Objective To study the effect of ghrelin gene knockout on the postsynaptic potential of dopaminergic neurons in the substantia nigra of mice. Methods Substantia nigra tissue was taken from 10-week-old ghrelin-/- male mice (ghrelin-/- group) and their male wild-type (WT) littermates (WT group). RNA-seq technique was used to screen differently expressed genes (DEGs). KEGG pathway enrichment analysis was performed for the neuronal synaptic activity-associated signaling pathways which DEGs might be involved in. Quantitative real-time PCR (RT-qPCR) was used for validation of the identified DEGs, and western blotting was used for protein expression of neuronal synaptic activity-related genes. Results Compared with the WT group, the ghrelin-/- group had significant changes in the expression levels of 23 genes in the synaptic signaling pathway of dopaminergic neurons. Specifically, Ionotropic glutamate receptors α-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid type subunit 3 (GluA3) and glycogen synthase kinase3-β (GSK-3β) regulated α-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid receptor and N-methyl-D-aspartic acid receptor on the postsynaptic membrane of neurons, respectively. GluA3 and GSK-3β genes were significantly downregulated in the substantia nigra tissue of mice in the ghrelin-/- group. To establish a rapid detection method for norovirus GII.2 genotype, this study employed reverse transcription recombinase polymerase amplification (RT-RPA) combined with CRISPR/Cas12a and lateral flow strip (RT-RPA-Cas12a-LFS). Here, the genome of norovirus GII.2 genotype was compared to identify highly conserved sequences, facilitating the design of RT-RPA primers and crRNA specific to the conserved regions of norovirus GII.2. Subsequently, the reaction parameters of RT-RPA were optimized and evaluated using agar-gel electrophoresis and LFS. The results indicate that the conserved sequences of norovirus GII.2 were successfully amplified through RT-RPA at 37°C for 25 minutes. Additionally, CRISPR/Cas12a-mediated cleavage detection was achieved through LFS at 37°C within 10 minutes using the amplification products as templates. Including the isothermal amplification reaction time, the total time is 35 minutes. The established RT-RPA-Cas12a-LFS method demonstrated specific detection of norovirus GII.2, yielding negative results for other viral genomes, and exhibited an excellent detection limit of 10 copies/μl. The RT-RPA-Cas12a-LFS method was further compared with qRT-PCR by analyzing 60 food-contaminated samples. The positive conformity rate was 100%, the negative conformity rate was 95.45%, and the overall conformity rate reached 98.33%. This detection method for norovirus GII.2 genotype is cost-effective, highly sensitive, specific, and easy to operate, offering a promising technical solution for field-based detection of the norovirus GII.2 genotype.

Vaccines play essential roles in the fight against the COVID-19 pandemic. The development and assessment of COVID-19 vaccines have generally focused on the induction and boosting of neutralizing antibodies targeting the SARS-CoV-2 spike (S) protein. Due to rapid and continuous variation in the S protein, such vaccines need to be regularly updated to match newly emerged dominant variants. T-cell vaccines that target MHC I- or II-restricted epitopes in both structural and non-structural viral proteins have the potential to induce broadly cross-protective and long-lasting responses. In this work, the entire proteome encoded by SARS-CoV-2 (Wuhan-hu-1) is subjected to immunoinformatics-based prediction of HLA-A*02:01-restricted epitopes. The immunogenicity of the predicted epitopes is evaluated using peripheral blood mononuclear cells from convalescent Wuhan-hu-1-infected patients. Furthermore, predicted epitopes that are conserved across major SARS-CoV-2 lineages and variants are used to construct DNA vaccines expressing multi-epitope polypeptides. Most importantly, two DNA vaccine constructs induce epitope-specific CD8 + T-cell responses in a mouse model of HLA-A*02:01 restriction and protect immunized mice from challenge with Wuhan-hu-1 virus after hACE2 transduction. These data provide candidate T-cell epitopes useful for the development of T-cell vaccines against SARS-CoV-2 and demonstrate a strategy for quick T-cell vaccine candidate development applicable to other emerging pathogens.

ABSTRACTTo explore field microclimate changes under maize/ginger intercropping and select a reasonable field configuration, this study used the major ginger variety ‘Zhugen’ from Southwest China as the experimental material. Two maize plant types; tall (Zhenghong 311) and short (Zhenghong 6), with 2, 3, and 4 m row spacings, shading treatment (ST), and a control (pure) without shading (CK) were used for the study. The effects of the maize plant type and row spacing on soil temperature, humidity, canopy temperature, light transmittance, plant growth, and intercropped ginger yield were investigated. Soil moisture, ginger canopy humidity, and shading rate in the maize/ginger intercropping mode were significantly higher than those in the non-shading treatment. Conversely, the soil and ginger canopy temperatures of the maize/ginger intercropping mode were significantly lower than those of the non-shading treatment. As a result, plant height, stem diameter, branch number, and ginger yield were significantly higher under the intercropping mode than the non-shading treatment. Although maize/ginger intercropping improves ginger growth and yield, these parameters are dependent on maize plant type and row spacing. The ginger yield was significantly higher when intercropped with the tall maize ZH 311 than with short maize ZH 6. The yield improved with decreasing row spacing and was the highest at a row spacing of 2 m. At the aforementioned field configurations, the soil and canopy humidity were the highest whereas the soil and canopy temperatures were the lowest, thereby giving best results on parameters like plant height, stem diameter, branch number, and yield.

Forkhead box P3 (FOXP3) plays a critical role in the pathogenesis of autoimmune disorders. In the present study, we genotyped three single-nucleotide polymorphisms, namely, rs2232365, rs3761548, and rs3761549, to determine the relationship between FOXP3 polymorphisms and neuromyelitis optica spectrum disorder (NMOSD) susceptibility among the Northern Chinese Han population.

The variation rule of deformation structure and magnetic properties of high strength non-oriented electrical steel with cold rolling were studied by field emission scanning electron microscopy(SEM+EBSD), rapid temperature tube furnace and AC magnetic properties measuring instrument. The results show that the rough strips with a large number of substructures and smooth strips with a small amount of deformation are formed in cold-rolled high strength non-oriented electrical steel. The rough strip has a γ orientation, mainly {111}〈110〉, and the smooth strip has a {112}〈110〉 and {001}〈110〉 orientation. In the rough strips, there are a lot of shear bands with width of about 2-3 μm, which show an angle of 20°-35°to the rolling direction. And the quantity increases gradually with the increase of the reduction rate. There is a certain misorientation between the shear band and the matrix. With the increase of the reduction rate, the shear band gradually changes from {111}〈110〉 to {223}〈110〉, and the misorientation between the shear band and the matrix gradually increases. After annealing, the texture of Goss and {111} increases, while the texture of {001} decreases. The rolling direction magnetic induction intensity increases, the transverse magnetic induction intensity decreases, the magnetic anisotropy is significant, and the iron loss decreases.

The aim of this study is designed an improved ResNet 50 network to achieve automatic classification model for pain expressions by elderly patients with hip fractures. This study built a dataset by combining the advantages of deep learning in image recognition, using a hybrid of the Multi-Task Cascaded Convolutional Neural Networks (MTCNN). Based on ResNet50 network framework utilized transfer learning to implement model function. This study performed the hyperparameters by Bayesian optimization in the learning process. This study calculated intraclass correlation between visual analog scale scores provided by clinicians independently and those provided by pain expression evaluation assistant(PEEA). The automatic pain expression recognition model in elderly patients with hip fractures, which constructed using the algorithm. The accuracy achieved 99.6% on the training set, 98.7% on the validation set, and 98.2% on the test set. The substantial kappa coefficient of 0.683 confirmed the efficacy of PEEA in clinic. This study demonstrates that the improved ResNet50 network can be used to construct an automatic pain expression recognition model for elderly patients with hip fractures, which has higher accuracy.

Pathogens detected by metagenomic next-generation sequencing (mNGS) and the laboratory blood culture flask method were compared to understand the advantages and clinical significance of mNGS assays in the etiological diagnosis of peritoneal dialysis-associated peritonitis (PDAP). The study involved a total of 37 patients from the hospital’s peritoneal dialysis centre, six of whom were patients with non-peritoneal dialysis-associated peritonitis. Peritoneal dialysis samples were collected from the 37 patients, who were divided into two groups. One group’s samples were cultured using conventional blood culture flasks, and the other samples underwent pathogen testing using mNGS. The results showed that the positive rate of mNGS was 96.77%, while that of the blood culture flask method was 70.97% (p < 0.05). A total of 29 pathogens were detected by mNGS, namely 24 bacteria, one fungus, and four viruses. A total of 10 pathogens were detected using the bacterial blood culture method, namely nine bacteria and one fungus. The final judgment of the PDAP’s causative pathogenic microorganism was made by combining the clinical condition, response to therapy, and the whole-genome sequencing findings. For mNGS, the sensitivity was 96.77%, the specificity was 83.33%, the positive predictive value was 96.77%, and the negative predictive value was 83.33%. For the blood culture flask method, the sensitivity was 70.97%, the specificity was 100%, the positive predictive value was 100%, and the negative predictive value was 0%. In conclusion, mNGS had a shorter detection time for diagnosing peritoneal dialysis-related peritonitis pathogens, with a higher positive rate than traditional bacterial cultures, providing significant advantages in diagnosing rare pathogens.

Abstract Disseminated Cryptococcosis infection typically occurs in immunocompromised patients, often manifested as pneumonia or meningoencephalitis. Cases with involvement of either prostate or adrenal glands are less frequent. We describe a case of an immunocompromised 62-year-old man with new-found Idiopathic CD4 + T lymphocytopenia who presented with urinary irritation symptoms followed by headache. The patient was finally diagnosed as disseminated cryptococcosis of prostate, adrenal gland involvement with the help of combining histopathology of formalin-fixed, paraffin-embedded tissue with metagenomic next-generation sequencing technique to identify C neoformans sensu stricto in prostate, adrenal gland tissues. Clinicians should be aware of atypical presentations of cryptococcal disease. In this case of cryptococcosis in immunocompromised patients, we find that cryptococcosis can affect varied organs simultaneously and should be considered in the differential of infectious diseases. And mNGS technology helps to confirm the diagnosis.

The Maillard reaction during the production of brown sugar is critical to the color, flavor and functional characteristics of brown sugar, but it also produces substances potentially harmful to human health such as glyoxal (GO), methylglyoxal (MGO), 3-deoxyglucosone (3-DG), 5-hydroxymethylfurfural (5-HMF), and acrylamide. This paper introduces the Maillard reaction at the key stages of the brown sugar production process, its influential factors, its possible harmful products and their effects on human health, and it puts forward some suggestions for controlling the production of harmful products from Maillard reaction during brown sugar production in order to provide a theoretical reference for improving the safety of brown sugar for consumption.

Objective To explore the construction of a nomogram model with good predictive value for community acquired pneumonia (CAP) in elderly patients using commonly used blood routine indicators in clinical practice. Methods A total of 88 elderly CAP patients and 80 healthy controls in Nanjing Integrated Traditional Chinese and Western Medicine Hospital from January to December 2022 were enrolled retrospectively. Univariate and multivariate logistic regression was used to screen the independent risk factors of CAP in the elderly, and nomogram model was constructed. ROC curve was used to evaluate the diagnostic efficiency of nomogram model. Results White blood cell count, red blood cell count, lymphocyte ratio, monocyte ratio, mean platelet volume and platelet distribution width were independent risk factors for CAP in the elderly. Based on this, nomogram model was constructed, and the area under the ROC curve was 0.869 (95%CI: 0.808-0.916). Conclusion The nomogram model constructed based on the clinical blood routine indicators in this study can accurately screen the elderly patients with CAP, assess the risk of CAP for elderly patients, and guide the clinical precise management.

Myasthenia gravis is a rare autoimmune disease, mainly manifested as muscle weakness and fatigue. Objective To compare the preparation efficiency of mouse pox and mouse hepatitis antibodies between two substrains of BALB/c and Big-BALB/c (B-BALB/c) mice, and to provide a theoretical basis and reference for the selection of laboratory animals in the preparation of monoclonal antibodies induced in vivo through hybridoma. Methods Individuals weighing more than 5% of the weight of normal animals at 4 weeks of age (the criterion for late selection is more than 10%) were selected from a population of conventionally bred BALB/c mice and bred individually, and a subline of B-BALB/c mice was prepared after 10 generations of selection. A total of 40 BALB/c mice and 40 B-BALB/c mice aged 10 to 11 weeks, half male and half female, were selected and inoculated with the mousepox monoclonal antibody hybridoma cell line G23 or the murine hepatitis monoclonal antibody hybridoma cell line Y15 pre-treated with liquid paraffin, respectively. Mice ascites containing monoclonal antibodies were obtained by in vivo induction. The antibody titer was tested by indirect ELISA. The mice were grouped based on the sub-strains, gender and inoculation type of hybridoma to analyze the ascites production, antibody titer and antibody production, and to evaluate the antibody preparation efficiency of the two BALB/c mouse sub-strains. Results After 10 generations of breeding, the body weight of 10-week-old male and female B-BALB/c mice increased by 22.3% and 12.8%, respectively, compared with BALB/c mice of the same age. Compared with BALB/c mice, B-BALB/c mice had better tolerance and adaptation to secondary ascites collection. Compared with BALB/c mice, the ascites production and antibody titer during the preparation of antibodies in B-BALB/c mice were significantly increased, especially in the hybridoma cell line G23 vaccination group (both P＜0.000 1) . After inoculation with the hybridoma cell lines G23 or Y15, the average antibody production of B-BALB/c mice (14.99×104 U and 33.22×104 U) was higher than that of BALB/c mice (5.33×104 U and 19.31×104 U) (both P＜0.01). After inoculation with hybridoma cell line G23, the average antibody production per unit body weight of B-BALB/c mice (0.55×104 U/g) was higher than that of BALB/c mice (0.23×104 U/g) (P＜0.000 1). And the antibody production per unit body weight of female B-BALB/c or BALB/c mice was higher than that of male B-BALB/c or BALB/c mice (both P＜0.01). Conclusion B-BALB/c mice can be used as an alternative to BALB/c mice in the in vivo induction of monoclonal antibody preparation, which can achieve the purpose of reducing the number of experimental animals used, lowering the labor cost, and improving the efficiency of antibody preparation.

Teaching quality evaluation is a process of evaluating the teaching quality of architectural majors. It can not only evaluate the teaching level of teachers, but also evaluate the learning effectiveness of students. Therefore, this study designs a teaching quality evaluation system for architecture majors based on fuzzy environment, in order to provide direction guidance for effectively evaluating the teaching quality of architecture majors by using this research. The teaching quality evaluation of higher vocational architecture majors based on enterprise platform is a multiple-attribute group decision-making (MAGDM). The spherical fuzzy sets (SFSs) provide more free space for decision makers to portray uncertain information during the teaching quality evaluation of higher vocational architecture majors based on enterprise platform. Therefore, this study expands the partitioned Maclaurin symmetric mean operator and induced ordered weighted average operator to SFSs based on the power average technique and construct induced spherical fuzzy power partitioned MSM (I-SFPPMSM) technique. Subsequently, a novel MAGDM method is put forward based on I-SFPPMSM technique and spherical fuzzy number weighted geometric technique under SFSs. Finally, a numerical example for teaching quality evaluation of higher vocational architecture majors based on enterprise platform is employed to verify the put forward method, and comparative analysis with some existing techniques to testy the validity and superiority of the I-SFPPMSM technique.

Abstract Background Early-onset pharyngeal airway collapse (PAC) in infants, which presents with onset within 6-months old is relatively rare. This disease has not been given enough attention in clinic. The aim of this study was to explore the clinical features, endoscopic findings and outcomes of early-onset PAC in infants. Methods The children of PAC with onset within 6-months old were included. A retrospective study was conducted. Results (1) Total 26 cases were included. The age of onset was neonatal period in 20 cases, 1 to 3-months old in 5 cases, and 4 to 6-months old in 1 case. (2) The main clinical manifestations were noisy breathing (26/26), suprasternal retraction (18/26), snoring (14/26) and hypoxic episode (13/26). (3) Based on the endoscopic findings, collapse at the retropalatal level was most common (24/26). (4) Twelve cases underwent pharyngolaryngeal CT examination, which revealed abnormal findings in 7 cases. (5) Fifteen cases were accompanied with the other airway malformations. (6) In the group with comorbidities of cerebral impairment or craniofacial abnormalities, 1 case was lost to follow up, 4 cases died, and 10 cases survived, in which 9 cases had neurodevelopmental disorders. In the group without comorbidities, 2 cases were lost to follow up, 9 cases survived, in which 1 case had neurodevelopmental disorders. Objective To investigate the association between retinal nerve fiber layer (RNFL) thickness and cognitive dysfunction in patients with mild ischemic stroke. Methods Total 146 patients with mild ischemic stroke that diagnosed and treated in Chengde Central Hospital in Hebei from January 2020 to December 2021 were included. Optical coherence tomography (OCT) was used to measure the RNFL thickness in each quadrant of both eyes, and Mini⁃Mental State Examination (MMSE) and Montreal Cognitive Assessment (MoCA) were used to evaluate cognitive function. Results In patients with mild ischemic stroke, the RNFL thickness at the superior side of left eye was greater than right eye (t = ⁃ 4.589, P = 0.000), and the RNFL thickness at the temporal side of right eye was greater than left eye (t = 3.639, P = 0.000). Correlation analysis showed that the correlation between the RNFL thickness at the superior side of the left eye and the MMSE score (r = 0.385, P = 0.000), the RNFL thickness at the temporal side of the left eye and the National Institutes of Health Stroke Scale (NIHSS) score at admission (r = 0.170, P = 0.020) were positively correlated. The correlation between the RNFL thickness at the inferior side of the left eye and the history of drinking (r = ⁃ 0.216, P = 0.011), the RNFL thickness at the temporal side of the right eye and the history of hypertension (r = ⁃ 0.194, P = 0.023) were negativley correlated. Conclusions The thinning of RNFL in patients with mild ischemic stroke may have a certain degree of association with cognitive dysfunction.

The magnitude is an important parameter that characterizes the size of earthquakes. However, in paleoearthquake studies, it is difficult to precisely determine the rupture parameters closely related to seismic moment, making it challenging to directly calculate the magnitude of events. Researchers often assume that event sequences consist of characteristic earthquakes with similar magnitudes or use empirical relationships based on known magnitudes of historical earthquakes to estimate magnitudes. However, previous studies have shown that the assumption of characteristic earthquakes is overly simplistic, and magnitude estimation based on empirical relationships is limited by various errors. Therefore, there is a pressing need to explore new methods to improve the reliability of magnitude assessments for ancient earthquake events. In recent years, the successful application of three-dimensional combination trenches has demonstrated that these trenches contain rich deformation information about events, confirming the feasibility of assessing event sizes within trenches. Using the example of the Copper Mine trench on the Altyn Tagh fault, this article utilizes the deformation intensity revealed within the trench, including vertical displacement, deformation zone width, and total tensile strain, to estimate the scale of the event sequence. Data analysis results indicate that the deformation intensity parameters have a certain positive correlation with the relative magnitude, and there is also some correlation among these parameters. Therefore, the information on deformation intensity within the trench can be used to assess the relative magnitude of events, and fully exploring the deformation information within trenches can provide valuable insights and references for the reasonable evaluation of the magnitude of paleoearthquake events. This underscores the importance of considering such information in paleoearthquake research.

Objective： This study aimed to explore the fatigue relieving effect of Lonicera caerulea Berry polyphenol, hawthorn polyphenol microcapsules, lactoferrin and vitamin C as polyphenol compound formula to alleviate fatigue. Methods： The commercial sports nutrition food with the compound formula of creatine, β-hydroxy-β-methylbutyrate, fructose diphosphate, taurine, vitamin B6 and watermelon powder was used as the positive control. The experiment of exhaustive swimming in mice was carried out. The fatigue related indexes were determined by the kit, and the microscopic pathological tissues of the organs were observed by H&E staining. Results： There was no significant difference in body weight of mice (P＞0.05). Compared with the normal group, no significant difference in organ index in the dietary supplement group (P＞0.05) was found. Polyphenol compound formula significantly prolonged the swimming time of mice at room temperature and low temperature by 106.01% and 140.94%, respectively (P＜0.01). Moreover, oxidative stress, metabolite accumulation and energy metabolism, and down-regulated secretion of inflammatory factors (P＜0.01 or P＜0.05) were also improved. It promoted the cell nucleus increase in muscle, inflammatory cell infiltration in the liver, and redness and shedding of tubular epithelial cells in the kidney. Conclusion： Polyphenol compound formula can relieve exercise fatigue at low temperature and room temperature.

In this paper, after the extraction of teaching resource features by mutual information method, the classification of Civics teaching resources is realized by constructing a decision tree classifier. According to the current situation of Civics education in university sports courses, the set segmentation correlation function is determined, the contribution of each factor is combined using the correlation matrix, the correlation coefficient weights are calculated, and the design of the Civics multimodal corpus is accomplished based on big data technology. Nine universities in Guangzhou City were selected as the objects of this study, and the research data were obtained through questionnaire surveys of physical education teachers in these nine schools. SPSS26.0 and Origin 2019 software were used to study the Civics and Politics of Physical Education Courses in Colleges and Universities. The results show that the convergence speed of the objective function of the algorithm in this paper and the classification accuracy has been improved substantially, compared with other algorithms, by about 0.3 to 0.7, i.e., it shows that the algorithm in this paper has a good performance of classification of the course Sijian. The scores of national sentiment and sports core literacy are 0.76 and 0.76, respectively, with a rank of medium (less than 0.79), and the score of physical and mental health is 0.82, with a rank of high (greater than or equal to 0.8). That is, the public physical education (volleyball) class has a relatively satisfactory effect on the cultivation of students’ physical and mental health, while there is still room for progress in the cultivation effect of family and national sentiment and sports core literacy. This study supplements, to a certain extent, the lack of an evaluation index system for the effect of sports courses’ ideology and nurturing and promotes the better development of public sports courses’ ideology.

Objective To investigate the checklist –based nursing in management of pediatric intravenous infusion via scalp vein set. Methods Totally 200 pediatric patients who visited the clinic between March 2022 to June 2022 were selected as the control group, another 200 pediatric patients who visited the clinic between July 2022 to October 2022 were selected as the observation group. All pediatric patients received routine nursing during intravenous infusion treatment, and those in the observation group were given checklist –based nursing interventions. The nursing defect events and intravenous infusion-related complications were observed and analyzed, and parents’ satisfaction with nursing was measured. Results The incidences of nursing defect events such as repeated puncture, omission of infusion fluid, improper infusion speed and delayed infusion fluid change in the observation group were lower than those in the control group (P＜0. 05). The incidences of complications such as leakage, catheter-related infection and phlebitis in the observation group were lower than those in the control group (P＜0. 05). The parents’ satisfaction rate in the observation group was higher than that in the control group. Conclusion The checklist –based nursing is potentially effective to reduce the adverse influence on pediatric patients with intravenous infusion via scalp vein set, and improve the quality of pediatric nursing. The high integration of the civil aircraft avionics systems will lead to an exponential rise in the size of airborne safety-critical software, and cause the numerous and inconsistent sources of its requirements, the transfer of requirements at each level of the avionics system software has duality and other problems. Therefore, how to ensure the consistency of the avionics software has become one of the core issues to be solved during the development of the system. On the basis of the syntax of Safety_SysML state machine, the Safety_SysML consistency verifier is designed, including static data detection and dynamic data detection. The test cases are designed for unit and integration test of the core algorithm and system. Based on the error inference and boundaries, the functional tests are designed and executed to find the defects in the verifier. The results show that the Safety_SysML consistency verifier can effectively identify the problem of duality in avionics system software, and is of significant importance for improving the reliability of the avionics software.

This paper considers the supply chain composed of altruistic retailers and selfish manufacturers under risk aversion. We use the mean variance (MV) method to construct two types of behavior models. One is a two-stage supply chain model with a single manufacturer and a single retailer, and the other is a competitive supply chain model with two retailers and two manufacturers. We discuss the decision-making problems under manufacturer Stackelberg (MS) game and retailer Stackelberg (RS) game, respectively. We analyze the role of risk aversion and power structure. Results show that the more risk aversion manufacturers are, the lower the emission reduction levels are. It also find that the prices increase with power shift from retailers to manufacturers. Finally, we point out that the competing can help the firms earn more benefits via numerical studies.

ObjectiveThis study was designed to investigate cadmium （Cd） pollution levels in wheat and wheat flour in different cities of Shandong Province， as well as conduct preliminary health risk assessment of Cd intake via wheat flour.Methods1 789 samples of wheat or wheat flour were collected from farmers’ markets， supermarkets， and farmers in 16 cities of Shandong Province. Inductively coupled plasma-mass spectrometry （ICP-MS） was performed to determine the Cd content in wheat. Previous studies suggest that the senescence and death of the replaceable bud of the Chinese chestnut cultivar (cv.) “Tima Zhenzhu” involves programmed cell death (PCD). However, the molecular network regulating replaceable bud PCD is poorly characterized. Here, we performed transcriptomic profiling on the chestnut cv. “Tima Zhenzhu” replaceable bud before (S20), during (S25), and after (S30) PCD to unravel the molecular mechanism underlying the PCD process. A total of 5,779, 9,867, and 2,674 differentially expressed genes (DEGs) were discovered upon comparison of S20 vs S25, S20 vs S30, and S25 vs S30, respectively. Approximately 6,137 DEGs common to at least two comparisons were selected for gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses to interrogate the main corresponding biological functions and pathways. GO analysis showed that these common DEGs could be divided into three functional categories, including 15 cellular components, 14 molecular functions, and 19 biological processes. KEGG analysis found that “plant hormone signal transduction” included 93 DEGs. Overall, 441 DEGs were identified as related to the process of PCD. Most of these were found to be genes associated with ethylene signaling, as well as the initiation and execution of various PCD processes.

Ginkgolic acid is a natural plant antifungal substance with important biological activities, present in the testa of Ginkgo biloba. This research aims to analyse the antifungal action of ginkgolic acid against pathogenic fungi. Ginkgolic acid was analyzed by HPLC and FTIR, and the in vitro inhibitory activities of ginkgolic acid against pathogenic fungi has been investigated. Results showed that ginkgolic acid was a small hydrophobic molecule composed of five ginkgolic acids and their derivatives, with the alkyl and phenolic hydroxyl groups being located on the skeleton of the aromatic ring. Among Nigrospora oryzae (N. oryzae), Alternaria alternata (A. alternata) and Penicillium expansum (P. expansum), ginkgolic acid showed the strongest inhibition against N. oryzae, and the inhibition rate of ginkgolic acid at 5 mg · mL-1 reached 65.82%. Ginkgolic acid lysed the cell membrane of N. oryzae, causing the leaking of the mycelial protein and enhancement of the permeability of the cell membrane, which caused the electrical conductivity of the liquid medium to increase. In addition, it also decreased the activities of superoxide dismutase and catalase in the mycelium of N. oryzae, thereby weakening the cell’s protective function by damaging the integrity of the biomembrane. Furthermore, ginkgolic acid can effectively protect the leaves of Epipremnum aureum from infection by N. oryzae. It was concluded that ginkgolic acid effectively inhibited the activity of N. oryzae.

Brain-computer interface (BCI) technology based on motor imagery electroencephalograph (EEG) signals has developed rapidly, and compared with traditional methods, deep learning has achieved competitive results. However, it is still a challenge to design and train an end-to-end network to fully extract the potential features of motor-imaging EEG signals. Based on the temporal and spatial characteristics of EEG, a multi-scale spatiotemporal self-attention network model based on attention mechanism is proposed to classify motor imagery EEG signals into four categories, such as left hand, right hand, foot and tongue/rest. Since the amplitude and response time of motor imagery EEG signal vary with the subjects, it is impossible to determine the most relevant brain region with motor imagery. Therefore, the self-attention mechanism is used to select the best channel by automatically weighting the higher weight to the motion-related channel and the lower weight to the non-motion-related channel in space. And parallel multi-scale TCN layer is used to extract the temporal feature at different scales and to eliminate the noise in time domain. The fusion module fuses the extracted spatial and temporal features, and finally inputs them to the classification module for classification. The model achieves accuracy of 79.26%, 85.90% and 96.96% on the BCI competition datasets IV-2a, IV-2b and HGD dataset. Compared with the baseline method, this method has a higher accuracy in subject classification. The results show that the method has good performance, robustness and transfer learning ability.

Objective: In order to expand the scope of cherry sales and achieve rapid grading of cherries under industrial conditions. Methods: Firstly, the YOLOX network was used to detect the defective fruit, in order to solve some problems where the defect was not obvious. The detection accuracy of the inconspicuous defect was improved by setting the appropriate fusion factor for the feature pyramid network, and in order to solve the problem of imbalance between various types of real samples, Focal Loss was integrated into the loss function. Then, the intact fruit was graded using the YOLOX network, and the attention mechanism CBAM was introduced to enhance the network feature extraction. Results: Experimental results showed that 97.59% of the mAP detected for cherry surface defects and 95.92% of the mAP of size and color grading. Conclusion: The accuracy of cherry defects and grading has been significantly improved by the improved YOLOX network.

Recent advances in inverse-design approaches for discovering optical structures based on desired functional characteristics have reshaped the landscape of nanophotonic structures, where most studies have focused on how light interacts with nanophotonic structures only. When quantum emitters (QEs), such as atoms, molecules, and quantum dots, are introduced to couple to the nanophotonic structures, the light–matter interactions become much more complicated, forming a rapidly developing field – quantum nanophotonics. Typical quantum functional characteristics depend on the intrinsic properties of the QE and its electromagnetic environment created by the nanophotonic structures, commonly represented by a scalar quantity, local-density-of-states (LDOS). In this work, we introduce a generalized inverse-design framework in quantum nanophotonics by taking LDOS as the bridge to connect the nanophotonic structures and the quantum functional characteristics. We take a simple system consisting of QEs sitting on a single multilayer shell–metal–nanoparticle (SMNP) as an example, apply fully-connected neural networks to model the LDOS of SMNP, inversely design and optimize the geometry of the SMNP based on LDOS, and realize desirable quantum characteristics in two quantum nanophotonic problems: spontaneous emission and entanglement. Our work introduces deep learning to the quantum optics domain for advancing quantum device designs; and provides a new platform for practicing deep learning to design nanophotonic structures for complex problems without a direct link between structures and functional characteristics.

ObjectiveTo assess the quality and safety of tea substitutes sold in Hebei Province.MethodsSixty-three samples of tea substitutes were collected from the markets of three cities in Hebei Province. Each sample was tested for moulds， coliforms， cadmium， moisture and ash content according to national standard methods. The test results were assessed in terms of qualification according to local safety standard of Hebei Province for tea substitutes.ResultsAmong the 63 samples， 16 samples had moulds counts exceeding the limit. The over-limit rate for moulds was 25.4%； the moulds contents varied from 140~18 000 CFU/g in over-limit samples with the median of 600 CFU/g. Four samples were found to exceed the limit of coliforms with a 6.3% （4/63） over-limit rate. The coliforms measurements of over-limit samples were between 7.5~110 MPN/g with the median of 110 MPN/g. One sample was over limit for cadmium with the content of 9.6 mg/kg. The test results for moisture and ash content of the 63 samples were all in accordance with the standard. Among the 63 samples， 46 samples were qualified for all measurements， and the total qualified rate was 73.0% （46/63）. There was no statistics difference in the qualified rate of tea substitutes among different producing areas and different product types.ConclusionThe unqualified rate of the tea substitutes sold in Hebei Province was high. Most unqualified samples exceeded the standard for mold， while a few samples exceeded the standard for coliforms and cadmium.

The nozzle is one of the key components in the integrated design of the aircraft body/propulsion,which directly affects the thrust-drag matching and pitching moment matching and other characteristics of the aircraft.In order to improve the efficiency of the optimal design of the nozzle,a multi-objective optimization model based on multi-fidelity surrogate model is established.The CFD results based on coarse mesh and inviscid model are used as low-fidelity data,and the CFD results based on fine mesh and SST k-ωturbulence model are used as high-fidelity data,the Cokriging models are constructed with the maximization of thrust coefficient,lift coefficient and pitching moment coefficient as optimization objectives,and the Pareto solution set is obtained by combining NSGA-Ⅱalgorithm.The optimization results show that the three aforementioned objectives are improved by 2.94%,13.0%and 40.6%respectively with the error less than 0.5%,and the sensitivity analysis is completed;the optimized flow field wave system structure is more complex which changes the wall pressure distribution law;compared with Kriging model,Cokriging model has considerable prediction performance and saves 62%of the construction cost.

Objective To evaluate the impacts of equal ratio ventilation (ERV) mode on lung morphology, gastric inflation and laryngeal mask airway ventilation during laparoscopic radical prostatectomy by lung ultrasound（LUS） and gastric ultrasound. Methods A prospective study was performed in 60 patients with prostate cancer undergoing laparoscopy from June 2021 to May 2022 in Nanjing First Hospital. The patients were randomly divided into ERP group (group E) and conventional ventilation group (group C,n=30, each). After induction of routine intravenous anesthesia and mechanical ventilation with laryngeal mask airway, and the ventilation mode was set as follows: the inspiratory-to-expiratory ratio in group E was 1∶1, while that in group C was 1∶2. After anesthesia induction (T0), 30 minutes after surgery begin (T1), 60 minutes after surgery begin (T2) and 10 minutes after surgery (T3), the peak airway pressure (Ppeak), airway plateau pressure(Pplat), mean airway pressure (Pmean),end-expiratory partial pressure of carbon dioxide (PETCO2) and minute ventilation (MV) were observed and compared between two groups. The LUS score, the cross-sectional area of gastric antrum and fundus and the incidence of airway complications were recorded in two groups. Results At T1 and T2, Ppeak in group E was significantly lower than that in group C(P＜0.05), and Pmean was significantly higher than that in group C(P＜0.05). There was no significant difference in PETCO2 and MV between two groups at different time points (P＞0.05). The LUS and the incidence of atelectasis in group E were statistically lower than those in group C (P＜0.05), and there was no significant difference in the cross-sectional area of gastric antrum and fundus（P＞0.05） and the incidence of airway complications（30.0% vs 32.1%，χ2=0.031，P=0.860) between two groups. Conclusion ERV mode during laparoscopic radical prostatectomy can reduce Ppeak, leakage rate of laryngeal mask, atelectasis and LUS scores without increasing the incidence of gastric inflation and laryngeal mask-related adverse complications.

Abstract Background Previous studies have shown that the Kyoto classification of gastritis can accurately predict H. pylori infection status on conventional gastroscopy. The aim of this study was to test whether the Kyoto classification of gastritis applies well to magnetic controlled capsule endoscopy (MCCE). Methods We consecutively recruited 227 participants who underwent both MCCE and urea breath tests (UBTs). Two physicians who were blinded to the UBT results independently made the diagnosis of H. pylori infection status according to 10 findings listed in the Kyoto classification of gastritis after reviewing MCCE images. We also developed 2 predictive models to assess H. pylori infection status by combining these 10 findings. Results The MCCE’s overall diagnostic accuracy for H. pylori infection status was 80.2%. The sensitivity, specificity and diagnostic odds ratio (DOR) for current infection were 89.4%, 90.1% and 77.1, respectively. Major specific findings were mucosal swelling and spotty redness for current infection, regular arrangement of collecting venules (RAC), streak redness, fundic gland polyp (FGP) for noninfection, and map-like redness for past-infection. In the two prediction models, the area under the curve (AUC) values for predicting noninfection and current infection were 84.7 and 84.9, respectively. Conclusions The Kyoto classification of gastritis applied well to MCCE. H. pylori infection status could be accurately assessed on MCCE according to the Kyoto classification of gastritis.

In order to study the internal cross-linking mechanism between cellulose and polysaccharides as wet-end additives in papermaking reconstituted tobacco, crystalline cellulose (MCC) was used as the target in this paper, sodium alginate (SA) , pullulan (Pul) , κ-carrageenan (κ-C), carboxymethyl chitosan (CMCS), hydroxypropyl chitosan (HPCS) and tamarind polysaccharide (TSP) were selected, the structure and composition of the monomer polysaccharides were determined by dynamic light scattering and ion chromatography, at the same time, the characteristics of the above-mentioned polysaccharides and MCC blend were analyzed, the results showed that SA contained a lot of negative charge, which resulted in weak adhesion and poor adsorption ability in cellulose blend; Pul was the most compact chain conformation, effectively improve the adhesion of cellulose, can be quickly adsorbed on cellulose; κ-C increased the viscosity of MCC blend and could adhere the 30μm size fragments of cellulose CMCS had strong adhesion to cellulose edge; due to the introduction of carboxymethyl group, the hydrophilicity of HPCS and the apparent viscosity of MCC blend increased greatly; the complex branched-chain structure of TSP results in high storage modulus of TSP/cellulose blend, and the amount of TSP adsorbed on cellulose is large and lasts long. Pul, TSP, κ-c, CMCS and TSP could adsorb cellulose well.

The article analyses the influencing factors of the anti-fouling properties of road traffic markings and researches a double-coated, anti-fouling road marking with good heat stability and high coating density. The anti-fouling performance of the road marking during its use period was experimentally analysed using SPSS software. The results indicate that the anti-fouling performance of the double-coated road marking is significantly better than that of thermoplastic markings and two-component markings, and it has a promising prospect for widespread application.

In the dynamic and rapid detection of retroreflection coefficient in road traffic markings, different factors may affect the detection results. This study investigates the significance of different levels of environmental light intensity and vehicle speed on the detection results of retroreflection coefficient in road markings using one-way ANOVA analysis. The results show that within a 95% confidence interval, there is no statistically significant difference in the detection results of retroreflection coefficient under different levels of environmental light intensity and vehicle speed. The variance effect size calculation indicates that for the detection of retroreflection coefficient in road markings, vehicle speed control is more important than light intensity. To obtain more reliable detection results, it is recommended to maintain a stable and uniform speed of no more than 80 km/h during the detection process.

As a major producer of oil and gas, China has historically adopted a variety of different approaches to managing its overall oil and gas supply chain. It also incorporates decisions and forecasts of trends in the overall petroleum supply chain in the context of existing domestic and international societal trends, such as the impact of national policies and the supply chains of China’s domestic petrochemical companies in the context of Sinopec’s past annual reports and assessments and recommendations for sustainable supply chains. This article uses the case study of Sinopec, a major Chinese chemical company, to provide statistics on the domestic market context and to propose and discuss risk management and sustainable supply chains for the company’s supply chain, as well as quantitative analysis to forecast and analyze China’s domestic oil consumption, with case studies and data on all indicators, and to show the impact of plans and trends on the chemical supply chain over time. The implications and trends of developing trends in the chemical supply chain are presented, and conclusions and final recommendations are drawn.

Influenza is a significant public health challenge because of the emergence of antigenically shifted or highly virulent strains. The neuraminidase inhibitor oseltamivir is used as an antiviral drug in clinical treatment. However, its therapeutic effects can be greatly compromised by the emergence of drug-resistant mutant viruses. Thus, there is an urgent need to distinguish drug-resistant strains with a simple method. To address this, in the present study, we develop a rapid, sensitive and convenient molecular diagnosis method based on CRISPR/Cas12a technology and lateral flow detection (LFD). By targeting mutant sequences amplified by recombinase polymerase amplification (RPA) reaction, crRNA is designed to develop the CRISPR/Cas12a assay, and 2000 copies can be directly observed by the naked eye under blue light-emitting diode (LED) light. Combined with LFD, the limit of detection of RPA-CRISPR/Cas12a-LFD is about 20 copies of target sequence per reaction. Collectively, RPA-CRISPR/Cas12a-LFD method provides a novel alternative for the sensitive, specific and portable detection to diagnose oseltamivir-resistant mutant strains.

Wheat tempering is an important step in wheat milling process. Appropriate tempering method is beneficial to shorten tempering time, reduce microbial content, improve the quality of flour and by-products, reduce energy consumption and save production cost. However, tempering process is often affected by multiple factors such as raw grain varieties, water quality, water addition, time, temperature, season and climate. At present, wheat flour processing enterprises are facing technical problems such as uneven tempering and easy breeding of microorganisms in the tempering process. In this paper, the basic principle of wheat tempering and the effects of raw grain, water addition, different temperature and humidity, time and other factors on wheat tempering were introduced in detail. The effects of different tempering methods such as heat treatment, vibration, ultrasound, vacuum, fracturing damage, biological enzyme preparation, salt water, acid water and other sterilization treatment on wheat processing and product quality were emphatically expounded. The advantages and disadvantages of different tempering methods and their application in current domestic production were also summarized in order to provide reference for wheat milling enterprises. Finally, it is pointed out that further research should be carried out on the accurate calculation and regulation of water content and the combination of different tempering methods and its effects on flour application quality for different wheat varieties.

ObjectiveTo investigate the prevalence and influencing factors of job burnout among nurses in a tertiary general hospital， providing a reference for medical institutions to develop effective management for job burnout.Methods684 nurses were investigated with general demographic questionnaire and Maslach burnout inventory （MBI）.ResultsAmong the three dimensions of job burnout， the scores ［median （25 percentile， 75 percentile）］ of emotional exhaustion （EE）， depersonalization （DP） and personal achievement （PA） were 25 （17， 33）， 5 （2， 9）， and 30 （23， 37）， respectively. Low PA was the most common problem （62.6%）， followed by EE （43.4%）， and DP （24.6%）. Objective To investigate the application value of conventional ultrasound typing and contrast-enhanced ultrasound (CEUS) in non-mass breast like lesions (NMLs). Methods A total of 50 patients (50 lesions) who were pathologically confirmed to have breast NMLs in Tianjin Fifth Central Hospital. The corresponding relationship between conventional ultrasound typing of NMLs and BI-RADS classification was analyzed, and CEUS was performed on breast NMLs to compare the diagnostic value of combined BI-RADS and CEUS. Results The NMLs conventional ultrasound classified Ⅰa and Ⅱa lesions corresponded to BI-RADS classification 4a; Ⅲ and Ⅳ lesions corresponded to BI-RADS classification 4b; Ⅰb and Ⅱb lesions corresponded to BI-RADS classification 4c. A statistically significant correlation was observed between the expansion of lesion range and the presence of "crab foot sign" in benign and malignant lesions after the enhancement (P < 0.05). Pearson correlation analysis showed that enlarged lesion area, "crab foot sign, " peak intensity, and area under the curve were associated with malignant breast NMLs. The diagnostic value of CEUS combined with BI-RADS was higher than that of conventional ultrasound BI-RADS. Conclusion NMLs conventional ultrasonographic classificationsⅠa andⅡa have the lowest malignant risk, whereas classifications Ⅰb and Ⅱb have the highest value. The expansion of lesion range and the presence of "crab foot sign" after enhancement are helpful for rapid differentiation of benign and malignant NMLs.

Objective To explore the expression of fibrinogen-like protein 1 (FGL1), the distribution of tumor-infiltrating lymphocytes (TILs), and their relationship with the prognosis of esophageal squamous-cell carcinoma (ESCC) patients. Methods We analyzed retrospectively the clinical data of 120 ESCC patients. The expression of FGL1 was detected through immunohistochemistry. The distributions of intratumoral TILs (iTILs) and stromal TILs (sTILs) were evaluated under a microscope. Survival analysis was used to evaluate the patient outcomes. Results The positive rate of FGL1 in ESCC was 18.3% (22/120), and it was connected to the TNM stage, lymph node status, and TILs. A total of 73 cases (60.8%) showed low levels of iTILs (iTILs≤10%), and 47 cases (39.2%) exhibited high iTIL levels (iTILs > 10%). Similarly, 82 cases (68.3%) presented low levels of sTILs (sTILs≤10%), and 38 cases (31.7%) manifested high sTIL levels (sTILs > 10%). The distribution of iTILs was associated with FGL1, tumor differentiation, and TNM stage, whereas the distribution of sTILs was associated with FGL1, tumor location, and TNM stage. The Kaplan–Meier survival analysis showed that tumor diameter, TNM stage, lymph node status, FGL1, and TILs were associated with the prognosis of patients with ESCC (P < 0.05). Multivariate Cox regression revealed that FGL1, TILs and TNM stage were the influencing factors of prognosis. Conclusion FGL1 expression is associated with the poor prognosis and may be a prognostic biomarker of ESCC. FGL1 combined with TILs can be used as a biomarker to predict ESCC.

In influenza vaccine development, Madin–Darby canine kidney (MDCK) cells provide multiple advantages, including large-scale production and egg independence. Several cell-based influenza vaccines have been approved worldwide. We cultured H5N1 virus in a serum-free MDCK cell suspension. The harvested virus was manufactured into vaccines after inactivation and purification. The vaccine effectiveness was assessed in the Wuhan Institute of Biological Products BSL2 facility. The pre- and postvaccination mouse serum titers were determined using the microneutralization and hemagglutination inhibition tests. The immunological responses induced by vaccine were investigated using immunological cell classification, cytokine expression quantification, and immunoglobulin G (IgG) subtype classification. The protective effect of the vaccine in mice was evaluated using challenge test. Antibodies against H5N1 in rats lasted up to 8 months after the first dose. Compared with those of the placebo group, the serum titer of vaccinated mice increased significantly, Th1 and Th2 cells were activated, and CD8+ T cells were activated in two dose groups. Furthermore, the challenge test showed that vaccination reduced the clinical symptoms and virus titer in the lungs of mice after challenge, indicating a superior immunological response. Notably, early after vaccination, considerably increased interferon-inducible protein-10 (IP-10) levels were found, indicating improved vaccine-induced innate immunity. However, IP-10 is an adverse event marker, which is a cause for concern. Overall, in the case of an outbreak, the whole-virion H5N1 vaccine should provide protection.

Objective: This study aims to explore the correlation between the types and contents of fatty acids and lecithin in fresh walnut. Methods: A kinds of 20 walnuts were selected from Chengdu in Sichuan Province. The method of the gas chromatography and the high performance liquid chromatography were used for quantitative determination. Correlation analysis and cluster analysis were used to analyze the results. Results: The fatty acid composition of 20 kinds of fresh walnut was the same basically, with significant differences in content （P＜0.05）. Longnan 15 had the highest oleic acid content. The contents of linoleic acid, arachidonic acid and cis-11-eicosenoic acid of Guangfeng 1 were the highest. The variation coefficient of lecithin was relatively small, and the content of lecithin was the highest in Xilo 3. The content of lecithin in fresh walnut had a high correlation with the content of fatty acids, and unsaturated fatty acids are highly associated. When the euclide distance was 15, the 20 walnut species could be divided into 3 categories. Conclusion: Longnan 15 and Guangfeng 1 are walnut varieties with high oil quality relatively; Xilo 3 is a walnut variety resource with high content of lecithin relatively.

Germination can improve the level of the free amino acid, and further improve the nutritious value of the foxtail millet. However, the effect of germination temperature and time on them is still unclear. The free amino acid of the germinated foxtail millet has been analyzed at different germination temperatures (20, 25, 30, 35, and 40 ℃) and time (0, 6, 12, 18, 24, and 30 h). Results reveal that 21 components of free amino acids increased. The lower temperature or higher temperature is not suitable for the accumulation of the free amino acid. The free amino acids increase to the maximum at the germination temperature of 30 ℃ for 30 h. The total free amino acids and total essential free amino acids can reach (810.7±69.0) and (367.8± 30.2) mg/100g, respectively. Among the amino acids, the contents of alanine and threonine increase to the highest level. The alanine has increased to (120.0±12.0) and (115.0±14.0) mg/100g at the temperature of 30 and 35 ℃, respectively. The threonine has increased to (106.0±1.0) mg/100g at the temperature of 35 ℃. The γ-aminobutyric acid reaches the maximum level at the germination temperature of 35 ℃ and reaches (60.1±1.8) mg/100g. In conclusion, the germination can promote the increase of the free amino acid and adjust the nutrition of the foxtail millet, and the germinated foxtail millet is a potential source for functional food.
