The aim of this study was to evaluate whether the indicator amino acid oxidation (IAAO) method could be applied in the domestic cat. Six adult male cats were used in a replicated 3 × 3 Latin square design. Three semi-synthetic diets were developed: a methionine (Met) and total sulfur AA (TSAA) deficient diet (T-BASAL; 0.24% Met+Cys − DM basis) and two Met and TSAA-sufficient diets in which either dl-Met (T-DLM) or 2-hydroxy-4-(methylthio)-butanoic acid (T-MHA) were supplemented, respectively, on an equimolar basis to meet the TSAA requirement (0.34%). After a 2-d diet adaptation, IAAO studies were performed. Cats were offered 13 small meals. The sixth meal contained a priming dose (4.8 mg/kg-BW) of l-[1-13C]-Phe and the remaining meals a constant dose (1.04 mg/kg-BW). Breath samples were collected every 25 min to measure 13CO2 enrichment. The following morning, fasted blood samples were collected. Cats returned to the T-BASAL top dressed with a dl-Met solution for 4 d prior to being fed a new dietary treatment. Isotopic steady state was evaluated through visual inspection. Data were analyzed using PROC GLIMMIX procedure in SAS 9.4. While 13CO2 enrichment was successfully captured in breath samples, cats failed to reach 13CO2 steady state. Thus, a non-steady state isotope model was developed and coded in ACSLX (V3.1.4.2) individually for each cat on each study day to predict 13CO2 enrichment, and then, calculate oxidation of l-[1-13C]-Phe (F13CO2). A higher predicted F13CO2 was observed for cats fed T-BASAL compared to the others (P < 0.05), while no differences were observed between T-DLM and T-MHA (P > 0.05). Cats fed T-DLM tended to have higher plasma Met concentrations compared to those fed T-BASAL with cats fed T-MHA intermediate (P = 0.0867). Plasma homocysteine concentrations were higher in cats fed T-BASAL compared to the others (P < 0.05), while threonine concentrations tended to be higher in cats fed T-BASAL compared to those fed T-MHA (P = 0.0750). In conclusion, short-term provision of a semi-synthetic diet deficient in Met may elicit a metabolic response aiming to conserve Met. The successful quantification of 13CO2 enrichment in breath and the higher predicted F13CO2 in cats fed a Met deficient diet suggest that the IAAO technique may be used in cats. Adaptations in the isotope protocol should be made to achieve 13CO2 steady state in breath and avoid mathematical modeling to predict F13CO2.

Milk replacers (MR) formulated to contain more lactose than whole milk could potentially reduce insulin sensitivity in dairy calves. This study evaluated how partially replacing lactose in MR with fat on a weight/weight basis affects glucose-insulin kinetics in neonatal dairy calves fed high quantities of MR. Thirty-four Holstein bull calves were blocked by dam parity (1.6 ± 0.3) and randomly assigned to 1 of 2 treatments (n = 17): a high-lactose MR (HL; 46.1% lactose, 18.0% crude fat, and 23.9% crude protein on a dry matter basis) or a high fat MR (HF; 39.9% lactose, 24.6% crude fat, and 24.0% crude protein on a dry matter basis). Calves were individually housed and fed pooled colostrum at 1.5 h and 12 h postnatal at 18 and 9% of metabolic body weight (BW0.75), respectively. From 24 h postnatal until the end of the study (d 7), calves were transitioned to MR (prepared at 150 g/L) at 18% of BW0.75 twice daily at 0700 and 1900 h. On d 4, an insulin-modified intravenous glucose tolerance test was conducted 8 h after the morning meal by intravenous infusion of glucose (0.54 g/kg of BW0.75, min 0) and insulin (3.9 µg/kg of BW0.75, min 20). During postprandial sampling on d 6, abomasal emptying was assessed by dosing acetaminophen (0.13 g/kg of BW0.75) with MR and measuring its appearance within plasma. Sequential blood samples were collected and analyzed for plasma glucose, insulin, and acetaminophen concentrations. Calves were killed on d 7 and liver and pancreatic tissues were collected for histomorphological analysis. Partially replacing lactose in MR with fat increased body weight gain (679.6 vs. 462.0 ± 69.68 g/d) and gain:metabolizable energy intake (186.6 vs. 120.5 ± 20.21 g of body weight/Mcal) compared with feeding HL MR. Furthermore, abomasal emptying rate was reduced in calves fed HF MR. Calculated variables based on postprandial glucose and insulin concentrations, such as maximum concentration or area under the curve, were or tended to be lower in HF than in HL calves. Postprandial insulin sensitivity tended to be reduced in HL compared with HF, although insulin sensitivity estimated from the insulin-modified intravenous glucose tolerance test was not different. Calves fed HF MR showed enhanced glucose-stimulated insulin secretion. In conclusion, partially replacing lactose in MR with fat resulted in smaller fluctuations in postprandial glucose and insulin concentrations and tended to increase postprandial but not fasting insulin sensitivity in neonatal dairy calves.

A variety of measures are currently used to evaluate feed efficiency in livestock, with some of the most popular being the linear regression of nutrient intake on the various energy sinks of the cohort being evaluated. The adoption of novel on-farm technologies has greatly increased the breadth and depth of data being collected on a daily basis, which creates the opportunity for the implementation of more advanced feed efficiency models. The objectives of the current study were to leverage data collected using high-throughput biometric technologies to evaluate changes of early-lactation feed efficiency on a daily basis and to develop a novel measure of feed efficiency based on the NRC net energy model. Daily records of nutrient intake, fat- and protein-corrected milk yield, body weight, and body condition score were used to model 7 different indices of feed efficiency (gross feed efficiency, return over feed costs, residual feed intake, residual net energy intake, and net energy efficiencies of lactation, maintenance, and body reserve flux) in primiparous Holstein heifers (n = 40) from 0 to 150 d in milk. We observed that feed efficiency was dynamic and fluctuated over the course of the experimental period regardless of the efficiency measure used, but most measures followed similar trends in changes of efficiency on a day-to-day basis. Of the measures evaluated, return over feed costs would be most suited for management purposes due to practicality and simplicity, whereas other measures, such as the efficiency of net energy flux to and from body reserves, present potential targets for inclusion in breeding goals. Net energy efficiency of lactation was estimated to have higher repeatability compared with residual feed or energy intake, which may reduce the time required to estimate whole-lactation efficiency. Because decreased net energy balance was strongly correlated with increased efficiency for all indices evaluated, future work investigating the associations of feed efficiency with the magnitude and duration of negative energy balance is warranted.

Multiparous, lactating Holstein cows (n = 32) were randomly assigned to one of two dietary treatments [TMR with rumen-protected Met (RPM) or TMR without RPM (CON)], and within each dietary treatment group cows were randomly assigned to one of two environmental treatment groups in a split-plot crossover design. In phase 1 (9 d), all cows were fed ad libitum and in thermoneutral conditions (TN). In phase 2 (9 d), group 1 (n = 16) was exposed to a heat stress (HS) challenge (HSC). Group 2 cows (n = 16) were pair-fed (PFTN) to HSC counterparts and remained in TN. After a 21-d washout period, the study was repeated (period 2) and the environmental treatments were inverted relative to treatments from phase 2 of period 1, while dietary treatments remained the same for each cow. During phase 1, cows in RPM had greater plasma Met concentration compared with cows in CON (59 and 30 µM, respectively; P < 0.001). Cows in PFTN had a greater decrease (P < 0.05) in plasma insulin than cows in HSC at 4 h (−2.7 µIU/mL vs. −0.7 µIU/mL) and 8 h (−7.7 µIU/mL vs. −0.4 µIU/mL) during phase 2. Compared with cows in PFTN, cows in HSC had an increase (P < 0.05) in plasma serum amyloid A (−59 µg/mL vs. +58 µg/mL), serum haptoglobin (−3 µg/mL vs. +33 µg/mL), plasma lipopolysaccharide binding protein (−0.27 and +0.11 µg/mL), and plasma interleukin-1β (−1.9 and +3.9 pg/mL) during phase 2. In conclusion, HSC elicited immunometabolic alterations; however, there were limited effects of RPM on cows in HSC.

Simple Summary Gastric emptying is the release of nutrients from the stomach into the small intestine. The rate at which gastric emptying occurs may be associated with diabetes and obesity risk in humans and could help prevent weight gain in dogs. The largest portion of carbohydrates in pet diets is provided by various starches that are digested and absorbed at different rates. This study investigated the effects of common starch ingredients found in commercial dog foods on the gastric emptying rate in dogs. Dogs received each test diet once (4 total) and a glucose control twice in a randomized order, along with acetaminophen. Blood samples were taken once prior to meal consumption and at multiple time points after to determine acetaminophen concentrations. A mathematical model was used to estimate the rate of gastric emptying using postprandial acetaminophen concentrations. Overall, more gastric emptying occurred at a faster rate in dogs when fed the diet containing the highest fraction of starch ingredients resistant to canine digestion. These findings suggest that the inclusion of different starch sources may be associated with altered digestion and absorption of nutrients, which consequently affects gastric emptying rate. The link between carbohydrate sources and gastric emptying may provide a mechanism to prevent weight gain in dogs. Abstract Gastric emptying rate (GER) may impact diabetes and obesity in humans and could provide a method to reduce canine weight gain. Starch, the most common source of carbohydrates (CHOs) in pet food, is classified as rapidly or slowly digestible, or resistant to digestion. This study investigated starch source effects in commercial extruded dog foods on the GER of 11 healthy adult Siberian Huskies. Test diets were classified as traditional, grain-free, whole-grain, and vegan. Dogs received each diet once, a glucose control twice, and acetaminophen (Ac) as a marker for GER in a randomized, partially replicated, 6 × 6 Latin square design. Pre- and post-prandial blood samples were collected at 16 timepoints from −15 to 480 min. Serum Ac concentrations were assessed via standard spectrophotometric assays and fitted with a mathematical model to estimate parameters of GER. Parameter values were subjected to ANOVA, with period and treatment as fixed effects and dog as a random effect. More total emptying (p = 0.074) occurred at a faster rate (p = 0.028) in dogs fed the grain-free diet, which contained the lowest total starch (34.03 ± 0.23%) and highest resistant starch (0.52 ± 0.007%). This research may benefit future diet formulations to reduce the prevalence of canine weight gain.

Estimating single nucleotide polymorphism (SNP) effects over time is essential to identify and validate candidate genes (or quantitative trait loci) associated with time-dependent variation of economically important traits and to better understand the underlying mechanisms of lactation biology. Therefore, in this study, we aimed to estimate time-dependent effects of SNP and identifying candidate genes associated with milk (MY), fat (FY), and protein (PY) yields, and somatic cell score (SCS) in the first 3 lactations of Canadian Ayrshire, Holstein, and Jersey breeds, as well as suggest their potential pattern of phenotypic effect over time. Random regression coefficients for the additive direct genetic effect were estimated for each animal using single-step genomic BLUP, based on 2 random regression models: one considering MY, FY, and PY in the first 3 lactations and the other considering SCS in the first 3 lactations. Thereafter, SNP solutions were obtained for random regression coefficients, which were used to estimate the SNP effects over time (from 5 to 305 d in lactation). The top 1% of SNP that showed a high magnitude of SNP effect in at least 1 d in lactation were selected as relevant SNP for further analyses of candidate genes, and clustered according to the trajectory of their SNP effects over time. The majority of SNP selected for MY, FY, and PY increased the magnitude of their effects over time, for all breeds. In contrast, for SCS, most selected SNP decreased the magnitude of their effects over time, especially for the Holstein and Jersey breeds. In general, we identified a different set of candidate genes for each breed, and similar genes were found across different lactations for the same trait in the same breed. For some of the candidate genes, the suggested pattern of phenotypic effect changed among lactations. Among the lactations, candidate genes (and their suggested phenotypic effect over time) identified for the second and third lactations were more similar to each other than for the first lactation. Well-known candidate genes with major effects on milk production traits presented different suggested patterns of phenotypic effect across breeds, traits, and lactations in which they were identified. The candidate genes identified in this study can be used as target genes in studies of gene expression.

We performed genome-wide association analyses for milk, fat, and protein yields and somatic cell score based on lactation stages in the first 3 parities of Canadian Ayrshire, Holstein, and Jersey cattle. The genome-wide association analyses were performed considering 3 different lactation stages for each trait and parity: from 5 to 95, from 96 to 215, and from 216 to 305 d in milk. Effects of single nucleotide polymorphisms (SNP) for each lactation stage, trait, parity, and breed were estimated by back-solving the direct breeding values estimated using the genomic best linear unbiased predictor and single-trait random regression test-day models containing only the fixed population average curve and the random genomic curves. To identify important genomic regions related to the analyzed lactation stages, traits, parities and breeds, moving windows (SNP-by-SNP) of 20 adjacent SNP explaining more than 0.30% of total genetic variance were selected for further analyses of candidate genes. A lower number of genomic windows with a relatively higher proportion of the explained genetic variance was found in the Holstein breed compared with the Ayrshire and Jersey breeds. Genomic regions associated with the analyzed traits were located on 12, 8, and 15 chromosomes for the Ayrshire, Holstein, and Jersey breeds, respectively. Especially for the Holstein breed, many of the identified candidate genes supported previous reports in the literature. However, well-known genes with major effects on milk production traits (e.g., diacylglycerol O-acyltransferase 1) showed contrasting results among lactation stages, traits, and parities of different breeds. Therefore, our results suggest evidence of differential sets of candidate genes underlying the phenotypic expression of the analyzed traits across breeds, parities, and lactation stages. Further functional studies are needed to validate our findings in independent populations.

Increasing evidence suggests that variability in hepatic glucuronidation is a critical determinant of mammalian species-specific sensitivity to the Fusarium mycotoxin deoxynivalenol (DON). Concurrently, ongoing work regarding the effects of DON on fish has led to broad classifications concerning species-specific sensitivity (e.g. rainbow trout are highly sensitive to DON and channel catfish are highly tolerant to DON). This study was designed to determine if sensitivity to DON in fish is related to inherent differences in UDP-glucuronosyltransferase (UDPGT) activity and to evaluate if increased dietary digestible starch content could be an effective strategy to increase glucuronidation capacity. Rainbow trout or Nile tilapia (initial average body weight = 21.9 and 8.1 g/fish, respectively) were fed two series (12 or 24% digestible starch) of three diets containing graded levels of naturally occurring DON (0.1, 0.7 and 1.3 ppm) for 10 weeks. Significant linear decreases in weight gain, thermal-unit growth coefficient (TGC, P ≤ .05), feed efficiency (P .05) with the exception of a quadratic increase in NRE (P .05, DON × digestible starch). A significant quadratic decrease (P ≤ .05) in UDPGT activity was associated with increasing levels of DON in rainbow trout fed the diet containing 12% digestible starch, whereas trout and tilapia fed the diet containing 24% digestible starch experienced significant quadratic or linear increases (P ≤ .05) in UDPGT activity, respectively. However, UDPGT activity was approximately 10-fold higher in rainbow trout than in Nile tilapia. This suggests that UDPGT activity is not responsible for the observed species-specific sensitivity to DON or it is not an appropriate biomarker of DON glucuronidation. A significantly higher relative fold expression of CYP1A in tilapia compared to trout (P = .0001) may offer potential opportunity for future study. We demonstrate for the first time that Nile tilapia are unaffected by practically relevant levels of DON which are detrimental to rainbow trout. This study also extends the current knowledge of DON metabolism and represents an important contribution to the development of efficacious nutritional mycotoxin mitigation strategies.

Hepatic glucuronidation represents an important phase II biotransformation reaction in both mammals and fish. The kinetic characteristics of uridine 5′-diphosphate (UDP) glucuronosyltransferases (UDPGTs) in rainbow trout liver microsomes were examined using p-nitrophenol (p-NP) as an aglycone and UDP-glucuronic acid (UDPGA) as a glucuronyl donor according to an existing protocol. The kinetic data obtained with varying concentrations of p-NP best fit the Hill equation and UDPGT activity was successfully induced following an i.p. injection of β-naphthoflavone (β-NF). The assay was subsequently adapted to a microplate method for determination of UDPGT activity in microsomal samples obtained from rainbow trout as well as Nile tilapia. In contrast to rainbow trout, UDPGT activity of Nile tilapia was best described by Michaelis-Menten kinetics. Based on the linearity of p-NP glucuronide formation, a p-NP concentration of 0.60 mM and a UDPGA concentration of 6.89 mM were determined to be suitable for assaying UDPGT activity in samples from rainbow trout and Nile tilapia. The microplate method offers several advantages over the historical assay; most notably it enables the observation of successive kinetics which ensures that enzyme activity is calculated in the most linear (initial) rate of the reaction. It also provides practical advantages in terms of ease-of-use and efficiency. This may be relevant to researchers investigating exposure of wild or farmed fish to environmental or feed-borne contaminants which are substrates of UDPGTs.

Dietary fiber affects canine physiology in many ways, such as increasing colonic absorption of water and improving gut health, both of which may positively impact exercise performance. The objectives of this study were to investigate the effects of increased dietary soluble fiber and incremental training on respiratory rate (RR), internal body temperature (BT), body composition, and fecal metabolites in mid-distance training sled dogs. Fourteen dogs (12 Siberian and 2 Alaskan Huskies) were blocked by age, sex, and body weight (BW) and then randomly allocated into one of two diet groups. Seven dogs were fed a dry extruded control diet (Ctl) with an insoluble:soluble fiber ratio of 4:1 (0.74% soluble fiber on a dry-matter basis), and seven dogs were fed a dry extruded treatment diet (Trt) with an insoluble:soluble fiber ratio of 3:1 (2.12% soluble fiber on a dry-matter basis). Fecal samples were taken once a week. All dogs underwent 9 weeks of incremental exercise conditioning where the running distance was designed to increase each week. Every 3 weeks, external telemetry equipment was used to non-invasively measure and record RR and internal BT at resting, working, and post-exercise recovery states. Body composition was measured on weeks −1 and 9 using quantitative magnetic resonance. Body composition, RR, BT, and fecal metabolites were analyzed using a mixed model with dog as a random effect and week and diet group as fixed effects. Dogs on Trt had lower working and post-exercise BT than Ctl (P < 0.05). In addition, Trt dogs had lower recovery BT at weeks 2 and 5 than Ctl dogs (P < 0.05). Treatment dogs had greater fecal short-chain fatty acid concentrations than Ctl (P < 0.05). Diet had no effect on RR or body composition (P > 0.10), but exercise resulted in an overall 7% increase in lean and 3.5% decrease in fat mass (P < 0.05). These data suggest that increasing dietary soluble fiber may positively influence BT and gut health; however, it has no effect on RR or body composition. Soluble fiber did not negatively impact any measures of overall health and performance and should be considered for use in performance dogs.

Genetic selection for improved feed efficiency in dairy cattle has received renewed attention over the last decade to address the needs of a growing global population. As milk yield is a critical component of feed efficiency metrics in dairy animals, our objective was to evaluate the associations between feed efficiency in primiparous Holstein cattle and parameters of a mathematical model describing individual lactation curves. The Dijkstra lactation curve model was fit to individual lactation records from 34 Holstein heifers with previously estimated measures of feed efficiency. We found that the optimal fit of the Dijkstra model was achieved using daily milk yield records up to 21 d in milk to capture the rise to peak milk yield and using monthly dairy herd improvement records for the remainder of lactation to accurately characterize lactation persistency. In the period of lactation before peak milk yield, improved feed efficiency was associated with a faster increase in daily milk yield over a shorter period of time at the expense of increased mobilization of body reserves; this serves to reinforce the concept that dairy cattle are primarily capital breeders versus income breeders. Feed efficiency in the period following peak lactation, as measured by gross feed efficiency, return over feed costs, and net energy efficiency of lactation, was positively associated with higher peak milk yield. The findings in early lactation suggest that estimates of feed efficiency could be improved by evaluating feed efficiency relative to conception, rather than parturition and lactation, to better account for the energy stored and released from body reserves in capital breeding.

Repetitive bouts of resistance and aerobic exercise can have dramatic effects on whole body physiology. Dietary tryptophan supplementation supports protein turnover and serotonin production, which assist in responses to exercise. The objective of this study was to investigate the effects of a 12-week incremental exercise regimen and supplemental dietary tryptophan on pre-, mid-, and post-exercise heart rate and respiratory rate in sled dogs. Sixteen dogs (mean age of 4.8 ± 2.5 years, body weight 24.3 ± 4.3 kg) were blocked for sex, age, and body weight, and randomly allocated to a control diet or the control plus tryptophan diet (tryptophan to large-neutral-amino-acid ratio of 0.075:1). All dogs participated in a 12-week conditioning regimen with controlled exercise challenges at week -1 and subsequently every 3 weeks. Electrocardiogram electrodes and thoracic rib bands were worn to record heart rate and respiratory rate (EMKA Tech., Falls Church, VA, USA) prior to, during, and following each exercise challenge. A trans regression, mixed, and correlation model were used where appropriate to assess the fixed effects of treatment and week, and explore linear relationships between recovery time and week in SAS (v 9.4). No differences were found in heart rate between treatment groups for any training level (P > 0.10). Working, recovery, and time required for heart rate to recover post-exercise decreased from week -1 to week 11 (P < 0.05). Correlation analysis indicated that treatment dogs recovered respiratory rate faster post-exercise compared to control (r = -0.421, P < 0.05). Resting, recovery and time required for respiratory rate to recover post-exercise decreased from baseline to week 11 (P < 0.05). This data suggests improvement in cardiorespiratory fitness was observed over 12 weeks of training and that tryptophan may support respiratory function during exercise recovery.

The purpose of the current study was to explore the effect of autologous adipose tissue on cartilage responses to lipopolysaccharide (LPS). We hypothesized that LPS elicits an inflammatory response in cartilage, and that response is augmented in the presence of adipose tissue. Furthermore, we hypothesized that this augmented inflammatory response is due, at least in part, to increased exposure of cartilage to adipose tissue-derived c3a. Porcine cartilage explants from market-weight pigs were cultured in the presence or absence of autologous adipose tissue for 96 hours, the final 48 hours of which they were stimulated with LPS (0 or 10 μg/mL). Adipose tissue explants were also cultured alone, in the presence or absence of LPS. Media from all cartilage treatments was assayed for c3a/c3a des Arg, PGE2, GAG, and NO, and the viability of cartilage tissue was determined by differential fluorescent staining. Media from adipose tissue explants was assayed for c3a/c3a des Arg and PGE2. LPS produced a significant increase in PGE2, GAG, and NO production when cartilage was cultured in the absence of adipose tissue. Coculture of adipose tissue prevented a significant increase in PGE2 in cartilage explants. There was no effect of adipose tissue on LPS-induced GAG or NO, but the presence of adipose tissue significantly reduced cell viability in LPS-stimulated cartilage explants. Adipose tissue explants from lean animals reduced inflammatory responses of cartilage to LPS via a c3a/c3a des Arg-independent mechanism and were associated with a significant decline in cell viability. Thus, contrary to our hypothesis, adipose tissue from lean animals does not augment the inflammatory response of cartilage to stimulation by LPS. The mechanism of modulatory effects of adipose tissue on LPS-induced increase in PGE2 and decline in chondrocyte viability requires further research but appears to have occurred via a mechanism that is independent of adipocentric c3a/c3a des Arg.

We evaluated the antioxidative and anti-inflammatory potential of daily oral supplementation with a proprietary powdered Cucumis melo pulp (CMP) on exercise-induced markers of articular and muscular oxidative stress and inflammation in 12 horses. Horses performed a high-intensity exercise test immediately prior to, and then following, 3 weeks of daily supplementation of 1 g powdered CMP (CMP; n=8). Controls (Co; n=8) underwent the same exercise and sampling regime but were not supplemented. Blood and synovial fluid (SF) samples were taken 24 h prior to exercise (BL), and at 1 and 24 h following exercise. Plasma and SF were analysed for prostaglandin E2 (PGE2), total antioxidant status (TAS), nitrite and superoxide dismutase (SOD) activity. SF was analysed for glycosaminoglycans (GAG), and plasma was analysed for thiobarbituric acid reactive substances (TBARS). Comparisons were made using repeated measures with the initial exercise test as a covariate. There was an increase in SF SOD activity in the CMP group. Compared to Co at 1 h, CMP reduced nitrite and GAG in SF, as well as maintained plasma TAS and lymphocyte levels. At 24 h, plasma PGE2 and creatine kinase were lower in horses receiving CMP. Three weeks of supplementation with CMP reduced markers of articular and skeletal muscle oxidative stress and inflammation in response to high-intensity exercise in horses. Nutritive antioxidants may provide a useful adjunct to the daily nutrition plan of horses undergoing regular exercise training and competition.

Thirty five barrows (initial body weight [BW]: 15.1 ± 1.0 kg) were used to determine the effect of partially replacing Gly + Ser with Thr in reduced crude protein (CP) diets on growth performance, protein deposition in carcass and viscera, and skin collagen abundance during the late nursery phase to 25 kg BW. Pigs were individually fed one of five iso-nitrogenous diets (n = 7) for 21 d. The basal diet met estimated essential amino acids (AA) requirements by using all essential AA plus Gly and Ser in free form (CON; 12.1% CP; as-fed, analyzed contents). The remaining four diets were formulated by reducing total Gly and Ser concentrations to 60% or 20% of the CON diet. The N removed with Gly and Ser was replaced with either crystalline Thr or Glu. Total analyzed Thr made up either 1.59% (T1; 12.5% CP) or 2.34% (T2; 12.2% CP) of the Thr-supplemented diets, and total analyzed Glu made up either 3.47% (G1; 12.7% CP) or 4.64% (G2; 12.9% CP) of the Glu-supplemented diets. Pigs were slaughtered on day 21 to determine body composition and skin collagen abundance via bright field microscopy. Overall, average daily gain (ADG) and G:F and final carcass weights were greater for pigs fed diets supplemented with Glu (G1 + G2) vs. those fed diets supplemented with Thr (T1 + T2; P < 0.05, P = 0.060, and P = 0.050 for ADG, G:F, and final carcass weight, respectively); intermediate values were observed for CON. Nitrogen retention in carcass plus viscera and the AA profile of deposited protein in the carcass were not influenced by dietary treatment. Pigs fed the T2 and G2 diets had greater retention of Thr (vs. CON and G2) and Glu (vs. CON and T2) in the viscera protein, respectively (P < 0.05). The apparent utilization efficiency of standardized ileal digestible Thr for protein deposition in carcass plus viscera was less for pigs fed T2 (15.1%) vs. those fed CON (56.7%) or G2 (58.6% ± 2.9%) diets (P < 0.001). Only pigs fed T1 had skin collagen abundance not different from CON; pigs fed G1, G2, and T2 had reduced skin collagen abundance compared with CON and T1 (P < 0.01). Using Glu as an N source when Gly and Ser were reduced to 60% and 20% of CON in reduced CP diets maintained ADG for pigs between 15 and 25 kg BW, whereas supplying Thr as a N source reduced ADG and carcass weight. When dietary Gly and Ser were supplied at 60% of CON, only Thr supplementation rescued skin collagen abundance. Therefore, supplemental Thr at excess levels is not sufficient to replace N from Gly and Ser in reduced CP diets fed to late nursery pigs, despite supporting skin collagen abundance as a secondary indicator of Gly status.

A total of 96 newly weaned barrows (initial body weight [BW]: 6.3 ± 0.5 kg) were used to determine the effect of a low crude protein (CP) diet supplemented with Gly and Ser on growth and skin collagen abundance. Barrows were assigned to one of three experimental diets in a three-phase feeding program fed for 35 days (n = 8; pen was the experimental unit): 1) corn–soybean meal diet (CON; 20.3% to 23.1% CP; as-fed, analyzed contents); 2) low CP diet (14.8% to 21.4% CP) supplemented with Gly and Ser (G + S) to the same concentrations as CON; 3) low CP diet supplemented with Glu to maintain the same CP concentration as the G + S diet (GLU; 15.0% to 22.1% CP). On days 21 and 35, eight pigs per treatment were euthanized for the determination of physical and chemical body composition and skin collagen abundance. Pigs fed the CON diet had greater overall ADG and final BW compared to pigs fed GLU and G + S (P < 0.01). Over the entire 35-day experimental period, ADFI was not influenced by dietary treatment but G:F tended to be greater for pigs fed CON than G + S (P = 0.084), while intermediate values were observed for GLU. Carcass weights on days 21 and 35 were greater for pigs fed CON than G + S or GLU (P < 0.01). Viscera weights on day 21 were greater for CON than G + S and GLU (P < 0.05) and on day 35 were greater for CON than G + S (P < 0.05) with intermediate values observed for GLU. The N intake (g/d) between days 0 and 35 was greater for CON than G + S or GLU (P < 0.05) and N retention in combined carcass and viscera was greater for CON than G + S (P < 0.01) with intermediate values observed for GLU. No treatment effects were observed for efficiency of N utilization. Between days 0 and 21 however, the efficiency of using dietary N for N retention in carcass and viscera tended to be less for pigs fed CON vs. GLU (73.8% vs. 91.6%), while intermediate values were observed for G + S (84.3%; P = 0.095). Pigs fed CON and G + S diets had greater skin collagen abundance than pigs fed GLU on days 21 and 35 (P < 0.01). Supplementing low CP diets with Glu or with Gly and Ser at the levels used in the current study did not maintain ADG or combined carcass and viscera N retention and only the G + S diet supported skin collagen abundance not different from pigs fed CON. The importance of meeting essential AA requirements for growth are well accepted, but supplementing specific NEAA may be needed when feeding reduced CP diets to newly weaned pigs to support secondary indicators of AA status, such as skin collagen abundance.

Mechanistic models (MMs) have served as causal pathway analysis and ‘decision-support’ tools within animal production systems for decades. Such models quantitatively define how a biological system works based on causal relationships and use that cumulative biological knowledge to generate predictions and recommendations (in practice) and generate/evaluate hypotheses (in research). Their limitations revolve around obtaining sufficiently accurate inputs, user training and accuracy/precision of predictions on-farm. The new wave in digitalization technologies may negate some of these challenges. New data-driven (DD) modelling methods such as machine learning (ML) and deep learning (DL) examine patterns in data to produce accurate predictions (forecasting, classification of animals, etc.). The deluge of sensor data and new self-learning modelling techniques may address some of the limitations of traditional MM approaches – access to input data (e.g. sensors) and on-farm calibration. However, most of these new methods lack transparency in the reasoning behind predictions, in contrast to MM that have historically been used to translate knowledge into wisdom. The objective of this paper is to propose means to hybridize these two seemingly divergent methodologies to advance the models we use in animal production systems and support movement towards truly knowledge-based precision agriculture. In order to identify potential niches for models in animal production of the future, a cross-species (dairy, swine and poultry) examination of the current state of the art in MM and new DD methodologies (ML, DL analytics) is undertaken. We hypothesize that there are several ways via which synergy may be achieved to advance both our predictive capabilities and system understanding, being: (1) building and utilizing data streams (e.g. intake, rumination behaviour, rumen sensors, activity sensors, environmental sensors, cameras and near IR) to apply MM in real-time and/or with new resolution and capabilities; (2) hybridization of MM and DD approaches where, for example, a ML framework is augmented by MM-generated parameters or predicted outcomes and (3) hybridization of the MM and DD approaches, where biological bounds are placed on parameters within a MM framework, and the DD system parameterizes the MM for individual animals, farms or other such clusters of data. As animal systems modellers, we should expand our toolbox to explore new DD approaches and big data to find opportunities to increase understanding of biological systems, find new patterns in data and move the field towards intelligent, knowledge-based precision agriculture systems.

Perennial forage production exists in Ontario to support the livestock industry, but also provides nesting habitat for grassland birds such as the threatened Bobolink (Dolichonyx oryzivorus) and Eastern Meadowlark (Sturnella magna). Delaying hay harvest until July 15 or later allows most nestling birds to leave the nest, but the nutritional value of hay decreases substantially. This project estimated the nutritional and economic impact of delaying the first hay cut until after July 15 on beef and dairy production in Ontario, Canada. Forage crops were sampled across Ontario, analysis of nutritional value performed, and effects on production and economics modelled. 634 samples were collected over 13 weeks at 16 sites from May 21 to August 14 during 2014 and 2015. As expected, nutritional quality declined over the season. Crude protein decreased by 5.2%, total digestible nutrients by 7.7%, neutral detergent fibre digestibility (NDFd48) by 20.1%, while lignin increased by 3.5%, neutral detergent fibre by 13.1%, and acid detergent fibre by 9.9%. Estimated yearly milk production decreased 10.9 kg or C$7.87/dairy cow for each day of delay in harvest (2017 values). Estimated growth of backgrounding beef steers decreased 1.56 kg or C$5.49/head for each day of delay in harvest. This translated into lost revenue per acre for backgrounding steers of C$31 per acre and C$45 per acre for over wintering beef cows for a delay from mid-June to mid-July. Some agri-environmental incentives in Canada, US and Europe offset the reduced revenue due to lower quality forages. This analysis informs farmers about the cost of practices to benefit grassland birds and provides empirical data on how to structure stewardship incentives for these practices.

Milk protein yield responses to changes in the profile of essential amino acids absorbed by the gastrointestinal tract or circulating in blood plasma do not follow the classic limiting amino acid response, in part because of an ability of the mammary glands to modify their blood flow rate and net clearance of amino acids out of plasma. The hypothesis that mammary blood flow is locally regulated to maintain ATP balance accounts for observed changes in flow due to postruminal glucose, insulin, and essential amino acid (EAA) infusions. An additional hypothesis that net mammary uptakes of metabolites from blood are affected by perturbations in their respective arterial concentrations and the rate of mammary blood flow also appears to hold for the energy metabolites glucose, acetate, β-hydroxybutyrate, and fatty acids. However, net EAA uptakes by the mammary glands are poorly predicted by models considering arterial concentrations and blood flow rates only. Evidence points to intramammary protein synthesis and secretion as the determinant of net EAA uptake. The intracellular signaling network anchored by the mechanistic target of rapamycin complex 1 stands as an excellent candidate to explain nutritional effects on milk protein synthesis because it integrates information on physiological and nutritional state to affect protein synthesis and cell metabolism, growth, proliferation, and differentiation in many cell types. In mammary cells in vitro and in vivo, the mechanistic target of rapamycin complex 1, integrated stress response, and glycogen synthase kinase-3 networks that contribute to regulation of initiation of mRNA translation are responsive to acute changes in nutrient supply and EAA profile. However, after several days of postruminal infusion of balanced and imbalanced EAA profiles, these signaling networks do not appear to continue to account for changes in milk protein yields. Gene expression evidence suggests that regulation of components of the unfolded protein response that control biogenesis of the endoplasmic reticulum and differentiation of a secretory phenotype may contribute to effects of nutrition on milk protein yield. Connections between early signaling events and their long-term consequences should be sought.

Twenty-eight lactating Yorkshire and Yorkshire × Landrace primiparous sows were used to test the hypothesis that feeding a diet with reduced CP concentration and supplemented with crystalline AA (CAA) does not decrease milk protein yield and litter growth but improves apparent N utilization for milk protein production. Sows were assigned to 1 of 2 dietary treatments: 1) control (CON; 16.2% CP; analyzed content) or 2) low CP with CAA to meet estimated requirements of limiting AA (LCP; 12.7% CP) over a 17-d lactation period. A N balance was conducted for each sow between days 13 and 17 of lactation. On day 17, a 12-h primed continuous infusion of l-[ring-2H5]-Phe was conducted on 12 sows (n = 6) with serial blood and milk sampling to determine plasma AA concentrations and Phe enrichment, and milk casein synthesis, respectively. Thereafter, sows were sacrificed and tissues were collected to determine tissue protein fractional synthesis rates (FSR). Litter growth rate and milk composition did not differ. Sows fed the LCP diet had reduced N intake (122.7 vs. 153.2 g/d; P < 0.001) and maternal N retention (13.5 vs. 24.6 g/d; P < 0.05) and greater apparent efficiency of using dietary N intake for milk production (85.1% vs. 67.5%; P < 0.001). On day 17 of lactation, all plasma essential AA concentrations exhibited a quartic relationship over time relative to consumption of a meal, where peaks occurred at approximately 1- and 4-h postprandial (P < 0.05). Protein FSR in liver, LM, gastrocnemius muscle, mammary gland, and in milk caseins did not differ between treatments. Feeding primiparous sows with a diet containing 12.7% CP and supplemented with CAA to meet the limiting AA requirements did not reduce milk protein yield or piglet growth rate and increased the apparent utilization of dietary N, Arg, Leu, Phe+Tyr, and Trp for milk protein production. The improved apparent utilization of N and AA appears to be related exclusively to a reduction in N and AA intake.

The objectives of this study were to: (a) select an ideal organogel for the oil phase of a novel gel encapsulation technology, (b) optimize the formulation of an organogel and sodium alginate-based gel complex, and (c) examine the rumen protective ability of the gel by measuring 48-h in vitro ruminal dry matter disappearance and gas production from encapsulated dried and ground holy basil leaves.; Results: A rice-bran wax and canola oil organogel was selected for the oil phase of the gel complex as this combination had a 48-h dry matter disappearance of 6%, the lowest of all organogels analyzed. The gel complex was formulated by homogenizing the organogel with a sodium alginate solution to create a low-viscosity oil-in-water emulsion. Average dry matter disappearance of gel-encapsulated holy basil was 19%, compared to 42% for the free, unprotected holy basil. However, gel encapsulation of holy basil stimulated gas production. Specifically, gas production of encapsulated holy basil was four times higher than the treatment with holy basil added on top of the gel prior to incubation rather than encapsulated within the gel.; Conclusion: Although the gel itself was highly degradable, it is speculated encapsulation thwarted holy basil's antimicrobial activity. © 2018 Society of Chemical Industry.; © 2018 Society of Chemical Industry.

Dietary restriction (DR) increases median life span and protects against age-related disease. Improved longevity can be achieved by restriction of dietary energy, protein, or amino acids (AAs), such as methionine (Met). Met requirements have been defined using methodologies that measure the dose response to Met when all other dietary variables are held constant and with outcomes focused on protein turnover. Here, we cover protein and sulfur AA requirements and discuss the terms "deficient," "optimal," and "excess" and how these need to be considered. We additionally discuss the effect of methyl-donating compounds on sulfur AA metabolism and outcomes. We will discuss how the mechanistic target of rapamycin complex 1 (mTORC1) signaling network regulates protein turnover, lipogenesis and cell growth, proliferation, differentiation, and metabolism in response to hormones, AAs, and cellular energy status. Inhibition of mTORC1 signaling with rapamycin or genetic mutation increases median life span in model organisms, and mTORC1 inhibition may be responsible for some of the life span-extending effects of DR. Finally, we discuss how the sulfur AAs may regulate aspects of reactive oxygen species (ROS) mitigation. Overall, we suggest that approaches evaluating AA intake need to consider whole-body protein synthesis and measures related to tissue-specific and whole-body metabolism that have been associated with longevity.

Elevated planes of nutrition in the preweaning period of dairy calf management can increase average daily gain, shorten age at puberty, and increase milk yield. In a previous study, 12 Holstein calves were fed 2 meals/d of 4 or 2 L milk replacer up to 7 wk of age. The objective of the current study was to estimate parameters of abomasal emptying and glucose-insulin dynamics in these calves by fitting a mechanistic model to postprandial appearances of plasma glucose, insulin, and the abomasal emptying marker acetaminophen measured at 4 and 7 wk of age. Higher intake of milk replacer resulted in longer bouts of abomasal emptying at a slower rate. Parameters of glucose and insulin dynamics were not affected by milk replacer intake. However, older calves had decreased insulin-stimulated glucose utilization indicating impaired insulin sensitivity, as well as increased pancreatic responsiveness. Neither of these effects were apparent from i.v. glucose tolerance tests on the calves and may have been related to postprandial gut hormone release. Effects of age on parameters of glucose-insulin dynamics were larger than effects of milk replacer intake. Conversely, effects of milk replacer intake on abomasal emptying were larger than effects of age.

Many chronic degenerative diseases have been linked to high intake of various trace and heavy metals. The presence of these compounds in drinking water may be a significant contributing factor to total dietary intake and deposition, resulting in the propagation of a disease cascade. Dairy farm families residing in rural Ontario completed a survey pertaining to the health status of the individuals living on-farm. Water samples were also collected at each location and analyzed for mineral content. Out of 200 surveys delivered to farms, 134 were returned, which formed a study group comprised of 218 adult and 230 children participants. Taking into consideration several factors, such as genetics, environment and diet, the data were analyzed for correlations between heavy metal and mineral status and the prevalence of neurodegenerative diseases. The findings from this study suggest phosphorus, potassium and magnesium concentrations may play a role in the development of neurodegenerative diseases. Total hardness and pH of water may also have an impact on the development of these diseases.

The objective of this study was to evaluate the influence of metabolizable protein (MP) level and supplemental rumen-protected methionine (RPM) on colostrum quality and passive immunity transfer from beef cows fed during late-gestation. One hundred and forty-seven pregnant Angus crossbred cows and heifers were randomly assigned to one of six dietary treatments based on a 3 x 2 factorial arrangement. Diets included feeding to 90%, 100% and 110% MP requirements (NRC, 2016), offered with (without) 9 g/d of RPM. Cattle were individually fed a partially mixed ration (60% haylage/40% straw for cows; 70% haylage/30% straw for heifers) with supplements top-dressed daily for approximately 56 d prior to calving. Immediately after calving, a colostrum sample was collected from each dam for analysis of milk components, total protein, and IgG concentrations. Two d post-calving, serum samples were collected from each calf for analysis of total protein and IgG concentrations. A refractometer was used to estimate total protein as a preliminary estimate of the transfer of maternal IgG and total IgG concentrations were later determined using Radial Immunodiffusion (RID). Data were analyzed as a completely randomized block design using Proc GLIMMIX in SAS. Level of MP, RPM, and their interaction did not influence concentrations of fat, protein, BHB or SCC in colostrum (P ≥ 0.12). Supplementation of RPM decreased concentrations of MUN (45.92 vs. 54.51 ±3.17 mg/dl; P = 0.04), but did not change IgG (14.66 vs. 15.90 ±0.87 g/dl; P = 0.29). While MP level during gestation did not affect total protein or IgG concentrations in calf serum (P ≥ 0.64), providing supplemental RPM decreased total protein (5.79 vs. 6.33 ±0.13 g/dl; P = 0.002) and IgG concentrations in calf serum (3.44 vs. 4.30 ±0.25 g/dl; P = 0.01). These results suggest that providing supplemental methionine for beef cows during gestation may alter colostrum quality and decrease serum protein and IgG in offspring.

A three-way factorial arrangement was used to assess impacts of protein/methionine nutrition during gestation and two lactational management systems on performance of cow-calf pairs pre-weaning. 140 crossbred beef cows were managed in drylot and randomly assigned to one of six nutritional treatments during the third trimester of gestation. Cows were fed to meet 110% (HP), 100% (MP), or 90% (LP) of metabolizable protein requirements for late-gestation (NRC, 2016), with(without) rumen-protected methionine (RPM) for 8 weeks prior to parturition. Post-partum, cow-calf pairs were managed in drylot (DL) and fed ad libitum, a ration formulated to meet lactational nutrient requirements, or rotationally grazed on pasture (PAS). Cow body weights (BW) and body condition scores (BCS) and calf BW were recorded at regular intervals. All data were analyzed using PROC GLIMMIX (SAS v9.4). HP cows were heaviest at calving (P = 0.02), but calf birthweight was unaffected by gestational nutrition (P > 0.19). Gestational nutrition did not impact cow conception rate (CR), or calf BW and ADG (P > 0.17). Lactational management system (PAS, DL) did not affect cow BCS or CR (P > 0.13), but impacted cow and calf BW and ADG. PAS calves were heavier, had greater ADG, and heavier weaning BW than DL calves (P < 0.02). This came at the expense of BW loss throughout lactation and lower ADG for PAS cows, versus BW gains and greater ADG for DL cows (P < 0.0001). An interaction between protein level and RPM supplementation was present for cow BW at calving and weaning, and BW change throughout lactation (P < 0.04), with tendencies for interactions for cow ADG and BCS (P < 0.09). Overall, prepartum protein/methionine supplementation did not affect calf performance prior to weaning, but may have affected cow performance during lactation. Rotationally grazing cow-calf pairs on pasture improved pre-weaning calf gains, but reduced cow gains throughout lactation.

Twenty-one of each pregnant (P) and nonserviced, nonpregnant (NP) sister-pairs of gilts were selected to investigate the effect of pregnancy on protein deposition (Pd; whole body and maternal), insulin sensitivity, and mRNA abundance of genes involved in energy and AA metabolism. Between breeding (study day 0) and day 111, P and NP gilts received 2.16 kg of the experimental diet (3.34 Mcal ME/kg, 17.6% crude protein, 0.78% standardized ileal digestible lysine) that was formulated to meet the estimated ME requirements of pregnant gilts (and meet or exceed AA requirements). Nitrogen balances were conducted on day 63 and 102 ± 0.2 of the study during 4-d periods. Blood samples were collected on day 43, 56, 71, 85, 98, and 108 ± 0.3 of the study to determine plasma concentrations of fasted IGF-1, estradiol (E2), and estrone sulfate (E1S). Frequently sampled intravenous glucose tolerance tests (FSIGTT) were conducted on day 75 ± 0.7 in 6 P and 5 NP gilts and on day 107 ± 0.4 in 17 P and 17 NP gilts and the MINMOD approach was applied to evaluate whole body insulin sensitivity and pancreatic responsiveness. Longissimus muscle (LM) and s.c. adipose tissue (AD) samples were excised from 12 P and 12 NP gilts at day 111 ± 0.4 of the study after euthanasia to determine mRNA abundance of key genes. Whole body Pd was greater (P < 0.001) at day 102 and maternal Pd was lower (P < 0.002) at day 63 and 102 for P compared to NP gilts. Plasma concentrations of E1S and E2 increased (P < 0.05) with study day for P gilts and remained constant for NP gilts, which coincided with reduced plasma concentrations of IGF-1 and increased estrogen receptor alpha (ESR1) mRNA abundance in LM of P gilts. Glucose effectiveness was not different between P and NP gilts, but whole body insulin sensitivity was lower (P = 0.004) in P compared to NP gilts on day 75 and 107, which corresponded with reduced mRNA abundances of SLC2A4, HK2, SREBF1, and FASN, and increased abundances of PDK4 and PPARGC1A in LM and AD. When fed identically, P gilts had greater whole body Pd at day 102, which reflects Pd in the pregnancy-associated tissues (at the expense of maternal Pd), likely driven by estrogen-stimulated insulin resistance in peripheral tissue and subsequent modulation of gene expression relating to glucose metabolism.

The objective of this study was to evaluate production responses to supplementation of 2 AA groups in a 2 × 2 factorial design. Eight cows (99 ± 18 days in milk) were assigned to 4 jugular infusion treatments consisting of saline (CON), methionine plus lysine plus histidine (MKH), isoleucine plus leucine (IL), or MKH plus IL, in a replicated 4 × 4 Latin square design. Periods were 18 d in length, comprising 8 d of rest followed by 10 d of jugular infusion. Daily infusion amounts were 21 g of methionine, 38 g of lysine, 20 g of histidine, 50 g of leucine, and 22 g of isoleucine. Cows were ad libitum fed a common diet consisting of 15.2% crude protein and 1.61 Mcal/kg NEL on a dry matter basis that was predicted to meet rumen degradable protein requirements but was 15% deficient in metabolizable protein. Milk and energy-corrected milk yields increased by 2.3 kg/d and 1.9 kg/d, respectively, with infused IL, and no change was observed for MKH. Milk protein concentration increased by 0.13 percentage units for MKH, whereas milk protein yield increased for both MKH and IL by 84 g/d and 64 g/d, respectively. The milk protein yield increase for MKH+IL was 145 g/d versus CON. Gross feed efficiency tended to increase with IL infusion, and N efficiency tended to increase with MKH infusion. Aggregate arterial EAA concentrations less Met, Lys, and His declined by 7.2% in response to MKH infusion. Arterial EAA less Ile and Leu also declined by 6.2% in response to IL infusion. Net total AA (TAA) and EAA uptake by the udder tended to increase in response to MKH infusion, whereas mammary blood flow increased in response to IL infusion, but TAA and EAA net uptakes were unaffected. Apparent udder affinity increased for TAA and EAA less Met, Lys, and His in response to MKH infusion, whereas affinity for EAA less Ile and Leu increased for IL infusion. Venous Met and Leu concentrations increased by 192% and 35% from the MKH and IL infusions, respectively, compared with CON, which indicates that intracellular concentration of these EAA changed substantially. Increases in milk protein yield were observed from 2 groups of amino acids independently and additively, which contradicts the single limiting amino acid theory that a single EAA will limit milk protein yield. Bovine mammary gland biopsies allow researchers to collect tissue samples to study cell biology including gene expression, histological analysis, signaling pathways, and protein translation. This article describes two techniques for biopsy of the bovine mammary gland (MG). Three healthy Holstein dairy cows were the subjects. Before biopsies, cows were milked and subsequently restrained in a cattle chute. An analgesic (flunixin meglumine, 1.1 to 2.2 mg/kg of body weight) was administered via jugular intravenous [IV] injection 15-20 min prior to biopsy. For standing sedation, xylazine hydrochloride (0.01-0.05 mg/kg of body weight) was injected via the coccygeal vessels 5-10 min before the procedure. Once adequately sedated, the biopsy site was aseptically prepared and locally anaesthetized with 6 mL of 2% lidocaine hydrochloride via subcutaneous injection. Using aseptic technique, a 2 to 3 cm vertical incision was made using a number 10 scalpel. Core and needle biopsy tools were used. The core biopsy tool was attached to a cordless drill and inserted into the MG tissue through the incision using a clock-wise drill action. The needle biopsy tool was manually inserted into the incision site. Immediately after the procedure, an assistant applied pressure on the incision site for 20 to 25 min using a sterile towel to achieve hemostasis. Stainless steel surgical staples were used to oppose the skin incision. The staples were removed 10 days post-procedure. The main advantages of core and needle biopsies is that both approaches are minimally invasive procedures that can be safely performed in healthy cows. Milk yield following the biopsy was unaffected. These procedures require a short recovery time and result in fewer risks of complications. Specific limitations may include bleeding after the biopsy and infection on the biopsy site. Applications of these techniques include tissue collection for clinical diagnosis and research purposes, such as primary cell culture.

Net energy accounts for the proportion of energy expenditure attributed to the digestion, metabolism, and absorption of ingested food. Currently, there are no models available to predict net energy density of food for domestic cats. Therefore, the objectives of this study were to measure the heat increment of feeding in cats, and to model the net energy of commercial diets. Metabolizable energy and calorimetry data from two previous studies was reanalyzed to create net energy models in the present study. Energy expenditure was calculated using measurements of CO2 production and O2 consumption. Net energy was determined as the metabolizable energy of the diets minus the heat increment of feeding. The heat increment of feeding was determined as the area under the energy expenditure curve above the resting fed metabolic rate. Eight net energy models were developed using metabolizable energy, 1 of 4 dietary parameters (crude protein, fat, fiber, and starch), and heat increment of feeding values from 0–2 h or 0–21 h. Two hours postprandial, and over the full calorimetry period, the heat increment of feeding amounted for 1.74, and 20.9% of the metabolizable energy, respectively. Of the models tested, the models using crude protein in combination with metabolizable energy as dietary parameters best fit the observed data, thus providing a more accurate estimate of dietary energy availability for cats.

Feed efficiency has been widely studied in many areas of dairy science and is currently seeing renewed interest in the field of breeding and genetics. A critical part of determining how efficiently an animal utilizes feed is accurately measuring individual dry matter (DM) intake. Currently, multiple methods are used to measure feed intake or determine the DM content of that feed, resulting in different levels of accuracy of measurement. Furthermore, the scale at which data need to be collected for use in genetic analyses makes some methodologies impractical. This systematic review aims to provide an overview of the current methodologies used to measure both feed intake in ruminants and DM content of feedstuffs, current methods to predict individual DM intake, and applications of large-scale intake measurements. Overall, advances in milk spectral data analysis present a promising method of estimating individual DM intake on a herd scale with further validation of prediction models. Although measurements of individual feed intake rely on the same underlying principle, the methods selected are largely dictated by the costs of capital, labor, and necessary analyses. Finally, DM methodologies were synthesized into a comprehensive protocol for use in a variety of feedstuffs.

Secretory capacity of bovine mammary glands is enabled by a high number of secretory cells and their ability to use a range of metabolites to produce milk components. We isolated RNA from milk fat to measure expression of genes involved in energy-yielding pathways and the unfolded protein response in mammary glands of lactating cows given supplemental energy from protein (PT) and fat (FT) tested in a 2 × 2 factorial arrangement. We hypothesized that PT and FT would affect expression of genes in the branched-chain AA catabolic pathway and tricarboxylic acid (TCA) cycle based on the different energy types (aminogenic versus lipogenic) used to synthesize milk components. We also hypothesized that the response of genes related to endoplasmic reticulum (ER) homeostasis via the unfolded protein response would reflect the increase in milk production stimulated by PT and FT. Fifty-six multiparous Holstein-Friesian dairy cows were fed a basal total mixed ration (34% grass silage, 33% corn silage, 5% grass hay, and 28% concentrate on a dry matter basis) for a 28-d control period. Experimental rations were then fed for 28 d, consisting of (1) low protein, low fat (LP/LF); (2) high protein, low fat (HP/LF); (3) low protein, high fat (LP/HF); or (4) high protein and high fat (HP/HF). To obtain the high-protein (HP) and high-fat (HF) diets, intake of the basal ration was restricted and supplemented isoenergetically (net energy basis) with 2.0 kg/d rumen-protected protein (soybean + rapeseed, 50:50 mixture on dry matter basis) and 0.68 kg/d hydrogenated palm fatty acids on a dry matter basis. RNA from milk fat samples collected on d 27 of each period underwent real-time quantitative PCR. Energy from protein increased expression of BCAT1 (branched-chain amino acid transferase 1) mRNA, but only at the LF level, and tended to decrease expression of mRNA encoding the main subunit of the branched-chain keto-acid dehydrogenase complex. mRNA expression of malic enzyme, a proposed channeling route for AA though the TCA cycle, was decreased by PT, but only at the LF level. Expression of genes associated with de novo fatty acid synthesis was not affected by PT or FT. Energy from fat had no independent effect on genes related to ER homeostasis. At the LF level, PT activated XBP1 (X-box binding protein 1) mRNA. At the HF level, PT increased mRNA expression of the gene encoding GADD34 (growth arrest and DNA damage-inducible 34). These findings support our hypothesis that mammary cells use aminogenic and lipogenic precursors differently for milk component production when dietary intervention alters AA and fatty acid supply. They also suggest that mammary cells respond to increased AA supply through mechanisms of ER homeostasis, dependent on the presence of FT.

The objective of this study was to investigate how preweaning plane of milk replacer intake and age can affect insulin and glucose kinetics as well as abomasal emptying rate in dairy calves fed twice a day. A total of 12 female Holstein Friesian calves were blocked by cow parity, paired by colostrum origin, and were randomly assigned to a high plane of milk replacer intake (8 L/d, 1.2kg of milk replacer/d; n=6) or a low plane of nutrition (4 L/d, 0.6kg of milk replacer/d; n=6). All calves received 4 L of colostrum over 2 meals (1 and 6h after birth) and were then directly transferred to their assigned feeding plans until they were stepped-down from milk by 50% during wk 7 and weaned on wk 8. Milk replacer (24% crude protein, 18% crude fat) was fed at 150g/L twice daily (0700 and 1700h) and all calves had ad libitum access to pelleted calf starter, chopped wheat straw, and water. Jugular catheters were placed in all calves at 4, 7, and 10wk of age. Then, postprandial response to plasma glucose, insulin, and acetaminophen (supplied with the meal) were determined to measure abomasal emptying. The next day, a glucose tolerance test was conducted by infusing glucose via the jugular catheter. At 4 and 7wk of age, the rate constant (%/h) for abomasal emptying of the meal was lower in high calves (0.21±0.02 in wk 4; 0.27±0.02 in wk 7) compared with low (0.34±0.02 in wk 4; 0.47±0.02 in wk 7). The postprandial plasma insulin area under the curve over 420min was greater in high calves (18,443±7,329; low=5,834±739 µU/mL) compared with low. We found no differences in glucose tolerance test kinetics between the high and low dairy calves at 4, 7, or 10wk of age. The findings from this study suggest that feeding dairy calves an elevated plane of nutrition in 2 meals of milk replacer per day does not decrease insulin sensitivity.

The objectives of this study were to determine the effect of dietary inclusion [0%–50% of diet dry matter (DM)] and form (dry and modified wet) of corn distillers grains plus solubles (DGS) on pancreatic α-amylase and trypsin activities in calves. Feeding up to 50% DGS did not negatively impact pancreatic exocrine function.

Ontario’s dairy goat industry has grown in recent years. The objective of the present study was to establish the current nutritional practices on dairy goat farms. A survey was sent out to 216 dairy goat producers in Ontario. Producers were asked questions regarding their general herd structure and mortality, voluntary culling reasons, lactation management, nutrition management, housing and space allowance, goat kid management, and opinions on future research. Nutritional practices varied considerably between farms. The most common feeding systems were complete feed (pellet) with forage and component feed for lactating does, dry does, transition does, and doelings. Alfalfa hay was the most common forage fed with the complete feed system. Most producers (84%) fed their goats pellets or grain at the parlor. Milk production was greatest in the summer months, whereas milk fat and protein levels were greater in the fall and winter months. Colostrum management varied between farms. Most producers (36.1%) did not allow goat kids to spend time with their dam. Few farms pasteurized or tested the quality of the colostrum. There was considerable variability when producers offered grain and starter, hay, and water to goat kids, ranging from younger than 5 d to after weaning. Nutrition and kid rearing were the 2 most common areas identified by producers that require further investigation. Future research is essential in providing the dairy goat industry with the information required to effectively meet the needs of the animal, improve production efficiency, and ultimately help support the growth of the industry.

Tryptophan (Trp), an indispensable amino acid for dogs, is the precursor of serotonin, a neurotransmitter with a variety of effects throughout the body, including the ability to modulate cardiac and pulmonary activity. This study aimed to investigate the effects of a 12-week incremental exercise regimen and supplemental dietary Trp on heart rate (HR) and respiratory rate (RR) in client-owned sled dogs. Sixteen Siberian huskies were randomly allocated to either treatment or control diet groups. Both groups were fed a control diet (Trp to large neutral amino acid ratio of 0.047:1), however, treatment dogs received a Trp supplement to achieve a Trp to large neutral amino acid ratio of 0.075:1. Every three weeks, external telemetry equipment was used to non-invasively measure and record HR and RR at a resting, working, and post-exercise state in a controlled exercise challenge. A mixed model was used to test differences between diet, activity parameter, and week. Dietary Trp supplementation had no effect on HR or RR. Independent of diet, resting, working, post-exercise HR, and time to recover post-exercise HR decreased from week &minus, 1 to week 11 (p &lt, 0.05). Resting HR had the greatest reduction from week &minus, 1 to week 11 (21%, p &lt, 0.05). Working RR did not change with exercise (p &gt, 0.10), but rRR and postRR decreased from week &minus, 0.05). These data suggest that the exercise regimen the dogs were subjected to may have positively impacted the dogs&rsquo, capacity to sustain aerobic exercise, whereas Trp supplementation had no effect on HR or RR.

Thirty-five barrows (BW=15.1 ± 2.72 kg) were used to determine the effect of supplementing dietary Thr above estimated requirements on pig growth and skin collagen abundance when fed low-CP low-Gly diets. Pigs were randomly assigned to 1 of 5 dietary treatments: 1) Control, semi-purified diet that met EAA requirements (CON; 12.1% CP as-fed); the final proportion of NEAA was similar to that found in whole-body protein. The remaining diets were formulated by reducing Gly and Ser content to 60 and 20%, respectively, of CON and supplementing with either Glu or Thr at 2 levels each, to maintain similar CP concentrations: For diets 2 and 3, Thr was included at 1.59% (LT) or 2.34% (HT) at the expense of the NEAA mix. For diets 4 and 5, Glu was included at 3.47% (LG) or 4.64% (HG) at the expense of the NEAA mix. Pigs were fed at 2.8 × estimated ME requirements for maintenance in 3 equal meals per day over a 21-day experimental period. At slaughter, skin samples were collected for collagen analysis. All statistical analyses were determined using Proc GLIMMIX. There was no difference in initial or final BW. Overall, ADG for pigs fed diets supplemented with Glu (378g/d) was greater than those fed diets supplemented with Thr (359 ± 9.45g/d; P=0.041). Feed efficiency for pigs supplemented with Glu (0.431) was greater than diets supplemented with Thr (0.411 ± 0.01; P=0.074). Stereotypic behavior in horses, including cribbing and weaving, can be attributed to a number of different factors including the number of hours in a stall, number of grazing opportunities, number of times fed hay in a day, and the ability to touch other horses. By providing environmental enrichment that allows for feed availability for a longer time period, a reduction of stereotypic behaviors could be seen. A behavioral tool, a tongue-activated liquid dispenser, was tested to determine whether the device itself could reduce stereotypic behaviors. Four horses, two weavers and two cribbers were studied over a 4-day period. Each horse acted as it's own control in a randomized block design. On average, data trended toward a treatment effect depending on the day.

Forty-two cross-bred steers (BW=357±5.8 kg) fed whole corn grain-based finishing diets were used in a completely randomized block (60, 120, or 180 d on feed) design (2×3 factorial arrangement of treatments plus control) to determine the effect of inclusion level [0 (control), 16.7, 33.3, and 50% of diet DM) and form (dry (DDGS) or modified wet (MWDGS)] of distillers’ grains plus solubles (DGS) on hepatic and renal glutathione peroxidase (GPx) and hepatic urea cycle enzyme activities. Kidney weight (g kg−1of BW) increased linearly (P=0.004) with increasing inclusion levels of DGS. There were no effects (P≥0.11) of dietary treatment on hepatic and renal GPx activity (U g−1, U mg−1of protein, and kU liver−1). Hepatic carbamoyl phosphate synthetase activity (kU liver−1and U kg−1of BW) tended to linearly increase (P=0.09 and P=0.10, respectively) with increasing inclusion level of DGS. Hepatic ornithine transcarbamoylase and argininosuccinate synthetase activity (kU liver−1and U kg−1of BW) increased linearly (P≤0.05) with increasing inclusion levels of DGS. These data indicate that steers adapt to feeding up to 50% DGS by increasing kidney mass and activity of urea cycle enzymes in liver to allow for clearance of excess nitrogen. Also, hepatic and renal GPx activity, as an indicator of Se status, is not affected when typical finishing diets are fed.

Background Dietary ammonia is rapidly absorbed but poorly used for urea synthesis in pigs fed low-crude-protein (low-CP) diets deficient in dispensable amino acid (DAA)-nitrogen. Objective We explored the effect of dietary ammonia on net amino acid (AA) balances in portal-drained viscera (PDV) and livers of pigs fed a diet deficient in DAA-nitrogen. Methods Eight barrows with an initial body weight (BW) of 26.5 ± 1.4 kg (mean + SD) were surgically fitted with 4 catheters each (portal, hepatic, and mesenteric veins and carotid artery). The pigs were restricted-fed (2.8 × 191 kcal/kg BW0.60) for 7 d, and every 8 h a diet deficient in DAA-nitrogen supplemented with increasing amounts of ammonia-nitrogen (CP = 7.76%, 9.27%, and 10.77% for the control and low- and high-ammonia diets, respectively). The treatment sequence was based on a 3 × 3 Latin-square design with 3 consecutive periods. On the last day of each period, blood flows in portal and hepatic veins were determined with a continuous infusion of ρ-amino hippuric acid into the mesenteric vein. Consecutive blood samples were taken for AA concentration in blood plasma, and AA balances were calculated for PDV and the liver. Results Cumulative release of citrulline (Cit) and proline (Pro) increased with ammonia supplementation in PDV but decreased for glutamine (Gln) and glycine (Gly) (Gln: -19.32 ± 3.56, -32.50 ± 3.73, and -42.11 ± 3.55 mmol/meal for the control and low- and high-ammonia groups, respectively; P ≤ 0.05). Cumulative release of alanine (Ala), glutamic acid (Glu), and Gln increased with ammonia supplementation across the liver (P ≤ 0.05). When combined, PDV+liver, the cumulative release of Ala, Cit, and Glu increased with ammonia-nitrogen supplementation (P ≤ 0.05). Conclusion Dietary ammonia could be used as a nitrogen supplement to increase the synthesis of Ala, Cit, and Glu across splanchnic organs in pigs fed a diet deficient in DAA-nitrogen.

Twenty-eight lactating Yorkshire and Yorkshire × Landrace primiparous sows were used to test the hypothesis that feeding a diet with reduced CP concentration and supplemented with crystalline AA (CAA) does not decrease milk protein yield and litter growth but improves apparent N utilization for milk protein production. Sows were assigned to 1 of 2 dietary treatments: 1) control (CON; 16.2% CP; analyzed content) or 2) low CP with CAA to meet estimated requirements of limiting AA (LCP; 12.7% CP) over a 17-d lactation period. A N balance was conducted for each sow between days 13 and 17 of lactation. On day 17, a 12-h primed continuous infusion of l-[ring-(2)H(5)]-Phe was conducted on 12 sows (n = 6) with serial blood and milk sampling to determine plasma AA concentrations and Phe enrichment, and milk casein synthesis, respectively. Thereafter, sows were sacrificed and tissues were collected to determine tissue protein fractional synthesis rates (FSR). Litter growth rate and milk composition did not differ. Sows fed the LCP diet had reduced N intake (122.7 vs. 153.2 g/d; P < 0.001) and maternal N retention (13.5 vs. 24.6 g/d; P < 0.05) and greater apparent efficiency of using dietary N intake for milk production (85.1% vs. 67.5%; P < 0.001). On day 17 of lactation, all plasma essential AA concentrations exhibited a quartic relationship over time relative to consumption of a meal, where peaks occurred at approximately 1- and 4-h postprandial (P < 0.05). Protein FSR in liver, LM, gastrocnemius muscle, mammary gland, and in milk caseins did not differ between treatments. Feeding primiparous sows with a diet containing 12.7% CP and supplemented with CAA to meet the limiting AA requirements did not reduce milk protein yield or piglet growth rate and increased the apparent utilization of dietary N, Arg, Leu, Phe+Tyr, and Trp for milk protein production. The improved apparent utilization of N and AA appears to be related exclusively to a reduction in N and AA intake.
