The advent of immune checkpoint inhibitors (ICIs) has represented a breakthrough in the treatment of many cancers, although a high number of patients fail to respond to ICIs, which is partially due to the ability of tumor cells to evade immune system surveillance. Non-coding microRNAs (miRNAs) have been shown to modulate the immune evasion of tumor cells, and there is thus growing interest in elucidating whether these miRNAs could be targetable or proposed as novel biomarkers for prognosis and treatment response to ICIs. We therefore performed an extensive literature analysis to evaluate the clinical utility of miRNAs with a confirmed direct relationship with treatment response to ICIs. As a result of this systematic review, we have stratified the miRNA landscape into (i) miRNAs whose levels directly modulate response to ICIs, (ii) miRNAs whose expression is modulated by ICIs, and (iii) miRNAs that directly elicit toxic effects or participate in immune-related adverse events (irAEs) caused by ICIs.

Abstract Background JUNB transcription factor contributes to the formation of the ubiquitous transcriptional complex AP-1 involved in the control of many physiological and disease-associated functions. The roles of JUNB in the control of cell division and tumorigenic processes are acknowledged but still unclear. Results Here, we report the results of combined transcriptomic, genomic, and functional studies showing that JUNB promotes cell cycle progression via induction of cyclin E1 and repression of transforming growth factor (TGF)-β2 genes. We also show that high levels of JUNB switch the response of TGF-β2 stimulation from an antiproliferative to a pro-invasive one, induce endogenous TGF-β2 production by promoting TGF-β2 mRNA translation, and enhance tumor growth and metastasis in mice. Moreover, tumor genomic data indicate that JUNB amplification associates with poor prognosis in breast and ovarian cancer patients. Conclusions Our results reveal novel functions for JUNB in cell proliferation and tumor aggressiveness through regulation of cyclin E1 and TGF-β2 expression, which might be exploited for cancer prognosis and therapy.

Activation of the aryl hydrocarbon receptor (AhR) has been shown to be important in physiological processes other than detoxification, including vascular homeostasis. Although AhR is highly expressed in the endothelium, its function has been poorly studied. This systematic review aims to summarise current knowledge on the AhR role in the endothelium and its cardiovascular implications. We focus on endogenous AhR agonists, such as some uremic toxins and other agonists unrelated to environmental pollutants, as well as studies using AhR knockout models. We conclude that AhR activation leads to vascular oxidative stress and endothelial dysfunction and that blocking AhR signalling could provide a new target for the treatment of vascular disorders such as cardiovascular complications in patients with chronic kidney disease or pulmonary arterial hypertension.

Pulmonary fibrosis (PF) is characterized by aberrant extracellular matrix (ECM) deposition, activation of fibroblasts to myofibroblasts and parenchymal disorganization, which have an impact on the biomechanical traits of the lung. In this context, the balance between matrix metalloproteinases (MMPs) and their tissue inhibitors of metalloproteinases (TIMPs) is lost. Interestingly, several MMPs are overexpressed during PF and exhibit a clear profibrotic role (MMP-2, -3, -8, -11, -12 and -28), but a few are antifibrotic (MMP-19), have both profibrotic and antifibrotic capacity (MMP7), or execute an unclear (MMP-1, -9, -10, -13, -14) or unknown function. TIMPs are also overexpressed in PF; hence, the modulation and function of MMPs and TIMP are more complex than expected. EMMPRIN/CD147 (also known as basigin) is a transmembrane glycoprotein from the immunoglobulin superfamily (IgSF) that was first described to induce MMP activity in fibroblasts. It also interacts with other molecules to execute non-related MMP aactions well-described in cancer progression, migration, and invasion. Emerging evidence strongly suggests that CD147 plays a key role in PF not only by MMP induction but also by stimulating fibroblast myofibroblast transition. In this review, we study the structure and function of MMPs, TIMPs and CD147 in PF and their complex crosstalk between them.

Patients with high cholesterol and glucose levels are at high risk for cardiovascular disease. The Sterol Regulatory Element Binding Protein (SREBP) system regulates genes involved in lipid, cholesterol and glucose pathways. Autosomal Dominant Hypercholesterolemias (ADHs) are a group of diseases with increased cholesterol levels. They affect 1 out of every 500 individuals. About 20–30% of patients do not present any mutation in the known genes (LDLR, APOB and PCSK9). ADHs constitute a good model to identify the genes involved in the alteration of lipid levels or possible therapeutic targets. In this paper, we studied whether a mutation in the SREBP system could be responsible for ADH and other metabolic alterations present in these patients. Forty-one ADH patients without mutations in the main responsible genes were screened by direct sequencing of SREBP system genes. A luciferase reporter assay of the found mutation and an oral glucose tolerance test in carriers and non-carriers were performed. We found a novel mutation in the SREBF2 gene that increases transcription levels and cosegregates with hypercholesterolemia, and we found increased glucose levels in one family. SREBP2 is known to be involved in cholesterol synthesis, plasma levels and glucose metabolism in humans. The found mutation may involve the SREBF2 gene in hypercholesterolemia combined with hyperglycemia.

Abstract Background Large-sequencing cancer genome projects have shown that tumors have thousands of molecular alterations and their frequency is highly heterogeneous. In such scenarios, physicians and oncologists routinely face lists of cancer genomic alterations where only a minority of them are relevant biomarkers to drive clinical decision-making. For this reason, the medical community agrees on the urgent need of methodologies to establish the relevance of tumor alterations, assisting in genomic profile interpretation, and, more importantly, to prioritize those that could be clinically actionable for cancer therapy. Results We present PanDrugs, a new computational methodology to guide the selection of personalized treatments in cancer patients using the variant lists provided by genome-wide sequencing analyses. PanDrugs offers the largest database of drug-target associations available from well-known targeted therapies to preclinical drugs. Scoring data-driven gene cancer relevance and drug feasibility PanDrugs interprets genomic alterations and provides a prioritized evidence-based list of anticancer therapies. Our tool represents the first drug prescription strategy applying a rational based on pathway context, multi-gene markers impact and information provided by functional experiments. Our approach has been systematically applied to TCGA patients and successfully validated in a cancer case study with a xenograft mouse model demonstrating its utility. Conclusions PanDrugs is a feasible method to identify potentially druggable molecular alterations and prioritize drugs to facilitate the interpretation of genomic landscape and clinical decision-making in cancer patients. Our approach expands the search of druggable genomic alterations from the concept of cancer driver genes to the druggable pathway context extending anticancer therapeutic options beyond already known cancer genes. The methodology is public and easily integratable with custom pipelines through its programmatic API or its docker image. The PanDrugs webtool is freely accessible at http://www.pandrugs.org.

Macronutrients represent risk factors for hyperlipidemia or diabetes. Lipid alterations and type 2 diabetes mellitus are global health problems. Overexpression of sterol regulatory element-binding factor (Srebf2) in transgenic animals is linked to elevated cholesterol levels and diabetes development. We investigated the impact of increased Srebf2 locus expression and the effects of control and high-fat, high-sucrose (HFHS) diets on body weight, glucose and lipid metabolisms in transgenic mice (S-mice). Wild type (WT) and S-mice were fed with both diets for 16 weeks. Plasma glucose, insulin and lipids were assessed (n = 25). Immunostainings were performed in liver, pancreas and fat (N = 10). Expression of Ldlr and Hmgcr in liver was performed by RT-PCR (N = 8). Control diet: S-mice showed reduced weight, insulin, total and HDL cholesterol and triglycerides (TG). HFHS diet widened differences in weight, total and HDL cholesterol, insulin and HOMA index but increased TG in S-mice. In S-mice, adipocyte size was lower while HFHS diet produced lower increase, pancreatic β-cell mass was lower with both diets and Srebf2, Ldlr and Hmgcr mRNA levels were higher while HFHS diet produced a rise in Srebf2 and Hmgcr levels. Srebf2 complete gene overexpression seems to have beneficial effects on metabolic parameters and to protect against HFHS diet effects.

El estrés laboral es una de las enfermedades laborales más extendidas a lo largo de los países desarrollados durante las últimas décadas del siglo XX y el siglo XXI. Los altos costes generados así como el aumento de la prevalencia han hecho que diversas instituciones a nivel mundial lo estudien y planteen la necesidad de crear leyes estatales para su prevención. El presente estudio experimental pretest-postest de tipo ensayo clínico aleatorizado tiene como objetivo principal evaluar si una intervención enfermera basada en la terapia cognitivo-conductual disminuye el efecto de los factores estresantes causantes del estrés laboral de los profesionales sanitarios que trabajan en los dispositivos de emergencias extrahospitalarias del servicio de urgencias médicas de la Comunidad de Madrid (SUMMA 112), ya que estos profesionales, debido a las características específicas de su puesto de trabajo, las situaciones de crisis en las que desarrollan su labor asistencial y la falta de adiestramiento en técnicas de superación y control del estrés les hace especialmente sensibles a padecer trastornos derivados del estrés laboral. The sleep disorders are among the most common behaviour problems in infancy and early childhood, not only affect children, but parents also suffer from lack of sleep which creates problems of anxiety and communication in the partner by exhaustion. One way to prevent insomnia in children is that parents know how to create the habit of sleeping with their children.The main aims of the protocol are: - To train parents to establish healthy habits for proper sleep hygiene.- Preventing sleep disorders caused by bad habits in the infant. - To prevent symptoms of anxiety due to lack of sleep in the parents.The program will consist of 6 sessions of 90 minutes long, 3 pre-natal where they thrive knowledge and 3 sessions after to exposure doubts and search for solutions. We think that cooperation between the mental health nurse specialist with the midwife it is necessary to create a program that establish a healthy dream pattern in both infants and their parents, will prevent the sleep disorders by incorrect habits and their effects

Los trastornos del sueño figuran entre los problemas del comportamiento más frecuentes en la lactancia y en la primera infancia, y o sólo afectan a los niños sino que los padres sufren también de falta de sueño lo que origina problemas de ansiedad y de comunicación en la pareja por agotamiento. Una forma de prevenir el insomnio de los niños es que los padres conozcan cómo deben crear el hábito del sueño a sus hijos. Los objetivos generales del protocolo son: - Capacitar a los padres para implantar hábitos saludables para una correcta higiene del sueño. - Prevenir trastornos del sueño por malos hábitos en el lactante. - Prevenir síntomas de ansiedad por falta de sueño en los padres. El programa constará de 6 sesiones de 90 minutos de duración, 3 previas al parto donde se desarrollan los conocimientos teóricos y 3 sesiones posteriores de exposición de dudas y búsqueda de soluciones. Creemos que la cooperación de la enfermera especialista en salud mental con la matrona para crear un programa psicoeducativo que favorezca el patrón sueño-descanso tanto de los lactantes como de sus padres, prevendrá la aparición de trastornos de sueño por hábitos incorrectos y sus efectos. Relaxation is a standard technique used by nurses to reduce the level of anxiety. It seems that their implementation on a group can bring certain benefits compared with individual relaxation. This outline is intended to raise this hypothesis in caring for individuals diagnosed with specific phobia, by approaching the problem from the cognitive behavioural therapy perspective. In addition, it seeks to evaluate the usefulness of the nurse intervention relaxation to reduce the level of anxiety, in turn comparing the results obtained using an indicator of the scale of results NOC and the Hamilton Anxiety Scale. The phobia is a specific entity underdiagnosed, whose prevalence is about 10%. Those affected can live a really limited and debilitating, deteriorating quality of life. The community mental health nurses are in a unique position to participate in the cognitive behavioural therapy through relaxation, which will allow them to reduce the level of anxiety when people establish contact with the phobic stimulus. Methodology: quasi-experimental study in specific phobia diagnosed, 20 to 40 years old adults attending for the first time to the mental health facility derived from primary care. Both the control group as the pilot will be treated by conducted cognitive-behavioural psychotherapy individualized according to the therapeutic protocol MSC, except in terms of relaxation, which in the experimental group will be conducted at the group level. The effectiveness of treatment will be assessed with the Hamilton anxiety scale and the likert type scale of outcome indicators NOC "stress level" with 3 measurements, before starting, immediately after completing the sessions of relaxation and three months later, checking the decline in the average level of anxiety.

La relajación es una técnica habitual utilizada por las enfermeras para disminuir el nivel de ansiedad. Parece que su puesta en práctica a nivel grupal puede aportar ciertos beneficios en comparación con la relajación individual. El presente proyecto pretende plantear esta hipótesis en la atención a las personas diagnosticadas de fobia específica, abordando el problema desde la perspectiva de la terapia cognitivo conductual. Asimismo, pretende evaluar la utilidad de la intervención enfermera de relajación para disminuir el nivel de ansiedad, comparando a su vez los resultados obtenidos al utilizar un indicador de la escala de resultados NOC y la Escala de Ansiedad de Hamilton. La fobia específica es una entidad infradiagnosticada, cuya prevalencia ronda el 10%. Pero las personas que la padecen pueden vivir una realidad limitada e incapacitante, deteriorando su calidad de vida de la persona. Las enfermeras de salud mental comunitaria están en una situación privilegiada de poder participar en la terapia cognitivo conductual mediante la relajación, que será la herramienta que les permita disminuir el nivel de ansiedad cuando las personas establezcan contacto con el estímulo fóbico. Metodología: Ensayo clínico cuasiexperimental, en adultos de 20 a 40 años diagnosticados de fobia específica que asistan por primera vez al centro de salud mental derivados de atención primaria. Tanto el grupo control como el experimental realizaran psicoterapia cognitiva-conductual individualizada según el protocolo terapéutico del CSM, salvo en lo referente a la relajación, que en el grupo experimental será llevada a cabo a nivel grupal. La eficacia del tratamiento se valorará con las escala de ansiedad de Hamilton y la escala tipo likert de los indicadores de resultado NOC “nivel de estrés” con 3 mediciones, antes de comenzar, inmediatamente después de terminar las sesiones de relajación y a los tres meses, verificando la disminución de la media del nivel de ansiedad. Presentamos una propuesta para aumentar la calidad de los cuidados enfermeros, mejorando la atención de salud mental de los pacientes hospitalizados, mediante la creación de la figura de la enfermera, dentro del equipo de interconsulta de psiquiatría de enlace. Esta enfermera, no sólo sería un referente de apoyo al personal de enfermería a nivel de cuidado del paciente, sino también del auto-cuidado psicoemocional del profesional. Objetivo: Justificar la necesidad de incluir la figura de la enfermera especialista en salud mental en el equipo de psiquiatría de enlace. Método: La rotación como residentes, durante un mes, con el equipo de interconsulta de psiquiatría del hospital Gregorio Marañón y revisión bibliográfica. Resultados: Se han observado necesidades parcialmente no satisfechas, estas podrían ser cubiertas con la existencia de una enfermera especialista en salud mental de interconsulta en este hospital. Discusión: Posiblemente se plantean dificultades a la hora de definir los roles de los distintos profesionales del equipo de enlace, que esperamos puedan ser definidos cuando se inicie la implantación del nuevo miembro. As residents of the mental health specialty, we take great care plan with methodology NANDA-NOC-NIC (NNN) in each of our rotations. Because of that there is still no consensus on how to use this methodology; we made this poster with our proposal. To do this, we propose a practical and standardized to carry out the care plans to guide future resident nurse of mental health in their care plans.Taking into account the review of various models of existing ratings of nursing at present, as well as taxonomy NNN, we carried out the development of a poster in which we show a practical example of how our proposed structure of a care plan:Evaluation: through functional patterns of M. Gordon Diagnosis: respecting the PES format: - (P) Problem: Using with the NANDA taxonomy - (E) Etiology: related factors- (S) Signs and symptoms: definitorial characteristics Planning: depending on the diagnosis set forth, the NOCs will agree. Performance indicators are also defined, specifying the current state, as well as the level to maintain /achieve and the time to be assessed. Lastly proposed interventions (NIC) and developed activities to be carried out to achieve the expected results. Implementation: implementation of interventions and activities Evaluation: we review the performance indicators, assessing the level reached in light of the current situation of the patient, and proposing corrective measures if necessary. As discussion could conclude that we used in our rotations this structure, so that we standardized "way we do." In turn, we have disseminated this structure in the National Congress of Mental Health Nursing.

Como enfermeras/os residentes de la especialidad de salud mental, realizamos un plan de cuidados con metodología NANDA-NIC-NOC (NNN) en cada uno de nuestros rotatorios. Debido a que todavía no hay un consenso sobre cómo utilizar esta metodología, realizamos este póster con nuestra propuesta. Para ello proponemos una forma práctica y estandarizada de llevar a cabo los planes de cuidados, de manera que podamos orientar a futuros residentes de enfermería en sus planes de cuidados Teniendo en cuenta la revisión de diversos modelos de valoraciones de enfermería existentes en la actualidad, así como la taxonomía NNN, llevamos a cabo la elaboración de un póster en el que mostramos un ejemplo práctico como nuestra propuesta de estructura de un plan de cuidados: Valoración: a través de los patrones funcionales de M. Gordon Diagnóstico: respetando el formato PES: - (P) Problema: enunciado con la taxonomía NANDA - (E) Etiología: factores relacionados - (S) Signos y síntomas: características definitorias Planificación: en función de los diagnósticos enunciados, se pactan los NOCs. También se definen los indicadores de resultados, concretando el estado actual, así como el nivel a mantener/conseguir y el tiempo en el que se volverán a evaluar. Por último se proponen las intervenciones (NIC) y se desarrollan las actividades a llevar a cabo para conseguir los resultados esperados. Ejecución: puesta en práctica de las intervenciones y actividades. Evaluación: se revisan los indicadores de resultados, evaluando el nivel alcanzado en función de la situación actual del paciente, y proponiendo medidas correctoras si fuese necesario. Como discusión podríamos concluir que hemos utilizado en nuestras rotaciones esta estructura, de forma que hemos estandarizado “nuestra forma de hacer”. A su vez, hemos difundido esta estructura en el Congreso Nacional de Enfermería de Salud Mental. Abstract: As residents of the mental health specialty, we take great care plan with methodology NANDA-NOC-NIC (NNN) in each of our rotations. Because of that there is still no consensus on how to use this methodology; we made this poster with our proposal. To do this, we propose a practical and standardized to carry out the care plans to guide future resident nurse of mental health in their care plans. Taking into account the review of various models of existing ratings of nursing at present, as well as taxonomy NNN, we carried out the development of a poster in which we show a practical example of how our proposed structure of a care plan: Evaluation: through functional patterns of M. Gordon Diagnosis: respecting the PES format: - (P) Problem: Using with the NANDA taxonomy - (E) Etiology: related factors - (S) Signs and symptoms: definitorial characteristics Planning: depending on the diagnosis set forth, the NOCs will agree. Performance indicators are also defined, specifying the current state, as well as the level to maintain /achieve and the time to be assessed. Lastly proposed interventions (NIC) and developed activities to be carried out to achieve the expected results. Implementation: implementation of interventions and activities Evaluation: we review the performance indicators, assessing the level reached in light of the current situation of the patient, and proposing corrective measures if necessary. The identification and selection of genetically superior animals for residual feed intake (RFI) could enhance productivity and minimize environmental impacts. The aim of this study was to use RNA-seq data to identify the differentially expressed genes (DEGs), known non-coding RNAs (ncRNAs), specific biomarkers and enriched biological processes associated with RFI of the liver in Nellore cattle in two genetic groups. In genetic group 1 (G1), 24 extreme RFI animals (12 low RFI (LRFI) versus 12 high RFI (HRFI)) were selected from a population of 60 Nellore bulls. The RNA-seq of the samples from their liver tissues was performed using an Illumina HiSeq 2000. In genetic group 2 (G2), 20 samples of liver tissue of Nellore bulls divergent for RFI (LRFI, n = 10 versus HRFI, n = 10) were selected from 83 animals. The raw data of the G2 were chosen from the ENA repository. A total of 1811 DEGs were found for the G1 and 2054 for the G2 (p-value ≤ 0.05). We detected 88 common genes in both genetic groups, of which 33 were involved in the immune response and in blocking oxidative stress. In addition, seven (B2M, ADSS, SNX2, TUBA4A, ARHGAP18, MECR, and ABCF3) possible gene biomarkers were identified through a receiver operating characteristic analysis (ROC) considering an AUC > 0.70. The B2M gene was overexpressed in the LRFI group. This gene regulates the lipid metabolism protein turnover and inhibits cell death. We also found non-coding RNAs in both groups. MIR25 was up-regulated and SNORD16 was down-regulated in the LRFI for G1. For G2, up-regulated RNase_MRP and SCARNA10 were found. We highlight MIR25 as being able to act by blocking cytotoxicity and oxidative stress and RMRP as a blocker of mitochondrial damage. The biological pathways associated with RFI of the liver in Nellore cattle in the two genetic groups were for energy metabolism, protein turnover, redox homeostasis and the immune response. The common transcripts, biomarkers and metabolic pathways found in the two genetic groups make this unprecedented work even more relevant, since the results are valid for different herds raised in different ways. The results reinforce the biological importance of these known processes but also reveal new insights into the complexity of the liver tissue transcriptome of Nellore cattle.

BACKGROUND/OBJECTIVE:Patients with non-small cell lung cancer (NSCLC) develop resistance to antitumor agents by mechanisms that involve the epithelial-to-mesenchymal transition (EMT). This necessitates the development of new complementary drugs, e.g., cannabinoid receptors (CB1 and CB2) agonists including tetrahydrocannabinol (THC) and cannabidiol (CBD). The combined use of THC and CBD confers greater benefits, as CBD enhances the effects of THC and reduces its psychotropic activity. We assessed the relationship between the expression levels of CB1 and CB2 to the clinical features of a cohort of patients with NSCLC, and the effect of THC and CBD (individually and in combination) on proliferation, EMT and migration in vitro in A549, H460 and H1792 lung cancer cell lines. METHODS:Expression levels of CB1, CB2, EGFR, CDH1, CDH2 and VIM were evaluated by quantitative reverse transcription-polymerase chain reaction. THC and CBD (10-100 μM), individually or in combination (1:1 ratio), were used for in vitro assays. Cell proliferation was determined by BrdU incorporation assay. Morphological changes in the cells were visualized by phase-contrast and fluorescence microscopy. Migration was studied by scratch recolonization induced by 20 ng/ml epidermal growth factor (EGF). RESULTS:The tumor samples were classified according to the level of expression of CB1, CB2, or both. Patients with high expression levels of CB1, CB2, and CB1/CB2 showed increased survival reaching significance for CB1 and CB1/CB2 (p = 0.035 and 0.025, respectively). Both cannabinoid agonists inhibited the proliferation and expression of EGFR in lung cancer cells, and CBD potentiated the effect of THC. THC and CBD alone or in combination restored the epithelial phenotype, as evidenced by increased expression of CDH1 and reduced expression of CDH2 and VIM, as well as by fluorescence analysis of cellular cytoskeleton. Finally, both cannabinoids reduced the in vitro migration of the three lung cancer cells lines used. CONCLUSIONS:The expression levels of CB1 and CB2 have a potential use as markers of survival in patients with NSCLC. THC and CBD inhibited the proliferation and expression of EGFR in the lung cancer cells studied. Finally, the THC/CBD combination restored the epithelial phenotype in vitro.

Interleukin-6 (IL-6) is involved in lung cancer tumorigenesis, tumor progression, metastasis, and drug resistance. Previous studies show that blockade of IL-6 signaling can inhibit tumor growth and increase drug sensitivity in mouse models. Clinical trials in non-small cell lung cancer (NSCLC) reveal that IL-6 targeted therapy relieves NSCLC-related anemia and cachexia, although other clinical effects require further study. We crossed IL-6(-/-) mice with Kras(G12D) mutant mice, which develop lung tumors after activation of mutant Kras(G12D), to investigate whether IL-6 inhibition contributes to tumor progression and survival time in vivo. Kras(G12D); IL-6(-/-) mice exhibited increased tumorigenesis, but slower tumor growth and longer survival, than Kras(G12D) mice. Further, in order to investigate whether IL-6 deletion contributes to suppression of lung cancer metastasis, we generated Kras(G12D); p53(flox/flox); IL-6(-/-) mice, which developed lung cancer with a trend for reduced metastases and longer survival than Kras(G12D); p53(flox/flox) mice. Tumors from Kras(G12D); IL-6(-/-) mice showed increased expression of TNFα and decreased expression of CCL-19, CCL-20 and phosphorylated STAT3(pSTAT3) than Kras(G12D) mice; however, these changes were not present between tumors from Kras(G12D); p53(flox/flox); IL-6(-/-) and Kras(G12D); p53(flox/flox) mice. Upregulation of pSTAT3 and phosphorylated AKT(pAKT) were observed in Kras(G12D) tumors with p53 deletion. Taken together, these results indicate that IL-6 deletion accelerates tumorigenesis but delays tumor progression and prolongs survival time in a Kras-driven mouse model of lung cancer. However, these effects can be attenuated by p53 deletion.
