Abstract

Este documento resume estudios realizados en variabilidad y control genético, así­ como efectos ambientales sobre la producción de semilla en alfalfa, y propone un criterio y un esquema de selección para aumentar esta producción. La producción de semilla y sus componentes presentan alta variabilidad genética inter e intra-varietal. Las mayores heredabilidades, en el sentido estricto, están en la producción de semilla por planta (0.52), peso de semillas por inflorescencia (0.56) y níºmero de semillas por vaina (0.56), siendo la varianza aditiva la principal fuente de variación genética. Existe la interacción genotipo x medio para la producción de semilla, por el contrario, el peso de semillas por inflorescencia presenta estabilidad al medio y está correlacionado genéticamente con la producción de semilla (0.92, P

4. Association study between the gibberellic acid insensitive gene and leaf length in a Lolium perenne L. synthetic variety

Abstract

Abstract Background Association studies are of great interest to identify genes explaining trait variation since they deal with more than just a few alleles like classical QTL analyses. They are usually performed using collections representing a wide range of variability but which could present a genetic substructure. The aim of this paper is to demonstrate that association studies can be performed using synthetic varieties obtained after several panmictic generations. This demonstration is based on an example of association between the gibberellic acid insensitive gene (GAI) polymorphism and leaf length polymorphism in 'Herbie', a synthetic variety of perennial ryegrass. Methods Leaf growth parameters, consisted of leaf length, maximum leaf elongation rate (LERmax) and leaf elongation duration (LED), were evaluated in spring and autumn on 216 plants of Herbie with three replicates. For each plant, a sequence of 370 bp in GAI was analysed for polymorphism. Results Genetic effect was highly significant for all traits. Broad sense heritabilities were higher for leaf length and LERmax with about 0.7 in each period and 0.5 considering both periods than for LED with about 0.4 in each period and 0.3 considering both periods. GAI was highly polymorphic with an average of 12 bp between two consecutive SNPs and 39 haplotypes in which 9 were more frequent. Linkage disequilibrium declined rapidly with distance with r 2 values lower than 0.2 beyond 150 bp. Sequence polymorphism of GAI explained 8-14% of leaf growth parameter variation. A single SNP explained 4% of the phenotypic variance of leaf length in both periods which represents a difference of 33 mm on an average of 300 mm. Conclusions Synthetic varieties in which linkage disequilibrium declines rapidly with distance are suitable for association studies using the "candidate gene" approach. GAI polymorphism was found to be associated with leaf length polymorphism which was more correlated to LERmax than to LED in Herbie. It is a good candidate to explain leaf length variation in other plant material.

5. Construction of two genetic linkage maps in cultivated tetraploid alfalfa (Medicago sativa) using microsatellite and AFLP markers

Abstract

Abstract Background Alfalfa (Medicago sativa) is a major forage crop. The genetic progress is slow in this legume species because of its autotetraploidy and allogamy. The genetic structure of this species makes the construction of genetic maps difficult. To reach this objective, and to be able to detect QTLs in segregating populations, we used the available codominant microsatellite markers (SSRs), most of them identified in the model legume Medicago truncatula from EST database. A genetic map was constructed with AFLP and SSR markers using specific mapping procedures for autotetraploids. The tetrasomic inheritance was analysed in an alfalfa mapping population. Results We have demonstrated that 80% of primer pairs defined on each side of SSR motifs in M. truncatula EST database amplify with the alfalfa DNA. Using a F1 mapping population of 168 individuals produced from the cross of 2 heterozygous parental plants from Magali and Mercedes cultivars, we obtained 599 AFLP markers and 107 SSR loci. All but 3 SSR loci showed a clear tetrasomic inheritance. For most of the SSR loci, the double-reduction was not significant. For the other loci no specific genotypes were produced, so the significant double-reduction could arise from segregation distortion. For each parent, the genetic map contained 8 groups of four homologous chromosomes. The lengths of the maps were 2649 and 3045 cM, with an average distance of 7.6 and 9.0 cM between markers, for Magali and Mercedes parents, respectively. Using only the SSR markers, we built a composite map covering 709 cM. Conclusions Compared to diploid alfalfa genetic maps, our maps cover about 88–100% of the genome and are close to saturation. The inheritance of the codominant markers (SSR) and the pattern of linkage repulsions between markers within each homology group are consistent with the hypothesis of a tetrasomic meiosis in alfalfa. Except for 2 out of 107 SSR markers, we found a similar order of markers on the chromosomes between the tetraploid alfalfa and M. truncatula genomes indicating a high level of colinearity between these two species. These maps will be a valuable tool for alfalfa breeding and are being used to locate QTLs.

7. Impact of intraspecific genetic variation on interspecific competition: a theoretical case study of forage binary mixtures.

Abstract

Introduction: Increasing intraspecific genetic variation (IV) has been identified as a potential factor to improve productivity and stabilise botanical composition in plant communities. In grasslands systems, this could offer a lever to manage uncertainties of production and variability in the harvested species balance. However, little is known about the conditions to favour IV impact and the mechanisms at play. Methods: The dependency of IV impact on traits holding it and environmental stressors were analysed using a spatially-explicit individual-based model (IBM) of grassland communities. Sixty-three binary mixtures were defined to reflect a gradient of functional divergence between species regarding light and nitrogen (N) acquisition. The growth and dynamics of these communities were simulated for one year with three possible IV levels under two environments contrasting in terms of soil N fertility. Results and discussion: The model predicted a positive impact of moderate and high IV levels on maintaining the species balance over time, but no marked effects on mixture productivity. This stabilising effect increased at higher IV levels and under low soil N fertility. It also tended to be more pronounced in communities with intermediate functional divergence offering a significant overlap between light and N acquisition parameter values of both species. The major traits involved in the plant response to neighbours differed depending on the most contested resource, as indicated by the within-population selection of individuals with favourable N-related parameters under low N and light-related parameters under high N environments. The hypothesis that IV favours a complementarity of resource use between species was not supported. Rather, a greater spatial heterogeneity in competitive interactions was demonstrated, leading to a higher probability of growth and survival for individuals within the subordinate species. Abstract

Legumes are essential for providing protein-rich products and introducing nitrogen into cropping systems. Perennial species, mainly grown for fodder production but also as service plants, contribute to the protein autonomy of livestock. Perennial legumes also contribute to many other ecosystem services: soil fertility through the supply of nitrogen coupled with organic matter, weed management, reduction of pollution emitted by ruminant livestock, and biodiversity preservation. Able to adapt to climate change, perennial legumes remain valuable under different food transition scenarios. The article suggests avenues of research in agronomy, plant breeding and animal husbandry to further increase these services, Les légumineuses sont indispensables pour fournir des produits riches en protéines et introduire de l’azote dans les systèmes de culture. Les espèces pérennes, essentiellement cultivées pour une production fourragère, mais aussi comme plantes de services, contribuent à l’autonomie protéique des élevages. Les légumineuses pérennes contribuent aussi à de nombreux autres services écosystémiques : fertilité des sols grâce à la fourniture d’azote couplé à la matière organique, gestion des adventices, réduction des pollutions émises par les élevages de ruminants, préservation de la biodiversité. Capables de s’adapter au changement climatique, les légumineuses pérennes restent précieuses sous différents scénarios de transition alimentaire. L’article suggère des pistes de recherche en agronomie, amélioration des plantes et zootechnie pour accroître encore ces services.

Abstract

International audience; GBS markers at the population level were used to study the genetic structure of lucerne in a set of 395 accessions from Europe, North and South America and China. More than 100K SNP without missing data were obtained. The structure revealed a certain genetic distance between European and American accessions, with a continuous variation between these groups. Contrastingly, Chinese accessions were genetically separated from the other accessions. This analysis is an invitation to go more deeply into a phenotypic analysis of these genetic resources to exploit them in breeding.

International audience; GBS is an efficient genotyping tool for heterozygous and polyploid species, yet the fraction of missing data, inherent to the method, may be detrimental to genetic analyses. We tested several restriction enzymes to choose the ones that were likely to provide the maximum number of loci with little missing data considering a certain sequencing effort. For lucerne and cocksfoot, the enzymes that met the target of about 10 000 loci were the combination of PstI-MseI for lucerne and PstI for cocksfoot. 1 066 lucerne accessions were genotyped with PstI-MseI, and more than 200 000 SNP with less than 5% missing data were obtained. Based on these results, we recommend performing such a test to optimize GBS genotyping efforts in forage species.

Abstract

International audience; Genomic prediction has proven its efficiency in numerous animal and plant species. In this study, we used diverse lucerne varieties and populations to test the predicting ability of genomic prediction. Several parameters, such as the number of markers, the population size and the addition of QTL effects, were tested for their effect on the quality of prediction. Based on a large number of SNPs (227 K) obtained by GBS and phenotypes observed in different locations, our results showed a good quality of predicting ability for dry matter yield, ADF (acid detergent fiber) and protein content, especially with a large training population size (around 0.6). The predicting ability is improved by the integration of QTL information directly in the model (above 0.8). A reduction of number of markers (less than 100K) did not alter much the predictive ability. Our results show an accurate prediction of the phenotype of populations via genomic prediction models that could speed up the creation of new lucerne varieties.

International audience; EUCLEG is a project funded by EU, from 2017 to 2021, entitled “Breeding forage and grain legumes to increase EU’s and China’s protein self-sufficiency”. EUCLEG is developing improved breeding strategies for legume species that are commercially important, it focuses on two main forage species (alfalfa and red clover) and three grain species (pea, faba bean and soybean). Objectives and first results are presented illustrating the huge diversity available and the genetic progress that could be achieved through molecular breeding.

Abstract

Wider and more profitable legume crop cultivation is an indispensable step for the agroecological transition of global agri-food systems but represents a challenge especially in Europe. Plant breeding is pivotal in this context. Research areas of key interest are represented by innovative phenotypic and genome-based selection procedures for crop yield, tolerance to abiotic and biotic stresses enhanced by the changing climate, intercropping, and emerging crop quality traits. We see outmost priority in the exploration of genomic selection (GS) opportunities and limitations, to ease genetic gains and to limit the costs of multi-trait selection. Reducing the profitability gap of legumes relative to major cereals will not be possible in Europe without public funding devoted to crop improvement research, pre-breeding, and, in various circumstances, public breeding. While most of these activities may profit of significant public-private partnerships, all of them can provide substantial benefits to seed companies. A favorable institutional context may comprise some changes to variety registration tests and procedures.
