Abstract

Abstract Iron is an essential mineral that supports numerous biological functions. Studies have reported associations between iron dysregulation and certain cardiovascular and neurodegenerative diseases, but the direction of influence is not clear. Our goal was to use computational approaches to better understand the role of genetically predicted iron levels on disease risk. We meta-analyzed genome-wide association study summary statistics for serum iron levels from two cohorts and two previous meta-analyses. We then obtained summary statistics from 11 neurodegenerative, cerebrovascular, cardiovascular or lipid traits to assess global and regional genetic correlation between iron levels and these traits. We used two-sample Mendelian randomization (MR) to estimate causal effects. Sex-stratified analyses were also carried out to identify effects potentially differing by sex. Overall, we identified three significant global correlations between iron levels and (i) coronary heart disease, (ii) triglycerides, and (iii) high-density lipoprotein (HDL) cholesterol levels. A total of 194 genomic regions had significant (after correction for multiple testing) local correlations between iron levels and the 11 tested traits. MR analysis revealed two potential causal relationships, between genetically predicted iron levels and (i) total cholesterol or (ii) non-HDL cholesterol. Sex-stratified analyses suggested a potential protective effect of iron levels on Parkinson’s disease risk in females, but not in males. Our results will contribute to a better understanding of the genetic basis underlying iron in cardiovascular and neurological health in aging, and to the eventual identification of new preventive interventions or therapeutic avenues for diseases which affect women and men worldwide.

Abstract

Abstract We assess performance and limitations of polygenic risk scores (PRSs) for multiple blood pressure (BP) phenotypes in diverse population groups. We compare “clumping-and-thresholding” (PRSice2) and LD-based (LDPred2) methods to construct PRSs from each of multiple GWAS, as well as multi-PRS approaches that sum PRSs with and without weights, including PRS-CSx. We use datasets from the MGB Biobank, TOPMed study, UK biobank, and from All of Us to train, assess, and validate PRSs in groups defined by self-reported race/ethnic background (Asian, Black, Hispanic/Latino, and White). For both SBP and DBP, the PRS-CSx based PRS, constructed as a weighted sum of PRSs developed from multiple independent GWAS, perform best across all race/ethnic backgrounds. Stratified analysis in All of Us shows that PRSs are better predictive of BP in females compared to males, individuals without obesity, and middle-aged (40-60 years) compared to older and younger individuals.

Abstract

Summary: Background: Identifying novel epigenetic signatures associated with serum immunoglobulin E (IgE) may improve our understanding of molecular mechanisms underlying asthma and IgE-mediated diseases. Methods: We performed an epigenome-wide association study using whole blood from Framingham Heart Study (FHS; n = 3,471, 46% females) participants and validated results using the Childhood Asthma Management Program (CAMP; n = 674, 39% females) and the Genetic Epidemiology of Asthma in Costa Rica Study (CRA; n = 787, 41% females). Using the closest gene to each IgE-associated CpG, we highlighted biologically plausible pathways underlying IgE regulation and analyzed the transcription patterns linked to IgE-associated CpGs (expression quantitative trait methylation loci; eQTMs). Using prior UK Biobank summary data from genome-wide association studies of asthma and allergy, we performed Mendelian randomization (MR) for causal inference testing using the IgE-associated CpGs from FHS with methylation quantitative trait loci (mQTLs) as instrumental variables. Findings: We identified 490 statistically significant differentially methylated CpGs associated with IgE in FHS, of which 193 (39.3%) replicated in CAMP and CRA (FDR < 0.05). Gene ontology analysis revealed enrichment in pathways related to transcription factor binding, asthma, and other immunological processes. eQTM analysis identified 124 cis-eQTMs for 106 expressed genes (FDR < 0.05). Abstract

Background: Plasma proteins are critical mediators of cardiovascular processes and are the targets of many drugs. Previous efforts to characterize the genetic architecture of the plasma proteome have been limited by a focus on individuals of European descent and leveraged genotyping arrays and imputation. Here we describe whole genome sequence analysis of the plasma proteome in individuals with greater African ancestry, increasing our power to identify novel genetic determinants. Methods: Proteomic profiling of 1301 proteins was performed in 1852 Black adults from the Jackson Heart Study using aptamer-based proteomics (SomaScan). Whole genome sequencing association analysis was ascertained for all variants with minor allele count ≥5. Results were validated using an alternative, antibody-based, proteomic platform (Olink) as well as replicated in the Multi-Ethnic Study of Atherosclerosis and the HERITAGE Family Study (Health, Risk Factors, Exercise Training and Genetics). Results: We identify 569 genetic associations between 479 proteins and 438 unique genetic regions at a Bonferroni-adjusted significance level of 3.8×10 -11 . These associations include 114 novel locus-protein relationships and an additional 217 novel sentinel variant-protein relationships. Novel cardiovascular findings include new protein associations at the APOE gene locus including ZAP70 (sentinel single nucleotide polymorphism [SNP] rs7412-T, β=0.61±0.05, P =3.27×10 -30 ) and MMP-3 (β=-0.60±0.05, P =1.67×10 -32 ), as well as a completely novel pleiotropic locus at the HPX gene, associated with 9 proteins. Further, the associations suggest new mechanisms of genetically mediated cardiovascular disease linked to African ancestry; we identify a novel association between variants linked to APOL1-associated chronic kidney and heart disease and the protein CKAP2 (rs73885319-G, β=0.34±0.04, P =1.34×10 -17 ) as well as an association between ATTR amyloidosis and RBP4 levels in community-dwelling individuals without heart failure. Conclusions: Taken together, these results provide evidence for the functional importance of variants in non-European populations, and suggest new biological mechanisms for ancestry-specific determinants of lipids, coagulation, and myocardial function.

Abstract

Variability in SARS-CoV-2 susceptibility and COVID-19 disease severity between individuals is partly due to genetic factors. Here, we identify 4 genomic loci with suggestive associations for SARS-CoV-2 susceptibility and 19 for COVID-19 disease severity. Four of these 23 loci likely have an ethnicity-specific component. Genome-wide association study (GWAS) signals in 11 loci colocalize with expression quantitative trait loci (eQTLs) associated with the expression of 20 genes in 62 tissues/cell types (range: 1:43 tissues/gene), including lung, brain, heart, muscle, and skin as well as the digestive system and immune system. We perform genetic fine mapping to compute 99% credible SNP sets, which identify 10 GWAS loci that have eight or fewer SNPs in the credible set, including three loci with one single likely causal SNP. Our study suggests that the diverse symptoms and disease severity of COVID-19 observed between individuals is associated with variants across the genome, affecting gene expression levels in a wide variety of tissue types., Graphical abstract, D’Antonio et al. characterize associations between GWAS signals for COVID-19 disease and eQTLs in 69 human tissues to identify causal variants and their underlying molecular mechanisms. They show that diverse symptoms and disease severity of COVID-19 are associated with variants affecting gene expression in a wide variety of tissues.

Abstract

In view of the insufficient heating capacity of a supercritical 600 WM unit, it is necessary to determine whether the equipment of the extraction and return heat pipe system can operate normally after the transformation, so as to check the adaptability of the extraction and return heat pipe system equipment of the heating unit. Based on the actual situation and heat balance before and after the transformation of the heating system, this study selects the extracting steam and heat return pipe system of the heating units at all levels, and makes adaptive accounting from the design parameters of the pipeline, and the calculations of steam flow rates or pipe diameters of the pipeline, which provide scientific basis and evaluation for the feasibility of the retrofit scheme.

Abstract

The first phase of a power plant has two subcritical 300 MW units, and each boiler feed water pump is equipped with two with 50% capacity steam driven feed pumps and one 50% capacity electric start-up feed pump. The two steam pumps are put into operation during normal operation. After the heat supply transformation in the unit of the first phase of this power plant and based on the calculation and analysis of the heat balance of the unit, it is found that there will be insufficient output of the feed pump due to the insufficient steam source after the flow-through transformation, which will affect the water supply of the unit and reduce the energy utilization rate. To avoid this, the steam source of feed pump turbine needs to be selected again. Taking the original steam turbine condition of feed pump and the original steam source of feed pump turbine as the research basis, this paper makes analysis on the steam source transformation, and concludes the scheme of steam source selection and reconstruction based on energy matching. The feasibility of the scheme is tested and verified finally.

Abstract

This paper presents the design of a low gain-error pseudo rail-to-rail indirect current feedback instrumentation amplifier (In-Amp). In order to reduce the gain error introduced by the input common mode voltage variations, MOS transconductors regulated by specially designed folded-cascoded amplifier are used. Simulated gain error is only about 0.2% with input common mode voltage ranging from half VDD to VDD.

Abstract

The substrate noise becomes more serious with the increasing frequency and decreasing process geometries. In this paper, a mathematics model of the substrate noise will be proposed. The current methods for solving the substrate noise will be analyzed using the model, and a new method is proposed based on the inverse relationship between the varying voltage of the power supply line and the one of ground line. In this method, there are two important units, which are high-pass filter and adder circuit. The noise is reduced by 26dB, compared with triple-well techniques.

Abstract

A Dual-Output voltage bandgap reference has been designed for high-accuracy pipelined ADC with CSMC0.6 process technology in this work. Based on the temperature and noise analyzing of voltage reference, the traditional circuit has been improved. The stucture with two PN junctions in series,non-operational amplifier stucture and PTAT (Proportional To Absolute Temperature) current supplied by “self-bias” cascode current mirror, were adopted in the circuit designed. A new method to divide voltage was suggested in this study. The simulation results showed that the dual-output voltages were 2.194V and 1.098V respectively when temperature varied from −10°C to 100°C,and the temperature coefficient is only 7.00ppm/°C;The change of V ref only was 5.326mV, and the PSRR was 49.3dB when source voltage varied from 6V to 9V.The Dual-Output Voltage Reference of the circuit designed can meet the requirement of comparator input voltage in high-accuracy pipelined ADC.

Abstract

The EMI/EMC problems appear in mixed-signal ICs with the increasing frequency and decreasing process geometries. In this paper, the amplitude modulation (AM) is taken into account the LC-VCO based on the TSMC 0.25um mixed-signal process, and a cascade amplifier circuit, which is not complex, is proposed to optimize the LC-VCO through analyzing the noise coupling transition function of the LC-VCO. Lastly, the efficiency and usefulness of the proposed method is proved in the simulations.

Abstract

Illustrated by the case of a lithium-ion battery protection IC, the paper focuses on the design of internal immunity to RFI. With analysis of the chip’s three major elements of electromagnetic compatibility (EMC), the qualitative and quasi-quantitative analysis results of RFI influence to the chip are given out. By using a simple filter circuit and available material physical construction which can isolate, absorb and consume the RFI energy, the protection IC’s electromagnetic susceptibility has been reduced effectively. The simulation of the devised structure in the time domain is gained by Winspice, and tool IC_EMC makes it possible of the conversion from time domain to frequency domain in which the spectrum analysis is completed. The design has passed the simulation verification and the layout implement of the devised construction designed is also available.
