In order to investigate the effects of different packaging materials on the quality changes of walnut oil, walnut oil was packaged in open glass beakers, tinplate cans, and glass oil bottles, respectively. The Schaal oven method was used to accelerate the oxidation of oil. The peroxide value, acid value, fatty acid composition and relaxation characteristics were used as evaluation indicators to study the effects of different packaging materials on the oxidative stability and predicted shelf life of walnut oil. When the peroxide value was limited to ≤0.25 g/100 g according to national standards for vegetable oils, the predicted shelf life of walnut oil packed in beakers, tinplate cans, glass oil bottles with added TBHQ, and the blank control were 160, 112, 336 and 64 days, respectively. Throughout the predicted shelf life, the acid values of the walnut oil remained within the range required for grade 2 walnut oil. The order of strength of the oxidation stability of walnut oil with TBHQ added by three packaging materials was glass oil bottle>beaker>tinplate can, and it is recommended to store walnut oil in glass oil bottles for long-term storage.

Abstract Objectives The performance of the new Respiratory Pathogen panel (fluorescent probe melting curve, FPMC) for the qualitative detection of 12 organisms (chlamydia pneumoniae, mycoplasma pneumoniae, adenovirus, influenza A virus, influenza B virus, parainfluenza virus, rhinovirus, etc.) was assessed. Methods Prospectively collected nasopharyngeal swab (NPS) and sputum specimens (n = 635) were detected by using the FPMC panel, with the Sanger sequencing method as the comparative method. Results The overall percent concordance between the FPMC analysis method and the Sanger sequencing method was 100% and 99.66% for NPS and sputum specimens, respectively. The FPMC testified an overall positive percent concordance of 100% for both NPS and sputum specimens. The FPMC analysis method also testified an overall negative percent concordance of 100% and 99.38% for NPS and sputum specimens, respectively. Conclusions The FPMC analysis method is a stable and accurate assay for rapid, comprehensive detecting for respiratory pathogens.

Abstract Background Certain occupations may predispose individuals to urolithiasis, a multi-factorial disease. The study aimed to evaluate the prevalence and related factors of nephrolithiasis in medical staff in Qingdao, China. Methods Physical examination results of 5115 in-service medical staff aged 22–60 years old were retrospectively analyzed. Multivariable logistic regression analysis and stratified analyses by age and gender were applied to explore the related factors of nephrolithiasis in these medical staff. Results The overall nephrolithiasis prevalence in medical staff in Qingdao, China was 4.65%. Doctors were more prone to nephrolithiasis than nurses (5.63% vs. 3.96%, P = 0.013) and the peak prevalence (6.69%) was observed in medical staff working in the emergency department (ED). ObjectiveThis research focused on examining the distinctive characteristics of nutrient intake and dietary patterns among long-lived elderly individuals. Additionally， the study was aimed to explore the specific dietary components that may impact the skeletal muscle mass in this particular group.MethodsThis study was conducted in the Chongming area of Shanghai， China. A total of 206 long-lived elderly individuals aged 90 or above were recruited. The 3-day 24-hour dietary recall method was used to collect dietary information and general demographic data through face-to-face interviews with professional nutritionists. The skeletal muscle mass index（SMI） was measured by bioelectrical impedance analysis（BIA）， and low skeletal muscle mass was diagnosed based on the 2019 Asian Working Group for Sarcopenia criteria. Abstract Background Saccharum spontaneum L. is a closely related species of sugarcane and has become an important genetic component of modern sugarcane cultivars. Stem development is one of the important factors for affecting the yield, while the molecular mechanism of stem development remains poorly understanding in S. spontaneum. Phenylalanine ammonia-lyase (PAL) is a vital component of both primary and secondary metabolism, contributing significantly to plant growth, development and stress defense. However, the current knowledge about PAL genes in S. spontaneum is still limited. Thus, identification and characterization of the PAL genes by transcriptome analysis will provide a theoretical basis for further investigation of the function of PAL gene in sugarcane. Results In this study, 42 of PAL genes were identified, including 26 SsPAL genes from S. spontaneum, 8 ShPAL genes from sugarcane cultivar R570, and 8 SbPAL genes from sorghum. Phylogenetic analysis showed that SsPAL genes were divided into three groups, potentially influenced by long-term natural selection. Notably, 20 SsPAL genes were existed on chromosomes 4 and 5, indicating that they are highly conserved in S. spontaneum. This conservation is likely a result of the prevalence of whole-genome replications within this gene family. The upstream sequence of PAL genes were found to contain conserved cis-acting elements such as G-box and SP1, GT1-motif and CAT-box, which collectively regulate the growth and development of S. spontaneum. Furthermore, quantitative reverse transcription polymerase chain reaction (qRT-PCR) analysis showed that SsPAL genes of stem had a significantly upregulated than that of leaves, suggesting that they may promote the stem growth and development, particularly in the + 6 stem (The sixth cane stalk from the top to down) during the growth stage. Conclusions The results of this study revealed the molecular characteristics of SsPAL genes and indicated that they may play a vital role in stem growth and development of S. spontaneum. Altogether, our findings will promote the understanding of the molecular mechanism of S. spontaneum stem development, and also contribute to the sugarcane genetic improving.

Abstract Background Pituitary neuroendocrine tumors (PitNETs) are common gland neoplasms demonstrating distinctive transcription factors. Although the role of immune cells in PitNETs has been widely recognized, the precise immunological environment and its control over tumor cells are poorly understood. Methods The heterogeneity, spatial distribution, and clinical significance of macrophages in PitNETs were analyzed using single-cell RNA sequencing (scRNA-seq), bulk RNA-seq, spatial transcriptomics, immunohistochemistry, and multiplexed quantitative immunofluorescence (QIF). Cell viability, cell apoptosis assays, and in vivo subcutaneous xenograft experiments have confirmed that INHBA-ACVR1B influences the process of tumor cell apoptosis. Results The present study evaluated scRNA-seq data from 23 PitNET samples categorized into 3 primary lineages. The objective was to explore the diversity of tumors and the composition of immune cells across these lineages. Analyzed data from scRNA-seq and 365 bulk RNA sequencing samples conducted in-house revealed the presence of three unique subtypes of tumor immune microenvironment (TIME) in PitNETs. These subtypes were characterized by varying levels of immune infiltration, ranging from low to intermediate to high. In addition, the NR5A1 lineage is primarily associated with the subtype characterized by limited infiltration of immune cells. Tumor-associated macrophages (TAMs) expressing CX3CR1 + , C1Q + , and GPNMB + showed enhanced contact with tumor cells expressing NR5A1 + , TBX19 + , and POU1F1 +, respectively. This emphasizes the distinct interaction axes between TAMs and tumor cells based on their lineage. Moreover, the connection between CX3CR1 + macrophages and tumor cells via INHBA-ACVR1B regulates tumor cell apoptosis. Conclusions In summary, the different subtypes of TIME and the interaction between TAM and tumor cells offer valuable insights into the control of TIME that affects the development of PitNET. These findings can be utilized as prospective targets for therapeutic interventions.

Abstract We aimed to identify the druggable cell-intrinsic vulnerabilities and target-based drug therapies for PitNETs using the high-throughput drug screening (HTS) and genomic sequencing methods. We examined 9 patient-derived PitNET primary cells in HTS. Based on the screening results, the potential target genes were analyzed with genomic sequencing from a total of 180 PitNETs. We identified and verified one of the most potentially effective drugs, which targeted the Histone deacetylases (HDACs) both in in vitro and in vivo PitNET models. Further RNA sequencing revealed underlying molecular mechanisms following treatment with the representative HDACs inhibitor, Panobinostat. The HTS generated a total of 20,736 single-agent dose responses which were enriched among multiple inhibitors for various oncogenic targets, including HDACs, PI3K, mTOR, and proteasome. Among these drugs, HDAC inhibitors (HDACIs) were, on average, the most potent drug class. Further studies using in vitro, in vivo, and isolated PitNET primary cell models validated HDACIs, especially Panobinostat, as a promising therapeutic agent. Transcriptional surveys revealed substantial alterations to the Nrf2 signaling following Panobinostat treatment. Moreover, Nrf2 is highly expressed in PitNETs. The combination of Panobinostat and Nrf2 inhibitor ML385 had a synergistic effect on PitNET suppression. The current study revealed a class of effective anti-PitNET drugs, HDACIs, based on the HTS and genomic sequencing. One of the representative compounds, Panobinostat, may be a potential drug for PitNET treatment via Nrf2-mediated redox modulation. Combination of Panobinostat and ML385 further enhance the effectiveness for PitNET treatment.

Issues at the top of global and national policy agendas, such as the COVID-19 pandemic, climate emergency, and energy and cost of living crises, are illustrating the interconnectivity of the economy, society, and the environment. Moreover, it is clear, that impacts of these issues are not uniformly felt across societies. Greater granularity of information is required to address inequalities. As a result, policy makers are taking a more holistic view of issues to address the interlinkages across domains. This is driving the demand for Statistics Canada to provide statistical insights that address these cross-cutting policy issues and provide more granular information across multiple domains. Linked data are necessary for understanding the interrelated nature of these issues and the scope of their impact. For example, recent Canadian research based on linked data has shown that COVID-19 led to changes in work arrangements that have implications for public transit use and greenhouse gas emissions. The employer-employee linked administrative data augmented with a further linkage to census data provide opportunities for Canadian researchers to produce multidimensional insights on a portrait of racialized groups and immigrants including refugees’ presence in the Canadian economy, their sociodemographic characteristics, and their performance over time relative to other Canadians, particularly post the COVID pandemic. Rapid growth in data availability for research also poses new challenges ranging from technical issues needed for the feasibility of data linkage to stewardship issues related to governance, access, and oversight. The presentation will also discuss the lessons learned and reflections from the Canadian experiences.

ObjectiveTo investigated drug resistance of Cronobacter sakazakii in infant food after biofilms formation， and provided suggestion for drug resistance monitoring.MethodsIn this study， 58 strains of Cronobacter sakazakii， which were isolated from infant formula in Liaoning Province from 2018 to 2021， were tested to determine drug sensitivity， biofilm forming ability， and drug resistance under different biological states with 15 antibiotics.ResultsCronobacterium sakazaki detected in infant food had different degrees of resistance to cefazolin， cefotaxime， compound sulfonamide， chloramphenicol， cefoxitin， naphthalic acid， and ampicillus. In fact， 88% of the tested strains exhibited resistance to polymyxin， with multiple resistant bacteria identified. The resistance of Cronobacterium sakazaki was higher in infant milk powder source than in infant and young children. The film forming rate of the biofilm was 100%， and most medium adhesion strains accounted for 61.02% of the rate. Based on the susceptibility test results of the biofilm state corresponding to the subjects， resistance to cefazolin， cefotaxime， imipenem， tetracycline， and ampicillin increased， while that to SXT disappeared.ConclusionCronobacter sakazakii detected in infant food in Liaoning Province was resistant to cefazolin， cefotaxime， compound sulfonamide， chloramphenicol， cefoxitin， naphdidixic acid， and ampicillin. Notably， 88% of the test strains exhibited intermediate resistance to polymyxin， and multidrug-resistant bacteria were identified. Owing to the resistance to different antibiotics in infant food in Liaoning Province， monitoring the resistance to Cronobacter sakazakii in infant food sources should be strengthened.

A low-noise Low Dropout Regulator (LDO) is designed in a Semiconductor Manufacturing International Corporation (SMIC) $0.18~\mu $ m process, aiming at supply of a clean and constant clamping voltage in the operation of Correlated Double Sampling (CDS) for elimination of switching and reset noise in CMOS Pixel Sensors (CPS) applied to high energy physics experiments. In order to decrease noise of the LDO, a noise model of the LDO consisting of blocks, such as a pass transistor, a resistor network, and an error amplifier, is constructed and noise contributions made by these blocks are analyzed in detail. According to results of the noise analysis, the error amplifier is identified as the major noise source. For the purpose of reduction in noise, especially 1/f noise, an error amplifier exploiting a chopper stabilization technique is proposed. Simulation results demonstrate the effectiveness of the proposed method. The Power Spectral Density (PSD) of the LDO’s output noise is only 60 nV/ $\surd $ Hz at 10 Hz and total integrated noise within the bandwidth is $40~\mu $ V.

Objective To observe the effect of modified Traditional Chinese Medicine external application in the treatment of pruritus in patients with chronic cholestatic liver disease, and to improve the quality of Tradition Chinese Medicine nursing of liver diseases. Methods Totally 112 patients with chronic cholestatic liver disease complicated with pruritus were randomly divided into the control group (n=55) and Traditional Chinese Medicine (TCM)group (n=57). According the occurrence time, frequency and influence of skin temperature on itch, the Traditional Chinese Medicine external application method was modified. Modified Traditional Chinese Medicine external application with calamine lotion and Qingre-Lishi-Zhiyang recipe was carried out in the control group and TCM group, respectively. The effect of nursing interventions was measured and compared between two groups. Results The rate of cured and remarkably improved outcome in the TCM group was 80. 70%(46/57) and 40%(22/55)in the control group, with a significant difference(P ＜0. 01). The levels of total bilirubin (TBIL) and Total bile acid (TBA)in the TCM group were lower than those in the control group after intervention (P＜0. 01). A lower pruritus symptom score and shorter duration of itching were found in the TCM group compared with those in the control group (P＜0. 01). Patients in the TCM group had a longer sleep time compared with that in the control group after nursing intervention (P＞0. 05). The lesion symptom score in the TCM group was lower than that in the control group (P＜0. 01). A better improvement of dry skin was observed in the TCM group compared with that in the control group after intervention (P＜0. 01). Conclusion The modified Traditional Chinese Medicine external application with Qingre-Lishi-Zhiyang recipe is effective in the treatment of pruritus in patients with chronic cholestatic liver disease. Iron (Fe) and methane (CH4) emissions play crucial roles in the carbon cycling of paddy field ecosystems. However, little is known about the effect and mechanism of Fe transformation on the CH4 emission from paddy fields, especially with different water management methods. In this study, the dynamic changes in CH4 emissions and different forms of Fe and soil organic carbon content were observed in paddy fields under flooded irrigation (FI) and water-saving irrigation (WSI) (including intermittent irrigation (II) and controlled irrigation (CI)). In addition, the structural equation model was used to clarify the response relationship between CH4 emissions and Fe ions in paddy fields. The results indicate that the cumulative CH4 emissions under WSI were reduced by 29.03–61.29%. The reasons are as follows (i) WSI increased the oxidation ability of Fe to CH4 by increasing the soil ferric iron (Fe3+) content in paddy fields; (ii) WSI could reduce the substrate utilization by methanogens by transforming the Fe oxide forms to increase the binding capacity of Fe to dissolved organic carbon, which reduced the CH4 production. In addition, compared with II, CI had higher soil Fe3+ content, Fe-bound organic carbon content, and lower acetate content, so it had lower CH4 production capacity. In summary, WSI promotes the direct oxidation of CH4 by Fe3+ in paddy fields and indirectly reduces CH4 production by facilitating the combination of Fe oxides with the substrates. These findings deepen our understanding of the mechanisms of CH4 emission in paddy fields and provide new insights for research on carbon sequestration and emission reduction in paddy field ecosystems.

Objective: Curcumin, a phenolic compound extracted from turmeric rhizomes, exhibits antitumour effects in preclinical models of tumours. However, its mechanism of action in prostate cancer remains unclear. Exploring the molecular mechanisms of curcumin in prostate cancer based on network pharmacology and molecular docking provides a new theoretical basis for prostate cancer treatment. Method: Using tools such as PharmMapper, SuperPred, TargetNet, and SwissTargetPrediction, we obtained information on curcumin-related targets. We comprehensively collected prostate cancer-related targets from several databases, including GeneCards, CTD, DisGeNET, OMIM, and PharmGKB. Cross-cutting drug-disease targets were then derived by screening using the Venny 2.1.0 tool. Subsequently, we used the DAVID platform to perform in-depth GO and KEGG enrichment analyses of the drug-disease-shared targets. To construct a PPI network map of the cross-targets and screen the 10 core targets, we combined the STRING database and Cytoscape 3.7.2. Molecular docking experiments were performed using AutoDockTools 1.5.7 software. Finally, we used several databases such as GEPIA, HPA, cBioPortal, and TIMER to further analyse the screened core targets in detail. Result: We identified 307 key targets of curcumin in cancer treatment. After GO functional enrichment analysis, we obtained 1119 relevant entries, including 782 biological progression (BP) entries, 112 cellular component (CC) entries, and 225 molecular function (MF) entries. In addition, KEGG pathway enrichment analysis revealed 126 signalling pathways, which were mainly involved in the cancer pathway, such as lipid and atherosclerosis pathway, PI3K-Akt signal pathway, MAPK signal pathway, Ras signal pathways, and chemical carcinogenesis-reactive oxygen species. By applying Cytoscape 3.7.2 software, we identified SRC, PIK3R1, STAT3, AKT1, HSP90AA1, ESR1, EGFR, HSP90AB1, MAPK8, and MAPK1 as core targets. Molecular docking experiments showed that the binding energies of curcumin to these core targets were all below −1.85 kJ mol−1, which fully demonstrated that curcumin could spontaneously bind to these core targets. Finally, these results were validated at multiple levels, including mRNA expression, protein expression, and immune infiltration. Conclusion: Through in-depth network pharmacology and molecular docking studies, we have found that curcumin may have anticancer potential by upregulating the expression of PIK3R1 and STAT3, and downregulating the binding ability of molecules such as SRC, AKT1, HSP90AA1, ESR1, EGFR, HSP90AB1, MAPK8, and MAPK1. In addition, curcumin may interfere with the cyclic process of prostate cancer cells by inhibiting key signalling pathways such as the PI3K-Akt signalling pathway, MAPK signalling pathway, and Ras, thereby inhibiting their growth. This study not only reveals the potential molecular mechanism of curcumin in the treatment of prostate cancer but also provides an important theoretical basis for subsequent research.

Abstract Smart city construction is crucial for regional economic development, and the micro-level impact of such construction on business operations cannot be ignored. Based on the panel data of Shanghai and Shenzhen A-share listed companies from 2008 to 2020, this paper investigates the relationship between smart city construction and firm performance and its mechanism of action. The study findings show that (1) smart city construction significantly improves enterprise performance; (2) the improvements are significant across all industries and regions; and (3) the impact of smart city construction on small and medium-sized enterprises (SMEs) and nonstate enterprises is more significant than that on large enterprises and state-owned enterprises. These findings provide a theoretical framework and fresh perspectives for encouraging coordinated regional development and new urbanization construction.

AbstractBackground Rheumatoid arthritis (RA) is a chronic autoimmune disease associated with an increased risk of death, but its underlying mechanisms are not fully understood. Circular RNAs (circRNAs) have recently been implicated in various biological processes. The aim of this study was to investigate the key circRNAs related to RA.Methods A microarray assay was used to identify the differentially expressed circRNAs (DEcircRNAs) in peripheral blood mononuclear cells (PBMCs) from patients with RA compared to patients with osteoarthritis (OA) and healthy controls. Then, quantitative real-time PCR was applied to verify the DEcircRNAs, and correlations between the levels of DEcircRNAs and laboratory indices were analysed. We also performed extensive bioinformatic analyses including Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genome (KEGG) pathway and potential circRNA–miRNA–mRNA network analyses to predict the function of these DEcircRNAs.Results A total of 35,342 and 6146 DEcircRNAs were detected in RA patients compared to controls and OA patients, respectively. Nine out of the DEcircRNAs in RA were validated by real-time PCR. There were correlations between the levels of DEcircRNAs and some of the laboratory indices. GO analyses revealed that these DEcircRNAs in RA were closely related to cellular protein metabolic processes, gene expression, the immune system, cell cycle, posttranslational protein modification and collagen formation. Objective·To investigate the distribution characteristics of vaginal flora in patients with cervical incompetence (CIC), and the effect of cervical cerclage on pregnancy outcome in patients with CIC with abnormal vaginal flora.Methods·A total of 1 261 pregnant women participated in the study. According to the diagnosis, they were divided into cervical incompetence group (CIC group) and normal pregnancy group (NP group). The CIC group patients randomly received cervical cerclage (CIC-C group) or expectation therapy (CIC-E group). Vaginal secretion samples of the patients in the groups were collected and analyzed in the laboratory. Meanwhile, through the results of vaginal flora, the CIC-C group and CIC-E group were further divided into normal vaginal flora groups (CIC-C-N group and CIC-E-N group) and abnormal vaginal flora groups (CIC-C-A group and CIC-E-A group). All groups were processed accordingly and analyzed on pregnancy outcomes. Objective To explore the effect of KCNQ1OT1 gene knockout combined with bruceine D on the proliferation, migration, and invasion of breast cancer MDA-MB-231 cells. Methods Cell Counting Kit-8, wound healing, and Transwell invasion assay were used to detect the effects of bruceine D and siKCNQ1OT1 on the viability, migration, and invasion of MDA-MB-231 cells. Effect of bruceine D and siKCNQ1OT1 on the expression of KCNQ1OT1 in MDA-MB-231 cells was detected by qRT-PCR. Western blot was used to detect the effect of bruceine D and siKCNQ1OT1 on the expression of EMT-related proteins and CDC42, p-MKK7, MKK7 proteins in MDA-MB-231 cells. Results Bruceine D and siKCNQ1OT1 could significantly inhibit the viability, migration, and invasion of MDA-MB-231 cells, and the inhibitory effect was enhanced when they were combined (all P < 0.05); bruceine D downregulated the expression of KCNQ1OT1 in MDA-MB-231 cells (all P < 0.05); bruceine D combined with siKCNQ1OT1 significantly decreased CDC42, p-MKK7, N-cadherin, and Vimentin expression in MDA-MB-231 cells and increased the expression of E-cadherin (all P < 0.05). Conclusion Bruceine D combined with siKCNQ1OT1 significantly inhibit the proliferation, migration, invasion, and EMT of human breast cancer MDA-MB-231 cells, and its molecular mechanism may be related to the blocking of CDC42/MKK7 signaling pathway.

The conventional energy-transition pathway, that is, reducing coal, increasing gas, and developing renewable energies, cannot fully satisfy the requirement of China for energy security under the new situation. Creating a novel dynamic carbon neutrality mode for coal-based energy systems is a forward-looking development approach to solving the problem of high carbon emissions and ensuring national energy security. This study summarizes the international energy transition modes and analyzes the urgency and importance of developing a low-carbon coal-based energy system in response to the challenges of energy security and emissions reduction. Moreover, it clarifies the scientific intension, establishes a system framework, and discloses the security guarantee and emissions reduction mechanisms of the dynamic carbon neutrality mode for coal-based energy systems. An effectiveness assessment model based on system dynamics is established to assess the effectiveness of the mode in terms of energy security, emission reduction,and social development. The results indicate that compared to the conventional coal-fueled systems, the coal-based energy system can potentially reduce carbon emissions by 46% to 55% and external dependence on oil and gas to be below 20% in 2060 under different scenarios; the carbon emissions can be reduced by 84% using the dynamic carbon neutrality mode and the carbon capture and storage technology, and is expected to be further lowered owing to the carbon capture, utilization, and storage technology and carbon sinks in mining areas. Coal-based energy development and application can serve as a strategic technology for oil and gas reserves, thereby ensuring energy security. However, China still faces a significant oil and gas gap before 2030, and thus the development of the coalbased energy is urgent. Furthermore, we propose that a novel coal-based energy system supported by dynamic carbon neutrality technologies should be built to achieve energy independence and security as well as achieve the carbon peaking and carbon neutrality goals. Coal underground gasification, tar-rich coal utilization, and coalbed gas development technologies should be regarded as a potential technology portfolio in the short and medium term and the coal in-situ fluidization mining technology could be a long-term choice.

Abstract We present numerical results for three-dimensional (3D) solitons with symmetries of the semi-vortex (SV) and mixed-mode (MM) types, which can be created in spinor Bose–Einstein condensates of Rydberg atoms under the action of the spin–orbit coupling (SOC). By means of systematic numerical computations, we demonstrate that the interplay of SOC and long-range spherically symmetric Rydberg interactions stabilize the 3D solitons, improving their resistance to collapse. We find how the stability range depends on the strengths of the SOC and Rydberg interactions and the soft-core atomic radius.

Abstract Background Serum exosomes play important roles in intercellular communication and are promising biomarkers of several autoimmune diseases. However, the biological functions and potential clinical importance of long non-coding RNAs (lncRNAs) and mRNAs from serum exosomes in rheumatoid arthritis (RA) have not yet been studied. Methods Serum exosomal lncRNAs and mRNAs were isolated from patients with RA and osteoarthritis (OA) and healthy controls. The differentially expressed lncRNAs (DE-lncRNAs) and mRNA profiles in the serum exosomes of patients with RA were analysed using high-throughput sequencing, and their functions were predicted using Gene Ontologyenrichment, Kyoto Encyclopedia of Genes and Genomes pathway, and gene set enrichment analysis. We constructed a DE-lncRNA-mRNA network and a protein–protein interaction network of differentially expressed mRNAs (DE-mRNAs) in RA using the Cytoscape software. The expression of several candidate a DE-lncRNAs and DE-mRNAs in the serum of patients with RA, patients with OA, and healthy controls was confirmed by qRT-PCR. We assessed the diagnostic ability of DE-lncRNAs and DE-mRNAs in patients with RA using receiver operating characteristic analysis. Furthermore, we analysed the characteristics of immune cell infiltration in RA by digital cytometry using the CIBERSORT algorithm and determined the correlation between immune cells and several DE-lncRNAs or DE-mRNAs in RA. Results The profiles of serum exosomal lncRNAs and mRNAs in patients with RA were different from those in healthy controls and patients with OA. The functions of both DE-lncRNAs and DE-mRNAs in RA are associated with the immune response and cellular metabolic processes. The RT-PCR results show that NONHSAT193357.1, CCL5, and MPIG6B were downregulated in patients with RA. The combination of three DE-mRNAs, CCL5, MPIG6B, and PFKP, had an area under the curve of 0.845 for differentiating RA from OA. Digital cytometry using the CIBERSORT algorithm showed that the neutrophil counts were higher in patients with RA than those in healthy controls and patients with OA. Conclusions These findings help to elucidate the role of serum exosomal lncRNAs and mRNAs in the specific mechanisms underlying RA.

The thermal expansion coefficient of two-dimensional (2D) materials is a very important physical parameter when forming heterostructures. Here, we have established a method that can be employed to evaluate the thermal expansion coefficient of 2D hexagonal materials by the computationally feasible elastic parameters, where the monolayer MoS2 is chosen as an example. Using the first-principles calculations, we calculate the elastic constants under different strains from the strain–stress relationship. Then, based on the calculated elastic parameters, the mechanical and thermal parameters of the monolayer MoS2, including bulk moduli, shear moduli, sound velocities, Debye temperature, heat capacity and thermal expansion coefficient, are obtained. The calculation results are in good agreement with those determined by quasi-harmonic phonon calculations and the experimental results. The monolayer WS2 is employed to further confirm the validity of the methodology. This method can also be widely used to investigate the thermal properties of other 2D materials and thin films.

In recent years, the accuracy and linguistic fluency of translation based on intelligent algorithms have been relatively close to that of human beings, and the application of intelligent algorithms has gradually become a research hotspot in the academic world. In this paper, it is found through relevant research that semantic retention, cultural appropriateness, linguistic fluency, content comprehensiveness and text readability are important factors in the application of intelligent algorithms for the translation of literary works for cross-cultural communication. To explore the degree of influence of these factors on the translation quality of literary works, this paper analyzes them using a multiple linear regression model. The first step in building the mathematical model for multiple linear regression is to construct it. Then, the formulas for calculating the goodness-of-fit and significance test of multiple linear regression equations are introduced. Finally, the regression model was tested using residual analysis and multicollinearity. The regression results show that the quality of literary translation = 0.165+0.124*Semantic retention + 0.356*Cultural appropriateness + 0.254*Linguistic fluency - 0.001*Content comprehensiveness + 0.256*Text accessibility. According to the results of the analysis, the impact of content comprehensiveness on the translation quality of literary works is not significant, and cultural appropriateness has the greatest impact on the translation quality of literary works.

Children are the hope and future of the country, and doing an excellent job for children is of great importance to social development. Social work program efficiency assessment, can help service providers and program implementers understand the effectiveness of program implementation. For this reason, this study uses data envelopment analysis and the Tobit regression model to construct a child protection social work program efficiency assessment model. The input-output indicators of child protection program efficiency are selected. The model is used to analyze the input-output efficiency of social work child protection programs in City H from 2018-2021 as an example. The variables that affect the efficiency of child protection programs are explored in depth to explore the effective ways of child protection. The results show that the comprehensive technical efficiency, pure technical efficiency, and scale efficiency of the child protection program in 2018-2021 are all less than 1 and have not reached DEA efficiency. The comprehensive technical efficiency of the program is significantly affected by the scale efficiency, which necessitates an increase in resource investment in the child protection program. In addition, the six selected influencing factors are significant, at least at the 10% level, all of which positively affect the efficiency of child protection programs and need to be focused on to better improve the efficiency of social work services.

This paper establishes a comprehensive framework for the in-class practical teaching curriculum tailored to majors in Chinese Language and Literature. It comprises four core components: a system of practical teaching content, specified practical teaching requirements, practical training modules, and a target index system for practical teaching outcomes. Through the application of single-factor and two-factor analyses, this study investigates the correlation among course outcomes to refine the scheduling arrangements and the model itself. It introduces a scheduling process designed using a genetic algorithm, which is also employed to allocate instructors and courses within the practical course curriculum, calculating the average teaching hours per instructor. Additionally, the research examines the impact of students’ gender and their varying levels of comprehension regarding professional practice abilities on their practical skills in the field. Specifically focusing on the discipline-related practical activities in Chinese language and literature, the study identifies key courses within the major. Notably, it found a significant correlation coefficient of 0.691 between literary writing and literary appreciation courses. Moreover, the overall average for career development capabilities, based on different levels of professional understanding, is 3.9975, indicating that a deeper comprehension of career development in this major positively influences students’ practical skills in Chinese Language and Literature.

IntroductionThere is currently no gold standard or specific nutritional assessment tool to assess malnutrition in patients with nasopharyngeal carcinoma (NPC). Our study aims to develop a new nutritional assessment tool for NPC patients.Methods and analysisNPC patients will be required to complete a risk factor questionnaire after obtaining their informed consent. The risk factor questionnaire will be used to collect potential risk factors for malnutrition. Univariate and multivariate logistic regression analyses will be used to identify risk factors for malnutrition. A new nutritional assessment tool will be developed based on risk factors. The new tool's performance will be assessed by calibration and discrimination. The bootstrapping will be used for internal validation of the new tool. In addition, external validation will be performed by recruiting NPC patients from another hospital.DiscussionIf the new tool is validated to be effective, it will potentially save medical staff time in assessing malnutrition and improve their work efficiency. Additionally, it may reduce the incidence of malnutrition and its adverse consequences.Strengths and limitations of this studyThe study will comprehensively analyze demographic data, disease status, physical examination, and blood sampling to identify risk factors for malnutrition. Furthermore, the new tool will be systematically evaluated, and validated to determine their effectiveness. However, the restricted geographical range may limit the generalizability of the results to other ethnicities. In the field of higher education in today’s era, cultivating vocal talents is crucial. This paper focuses on an innovative mechanism in vocal talent cultivation, i.e., enhancing vocal education by constructing a music recognition model based on Fourier algorithm. The Model is further applied to design a teaching aid system incorporating reinforcement learning to realize the synergistic development of digital media art and vocal talent cultivation. After rigorous testing, we found that the music recognition model has an accuracy rate of over 90% and is highly resistant to noise. In addition, by analyzing the practices of several colleges and universities’ music schools, it is observed that there are significant differences among different institutions in the four comparative items, with standard deviations exceeding 0.6. In particular, the average scores using this study’s teaching system for vocal talent cultivation are 4.312, 4.335, 4.267, and 4.354, respectively, generally higher than those of the other colleges. The mean values of these four different vocal music majors were all above 4 points. Therefore, the teaching system for vocal talent training proposed in this paper effectively improves the quality of vocal talent training and provides a new path for the practical reform and optimization of said education.

With the rapid development of digital technology, Chinese modern and contemporary literature is facing unprecedented challenges of transformation and innovation, and how to realize the innovation of literature in the context of the digital era has become the key. This study adopts a combination of natural language processing and knowledge mapping to analyze literary works’ narrative vein and character relationships. The research process includes text preprocessing, constructing co-occurrence matrix, LDA topic model analysis, knowledge graph construction and narrative line visualization. The results show that the performance of characters, emotions and plots in modern and contemporary literary works has significant characteristics, and the co-occurrence analysis reveals the close connection between literary works. Through clustering analysis and association rule mining, a significant correlation was found between the style of literary works and narrative content. It is concluded that Chinese modern and contemporary literature shows diverse narrative styles and thematic contents in the digital transformation, an important revelation for literary innovation and development.

Abstract The Parkinson's Progression Marker Initiative (PPMI) aims to identify biomarkers for Parkinson's disease (PD) risk, onset, and progression. This study focuses on the G2019S missense mutation in the LRRK2 gene, which is associated with hereditary and sporadic PD. Utilizing data from the PPMI database, we conducted an analysis of baseline clinical characteristics, as well as serum and cerebrospinal fluid levels in two groups: patients with PD with the G2019S mutation (PD + G2019S) and patients with PD without the mutation (PD‐G2019S). Multiple linear regression and longitudinal analysis were performed, controlling for confounding factors. The establishment of an industrial benefit distribution mechanism for reservoir migrants in water conservancy and hydropower projects is a crucial part. This paper establishes the logistic regression model with gradient descent by deriving the objective function and using the gradient descent method to solve its approximate solution. Then, the model is applied to predict the total income and total expenditure of reservoir migrants before and after agricultural resettlement and tertiary resettlement, and the criteria for benefit compensation distribution are given based on the comparison before and after relocation. In the case of agricultural resettlement, the average share of equal compensation in the first 5 years accounts for 54.67% of the predicted income, and the predicted average annual growth rate of net income per capita is 17.86% with a discount rate of 3.12%. In the tertiary placement case, the predicted growth rates of wage income in the first 5 years are 7.49%, 4.84%, 10.56%, and 1.15%, respectively, and the average annual growth rate of transfer income is 7.01%, and the average annual growth rate of government subsidies is 0.80%. The prediction accuracy of the logistic regression model based on gradient decline reached 84.14%, and the analysis of the distribution of industrial benefits for reservoir migrants was credible. To let the living standard of migrants recover and exceed the level of non-relocation as soon as possible, it is necessary to give migrants certain compensation for production and life recovery based on the living standard measurement index.

IntroductionEKLF/Klf1 is a tissue-restricted transcription factor that plays a critical role in all aspects of erythropoiesis. Of particular note is its tissue-restricted pattern of expression, a property that could prove useful for expression control of a linked marker or enzymatic gene.Methods and resultsWith this in mind, we fused the CRE recombinase to the genomic EKLF coding region and established mouse lines. We find by FACS analyses that CRE expression driven by the EKLF transcription unit recapitulates erythroid-restricted expression with high penetrance in developing embryos. We then used this line to test its properties in the adult, where we found EKLF/CRE is an active and is a robust mimic of normal EKLF expression in the adult bone marrow. EKLF/CRE is also expressed in erythroblastic island macrophage in the fetal liver, and we demonstrate for the first time that, as seen during embryonic development, EKLF is also expressed in adult BM-derived erythroblastic island macrophage. Our data also support lineage studies showing EKLF expression at early stages of hematopoiesis.DiscussionThe EKLF/CRE mouse lines are novel reagents whose availability will be of great utility for future experiments by investigators in the red cell field.

ObjectiveTo provide a basis for the tracing and early warning of foodborne diseases caused by Vibrio parahaemolyticus （VP） in Liaoning Province， the distribution and epidemic trend of VP in Liaoning Province combined with serotype analysis were elucidated.MethodsOne hundred and fifty-one VP isolates from Liaoning Province in 2019 were serotyped， and PFGE， REP-PCR， and ERIC-PCR molecular typing and cluster analysis were performed to evaluate the correlation between the three methods and explore the kinship between the strains.ResultsOne hundred and twenty-four isolates were divided into 18 serotypes and 27 isolates could not be classified. The main serotypes were the O3， O1， and O2 groups. The resolutions （DI） of PFGE， REP-PCR， and ERIC-PCR were 0.97， 0.95， and 0.93， respectively. There was a higher similarity in the molecular type between the serotype O3 group strain and the O1 group strain.ConclusionThe epidemic serogroup of clinical VP isolates in Liaoning Province is still O3：K6， and the epidemic serogroup of food VP isolates is still O2. The cluster analysis results of PFGE， REP-PCR， and ERIC-PCR are consistent， and the resolution and reproducibility of PFGE are better than those of the other two methods. These results can provide novel technical means for the traceability of foodborne diseases caused by VP.

Aiming at the higher correlation between the objective evaluation of computer English speech and the subjective evaluation of experts, an acoustic model based on discriminative training is proposed to improve the confidence score of objective evaluation. First, the process of obtaining the pronunciation quality evaluation score of the speech vector by the forced matching algorithm is introduced, and then the mathematical theory of hypothesis testing is used to prove that the acoustic model trained based on the discriminative algorithm ‘minimum phoneme error’ is more effective than the acoustic model based on the traditional maximum likelihood algorithm. Confidence scores close to subjective assessments are obtained. By calculating the correlation coefficient of the subjective and objective evaluation results, the experiment verifies that the speech evaluation system using the discriminative acoustic model can give a higher confidence score and proposes a data selection method based on dynamic weighting, which is applied to continuous speech recognition in the discriminative training of the acoustic model. This method combines the posterior probability and the phoneme accuracy rate to select the data. First, the Beam algorithm of the posterior probability is used to trim the word graph. On this basis, according to the error rate of the candidate path where the candidate word is located, the probability dynamically assigns different weights to the candidate words; second, by calculating the confusion degree between the phoneme pairs, different penalty weights is dynamically added to the easily confused phoneme pairs to calculate the phoneme accuracy; finally, the expected accuracy of the obtained arc is calculated on the basis of the probability distribution. The Gaussian function is used to softly weight the expected phoneme accuracy of all competing arcs. The experimental results show that compared with the minimum phoneme error criterion, the dynamic weighting method has higher recognition accuracy and can effectively reduce the training time.

To improve nitrogen use efficiency (NUE) during rice cultivation, it is essential to comprehend the morphological and physiological traits of rice roots. However, in high-fertility black soil regions of Northeast China, the effects of combining biochar application with water-saving irrigation (WSI) conditions on rice root development and nitrogen utilization are still unknown. To address this knowledge gap, a combination of field experiments and 15N tracer micro-area investigations was conducted in this study. Four treatments were implemented: (i) controlled irrigation without biochar application (CB0); (ii) controlled irrigation with 2.5 t ha−1 biochar application (CB1); (iii) controlled irrigation with 12.5 t ha−1 biochar application (CB2); and (iv) controlled irrigation with 25 t ha–1 biochar application (CB3). Flooded irrigation conditions without biochar treatment (FB0) were used as the control. The primary objective of this research was to identify the mechanisms by which combined WSI conditions and biochar application affect rice root development and nitrogen utilization. Biochar application enhanced rice root morphological and physiological characteristics. Optimal biochar application increased the longest root length (RL), root volume (RV), root fresh weight (RFW), root active absorption area, root bleeding intensity, and root activity (RA) of rice while also optimizing the root–shoot ratio and facilitating nitrogen absorption by roots. These changes in root morphological and physiological characteristics facilitated the absorption of fertilizer-15N and soil nitrogen by rice roots, ultimately leading to improvements in rice yields and NUEs. Notably, the rice yields, NUE, nitrogen agronomic efficiency (NAE), and nitrogen partial factor productivity (NPFP) of CB2 plants were 16.45%, 39.42%, 24.48%, and 16.45% higher than those of FB0 plants, respectively. These results highlight the effectiveness of biochar application as a strategy to ensure food security and enhance NUE under WSI conditions. Furthermore, this study suggests that the recommended optimal application amount of biochar for the black soil area of Northeast China is 12.5 t ha−1.

With the development of sustainable cities, densely populated higher education institutions increasingly emphasize the sustainability of campuses and their impact on the environment. However, there is a lack of means to quantify emission reduction measures. This study aims to propose an evaluation framework that can quantify energy conservation and emission reduction measures and incentive policies. To this end, this study adopts a mixed methods approach, using questionnaires to assess the effectiveness of management and communication interventions and the impact of incentives on residents’ willingness to participate in emission reduction efforts. The survey results show that although the support for the intervention measures is slightly higher than the average, specific measures such as adjusting dormitory lights-out time and providing sports equipment show superior emission reduction potential. Universities could reduce carbon emissions by about 560 tons per year without incentives and just using interventions. However, when incentives and interventions are combined, the university’s annual emissions reductions are expected to increase to 800 to 1045 tons. Research also highlights the importance of understanding the relationship between occupant behavior, energy consumption, and building carbon emissions. By quantifying the impact of carbon reduction measures and incentives on the daily behaviors of residents, universities can more effectively implement sustainable campus strategies.

Corrosion failure is the bottleneck for restricting the application and development of copper-based metal materials. As a new technology of improving corrosion resistance, superhydrophobic surface provides an effective way to solve the problem of copper corrosion. In this work, the superhydrophobic copper surface was prepared by wire electrical discharge machining (WEDM) and surface modification with stearic acid. Subsequently, scanning electron microscope was used to observe the surface micro morphology, video optical contact angle meter was used to measure the wettability, and the corrosion behavior was further tested through the electrochemical workstation. Results showed that the micron strip nano stalactite graded composite structure on the copper surface had been successfully prepared, and the surface of the composite structure exihibited excellent superhydrophobic properties. In addition, compared with the substrate, the corrosion resistance of the prepared surface was increased by 99.35%. The corrosion prevention mechanism of the superhydrophobic surface was systematically studied and analyzed, and it was found that the formation of micro nano composite structure could effectively capture air to form a solid gas liquid interface on the surface of the sample, and the existence of the air layer further impeded the electron transfer and material transfer rate between the substrate and the electrolyte. Thereby, the electrochemical corrosion rate of the substrate was inhibited, and the corrosion resistance of the superhydrophobic copper sample was significantly improved. Overall, the method was simple, efficient and widely used, and the preparation process was environmentally friendly, which could be applied to large-scale production.

In case that the mixed oil treatment method of offloading from the intermediate station along the line and cutting at the terminal station of pipeline transportation is adopted for the product oil pipeline, the mixed oil offloading from the intermediate station would disturb the development of the mixed oil section along the line, and thus the distribution of mixed oil becomes more complicated. For this reason,a one-dimensional mixed oil prediction model was constructed by analyzing the oil mixing mechanism, introducing the effective diffusion coefficient, and comprehensively considering the law of mass conservation and Fick's laws of diffusion. Meanwhile, ignoring the influence of terrain on oil mixing ignored, the change of mixed oil along the whole line was predicted and calculated under the condition of offloading from the intermediate station and mixed oil backfeed at 0, 40 m3, 60 m3 and 70 m3 in an actual pipeline. On this basis, the influence rule of offloading from the intermediate station on oil mixing along the line was analyzed. The results indicate that offloading the mixed oil from the intermediate station does not affect the concentration distribution of mixed oil. Offloading the mixed oil from the intermediate station reduces the cutting amount of mixed oil in the terminal station of pipeline transportation, but it increases the total amount of mixed oil in the whole line. The change range increases with the increasing of mixed oil offloaded from the intermediate station. Hence, the mixed oil is fed back to reduce the quality of oil in the whole line. Generally, the research results could provide reference basis for the preparation of offloading and cutting scheme of mixed oil in the product oil pipeline for batch transportation.

ObjectiveWe conducted a two-sample Mendelian randomization （MR） study to assess the causal relationship between circulating isoleucine， leucine and valine levels and the risk of peripheral atherosclerosis.MethodsBased on the large-scale genome-wide association study （GWAS） database， single nucleotide polymorphisms （SNPs） closely related to the circulating levels of isoleucine， leucine and valine were identified as instrumental variables （IVs）. Two-sample MR analysis applying the inverse variance weighted （IVW） method and the weighted median estimator （WME） method were performed to estimate the causal relationship between the risk of peripheral atherosclerosis and the exposure with more than three SNPs that were available as IVs. The pleiotropy was evaluated by using the MR-Egger regression and MR-PRESSO method， and the leave-one-out method was used in sensitivity analysis.ResultsFour， one and one SNPs were identified as IVs for circulating isoleucine， leucine and valine levels， respectively. For isoleucine， the IVW model demonstrated there was no evidence of heterogeneity among the IVs （P=0.715）， and there was a significant causal relationship between the increase of circulating isoleucine level and a higher risk of peripheral atherosclerosis risk. Per every 1 elevated standard deviation （SD） of circulating isoleucine level resulted in increasing 31% of peripheral atherosclerosis risk （OR=1.31， 95%CI： 1.07‒1.61）. Similarly， the OR（95%CI） was 1.33 （1.04‒1.71） in the WME model. The MR-Egger regression and MR-PRESSO analysis indicated no evidence of pleiotropy in IVs （all P>0.05）. The result of the leave-one-out sensitivity analysis was stable. The Wald ratio model displayed that the causal relationship between circulating leucine and valine levels and the risk of peripheral atherosclerosis was not statistically significant. The OR （95%CI） for leucine and valine was 1.13 （0.78‒1.63） and 1.11 （0.82‒1.50）， respectively.ConclusionThere is a significant causal relationship between the increase of circulating isoleucine level and a higher peripheral atherosclerosis risk. The causal relationships between circulating leucine and valine levels and the risk of peripheral atherosclerosis need to be further confirmed in future studies.

Abstract Urethral stricture secondary to urethral injury, afflicting both patients and urologists, is initiated by excessive deposition of extracellular matrix in the submucosal and periurethral tissues. Although various anti-fibrotic drugs have been applied to urethral stricture by irrigation or submucosal injection, their clinical feasibility and effectiveness are limited. Here, to target the pathological state of the extracellular matrix, we design a protein-based nanofilm-controlled drug delivery system and assemble it on the catheter. This approach, which integrates excellent anti-biofilm properties with stable and controlled drug delivery for tens of days in one step, ensures optimal efficacy and negligible side effects while preventing biofilm-related infections. In a rabbit model of urethral injury, the anti-fibrotic catheter maintains extracellular matrix homeostasis by reducing fibroblast-derived collagen production and enhancing metalloproteinase 1-induced collagen degradation, resulting in a greater improvement in lumen stenosis than other topical therapies for urethral stricture prevention. Such facilely fabricated biocompatible coating with antibacterial contamination and sustained-drug-release functionality could not only benefit populations at high risk of urethral stricture but also serve as an advanced paradigm for a range of biomedical applications.

Objective: This study aimed to optimize the wet processing of coffee. Methods: The effects of pectinase concentration, temperature and drying method on the contents of flavor precursors and flavor substances in coffee beans were investigated. The quality ranking was obtained by grey correlation analysis and the volatile substances were analyzed. Results: There were significant differences in the contents of protein, crude fat, caffeic acid, caffeine, chlorogenic acid and volatile compounds among different pectinase concentration, temperature, sun drying and heat pump drying. Grey correlation analysis showed that the quality ranking of coffee in the 2% pectinase + 27 ℃+ sun-drying treatment group was the first. The protein content in green coffee beans was 14.04%, the crude fat content was 9.05%, the caffeic acid content in roasted coffee beans was 1.05 mg/g, the caffeine content was 1.41%, and the chlorogenic acid content was 13.89 mg/g. The relative contents of the main volatile substances esters and acids were 40.87 μg/g and 7.62 μg/g, respectively. Conclusion: The comprehensive quality of coffee obtained by 2% pectinase + 27 ℃ + sun-drying was the best.

BackgroundThe management of acne requires the consideration of its severity; however, a universally adopted evaluation system for clinical practice is lacking. Artificial intelligence (AI) evaluation systems hold the promise of enhancing the efficiency and reproducibility of assessments. Artificial intelligence (AI) evaluation systems offer the potential to enhance the efficiency and reproducibility of assessments in this domain. While the identification of skin lesions represents a crucial component of acne evaluation, existing AI systems often overlook lesion identification or fail to integrate it with severity assessment. This study aimed to develop an AI-powered acne grading system and compare its performance with physician image-based scoring.MethodsA total of 1,501 acne patients were included in the study, and standardized pictures were obtained using the VISIA system. The initial evaluation involved 40 stratified sampled frontal photos assessed by seven dermatologists. Subsequently, the three doctors with the highest inter-rater agreement annotated the remaining 1,461 images, which served as the dataset for the development of the AI system. The dataset was randomly divided into two groups: 276 images were allocated for training the acne lesion identification platform, and 1,185 images were used to assess the severity of acne.ResultsThe average precision of our model for skin lesion identification was 0.507 and the average recall was 0.775. The AI severity grading system achieved good agreement with the true label (linear weighted kappa = 0.652). After integrating the lesion identification results into the severity assessment with fixed weights and learnable weights, the kappa rose to 0.737 and 0.696, respectively, and the entire evaluation on a Linux workstation with a Tesla K40m GPU took less than 0.1s per picture.ConclusionThis study developed a system that detects various types of acne lesions and correlates them well with acne severity grading, and the good accuracy and efficiency make this approach potentially an effective clinical decision support tool.

Modern sugarcane cultivars (Saccharum spp., 2n = 100~120) are complex polyploids primarily derived from interspecific hybridization between S. officinarum and S. spontaneum. Nobilization is the theory of utilizing wild germplasm in sugarcane breeding, and is the foundation for utilizing S. spontaneum for stress resistance. However, the exact chromosomal transmission remains elusive due to a lack of chromosome-specific markers. Here, we applied chromosome-specific oligonucleotide (oligo)-based probes for identifying chromosomes 1-10 of the F1 hybrids between S. officinarum and S. spontaneum. Then, S. spontaneum-specific repetitive DNA probes were used to distinguish S. spontaneum in these hybrids. This oligo- fluorescence in situ hybridization (FISH) system proved to be an efficient tool for revealing individual chromosomal inheritance during nobilization. We discovered the complete doubling of S. officinarum-derived chromosomes in most F1 hybrids. Notably, we also found defective S. officinarum-derived chromosome doubling in the F1 hybrid Yacheng75-4191, which exhibited 1.5n transmission for all nonhomologous chromosomes. Altogether, these results highlight the presence of variable chromosome transmission in nobilization between S. officinarum and S. spontaneum, including 1.5n + n and 2n + n. These findings provide robust chromosome markers for in-depth studies into the molecular mechanism underlying chromosome doubling during the nobilization, as well as tracing chromosomal inheritance for sugarcane breeding.

Abstract Despite the development of advanced technologies for interventional coronary reperfusion after myocardial infarction, a substantial number of patients experience high mortality due to myocardial ischemia‐reperfusion (MI/R) injury. An in‐depth understanding of the mechanisms underlying MI/R injury can provide crucial strategies for mitigating myocardial damage and improving patient survival. Here, it is discovered that the 4‐hydroxy‐2‐nonenal (4‐HNE) accumulates during MI/R, accompanied by high rates of myocardial ferroptosis. The loss‐of‐function of aldehyde dehydrogenase 2 (ALDH2), which dissipates 4‐HNE, aggravates myocardial ferroptosis, whereas the activation of ALDH2 mitigates ferroptosis. Mechanistically, 4‐HNE targets glutathione peroxidase 4 (GPX4) for K48‐linked polyubiquitin‐related degradation, which 4‐HNE‐GPX4 axis commits to myocyte ferroptosis and forms a positive feedback circuit. 4‐HNE blocks the interaction between GPX4 and ovarian tumor (OTU) deubiquitinase 5 (OTUD5) by directly carbonylating their cysteine residues at C93 of GPX4 and C247 of OTUD5, identifying OTUD5 as the novel deubiquitinase for GPX4. Consequently, the elevation of OTUD5 deubiquitinates and stabilizes GPX4 to reverse 4‐HNE‐induced ferroptosis and alleviate MI/R injury. The data unravel the mechanism of 4‐HNE in GPX4‐dependent ferroptosis and identify OTUD5 as a novel therapeutic target for the treatment of MI/R injury.

Malignant tumor seriously endangers human life and health. Radiotherapy is currently one of the most common treatments for malignant tumor. However, ionizing radiation (IR) can cause radiation resistance of tumor cells and damage normal tissue cells, thereby limiting the clinical application of radiotherapy. Natural polyphenols not only can improve the radiosensitivity of tumor cells, but also can alleviate the negative effects of IR on the body, thus playing an important role in tumor treatment. Dysregulation of non-coding RNA plays an important role in the occurrence and development of many diseases. In recent years, studies have found that the anti-tumor and radiosensitization effects of polyphenols are associated with non-coding RNA regulation. In this context, this paper reviews the anti-tumor and radiosensitization effects of natural polyphenols and their mechanisms, especially from the perspective of non-coding RNA regulation, in order to provide new ideas for the intervention and treatment of tumors and other diseases, as well as the development of food products for special medical purposes.

Abstract Background Prostate cancer (PCa), the second most prevalent solid tumor among men worldwide, has caused greatly increasing mortality in PCa patients. The effects of lipid metabolism on tumor growth have been explored, but the mechanistic details of the association of lipid metabolism disorders with PCa remain largely elusive. Methods The RNA sequencing data of the GSE45604 and The Cancer Genome Atlas-Prostate Adenocarcinoma (TCGA-PRAD) datasets were extracted from the Gene Expression Omnibus (GEO) and UCSC Xena databases, respectively. The Molecular Signatures Database (MSigDB) was utilized to identify lipid metabolism-related genes. The limma R package was used to identify differentially expressed lipid metabolism-related genes (DE-LMRGs) and differentially expressed microRNAs (DEMs). Moreover, least absolute shrinkage and selection operator (LASSO), extreme gradient boosting (XGBoost), and support vector machine-recursive feature elimination (SVM-RFE) were applied to select signature miRNAs and construct a lipid metabolism-related diagnostic model. The expression levels of selected differentially expressed lipid metabolism-related miRNAs (DE-LMRMs) in PCa and benign prostate hyperplasia (BPH) specimens were verified using quantitative real-time polymerase chain reaction (qRT‒PCR). Furthermore, a transcription factor (TF)-miRNA‒mRNA network was constructed. Eventually, Kaplan‒Meier (KM) curves were plotted to illustrate the associations between signature miRNA-related mRNAs and TFs and overall survival (OS) along with biochemical recurrence-free survival (BCR). Results Forty-seven LMRMs were screened based on the correlation analysis of 29 DE-LMRGs and 56 DEMs, in which 27 LMRMs were stably expressed in the GSE45604 dataset. Subsequently, receiver operating characteristic (ROC) curves and machine learning methods were employed to develop a lipid metabolism-related diagnostic signature, which may be of diagnostic value for PCa patients. qRT‒PCR results showed that all seven key DE-LMRMs were differentially expressed between PCa and BPH tissues. Eventually, a TF-miRNA‒mRNA network was constructed. Conclusions These results suggested that 7 key diagnostic miRNAs were closely related to PCa pathological processes and provided new targets for the diagnosis and treatment of PCa. Moreover, CLIC6 and SCNN1A linked to miR-200c-3p had good prognostic potential and provided valuable insights into the pathogenesis of PCa.

As important supplementary to major edible oils, comparative chemical advantages of minor edible oils decide their development and usage. In this study, chemical composition of 13 kinds of specific edible vegetable oils were investigated. The comparative advantages of chemical compositions of these edible oils were obtained as follows: (1) camellia, tiger nut and almond oil were rich in oleic acid, the contents of which accounted for 79.43%, 69.16% and 66.26%, respectively; (2) safflower oil contained the highest content of linoleic acid (76.69%), followed by grape seed (66.85%) and walnut oil (57.30%); (3) perilla seed, siritch, peony seed and herbaceous peony seed oil were rich in α-linolenic acid (59.61%, 43.74%, 40.83% and 30.84%, respectively); (4) the total phytosterol contents of these oils ranged from 91.46 mg/100 ​g (camellia oil) to 506.46 mg/100 ​g (siritch oil); and (5) The best source of tocopherols was sacha inchi oil (122.74 mg/100 ​g), followed by perilla seed oil (55.89 mg/100 ​g), peony seed oil (53.73 mg/100 ​g) and herbaceous peony seed oil (47.17 mg/100 ​g). The comparative advantages of these specific edible oils indicated that they possess the high potential nutritional values and health care functions.

ObjectiveTo give a solid foundation for the epidemiological data of diarrheagenic Escherichia coli （DEC）， and provide a basis for the rational drug use for foodborne diseases caused by DEC， the continuous monitoring of DEC from 2018 to 2020 in Liaoning Province was carried out. The pathogenic characteristics and drug sensitivity characteristics of DEC were investigated.MethodsA total of 208 infant food in Liaoning Province were collected， from which Enterobacteriaceae was isolated， and 16S rRNA gene sequencing analysis and biochemical identification were carried out. The virulence genes of E.coli were detected. The serotype and drug resistance spectrum of EAEC were identified.ResultsIdentified species of Enterobacteriaceae by 16S rRNA gene sequencing method was consistent with the biochemical results. The detection rate of EAEC in infant food was high， and the detection rate of virulence gene pic of EAEC was high. Virulence genotyping was consistent with the serum typing. According to the virulence gene carried by Escherichia coli， 25 strains of EAEC were detected， of which 40% of the strains （serotype O134：H9） carried virulence gene pic， 28% of the strains （serotype O3：H2） carried virulence gene aggR and astA， 16% of the strains （serotype O9：H6） carried virulence gene aggR， and 16% of the strains （serotype O62：H7） carried virulence gene astA. The carrying rate of EAEC virulence gene in 78 strains was 32.1%. The drug resistance of 25 EAEC strains was not optimistic， and there were multiple drug-resistant strains. The resistance was mainly for β lactams， macrolides， quinolones and tetracyclines.ConclusionEAEC was the main contamination of Escherichia coli in infant food in Liaoning Province， and had high drug resistance， which need more attention to be paid to.
