Objective: Despite advancements in spinal metastasis surgery techniques and the rapid development of multidisciplinary treatment models, we aimed to explore the clinical efficacy of spinal metastasis surgery performed by a combined NOMS decision system-utilizing multidisciplinary team and Revised Tokuhashi scoring system, compared with the Revised Tokuhashi scoring system. Methods: Clinical data from 102 patients with spinal metastases who underwent surgery at three affiliated hospitals of Zunyi Medical University from December 2017 to June 2022 were analysed. The patients were randomly assigned to two groups: 52 patients in the treatment group involving the combined NOMS decision system-utilizing multidisciplinary team and Revised Tokuhashi scoring system (i.e., the combined group), and 50 patients in the treatment group involving the Revised Tokuhashi scoring system only (i.e., the revised TSS-only group). Moreover, there were no statistically significant differences in preoperative general data or indicators between the two groups. Intraoperative and postoperative complications, average hospital stay, mortality rate, and follow-up observation indicators, including the visual analogue scale (VAS) score for pain, Eastern Cooperative Oncology Group (ECOG) performance status, Karnofsky Performance Status (KPS) score, negative psychological assessment score (using the Self-Rating Anxiety Scale, [SAS]), and neurological function recovery score (Frankel functional classification) were compared between the two groups. Results: All 102 patients successfully completed surgery and were discharged. The follow-up period ranged from 12 to 24 months, with an average of (13.2 ± 2.4) months. The patients in the combined group experienced fewer complications such as surgical wound infections 3 patients(5.77%), intraoperative massive haemorrhage 2 patients(3.85%), cerebrospinal fluid leakage 2 patients(3.85%), deep vein thrombosis 4 patients(7.69%),and neurological damage 1 patient(1.92%), than patients in the revised TSS-only group (wound infections,11 patients(22%); intraoperative massive haemorrhage, 8 patients(16%);cerebrospinal fluid leakage,5 patients(10%);deep vein thrombosis,13 patients (26%); neurological damage,2 patients (4%). Significant differences were found between the two groups in terms of surgical wound infections, intraoperative massive haemorrhage, and deep vein thrombosis (P < 0.05). The average postoperative hospital stay in the combined group (7.94 ± 0.28 days) was significantly shorter than that in the revised TSS-only group (10.33 ± 0.30 days) (P < 0.05). Long-term follow-up (1 month, 3 months, 6 months, and 1 year postoperatively) revealed better clinical outcomes in the combined group than in the revised TSS-only group in terms of VAS scores, overall KPS%, neurological function status Frankel classification, ECOG performance status, and SAS scores.(P < 0.05). Conclusion: A multidisciplinary team using the NOMS combined with the Revised Tokuhashi scoring system for spinal metastasis surgery showed better clinical efficacy than the sole use of the Revised Tokuhashi scoring system. This personalized, precise, and rational treatment significantly improves patient quality of life, shortens hospital stay, reduces intraoperative and postoperative complications, and lowers mortality rates. Substantial research indicates that students' beliefs mediate learning strategies. Nevertheless, students' strategy-related beliefs about feedback and their mediation on strategies to act on feedback are insufficiently addressed. This case study aimed to examine: (1) students' strategy-related beliefs about feedback in L2 writing and (2) how beliefs mediated students' actions when a feedback intervention was implemented to facilitate students' use of feedback strategies. Data were collected from semi-structured interviews and reflective journals from 10 Year-1 participants who studied in an associate degree programme at a higher education institute in Hong Kong. Students' strategy-related beliefs about feedback encompass the evaluation of strategies about: (1) understanding feedback, (2) addressing areas for improvement, (3) seeking feedback, and (4) implementing feedback. Nine participants adopted a more comprehensive range of strategies to act on feedback consistent with their strategy-related beliefs. One participant, however, did not appreciate and use feedback. Background: Enumeration of circulating tumor cells (CTCs) has proven clinical significance for monitoring patients with metastatic cancers. Multiplexed gene expression profiling of CTCs is a potential tool for assessing disease status and monitoring treatment response. The Parsortix® technology enables the capture and harvest of CTCs from blood based on cell size and deformability. The HyCEAD™ (Hybrid Capture Enrichment Amplification and Detection) assay enables simultaneous amplification of short amplicons for up to 100 mRNA targets, and the Ziplex™ instrument quantifies the amplicons for highly sensitive gene expression profiling down to single cell levels. The aim of the study was to functionally assess this system. Methods: The HyCEAD/Ziplex platform was used to quantify the expression levels for 72 genes using as little as 20 pg of total RNA or a single cultured tumor cell. Assay performance was evaluated using cells or total RNA spiked into Parsortix harvests of healthy donor blood. The assay was also evaluated using total RNA obtained from Parsortix harvests of blood from metastatic breast cancer (MBC) patients or healthy volunteers (HVs). Results: Using genes with low expression in WBC RNA and/or in unspiked Parsortix harvests from HVs, the assay distinguished between the different breast cancer and ovarian cancer cell lines with as little as 20 pg of total RNA (equivalent to a single cell) in the presence of 1 ng of WBC RNA. Single cultured cells spiked into Parsortix harvests from 10 mL of HV blood were also detected and distinguished from each other. CVs from repeatability experiments were less than 20%. Hierarchical clustering of clinical samples differentiated most MBC patients from HVs. Conclusion: HyCEAD/Ziplex provided sensitive quantification of expression of 72 genes from 20 pg of total RNA from cultured tumor cell lines or from single cultured tumor cells spiked into lysates from Parsortix harvests of HV blood. The HyCEAD/Ziplex platform enables the quantification of selected genes in the presence of residual nucleated blood cells in Parsortix harvests. The social organization of groups varies greatly across primate species, ranging from egalitarian to despotic. Moreover, the typical or average size of groups varies greatly across primate species. Yet we know little about how group size affects social organization across primate species. Here we used the hawk-dove game (HDG) to model the evolution of social organization as a function of maximum group size and used the evolved frequency of hawks as a measure of egalitarian/despotism in societies. That is, the lower the frequency of hawks, the more egalitarian a society is, and the higher the frequency of hawks, the more despotic it is. To do this, we built an agent-based model in which agents live in groups and play the HDG with fellow group members to obtain resources to reproduce offspring. Offspring inherit the strategy of their parent (hawk or dove) with a low mutation rate. When groups reach a specified maximum size, they are randomly divided into two groups. We show that the evolved frequency of hawks is dramatically lower for relatively small maximum group sizes than predicted analytically for the HDG. We discuss the relevance of group size for understanding and modeling primate social systems, including the transition from hunter-gather societies to agricultural societies of the Neolithic era. The N-acyl-D-amino acid amidohydrolase (N-d-AAase) of Variovorax paradoxus Iso1 can enantioselectively catalyze the zinc-assisted deacetylation of N-acyl-Damino acids to yield consistent D-amino acids. A putative FAD-binding glycine/Damino acid oxidase was located immediately upstream of the N-d-AAase gene. The gene encoding this protein was cloned into Escherichia coli BL21 (DE3)pLysS and overexpressed at 25°C for 6 h with 0.5 mM isopropyl β-D-1-thiogalactopyranoside induction. After purification, the tag-free recombinant protein was obtained. The enzyme could metabolize glycine, sarcosine, and D-alanine, but not L-amino acids or bulky D-amino acids. Protein modeling further supported these results, demonstrating that glycine, sarcosine, and D-alanine could fit into the pocket of the enzyme's activation site, while L-alanine and D-threonine were out of position. Therefore, this protein was proposed as a glycine oxidase, and we designated it VpGO. Interestingly, VpGO showed low sequence similarity to other well-characterized glycine oxidases. We found that VpGO and N-d-AAase were expressed on the same mRNA and could be transcriptionally induced by various N-acetyl-D-amino acids. Western blotting and zymography showed that both proteins had similar expression patterns in response to different types of inducers. Thus, we have identified a novel glycine oxidase that is co-regulated with N-d-AAase in an operon, and metabolizes N-acyl-D-amino acids in the metabolically versatile V. paradoxus Iso1. • The resting effect is another version of the spacing effect, which is explained by the working memory resource depletion. • Under different conditions, the spacing effect can be explained by both working memory resource depletion and by mental rehearsal. • Working memory resource depletion is more sensitive to materials high in element interactivity. • For low element interactivity information, mental rehearsal is likely to happen automatically during wakeful resting. • Measures of working memory resource depletion can be used to determine whether it is a relevant cause. The spacing effect occurs when learning with rest periods is superior to learning without rest periods. Cognitive load theory has explained this superiority by working memory resource depletion, under which resources are depleted during cognitive activity but restored with rest. In a series of four experiments involving 341 participants, we explored the relationships between the spacing effect, depletion of working memory resources, and mental rehearsal, particularly focusing on how these dynamics are influenced by task complexity as defined by element interactivity. Experiment 1 showed that materials with higher element interactivity had a greater impact on working memory resource depletion. In Experiment 2, using materials low in element interactivity, a spacing effect was obtained with no evidence of working memory resource depletion. Instead, results suggested that the effect might be due to mental rehearsal occurring during rest periods. Experiment 3, using more complex information, obtained both the spacing and working memory resource depletion effects for less knowledgeable learners for whom the information was high in element interactivity. In Experiment 4, testing more knowledgeable learners for whom the same information was lower in element interactivity, both effects disappeared. The results indicated that working memory resource depletion and recovery may be more sensitive to materials high in element interactivity and suggest that it is only one of multiple causes of the spacing effect. In data-driven residential care settings, it is sometimes necessary to change an established assessment tool due to cost and respondent burden. However, accurate assessment of youth functioning over time or across samples can be compromised when different assessments are used. Hence, the ability to crosswalk between assessments is important. The Child Behavior Checklist (CBCL) and Strengths and Difficulties Questionnaire (SDQ) screen for similar attributes and are highly correlated. The SDQ is shorter and free and can be an alternative to the CBCL. This study links the CBCL and SDQ total problem scores in a sample of 284 high-risk youth in a residential care facility, using equipercentile equating. Youth had both assessments completed on them concurrently by caregivers. The "equate" package in R Studio was used to conduct the analysis. Concordance tables were created for crosswalks from CBCL to SDQ and vice versa. Results showed linked scores that were consistent when equating from each assessment to the other. The concordance tables can serve as a comparison tool between total problem scores of each assessment. Low birthweight and chronic low-grade inflammation have been significantly associated with an increased risk of cardiovascular disease (CVD) [[1]]. Adults born preterm at a very low birthweight (VLBW) of <1500 g represent the low end of the low birthweight spectrum and have increased levels of CVD risk factors [4]. Follow-up population studies have reported higher CRP concentrations in low birthweight children and adults [[5], [7]] but the effect of VLBW on CRP in young adulthood needs to be further explored. This difference may be related to study design, as our study focused on those born preterm at a VLBW, while population-based studies have included the whole range of birthweights but included very few VLBW individuals by design. Collective cell migration plays important roles in various physiological processes. To investigate this collective cellular movement, various wound-healing assays have been developed. In these assays, a "wound" is created mechanically, chemically, optically, or electrically out of a cellular monolayer. Most of these assays are subject to drawbacks of run-to-run variations in wound size/shape and damages to cells/substrate. Moreover, in all these assays, cells are cultured in open, static (non-circulating) environments. In this study, we reported a microfluidics-based wound-healing assay by using the trypsin flow-focusing technique. Fibroblasts were first cultured inside this chip to a cellular monolayer. Then three parallel fluidic flows (containing normal medium and trypsin solution) were introduced into the channels, and cells exposed to protease trypsin were enzymatically detached from the surface. Wounds of three different widths were generated, and subsequent wound-healing processes were observed. This assay is capable of creating three or more wounds of different widths for investigating the effects of various physical and chemical stimuli on wound-healing speeds. The effects of shear stresses, wound widths, and β-lapachone (a wound healing-promoting chemical) on wound-healing speeds were studied. It was found that the wound-healing speed (total area healed per unit time) increased with increasing shear stress and wound width, but under a shear stress of 0.174 mPa the linear healing speed (percent area healed per unit time) was independent of the wound width. Also, the addition of β-lapachone up to 0.5 μM did not accelerate wound healing. Objective: The purpose of this paper is to introduce a rubric for both researchers and consumers of research to guide best practices in the conduct of exploratory and confirmatory subgroup analyses studies. Methods: Paper draws on a review of the literature on subgroup analyses techniques and studies on racial and ethnic disparities in behavioral, mental, and physical health outcomes. Results: The paper highlights theoretical, methodological, and interpretational challenges in the use of subgroup analyses and illustrates them with specific examples. Conclusion: The paper concludes with a series of specific recommendations for the development of subgroup analyses specifically within three practice areas: theoretical, methodological, and interpretational. Such recommendations are captured in a rubric of best practices that aim to strengthen the evidence-base derived from subgroup studies to guide treatment and prevention interventions as well as the design and evaluation of effective policy recommendations to ameliorate health disparities. Developing a multiple functional wound dressing suitable for different stages of wound healing is important for patients with special wound such as burn or decubital ulcers. In this study, poly (vinyl alcohol) (PVA), dextran, and chitosan are integrated to produce ideal wound dressing where glutaraldehyde (GA) is used as the cross-linker. The result demonstrated that 6% PVA hydrogel with 0.25% chitosan was found to provide antimicrobial ability. The PVA/chitosan hydrogel combined with 4% dextran utilizing GA cross-linking also presents the high cell proliferation ability, which suggests that the hydrogel is potential as a wound dressing. In the following physical analyses, the addition of chitosan and dextran appeared to promote the thermostability, mechanical properties, water retention, and moisturizing ability in the PVA hydrogel. In conclusion, the PVA/chitosan/dextran hydrogel has promising potential such as high water content, antimicrobial property, and well cell proliferation, which can be applied to wound healing application. Abstract Ethnopharmacological relevance We know little about US use of Chinese herbal products (CHP). Aim of the study: To understand CHP users' sociodemographic characteristics, CHP indications for use, and prescribing patterns in an acupuncture teaching center in the United States. Materials and methods Retrospective chart review and descriptive statistics. Results CHP users were mostly female and white with at least a college education. The most common main complaint for CHP treatment was genitourinary system. Most of the CHP users were given thirteen different herbs per prescription, and the mean number of herbs per CHP prescription was twelve. The most common dosing frequency was twice a day (BID) and the most common duration of prescription was seven days. Dang Gui / Angelicae sinensis (Oliv.) Diels was the most frequently prescribed Chinese herb. Conclusion We identified CHP users' sociodemographic characteristics and CHP prescription patterns in a teaching center in the United States. The introduction of left ventricular assist device (LVAD) has improved survival rates for patients with end-stage heart failure. Two categories of VADs exist: one generates pulsatile flow and the other produces nonpulsatile continuous flow. Survival is better for patients with continuous-flow LVADs. With improved survival, more of such patients now present for noncardiac surgery (NCS). This review, written for the general anesthesiologists, addresses the perioperative considerations when the patient undergoes NCS. Standard wound healing assays often deal with just one initial wound shape, and it is unclear whether varying the initial wound shape might impact how we interpret results from these experiments. Here, we use a new kind of wound healing assay, called a sticker assay , to explore the role of initial wound shape. We examine wound healing with square, circular and triangular wounds. Measurements of wound area as a function of time show that the rate of wound closure depends on the initial wound shape. This is interesting because the only aspect of the assay we change is the initial wound shape, and the reason for different closure rates is unclear. To provide more insight we calibrate a typical mathematical model to match the data. This shows that the rates of cell motility and proliferation associated with different initial wound shapes are approximately the same, implying that the differences in the closure rates are consistent with a typical mathematical model of wound healing. In this work, a scalable method was developed to produce graphene oxide (GO) via high-shearing of pre-oxidized graphite. Various oxidation states of pre-oxidized graphite were obtained following different oxidation periods through a modified Hummers’ method. The oxidized graphite flakes were investigated by X-ray diffractometry, Fourier transform infrared spectroscopy, and Raman spectroscopy. Instead of a prolonged oxidation period of more than 24 hours, the graphite sample that underwent 6 hours of oxidation was selected for the high-shearing process to obtain exfoliated GO. The transmission electron microscopy and atomic force microscopy results confirmed that large-area and single-layer GO can be produced by the high-shear exfoliation method with pre-oxidized graphite flakes. A novel and non-invasive sampling method was developed for use in electrospray ionization/mass spectrometry (ESI/MS). The sampling tool, a watercolor pen (brush) that had been rinsed with ethanol, was used to collect analytes from the eyelids of volunteers. Following this, the brush was quickly moved to the front of the mass inlet, where the ethanol as well as the analytes evaporated very fast and escaped from the brush surface. When the analytes make contact with the ESI plume, which arises from the ESI needle tip, they are ionized and then detected by a mass spectrometer. The findings show that this new technique is applicable for the analysis of caffeine and its metabolites in samples obtained from eyelids after the volunteers consumed coffee. The methodology is simple and economical, non-invasive, and is suitable for use in monitoring the levels of caffeine and its metabolites over an extended period of time. [ABSTRACT FROM AUTHOR]

Due to the presence of unique micro/nanostructures on the surface, fish's scale exhibits underwater superoleophobicity and keeps clean even in oil-polluted water. Inspired from this, we propose a facile method for the fabrication of underwater superoleophobic and anti-oil sapphire surface with line-patterned nanostructures by femtosecond laser. The as-prepared surface shows great superoleophobicity that the oil contact angles can reach up to 153° for 1, 2-dichloroethane droplets in water and low oil-adhesion. At the same time, the relationship between the microgrooves' period and surface wettability is studied, and the results indicate that the underwater superoleophobicity and low oil-adhesion can be achieved using a wide range of processing parameters. Meanwhile, the obtained surface is demonstrated to exhibit excellent stability. Moreover, the self-cleaning anti-oil ability of the as-prepared surface is conducted, and the potential mechanism of which is discussed. Glaucoma is the most common optic neuropathy in humans and the leading cause of irreversible blindness worldwide. Its prevalence and incidence increase exponentially with ageing and raised intraocular pressure (IOP), while increasing evidence suggests that systemic mitochondrial abnormalities may also be implicated in its pathogenesis. We have recently shown that patients who have not developed glaucoma despite being exposed for many years to high IOP (ocular hypertension - OHT) have more efficient mitochondria, measured in peripheral blood lymphocytes, when compared to age-similar controls and fast progressing normal tension glaucoma (NTG) patients. In this prospective case series we aimed to explore some of the molecular pathways involved in mitochondrial efficiency in glaucoma resistance by measuring the systemic activity (in peripheral blood) of key mitochondrial regulators: the mammalian target of rapamycin (mTOR) and its major upstream regulators and downstream effectors that form the PTEN-Akt1-mTOR signalling pathway. We found no statistically significant difference in the systemic mTOR activity between the three groups (control, NTG and OHT). In line with the mTOR results, there was no significant difference in the activity of both the two major upstream mTOR regulators (PTEN and Akt1) and its two main downstream effectors (S6K and 4EBP1). In a single NTG patient, with history of Raynaud's, significantly higher mTOR activity was noted. The worked example effect has been well documented within the framework of Cognitive Load Theory (CLT), which suggests that teaching with examples would be superior to engaging in unguided problem solving, particularly for novices, as using worked examples would reduce their cognitive load, compared to solving problems, thus facilitating knowledge retention. This paper, using multiple-step mathematics problems, reports an experiment investigating the micro level of the worked example effect on learning solution steps, from the perspective of cognitive load and challenge (as a relevant affective, motivational factor), testing the worked example effect with a transfer test. The results favoured worked examples on both the retention and transfer tests after learning and showed that using worked examples would reduce cognitive load and impose less challenge on each step during learning. Background: Directed cell migration can be driven by a range of external stimuli, such as spatial gradients of: chemical signals (chemotaxis); adhesion sites (haptotaxis); or temperature (thermotaxis). Continuum models of cell migration typically include a diffusion term to capture the undirected component of cell motility and an advection term to capture the directed component of cell motility. However, there is no consensus in the literature about the form that the advection term takes. Some theoretical studies suggest that the advection term ought to include receptor saturation effects. However, others adopt a much simpler constant coefficient. One of the limitations of including receptor saturation effects is that it introduces several additional unknown parameters into the model. Therefore, a relevant research question is to investigate whether directed cell migration is best described by a simple constant tactic coefficient or a more complicated model incorporating saturation effects. Results: We study directed cell migration using an experimental device in which the directed component of the cell motility is driven by a spatial gradient of electric potential, which is known as electrotaxis. The electric field (EF) is proportional to the spatial gradient of the electric potential. The spatial variation of electric potential across the experimental device varies in such a way that there are several subregions on the device in which the EF takes on different values that are approximately constant within those subregions. We use cell trajectory data to quantify the motion of 3T3 fibroblast cells at different locations on the device to examine how different values of the EF influences cell motility. The undirected (random) motility of the cells is quantified in terms of the cell diffusivity, D, and the directed motility is quantified in terms of a cell drift velocity, v. Estimates D and v are obtained under a range of four different EF conditions, which correspond to normal physiological conditions. Our results suggest that there is no anisotropy in D, and that D appears to be approximately independent of the EF and the electric potential. The drift velocity increases approximately linearly with the EF, suggesting that the simplest linear advection term, with no additional saturation parameters, provides a good explanation of these physiologically relevant data. Conclusions: We find that the simplest linear advection term in a continuum model of directed cell motility is sufficient to describe a range of different electrotaxis experiments for 3T3 fibroblast cells subject to normal physiological values of the electric field. This is useful information because alternative models that include saturation effects involve additional parameters that need to be estimated before a partial differential equation model can be applied to interpret or predict a cell migration experiment. Stakeholder engagement in Environmental Impact Assessment (EIA) and Health Impact Assessment (HIA) provides opportunities for inclusive environmental decision-making contributing to the attainment of agreement about the potential environmental and health impacts of a plan. A case evaluation of stakeholder engagement was carried out to assess its effect in terms of consensus-building. The case consisted in two health impact scoping workshops engaging 20 stakeholders: policy-makers, experts and residents. A Participatory Action Research approach was adopted. Methods included observation, semi-structured questionnaires and interviews. Analysis methods consisted of several coding rounds, in-depth reading and discussion of Atlas.ti output reports, as well as studying questionnaire results. Participants reported a broadening of perspectives on health in relation to the environment and attainment of shared perspectives. Still, meaningful differences remained, indicating that joint learning experiences, trust and mutual respect created a ‘sense of consensus’ rather than a joint view on the issues at stake. To avoid disappointment and conflict in later project development, explicit acknowledgment and acceptance of disagreements should be included as a ground rule in future stakeholder engagement processes. Microvesicles are formed under many circumstances, especially in atheromatous plaques. Erythrocyte-derived microvesicles (ErMVs) have been proved to promote atherosclerosis by promoting hypercoagulation, mediating inflammation and inducing cell adhesion. Several clinical studies have reported potential roles of ErMVs in cardiovascular disease diagnosis, but the current understanding of ErMVs remains insufficient. In this paper, we will review current research on the formation and degradation of ErMVs and the possible effects of ErMVs in atherosclerosis, discuss potential clinical applications in cardiovascular disease, and hope to raise awareness of the relation with atherosclerosis. Context • The trend of people using complementary and alternative medicine (CAM) has been increasing globally. To enact policies and regulations that promote the safe use of CAM, a detailed investigation is needed into people’s knowledge, practices, and beliefs related to CAM. Food therapy has long been regarded as an important part of traditional Chinese medicine (TCM), a type of CAM. Objective • This study intended to empirically examine the public’s knowledge about TCM food therapy, including the sources of that knowledge, actual practices, and the public’s beliefs about the therapy. Design • The research team designed a questionnaire to test the public’s understanding of food properties as designated in TCM food therapy and to identify the public’s sources of knowledge and experience in using food therapy. Setting •The study took place at the College of Professional and Continuing Education, the Hong Kong Polytechnic University, and the University of Hong Kong in Hong Kong, China. Participants • Participants were 447 Chinese students participating in postsecondary or tertiary education at the two higher education institutions mentioned above. Outcome Measures • The survey’s quantitative questions examined respondents’ prior TCM knowledge, sources of knowledge, and food therapy experience compared to their ability to correctly categorize 24 foods. The survey’s qualitative questions examined the general practice of food therapy and the cultural beliefs underlying the food therapy strategies that participants used. Results • The study showed that: (1) having prior and partial knowledge of food properties (P=.000); (2) seeking information from the internet (P = .000), television (P = .0473) and friends (P = .0181); and (3) having used food therapy for chronic conditions (P=.0034) significantly improved participants ability to correctly categorize foods as to their food therapy characteristics. Conclusions • Participants relied less on traditional medical sources such as parents and health professionals but more on the internet, which may have empowered them through wider access to information. The study has further contributed to the study of Complementary and Alternative Medicine (CAM) by highlighting the role of cultural practices and beliefs in TCM food therapy. [ABSTRACT FROM AUTHOR]

In this letter, we perform a comprehensive study of micro torch effect on the formation of femtosecond laser-induced nanostructures on nickel. Under identical experimental conditions, laser induced nanostructures and periodic surface patterns exhibit distinctly different level of morphology with and without the micro torch. In addition, assisted by the micro torch, the ablation threshold is considerably reduced and the content of oxygen in the textured nanostructures keeps a stable low level. It is suggested that the change on the surface directly relates to the status of plasma plume and substrate heating. With the assistance of the micro torch, laser induced plasma plume is confined and its density at center region is raised, which results in the increase of the central plasma's temperature, more energy deposited on the nickel surface, and ultimately leading to the changes in the nanostructures' morphology and ablation threshold. Background: Alzheimer's disease (AD) is characterized with progressive memory loss and severe cognitive impairments, which affect everyday life and human health in the elderly. It is required that an effective and safe protective reagent against AD should be developed. It has been reported that humanin (HN) exerts neuroprotective effects against AD. In this study, we investigated the effect of a novel and more effective HN derivative, Colivelin (CLN) on AD. Methods: PDAPPV717I transgenic AD model mice (derived from parental C57/BL6 mice) were used in our study as AD model. Morris water maze test was used to test the memory impairment of AD mice and the levels of Aβ40 and Aβ42 were determined by an Elisa assay. We used an Immunohistochemistry and Immunofluorescence staining method to check the GFAP and MAP2 positive cells, and TUNEL to assess the apoptotic cells. Western blot assay was used to check the expression and phosphorylation level of p38. Results: We found that CLN improved the memory impairment induced by AD and reduced the deposit of Aβ40 and Aβ42. CLN also inhibited cell apoptosis and activation of caspase 3 in brain tissues of AD mice. Inflammation in AD mice was alleviated by CLN treatment, including the accumulation of GFAP positive cells and the inflammatory cytokines. With both structure of AGA-HNG and ANDF, CLN exhibited significantly stronger effects than synchronously administration of AGA-HNG and ADNF, suggesting CLN as a novel potential effective therapeutic reagent for AD patients. Finally, we found that CLN inhibited phosphorylation of p38 in AD mice and p38 inhibitor, SB203580 weakened the therapeutic effect of CLN. Conclusion: CLN effectively improved the memory dysfunction in PDAPP mice, and our data suggests CLN as a novel and effective reagent which may have great potentials in AD therapy. Impairment of the autophagy-lysosome pathway is implicated with the changes in α-synuclein and mitochondrial dysfunction observed in Parkinson's disease ( PD). Damaged mitochondria accumulate PINK1, which then recruits parkin, resulting in ubiquitination of mitochondrial proteins. These can then be bound by the autophagic proteins p62/ SQSTM1 and LC3, resulting in degradation of mitochondria by mitophagy. Mutations in PINK1 and parkin genes are a cause of familial PD. We found a significant increase in the expression of p62/ SQSTM1 m RNA and protein following mitophagy induction in human neuroblastoma SH- SY5Y cells. p62 protein not only accumulated on mitochondria, but was also greatly increased in the cytosol. Increased p62/ SQSMT1 expression was prevented in PINK1 knock-down cells, suggesting increased p62 expression was a consequence of mitophagy induction. The transcription factors Nrf2 and TFEB, which play roles in mitochondrial and lysosomal biogenesis, respectively, can regulate p62/ SQSMT1. We report that both Nrf2 and TFEB translocate to the nucleus following mitophagy induction and that the increase in p62 m RNA levels was significantly impaired in cells with Nrf2 or TFEB knockdown. TFEB translocation also increased expression of itself and lysosomal proteins such as glucocerebrosidase and cathepsin D following mitophagy induction. We also report that cells with increased TFEB protein have significantly higher PGC-1α m RNA levels, a regulator of mitochondrial biogenesis, resulting in increased mitochondrial content. Our data suggests that TFEB is activated following mitophagy to maintain autophagy-lysosome pathway and mitochondrial biogenesis. Glaucomatous optic neuropathy, an important neurodegenerative condition and the commonest optic neuropathy in humans, is the leading cause of irreversible blindness worldwide. Its prevalence and incidence increase exponentially with ageing and raised intraocular pressure (IOP). Using glaucomatous optic neuropathy as an exemplar for neurodegeneration, this study investigates putative factors imparting resistance to neurodegeneration. Systemic mitochondrial function, oxidative stress and vascular parameters were compared from isolated lymphocytes, whole blood and urine samples between 30 patients who have not developed the neuropathy despite being exposed for many years to very high IOP (‘resistant’), 30 fast deteriorating glaucoma patients despite having low IOP (‘susceptible’), and 30 age-similar controls. We found that ‘resistant’ individuals showed significantly higher rates of ADP phosphorylation by mitochondrial respiratory complexes I, II and IV, hyperpolarised mitochondrial membrane potential, higher levels of mitochondrial DNA, and enhanced capacity to deal with cytosolic calcium overload and exogenous oxidative stress, as compared to both controls and glaucoma patients. While it has been known for some years that mitochondrial dysfunction is implicated in neurodegeneration, this study provides a fresh perspective to the field of neurodegeneration by providing, for the first time, evidence that systemic mitochondrial efficiency above normal healthy levels is associated with an enhanced ability to withstand optic nerve injury. These results demonstrate the importance of cellular bioenergetics in glaucomatous disease progression, with potential relevance for other neurodegenerative disorders, and raise the possibility for new therapeutic targets in the field of neurodegeneration. Purpose: Breast cancer cases with four or more involved axillary lymph nodes (ALNs) feature an aggressive clinical history despite intensive treatment. However, therapies for improving the prognosis for these high-risk patients and the prognostic role of clinical characteristics have been little investigated. Therefore, we sought to assess potential prognostic factors for these patients in female Chinese patients and identify the treatment modalities they might benefit from, which offers implications for clinical practice. Patients and methods: A total of 518 patients with four or more involved ALNs were retrospectively analyzed. Survival-curve analysis was performed with the Kaplan-Meier method, and Cox proportional hazard regression was applied to identify independent variables for disease-free survival (DFS) and overall survival (OS). Results: The patients were divided into groups depending on the number of ALNs, with 38.22% having four to six positive ALNs and 61.78% having seven or more ALNs. Compared with the seven or more-positive ALN subgroup, patients with four to six positive ALNs tended to have smaller tumors and were more likely to undergo modified radical mastectomy rather than radical mastectomy (both P<0.001). Univariate analysis revealed that a fluorouracil/doxorubicin (epirubicin)/cyclophosphamide (CA[E]F) regimen or a CA(E)F followed by docetaxel (CA[E] F > T) regimen conferred significantly better DFS (P=0.0075) and OS (P<0.0001) than those achieved from a cyclophosphamide/methotrexate/fluorouracil regimen, which was almost completely generated by the seven or more ALN subgroup (P=0.0088 and P=0.0001, respectively). Postoperative radiotherapy was associated with better DFS (P=0.0360), which was also generated by the seven or more ALN subgroup (P=0.0107). Subgroup analysis also clarified that the type of surgery conferred a modest effect on DFS in the seven or more ALN subgroup (P=0.0305). Multivariate survival analysis revealed that ALN status (hazard ratio [HR] 2.00, 95% confidence interval [CI] 1.31-3.05; P=0.001), tumor size (HR 1.48, 95% CI 1.06-2.08; P=0.022), and type of surgery (HR 0.47, 95% CI 0.30-0.74; P=0.001) were independent prognostic factors for DFS. Meanwhile, ALN status (HR 2.96, 95% CI 1.51-5.77; P=0.002), tumor size (HR 2.32, 95% CI 1.38-3.89; P=0.001), type of surgery (HR=0.39, 95% CI 0.20-0.76; P=0.006), and regimen of chemotherapy (HR=0.64, 95% CI 0.50-0.85; P=0.002) were identified as independent prognostic factors for OS. Conclusion: Besides the classical prognostic factors and the improvement of prognosis achieved from the anthracycline-based or anthracycline-taxane combination chemotherapy compared to cyclophosphamide/methotrexate/fluorouracil chemotherapy, our findings showed benefits on DFS and OS for appropriate local treatments, including radiotherapy and sufficient ALN dissection for high-risk breast cancer patients with four or more ALNs involved, which suggests that much importance should also be attached to local treatment besides adjuvant systemic therapy. Land use and land cover (LULC) datasets for Beijing in 1978, 1987, 1992, 2000 and 2010 were developed from Landsat images using the object-oriented classification approach. The relationships between social-economic, demographic and political factors and time-series LULC data were examined for the periods between 1978 and 2010. The results showed the effectiveness of using the object-oriented decision tree classification method for LULC classification with time series of Landsat images. Combined with anthropogenic driving forces, our research can effectively explain the detailed LULC change trajectories corresponding to different stages and give new insights for Beijing LULC change patterns. The results show a significant increase in forest and built-up areas, but a decrease in arable lands, due to urbanization and reforestation. Large ecological projects result in an increase of forest areas and population, and economic conditions result in urban expansion. The anthropogenic driving forces analysis results further prove that both population increase and economic development played important roles in the expansion of built-up areas. Both the qualitative and quantitative anthropogenic driving forces analysis methods were helpful for better understanding the mechanisms of LULC change. [ABSTRACT FROM AUTHOR]

Two anaerobic forward osmosis (FO) membrane bioreactors (AnFOMBRs), Rchloride and Rsulfate, were operated for 100 days using NaCl and Na2SO4 as the draw solution, respectively. The operating conditions were identical for both systems, with a solids retention time of 30 d, hydraulic retention time of 8 h and using cellulose triacetate FO membrane. High rejection performance of FO membranes resulted in salinity accumulation in the bioreactors. Rchloride and Rsulfate reached a stable conductivity of about 35 and 11 mS/cm, respectively, at the end of the experimental run. Hypersalinity of Rchloride undesirably impacted biological growth; mixed liquor volatile suspended solids in Rchloride was much lower at 376 mg/L, whereas that of Rsulfate was 1,170 mg/L. Organic removals were excellent due to reduced organic loadings at low fluxes and thus, Rsulfate and Rchloride achieved secondary total organic carbon (TOC) removal efficiencies of at least 75%. Both AnFOMBRs started with an initial flux of 5 LMH. Flux for Rchloride stabilized at 0.25 LMH, while Rsulfate at 0.96 LMH. The high salinities of both reactors negatively impacted methanogenic growth. Application of the fluorescence in-situ hybridization (FISH) technique confirmed the ousting of methanogens by sulfate reducing bacteria from the anaerobic consortium. Background Type 2 diabetes is one of the most common causes of cardiovascular disease as it causes arterial stiffness changes. The purpose of this study is to characterize, in vivo, carotid arterial structural and functional changes by applying radio frequency and X-strain ultrasound techniques. Methods Ninety-one subjects were assigned into two groups; a diabetes group and a control group. Structural and functional changes in the common carotid arterial wall were investigated by quality intima-media thickness (QIMT), quality arterial stiffness (QAS), and X-strain analysis with a Mylab Twice ultrasound instrument. The relationships among variables between the two groups were analyzed in this study. Results There was no significant difference in carotid IMT (626.5 ± 169.1 μm vs. 568.5 ± 122.6 μm, P = 0.1506) between two groups. Pulse wave velocity (PWV) and stiffness index (β) were remarkably greater (8.388 ± 3.254 m/s vs. 7.269 ± 1.332 m/s; 12.51 ± 14.16 vs.9.279 ± 2.871), while compliance coefficient (CC) decreased significantly in the diabetes group (0.802 ± 0.3094 mm2/Kpa vs. 0.968 ± 0.3992 mm2/Kpa) (P < 0.05). The displacement difference of radial (RD-D), longitudinal (LD-D) and rotation (ROT-D) directions were significantly different between two groups' comparison (P = 0.0212, P = 0.0235 and P = 0.0072, respectively). The time of circumferential peak strain difference (CS-DT) and the time of radial peak strain rate (RSR-T) were found to be significantly different between the two groups (341.9 ± 77.56 ms vs. 369.0 ± 78.26 ms, P = 0.0494; 142.7 ± 22.43 ms vs. 136.2 ± 30.70 ms, P = 0.0474). CS-TD and RSR-T were also found to be positively correlated with CC value (r = 0.3908, P < 0.005 and r = 0.3027, P = 0.0326, respectively). Finally, PWV was negatively correlated with CC with (r = -0.6177, P < 0.001). Conclusions In type 2 diabetes, the functional changes in CCA can be identified using the methods presented in this article earlier than the structural changes. Reactive oxygen species (ROS) are known to be a key factor in the development of cancer, and many exogenous sources are supposed to be related to the formation of ROS. In this paper, a microfluidic chip was developed for studying the production of ROS in lung cancer cells under different chemical and physical stimuli. This chip has two unique features: (1) five relative concentrations of 0, 1/8, 1/2, 7/8, and 1 are achieved in the culture regions; (2) a shear stress gradient is produced inside each of the five culture areas. Lung cancer cells were seeded inside this biocompatible chip for investigating their response to different concentrations of H2O2, a chemical stimulus known to increase the production of ROS. Then the effect of shear stress, a physical stimulus, on lung cancer cells was examined, showing that the production of ROS was increased in response to a larger shear stress. Finally, two antioxidants, α-tocopherol and ferulic acid, were used to study their effects on reducing ROS. It was found that high-dose α-tocopherol was not able to effectively eliminate the ROS produced inside cells. This counter effect was not observed in cells cultured in a traditional chamber slide, where no shear stress was present. This result suggests that the current microfluidic chip provides an in vitro platform best mimicking the physiological condition where cells are under circulating conditions. [ABSTRACT FROM AUTHOR]

Highlights: The state-of-the-art progress of flexible thermally conductive films with ultrahigh in-plane isotropic thermal conductivity (k) and potential application are summarized. The heat transfer mechanism, processing methods to enhance k, optimization strategies to reduce interface thermal resistance of flexible thermally conductive films are reviewed. The limitations and opportunities for the future development of flexible thermally conductive films are proposed.Effective thermal management is quite urgent for electronics owing to their ever-growing integration degree, operation frequency and power density, and the main strategy of thermal management is to remove excess energy from electronics to outside by thermal conductive materials. Compared to the conventional thermal management materials, flexible thermally conductive films with high in-plane thermal conductivity, as emerging candidates, have aroused greater interest in the last decade, which show great potential in thermal management applications of next-generation devices. However, a comprehensive review of flexible thermally conductive films is rarely reported. Thus, we review recent advances of both intrinsic polymer films and polymer-based composite films with ultrahigh in-plane thermal conductivity, with deep understandings of heat transfer mechanism, processing methods to enhance thermal conductivity, optimization strategies to reduce interface thermal resistance and their potential applications. Abstract: Mutations of the gene for glucocerebrosidase 1 (GBA) cause Gaucher disease (GD), an autosomal recessive lysosomal storage disorder. Individuals with homozygous or heterozygous (carrier) mutations of GBA have a significantly increased risk for the development of Parkinson’s disease (PD), with clinical and pathological features that mirror the sporadic disease. The mechanisms whereby GBA mutations induce dopaminergic cell death and Lewy body formation are unknown. There is evidence of mitochondrial dysfunction and oxidative stress in PD and so we have investigated the impact of glucocerebrosidase (GCase) inhibition on these parameters to determine if there may be a relationship of GBA loss-of-function mutations to the known pathogenetic pathways in PD. We have used exposure to a specific inhibitor (conduritol-β-epoxide, CβE) of GCase activity in a human dopaminergic cell line to identify the biochemical abnormalities that follow GCase inhibition. We show that GCase inhibition leads to decreased ADP phosphorylation, reduced mitochondrial membrane potential and increased free radical formation and damage, together with accumulation of alpha-synuclein. Taken together, inhibition of GCase by CβE induces abnormalities in mitochondrial function and oxidative stress in our cell culture model. We suggest that GBA mutations and reduced GCase activity may increase the risk for PD by inducing these same abnormalities in PD brain. [Copyright &y& Elsevier]

This paper studies the dynamic relationships between GDP, residential property prices, and stock prices in Hong Kong. The study is interesting because most people put their wealth into these two markets. We find that there are long-run feedback effects between the two asset markets, providing evidence of wealth and credit-price effects in Hong Kong. There are also long-run, bi-directional causal links between real GDP and real asset prices. Hence, real asset prices can drive long-run economic growth and vice versa. Abstract: The recombinant N-acyl-d-amino acid amidohydrolase (N-d-AAase) of Variovorax paradoxus Iso1 was unstable during protein purification and storage at 4°C. Since the methionine oxidation might be the artificial factor leading to the inactivation of N-d-AAase, eight potential oxidation sensitive methionine residues of the enzyme were individually substituted with leucine utilizing site-directed mutagenesis. Among them, five mutants, M39L, M56L, M221L, M254L, and M352L remained at least 70% of wild-type specific activity. The enzyme kinetic parameters of M221L revealed a 44% decrease in K m, and finally reflected a 2.4-fold increase in k cat/K m. Moreover, its half-life at 4°C increased up to 6-fold longer than that of the wild-type. Structural analysis of each methionine substitution was carried out based on the crystal structure of N-d-AAase from Alcaligenes faecalis DA1. Met221 spatial closeness to the zinc-assistant catalytic center is highly potential as the primary site for oxidative inactivation. We conclude that the replacement of methionine M221 with leucine in N-d-AAase successfully enhances the oxidative resistance, half-life, and enzyme activity. Reverse cholesterol transport (RCT) has been characterized as a crucial step for antiatherosclerosis, which is initiated by ATPbinding cassette A1 (ABCA1) to mediate the efflux of cellular phospholipids and cholesterol to lipid-free apolipoprotein A-I (apoA-I). However, the mechanisms underlying apoA-I/ABCA1 interaction to lead to the lipidation of apoA-I are poorly understood. There are several models proposed for the interaction of apoA-I with ABCA1 as well as the lipidation of apoA-I mediated by ABCA1. ApoA-I increases the levels of ABCA1 protein markedly. In turn, ABCA1 can stabilize apoA-I. The interaction of apoA-I with ABCA1 could activate signaling molecules that modulate posttranslational ABCA1 activity or lipid transport activity. The key signaling molecules in these processes include protein kinase A (PKA), protein kinase C (PKC), Janus kinase 2 (JAK2), Rho GTPases and Ca2+, and many factors also could influence the interaction of apoA-I with ABCA1. The optical light curve of the BL Lac object OQ 530 is constructed based on published data. Diagrams showing the correlations of fluxes in different wavelength ranges are constructed and used to analyze the color variations. The relative spectral energy distribution of the variable source in the optical can be represented by a power law that is stable on long time scales. Geometry theorem proving involves skills that are difficult to learn. Instead of working with abstract and complicated representations, students might start with concrete, graphical representations. A proof tree is a graphical representation of a formal proof, with each node representing a proposition or given conditions. A computer-assisted learning environment called MR Geo is proposed to help students in learning to do theorem proving, with the help of multiple representations including problem description, static figure, dynamic geometry figure, formal proof and proof tree. Empirical results indicated that medium-achievement students enjoyed most in interacting with these representations and found them most helpful in learning geometry proofs while low-achievement students changed their attitudes of hating geometry theorem proving. The Trömner sign is commonly used as a clinical neurological examination for upper motor neuron lesions above the fifth or sixth cervical segments of the spinal cord. This study aims to assess and quantify the Trömner signs utilizing electrophysiological test, and correlate to the severity of cord compression in cervical spondylotic myelopathy (CSM). We enlisted 46 CSM patients, and 30 healthy persons as controls. Manual Trömner and Hoffmann signs were tested in all subjects. By using a self-designed instrument, we performed electrophysiological assessments for the Trömner signs in patients and controls. Parameters of conduction latencies and amplitude of muscle action potentials were measured and compared with the cord compression ratios in CSM patients. The results showed a greater diagnostic sensitivity for the quantified Trömner signs in comparison to those of manual Trömner signs and Hoffmann signs. We found a positive correlation between the amplitude of muscle action potentials obtained in the Trömner signs and the cord compression ratios in the patients with CSM. In conclusion, the Trömner signs can be measured by electrophysiological assessments. We demonstrate a new quantification method for an established neurological sign. Not only is Trömner sign a highly sensitive test in clinical neurological examination, the electrophysiological assessment of this sign can also serve as an objective marker for evaluation of the severity of cervical cord compression. Atherosclerosis is an inflammatory disease characterized by the accumulation of macrophages in the arterial intima. The activated macrophages secreted more pro-inflammatory cytokines, such as tumor necrosis factor (TNF)-α, which promote the development of the disease. Apolipoprotein A-I (apoA-I), the major component of high density lipoprotein, is involved in reverse cholesterol transport of lipid metabolism. Recently, it has been found that apoA-I suppresses inflammation via repression of inflammatory cytokine expression; the mechanisms of the apoA-I-suppressive action, however, are not yet well characterized. In this study, we have for the first time found that apoA-I suppresses the expression of some inflammatory cytokines induced by lipopolysaccharide via a specific post-transcriptional regulation process, namely mRNA destabilization, in macrophages. Our further studies have also shown that AU-rich elements in the 3′-untranslated region of TNF-α mRNA are responsive to the apoA-I-mediated mRNA destabilization. The apoA-I-induced inflammatory cytokine mRNA destabilization was associated with increased expression of mRNA-destabiizing protein tristetraprolin through aJAK2/STAT3 signaling pathway-dependent manner. When blocking interaction of apoA -I with ATP-binding membrane cassette transporter Al (ABCA1), a major receptor for apoA -I in macrophages, it would almost totally abolish the effect of apoA-I on tristetraprolin expression. Atherosclerosis is characterized by a chronic inflammatory condition that involves numerous cellular and molecular inflammatory components. A wide array of inflammatory mediators, such as cytokines and proteins produced by macrophages and other cells, play a critical role in the development and progression of the disease. ATP-binding membrane cassette transporter A1 (ABCA1) is crucial for cellular cholesterol efflux and reverse cholesterol transport (RCT) and is also identified as an important target in antiatherosclerosis treatment. Evidence from several recent studies indicates that inflammation, along with other atherogenic- related mediators, plays distinct regulating roles in ABCA1 expression. Proatherogenic cytokines such as interferon (IFN)-γ and interleukin (IL)-1β have been shown to inhibit the expression of ABCA1, while antiatherogenic cytokines, including IL-10 and transforming growth factor (TGF)-β1, have been shown to promote the expression of ABCA1. Moreover, some cytokines such as tumor necrosis factor (TNF)-α seem to regulate ABCA1 expression in species-specific and dose-dependent manners. Inflammatory proteins such as C-reactive protein (CRP) and cyclooxygenase (COX)-2 are likely to inhibit ABCA1 expression during inflammation, and inflammation induced by lipopolysaccharide (LPS) was also found to block the expression of ABCA1. Interestingly, recent experiments revealed ABCA1 can function as an antiinflammatory receptor to suppress the expression of inflammatory factors, suggesting that ABCA1 may be the molecular basis for the interaction between inflammation and RCT. This review aims to summarize recent findings on the role of inflammatory cytokines, inflammatory proteins, inflammatory lipids, and the endotoxin-mediated inflammatory process in expression of ABCA1. Also covered is the current understanding of the function of ABCA1 in modulating the immune response and inflammation through its direct and indirect antiinflammatory mechanisms including lipid transport, high-density lipoprotein (HDL) formation and apoptosis. Based on grey relational analysis and stepwise regression, this study analysed the socio-economic driving forces of Xiamen since the reform and opening of China. The research aimed to explore the deep dynamic mechanism of urban space expansion of Xiamen and provide scientific support for its sustainable development. The results indicate resource consumption factors and population factors have had a significant driving effect on the expansion of built-up area, and have contributed more than the economic development factors. It was revealed that the main urban socio-economic driving forces have gradually changed during the middle and late stage of urbanisation, and the city has also shifted from an original construction and developing city type into a more spatially stable, developed city type. Summary: In eukaryotic cells the final maturation of ribosomes occurs in the cytoplasm, where trans-acting factors are removed and critical ribosomal proteins are added for functionality. Here, we have carried out a comprehensive analysis of cytoplasmic maturation, ordering the known steps into a coherent pathway. Maturation is initiated by the ATPase Drg1. Downstream, assembly of the ribosome stalk is essential for the release of Tif6. The stalk recruits GTPases during translation. Because the GTPase Efl1, which is required for the release of Tif6, resembles the translation elongation factor eEF2, we suggest that assembly of the stalk recruits Efl1, triggering a step in 60S biogenesis that mimics aspects of translocation. Efl1 could thereby provide a mechanism to functionally check the nascent subunit. Finally, the release of Tif6 is a prerequisite for the release of the nuclear export adaptor Nmd3. Abstract: Two novel alternating phenylenevinylene copolymers P6 and P12 as well as one fluorenevinylene copolymer F connected at the 1,7 bay positions with perylene bisimide were synthesized by Heck coupling. They were characterized by GPC, FT-IR, 1H NMR, TGA, TMA, UV–vis, cyclic voltammetry and photoluminescence (PL) emission spectra. The copolymers were soluble in common organic solvents and thermally stable up to ∼300°C. Their glass transition temperatures were 48–60°C. The long wavelength absorption maximum was located at 510–542nm with optical band gaps of ∼2.0eV. The PL emission maximum of P6 and P12 was red-shifted relative to F. The ribosome stalk is essential for recruitment of translation factors. In yeast, P0 and Rpl12 correspond to bacterial L10 and L11 and form the stalk base of mature ribosomes, whereas Mrt4 is a nuclear paralogue of P0. In this study, we show that the dual-specificity phosphatase Yvh1 is required for the release of Mrt4 from the pre-60S subunits. Deletion of YVH1 leads to the persistence of Mrt4 on pre-60S subunits in the cytoplasm. A mutation in Mrt4 at the protein-RNA interface bypasses the requirement for Yvh1. Pre-60S subunits associated with Yvh1 contain Rpl12 but lack both Mrt4 and P0. These results suggest a linear series of events in which Yh1 binds to the pre-60S subunit to displace Mrt4. Subsequently, P0 loads onto the subunit to assemble the mature stalk, and Yvh1 is released. The initial assembly of the ribosome with Mrt4 may provide functional compartmentalization of ribosome assembly in addition to the spatial separation afforded by the nuclear envelope. Alpha synuclein can be phosphorylated at serine129 (P-S129), and the presence of highly phosphorylated α-synuclein in Lewy bodies suggests changes to its phosphorylation status has an important pathological role. We demonstrate that the kinase(s) responsible for α-synuclein S129 phosphorylation is constitutively active in SH-SY5Y cells and involves casein kinase 2 activity. Increased oxidative stress or proteasomal inhibition caused significant elevation of P-S129 α-synuclein levels. Under these conditions, similar increases in P-S129 α-synuclein were found in both sodium dodecyl sulphate lysates and Triton extracts indicating the phosphorylated protein was soluble and did not lead to aggregation. The rate of S129 phosphorylation was increased in response to proteasomal inhibition indicating a higher activity of the relevant kinase. Cells expressing the phosphorylation mimic, S129D α-synuclein increased cell death and enhanced sensitivity to epoxomycin exposure. Proteasomal inhibition markedly decreased S129D α-synuclein turnover suggesting proteasomal inhibition leads to the accumulation of P-S129 α-synuclein through an increase in the kinase activity and a decrease in protein turnover resulting in increased cell death. We conclude that S129 phosphorylation is toxic to dopaminergic cells and both the levels of S129 phosphorylated protein and its toxicity are increased with proteasomal inhibition emphasising the interdependence of these pathways in Parkinson’s disease pathogenesis.
