Maritime archaeological investigations of the wreck of the medieval warship Gribshunden (1495), flagship of King Hans of Denmark and Norway, have revealed diverse artifacts including exotic spices imported from far distant origins: saffron, ginger, clove, peppercorns, and almond. The special circumstances of the vessel's last voyage add unique context to the assemblage. Gribshunden and an accompanying squadron conveyed the king, courtiers, noblemen, and soldiers from Copenhagen to a political summit in Kalmar, Sweden. At that conference, Hans expected the Swedish Council to elect him king of Sweden, and thereby fulfill his ambition to reunify the Nordic region under a single crown. To achieve this, Hans assembled in his fleet and particularly aboard his flagship the people and elite cultural signifiers that would convince the Swedish delegation to accept his rule. Along the way, the ships anchored near Ronneby, Blekinge. Written sources record that an explosion and fire caused Gribshunden to sink off Stora Ekön (Great Oak Island). Exotic spices were status markers among the aristocracy in Scandinavia and around the Baltic Sea during the Middle Ages (1050–1550 CE). Until the Gribshunden finds, these extravagances have rarely or never been represented archaeologically. Evidence of their use and consumption in medieval Scandinavia has been limited to sparse written references. We present here the botanical remains from the Gribshunden shipwreck and compare them to previous archaeobotanical finds from the medieval Baltic region. These opulent status symbols traveled with a medieval king en route to a major historical event. Stable isotope analysis (N and C) of fossil cereal grains is regularly explored as a means of obtaining insights to past crop growing conditions and agricultural practices. In this study, we assessed how several growth conditions can affect δ15N and δ13C values of modern cereal grain with the aim to help understand isotopic values from ancient cereal remains. We investigated the impact of fertilizer intensity, plant density, and soil type on cereal grain δ15N and δ13C values in a short-term field experiment using hulled and naked barley, bread wheat, emmer wheat, einkorn, spelt wheat, rye, and oat. We found the following key results: 1) while fertilizing had a significant effect on δ15N values in grain, the impact varied between species, and cereals grown in heavy clay had consistently higher δ15N values in grain compared to those grown in light sandy soil, and 2) the δ13C values were significantly different between cereal species, but the difference was very similar between the two experimental site-locations. In this paper, we examine the exchange of crops and livestock through the application of strontium (87Sr/86Sr) isotope analysis on cereal grains and faunal tooth enamel from the regional center of Uppåkra and three nearby settlements in Scania, southern Sweden, dating to the first millennium AD. Around a third of the fauna have non-local 87Sr/86Sr values, indicating the import of livestock from several different regions. After cleaning, almost all of the cereal grains have non-local 87Sr/86Sr values, which is surprising given the nearby abundance of fertile agricultural soils. We therefore suggest considering non-locally grown crops to be those whose 87Sr/86Sr values fall outside the normal distribution; if this approach is used, around 20% of the analyzed crop samples are interpreted as having grown non-locally. This study demonstrates the potential of combining strontium isotopic data of archaeobotanical and zooarchaeological material for gaining insights into the movement of agricultural products in prehistory. The excavation of a low-temperature kiln structure at an affluent Iron Age regional center, Uppåkra, located in southern Sweden, revealed from archeobotanical samples and its context evidence of malting in the process to make beer. Carbonized germinated hulled barley grain (Hordeum vulgare) was recovered from the kiln structure itself and from the surrounding occupational surface. Located somewhat from the central area of the site, where previous excavations have uncovered hall-buildings, a ceremonial structure, and several smaller houses, the investigated kiln was situated in an area on the site that is absent of remains to indicate a living quarter. Activities using kilns have instead primarily been linked to this area and archeological finds are mainly of charred crops remains. In this paper, we argue that the germination of grain was deliberate and that the kiln was used to stop the germination process by drying or roasting the grain. If the malting process for large-scale beer production was carried out at a designated area of the site is discussed, as well as if this activity area was part of a structural organization observed elsewhere on the settlement. This study uses crop stable nitrogen isotope analysis of charred grain to explore manuring practices in arable production at the affluent regional center Uppåkra and a set of smaller surrounding sites, dating to the first millennium AD in southern Sweden. The isotopic analysis focuses on hulled barley, the principle crop in the Scandinavian Iron Age, and the minor crops: bread wheat, emmer wheat, rye and oat, are included to compare manuring practices in cultivation of other crop species during this period. A field experiment was first conducted to establish relationships between manuring and δ15N values in modern grain from known growing conditions. The data formed an interpretive framework to reconstruct past agricultural practices and manuring intensity in the archaeological study area. Our results from the ancient grains have demonstrated that barley from the early phase in the study area (AD 0–200) varies widely in its δ15N values, reflecting mixed manuring regimes. In the following periods (AD 200–1000), isotopic values are relatively high overall, indicating systematic input of manure. In this paper, we explore whether the isotopic data that indicates sustained and high manuring levels could reflect the wealth of Uppåkra and its surrounding areas by showing prosperity also in its agricultural production, since intensive manuring would have required more resource and labor investments. Films engineered to control the transport of liquids are widely used through society. Examples include barriers in packaging, wound care products, and controlled release coatings in pharmaceutics. When observed at the macroscopic scale such films commonly appear homogeneous, however, a closer look reveals a complex nano- and microstructure that together with the chemical properties of the different domains control the transport properties. In this review we compare and discuss macroscopic transport properties, measured using the straightforward, yet highly powerful technique “modified Ussing chambers”, also denoted side-by-side diffusion cells, for a wide range of structured polymer films and composites. We also discuss and compare the macroscopic observations and conclusions on materials properties with that of lattice Boltzmann simulations of transport properties based on underlying material structure and chemistry. The survey of the field: (i) highlights the use and power of modified Ussing Chambers for determining liquid transport properties of polymer films, (ii) demonstrates the predictability in both directions between macroscopic observations of transport using modified Ussing chambers and structure-based simulations, and (iii) provides experimental and theoretical insights regarding the transport-determining properties of structured polymer films and composites. A metric analysis on hulled barley grain from the Iron Age regional centre of Uppåkra and surrounding sites in southern Sweden has identified a variation in the size of the grain found on these archaeological sites. Large, high-quality grain was found more frequently at Uppåkra when compared to sites in the surrounding area, where smaller grain was more frequent. The observed large grain found at Uppåkra was, however, restricted to only a few house contexts, including hall-buildings, while other contexts on the site, such as areas dedicated to craft production, had barley assemblages containing smaller grain, similar to the size range found on the surrounding sites. The intra-site variation between different contexts at Uppåkra points to a degree of sorting for larger grain and that this variation between grain assemblages was the result of selection after the crop processing was completed. The distribution of grain size at Uppåkra shows a pattern that indicates that the high-quality barley grain was indented for specific individuals or households. The different contexts at Uppåkra have together produced a record spanning the first millennium ad, representing almost the whole existence of the site. The evidence for selection of larger grain can be seen in the hall-buildings throughout most of the first millennium ad, although less prominently during the Late Roman Iron Age (ad 200-400), while during the Migration Period (ad 400-550) several houses on the main site Uppåkra had assemblages of large grain size. The distribution of grain size at the regional centre Uppåkra shows a pattern that indicates that the handling of large high-quality barley grain was part of a spatial organization, and such organization is similar to other functions observed on the site. The long-term record of grain size patterns across time shows that a structure for handling grain was already in place during the early phase of the settlement and that it remained for centuries. This study indicates that the affluence otherwise seen at the regional centre Uppåkra from an abundance of high-status objects, could also include agricultural wealth, with extensive access to high-quality grain. To date, the quest for cost-effective methods for removal of dissolved metals from aqueous solutions remains a daunting challenge for many industries. This paper reports on the development of an effective, hybrid adsorbent for selective copper recovery from aqueous solutions under industrially relevant conditions. The work involved (i) purification and functionalization of diatomaceous earth (DE) particles with glutaraldehyde(GA)-crosslinked polyethyleneimine(PEI), (ii) physicochemical characterization of the product and (iii) metal adsorption from solutions containing Cu(II)/Ni(II)/Fe(II)/Ca(II)/Mg(II)/Mn(II)/Al(III)/Na ions and their subsequent elution behaviour. Acid leaching of the pristine DE led to a significant reduction in its metal oxide (e.g., Al 2 O 3 , Fe 2 O 3 ) constituents and a concomitant increase in both SiO 2 content and specific surface area. Upon functionalization with GA-crosslinked PEI, the DE particles' interfacial chemistry was completely altered to that of the polymer with no change in specific surface area. Isothermal batch adsorption from saline (15 g/dm 3 NaCl) and non-saline solutions containing 500 and 1000 mg/dm 3 of Cu at ~ pH 4 revealed > 97% Cu(II) removal by functionalized DE within 3 min in both cases. Subsequent water elution tests at pH 1 showed complete release of the adsorbed Cu confirming pH-dependent interaction between dissolved Cu and GA-crosslinked PEI. The preliminary batch adsorption/elution tests involving 1000 mg/dm 3 solutions of Ni, Ca, Mg, Mn, Fe, Na and Al showed little or negligible affinity of the functionalized DE towards these elements, suggesting good selectivity for Cu. Furthermore, it is shown that the functionalized particles are chemically stable at H 2 SO 4 concentrations up to 2 M and may be recycled > 10 times without loss in their Cu adsorption/desorption performance. Measurements of archaeological cereal grains has the potential to improve interpretations of archaeobotanical assemblages and to address a broad range of research questions related to agricultural practices in the past. However, understanding the role of the many factors that can influence the morphometrics of cereal grains is of importance to validate the application of morphometrics in archaeobotany. The aim of this study was to investigate the potential effects of growing conditions on cereal grain size using three experimental factors: site-location with different soil types, manure intensity and plant density. A short-term field experiment was conducted at two locations (Alnarp and Holma) in southern Sweden with hulled barley, naked barley, bread wheat, emmer wheat, einkorn, spelt wheat, rye and oat. Our results show that, while all three factors investigated affect grain size, most significant effect was from manuring (p < 0.0001) and site location with different soil types (p < 0.0001), plant density had the weakest effect (p = 0.005), and that cereal species reacted differently to experimental factors. We further use the morphometric dataset to assess the relationship between grain size variation and growing conditions, using the sample range of grain size from different experimental factors. Metallic nanoparticles have been utilized as analytical tools to detect a wide range of organic analytes. In most reports, gold (Au)-based nanosensors have been modified with ligands to introduce selectivity towards a specific target molecule. However, in a recent study a new concept was presented where bare Au-nanorods on self-assembled carboxymethyl-hexanoyl chitosan (CHC) nanocarriers achieved sensitive and selective detection of human serum albumin (HSA) after manipulation of the solution pH. Here this concept was further advanced through optimization of the ratio between Au-nanorods and CHC nanocarriers to create a nanotechnology-based sensor (termed CHC-AuNR nanoprobe) with an outstanding lower detection limit (LDL) for HSA. The CHC-AuNR nanoprobe was evaluated in simulated urine solution and a LDL as low as 1.5 pM was achieved at an estimated AuNR/CHC ratio of 2. Elemental mapping and protein adsorption kinetics over three orders of magnitude in HSA concentration confirmed accumulation of HSA on the nanorods and revealed the adsorption to be completed within 15 min for all investigated concentrations. Lung cancer kills more humans than any other cancer and multidrug resistance (MDR) in cancer stem-like cells (CSC) is emerging as a reason for failed treatments. One concept that addresses this root cause of treatment failure is the utilization of nanoparticles to simultaneously deliver dual drugs to cancer cells with synergistic performance, easy to envision — hard to achieve. (1) It is challenging to simultaneously load drugs of highly different physicochemical properties into one nanoparticle, (2) release kinetics may differ between drugs and (3) general requirements for biomedical nanoparticles apply. Here self-assembled nanoparticles of amphiphilic carboxymethyl-hexanoyl chitosan (CHC) were shown to present nano-microenvironments enabling simultaneous loading of hydrophilic and hydrophobic drugs. This was expanded into a dual-drug nano-delivery system to treat lung CSC. CHC nanoparticles were loaded/chemically modified with the anticancer drug cisplatin and the MDR-suppressing Chinese herbal extract demethoxycurcumin, followed by biofunctionalization with CD133 antibody for enhanced uptake by lung CSC, all in a feasible one-pot preparation. The nanoparticles were characterized with regard to chemistry, size, zeta potential and drug loading/release. Biofunctionalized and non-functionalized nanoparticles were investigated for uptake by lung CSC. Subsequently the cytotoxicity of single and dual drugs, free in solution or in nanoparticles, was evaluated against lung CSC at different doses. From the dose response at different concentrations the degree of synergy was determined through Chou-Talalay’s Plot. The biofunctionalized nanoparticles promoted synergistic effects between the drugs and were highly effective against MDR lung CSC. This review summarizes important advances in polymer-based nanotechnologies for the treatment and diagnosis of cardiovascular diseases, focusing on nanoparticles and nanostructured materials/devices. The current state of the art and design parameters are discussed with the aim of providing a source of information that can aid in the development of new treatments by bridging between materials development, preclinical, and clinical trials. This is highly relevant as cardiovascular diseases remain a leading cause of death worldwide and as current treatments burden the healthcare systems with great costs. Different strategies, from targeting theranostic nanocarriers, to dual-drug depot systems, injectable cell scaffolds, and LDL acceptors are treated. Diabetes is a common global disease that causes immense suffering for individuals and huge costs for the health care system. To minimize complications such as organ degeneration, diabetic patients are required to undergo treatments to maintain the blood glucose level in the normal range, ideally mimicking normal insulin secretion. The normal physiological insulin secretion pattern in healthy individuals consists of a base (basal) level through the day and increased secretion after meals (bolus insulin). Thus effective treatments may combine long acting, low-level insulin therapy with boosts of short acting insulin and/or oral agents. To achieve long term management of basal insulin level, an injectable insulin-loaded gel composed of self-assembled nanoparticles from carboxymethyl-hexanoyl chitosan (CHC) and integrated lysozyme for controlled biodegradation and insulin release was developed. In vitro characterizations and evaluations confirmed that lysozyme was active on CHC and that the amount of lysozyme in a CHC hydrogel determined the degradation and insulin release rate. The degradation products were found to be highly cytocompatible using a cell assay. In vivo evaluation of the system in a diabetic mouse model revealed that the fasted blood glucose level could be maintained in the normal range for 10 days with a single injection of insulin-loaded CHC-lysozyme gel. The insulin-loaded CHC-lysozyme gels clearly show promise for use as a novel injectable long-acting insulin delivery system, with potential to manage the basal insulin level for many days with a single injection. The risk of cardiovascular events increases in individuals with elevated plasma triglyceride (TG) levels, therefore advocating the need for efficient TG-lowering drugs. In the blood circulation, TG levels are regulated by lipoprotein lipase (LPL), an unstable enzyme that is only active as a non-covalently associated homodimer. We recently reported on a N-phenylphthalimide derivative ( 1 ) that stabilizes LPL in vitro , and moderately lowers triglycerides in vivo ( Biochem. Biophys. Res. Commun. 2014 , 450 , 1063). Herein, we establish structure–activity relationships of 51 N-phenylphthalimide analogs of the screening hit 1 . In vitro evaluation highlighted that modifications on the phthalimide moiety were not tolerated and that lipophilic substituents on the central phenyl ring were functionally essential. The substitution pattern on the central phenyl ring also proved important to stabilize LPL. However, in vitro testing demonstrated rapid degradation of the phthalimide fragment in plasma which was addressed by replacing the phthalimide scaffold with other heterocyclic fragments. Apolipoprotein C-II (apoC-II) is the co-factor for lipoprotein lipase (LPL) at the surface of triacylglycerol-rich lipoproteins. LPL hydrolyzes triacylglycerol, which increases local surface pressure as surface area decreases and amphipathic products transiently accumulate at the lipoprotein surface. To understand how apoC-II adapts to these pressure changes, we characterized the behavior of apoC-II at multiple lipid/water interfaces. ApoC-II adsorption to a triacylglycerol/water interface resulted in large increases in surface pressure. ApoC-II was exchangeable at this interface and desorbed on interfacial compressions. These compressions increase surface pressure and mimic the action of LPL. Analysis of gradual compressions showed that apoC-II undergoes a two-step desorption, which indicates that lipid-bound apoC-II can exhibit at least two conformations. We characterized apoC-II at phospholipid/triacylglycerol/water interfaces, which more closely mimic lipoprotein surfaces. ApoC-II had a large exclusion pressure, similar to that of apoC-I and apoC-III. However, apoC-II desorbed at retention pressures higher than those seen with the other apoCs. This suggests that it is unlikely that apoC-I and apoC-III inhibit LPL via displacement of apoC-II from the lipoprotein surface. Upon rapid compressions and re-expansions, re-adsorption of apoC-II increased pressure by lower amounts than its initial adsorption. This indicates that apoC-II removed phospholipid from the interface upon desorption. These results suggest that apoC-II regulates the activity of LPL in a pressure-dependent manner. ApoC-II is provided as a component of triacylglycerol-rich lipoproteins and is the co-factor for LPL as pressure increases. Above its retention pressure, apoC-II desorbs and removes phospholipid. GPIHBP1 is an endothelial membrane protein that transports lipoprotein lipase (LPL) from the subendothelial space to the luminal side of the capillary endothelium. Here, we provide evidence that two regions of GPIHBP1, the acidic N-terminal domain and the central Ly6 domain, interact with LPL as two distinct binding sites. This conclusion is based on comparative binding studies performed with a peptide corresponding to the N-terminal domain of GPIHBP1, the Ly6 domain of GPIHBP1, wild type GPIHBP1, and the Ly6 domain mutant GPIHBP1 Q114P. Although LPL and the N-terminal domain formed a tight but short lived complex, characterized by fast on- and off-rates, the complex between LPL and the Ly6 domain formed more slowly and persisted for a longer time. Unlike the interaction of LPL with the Ly6 domain, the interaction of LPL with the N-terminal domain was significantly weakened by salt. The Q114P mutant bound LPL similarly to the N-terminal domain of GPIHBP1. Heparin dissociated LPL from the N-terminal domain, and partially from wild type GPIHBP1, but was unable to elute the enzyme from the Ly6 domain. When LPL was in complex with the acidic peptide corresponding to the N-terminal domain of GPIHBP1, the enzyme retained its affinity for the Ly6 domain. Furthermore, LPL that was bound to the N-terminal domain interacted with lipoproteins, whereas LPL bound to the Ly6 domain did not. In summary, our data suggest that the two domains of GPIHBP1 interact independently with LPL and that the functionality of LPL depends on its localization on GPIHBP1. [ABSTRACT FROM AUTHOR]

Patients at increased cardiovascular risk commonly display high levels of plasma triglycerides (TGs), elevated LDL cholesterol, small dense LDL particles and low levels of HDL-cholesterol. Many remain at high risk even after successful statin therapy, presumably because TG levels remain high. Lipoprotein lipase (LPL) maintains TG homeostasis in blood by hydrolysis of TG-rich lipoproteins. Efficient clearance of TGs is accompanied by increased levels of HDL-cholesterol and decreased levels of small dense LDL. Given the central role of LPL in lipid metabolism we sought to find small molecules that could increase LPL activity and serve as starting points for drug development efforts against cardiovascular disease. Using a small molecule screening approach we have identified small molecules that can protect LPL from inactivation by the controller protein angiopoietin-like protein 4 during incubations in vitro. One of the selected compounds, 50F10, was directly shown to preserve the active homodimer structure of LPL, as demonstrated by heparin-Sepharose chromatography. On injection to hypertriglyceridemic apolipoprotein A-V deficient mice the compound ameliorated the postprandial response after an olive oil gavage. This is a potential lead compound for the development of drugs that could reduce the residual risk associated with elevated plasma TGs in dyslipidemia. [ABSTRACT FROM AUTHOR]

Biodegradable cell scaffolds and local drug delivery to stimulate cell response are currently receiving much scientific attention. Here we present a nanocomposite that combines biodegradation with controlled release of lithium, which is known to enhance bone growth. Nanogels of lithium neutralized polyacrylic acid were synthesized by microemulsion-templated polymerization and were incorporated into a biodegradable polyhydroxybutyrate (PHB) matrix. Nanogel size was characterized using dynamic light scattering, and the nanocomposites were characterized with regard to structure using scanning electron microscopy, mechanical properties using tensile testing, permeability using tritiated water, and lithium release in PBS using a lithium specific electrode. The nanogels were well dispersed in the composites and the mechanical properties were good, with a decrease in elastic modulus being compensated by increased tolerance to strain in the wet state. Approximately half of the lithium was released over about three hours, with the remaining fraction being trapped in the PHB for subsequent slow release during biodegradation. The prepared nanocomposites seem promising for use as dual functional scaffolds for bone regeneration. Here lithium ions were chosen as model drug, but the nanogels could potentially act as carriers for larger and more complex drugs, possibly while still carrying lithium. [ABSTRACT FROM AUTHOR]

Apolipoproteins (apo) C-I and C-III are known to inhibit lipoprotein lipase (LPL) activity, but the molecular mechanisms for this remain obscure. We present evidence that either apoC-I or apoC-III, when bound to triglyceride-rich lipoproteins, prevent binding of LPL to the lipid/water interface. This results in decreased lipolytic activity of the enzyme. Site-directed mutagenesis revealed that hydrophobic amino acid residues centrally located in the apoC-III molecule are critical for attachment to lipid emulsion particles and consequently inhibition of LPL activity. Triglyceride-rich lipoproteins stabilize LPL and protect the enzyme from inactivating factors such as angiopoietin-like protein 4 (angptl4). The addition of either apoC-I or apoC-III to triglyceride-rich particles severely diminished their protective effect on LPL and rendered the enzyme more susceptible to inactivation by angptl4. These observations were seen using chylomicrons as well as the synthetic lipid emulsion Intr-alipid. In the presence of the LPL activator protein apoC-II, more of apoC-I or apoC-III was needed for displacement of LPL from the lipid/water interface. In conclusion, we show that apoC-I and apoC-III inhibit lipolysis by displacing LPL from lipid emulsion particles. Charred seed conglomerates of Linum usitatissimum (flax) and Camelina sativa (gold of pleasure) were found at Uppåkra 2:25, a Roman Iron Age site in Skåne, southern Sweden. The conglomerates showed no mixing with each other, as they were almost pure flax and gold of pleasure respectively. Together with other archaeobotanical data from the site, they provide new evidence on the use, processing and cultivation of these two plants in early Iron Age in Scandinavia. Metric analyses were applied to flax seeds from both conglomerates and other contexts at this site, and compared to seed assemblages from other Roman Iron Age sites in Europe. The comparison showed that the flax cultivation at Uppåkra 2:25 was intended for the production of oil-rich seeds. The contextual relationship indicates that both flax and gold of pleasure seeds were processed in a similar way and used for oil. Furthermore, the pure seed conglomerate of gold of pleasure suggests that this plant was not a weed, but rather an intentionally grown crop which was cultivated separately from flax. Abstract: Chitosan is among the most abundant biopolymers on earth and has been either used or exhibited potential in a wide variety of industrial and biomedical applications. With the advancement of materials technologies, chitosan has been chemically modified to self-assemble into nanoarchitectures that are usable in advanced biomedical applications, such as drug nanocarriers, macroscopic injectables, tissue-engineering scaffolds, and nanoimaging agents. Colloidal amphiphilically modified chitosan (AMC) is a relatively recent material receiving increased attention with numerous publications addressing the medical advantages of specific systems. To date, many reviews have focused on the synthesis and biomedical properties of chitosan-based biomaterials, but a comprehensive study focusing on the colloidal properties of AMC in relation to biomedical performance appears to be lacking. This review provides a survey of the field, critically reviewing the colloidal properties and biomedical performance of AMC systems, such as nanoparticle drug delivery systems and macroscopic medical devices. Angiopoietin-like protein 4 (Angptl4), a potent regulator of plasma triglyceride metabolism, binds to lipoprotein lipase (LPL) through its N-terminal coiled-coil domain (ccd-Angptl4) inducing dissociation of the dimeric enzyme to inactive monomers. In this study, we demonstrate that fatty acids reduce the inactivation of LPL by Angptl4. This was the case both with ccd-Angptl4 and full-length Angptl4, and the effect was seen in human plasma or in the presence of albumin. The effect decreased in the sequence oleic acid > palmitic acid > myristic acid > linoleic acid > linolenic acid. Surface plasmon resonance, isothermal titration calorimetry, fluorescence, and chromatography measurements revealed that fatty acids bind with high affinity to ccd-Angptl4. The interactions were characterized by fast association and slow dissociation rates, indicating formation of stable complexes. The highest affinity for ccd- Angptl4 was detected for oleic acid with a subnanomolar equilibrium dissociation constant (Kd). The Kd values for palmitic and myristic acid were in the nanomolar range. Linoleic and linolenic acid bound with much lower affinity. On binding of fatty acids, ccd-Angptl4 underwent conformational changes resulting in a decreased helical content, weakened structural stability, dissociation of oligomers, and altered fluorescence properties of the Trp-38 residue that is located close to the putative LPL-binding region. Abstract: Thermo-gelling injectable nanogels, with no burst release of loaded drug, were prepared by a simple route by combining self assembled nanocapsules of amphiphilically modified chitosan with glycerophosphate di-sodium salt and glycerol. The potential as a depot drug delivery system was demonstrated in vivo through the therapeutic effect of ethosuximide (ESM) loaded nanogels, suppressing spike wave discharges (SWDs) in Long Evan rat model. Simultaneously clearance of gels from the site of administration was monitored non-invasively using MRI. The gel structure was characterized using TEM and SEM, confirming the gels to be an assembly of nanocapsules and using two-photon microscopy to visualize the network structure. In vitro drug release studies using ESM revealed that the nanogels exhibited extended, mostly Fickian release. Finally, all investigated formulations displayed excellent cytotoxicity data determined by MTT assay using human retinal pigmented epithelium cells. Abstract: Microfibrillated cellulose (MFC) films containing the water soluble and pharmaceutically approved polymer hydroxypropyl methylcellulose (HPMC) exhibited an unexpected decrease in permeability and excellent one-dimensional swelling properties above a threshold in HPMC content. It is proposed that the observed material characteristics derive from the influence of HPMC on the aggregation behavior of MFC in such a way that above a critical HPMC content the films are created through self assembly into a layered structure, composed of low swelling layers with swellable inter layer regions. The suggested structures were supported by high resolution microscopy. The findings should hold potential for direct applications, but even more as a concept for future material design. Abstract: In this paper a new method for increased time resolution in multi-period Mixed Integer Linear Programming (MILP) optimisation is presented and applied to a district heating system. The proposed method facilitates the analysis of many time periods in multi period MILP optimisation projects. In the paper, a 365 time period model spanning 1year developed with the novel method is compared to a 12 time period model developed with a more conventional methodology. The new method offers a significant decrease in the amount of input data for multi period models and facilitates changes to the analysed time span or resolution in time. In the application of the new method oil savings of 7% compared to the current operational strategy of the district heating system are revealed. [Copyright &y& Elsevier]

Three types of microfibrillated cellulose (MFC) with differences in structure and surface charge were used at low concentration as filler materials in polysodium acrylate superabsorbents (SAPs). The swelling of the composite hydrogels was determined in 0.9% NaCl solution as well as in deionized water. The shear modulus of the samples was determined through uniaxial compression analysis after synthesis and after swelling in 0.9% NaCl solution. Furthermore, the ability to retain filler effects after washing was investigated. The results showed that all of the investigated MFCs had a strong reinforcing effect on the shear modulus after synthesis. The filler effect on swelling and on the associated shear modulus of swollen samples showed a more complicated dependence on structure and surface charge. Finally, it was found that the filler effects were reasonably retained after washing and subsequent drying. The results confirm that MFC holds great potential as a filler material in superabsorbent applications. Furthermore, the results provide some insight on how the structural properties and surface charge of MFC will affect gel properties depending on swelling conditions. Abstract: The solid-state properties were studied for different batches of hydroxypropyl methylcellulose (HPMC). The batches had similar chemical composition, but different degree of heterogeneity with regard to the distribution of the substituents along the polymer chains. The glass transition temperature, T g, was analysed using a new developed method where dynamic mechanic analysis, DMA, was performed in compression mode on compacts, utilizing a wedge-shaped probe. The method was verified by conventional DMA on films. Molecular interactions were studied using FT-IR. In addition, the water vapour sorption was determined by gravimetric measurements and the plasticization by water vapour was studied on film samples using DMA. The results revealed a linear relationship between increasing T g and increasing percent glucose liberated after enzyme hydrolysis. The percent glucose liberated can in turn be considered to account for both the heterogeneity of the substituents and the total degree of substitution. The results indicated that more heterogeneously substituted cellulose derivatives and derivates with a lower degree of substitution had stronger interactions between polymer chains. As expected from these results, some small difference in the plasticization by water vapour could be detected. However, no significant differences were found in molecular interactions using FT-IR or in the sorption of water vapour. The correlation between heterogeneity in the distribution of the substituents and T g is of much interest as heterogeneously substituted batches of HPMC have been previously shown to exhibit very different behaviour in solution and in gelling tablets. Abstract: The robustness of controlled release formulations when co-ingested with alcohol is a current concern expressed by regulatory authorities, especially with regard to dose dumping. One such controlled release formulation commonly used is film coating composed of ethyl cellulose (EC) and hydroxypropyl cellulose (HPC). The aim of this study was to investigate how the presence of ethanol in the dissolution medium affects the water permeability of such films. Film samples were prepared in various EC–HPC compositions, and the effect of different ethanol concentrations in the dissolution medium on the permeability was studied using a modified Ussing chamber and tritiated water. It was found that the effect of ethanol on the film permeability varied depending on the composition of the films. The results were interpreted in terms of swelling of the EC in the films, where the swelling increased with increasing ethanol concentration. Thus, for films with low HPC content (non-interconnected pores), the water permeability of the films increased with increasing ethanol concentration as the diffusion through the ethyl cellulose increased due to swelling. However, for films with higher HPC content (having interconnected pores through the films), the permeability decreased, likely due to the swelling of the ethyl cellulose blocking the pores. Microfibrillated cellulose was utilized at low concentrations as a filler material, added prior to free radical polymerization, in cross-linked superabsorbent polysodium acrylate hydrogels. The effect of microfibrillated cellulose concentration on equilibrium swelling, shear modulus after synthesis, and shear modulus at equilibrium swelling was studied at different degree of cross-linking. For the characterization of the microfibrillated cellulose optical microscopy, atomic force microscopy, and transmittance analysis were used. The shear modulus of the samples was determined using uniaxial compression analysis. The swelling of the gels was determined using classical gravimetrical measurements. It was found that microfibrillated cellulose was highly efficient in increasing the shear modulus of the gels. Furthermore, the microfibrillated cellulose was found to have the same effect on the swelling and shear modulus at equilibrium swelling as the same mass of the conventional covalent cross-linker N,N'-methylenebisacrylamide (MBA), while in fact improving the fracture resistance of the gels. Many species have suffered population declines through loss of suitable habitat. In addition, current agricultural land use and human settlements favor generalist predators, which pose an increasing threat to ground-nesting bird species such as shorebirds (waders). During the last 2 decades, nest exclosures have been used to control nest-predation rates and often improved hatching success. We evaluated the effectiveness of protective nest-cages to boost reproductive success in the endangered southern dunlin (Calidris alpina schinzii) and ultimately to halt the population decline. We found that exclosures successfully increased the survival probability of nests and, thereby, the number of hatchlings, without markedly elevating the predation rate on incubating adults. Nest exclosures did not, however, translate into an increased number of fledglings and recruits produced/breeding adult in the population, showing that factors other than nest survival are also important for population development. Our results highlight that conservation efforts aimed only at removing high nest-predation pressure may be insufficient to preserve declining species such as the southern dunlin. The coke oven plant has a central role in the iron and steel making process in an integrated steel plant. The subject of this research is to study how the production and energy system at the steel industry, with a connected combined heat and power plant, is affected by renovation of the coke oven. The aim is to investigate the interaction between the different processes and how the choice of system boundary affects the operation practice for the steel plant. MILP-based optimization models have been developed and used for the evaluation. The analysis shows that it is very important to take the interactions between the different production units in the system into consideration when making the analysis. A system optimization with a boundary including the whole system has a greater potential for minimizing the total system cost than one that only includes the processes where the actual changes are made. Maintaining soil health is integral to agricultural production, and the archaeological record contains multiple lines of palaeoclimatic and palaeoenvironmental proxy evidence that can contribute to the understanding and analysis of long-term trajectories of change that are key for contextualizing 21st century global environmental challenges. Soil is a capital resource and its nutrient balance is modified by agricultural activities, making it necessary to ensure soil productivity is maintained and managed through human choices and actions. Since prehistory this has always been the case; soil is a non-renewable resource within a human lifetime. Here, we present and interpret carbon and nitrogen isotope analysis of charred cereals from southern Scandinavia. Anthropogenic effects on soils are evident from the initiation of farming 6000 years ago, as is amendment to counteract its effects. The earliest cereals were planted on pristine soils, and by the late Neolithic, agriculture extensified. By the Iron Age it was necessary to significantly amend depleted soils to maintain crop yields. We propose that these data provide a record of soil water retention, net precipitation and amendment. From the start of the Neolithic there is a concurrent decrease in both Δ13C and δ15N, mitigated only by the replacement of soil organic content in the form of manure in the Iron Age. The cereal isotopes provide a record of trajectories of agricultural sustainability and anthropogenic adaptation for nearly the entire history of farming in the region. • Isotope analyses of charred cereals are used to explore long-term soil health. • Soil amendment was practiced from the start of the Scandinavian Neolithic. • Anthropogenic soil depletion was only mitigated in the Iron Age. Lipoprotein lipase (LPL) undergoes spontaneous inactivation via global unfolding and this unfolding is prevented by GPIHBP1 (Mysling et al., 2016). We now show: (1) that ANGPTL4 inactivates LPL by catalyzing the unfolding of its hydrolase domain; (2) that binding to GPIHBP1 renders LPL largely refractory to this inhibition; and (3) that both the LU domain and the intrinsically disordered acidic domain of GPIHBP1 are required for this protective effect. Genetic studies have found that a common polymorphic variant in ANGPTL4 results in lower plasma triglyceride levels. We now report: (1) that this ANGPTL4 variant is less efficient in catalyzing the unfolding of LPL; and (2) that its Glu-to-Lys substitution destabilizes its N-terminal α-helix. Our work elucidates the molecular basis for regulation of LPL activity by ANGPTL4, highlights the physiological relevance of the inherent instability of LPL, and sheds light on the molecular defects in a clinically relevant variant of ANGPTL4. GPIHBP1 is a glycolipid-anchored membrane protein of capillary endothelial cells that binds lipoprotein lipase (LPL) within the interstitial space and shuttles it to the capillary lumen. The LPL·GPIHBP1 complex is responsible for margination of triglyceride-rich lipoproteins along capillaries and their lipolytic processing. The current work conceptualizes a model for the GPIHBP1·LPL interaction based on biophysical measurements with hydrogen-deuterium exchange/mass spectrometry, surface plasmon resonance, and zero-length cross-linking. According to this model, GPIHBP1 comprises two functionally distinct domains: (1) an intrinsically disordered acidic N-terminal domain; and (2) a folded C-terminal domain that tethers GPIHBP1 to the cell membrane by glycosylphosphatidylinositol. We demonstrate that these domains serve different roles in regulating the kinetics of LPL binding. Importantly, the acidic domain stabilizes LPL catalytic activity by mitigating the global unfolding of LPL’s catalytic domain. To identify differences in seed lignin content, 520 randomly chosen independent lines were screened in mutagenized oat population and lines with the seed lignin levels ranging from 20 to 63gkg-1 were identified. In the commercial variety Belinda, from which the mutated population was developed, seed lignin level was determined to be 41gkg-1. In Assiniboia, a Canadian low lignin variety, it was found to be 21gkg-1. To investigate if extracted lignin from the mutated lines were also qualitatively different from Belinda, two lines with the lowest and highest lignin levels were selected for structural analyses using XRD, UV and FT-IR spectroscopy. Results showed that there were significant qualitative differences in seed lignin in the mutated lines and in Belinda, and based on this, we predict that lignin from the mutated seeds will be more digestible in ruminant animals than Belinda seeds. Aims  Fibroblast growth factor (FGF) 21, a key regulator of energy metabolism, is currently evaluated in humans for treatment of type 2 diabetes and non-alcoholic steatohepatitis. However, the effects of FGF21 on cardiovascular benefit, particularly on lipoprotein metabolism in relation to atherogenesis, remain elusive. Methods and results   Here, the role of FGF21 in lipoprotein metabolism in relation to atherosclerosis development was investigated by pharmacological administration of a half-life extended recombinant FGF21 protein to hypercholesterolaemic APOE*3-Leiden.CETP mice, a well-established model mimicking atherosclerosis initiation and development in humans. FGF21 reduced plasma total cholesterol, explained by a reduction in non-HDL-cholesterol. Mechanistically, FGF21 promoted brown adipose tissue (BAT) activation and white adipose tissue (WAT) browning, thereby enhancing the selective uptake of fatty acids from triglyceride-rich lipoproteins into BAT and into browned WAT, consequently accelerating the clearance of the cholesterol-enriched remnants by the liver. In addition, FGF21 reduced body fat, ameliorated glucose tolerance and markedly reduced hepatic steatosis, related to up-regulated hepatic expression of genes involved in fatty acid oxidation and increased hepatic VLDL-triglyceride secretion. Ultimately, FGF21 largely decreased atherosclerotic lesion area, which was mainly explained by the reduction in non-HDL-cholesterol as shown by linear regression analysis, decreased lesion severity, and increased atherosclerotic plaque stability index. Conclusion  FGF21 improves hypercholesterolaemia by accelerating triglyceride-rich lipoprotein turnover as a result of activating BAT and browning of WAT, thereby reducing atherosclerotic lesion severity and increasing atherosclerotic lesion stability index. Abstract: Vanadium in raw materials used in iron- and steelmaking, a particular challenge for Nordic steel producers, affects the composition of the generated slag from the steelmaking vessel, i.e. the basic oxygen furnace (BOF) adversely and reduces the potential for recycling and external utilisation. A process concept under development aims to enrich and extract the vanadium content of slag from the BOF, making use of the vanadium in the slag and also increasing the overall slag use potential. Applications of this concept affect slag compositions and internal material flows in the iron and steel production system, especially when recycling BOF slags as flux in the blast furnace (BF). This paper will present a case study, based on a Process Integration (PI) approach, using a designated system model to simulate the steel production system and the implementation of the process concept, thereby analysing how to obtain maximum usage of metallurgical slags without compromising the quality of the main product, i.e. liquid steel. Different approaches were studied to improve the environmental sustainability in the production system by maximising the material efficiency through vanadium recovery (as FeV alloy) and the use of slags, thereby minimising the stored/deposited slag amounts. At its integrated steel plant in Luleå, SSAB EMEA produces high strength steel via two basic oxygen furnaces (BOFs), type LD/LBE. The BOFs are charged with a mix of hot metal, scrap, and slag formers. The scrap has several functions, for example, as coolant to balance excess heat, and it contributes to high steel production rate and decreased CO2 emission. The optimal scrap to hot metal ratio is influenced by several factors, for example, the excess heat generated in the BOF versus target value of tapping temperature, content of contamination elements versus contents allowed in the steel, possible use of alloys in scrap to decrease the need of alloy addition and the scrap price versus the production cost of hot metal. The first two factors also affect the maximum amount of scrap to be charged. Furthermore, the available scrap exists as several types with different composition, properties, size, and price. For most scrap types there are also uncertainties in composition, which has to be considered. An optimization model has been further developed in combination with some statistic analysis techniques. The present work is focusing on the possibility to use the model as a tool to optimize and control raw material/scrap blending into the BOFs. On the basis of the statistical analysis technique, the scrap sorting in the model will be described, as well as development and introduction of an extended BOF sub-model. This model includes a scrap sorting function and a response on deviations in steel quality. Real production data is used to identify steel quality parameters with consideration of different combination of elements, for example, S, Cr, Ni, and Cu. Plant mutagenesis is rapidly coming of age in the aftermath of recent developments in high-resolution molecular and biochemical techniques. By combining the high variation of mutagenised populations with novel screening methods, traits that are almost impossible to identify by conventional breeding are now being developed and characterised at the molecular level. This paper provides a comprehensive overview of the various techniques and workflows available to researchers today in the field of molecular breeding, and how these tools complement the ones already used in traditional breeding. Both genetic (Targeting Induced Local Lesions in Genomes; TILLING) and phenotypic screens are evaluated. Finally, different ways of bridging the gap between genotype and phenotype are discussed. Background: Inbreeding and loss of genetic diversity are expected to increase the extinction risk of small populations, but detailed tests in natural populations are scarce. We combine long-term population and fitness data with those from two types of molecular markers to examine the role of genetic effects in a declining metapopulation of southern dunlins Calidris alpina schinzii, an endangered shorebird. Results: The decline is associated with increased pairings between related individuals, including close inbreeding (as revealed by both field observations of parentage and molecular markers). Furthermore, reduced genetic diversity seems to affect individual fitness at several life stages. Higher genetic similarity between mates correlates negatively with the pair's hatching success. Moreover, offspring produced by related parents are more homozygous and suffer from increased mortality during embryonic development and possibly also after hatching. Conclusions: Our results demonstrate strong genetic effects in a rapidly declining population, emphasizing the importance of genetic factors for the persistence of small populations. Similar to blood types, human plasma haptoglobin (Hp) is classified into three phenotypes: Hp 1-1, 2-1 and 2-2. They are genetically inherited from two alleles Hp 1 and Hp 2 (represented in bold), but only the Hp 1-1 phenotype is found in almost all animal species. The Hp 2-2 protein consists of complicated large polymers cross-linked by α2-β subunits or (α2-β) n (where n ≥ 3, up to 12 or more), and is associated with the risk of the development of diabetic, cardiovascular and inflammatory diseases. In the present study, we found that deer plasma Hp mimics human Hp 2, containing a tandem repeat over the α-chain based on our cloned cDNA sequence. Interestingly, the isolated deer Hp is homogeneous and tetrameric, i.e. (α-β)4, although the locations of −SH groups (responsible for the formation of polymers) are exactly identical to that of human. Denaturation of deer Hp using 6 m urea under reducing conditions (143 mmβ-mercaptoethanol), followed by renaturation, sustained the formation of (α-β)4, suggesting that the Hp tetramers are not randomly assembled. Interestingly, an α-chain monoclonal antibody (W1), known to recognize both human and deer α-chains, only binds to intact human Hp polymers, but not to deer Hp tetramers. This implies that the epitope of the deer α-chain is no longer exposed on the surface when Hp tetramers are formed. We propose that steric hindrance plays a major role in determining the polymeric formation in human and deer polymers. Phylogenetic and immunochemical analyses revealed that the Hp 2 allele of deer might have arisen at least 25 million years ago. A mechanism involved in forming Hp tetramers is proposed and discussed, and the possibility is raised that the evolved tetrameric structure of deer Hp might confer a physiological advantage. Ground-nesting birds have declined world-wide, probably partly due to high nest predation. A non-lethal method for decreasing predation uses protective cages at nests. Tests have mainly looked at the effect of such nest exclosures on hatching success and adult predation, but several additional aspects need to be explored for a comprehensive evaluation of this conservation technique. Here, we test the effect of nest exclosures in two common European shorebirds: northern lapwing (Vanellus vanellus) and redshank (Tringa totanus), measuring hatching success, incubation length, hatching synchrony, hatchability, partial clutch loss, chick condition, and adult predation. In both species, protected nests had higher hatching success than unprotected nests. Taking into account incubation time, nest abandonment, hatchability and partial clutch loss, protected nests still hatched more young than unprotected controls. In lapwings, but not in redshanks, protected nests were incubated longer, but this did not impair the condition of lapwing chicks. Protected redshanks suffered increased predation on incubating adults, which often sit on the nest until a predator is close by. Our results emphasize the need for caution in the use of nest exclosures, particularly in redshanks and other species with similar incubation behaviour. Exclosures can, however, be a useful management tool in shorebirds that leave their nest early, when an approaching predator is still far away. [Copyright &y& Elsevier]

Isothermal storage and reduction of NO2 with CO, C3H6 and H2 as reducing agents on a lean NOx adsorber was investigated by temperature programmed desorption (TPD) and temperature programmed reduction (TPR) studies. The reduction of NOx was clearly favoured with H2 as reducing agent. Carbon monoxide and C3H6 showed fairly low reduction of NOx. Here we report a newly-synthesized amphiphilic Gadodiamide-conjugated carboxymethyl-hexanoyl chitosan (termed as CHC-Gd) hybrid. The gadodiamide was selected is due to its smallest molecular size among other Gd-based complexes reported in literature, which assumed to give least influence on the resulting physicochemical properties such as colloidal stability, nanostructural evolution, and cytocompability, particularly self-assembly capability, of the resulting hybrid upon practical uses. Experimental outcomes showed a successful synthesis of the CHC-Gd hybrid using a one-pot synthesis protocol, where the gadodiamide complexes were covalently attached to the carboxyl groups along the CHC backbone. Self-assembly behavior can be observed to form a sphere-like nanoparticle of 100–200 nm in size as of amphiphilic native CHC macromolecule. Experimental outcomes indicated a largely improved cytocompatibility of the hybrid, compared with free Gd, suggesting the Gd+3 ions were well stabilized in the CHC nanostructure. Excellent contrastability in-vitro and in particular in vivo were measured, where for in-vivo test, a 10-40-folded reduction in dosage, compared with clinical Gd dose, was used and demonstrated a comparative-to-better imaging resolution and brightness. Therefore, from this preliminary investigation, a potential translation to clinical practice through the use of newly-synthesized amphiphilic CHC-Gd hybrid appears to be relatively promising. The iron and steel industry accounts for about 20% of the annual industrial energy utilization. The intensive fossil fuel consumption in steel industry is associated with CO 2 emission. In the absence of economically feasible and efficient methods for capture and storage of enormous quantities of CO 2 emissions from steel industry, the use of biomass products as a source of energy and reducing agents provides a promising alternative solution for green steel production. However, the biomass application in iron and steel industry is still limited and it suffers strong competition from fossil fuels. The challenges of biomass usage in steel industry are included technical and economic aspects which required synergy between steelmaking and bioenergy sectors. Although intensive work has been carried out separately, there is a lack of link between these two vital sectors. The present article provides a comprehensive review of recent research progresses which have been conducted on biomass upgrading and analysing the opportunities and obstacles for biomass implementation in iron and steel industry. In the first part, an overview on the energy consumption and CO 2 emissions in different iron and steelmaking routes is clarified. Moreover, the potential approaches of biomass conversion processes and upgrading technologies are reviewed. In the second part, an attention has been paid to the utilization of torrefied/pyrolyzed biomass in the energy-intensive ironmaking processes. Biomass addition to coal blend during cokemaking and its influence on the product coke quality is discussed. The partial and complete substitution of coke breeze with biochar in sintering process and its influence on the product sinter quality is explained. The impact of charcoal top charging or injection into blast furnace has been elaborated. Benefits and limitations of biomass application in each process are thoroughly discussed. The use of Zinc-coated steel (e.g. galvanized steel) in melting cycles based on Electric Arc Furnaces can increase the production of harmful dust and hazardous air emissions. This article describes a novel process to simultaneously preheat and remove the coating from the scrap surface before the melting phase. The zinc in coating is removed in the gas phase by chloride containing syngas combustion and collected in a dedicated recovery system. Two possible innovative process routes are described, which involve plastic waste pre-treatment, shredded plastic gasification/pyrolysis, steel scrap preheating and zinc recovery processes. The routes have been modeled in an integrated flowsheet, in order to allow a comprehensive simulation and optimization of the pretreatment processes. The process optimization results in possible energy savings of over 300 MJ/t of preheated scrap charged in the Electric Arc Furnace for steel production. We have investigated and modeled the injection of biomass into blast furnaces (BF), in place of pulverized coal (PC) from fossil sources. This is the easiest way to reduce CO 2 emissions, beyond efficiency-improvements. The considered biomass is either pelletized, torrefied or pyrolyzed. It gives us three cases where we have calculated the maximum replacement ratio for each. It was found that charcoal from pyrolysis can fully replace PC, while torrefied material and pelletized wood can replace 22.8% and 20.0% respectively, by weight. Our energy and mass balance model (MASMOD), with metallurgical sub-models for each zone, further indicates that (1) more Blast Furnace Gas (BFG) will be generated resulting in reduced fuel consumption in an integrated plant, (2) lower need of limestone can be expected, (3) lower amount of generated slag as well, and (4) reduced fuel consumption for heating the hot blast is anticipated. Overall, substantial energy savings are possible, which is one of the main findings in this paper. Due to the high usage of PC in Sweden, large amounts of biomass is required if full substitution by charcoal is pursued (6.19 TWh/y). But according to our study, it is likely available in the long term for the blast furnace designated M3 (located in Luleå). Finally, over a year with almost fully used production capacity (2008 used as reference), a 28.1% reduction in on-site emissions is possible by using charcoal. Torrefied material and wood pellets can reduce the emissions by 6.4% and 5.7% respectively. GPIHBP1, a glycosylphosphatidylinositol-anchored glycoprotein of microvascular endothelial cells, binds lipoprotein lipase (LPL) within the interstitial spaces and transports it across endothelial cells to the capillary lumen. The ability of GPIHBP1 to bind LPL depends on the Ly6 domain, a three-fingered structure containing 10 cysteines and a conserved pattern of disulfide bond formation. Here, we report a patient with severe hypertriglyceridemia who was homozygous for a GPIHBP1 point mutation that converted a serine in the GPIHBP1 Ly6 domain (Ser-107) to a cysteine. Two hypertriglyceridemic siblings were homozygous for the same mutation. All three homozygotes had very low levels of LPL in the preheparin plasma. We suspected that the extra cysteine in GPIHBP1-S107C might prevent the trafficking of the protein to the cell surface, but this was not the case. However, nearly all of the GPIHBP1-S107C on the cell surface was in the form of disulfide-linked dimers and multimers, whereas wild-type GPIHBP1 was predominantly monomeric. An insect cell GPIHBP1 expression system confirmed the propensity of GPIHBP1-S107C to form disulfide-linked dimers and to form multimers. Functional studies showed that only GPIHBP1 monomers bind LPL. In keeping with that finding, there was no binding of LPL to GPI-HBP1-S107C in either cell-based or cell-free binding assays. We conclude that an extra cysteine in the GPIHBP1 Ly6 motif results in multimerization of GPIHBP1, defective LPL binding, and severe hypertriglyceridemia. [ABSTRACT FROM AUTHOR]

Abstract: Lipoprotein lipase (LPL) is important for clearance of triacylglycerols (TG) from plasma both as an enzyme and as a bridging factor between lipoproteins and receptors for endocytosis. The amount of LPL at the luminal side of the capillary endothelium determines to what extent lipids are taken up. Mechanisms to control both the activity of LPL and its transport to the endothelial sites are regulated, but poorly understood. Angiopoietin-like proteins (ANGPTLs) 3 and 4 are potential control proteins for LPL, but plasma concentrations of ANGPTLs do not correlate with plasma TG levels. We investigated the effects of recombinant human N-terminal (NT) ANGPTLs3 and 4 on LPL-mediated bridging of TG-rich lipoproteins to primary mouse hepatocytes and found that the NT-ANGPTLs, in concentrations sufficient to cause inactivation of LPL in vitro, were unable to prevent LPL-mediated lipoprotein uptake. We therefore investigated the effects of lipoproteins (chylomicrons, VLDL and LDL) on the inactivation of LPL in vitro by NT-ANGPTLs3 and 4 and found that LPL activity was protected by TG-rich lipoproteins. In vivo, postprandial TG protected LPL from inactivation by recombinant NT-ANGPTL4 injected to mice. We conclude that lipoprotein-bound LPL is stabilized against inactivation by ANGPTLs. The levels of ANGPTLs found in blood may not be sufficient to overcome this stabilization. Therefore it is likely that the prime site of action of ANGPTLs on LPL is in subendothelial compartments where TG-rich lipoprotein concentration is lower than in blood.
