alginate(3%) added with GA3(0.1 mg x L(-1)), sodium benzoate(0.2%), badistan(1.0%), ClO2(0.1%) and chitosan(0.2%), were used as artificial seed vessel. Stored under (25 +/- 1) degrees C for 30 days, and the germination rate was over 65%. The sodium alginate(3%), added with GA3(0.1 mg x L(-1)), sodium benzoate(0.2%), badistan(1.0%), ClO2(0.2%) and chitosan(0.1%), were used as artificial seed vessel. Stored under 4 degrees C for 30 days, and the germination rate was over 80%.

This is a review of the advantages of polyploid in medicinal plants, their applications and identification. The common methods for the induction of polyploidy are summarized, and the problems to be dealt with are discussed.

To study the effects of different factors on direct induction of microtubers. Optimal media to directly induce microtubers from tubers were MS + 6-BA 1.0 mg x L(-1) + PEG 5% + sucrose 5% and MS+ CH 500 mg x L(-1) + MET 0.5 mg x L(-1) + 6-BA 0.5 mg x L(-1) + AC 0.5% + sucrose 5%. Media suitable for plantlet growth were MS + 6-BA 0.5 mg x L(-1) + IAA 0.5 mg x L(-1) + AC 0.5% + sucrose 5% and MS + MET 0.5 mg x L(-1) + 6-BA 0.5 mg x L(-1) + sucrose 3%.

To set up the fittest medium for directly inducing regenerated plantlet from leaf of Chrysanthemum morifolium.Leaves of different phases obtained from the virus-free plantlets were cut and cultured in some kinds of media.To Bo morifolium and Chu morifolium, the fittest medium for the formation of regenerated plantlet was MS + 6-BA 0.1 mg x L(-1) + NAA 0.1 mg x L(-1), and to be incubated on the right side was better than on the back right.

To study the morphogenesis in formative process of tuberous root in vitro from Rehmannia glutinosa and compare the anatomical shape of tuberous root with nature term R. glutinosa.Tuberous roots of different vegetal phase were cut and dyed, then made into paraffin cuts and observed microscope.In anatomical shape, nature R. glutinosa and tuberous root were the same, which showed that no structural variation occurred in tuberous root induced process.

To study the technique on artificial seeds of Pinellia ternata.Microtubers were used as materials. Sodium alginate or chitosan, with GA3, penicillin, streptomycin sulphate, sodium benzoate, badistan, CuCl2, NiCl and CoCl2 added to were used as seed vessel. The seeds were cultured respectively under artificial conditions and natural conditions.The sodium alginate (3%) and chitosan (2%) added with GA3 (0.1 mg x L(-1)), badistan (0.3%), sodium benzoate (0.2%), Ca2+ (0.05%), Ni+ (0.05%) added to, were used as artificial seed vessel, and the germination rate was over 95% and the seedling rate was over 90%.

To compare and explore the physiological and biochemical index of virus-free seedling and common seedling of Chrysanthemum morifolium.Leaves of virus-free seedling and common seedling were cut down and the contents of chlorophyll a and b, MDA, and the activity of SOD, POD, CAT, and the net photosynthetic rate (Pn) were measured.The contents of chlorophyll and Pn of virus-free seedling were higher than those of common seedling. But were measured, the contents of MDA and the activity of SOD, POD, CAT related to the physiological resistance were lower than the latter. These could explain the phenomenon of virus-free seedling grow better than the common seedling under the same condition.

To set up the optimums for the petal tissue culture of Chrysanthemum morifolium cultivated in Anhui Province.Petals were cut and cultured onto the same kind of media on different sides and different kinds of media on the same side, and induced to form the whole plantlet.All the media used could induce callus, while the results of callus regeneration was distinct. The MS medium with KT 2 mg x L(-1) and NAA 0.2 mg x L(-1) was much better than others when callus were induced. The MS medium with KT 2 mg x L(-1) + NAA 0.2 mg x L(-1) and AgNO3 5 mg x L(-1) was the preferable medium for the bud sprouting. The frequency of plantlets regeneration which was induced by flower bud was higher than that by opening petal. The regenerated plantlets shod produced variation of leaf and plant shape.

To select the sodium chloride tolerant mutants of the Chrysanthemum morifolium callus through tissue culture and EMS (ethylmethane sulfonate) treatment.Calli were induced from the leaves of C. morifolium. The calli were treated with 0.2% and 0.5% EMS, respectively. After 15 days' culture, the calli were transplanted to selection media with 0.5%, 1.0% and 1.5% NaCl, and the sodium chloride tolerant mutants were selected out.After tested the sodium chloride tolerant stability, the callus selected are found to be the mutants indeed.

To study the change of the activity of protective enzyme to avoid sprout tumble of Pinellia ternata.A high temperature stress was given to P. ternata. Then the activity of protective enzyme (SOD, POD and CAT) and the content of membrane lipid peroxidation (MDA) were detected after different treating days.By stress time increasing, the content of MDA in lamina and petiole increased obviously, but it increased invisibly in tuber. After high temperature stress, the activity of both SOD and CAT decreased gradually. The activity of POD increased at first but then decreased rapidly.

To set up the optimums of tissue culture for Cornus officinalis.A section of stems of the fine varieties of Cornus officinalis were strictly sterilized, cultivated and geminated in medium with addition of different kinds of hormone.The WPM medium with 6-BA 2.0 mg.L-1 + ZT 0.1 mg.L-1 + NAA 0.1 mg.L-1 was the optimum medium for the multiplication of adventitious buds. The WPM medium with 6-BA 1.0 mg.L-1 + ZT 0.1 mg.L-1 + GA3 0.5 mg.L-1 + NAA 0.1 mg.L-1 was the optimum medium for cultivation in strength. The plantlet could root well in 1/2 MS medium with IBA 1.0 mg.L-1 + 6-BA 0.1 mg.L-1.

To set up the fittest medium and optimum condition for leaves from Rehmannia glutinosa to form regenerated plantlets directly and to induce microtubers.Leaves of different phases obtained from the virus-free plantlets were incubated in some kinds of media on the back side.The fittest medium for the formation of regenerated plantlet was MS + 6-BA 2.0 mg.L-1 and the fittest medium for the induction of microtuber was MS + 6-BA 1.0 mg.L-1 + NAA 0.5 mg.L-1. The optimum incubation temperaue was 25 degrees C and the light length was (2,000-3,000 lx) 12 h.d-1.

To set up the optimums for the stem-tip tissue culture of Chrysanthemum morifolium cultivated in Anhui Province.Small sections (about 0.5 mm in length) from the stem-tips were isolated and inoculated with different media, and induced to form the whole plantlet formation.The MS medium added with 6-BA 2 mg.L-1 + NAA 0.2 mg.L-1 was the optimum medium for the bud sprouting and the inducing rate was over 80% after 40 d cultivation on this modified medium. The MS medium supplemented with 6-BA 2 mg.L-1 and NAA 0.5 mg.L-1 was the optimum medium for the multiplication of the adventitious buds in which the bud multiplication was about 4-7 times higher after 25-30 d cultivation. The plantlet could root well on the MS medium with NAA 0.5 mg.L-1.
