The hydroxyapatite (HAp)/gelatin (GEL) nanocomposite was prepared through the coprecipitation and then cross-linked by using glutaraldehyde (GA). From FT-IR measurement the spectral features for amide bands and phosphate bands were severely modified by the cross-linkage and the organic content increased with the degree of cross-linkage. From Transmission Electron Microscopy (TEM) and electron diffraction analyses we could confirm the preferentially directional growth of needle-like HAp particles, which were embedded in GEL by the mineralization. Also we could observe worm-like stria patterns of contrast, which were revealed by the mineralization on the individual GEL fibrils. Moire´ images were observed in a highly cross-linked HAp/GEL nanocomposite sample and we think the cross-linkage induced the assembly of unordered individual fibrils along the preferential direction. A nanocomposite of gelatin[GEL]-hydroxyapatite[HAp] was prepared using the biomimetic process. The hydroxyapatite nanocrystals were precipitated in aqueous solution of gelatin at pH 8 and 38°C. The chemical bonding between calcium ions of HAp and carboxyl ions of GEL molecules induced a red-shift of the 1339 cm−1 band of GEL in FT-IR analysis. TEM images and electron diffraction patterns for the nanocomposite strongly indicate the self-organization of HAp nanocrystals along the GEL fibrils. Electron diffraction for the nanocomposites showed a strong preferred orientation of the (0 0 2) plane in HAp nanocrystals. The development of HAp nanocrystals in an aqueous GEL solution was highly influenced by the concentration ratio of GEL to HAp. A higher concentration of GEL induced the formation of tiny crystallites (4 nm×9 nm size), while a lower concentration of GEL contributed to the development of bigger crystallites (30 nm×70 nm size). From DT/TGA data, the HAp-GEL nanocomposite showed typically three exothermic temperatures. The increase in decomposition temperatures indicates the formation of a primary chemical bond between HAp and GEL. The higher concentration of GEL supplies abundant reaction sites containing groups such as carboxyl, which can bind with calcium ions. The abundant supply of reaction sites leads to a very large number of HAp nuclei. However, the formation of a large number of nuclei depletes the concentration of calcium ions that available for growth to the extent that the nuclei cannot grow very large. This in turn will lead to the creation of a large number of tiny nanocrystals at this higher GEL concentration. [Copyright &y& Elsevier]

This study reports a method for controlling loads on an in vivo dental implant and its application for the investigation of early loading versus delayed loading of dental implants. The method was developed for the purpose of studying an ongoing hypothesis that amounts to bone loss around dental implants are related to mechanical-mediated adaptation of the alveolar bone. Using a customized intra-oral hydraulic system, the daily loading over a dental implant has been completed and recorded for six Sinclair swine. Each pig had a 5-month duration implant loading. During the experiments (loading), no analgesic treatment was supplied. The mean of the in vivo daily loadings was confirmed through an in vitro bench test after each animal was euthanized. Variations of the averaged loading input among the six animals were smaller than 10%. Preliminary data produced by the model suggests that cervical bone loss is less for early loading than for delayed loading. The current system is expected to provide a useful load control model for the study of alveolar bone adaptation around dental implants in relation to various loadings. [Copyright &y& Elsevier]

The objective of this study was to explore the feasibility of current 3D reconstruction in assessing the position of maxillary impacted canines from 2D panoramic X-rays. A dataset was created using pre-treatment CBCT data from a total of 123 patients, comprising 74 patients with impacted canines and 49 patients without impacted canines. From all 74 subjects, we generated a dataset containing paired 2D panoramic X-rays and pseudo-3D images. This pseudo-3D image contained information about the location of the impacted canine in the buccal/lingual, mesial/distal, and apical/coronal positions. These data were utilized to train a deep-learning reconstruction algorithm, a generative AI. The location of the crown of the maxillary impacted canine was determined based on the output of the algorithm. The reconstruction was evaluated using the structure similarity index measure (SSIM) as a metric to indicate the quality of the reconstruction. The prediction of the impacted canine's location was assessed in both the mesiodistal and buccolingual directions. The reconstruction algorithm predicts the position of the impacted canine in the buccal, middle, or lingual position with 41% accuracy, while the mesial and distal positions are predicted with 55% accuracy. The mean SSIM for the output is 0.71, with a range of 0.63 to 0.84. Our study represents the first application of AI reconstruction output for multidisciplinary care involving orthodontists, periodontists, and maxillofacial surgeons in diagnosing and treating maxillary impacted canines. Nuclear factor erythroid-derived factor 2-related factor 2 (Nrf2) is a master regulator of antioxidant response and protects cells from excessive oxidative stress. Nrf2 emerges as a prospective therapeutic target for metabolic bone disorders, in which the balance between osteoblastic bone formation and osteoclastic bone resorption is disrupted. However, the molecular mechanism through which Nrf2 modulates bone homeostasis remains unclear. In this study, we compared the differences in Nrf2-mediated antioxidant response and ROS regulation in osteoblasts and osteoclasts, both in vitro and in vivo. Findings indicated a close connection between the Nrf2 expression and its related antioxidant response with osteoclasts than osteoblasts. We next pharmacologically manipulated the Nrf2-mediated antioxidant response during osteoclast or osteoblast differentiation. Nrf2 inhibition enhanced osteoclastogenesis, while its activation suppressed it. In contrast, osteogenesis decreased irrespective of whether Nrf2 was inhibited or activated. These findings highlight the distinct ways in which the Nrf2-mediated antioxidant response regulates osteoclast and osteoblast differentiation, thereby contributing to the development of Nrf2 targeted therapies for metabolic bone diseases. • The Nrf2-induced antioxidant response is more actively regulated in osteoclasts than in osteoblasts, both in vitro and in vivo. • ROS plays a critical role in osteoclastogenesis regulated by Nrf2. • Pharmacological inhibition of Nrf2 by ML385 leads to increased osteoclastogenesis and decreased osteogenesis. • Pharmacological activation of Nrf2 by curcumin leads to a decrease in both osteoclastogenesis and osteogenesis. Objective: Visual differentiation of resin and tooth in a tooth cavity is not simple due to their highly similar shade. The purpose of the present study was to find any noninvasive method which can effectively differentiate resin from sound tooth in a resin-imbedded tooth for resin repair.Materials and methods: For the study, various resin products were imbedded into the cavity of sound tooth. By applying laser of different wavelengths, autofluorescence (AF) of sound tooth and resin products were obtained. Microhardness, X-ray radiograph, and DIAGNOdent were tested for each tooth, resin product, and resin-imbedded tooth.Results: For the AF spectra obtained using the 405-nm wavelength, sound tooth has emission peak at 440-470 nm and near 490 nm. Sound tooth has several times higher microhardness than resin products regardless of position in tooth subsurface. Due to the difference of radiopaque fillers' composition and concentration, resin products have different brightness in the X-ray radiograph. DIAGNOdent readings for tooth and resin products were inconsistently different, and the difference of obtained values was slightly not to be applicable for the differentiation.Conclusion: Among the tested methods, with noninvasive treatment, AF spectrum by the 405-nm wavelength showed the apparent difference between resin and tooth.Clinical significance: For the resin repair in a resin-imbedded tooth cavity, AF spectrum produced by 405-nm wavelength could be a useful method in tracing the resin-tooth boundary if combined with conventional X-ray radiography. The author discusses the result of a study which evaluates the vertical positions of anterior teeth on smile esthetics determined by the Japanese and Korean orthodontists and orthodontic patients. The author states that the anterior teeth's vertical positions were assessed after digital modification in 1-millimeter (mm) increments, from 5mm upper lip coverage to 5mm gingival exposure. He notes that orthodontists and the female patients shared the same preference in the smile evaluation.

Accurately segmenting teeth and identifying the corresponding anatomical landmarks on dental mesh models are essential in computer-aided orthodontic treatment. Manually performing these two tasks is time-consuming, tedious, and, more importantly, highly dependent on orthodontists’ experiences due to the abnormality and large-scale variance of patients’ teeth. Some machine learning-based methods have been designed and applied in the orthodontic field to automatically segment dental meshes (e.g., intraoral scans). In contrast, the number of studies on tooth landmark localization is still limited. This paper proposes a two-stage framework based on mesh deep learning (called TS-MDL) for joint tooth labeling and landmark identification on raw intraoral scans. Our TS-MDL first adopts an end-to-end iMeshSegNet method (i.e., a variant of the existing MeshSegNet with both improved accuracy and efficiency) to label each tooth on the downsampled scan. Guided by the segmentation outputs, our TS-MDL further selects each tooth’s region of interest (ROI) on the original mesh to construct a light-weight variant of the pioneering PointNet (i.e., PointNet-Reg) for regressing the corresponding landmark heatmaps. Our TS-MDL was evaluated on a real-clinical dataset, showing promising segmentation and localization performance. Specifically, iMeshSegNet in the first stage of TS-MDL reached an averaged Dice similarity coefficient (DSC) at ${0.964}\pm {0.054}$ , significantly outperforming the original MeshSegNet. In the second stage, PointNet-Reg achieved a mean absolute error (MAE) of ${0}.{597}\pm {0}.{761} \, mm$ in distances between the prediction and ground truth for 66 landmarks, which is superior compared with other networks for landmark detection. The article presents the author's views on a case report concerning anterior crossbite correction of a 28-year-old Korean female using orthodontic clear aligner. In the case report, the authors have mentioned esthetics, quickness, comfort, low cost and availability to change plans during treatment as some of the advantages of clear aligner. According to the author, the clear aligner is an effective alternative for patients who are reluctant to wear fixed appliances.

Three-dimensional finite element (FE) models of splinted prosthetic crowns were studied and stress analyses were evaluated with different types of implant support, including standard, wide or two implant(s) for partial, posterior edentulous restorations. The FE models were constructed based on a cadaver mandible containing the 2nd premolar and the 1st molar. The crowns of these two teeth were modeled as connected and disconnected to mimic the splinted and non-splinted designs, respectively. One standard implant was placed at the premolar region, while three types of implant support, one at a time (the standard implant, wide implant and two implants), were used to support the molar crown. A 100 N oblique load was applied to the buccal cusp on each crown. The FE simulation was validated experimentally via strain gauge measurement. The experimental data were well correlated with the FE predictions ( r2=0.97). When compared with the standard implant used in the molar area, the wide implant and two implants reduced the peak stress in crestal bone by 29–37% for both splinted and non-splinted cases. Inserting the standard implant into both the premolar and molar area, the bone stresses were identical for splinted and non-splinted designs. However, splinting the adjacent crowns has shown to decrease the bone stresses at the premolar region by 25%, while the wide implant or two implants were placed at the molar region. The biomechanical advantages of using the wide implant or two implants are almost identical. The benefit of load sharing by the splinted crowns is notable only when the implants on the premolar and molar regions have different supporting ability. [ABSTRACT FROM AUTHOR]

The use of an mesial-occlusal-distal (MOD) restoration in repairing a large carious lesion depends on many factors. Biomechanical performance is one of the most important. It has been recognized that resistance to restoration failure is not solely a biological concern (e.g. toxicity), but that the cavity shape, dimensions, and the state of stress must all be taken into account. In the present study, a newly developed auto-mesh program was used to generate 30 three-dimensional (3D) finite element (FE) models simulating the biomechanics for multiple factorial design of the MOD gold restoration in a maxillary second premolar. Stress levels were related to individual design factors (e.g. pulpal wall depth [P], isthmus width [W] and interaxial thickness [T]) and to their interactions under the worst physiological scenario: a concentrated bite force acting on lingual cusp with debonded interfaces between cavity walls and restorations. The results showed that enlarging the volume of the MOD cavity significantly increased stresses in enamel but did not intentionally affect stresses in dentin. The alternation of individual design parameters significantly changed the peak stresses (P < 0·05). For all three parameters, except for the width, the peak stress increased as the cavity dimension increased. Stress elevation rate (termed as ‘volumetric stress rate’ – stress elevation by increasing one unit volume of the restored materials) was different among three design factors. Depth was the most critical factor governing the stress elevation in enamel (1·76 MPa mm-3) while length (interaxial thickness) was the most important parameter in dentin (0·49 MPa mm-3). Width was the least compromising factor to the remaining tooth, 0·32 MPa mm-3 for enamel and -0·23 MPa mm-3 for dentin. The findings, at its core, did not fully agree with the traditional concept that the preservation of tooth substances will reduce risk of tooth fracture. Bioluminescent imaging (BLI) has emerged as a popular in vivo tracking modality in bone regeneration studies stemming from its clear advantages: non-invasive, real-time, and inexpensive. We recently adopted bioluminescence resonance energy transfer (BRET) principle to improve BLI cell tracking and generated the brightest bioluminescent signal known to date, which thus enables more sensitive real-time cell tracking at deep tissue level. In the present study, we brought BRET-based cell tracking strategy into the field of bone tissue engineering for the first time. We labeled rat mesenchymal stem cells (rMSCs) with our in-house BRET-based GpNLuc reporter and evaluated the cell tracking efficacy both in vitro and in vivo. In scaffold-free spheroid 3D culture system, using BRET-based GpNLuc labeling resulted in significantly better correlation to cell numbers than a fluorescence based approach. In scaffold-based 3D culture system, GpNLuc-rMSCs displayed robust bioluminescence signals with minimal background noise. Furthermore, a tight correlation between BLI signal and cell number highlighted the robust reliability of using BRET-based BLI. In calvarial critical sized defect model, robust signal and the consistency in cell survival evaluation collectively supported BRET-based GpNLuc labeling as a reliable approach for non-invasively tracking MSC. In summary, BRET-based GpNLuc labeling is a robust, reliable, and inexpensive real-time cell tracking method, which offers a promising direction for the technological innovation of BLI and even non-invasive tracking systems, in the field of bone tissue engineering. Paget's disease of bone (PDB) is a degenerative disorder affecting the skull and bones. Hyperactive osteoclasts (OCs) initiate bone degradation in the early stage, followed by increased bone formation by osteoblasts (OBs) in trabecular bones during the advanced stage. This OB-OC uncoupling results in bone deformations and irregular trabecular bone patterns. Current mouse models poorly replicate the advanced-stage characteristics of PDB. Optineurin (Gene: OPTN in humans, Optn in mice, protein: OPTN) has been implicated in PDB by genetic analyses. We identified PDB-like cortical lesions associated with OC hyperactivation in an Optn knockout (Optn −/−) mouse model. However, the effects of OPTN dysfunction on OBs and trabecular bone in advanced PDB remain unclear. In this study, we used the Optn −/− mouse model to investigate trabecular bone abnormalities and OB activity in PDB. Micro-computed tomography analysis revealed severe pagetic alterations in craniofacial bones and femurs of aged Optn −/− mice, resembling clinical manifestations of PDB. Altered OB activity was observed in aged Optn −/− mice, implicating compensatory OB response in trabecular bone anomalies. To elucidate the role of OC-OB interactions in PDB, we conducted in vitro experiments using OC conditioned media (CM) to examine the effects on OB osteogenic potential. We found OC CM restored compromised osteogenic induction of Optn −/− bone marrow stromal cells (BMSCs) from young mice, suggesting OCs maintain OB activity through secreted factors. Strikingly, OC CM from aged Optn −/− mice significantly enhanced osteogenic capability of Optn −/− BMSCs, providing evidence for increased OB activity in advanced stages of PDB. We further identified TGF-β/BMP signaling pathway in mediating the effects of OC CM on OBs. Our findings provide insights into Optn's role in trabecular bone abnormalities and OB activity in PDB. Scaffolds that are used for bone repair should provide an adequate environment for biomineralization by mesenchymal stem cells (MSCs). Recently, decellularized pulp matrices (DPM) have been utilized in endodontics for their high regenerative potential. Inspired by the dystrophic calcification on the pulp matrix known as pulp stone, we developed acellular pulp bioscaffolds and examined their potential in facilitating MSCs mineralization for bone defect repair. Pulp was decellularized, then retention of its structural integrity was confirmed by histological, mechanical, and biochemical evaluations. MSCs were seeded and proliferation, osteogenic gene expression, and biomineralization were assessed to verify DPM's osteogenic effects in vitro. MicroCT, energy-dispersive X-ray (EDX), and histological analyses were used to confirm that DPM seeded with MSCs result in greater mineralization on rat critical-sized defects than that without MSCs. Precisely labeling teeth on digitalized 3D dental surface models is the precondition for tooth position rearrangements in orthodontic treatment planning. However, it is a challenging task primarily due to the abnormal and varying appearance of patients’ teeth. The emerging utilization of intraoral scanners (IOSs) in clinics further increases the difficulty in automated tooth labeling, as the raw surfaces acquired by IOS are typically low-quality at gingival and deep intraoral regions. In recent years, some pioneering end-to-end methods (e.g., PointNet) have been proposed in the communities of computer vision and graphics to consume directly raw surface for 3D shape segmentation. Although these methods are potentially applicable to our task, most of them fail to capture fine-grained local geometric context that is critical to the identification of small teeth with varying shapes and appearances. In this paper, we propose an end-to-end deep-learning method, called MeshSegNet, for automated tooth labeling on raw dental surfaces. Using multiple raw surface attributes as inputs, MeshSegNet integrates a series of graph-constrained learning modules along its forward path to hierarchically extract multi-scale local contextual features. Then, a dense fusion strategy is applied to combine local-to-global geometric features for the learning of higher-level features for mesh cell annotation. The predictions produced by our MeshSegNet are further post-processed by a graph-cut refinement step for final segmentation. We evaluated MeshSegNet using a real-patient dataset consisting of raw maxillary surfaces acquired by 3D IOS. Experimental results, performed 5-fold cross-validation, demonstrate that MeshSegNet significantly outperforms state-of-the-art deep learning methods for 3D shape segmentation. Objectives: To synthesize two different sizes of bioactive glass-coated mesoporous silica nanoparticles (BGN@MSNs) and to investigate their effects on dentinal tubule occlusion and remineralization. Materials and methods: Two different sizes of mesoporous silica nanoparticles (MSNs) were synthesized using the Stöber method (368A, 1840A) and coated with bioactive glass nanoparticles (BGNs) using a modified quick alkali-mediated sol-gel method (368B, 1840B). Sensitive tooth disc models were prepared and divided into six groups and the following treatments were applied: group 1—no treatment, group 2—bioglass, group 3—368A, group 4—368B, group 5—1840A, and group 6—1840B. Then, five discs were selected from each group and soaked in 6 wt% citric acid to test acid resistance. Dentinal tubule occlusion and occlusion ratio were observed using field-emission scanning electron microscopy. In vitro mineralization tests using simulated body fluid solution were performed to evaluate the remineralization effect of the treatment. Results: All samples effectively occluded the dentinal tubule and formed a membrane-like layer. After acid treatment, 1840B (group 6) exhibited the highest rate of dentinal tubule occlusion. Remineralization was observed in 368B and 1840B, and 1840B exhibited the fastest remineralization. Conclusions: Dentinal tubule remineralization induced by the BGN@MSN biocomposite can be used to stabilize long-term prognosis in dentin hypersensitivity. The 1840B induced the most efficient remineralization, and its smaller size and larger surface area were effective for remineralization. Abstract Metabolic bone diseases are global public health concerns and are primarily caused by uncontrolled osteoclast (OC) formation and activation. During OC differentiation, intracellular reactive oxygen species (ROS) stimulated by receptor activator of nuclear factor kappa-B ligand (RANKL) can serve as the signaling molecules to promote osteoclastic genes expression. Nuclear factor erythroid-2 related factor 2 (NRF2), a master mediator of cellular antioxidant response, also plays a critical role in OC differentiation through the regulation of redox homeostasis. In this study, we investigated the effects of three NRF2 inducers on osteoclastogenesis, including Bardoxolone methyl (CDDO-Me), Sulforaphane (SFN), and tert-butylhydroquinone (tBHQ). By treating RAW cells with three compounds, we found that NRF2 was activated and its downstream antioxidant genes were upregulated, and the RANKL-induced intracellular ROS production and osteoclastogenesis were impaired. Additionally, the expression of nuclear factor of activated T cells c1 (NFATC1), C-FOS and tumor necrosis factor alpha (TNFα) were inhibited after acute exposures (6 h) to the three compounds. Furthermore, suppressed the expression of osteoclast differentiation-associated genes, tartrate-resistant acid phosphatase (TRAP), cathepsin K (CTSK), matrix metalloproteinase-9 (MMP-9) and dendritic cell-specific transmembrane protein (DC-STAMP) were observed after prolonged exposures (5 days) to the compounds. Taken together, these results suggest that CDDO-Me, SFN and tBHQ attenuate RANKL-induced osteoclastogenesis via activation of NRF2-mediated antioxidant response. Among these compounds, relatively low concentrations of CDDO-Me showed stronger active and inhibitory effects on antioxidant response and osteoclastogenesis, respectively. Objectives: To synthesize a silver-doped bioactive glass/mesoporous silica nanoparticle (Ag-BGN@MSN), as well as to investigate its effects on dentinal tubule occlusion, microtensile bond strength (MTBS), and antibacterial activity.Materials and methods: Ag-BGN@MSN was synthesized using a modified "quick alkali-mediated sol-gel" method. Demineralized tooth disc models were made and divided into four groups; the following treatments were then applied: group 1—no treatment, group 2—bioglass, group 3—MSN, group 4—Ag-BGN@MSN. Next, four discs were selected from each group and soaked into 6 wt% citric acid to test acid-resistant stability. Dentinal tubule occlusion, as well as the occlusion ratio, was observed using field-emission scanning electron microscopy. The MTBS was also measured to evaluate the desensitizing effect of the treatments. Cytotoxicity was examined using the MTT assay. Antibacterial activity was detected against Lactobacillus casei, and ion dissolution was evaluated using inductively coupled plasma optical emission spectrometry.Results: Ag-BGN@MSN effectively occluded the dentinal tubule and formed a membrane-like layer. After the acid challenge, Ag-BGN@MSN had the highest rate of dentinal tubule occlusion. There were no significant differences in MTBS among the four groups (P > 0.05). All concentrations of Ag-BGN@MSN used had a relative cell viability above 72%.Conclusions: Ag-BGN@MSN was successfully fabricated using a modified sol-gel method. The Ag-BGN@MSN biocomposite effectively occluded dentinal with acid-resistant stability, did not decrease bond strength in self-etch adhesive system, had low cytotoxicity, and antibacterial effect.Clininal relevance: Dentinal tubule sealing induced by Ag-BGN@MSN biocomposite with antibacterial effect is likely to increase long-term stability in DH. Objective: The correlations between morphology of the temporomandibular joint structure, the anterior guidance angle, and occlusal plane were investigated.Materials and Methods: A cone beam computed tomography analysis was performed in 158 patients (86 women and 72 men). 3D software was employed to obtain the coordinates of the shape of the incisal guidance angle, occlusal guidance angle, articular fossa, and mandibular condyle. Generalized Procrustes analysis including principal components analysis (PCA) were performed and produced principal components (PCs) scores of each shape and their centroid size (CS).Results: A significant Pearson correlation coefficient of 0.3451 (p < .001) was observed between the incisal guidance angle and occlusal plane. The CS also showed a correlation with the incisal guidance angle, but not with the occlusal plane angle. The PCA results revealed that there were no significant correlations between the temporomandibular joint structure (TMJ) shape (fossa and condyle) and the incisal guidance angle.Conclusions: Incisor guidance angle and occlusal plane angle were correlated. In addition, there was a correlation between CS and incisal guidance angle. Abstract: Objective: To compare the cuspal strain in Class II restorations made with bulk‐fill and conventional composite resins. Materials and methods: Fifty extracted maxillary premolars were mounted into phenolic rings and divided into five groups (n = 10). Specimens received standardized MOD preparations. A two‐step self‐etch adhesive was applied and the preparations were restored using a custom matrix as follows: Filtek Supreme Ultra in eight 2‐mm increments (FSUI); Filtek Supreme Ultra in bulk (FSUB); SonicFill in bulk (SF); SureFil SDR flow in bulk, covered with a 2‐mm occlusal layer of Filtek Supreme Ultra (SDR/FSU); Tetric EvoCeram Bulk Fill in bulk (TEBF). Strain gages bonded to the buccal and lingual cusps recorded cuspal strain during restorations. End strain values were determined and data were subjected to Kruskal‐Wallis testing, followed by one‐way ANOVA and Tukey´s post hoc test. Results: Combined strain values and standard deviations (in µɛ) were: FSUI: 723 ± 102.8, FSUB: 929.2 ± 571.9, SF: 519.1 ± 80.2, SDR‐FSU: 497.4 ± 67.6 and TEBF: 604.5 ± 127.1. A significant difference was found between group FSUI and groups SF, SDR‐FSU, and TEBF. Group FSUB showed significantly higher mean strain and greater standard deviation than all other groups due to cuspal fractures, and was thus excluded from the statistical analysis. Conclusions: The tested bulk‐fill composite resins exerted less strain onto tooth structure than the incrementally placed conventional composite resin, although the magnitude of generated strain was product‐dependent. Bulk‐filling with conventional composite resins is contraindicated. Clinical significance: Bulk‐fill composite resins exerted less strain onto adjacent tooth structure than a traditional composite, even when that composite is was placed incrementally. Objectives: The purpose of this study was to investigate the microhardness, polymerization shrinkage, and translucency of bulk-fill composites (BFCs) which have different light attenuation properties and filler contents. Materials and methods: Five BFCs [Filtek Bulk Fill (FB), SureFil SDR (SS), Venus Bulk Fill (VB), SonicFill (SF), and Tetric N-Ceram Bulk Fill (TB)] and two resin-based composites (RBCs) [Tetric N-Ceram (TN) and Filtek Z350XT Flowable (ZF)] were chosen. Numbers of transmitted photons, refractive index, microhardness at different thicknesses, polymerization shrinkage, and translucency parameter (TP) were evaluated. Results: Attenuation coefficients (ACs) based on measured photons ranged from −0.88 to −1.90. BFCs, except SF, had lower AC values than TN or ZF, and BFCs, except TN, had smaller refractive index decreases between top and bottom surfaces. Regardless of an exponential decrease in photon counts, microhardness decreased linearly as specimen thickens. Moreover, microhardness of BFCs showed smaller top-to-bottom decreases (11.5-48.8 %) than TN or ZF (57.3 and 71.5 %, respectively). BFCs with low filler contents showed lower microhardness and higher polymerization shrinkage than those of high filler contents. Also, BFCs had significantly higher ( p < 0.001) TP values than TN or ZF. Conclusion: BFCs attenuated light less than RBCs. However, some BFCs had much lower top surface microhardness and higher polymerization shrinkage than tested RBCs. Clinical relevance: Despite the convenience of bulk filling, careful selection of BFC is advised because some BFCs have worse microhardness and polymerization shrinkage than RBCs due to their lower filler contents. This study aimed to determine the correlation between the volumetric parameters derived from the images of the second, third, and fourth cervical vertebrae by using cone beam computed tomography with skeletal maturation stages and to propose a new formula for predicting skeletal maturation by using regression analysis. We obtained the estimation of skeletal maturation levels from hand-wrist radiographs and volume parameters derived from the second, third, and fourth cervical vertebrae bodies from 102 Japanese patients (54 women and 48 men, 5–18 years of age). We performed Pearson’s correlation coefficient analysis and simple regression analysis. All volume parameters derived from the second, third, and fourth cervical vertebrae exhibited statistically significant correlations (P<0.05). The simple regression model with the greatest R-square indicated the fourth-cervical-vertebra volume as an independent variable with a variance inflation factor less than ten. The explanation power was 81.76%. Volumetric parameters of cervical vertebrae using cone beam computed tomography are useful in regression models. The derived regression model has the potential for clinical application as it enables a simple and quantitative analysis to evaluate skeletal maturation level. [ABSTRACT FROM AUTHOR]

Veneer fracture is the most common complication in zirconia-based restorations. The aim of this study was to evaluate the mechanical behavior of a zirconia-based crown in a lower canine tooth supporting removable partial denture (RPD) prosthesis, varying the bond quality of the veneer/coping interface. Microtomography (μCT) data of an extracted left lower canine were used to build the finite element model (M) varying the core material (gold core – MAu; zirconia core – MZi) and the quality of the veneer/core interface (complete bonded – MZi; incomplete bonded – MZi-NL). The incomplete bonding condition was only applied for zirconia coping by using contact elements (Target/Contact) with 0.3 frictional coefficients. Stress fields were obtained using Ansys Workbench 10.0. The loading condition (L = 1 N) was vertically applied at the base of the RPD prosthesis metallic support towards the dental apex. Maximum principal (σmax) and von Mises equivalent (σvM) stresses were obtained. The σmax(MPa) for the bonded condition was similar between gold and zirconia cores (MAu, 0.42; MZi,0.40). The incomplete bonded condition (MZi-NL) raised σmaxin the veneer up to 800% (3.23 MPa) in contrast to the bonded condition. The peak of σvMincreased up to 270% in the MZi-NL. Objectives: The purpose of this study was to establish multivariable regression models for the estimation of skeletal maturation status in Japanese boys and girls using the cone-beam computed tomography (CBCT)-based cervical vertebral maturation (CVM) assessment method and hand-wrist radiography. Materials and methods: The analyzed sample consisted of hand-wrist radiographs and CBCT images from 47 boys and 57 girls. To quantitatively evaluate the correlation between the skeletal maturation status and measurement ratios, a CBCT-based CVM assessment method was applied to the second, third, and fourth cervical vertebrae. Pearson's correlation coefficient analysis and multivariable regression analysis were used to determine the ratios for each of the cervical vertebrae ( p < 0.05). Results: Four characteristic parameters ((OH + PH)/W, (OH + AH)/W, D, AH/W), as independent variables, were used to build the multivariable regression models: for the Japanese boys, the skeletal maturation status according to the CBCT-based quantitative cervical vertebral maturation (QCVM) assessment was 5.90 + 99.11 × AH/W − 14.88 × (OH + AH)/W + 13.24 × D; for the Japanese girls, it was 41.39 + 59.52 × AH/W − 15.88 × (OH + PH)/W + 10.93 × D. Conclusions: The CBCT-generated CVM images proved very useful to the definition of the cervical vertebral body and the odontoid process. The newly developed CBCT-based QCVM assessment method showed a high correlation between the derived ratios from the second cervical vertebral body and odontoid process. Clinical relevance: There are high correlations between the skeletal maturation status and the ratios of the second cervical vertebra based on the remnant of dentocentral synchondrosis. The selection of a light-curing unit for the curing composite resins is important to achieve best outcomes. The purpose of the present study was to test lasers of 457 and 473 nm alone or in combination under different light conditions with respect to the cure of composite resins. Four different composite resins were light cured using five different laser combinations (530 mW/cm 457 nm only, 530 mW/cm 473 nm only, 177 mW/cm 457 + 177 mW/cm 473 nm, 265 mW/cm 457 + 265 mW/cm 473 nm, and 354 mW/cm 457 + 354 mW/cm 473 nm). Microhardness and polymerization shrinkage were evaluated. A light-emitting diode (LED) unit was used for comparison purposes. On top surfaces, after aging for 24 h, microhardness achieved using the LED unit and the lasers with different conditions ranged 42.4-65.5 and 38.9-67.7 Hv, respectively, and on bottom surfaces, corresponding ranges were 25.2-56.1 and 18.5-55.7 Hv, respectively. Of the conditions used, 354 mW/cm 457 nm + 354 mW/cm 473 nm produced the highest bottom microhardness (33.8-55.6 Hv). On top and bottom surfaces, microhardness by the lowest total light intensity, 354 (177 × 2) mW/cm, ranged 39.0-60.5 and 18.5-52.8 Hv, respectively. Generally, 530 mW/cm at 457 nm produced the lowest polymerization shrinkage. However, shrinkage values obtained using all five laser conditions were similar. Objective: The purpose of the present study was to investigate the usefulness of 457 and 473 nm lasers for the curing of composite resins during the restoration of damaged tooth cavity. Background data: Monochromaticity and coherence are attractive features of laser compared with most other light sources. Better polymerization of composite resins can be expected. Materials and methods: Eight composite resins were light cured using these two lasers and a light-emitting diode (LED) light-curing unit (LCU). To evaluate the degrees of polymerization achieved, polymerization shrinkage and flexural and compressive properties were measured and compared. Results: Polymerization shrinkage values by 457 and 473 nm laser, and LED ranged from 10.9 to 26.8, from 13.2 to 26.1, and from 11.5 to 26.3 μm, respectively. The values by 457 nm laser was significantly different from those by 473 and LED LCU ( p<0.05). However, there was no statistical difference between values by 473 and LED LCU. Before immersion in distilled water, flexural strength (FS) and compressive modulus (CM) of the specimens were inconsistently influenced by LCUs. On the other hand, flexural modulus (FM) and compressive strength (CS) were not significantly different for the three LCUs ( p>0.05). For the tested LCUs, no specific LCU could consistently achieve highest strength and modulus from the specimens tested. Conclusions: Two lasers (457 and 473 nm) can polymerize composite resins to the level that LED LCU can achieve despite inconsistent trends of polymerization shrinkage and flexural and compressive properties of the tested specimens. Objective: The purpose of the present study was to test the usefulness of 457 nm diode-pumped solid state (DPSS) laser as a light source to cure composite resins. Materials and methods: Five different composite resins were light cured using three different light-curing units (LCUs): a DPSS 457 nm laser (LAS), a light-emitting diode (LED), and quartz-tungsten-halogen (QTH) units. The light intensity of LAS was 560 mW/cm2, whereas LED and QTH LCUs was ∼900 mW/cm2. The degree of polymerization was tested by evaluating microhardness, cross-link density, and polymerization shrinkage. Results: Before water immersion, the microhardness of laser-treated specimens ranged from 40.8 to 84.7 HV and from 31.7 to 79.0 HV on the top and bottom surfaces, respectively, and these values were 3.3-23.2% and 2.9-31.1% lower than the highest microhardness obtained using LED or QTH LCUs. Also, laser-treated specimens had lower top and bottom microhardnesses than the other LCUs treated specimens by 2.4-19.4% and 1.4-27.8%, respectively. After ethanol immersion for 24 h, the microhardness of laser-treated specimens ranged from 20.3 to 63.2 HV on top and bottom surfaces, but from 24.9 to 71.5 HV when specimens were cured using the other LCUs. Polymerization shrinkage was 9.8-14.7 μm for laser-treated specimens, and these were significantly similar or lower (10.2-16.0 μm) than those obtained using the other LCUs. This study evaluated the shear bond strength (SBS) and adhesive remnant index (ARI) of ceramic brackets with different base designs using a 473-nm diode-pumped solid-state (DPSS) laser to test its usefulness as a light source. A total of 180 caries-free human premolars were divided into four groups according to the base designs: microcrystalline, crystalline particle (CP), dovetail, and mesh. For each base design, teeth were divided into three different subgroups for light curing using three different light-curing units (LCUs) (quartz-tungsten-halogen unit, light-emitting diode unit, and a DPSS laser of 473 nm). Applied light intensities for the DPSS laser and the other LCUs were approximately 630 and 900 mW/cm, respectively. Stainless steel brackets with a mesh design served as controls. The failure modes of debonded brackets were scored using ARI. As a result, brackets bonded using the DPSS laser had the highest SBS values (16.5-27.3 MPa) among the LCUs regardless of base design. Regarding base designs, the CP groups showed the highest SBS values (22.9-27.3 MPa) regardless of LCU. Furthermore, stainless steel brackets with a mesh design had the lowest SBS values regardless of LCU. In many cases, brackets bonded using the DPSS laser had higher ARI scores and had more adhesive on their bases than on tooth surfaces. Lasers have many favorable features as a light source owing to their monochromaticity and coherence. This study examined the mechanical properties of composite resins that were light-cured using a diode-pumped solid state (DPSS) laser. Eight composite resins were light-cured using four different light sources (one quartz-tungsten-halogen (QTH), two light-emitting diodes (LEDs), and one DPSS laser with a wavelength of 473 nm). The light intensity of the DPSS laser and remaining light-curing units were approximately 500 and 900 mW/cm, respectively. The microhardness, flexural properties, and compressive properties were evaluated using the Vickers hardness test, three-point bending test, and compression test, respectively. In most cases, the microhardness, flexural properties, and compressive properties of the specimens light-cured using the DPSS laser were similar to those obtained using the other light-curing units. Within the limits of the study, the microhardness, flexural modulus, and compressive strength were linearly correlated with the filler content (in weight percent). The flexural modulus and compressive modulus were also linearly correlated with the microhardness. Even with a much lower light intensity, the DPSS laser with a wavelength of 473 nm can polymerize composite resins and give comparable mechanical properties to those obtained using the other light-curing units. Abstract: Objective: Cells derived from the neural crest (NC) contribute to the development of several adult tissues, including tooth and periodontal tissue. Here, two transgenic lines, Wnt1-Cre/ZEG and P0-Cre/ZEG, were analysed to determine the fate and distribution of neural crest cells (NCCs) in adult mouse periodontal ligament (PDL). Design: Paraffin-embedded and decalcified histology samples were prepared from Wnt1-Cre/ZEG and P0-Cre/ZEG mice that were 4-, 8-, or 12-weeks old. Expression of GFP (NC-derived cells), NC-markers (Slug, AP-2 alpha, HNK-1, p75NTR and Nestin), and mesenchymal stem cell markers (CD29 and STRO-1) were examined using immunohistochemistry. Results: In four-week-old Wnt1-Cre/ZEG mice, GFP(+) NC-derived cells were specifically detected in the mid-zone of PDL. The GFP(+) cells constituted 1.4% of all cells in PDL, and this percentage decreased as the mice aged. The distribution and prevalence of GFP(+) cells were comparable between Wnt1-Cre/ZEG and P0-Cre/ZEG mice. NC-marker(+) cells were expressed only in GFP(+) cells while MSC markers were detected only in GFP(−) cells. Conclusion: The prevalence and specific distribution of NC-derived cells in adult PDL of Wnt1-Cre/ZEG and P0-Cre/ZEG mouse were examined. Interestingly, various NC markers, including markers for undifferentiated NCCs, were still expressed at high levels in GFP(+) cells. These observations may indicate that labelled cells in the Wnt1-Cre/ZEG and P0-Cre/ZEG mice did not constituted all NC-derived cells, but rather an interesting subset of NC-derived cells. This study evaluated the shear bond strength (SBS) of orthodontic brackets bonded to teeth using a diode-pumped solid state (DPSS) laser of 473 nm with various light intensity and light-curing settings. For the study, a total of 150 extracted human teeth were divided into ten groups. In the control group, the brackets were bonded to the teeth using a quartz-tungsten-halogen (QTH) light with an intensity of 900 mW/cm(2). In the experimental groups, the brackets were bonded using a DPSS laser with three different light intensities and light-curing times. The same bracket type and adhesive were used in all groups throughout the study. Statistical analysis was performed to compare the SBS, and adhesive remnant index (ARI) among the groups. As results, brackets bonded using the DPSS laser with an intensity of 700 mW/cm(2) for 40 s (totally) showed a slightly higher SBS (12.2 ± 1.8 MPa) than that of those bonded using a QTH light (control; 11.6 ± 1.6 MPa). The SBS values linearly increased with increasing energy density (light intensity × light-curing time) of the DPSS laser (R = 0.95, p < 0.001). However, the SBS values among the test groups were similar regardless of the difference in light-curing conditions. Abstract: Objectives: This study investigated the surrounding bone stress and the implant–bone interfacial sliding of implant designs and implant sizes of immediately loaded implant with maxillary sinus augmentation by using three-dimensional (3D) non-linear finite element (FE) analysis. Methods: Twenty-four FE models including four implant designs (cylindrical, threaded, stepped and step-thread implants) and three implant dimensions (standard, long and wide threaded implants) with a bonded and three levels of frictional contact of implant–bone interfaces were analyzed. The maxillary model was constructed from computer tomography (CT) images of a human skull and all 3D implant models were created via the computer-aided design (CAD) software. Results: The use of threaded implants decreased the bone stress and sliding distance obviously about 30% as compared with non-threaded (cylindrical and stepped) implants. Increasing the implant''s length or diameter reduced the bone stress by 13–26%. Employing a immediately loaded implant with smooth machine surface (μ =0.3, μ represents frictional coefficient) increased the bone stress by 28–63% as compared with the osseointegrated (bonded interfaces) implants. Roughening the implant surface (μ >0.3) did not reduced the bone stress, however it did decrease the interfacial sliding between implant and bone. Conclusions: For an immediately loaded implant placed with sinus augmentation, using threaded implant could decrease both the bone stress and implant–bone sliding distance which may improve the implant initial stability and long-term survival. Rough surface of implants shows no benefit to reduce the bone stress but they could lower the interfacial sliding. On the contrary, employing long or wide implants decrease the bone stress but they cannot diminish the interfacial sliding. [Copyright &y& Elsevier]

Many researches have addressed the high correlation between the fracture of restored teeth and the prepared cavity geometry. In addition, concerns about bonding versus debonding dental materials from cavity walls and different occlusal force conditions could also alter the mechanical responses in a restored tooth. This study employed an automatic mesh procedure to investigate the mechanical interactions between different interfacial conditions and cavity parameters such as pulpal wall depth under different chewing functions. The results indicated that when occlusal force was applied directly on the tooth, it could increase unfavourable stress dramatically. When interfacial fixation was simulated as the contact condition between the tooth tissue and restorative material, it might increase the fracture potential exponentially compared with the bonded interface. For pulpal wall depth analyses, greater risks of fracture for the remaining tooth were observed in deeper cavity of mesio-occlusal-distal (MOD) restorations and the existence of a pulpal wall is essential even it is only 1 mm above the gingival wall. How the nervous system regulates bone remodeling is an exciting area of emerging research in bone biology. Accumulating evidence suggest that neurotransmitter-mediated inputs from neurons may act directly on osteoclasts. Dopamine is a neurotransmitter that can be released by hypothalamic neurons to regulate bone metabolism through the hypothalamic-pituitary-gonadal axis. Dopamine is also present in sympathetic nerves that penetrate skeletal structures throughout the body. It has been shown that dopamine suppresses osteoclast differentiation via a D2-like receptors (D2R)-dependent manner, but the intracellular secondary signaling pathway has not been elucidated. In this study, we found that cAMP-response element binding protein (CREB) activity responds to dopamine treatment during osteoclastogenesis. Considering the critical role of CREB in osteoclastogenesis, we hypothesize that CREB may be a critical target in dopamine's regulation of osteoclast differentiation. We confirmed that D2R is also present in RAW cells and activated by dopamine. Binding of dopamine to D2R inhibits the cyclic adenosine monophosphate (cAMP)/protein kinase A (PKA) signaling pathway which ultimately decreases CREB phosphorylation during osteoclastogenesis. This was also associated with diminished expression of osteoclast markers that are downstream of CREB. Pharmacological activation of adenylate cyclase (to increase cAMP production) and PKA reverses the effect of dopamine on CREB activity and osteoclastogenesis. Therefore, we have identified D2R/cAMP/PKA/CREB as a candidate pathway that mediates dopamine's inhibition of osteoclast differentiation. These findings will contribute to our understanding of how the nervous and skeletal systems interact to regulate bone remodeling. This will enable future work toward elucidating the role of the nervous system in bone development, repair, aging, and degenerative disease. Dentin hypersensitivity (DH) is one of the most common clinical conditions usually associated with exposed dentinal surfaces. In this study, we identified the effectiveness of poly(amidoamine) (PAMAM) dendrimer-coated mesoporous bioactive glass nanoparticles (MBN) (PAMAM@MBN) on DH treatment, examining the ion-releasing effect, dentin remineralization, and the occluding effect of dentinal tubules. We synthesized MBN and PAMAM@MBN. After soaking each sample in simulated body fluid (SBF), we observed ion-releasing effects and dentin remineralization effects for 30 days. Also, we prepared 30 premolars to find the ratio of occluded dentinal tubules after applying MBN and PAMAM@MBN, respectively. The results showed that PAMAM did not disrupt the calcium ion-releasing ability or the dentin remineralization of MBN. The PAMAM@MBN showed a better occluding effect for dentinal tubules than that of MBN (p < 0.05). In terms of dentinal tubule occlusion, the gap between MBN was well occluded due to PAMAM. Although bone marrow–derived mesenchymal stem cells (MSCs) have been extensively explored in bone tissue engineering, only few studies using mesenchymal stem cells from mandible (M-MSCs) have been reported. However, mesenchymal stem cells from mandible have the potential to be as effective as femur-derived mesenchymal stem cells (F-MSCs) in regenerating bone, especially in the orofacial regions, which share embryonic origin, proximity, and accessibility. M-MSCs were isolated and characterized using mesenchymal stem cell–specific markers, colony forming assay, and multi-potential differentiation. In vitro osteogenic potential, including proliferation, osteogenic gene expression, alkaline phosphatase activity, and mineralization, was examined and compared. Furthermore, in vivo bone formations of F-MSCs and M-MSCs in rat critical sized defect were evaluated using microCT and histology. M-MSCs from rat could be successfully isolated and expanded while preserving their MSC's characteristics. M-MSCs demonstrated a comparable proliferation and mineralization potentials and in vivo bone formation as F-MSCs. White spot lesions (WSLs), a side effect of orthodontic treatment, can result in reversible and unaesthetic results. Graphene oxide (GO) with a bioactive glass (BAG) mixture (BAG@GO) was added to Low-Viscosity Transbond XT (LV) in a ratio of 1, 3, and 5%. The composite's characterization and its physical and biological properties were verified with scanning electron microscopy (SEM) and X-ray diffraction (XRD); its microhardness, shear bond strength (SBS), cell viability, and adhesive remnant index (ARI) were also assessed. Efficiency in reducing WSL was evaluated using antibacterial activity of S. mutans. Anti-demineralization was analyzed using a cycle of the acid-base solution. Adhesives with 3 wt.% or 5 wt.% of BAG@GO showed significant increase in microhardness compared with LV. The sample and LV groups showed no significant differences in SBS or ARI. The cell viability test confirmed that none of the sample groups showed higher toxicity compared to the LV group. Antibacterial activity was higher in the 48-h group than in the 24 h group; the 48 h test showed that BAG@GO had a high antibacterial effect, which was more pronounced in 5 wt.% of BAG@GO. Anti-demineralization effect was higher in the BAG@GO-group than in the LV-group; the higher the BAG@GO concentration, the higher the anti-demineralization effect. [ABSTRACT FROM AUTHOR]

Our laboratory utilized biomimicry to develop a synthetic bone scaffold based on hydroxyapatite-gelatin-calcium silicate (HGCS). Here, we evaluated the potential of HGCS scaffold in bone formation in vivo using the rat calvarial critical-sized defect (CSD). Twelve Sprague-Dawley rats were randomized to four groups: control (defect only), decellularized bone matrix (DECBM), and HGCS with and without multipotent adult progenitor cells (MAPCs). DECBM was prepared by removing all the cells using SDS and NH4OH. After 12 weeks, the CSD specimens were harvested to evaluate radiographical, histological, and histomorphometrical outcomes. The in vitro osteogenic effects of the materials were studied by focal adhesion, MTS, and alizarin red. Micro-CT analysis indicated that the DECBM and the HGCS scaffold groups developed greater radiopaque areas than the other groups. Bone regeneration, assessed using histological analysis and fluorochrome labeling, was the highest in the HGCS scaffold seeded with MAPCs. The DECBM group showed limited osteoinductivity, causing a gap between the implant and host tissue. The group grafted with HGCS+MAPCs resulting in twice as much new bone formation seems to indicate a role for effective bone regeneration. In conclusion, the novel HGCS scaffold could improve bone regeneration and is a promising carrier for stem cell-mediated bone regeneration. [ABSTRACT FROM AUTHOR]
