Abstract Background Paroxysmal atrial fibrillation (PAF) usually recurs after radiofrequency ablation (RFA). This study probed the predictive value of monocyte count to high-density lipoprotein cholesterol (HDL-C) ratio (MHR) with left atrial diameter (LAD) for post-RFA recurrence in PFA patients. Methods Totally 210 RFA-treated PAF patients were selected and assigned into Recurrence and Non-Recurrence groups, with clinical baseline data recorded. Preoperative HDL-C, hypersensitive C-reactive protein, interleukin-6, tumor necrosis factor alpha, brain natriuretic peptide, low-density lipoprotein cholesterol, total cholesterol, and total cholesterol were determined. Monocyte count and MHR were examined. The effects of preoperative MHR and LAD on post-RFA PAF recurrence was analyzed by COX regression analysis. Patients were arranged into high and low MHR and high and low LAD groups. The recurrence of PAF patients with different preoperative MHR and LAD within 12 month was analyzed by Kaplan-Meier (KM) analysis. The predictive value of preoperative MHR, LAD, and their combination for post-RFA recurrence in PAF patients was analyzed. Results MHR and LAD were independent risk factors for post-RFA recurrence in PAF patients. KM curve shift left in the high MHR and LAD groups versus the low MHR and LAD groups. MHR and LAD could help predict post-RFA recurrence in PAF patients, and the combined predictive value of MHR and LAD was greater than that of either factor alone. Conclusion MHR and LAD are independent risk factors for post-RFA PAF recurrence, and can help predict post-RFA recurrence in PAF patients, with their combined predictive value higher than that of each factor individually.

The building blocks-based molecular network (BBMN) strategy was applied to the phytochemical investigation of Cleistocalyx operculatus, leading to the targeted isolation of eighteen novel cinnamoylphloroglucinol-terpene adducts (CPTAs) with diverse skeleton types (cleistoperones A–R, 1–18). Their structures including absolute configurations were determined by extensive spectroscopic methods, quantum chemical calculations, and single-crystal X-ray crystallographic experiments. Cleistoperone A (1), consisting of a cinnamoylphloroglucinol motif and two linear monoterpene moieties, represents an unprecedented macrocyclic CPTA, whose densely functionalized tricyclo[15.3.1.03,8]heneicosane bridge ring skeleton contains an enolizable β,β′-triketone system and two different kinds of stereogenic elements (including five point and three planar chiralities). Cleistoperones B and C (2 and 3) are two new skeletal CPTAs with an unusual coupling pattern between the (nor)monoterpene moiety and the cinnamoyl chain of the cinnamoylphloroglucinol unit. Cleistoperone D (4) possesses an unprecedented cage-like 6/6/6/4/6-fused heteropentacyclic scaffold. The plausible biosynthetic pathways for 1–18 were also proposed. Notably, compounds 1, 4, 7, 8, and 18 exhibited significant antiviral activity against respiratory syncytial virus (RSV). The most potent one, cleistoperone A (1) with IC50 value of 1.71 ± 0.61 μmol/L, could effectively inhibit virus replication via affecting the Akt/mTOR/p70S6K signaling pathway.

Glutathione S-transferases (GSTs) play crucial roles in crop stress tolerance through protection against oxidative damage. In this study, we conducted genome-wide identification and expression analysis of the GST gene family in the autotetraploid potato cultivar Cooperative-88 (C88) using bioinformatic approaches. We identified 366 GST genes in the potato genome, which were classified into 10 subfamilies. Chromosomal mapping revealed that StGSTs were distributed across all 12 chromosomes, with 13 tandem duplication events observed in three subfamilies. Analysis of protein sequences identified 10 conserved motifs, with motif 1 potentially representing the GST domain. Analysis of cis-acting elements in the StGSTs promoter regions suggested their involvement in stress response pathways. RNA-seq analysis revealed that most StGSTs responded to both drought stress and DNA demethylation treatments. Quantitative PCR validation of 16 selected StGSTs identified four members that showed strong responses to both treatments, with distinct expression patterns between drought-tolerant (QS9) and drought-sensitive (ATL) varieties. Transient expression assays in tobacco demonstrated that these four StGSTs enhanced drought tolerance and may be regulated through DNA methylation pathways, though the precise mechanisms require further investigation. These findings provide a theoretical foundation for understanding the response and epigenetic regulation of potato GST genes under drought stress.

ABSTRACT Infection with respiratory syncytial virus (RSV) is a major cause of acute lower respiratory tract disease in young children and older people. Despite intensive efforts over the past few decades, no direct-acting small-molecule agents against RSV are available. Most small-molecule candidates targeting the RSV fusion (F) protein pose a considerable risk of inducing drug-resistant mutations. Here, we explored the in vitro and in vivo virological properties of the K394R variant, a cross-resistant mutant capable of evading multiple RSV fusion inhibitors. Our results demonstrated that the K394R variant is highly fusogenic in vitro and more pathogenic than the parental strain in vivo. The small molecule (2E,2′E)-N,N′-((1R,2S,3S)−3-hydroxycyclohexane-1,2-diyl)bis(3-(2-bromo-4-fluorophenyl) acrylamide) (CL-A3-7), a structurally optimized compound derived from a natural caffeoylquinic acid derivative, substantially reduced in vitro and in vivo infections of both wild-type RSV and the K394R variant. Mechanistically, CL-A3-7 significantly inhibited virus–cell fusion during RSV entry by blocking the interaction between the viral F protein and the cellular insulin-like growth factor 1 receptor (IGF1R). Collectively, these results indicate severe disease risks caused by the K394R variant and reveal a new anti-RSV mechanism to overcome K394R-associated resistance.IMPORTANCERespiratory syncytial virus (RSV) infection is a major public health concern, and many small-molecule candidates targeting the viral fusion (F) protein are associated with a considerable risk of inducing drug-resistant mutations. Abstract Treatment of highly aggressive triple‐negative breast cancer (TNBC) in the clinic is challenging. Here, a liposome nanodrug (LP@PFH@HMME) integrating imaging agents and therapeutic agents for bimodal imaging‐guided sonodynamic therapy (SDT) is developed, which boosted immunogenicity to enable potent immunotherapy via immune checkpoint blockade (ICB) in TNBC. In the acidic tumor microenvironment (TME), LP@PFH@HMME undergoes “nano‐to‐micro” transformation due to a pH‐responsive lipid fusion, which makes droplets much more sensitive to ultrasound (US) in contrast‐enhanced ultrasound (CEUS) and SDT studies. The nanodrug demonstrates robust bimodal imaging ability through fluorine‐19 magnetic resonance imaging (19F MRI) and CEUS bimodal imaging, and it exhibits excellent solubility in aqueous solution with relatively high 19F content and desirable long transverse relaxation time (T2 = 1.072 s), making it suitable for high‐performance 19F MRI, in addition to effective accumulation of nanodrugs after tail vein injection. Thus, 19F MRI/CEUS dual imaging is achievable to show adequate time points for US irradiation of tumor sites to induce highly effective SDT, which produces abundant reactive oxygen species (ROS) triggering immunogenic cell death (ICD) to assist ICB‐based immunotherapy. The combination treatment design of sonodynamic therapy with immunotherapy effectively inhibited TNBC growth and recurrence, highlighting the promise of multifunctional nanodrugs in treating TNBC.

This study aims to address the challenges of apple recognition in complex environments by proposing a solution based on the FC-DETR model. Apple cultivation is an important part of agriculture in Xinjiang, but the increasing shortage of labor has driven the demand for automated harvesting technologies. Therefore, research and development of apple recognition technology in complex environments have become crucial. While CNN architecture models can accurately identify apples in conventional settings, their inherent receptive field limitations prevent them from fully capturing global features in noisy and complex environments, making it difficult to achieve the accuracy and robustness required for practical applications. To solve this problem, this study proposes a real-time object detection model, FC-DETR. This model incorporates the innovative FEMA-BasicBlock residual module, the CAFM cross-scale adaptive feature fusion module, and the novel Inner-WIoU loss function, thereby enhancing feature processing, multi-scale feature selection and integration, and detection accuracy. Experimental results show that the FC-DETR model achieves an apple recognition accuracy of 87 % and a recall rate of 82 % in complex backgrounds while maintaining a lightweight design. This study not only makes significant advances in automated apple harvesting technology but also contributes to improving the efficiency and sustainability of the apple industry in Xinjiang and globally.

Abstract Diazo compounds are rare natural products possessing various biological activities. Kinamycin and lomaiviticin, two diazo natural products featured by the diazobenzofluorene core, exhibit exceptional potency as chemotherapeutic agents. Despite the extensive studies on their biosynthetic gene clusters and the assembly of their polyketide scaffolds, the formation of the characteristic diazo group remains elusive. l-Glutamylhydrazine was recently shown to be the hydrazine donor in kinamycin biosynthesis, however, the mechanism for the installation of the hydrazine group onto the kinamycin scaffold is still unclear. Here we describe an O-methyltransferase-like protein, AlpH, which is responsible for the hydrazine incorporation in kinamycin biosynthesis. AlpH catalyses a unique SAM-independent coupling of l-glutamylhydrazine and polyketide intermediate via a rare Mannich reaction in polyketide biosynthesis. Our discovery expands the catalytic diversity of O-methyltransferase-like enzymes and lays a strong foundation for the discovery and development of novel diazo natural products through genome mining and synthetic biology.

IntroductionThe objective of this study was to analyze antimicrobial resistance (AMR), stress resistance, and virulence factors through whole genome sequencing (WGS) of 192 isolates comprising 164 Salmonella isolates, 8 non-O157 Shiga toxin-producing Escherichia coli (STEC) isolates, and 20 Listeria monocytogenes isolates.MethodsThese isolates were sourced from a national survey conducted between 2010 and 2017, involving 31,322 samples of produce (31 isolates), nuts (43 isolates), and spices (118 isolates).ResultsThe findings yielded several key insights: (1) Within all Salmonella isolates studied, the most prevalent Salmonella serotypes included Give, Kentucky, Senftenberg, Mbandaka, Anatum, Newport, and Weltevreden. (2) All eight non-O157 STEC isolates were found to carry the genes blaEC, acrF, and mdtM, while all 20 L. monocytogenes isolates possessed fosX and lin genes. The Salmonella isolates displayed diverse AMR gene profiles, with 3.65% exhibiting multi-drug resistance. (3) Both Salmonella and non-O157 STEC isolates were discovered to carry stress genes associated with acid resistance, but none of the L. monocytogenes isolates carried an acid resistance gene. Salmonella isolates were found to carry multiple metal-resistance genes. The non-O157 STEC isolates universally exhibited acid resistance genes, and 4 out of the 20 L. monocytogenes isolates were equipped with resistance genes against biocides. (4) All of the STEC isolates (100%) carried stx1 and stx2 genes, while none of them carried eae and wyz genes. Most L. monocytogenes isolates were found to contain 29 virulence genes and 1 pathogenicity island. All Salmonella isolates carried SPI-9, but lacked SPI-7, SPI-10, SPI-11, SPI-12, mig-5, prefA, tviA, and viaB genes.ConclusionThese findings on AMR, stress resistance, and virulence factors among the investigated isolates highlight the potential risks they pose to public health and provide the scientific foundation for the development of preventative and control strategies and guidance pertaining to these major foodborne pathogens.

Soil moisture content plays a vital role in agricultural production, significantly influencing crop growth, development, and yield. Thoroughly understanding the specific soil moisture content in cotton fields is crucial for enhancing agricultural efficiency and driving sustainable agricultural development. This study utilized the gradient-boosting regression–random forest (GBR-RF) algorithm and the GBR and RF algorithms separately, in conjunction with Sentinel-2 satellite images, to estimate cotton soil moisture content, focusing on the B1–B8 bands and in particular the sensitive B6, B7, and B8 bands. The soil data in the jujube orchard of the study area were collected using soil augers at a depth of 30 cm, with soil data collected from a depth of 20 to 30 cm. The findings revealed that the integrated learning algorithm GBR-RF demonstrated high accuracy, with R2, MAE, and MSE results of 0.8838, 1.0121, and 1.6168, respectively. In comparison, the results using just the GBR algorithm yielded R2, MAE, and MSE values of 0.8158, 1.1327, and 1.9645, respectively, while those obtained from the RF algorithm were 0.8415, 1.0680, and 1.8331, respectively. These results indicate that the algorithms exhibited strong generalization, robustness, and accuracy, with GBR-RF outperforming GBR and RF by 8.34% and 5.03%, respectively, in combination with using the B1–B8 bands for inversion. Furthermore, utilizing the full-band data resulted in R2 values that were up to 24.27% higher than those of the individual bands, affirming the efficacy of band combinations for improved accuracy. This study’s demonstration of the positive impact of integrated learning algorithms on estimating cotton soil moisture content underscores the advantages of multi-band data combinations over single-band data, highlighting their ability to enhance accuracy without significantly impacting errors. Importantly, this study’s findings, while not limited to a single experimental field, have broad applicability in cotton precision agriculture, offering valuable insights for research on yield enhancement and agricultural efficiency.

Abstract The crystal phases are essential to the physicochemical properties and functionalities of materials. Copper selenide has emerged as an important and appealing semiconductor, which can exist in a variety of polymorphic phases. However, the richness of polymorphs also makes it a challenge to the direct preparation of copper selenide nanocrystals with tunable phases. Herein, two polymorphs, that is, quasi‐tetragonal Cu2−xSe nanocubes and metastable wurtzite Cu2Se nanodisks, are successfully synthesized by using a single precursor, copper(I) selenocyanate (CuSeCN), as the Cu and Se sources. The key to phase modulation is the optimal choice of the ligand in the synthesis. The as‐prepared nanocrystals possess different morphologies and compositions, giving rise to distinct optical properties and electrical conductivities. Interestingly, the copper selenide nanocrystals can provide a platform for the rational construction of two types of amorphous hollow Au─Cu─Se nanostructures by reaction with Au(I) precursor, in which their final shapes are well kept as that of the original nanocrystal templates. This work provides an easy strategy for the phase‐controlled synthesis of copper selenide nanocrystals and enables the design of new materials for broad applications.

Abstract A defect‐rich 2D p‐n heterojunction, CoxNi3‐x(HITP)2/BNSs‐P (HITP: 2,3,6,7,10,11‐hexaiminotriphenylene), is constructed using a semiconductive metal–organic framework (MOF) and boron nanosheets (BNSs) by in situ solution plasma modification. The heterojunction is an effective catalyst for the electrocatalytic nitrogen reduction reaction (eNRR) under ambient conditions. Interface engineering and plasma‐assisted defects on the p‐n CoxNi3‐x(HITP)2/BNSs‐P heterojunction led to the formation of both Co‐N3 and B…O dual‐active sites. As a result, CoxNi3‐x(HITP)2/BNSs‐P has a high NH3 yield of 128.26 ± 2.27 µg h−1 mgcat.−1 and a Faradaic efficiency of 52.92 ± 1.83% in 0.1 m HCl solution. The catalytic mechanism for the eNRR is also studied by in situ FTIR spectra and DFT calculations. A CoxNi3‐x(HITP)2/BNSs‐P‐based Zn‐N2 battery achieved an unprecedented power output with a peak power density of 5.40 mW cm−2 and an energy density of 240 mA h gzn−1 in 0.1 m HCl. This study establishes an efficient strategy for the rational design, using defect and interfacial engineering, of advanced eNRR catalysts for ammonia synthesis under ambient conditions.

Abstract Rapid and sensitive detection of Salmonella is a critical step in routine food quality control, outbreak investigation, and food recalls. Although various genes have been the targets in the design of rapid molecular detection methods for Salmonella, there is limited information on the diversity of these target genes at the level of DNA sequence and the encoded protein structures. In this study, we investigated the diversity of ten target genes (invA, fimA, phoP, spvC, and agfA; ttrRSBCA operon including 5 genes) commonly used in the detection and identification of Salmonella. To this end, we performed whole genome sequencing of 143 isolates of Salmonella serotypes (Enteritidis, Typhimurium, and Heidelberg) obtained from poultry (eggs and chicken). Phylogenetic analysis showed that Salmonella ser. Typhimurium was more diverse than either Enteritidis or Heidelberg. Forty-five non-synonymous mutations were identified in the target genes from the 143 isolates, with the two most common mutations as T ↔ C (15 times) and A ↔ G (13 times). The gene spvC was primarily present in Salmonella ser. Enteritidis isolates and absent from Heidelberg isolates, whereas ttrR was more conserved (0 non-synonymous mutations) than ttrS, ttrB, ttrC, and ttrA (7, 2, 2, and 7 non-synonymous mutations, respectively). Notably, we found one non-synonymous mutation (fimA-Mut.6) across all Salmonella ser. Enteritidis and Salmonella ser. Heidelberg, C → T (496 nt postion), resulting in the change at AA 166 position, Glutamine (Q) → Stop condon (TAG), suggesting that the fimA gene has questionable sites as a target for detection. Using Phyre2 and SWISS-MODEL software, we predicted the structures of the proteins encoded by some of the target genes, illustrating the positions of these non-synonymous mutations that mainly located on the α-helix and β-sheet which are key elements for maintaining the conformation of proteins. These results will facilitate the development of sensitive molecular detection methods for Salmonella.

Tenderness is a key attribute of meat quality that affects consumers’ willingness to purchase meat. Changes in the physiological environment of skeletal muscles following slaughter can disrupt the balance of redox homeostasis and may lead to cell death. Excessive accumulation of reactive oxygen species (ROS) in the myocytes causes DNA damage and activates poly ADP-ribose polymerase 1 (PARP1), which is involved in different intracellular metabolic pathways and is known to affect muscle tenderness during post-slaughter maturation. There is an urgent requirement to summarize the related research findings. Thus, this paper reviews the current research on the protein structure of PARP1 and its metabolism and activation, outlines the mechanisms underlying the function of PARP1 in regulating muscle tenderness through cysteine protease 3 (Caspase-3), oxidative stress, heat shock proteins (HSPs), and energy metabolism. In addition, we describe the mechanisms of PARP1 in apoptosis and necrosis pathways to provide a theoretical reference for enhancing the mature technology of post-mortem muscle tenderization.

In this study, we aim to investigate the predictive value of serum vascular endothelial growth factor (VEGF) in evaluating treatment efficacy and long-term prognosis for patients with non-surgical esophageal squamous cell carcinoma (ESCC). The patients diagnosed with ESCC by histopathology who didn’t receive surgical treatment were retrospectively analyzed. Through follow-up and prognostic analysis, we explored the value of serum VEGF changes before, during, and after radiotherapy for predicting treatment efficacy, and identified important indicators to construct the predictive model. Eighty-four patients were enrolled in this study, and the objective response rate (ORR) after treatment was 75.0%. The serum VEGF before, during and after radiotherapy were 108.2 ± 38.4, 98.6 ± 20.3 and 96.9 ± 20.0pg/ml, respectively. Sea fog poses a considerable challenge to port operations, impacting maritime safety and efficiency. During the past five years, the average annual downtime of the navigation dispatch department in Ningbo Zhoushan Port due to weather was 800–1000 h, of which approximately 300 h can be attributed to sea fog. This study addresses the issue by developing a comprehensive sea fog monitoring system for Ningbo Zhoushan Port. The system utilizes automatic weather stations (AWS) and visibility laser imaging, detection, and ranging (LIDAR) to assess sea fog severity and improve monitoring accuracy. By increasing monitoring frequency and adopting corresponding warning measures, the system aims to enhance maritime safety and efficiency in Ningbo Zhoushan Port. The results showed that the implemented system successfully determines sea fog severity, enables real-time monitoring, and provides precise visibility assessments. Joint assessments revealed a substantial increase in the annual operating time and revenue of the port. These findings underscore the importance of advanced monitoring techniques in optimizing port operations, reducing collision risks, and mitigating economic losses caused by sea fog.

Abstract Background The implementation of early intervention (EI) in medical settings is time-consuming and resource-intensive, which limits its extensive use. In 2018, the Chinese Eugenics Association developed a home-based, post-discharge EI program. This study aims at evaluating the impact of this EI program on neurodevelopment and physical growth of early preterm infants. Methods This study was a prospective, partially blinded, randomized controlled trial (RCT), followed by an open phase. A total of 73 infants born at 28+ 0 ~ 31+ 6 weeks’ gestation who were admitted to the Children’s Hospital of Chongqing Medical University between December 1, 2019, and June 31, 2020, were enrolled. Another 33 infants were retrospectively recruited as the reference group. Thirty-seven infants randomized in the first early intervention, then standard care (EI-SC) group performed a 30-day EI during RCT period, while 36 infants allocated to SC-EI group were given EI in the following open phase. The test of infant motor performance (TIMP), development quotient (DQ), and anthropometric measures (length, weight, head circumference) were measured at the baseline (T0), termination of the RCT (T1), and termination of the open phase (T2). Repeated measures analysis was performed for comparison among groups. Results From T0 to T1, both groups had significant improvements in all outcome measures (all p 0.05). Whole-genome sequencing (WGS) is a valuable tool in research on foodborne pathogens. In this study, a total of 143 isolates of Salmonella serotypes Enteritidis, Typhimurium, and Heidelberg sourced from eggs and chickens were analyzed for their antimicrobial resistance profiles using WGS data. The isolates carried high rate of genes resistant to aminoglycoside (70.63%), tetracycline (26.57%), fosfomycin (25.17%), sulfonamides (23.78%), and β-lactamases (15.38%); and aadA was the most frequently observed antimicrobial resistance gene (ARG). Antimicrobial resistance varies by Salmonella serotypes, with Salmonella enterica serovar Enteritidis (Salmonella ser. Enteritidis) isolates being highly resistant to aminoglycoside (particularly streptomycin); Salmonella ser. Typhimurium more resistant to aminoglycoside, tetracycline, and sulfonamides; and Salmonella ser. Heidelberg more resistant to aminoglycoside and fosfomycin. Salmonella ser. Typhimurium isolates presented more varieties of ARG than Salmonella ser. Enteritidis and Salmonella ser. Heidelberg. Our data showed that 5 isolates of Salmonella ser. Typhimurium and Salmonella ser. Heidelberg contained ARG resistant to ≥ 5 antimicrobials. In addition, 23 Salmonella isolates carried ARG resistant to 4 antimicrobials.

Abstract Background Nonalcoholic fatty liver disease (NAFLD) is a multisystem disease that affects the liver and a variety of extra-hepatic organ systems. This study aimed to investigate the relationship between hepatic steatosis and glucose metabolism in liver and extra-hepatic tissues and organs. Methods The whole body 18F-fluorodeoxyglucose (FDG) positron emission tomography (PET)/computed tomography (CT) images of 191 asymptomatic tumor screening patients were retrospectively analyzed. Patients with the ratio of spleen/liver CT densities > 1.1 were defined to have NAFLD, and their clinical symptoms, laboratory markers, FDG uptake in a variety of tissues and organs including heart, mediastinal blood pool, liver, spleen, pancreas, and skeletal muscle, as well as abdominal adipose tissue volumes including visceral adipose tissue (VAT) volume and subcutaneous adipose tissue (SAT) volume were compared with those of the non-NAFLD patients and used to analyze the independent correlation factors of NAFLD. Results Among the 191 patients, 33 (17.3%) were NAFLD, and 158 (82.7%) were non-NAFLD. There was no significant correlation between the mean standardized uptake value (SUVmean) and CT density of liver as well as the ratio of spleen/liver CT densities. Hepatic steatosis, but not FDG intake, was more significant in NAFLD patients with abnormal liver function than those with normal liver function. Harmful algal blooms caused by Karenia mikimotoi frequently occur worldwide and severely threaten the marine environment. In this study, the biochemical and physiological responses of K. mikimotoi to the algicidal bacterium Paracoccus homiensis O-4 were investigated, and the effects on the levels of reactive oxygen species (ROS), malondialdehyde content, multiple antioxidant systems and metabolites, photosynthetic pigments, and photosynthetic index were examined. The cell-free supernatant in strain O-4 significantly inhibited K. mikimotoi cell growth. The bacterium caused the K. mikimotoi cells to activate their antioxidant defenses to mitigate ROS, and this effect was accompanied by the upregulation of intracellular antioxidant enzymes and non-enzyme systems. However, the overproduction of ROS induced lipid peroxidation and oxidative damage within K. mikimotoi cells, ultimately leading to algal death. In addition, the photosynthetic efficiency of the algal cells was significantly inhibited by O-4 and was accompanied by a reduction in photosynthetic pigments. This study indicates that O-4 inhibits K. mikimotoi through excessive oxidative stress and impaired photosynthesis. This research into the biochemical and physiological responses of K. mikimotoi to algicidal bacteria provides insights into the prophylaxis and control of harmful algal blooms via interactions between harmful algae and algicidal bacteria.

Abstract Background To systematically review and assess the in vivo effectiveness and safety of probiotics for prophylaxis and treating oral candidiasis. Methods A literature search for studies published in English until August 1, 2018 was conducted in the following databases: PubMed, EMBASE, Cochrane Library, and Web of Science. Randomized controlled clinical trials and experimental mouse animal model studies comparing probiotics (at any dosage and in any form) with control groups (placebo, blank control or other agents) and reporting outcomes of the prophylactic and therapeutic effects were considered for inclusion. A descriptive study and, potentially, a meta-analysis were planned. Results Six randomized controlled clinical trials and 5 controlled experiments of mouse animal models were included in the systematic review. In this study, a deep learning method called Lightning-SN was developed and used for cloud-to-ground (CG) lightning identification. Based on artificial scenarios, this network model selects radar products that exhibit characteristic factors closely related to lightning. Advanced time of arrival and direction lightning positioning data were used as the labeling factors. The Lightning-SN model was constructed based on an encoder–decoder structure with 25 convolutional layers, five pooling layers, five upsampling layers, and a sigmoid activation function layer. Additionally, the maximum pooling index method was adopted in Lightning-SN to avoid characteristic boundary information loss in the pooling process. The gradient harmonizing mechanism was used as the loss function to improve the model performance. The evaluation results showed that the Lightning-SN improved the segmentation accuracy of the CG lightning location compared with the traditional threshold method, according to the 6-minute operating period of the current S-band Doppler radar, exhibiting a better performance in terms of lightning location identification based on high-resolution radar data. The model was applied to the Ningbo area of Zhejiang Province, China. It was applied to the lightning hazard prevention in the hazardous chemical park in Ningbo. The composite reflectivity and radial velocity were the two dominant factors, with a greater influence on the model performance than other factors.

This study is the first to use the observation data of a fog monitor, a visibility meter, and an automatic weather station to carry out a comprehensive observation experiment from the perspective of microphysics on a severe sea fog process in Beilun District, China, from 14 to 15 June 2021. The results show the following: (1) Temperature is closely related to nucleation, condensation growth, and other processes. The decrease (increase) in temperature is the main reason for the enhancement (weakening) of nucleation and the growth of condensation (evaporation of droplets), which leads to an increase (or decrease) in microphysical quantities, such as droplet number concentration and liquid water content. (2) The average droplet number spectral distribution roughly conforms to the Gamma distribution, and the spectral distribution of the fog process presents a ”multi-peak” structure, with peak diameters of 6 μm, 12 μm, 16 μm, 24 μm, and 44 μm. Droplets with a diameter of less than 16 μm account for 75% of the droplet size distribution. (3) During this sea fog process, three microphysical parameters, namely, number concentration, liquid water content, and average diameter, are all positively correlated in pairs, but the positive correlation between the number concentration and the average diameter is weak. This shows that the condensation nucleation and the condensation growth of droplets are the main processes in this sea fog process and that the collision process occurs but is not the dominant process. The sea fog comprehensive observation experiment provides an important demonstration of the microphysics research of sea fog in the eastern coastal areas of China and provides more reference information for sea fog research and equipment comparisons between different regions. At the same time, it also provides an essential scientific basis for the short-term forecast of sea fog in the future and for the optimization of the microphysical parameters of related models.

Fusobacterium nucleatum (Fn) has been considered as a significant contributor in promoting colorectal carcinoma (CRC) development by suppressing host anti-tumor immunity. Recent studies demonstrated that the aggregation of M2 macrophage (Mφ) was involved in CRC progress driven by Fn infection. However, the underlying molecular mechanisms are poorly characterized. Here, we investigated the role of Fn in Mφ polarization as well as its effect on CRC malignancy. Fn infection facilitated differentiation of Mφ into the M2-like Mφ phenotype by in vitro study. Histological observation from Fn-positive CRC tissues confirmed the abundance of tumor-infiltrating M2-like Mφ. Fn-induced M2-like Mφ polarization was weakened once inhibiting a highly expressed damage-associated molecular pattern (DAMP) molecule S100A9 mainly derived from Fn-challenged Mφ and CRC cells. In addition, Fn-challenged M2-like Mφ conferred CRC cells a more malignant phenotype, showing stronger proliferation and migration characteristics in vitro and significantly enhanced tumor growth in vivo, all of which were partially inhibited when S100A9 was lost. Mechanistic studies further demonstrated that activation of TLR4/NF-κB signaling pathway mediated Fn-induced S100A9 expression and subsequent M2-like Mφ activation. Collectively, these findings indicate that elevated S100A9 in Fn-infected CRC microenvironment participates in M2-like Mφ polarization, thereby facilitating CRC malignancy. Furthermore, targeting TLR4/NF-κB/S100A9 cascade may serve as promising immunotherapeutic strategy for Fn-associated CRC.

Oxygen vacancy manipulation and hierarchical morphology construction in oxygen-containing semiconductors have been demonstrated to be effective strategies for developing high efficiency photocatalysts. In most studies of bismuth-based photocatalysts, hierarchical morphology and crystal defects are achieved separately, so the catalysts are not able to benefit from both features. Herein, using boiling ethylene glycol as the treatment solution, we developed an etching-recrystallization method for the fabrication of 3D hierarchical defective BiOCl at ambient pressure. The target hierarchical 3D-BiOCl is composed of self-assembled BiOCl nanosheets, which exhibit a hexagonal prism-like morphology on a micron scale, while simultaneously containing numerous oxygen vacancies within the crystal structure. Consequently, the target catalyst was endowed with a higher specific surface area, greater light harvesting capability, as well as more efficient separation and transfer of photo-excited charges than pristine BiOCl. As a result, 3D-BiOCl presented an impressive photocatalytic activity for the degradation of tetracycline hydrochloride in both visible light and natural white light emitting diode (LED) irradiation. Moreover, an extraordinary recycling property was demonstrated for the target photocatalyst thanks to its hierarchical structure. This study outlines a simple and energy-efficient approach for producing high-performance hierarchically defective BiOCl, which may also open up new possibilities for the morphological and crystal structural defect regulation of other Bi-based photocatalysts.

Abstract Background In diabetes, dysregulated substrate utilization and energy metabolism of myocardium can lead to heart failure. To examine the dynamic changes of myocardium, most of the previous studies conducted dynamic myocardial PET imaging following euglycemic-hyperinsulinemic clamp, which involves complicated procedures. In comparison, the whole-body 18F-FDG PET/CT scan is a simple and widely used method. Therefore, we hope to use this method to observe abnormal myocardial glucose metabolism in diabetes and determine the influencing factors. Methods We retrospectively analyzed PET/CT images of 191 subjects from our medical examination center. The levels of FDG uptake in myocardium were visually divided into 4 grades (Grade 0–3, from low to high). The differences in clinical and metabolic parameters among diabetes mellitus (DM), impaired fasting glucose (IFG), and normal fasting glucose (NFG) groups were analyzed, as well as their associations with myocardial FDG uptake. Results Compared with NFG and IFG groups, DM group had more cardiovascular-related risk factors. The degree of myocardial FDG uptake was significantly decreased in DM group; when myocardial FDG uptake ≤ Grade 1, the sensitivity of DM prediction was 84.0%, and the specificity was 58.4%. Loop-mediated isothermal amplification (LAMP) has gained wide popularity in the detection of Salmonella in foods owing to its simplicity, rapidity, and robustness. This multi-laboratory validation (MLV) study aimed to validate a Salmonella LAMP-based method against the United States Food and Drug Administration (FDA) Bacteriological Analytical Manual (BAM) Chapter 5 Salmonella reference method in a representative animal food matrix (dry dog food). Fourteen independent collaborators from seven laboratories in the United States and Canada participated in the study. Each collaborator received two sets of 24 blind-coded dry dog food samples (eight uninoculated; eight inoculated at a low level, 0.65 MPN/25 g; and eight inoculated at a high level, 3.01 MPN/25 g) and initiated the testing on the same day. The MLV study used an unpaired design where different test portions were analyzed by the LAMP and BAM methods using different preenrichment protocols (buffered peptone water for LAMP and lactose broth for BAM). All LAMP samples were confirmed by culture using the BAM method. BAM samples were also tested by LAMP following lactose broth preenrichment (paired samples). Statistical analysis was carried out by the probability of detection (POD) per AOAC guidelines and by a random intercept logistic regression model. Overall, no significant differences in POD between the Salmonella LAMP and BAM methods were observed with either unpaired or paired samples, indicating the methods were comparable. LAMP testing following preenrichment in buffered peptone water or lactose broth also resulted in insignificant POD differences (P > 0.05). The MLV study strongly supports the utility and applicability of this rapid and reliable LAMP method in routine regulatory screening of Salmonella in animal food.

PURPOSE: The aims of this study were to evaluate the clinical significance and potential prognostic value of pregnancy up-regulated non-ubiquitous calmodulin kinase (PNCK) in clear cell renal cell carcinoma (ccRCC) patients. MATERIALS AND METHODS: The expression of PNCK mRNA was determined in 24 paired samples of ccRCCs and adjacent normal tissues using real-time RT-PCR. The expression of PNCK was determined in 248 samples of ccRCCs and 92 paired samples of adjacent normal tissues by immunohistochemical analysis. Abstract Objectives This study investigated the prevalence of temporomandibular disorders (TMD) in patients with malocclusion through a systematic review and meta-analysis. Materials and methods A comprehensive literature search was conducted up to November 15, 2024. Cross-sectional studies providing data on TMD prevalence among malocclusion patients were included. Study quality was assessed using the Joanna Briggs Institute (JBI) checklist. A random-effects model was used for meta-analysis, with subgroup and meta-regression analyses to explore heterogeneity. Sensitivity analyses were performed to evaluate result robustness. Results Thirty-two studies were included, showing an overall TMD prevalence of 43% (95% CI: 35%-50%; I2 = 97.9%). Prevalence was higher in females (44%) than males (33%) and in adults (42%) than adolescents (39%). Among malocclusion types, Class II (40%) and posterior unilateral crossbite (59%) had the highest TMD prevalence. Sensitivity analysis confirmed the robustness of findings, though diagnostic criteria variations contributed to heterogeneity. Conclusions The prevalence of TMD in malocclusion patients was 43%, with higher rates in females, adults, and specific malocclusion types such as Class II, open bite, overjet, and crossbite. Variations in diagnostic criteria and malocclusion classification contributed to heterogeneity, emphasizing the need to consider individual patient characteristics when assessing TMD risk. Standardized diagnostic criteria, representative sampling, and multilingual search strategies are essential for future research to minimize bias and improve data reliability.

Abstract Background FOLFOX is the recommended chemotherapy regimen for colorectal cancer (CRC), but its response rate remains low. Our previous studies have established a close relationship between gut microbiota and the anti-CRC effect of FOLFOX, though the underlying mechanisms remain unclear. Diet has been confirmed as a key factor influencing gut microbiota, and high-salt diets, representative of western dietary habits, has been shown to affect gut microbiota, immune function, and the risk of developing CRC. However, the impact of high-salt diets on the anti-CRC efficacy of FOLFOX remains unstudied. Therefore, we aimed to investigate the effect and mechanism of high-salt diets on the anti-CRC effect of FOLFOX. Methods We performed 16 S rRNA sequencing and T500 targeted metabolomics analysis on fecal samples from CRC patients and healthy adults. A CRC orthotopic xenograft mouse model was used to study the effect of a high-salt diet on FOLFOX’s anti-CRC efficacy. 16 S rRNA sequencing and non-targeted metabolomics were conducted on mouse fecal samples. Flow cytometry was used to assess immune cell infiltration in tumor and paracancerous tissues. A mouse macrophage conditioned medium system, with tryptophan metabolites, was employed to annotate the functional metabolites, followed by in vivo verification using the orthotopic xenograft mouse model. Results The structure and metabolic profiles of gut microbiota are significantly different between 9 healthy adults and 6 CRC patients. A high-salt diet significantly reduced the efficacy of FOLFOX in mice, with notable changes in gut microbiota and related metabolites. Correlation analysis revealed a significant relationship between gut microbiota, tryptophan metabolites and FOLFOX efficacy. Flow cytometry indicated that a high-salt diet altered macrophage infiltration (CD45+F4/80+) in both the tumor and paracancerous tissues. In vitro experiments confirmed that the tryptophan metabolite SK reduced FOLFOX efficacy, while IPA enhanced it through macrophage-conditioned medium. In vivo, we verified that under a high-salt diet, SK inhibited the efficacy of FOLFOX, while IPA promoted it. Conclusion A high-salt diet reduces the anti-CRC efficacy of FOLFOX through gut bacterial tryptophan metabolism mediated macrophage immunomodulation.

Objective: The aim of this study was to investigate the effect of bovine trypsin on the adhesion and pH of dental plaque biofilms. Methods: A multispecies dental plaque biofilm model and a single-species dental plaque biofilm model were established in vitro. Three groups were tested: (1) blank control group (aseptic ultrapure water); (2) negative control group (1M Tris-HCl buffer, pH = 7.4); and (3) experimental group (bovine trypsin). Adhesion ability was measured using an automatic microplate reader and visualised by confocal laser scanning microscopy (CLSM). The pH was measured using a pH meter. The expression of gtfB, gtfC, and gtfD was analysed using quantitative real-time polymerase chain reaction. Results: Adhesion ability in the experimental group was significantly lower than that in the blank group and the negative control group (P < .05); readhesion ability in the experimental group was inhibited for a certain period of time (24-hour multispecies biofilms were inhibited from 4 to 8 hours, and the 48- and 72-hour multispecies biofilms were inhibited from 2 to 6 hours; P < .05). The decrease in pH was inhibited for a certain period of time (24-hour multispecies biofilms were inhibited from 2 to 8 hours, and the 48- and 72-hour multispecies biofilms were inhibited from 1 to 8 hours; P < .05). Expression levels of gtfB, gtfC, gtfD, and ldh in the experimental group were significantly lower than those in the blank group (P < .05). Conclusions: Bacterial adhesion, and readhesion, decreasd pH, and expression of adhesion- and acid-related genes by Streptococcus mutans in biofilms could be reduced by bovine trypsin for a certain period of time.
