Abstract

Thymosin beta 4 (T beta 4) is a key factor in cardiac development, growth, disease, epicardial integrity, blood vessel formation and has cardio-protective properties. However, its role in murine embryonic stem cells (mESCs) proliferation and cardiovascular differentiation remains unclear. Thus we aimed to elucidate the influence of T beta 4 on mESCs. Target genes during mESCs proliferation and differentiation were detected by real-time PCR or Western blotting, and patch clamp was applied to characterize the mESCs-derived cardiomyocytes. It was found that T beta 4 decreased mESCs proliferation in a partial dose-dependent manner and the expression of cell cycle regulatory genes c-myc, c-fos and c-jun. However, mESCs self-renewal markers Oct4 and Nanog were elevated, indicating the maintenance of self-renewal ability in these mESCs. Phosphorylation of STAT3 and Akt was inhibited by T beta 4 while the expression of RAS and phosphorylation of ERK were enhanced. No significant difference was found in BMP2/BMP4 or their downstream protein smad. Wnt3 and Wnt11 were remarkably decreased by T beta 4 with upregulation of Tcf3 and constant beta-catenin. Under mESCs differentiation, T beta 4 treatment did not change the expression of cardiovascular cell markers alpha-MHC, PECAM, and alpha-SMA. Neither the electrophysiological properties of mESCs-derived cardiomyocytes nor the hormonal regulation by Iso/Cch was affected by T beta 4. In conclusion, T beta 4 suppressed mESCs proliferation by affecting the activity of STAT3, Akt, ERK and Wnt pathways. However, T beta 4 did not influence the in vitro cardiovascular differentiation.

Abstract

The Gram-negative bacterium Riemerella anatipestifer CH-2 is resistant to lincosamides, having a lincomycin (LCM) minimum inhibitory concentration (MIC) of 128 µg/mL. The G148_1775 gene of R. anatipestifer CH-2, designated lnu (H), encodes a 260-amino acid protein with ≤41% identity to other reported lincosamide nucleotidylyltransferases. Escherichia coli Rosetta TM (DE3) containing the pBAD24- lnu (H) plasmid showed four- and two-fold increases in the MICs of LCM and clindamycin (CLI), respectively. A kinetic assay of the purified Lnu(H) enzyme for LCM and CLI showed that the protein could inactive lincosamides. Mass spectrometry analysis demonstrated that the Lnu(H) enzyme catalysed adenylylation of lincosamides. In addition, an lnu (H) gene deletion strain exhibited 512- and 32-fold decreases in LCM and CLI MICs, respectively. The wild-type level of lincosamide resistance could be restored by complementation with a shuttle plasmid carrying the lnu (H) gene. The transformant R. anatipestifer ATCC 11845 [ lnu (H)] acquired by natural transformation also exhibited high-level lincosamide resistance. Moreover, among 175 R. anatipestifer field isolates, 56 (32.0%) were positive for the lnu (H) gene by PCR. In conclusion, Lnu(H) is a novel lincosamide nucleotidylyltransferase that inactivates LCM and CLI by nucleotidylylation, thus conferring high-level lincosamide resistance to R. anatipestifer CH-2. 37. Hyposmotic challenge modulates function of L-type calcium channel in rat ventricular myocytes through protein kinase C

Abstract

Aim: To study the effects and mechanisms by which hyposmotic challenge modulate function of L-type calcium current (I-Ca,I-L) in rat ventricular myocytes. Methods: The whole-cell patch-clamp techniques were used to record I-Ca,I-L in rat ventricular myocytes. Results: Hyposmotic challenge(similar to 220 mosmol/L) induced biphasic changes of I-Ca,I-L, a transient increase followed by a sustained decrease. I-Ca,I-L increased by 19.1%+/- 6.1% after short exposure (within 3 min) to hyposmotic solution. On the contrary, long hyposmotic challenge (10 min) decreased I-Ca,I-L to 78.1%+/- 11.0% of control, caused the inactivation of I-Ca,I-L, and shifted the steady-state inactivation curve of I-Ca,I-L to the right. The decreased I-Ca,I-L induced by hyposmotic swelling was reversed by isoproterenol or protein kinase A (PKA) activator foskolin. Hyposmotic swelling also reduced the stimulated I-Ca,I-L by isoproterenol or foskolin. PKA inhibitor H-89 abolished swelling-induced transient increase of I-Ca,I-L, but did not affect the swelling-induced sustained decrease of I-Ca,I-L. NO donor SNAP and protein kinase G (PKG) inhibitor Rp-8-Br-PET-cGMPS did not interfere with swelling-induced biphasic changes of I-Ca,I-L. Protein kinase C (PKC) activator PMA decreased I-Ca,I-L and hyposmotic solution with PMA reverted the decreased I-Ca,I-L by PMA. PKC inhibitor BIM prevented the swelling-induced biphasic changes of I-Ca,I-L. Conclusion: Hyposmotic challenge induced biphasic changes of I-Ca,I-L, a transient increase followed by a sustained decrease, in rat ventricular myocytes through PKC pathway, but not PKG pathway. PKA system could be responsible for the transient increase of I-Ca,I-L during short exposure to hyposmotic solution.

Abstract

ABSTRACTThe hyperpolarization-activated current (If) plays an important role in determining the spontaneous rate of cardiac pacemaker cells. The automatic rhythmicity also exists in working cells of embryonic heart, therefore we studied developmental changes in functional expression and β-adrenergic regulation of Ifin embryonic mouse heart. The expression of Ifis high in early developmental stage (EDS) (10.5 d after coitus) ventricular myocytes, low in intermediate developmental stage (IDS) (13.5 d) atrial or ventricular myocytes and even lower in late developmental stage (LDS) (16.5 d) atrial or ventricular myocytes, indicating that these cells of the EDS embryonic heart have some properties of pacemaker cells. β-adrenergic agonist isoproterenol (ISO) stimulates Ifin LDS but not in EDS cardiomyocytes, indicating that the b-adrenergic regulation of If is not mature in EDS embryonic heart. But forskolin (a direct activator of adenylate cyclase) and 8-Br-cAMP (a membrane-permeable analogue of cAMP) increase the amplitude of Ifin EDS cells, indicating that adenylate cyclase and cAMP function fairly well at early stage of development. Furthermore, the results demonstrate that Ifis modulated by phosphorylation via cAMP dependent PKA both in EDS and LDS cells.

Abstract

As an important part of the soil microbial system, fungi can clearly indicate changes in the soil environment.Human activities in the city can easily affect the soil condition, so the phenomenon of artificial heavy metal enrichment often appears in urban soil. The objective of this study was to analyze the fungal community structure in different urban functional areas and to determine the effect of heavy metal content in different urban functional areas on the soil fungal community structure. This study provides theoretical basis for protecting and repairing the urban soil ecosystem and transforming and improving urban environmental quality. Soil samples from eight sampling sites in five functional areas of Beibei District in Chongqing were taken as the research objects. The diversity and community structure of fungi in soil were studied using high-throughput sequencing technology. The content of Cd and Hg in the soil of different functional areas in Beibei District exceeded the environmental background value of Chongqing. The 20-40 cm and 40-60 cm soil layers of JD were slightly polluted. The 20-40 cm soil layer and 0-20 cm soil layer of JLD and ZYY, respectively, were in the alerting state of heavy metal pollution. The Sobs index, Chao 1 index, and Ace index of most sampling points decreased with the increase in soil depth. The NMDS analysis showed that the composition of fungal communities between the 0-20 cm and 20-40 cm soil depths in both JD and ZYY were quite different. From the perspective of community composition, Ascomycota was the most abundant phylum in the soil, followed by unclassified _k _Fungi and Basidiomycota. At the genus level, unclassified_k_Fungi , unclassified_p_Ascomycota , unclassified_o_Sordariales , Scopuloides , Robillarda , and Dactylonectria had higher abundances. The redundancy analysis (RDA) showed that Pb and Zn had the greatest effect on the samples, and the effect on the fungal community structure was significantly different. This study has deepened the understanding of the relationship between the content of heavy metals in different urban functional areas and the structure of fungal communities and has provided a scientific basis for the rational use and planning of urban soil.
