The robust use of osmolytes (i.e., polyols and sugars) in the key therapeutic regimens/formulations has questioned their impact beyond the stability of therapeutic proteins as these osmolytes trigger structural alterations into proteins including misfolding and subsequent aggregation into amyloid fibrils. Therefore, the current study is the first to delineate the inhibitory effect of carvacrol (CRV) on the carbonyl osmolyte-induced aggregation as well as structural alterations to the bovine serum albumin (BSA) via a set of physicochemical as well as artificial intelligence (AI)-based molecular docking studies. Our initial findings from physicochemical investigations revealed that CRV exhibits substantial protection to BSA under carbonyl osmolyte stress as evident by the compromised hyperchromicity, Schiff's bases, carbonyl and hydroxymethyl furfural content, reduced fluorescent signals, low Rayleigh scattering and prevention of covalent modifications at Lys and Arg residues. The protection against aggregate formation by CRV was further confirmed through the reduced amyloid-specific congo red absorbance as well as fluorescent signals recorded after adding the fibril-specific extrinsic fluorophore probes (i.e., ThT and ANS). The AI-based molecular docking analysis further revealed that CRV (ΔG: -4.96 kcal/mol) competes with d-fructose (ΔG: -4.40 kcal/mol) to mask the Lys and Arg residues to restrict the osmolyte-mediated protein modifications. In conclusion, CRV exhibits substantial protective impact against carbonyl osmolyte-induced structural alterations and protein misfolding and aggregation.

Hyperglycemia is a poorly controlled diabetic condition, affects about 70 percent of people all round the world. In the year 2015, about 41.5 crore people were diabetic and is expected to reach around 64.3 crore by the year 2040. Cardiovascular diseases (CVDs) are considered as one of the major risk factors that cause more than half of the death of diabetic patients and promote related comorbidities. Atherosclerosis and amyloidosis are the prime rudimentary cause associated with CVDs. Apolipoprotein A-I (ApoA-I) of high-density lipoprotein (HDL) have protective action against CVDs, participate in reverse cholesterol transport (RCT) mechanism and lipid metabolism, but gets easily glycated under prolonged hyperglycemic aura i.e. glycation. ApoA-I have a potent role in maintenance of glucose level, providing a compelling link between diabetes and CVDs. Increased protein glycation in people with diabetes promote atherosclerosis which might play possible role in promotion of protein aggregation by altering the protein structure and its confirmation. Here, we intend to investigate the mechanistic behavior of ApoA-Iunder the menace of glycation and its impact on ApoA-I structure and function that possibly link with aggregation or amyloidosis.

We aimed to identify and evaluate natural organosulfur compounds (OSCs) from the genus Allium as potent dual inhibitors of α-amylase and α-glucosidase via a set of molecular simulations and in-vitro analysis. We reported that all OSCs fall under the acceptable scores of Lipinski’s rules and desirable ADME parameters. In-silico screening revealed that OSCs strongly interacted with α-amylase and α-glucosidase crystal structures. Our in-vitro studies with four OSCs having best ΔG-scores demonstrated that alliin, S-allyl-L-cysteine (SAC), N-acetylcysteine (NAC), and S-ethyl-L-cysteine (SEC) exhibit a marked in-vitro inhibition of α-amylase activity, whereas, γ-glutamylcysteine (GGC), SEC, alliin, and SAC potentially inhibited the in-vitro α-glucosidase activity. Our enzyme kinetics studies depicted that the selected OSCs competitively inhibited the α-amylase and α-glucosidase activity. In conclusion, this initial in-silico and the in-vitro report demonstrates the drug-likeness and ADME indices of OSCs and identifies alliin, SAC, NAC, SEC, and GGC as potential dual inhibitors of α-amylase and α-glucosidase.

Aims: Hyptis suaveolens (L.) Poit, is one of the natural herbs with several medicinal properties. However, many medicinal aspects of this herb still need to be explored. Therefore, our aim was to examine the antioxidant, antimicrobial properties and genoprotective effect of H. suaveolens methanolic extracts (HSME) of seed, stem, and root. Study design: extraction and therapeutic aspects of H. suaveolens. Place and Duration of Study: 1) Department of Medical Laboratory Sciences, College of Applied Medical Sciences, Majmaah University, Al-Majmaah and 2) Clinical Biochemistry & Natural Product research laboratory, Department of Biosciences, Integral University, lucknow between 2018-2020. Methodology: HSME were extracted through soxhlet extractor and further analyzed for TPC, antioxidant activity through DPPH and FRAP assay followed by antimicrobial potential through zone of inhibition and MIC/MBC assay. We also examined the genoprotective properties of HSME on oxidative DNA damage. Results: Our results showed that TPC (180±5 mg GAE/g dw), DPPH scavenging activity (IC50 value = 72±0.45 µg/ml) and FRAP value (1.443±0.02 µM ferrous ion/mg extract) was highest in HSME seeds followed by root and stem. The results also illustrated that the antimicrobial activity of HSME (seed and stem) against five bacterial strain were found very effective than root part. Moreover, genoprotective effect of HSME seeds (80±3 % retention) was better than stem (41±2 %) and root (32±2 %) extract. Conclusion: The study revealed that HSME seed extract showed potential bioactivities might be due to presence of high TPC and can be used to treat diseases related with oxidative stress or microbial infections.

The current in-vivo study was premeditated to uncover the protective role of ezetimibe (EZ) against advanced glycation endproducts (AGEs)-related pathologies in experimental diabetes. Our results showed that EZ markedly improved the altered biochemical markers of diabetes mellitus (DM) (FBG, HbA1c, insulin, microalbumin, and creatinine) and cardiovascular disease (in-vivo lipid/lipoprotein level and hepatic HMG-CoA reductase activity) along with diminished plasma carboxymethyl-lysine (CML) and renal fluorescent AGEs level. Gene expression study revealed that EZ significantly down-regulated the renal AGEs-receptor (RAGE), nuclear factor-κB (NFκB-2), transforming growth factor-β (TGF-β1), and matrix metalloproteinase-2 (MMP-2) mRNA expression, however, the neuropilin-1 (NRP-1) mRNA expression was up-regulated. In addition, EZ also maintained the redox status via decreasing the lipid peroxidation and protein-bound carbonyl content (CC) and increasing the activity of high-density lipoprotein (HDL)-associated-paraoxonase-1 (PON-1) and renal antioxidant enzymes as well as also protected renal histopathological features. We conclude that EZ exhibits antidiabetic and reno-protective properties in diabetic rats.

Advanced glycation end-products (AGEs) induce the production of reactive oxygen species (ROS) and extra cellular matrix (ECM) degradation via suppression of neuropilin-1 (NRP-1) and interaction with AGE-receptors (RAGE). This study aimed to reveal whether modulation of NRP-1 by rosuvastatin (RT) prevents AGE-induced renal injury via targeting RAGE/matrix metalloproteinase-2 (MMP-2) signaling in diabetic rats. Treatment with RT ameliorated the altered level of markers of glycemic control, renal injury, cholesterol, triglyceride (TG) and hepatic HMG-CoA reductase activity; the level of circulatory carboxymethyl-lysine (CML) and the accumulation of fluorogenic-AGEs in renal tissue was reduced; the expression of renal NRP-1, a checkpoint target, was stimulated; the transcription of RAGE, NF?B-2, TGF-?1 and MMP-2 was suppressed; the circulatory carbonyl content (CC) and paraoxonase-1 (PON-1) activity was ameliorated, and renal histopathological features were attenuated as evidenced by improved glomerular appearance, Bowman?s space and abundant podocytes in kidneys. In conclusion, RT exhibited the potential to counteract diabetes and AGE-induced renal pathologies via stimulation of NRP-1, suppression of RAGE, and of genes responsible for ECM disintegration (MMP-2) and the inflammatory response (NF?B-2).

An experiment was conducted to study the influence of combinations of different fodder crops on nutritional quality under hydroponic condition at Agronomy field unit, Zonal Agricultural Research Station, Gandhi Krishi Vignana Kendra, University of Agricultural Sciences, Bangalore during the year 2018. The experiment was conducted with 13 treatment combinations involving maize, finger millet, little millet and cowpea at different proportions by using completely randomised design (CRD) and repeated three times. The results revealed that the crude protein and carbohydrate yield was highest in 100% Cowpea and 25% Cowpea + 75% Maize treatments compared to other treatments.

Alzheimer’s disease (AD) is a progressive neurological disorder that affects 50 million people. Despite this, only two classes of medication have been approved by the FDA. Therefore, we have planned to develop therapeutics by multitarget approach. We have explored the library of 2029 natural product-like compounds for their multi-targeting potential against AD by inhibiting AChE, BChE (cholinergic pathway) MAO-A, and MOA-B (oxidative stress pathway) through in silico high-throughput screening and molecular dynamics simulation. Based on the binding energy of these target enzymes, approximately 189 compounds exhibited a score of less than −10 kcal/mol against all targets. However, none of the control inhibitors exhibited a binding affinity of less than −10 kcal/mol. Among these, the top 10 hits of compounds against all four targets were selected for ADME-T analysis. As a result, only F0850-4777 exhibited an acceptable range of physicochemical properties, drug-likeness, pharmacokinetics, and suitability for BBB permeation with high GI-A and non-toxic effects. The molecular dynamics study confirmed that F0850-4777 remained inside the binding cavity of targets in a stable conformation throughout the simulation and Prime-MM/GBSA study revealed that van der Waals’ energy (ΔGvdW) and non-polar solvation or lipophilic energy (ΔGSol_Lipo) contribute favorably towards the formation of a stable protein–ligand complex. Thus, F0850-4777 could be a potential candidate against multiple targets of two pathophysiological pathways of AD and opens the doors for further confirmation through in vitro and in vivo systems.

Introduction: We have previously demonstrated that sequentially extracted methanolic fractions of Boerhaavia diffusa (Linn.) showed marked antioxidant, antidiabetic and oxidative-DNA damage protective properties in vitro. The present study was undertaken to evaluate the beneficial effects of B.diffusa (Linn.) methanolic root extract and its partially purified bioactive fraction on streptozotocin (STZ)-induced hyperglycemia and hyperlipidemia in rats. Methods: The diabetic rats were treated for fourteen weeks either with methanolic extract of B. diffusa root (D-MT1, D-MT2, and D-MT3 : doses of 50, 150, and 300 mg/rat/day, respectively), partially isolated bioactive fraction (DBT: 0.5 mg/rat/day), or glibenclamide (D-GT: 0.5 mg/rat/day). Results: The level of fasting blood glucose (FBG) and glycated hemoglobin (HbA1c) were significantly alleviated in D-MT- and D-BT treated groups after fourteen weeks of administration. Moreover, plasma lipid profile, free fatty acids (FFAs), phospholipids (PLs), HMG-CoA reductase (HMG-R) activity, conjugated diene (CD), lipid hydroperoxide (LOOH), and malondialdehyde (MDA) were also markedly ameliorate d in all treatment groups. In addition, the activity of antioxidant enzymes, e.g., superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx), glutathione reductase (Gred), and glutathione-S-transferase (GST), were also significantly restored by D-MT — and D-BT — treated groups. Furthermore, histologically, all the unseemly features of nephropathy were extensively regressed and normalized by the administration of B. diffusa and its bioactive fraction. Conclusions: Our results demonstrate a strong antidiabetic and hypolipidemic impact of B. diffusa extract an ideal alternative therapeutic agent in the prevention and treatment of diabetes as well as diabetes-linked hyperlipidemia.

Japanese encephalitis (JE), a viral disease has seen a drastic and fatal enlargement in the northern states of India in the current decade. The better and exact cure for the disease is still in waiting. For the cause an in silico strategy in the development of the peptide vaccine has been taken here for the study. A computational approach to find out the Major Histocompatibility Complex (MHC) binding peptide has been implemented. The prediction analysis identified MHC class I (using propred I) and MHC class II (using propred) binding peptides at an expectable percent predicted IC (50) threshold values. These predicted Human leukocyte antigen [HLA] allele binding peptides were further analyzed for potential conserved region using an Immune Epitope Database and Analysis Resource (IEDB). This analysis shows that HLA-DRB1*0101, HLA-DRB3*0101, HLA-DRB1*0401, HLA-DRB1*0102 and HLA-DRB1*07:01% of class II (in genotype 2) and HLA-A*0101, HLA-A*02, HLA-A*0301, HLA-A*2402, HLA-B*0702 and HLA-B*4402% of HLA I (in genotype 3) bound peptides are conserved. The predicted peptides MHC class I are ILDSNGDIIGLY, FVMDEAHFTDPA, KTRKILPQIIK, RLMSPNRVPNYNLF, APTRVVAAEMAEAL, YENVFHTLW and MHC class II molecule are TTGVYRIMARGILGT, NYNLFVMDEAHFTDP, AAAIFMTATPPGTTD, GDTTTGVYRIMARGI and FGEVGAVSL found to be top ranking with potential super antigenic property by binding to all HLA. Out of these the predicted peptide FVMDEAHFTDPA for allele HLA-A*02:01 in MHC class I and NYNLFVMDEAHFTDP for allele HLA-DRB3*01:01 in MHC class II was observed to be most potent and can be further proposed as a significant vaccine in the process. The reported results revealed that the immune-informatics techniques implemented in the development of small size peptide is useful in the development of vaccines against the Japanese encephalitis virus (JEV). The current study is aimed at studying the inhibitory effect of glycyrrhizic acid (GA) on D-ribose-mediated protein glycation via various physicochemical analyses and in silico approaches. Being a potent free radical scavenger and a triterpenoid saponin, GA plays a vital role in diminishing the oxidative stress and thus could be an effective inhibitor of the nonenzymatic glycation process. Our data showed that varying concentrations of GA inhibited the in vitro BSA-AGEs via inhibiting the formation of fructosamines, fluorescent AGEs, scavenging protein carbonyl and hydroxymethyl furfural (HMF) content, and protection against D-ribose-induced modification of BSA as evident by increased free Arg and Lys residues in GA-treated Gly-BSA samples. Moreover, GA also attenuated D-ribose-induced alterations in the secondary structure of BSA by protecting the α-helix and β-sheet conformers and amide-I band delocalization. In addition, GA attenuated the modification in β-cross amyloid structures of BSA and in silico molecular interaction study too showed strong binding of GA with higher number of Lys and Arg residues of BSA and binding energy (ΔG) of -8.8 Kcal/mol, when compared either to reference standard aminoguanidine (AG)-BSA complex (ΔG: -4.3 Kcal/mol) or D-ribose-BSA complex (ΔG: -5.2 Kcal/mol). Therefore, GA could be a new and favorable inhibitor of the nonenzymatic glycation process that ameliorates AGEs-related complications via attenuating the AGE formation and glycation-induced multiple protein modifications with a reduced risk of adverse effects on protein structure and functionality; hence, it could be investigated at further preclinical settings for the treatment and management of diabetes and age-associated complications.

Diabetic retinopathy and nephropathy are questionably the most dreaded complications of diabetes; contribute to serious morbidity and mortality. The current study was undertaken with the aim of exploring the anti-lipoperoxidative and antioxidant status including nephroprotective and retinoprotective potential of Phyllanthus virgatus methanolic extract and its partially purified fraction in streptozotocin (STZ)-induced diabetic stressed rats. Diabetes was induced by intraperitoneal injection of Streptozotocin (60mg/kg B. Wt of rat). Among all the treatment groups, P. virgatus methanolic extract and its partially purified fraction at a dose of 50mg/kg (PET-1) and 0.5mg/kg (CT-1), respectively, showed significant protection against STZ-induced diabetic oxidative stress in rats with marked amelioration in lipid peroxidation byproducts level, antioxidant enzymes, nephroprotective and retinoprotective effects and plasma total antioxidant levels after treatment of 28 days. The combined results demonstrated significant protection against STZ-induced oxidative stress, nephropathy and retinopathy condition by P. virgatus methanolic extract and its bioactive compound.

Polydatin (PD) is a monocrystalline metabolite from the underground parts of Polygonum cuspidatum Sieb. et Zucc., a member of the Polygonaceae family, which has been traditionally used in Asian countries as both foodstuffs and medicine. PD, also reckoned as pieceid, 3,4',5-trihydroxystilbene-3-β-D-glucoside, (E)-piceid, (E)-polydatin, and trans-polydatin. It possesses potent biological activities i.e. analgesic, anti-inflammatory, antidiabetic, anticancer, and anti-atherosclerotic properties. The initial part of this report specifically explains distinct sequential mechanisms underlying the initiation and development of atherosclerotic plaques and later part deals with the pharmacological efficacy of PD in the management of major cardiac event i.e. atherosclerotic cardiovascular diseases (ASCVD) via modulation of a set of molecular mechanisms i.e. antioxidant potential, lipid and lipoprotein metabolism including total cholesterol (TC) and low density lipoprotein (LDL) levels, β-hydroxy-β-methyl-glutaryl-CoA reductase (HMG-R) expression and functionality, SIRT signalling, LDL-receptor (LDL-R), LDL oxidation status (Ox-LDL), effects on endothelial cells (ECs), smooth muscle cells (SMCs), macrophage, foam cell formation and plaque stabilization, inflammatory signalling pathways and hypertension. In contrast, one of the major insight into the potential cardioprotective molecular mechanism is the PD-mediated targeting of proprotein convertase subtilisin/kexin type-9 (PCSK-9) and LDL-R pathway, both at transcriptional and protein functional level, which makes it a better alternative therapeutic medicinal candidate to treat hypercholesterolemia, especially for the patients facing inadequate lipid lowering with classical HMG-R inhibitors (statins) and statin intolerance. Finally, to sum up the whole, we concluded that PD may be promoted from alternative to mainstream medicine in targeting risk factors mediated ASCVD.

The current study was undertaken to delineate the protective effect of Ginkgolide B, a phyto-constituent from Ginkgo biloba, on oxidized (ox)-LDL-induced endothelial dysfunction via targeting Lectin-like ox-LDL-receptor-1 (LOX-1), NADPH oxidase 4 (NOX-4), and other inflammatory proteins. Our results have shown that Ginkgolide B downregulated the expression of LOX-1 in ox-LDL-treated human umbilical vein endothelial cells (HUVECs) and RAW246.7 murine macrophages which ultimately resulted in decreased cholesterol deposits in HUVECs and RAW264.7. Moreover, Ginkgolide B suppressed the enhanced NOX4 expression, which was associated with attenuation of ROS generation in ox-LDL-stimulated HUVECs and RAW264.7 cells. Ginkgolide B also ameliorated the endothelial dysfunction by inhibiting the augmented expression of monocyte chemotactic protein-1 (MCP-1), intercellular adhesion molecule-1 (ICAM-1), and vascular cell adhesion molecule-1 (VCAM-1) in ox-LDL-activated HUVECs. Furthermore, the enhanced expression of many inflammatory cytokines in ox-LDL-induced RAW264.7 macrophages, both at transcription and protein level, was significantly down-regulated after Ginkgolide B treatment. Ginkgolide B also illustrated atheroprotective property via suppressing the augmented expression of matrix metalloproteinase-1 and cyclooxygenase-2 in ox-LDL-stimulated RAW264.7 macrophages. In summary, our study has established that Ginkgolide B ameliorates endothelial dysfunction via targeting LOX-1, NOX-4, MCP-1, ICAM-1, and VCAM-1 along with the markers associated with inflammatory cascades and thus could be promoted as a valuable therapeutic agent in prevention and management of atherosclerosis.

Methylglyoxal (MGO), a reactive dicarbonyl metabolite is a potent arginine directed glycating agent which has implications for diabetes-related complications. Dicarbonyl metabolites are produced endogenously and in a state of misbalance, they contribute to cell and tissue dysfunction through protein and DNA modifications causing dicarbonyl stress. MGO is detoxified by glyoxalase 1 (GLO1) system in the cytoplasm. Reactive oxygen species (ROS) are known to aggravate the glycation process. Both the processes are closely linked, and their combined activity is often referred to as "glycoxidation" process. Glycoxidation of proteins has several consequences such as type 2 diabetes mellitus (T2DM), aging etc. In this study, we have investigated the glycation of low-density lipoprotein (LDL) using different concentrations of MGO for varied incubation time periods. The structural perturbations induced in LDL were analyzed by UV-Vis, fluorescence, circular dichroism spectroscopy, molecular docking studies, polyacrylamide gel electrophoresis, FTIR, thermal denaturation studies, Thioflavin T assay and isothermal titration calorimetry. The ketoamine moieties, carbonyl content and HMF content were quantitated in native and glycated LDL. Simulation studies were also done to see the effect of MGO on the secondary structure of the protein. We report structural perturbations, increased carbonyl content, ketoamine moieties and HMF content in glycated LDL as compared to native analog (native LDL). We report the structural perturbations in LDL upon modification with MGO which could obstruct its normal physiological functions and hence contribute to disease pathogenesis and associated complications.

This study was undertaken to uncover the regulatory role of lycopene in targeting lipopolysaccharide (LPS) induced oxidative stress and inflammatory cascades and subsequent regulation of proprotein convertase subtilisin/kexin type-9 (PCSK-9) expression via sterol regulatory element binding protein-2 (SREBP-2) and hepatocyte nuclear factor-1α (HNF-1α). Further, protein-protein interaction (PPI) studies for Lycopene-Apo-CIII complex against lipoprotein lipase (LPL) were also performed to assess its regulatory role behind the enhanced circulatory TG/TRLs clearance. Lycopene treatment down-regulated hepatocyte PCSK-9 expression via down-regulation of HNF-1α, whereas, LDL-receptor (LDL-R) was up-regulated by subsequent up-regulation of SREBP-2. PPI studies showed that lycopene diminishes the affinity of Apo-CIII to complex with LPL (ΔG: −917.1 Kcal/mol) resulting in increased LPL functionality and TRLs clearance. Moreover, lycopene also ameliorated LPS stimulated oxidative-stress via enhanced total antioxidant and HDL associated PON-1 activity in addition to down-regulate the expression and plasma level of inflammatory mediators. Based on above findings, we concluded that lycopene exhibits dual role in targeting LPS induced oxidative stress and hypertriglyceridemia via down-regulation of PCSK-9, making greater no. of surface LDL-R available for LPS processing and clearance, as well as increased LPL activity through inhibition of Apo-CIII.

Garlic (Allium sativum Linn), a member of Liliaceae family has been used in different cultures as foodstuffs and medicine. It possesses potent biological activities, i.e., anti-inflammatory, antidiabetic, anticancer, and anti-atherosclerotic properties that are accredited to the organosulfur compound (OSCs) as well as other phytochemicals content of garlic, viz., phenolic acids, flavonoids, thiosulfinates, and anthocyanins that either work alone or synergistically by complex mechanisms. Among all the dietary phytochemicals from A. sativum Linn, OSCs have been found to show greater effects against various metabolic disorders. Major OSCs from garlic owing to its high therapeutic and pharmacological properties are allicin, diallyl trisulfide (DAT), allyl methyl trisulfide, diallyl disulfide (DAD), ajoene, and many others. The current chapter summarizes the therapeutic efficacy of garlic and its bio-active OSCs against risk factor-mediated atherosclerotic cardiovascular diseases (ASCVD) via modulation of distinct molecular mechanisms, i.e., antioxidant potential, lipid and lipoprotein metabolism, HDL functionality through paraoxonase-1 (PON-1) activity, HMG-CoA reductase (HMG-R) expression and functionality, LDL-receptor (LDL-R), LDL oxidation status, effects on macrophage, foam cell formation and plaque stabilization, inflammatory signalling pathways and hypertension, etc. We also hypothesized that garlic and its OSCs could be beneficial in targeting proprotein convertase subtilisin/kexin type-9 (PCSK-9) and LDL-R pathway in order to treat and manage hypercholesterolemia especially for the patients facing inadequate lipid lowering with classical HMG-R inhibitors (statins) and statin intolerance. Finally to sum up the whole, we conclude that the garlic and its OSCs may be promoted from alternative to mainstream medicine in targeting risk factor-mediated ASCVD.

Rosemary (Rosmarinus officinalis L.), the representative of Lamiaceae family is known for its various medicinal uses that are accompanied by their hallmark secondary metabolites, i.e., carnosol, carnosic acid and rosmarinic acid (mostly the polyphenolic diterpenes). In the age of medicines and methodologies, when we are floating through the advancements and achievements, we are being hijacked by various diseases leading to increased number of young deaths. Neurological disorders are one of them and characterized by any impairment in the nervous system, brain or spinal cord. The majority of young and aged people around the globe are manifested by neurological disorders, i.e., stroke, epilepsy, dementia, Alzheimer’s disease (AD), Parkinson’s disease (PD) and migraine. A large number of therapeutic approaches mend the symptoms in early stages of these disorders, but with the span of time, patients become progressively more disabled as they may suffer from drug-associated adverse effects. Emphasizing on the urgent need of alternative therapeutic regimens, natural products are encouraged worldwide in terms of safety and to minimize the aforesaid loss. In this order, the current chapter summarizes the protective role of R. officinalis L. and its bio-active metabolites against various neurological disorders via targeting amyloid-beta (A-β) aggregation, neuronal cell death, acetylcholinesterase (AChE), neuroinflammation, β-secretase (BACE-1) activity, mitochondrial redox status, etc. Based on the multifunctional nature due to effective bio-active secondary metabolites, R. officinalis can be a terrific alternative therapeutic source against many neurodegenerative diseases.

The potential of oxidized-LDL (Ox-LDL) to elicit inflammatory responses in macrophages leading to the atherosclerosis (AS) progression is well known. Since proprotein convertase subtilisin/Kexin-9 (PCSK-9), the posttranslational regulator of LDL-receptor, is associated with elevated LDL in the circulation, the present report was aimed to uncover the ameliorative effects of Ginkgolide B, a terpenic lactone fromGinkgo biloba, against Ox-LDL-induced alterations in cholesterol metabolism in HUVECs. Consequently, our results demonstrated that incubation with Ox-LDL significantly upregulated the PCSK-9 expression in HUVECs, which was significantly downregulated, both at mRNA and protein level, after Ginkgolide B treatment via subsequent suppression of sterol element binding protein (SREBP-2) expression. Moreover, Ginkgolide B-mediated inhibition of PCSK-9 activity was also validated byin silicomethods which revealed that it interferes the PSCK-9 interaction with LDL-receptor (LDL-R). Interestingly, Ox-LDL-induced LDL-R expression was further enhanced by Ginkgolide B treatment in HUVECs. Moreover, Ginkgolide B treatment lead to downregulation of lectin-like Ox-LDL receptor (LOX-1) and NADPH oxidase (NOX-4) expression which was upregulated in Ox-LDL-treated HUVECs, along with the attenuation of mitochondrial ROS generation. Furthermore, Ginkgolide B significantly inhibited the augmented expression of intercellular adhesion molecule-1 (ICAM-1) and vascular adhesion molecule-1 (VCAM-1) in Ox-LDL-activated HUVECs. Ginkgolide B also significantly ameliorated the inflammatory response in Ox-LDL-activated HUVECs by suppressing the expression of IL-1α, IL-1β, IL-6, CXCL-1, CXCL-2, and monocyte chemotactic protein (MCP-1), at mRNA and protein level. Ourin vitroandin silicostudy established that Ginkgolide B alleviated the Ox-LDL-induced inflammatory cascades and altered lipid metabolism in HUVECs by suppressing the PCSK-9 and, thus, could be established as a treasured alternative therapeutic candidate in the atherosclerosis management.

Introduction Triphala is widely prescribed herbal drug in the Indian traditional system of medicine. It is rich in antioxidants and possesses diverse beneficial properties. It is used to treat many diseases such as anemia, jaundice, constipation, asthma, fever, chronic ulcer and various gastrointestinal disorders. The aim of this study was to investigate the ulcer ameliorative effect of hydro-alcoholic extract of Triphala in indomethacin-induced gastric ulcer in Wistar rats. Methods Gastric ulcer was induced by indomethacin (10 mg/kg b wt, po) in Wistar rats. Triphala extract (1000 mg/kg b wt, po) or vehicle (1 ml/kg/day po of 1% CMC) was given to the rats for 15 days by oral gavage ( n = 6). The ulcer ameliorative effect of Triphala extract was compared with standard drug ranitidine (50 mg/kg b wt, po for 15 days). The ulcer index was calculated. Gastric histopathology and biochemical parameters like mucus, lipid peroxide, glutathione (GSH), catalase (CAT), superoxide dismutase (SOD) were determined. Results Treatment with Triphala extract significantly decreased the levels of ulcer index and lipid peroxide in the treated rats. The extract also significantly elevated the levels of mucus, GSH, CAT and SOD. These findings matched the results of histopathological studies. Conclusion Triphala extract possesses an ulcer ameliorative effect in gastric ulcer through strengthening the gastric mucosa, restoring the free radical scavenging enzymes and reducing the lipid peroxide production.

Glycation of proteins leads to the formation of advanced glycation end products (AGEs, which have significant role in the pathophysiology of diabetes complications. d-ribose appears to be the most reactive among the naturally occurring sugars and contribute significantly to the generation of AGEs. Glycation also results in the generation of free radicals causing structural modification which leads to the generation of neoantigenic epitopes. The aim of the present study was to investigate whether LDL modification results in auto-antibodies formation against its glycated conformer in diabetes and atherosclerosis patients.The binding characteristics of circulating auto-antibodies in patients against native and modified LDL were assessed. T2D (n=105), ATH (n=106) and T2D-ATH patients (n=72) were examined by direct binding ELISA as well as inhibition ELISA, compared with healthy age-matched controls (n=50). Furthermore, ketoamine moieties, HMF and carbonyl content were also estimated in these patient's and healthy subjects.High degree of specific binding was observed by 41.91% of T2D, 54.72% of ATH and 70.83% T2D-ATH patient's sera towards d-ribose glycated LDL, in comparison to its native analog (P0.05). Normal human sera showed negligible binding with either antigen. Competitive inhibition ELISA reiterates the direct binding results. The higher concentration of HMF, ketoamine and carbonyl content was observed in patient's sera than healthy subjects.LDL glycation results in structural perturbation causing generation of neoantigenic epitopes that are better antigens for antibodies in T2D, ATH and T2D-ATH patients where T2D-ATH subjects showed higher prevalence in auto-antibodies against ribosylated LDL.

Non-enzymatic protein glycation results in the formation of advanced glycation end products (AGEs) leads to the pathogenesis of long-term diabetic complications. Iridin (ID), an antioxidant, plays an important role in protecting against oxidative stress and could therefore be an efficacious anti-glycating regimen. Herein, we assessed the anti-glycating potential of ID against d-ribose induced glycation of bovine serum albumin (BSA) by various biophysical and biochemical techniques. Our results from several physicochemical assays advocated that ID was able to evidently prevent the AGEs generation via reducing hyperchromicity, early glycation products (EGPs), carbonyl content (CC), hydroxymethyl furfural (HMF) content, production of fluorescent AGEs, protection against loss of secondary structure (i.e. α-helix and β-sheets) of proteins, increasing the free lysine and free arginine content, reduced binding of congo red (CR), and reduced thioflavin T (ThT) and 8-aninilo-1-napthalene sulphonate (ANS)-specific fuorescence in glycated-BSA (Gly-BSA). On the basis of these findings, we concluded that ID possesses the significant anti-glycation potential and may be established as a remarkable anti-AGEs therapeutic agent. Further in-vivo and clinical studies are still warranted to uncover the therapeutic effects of ID against age-related as well as metabolic diseases.

The current study was undertaken to delineate the protective effect of Ginkgolide B, a phyto-constituent from Ginkgo biloba, on oxidized (ox)-LDL-induced endothelial dysfunction via targeting Lectin-like ox-LDL-receptor-1 (LOX-1), NADPH oxidase 4 (NOX-4), and other inflammatory proteins. Our results have shown that Ginkgolide B downregulated the expression of LOX-1 in ox-LDL-treated human umbilical vein endothelial cells (HUVECs) and RAW246.7 murine macrophages which ultimately resulted in decreased cholesterol deposits in HUVECs and RAW264.7. Moreover, Ginkgolide B suppressed the enhanced NOX4 expression, which was associated with attenuation of ROS generation in ox-LDL-stimulated HUVECs and RAW264.7 cells. Ginkgolide B also ameliorated the endothelial dysfunction by inhibiting the augmented expression of monocyte chemotactic protein-1 (MCP-1), intercellular adhesion molecule-1 (ICAM-1), and vascular cell adhesion molecule-1 (VCAM-1) in ox-LDL-activated HUVECs. Furthermore, the enhanced expression of many inflammatory cytokines in ox-LDL-induced RAW264.7 macrophages, both at transcription and protein level, was significantly down-regulated after Ginkgolide B treatment. Ginkgolide B also illustrated atheroprotective property via suppressing the augmented expression of matrix metalloproteinase-1 and cyclooxygenase-2 in ox-LDL-stimulated RAW264.7 macrophages. In summary, our study has established that Ginkgolide B ameliorates endothelial dysfunction via targeting LOX-1, NOX-4, MCP-1, ICAM-1, and VCAM-1 along with the markers associated with inflammatory cascades and thus could be promoted as a valuable therapeutic agent in prevention and management of atherosclerosis.

The present study is premeditated to extenuate the role of Ficus virens extract and its bioactive compound on cigarette smoke, an important risk factor for CVD, induced oxidative stress and hyperlipidemia. Cigarette smoke (CS) exposure to rats results in significant loss of body weight and increases blood carbon monoxide saturation (carboxyhemoglobin), nicotine, plasma TC, TG, and LDL-C levels but reduced level of antiatherogenic HDL-C. Moreover, owing to substantial oxidative stress generated in rats due to cigarette smoke a significant increase in plasma and erythrocytes lipid peroxidation products were observed which was well correlated with increase in ex-vivo BDC (48%) and MDA (53%) level (p < 0.001). Simultaneous administration of FVBM extract at higher dose (100 mg/rat) and F18 (n-Octadecanyl-O-α-D-glucopyranosyl(6’→1’’)-O-α-D-glucopyranoside) compound to CS-exposed rats effectively blocked the increase in plasma lipid and lipoprotein levels (p < 0.001) which was due to the marked suppression in the hepatic HMG-CoA reductase activity (p < 0.001) and significantly inhibit the lipid peroxidation process thus preventing the membrane damage, LDL oxidation, and in turn subsequent atherosclerosis. Thus, the results clearly demonstrated the protective role of FVBM extract and F18 compound in risk factor induced cardiovascular disease.

Non-enzymatic glycation mediated advanced glycation end products (AGEs) generation results in the pathogenesis of diabetic complications and atherosclerotic cardiovascular disease (ASCVD) which is greatly influenced by 3-hydroxy-3-methyl-glutaryl Co-A reductase (HMG-R) activity. HMG-R inhibitors, statins, are well known for reducing mortality and morbidity of ASCVD in patients with diabetes due to their pleiotropic effects independent of cholesterol lowering. Due to distinct chemical structures, various statins may play important role in the inhibition of AGEs mediated pathologies. Herein, we evaluated the anti-glycating potential of atorvastatin (AT), rosuvastatin (RT), pitavastatin (PT), fluvastatin (FT), simvastatin (ST) alone as well in combination with ezetimibe (EZ) and tocotrienol (TT) against d-ribose mediated BSA and LDL glycation by various physicochemical approaches. Our data suggested that AT, TT, RT, EZ, EZ-AT, and EZ-RT were able to substantially inhibit the AGEs formation via modulation of hyperchromicity, fluorogenic AGEs, % contribution of α-helix and β-sheets to protein secondary structure, amide-I band stretching, carbonyl and HMF content in Gly-BSA as well as Gly-LDL. On the basis of above findings, we concluded that HMG-R inhibitors and TT, alone or in combination with EZ, may be established as terrific therapeutic agents for the patients suffering from AGEs induced diabetic cum ASCVD complications.

Gall bladder cancer (GBC) is a gastro-intestinal cancer with high prevalence among north Indian women. Platelet derived growth factor-B (PDGFB) and human epidermal growth factor receptor-2 (HER2) may play roles in the etiology of GBC through the inflammation-hyperplasia-dysplasia-carcinoma pathway. To study the association of PDGFB and HER2 polymorphisms with risk of GBC, 200 cases and 300 controls were considered. PDGFB +286AG and +1135AC polymorphisms were investigated with an amplification refractory mutation system and the HER2 Ile655Val polymorphism by restriction fragment length polymorphism. Significant risk associations for PDGFB +286 GG (OR=5.25) and PDGFB +1135 CC (OR=3.19) genotypes were observed for GBC. Gender wise stratification revealed susceptibility for recessive models of PDGFB +1135AC (OR=3.00) and HER2 Ile655Val (OR=2.52) polymorphisms among female GBC cases. GBC cases with gall stones were predisposed to homozygous +286 GG and +1135 CC genotypes. Significant risk associations were found for ACIle (OR=1.48), GAVal (OR=1.70), GAIle (OR=2.00) haplotypes with GBC cases and GCIle haplotype with female GBC cases (OR=10.37, P=0.0001). Pair-wise linkage disequilibrium revealed negative associations among variant alleles. On multi-dimensional reduction analysis, a three factor model revealed significant gene-gene interaction for PDGFB +286AG, PDGFB +1135AC and HER2 Ile165Val SNPs with GBC. Protein-protein interaction showed significant association of PDGFB and HER2 with the epidermal growth factor receptor signaling pathway.

In the present density functional studies, structural, mechanical and magneto-electronic properties of CaTaO3, SrTaO3 and BaTaO3 perovskites have been investigated. The calculated structural parameters by DFT and analytical methods are found consistent with the experiments. The analytically calculated tolerance factors of these compounds as well as their mechanical properties show that they are stable in the cubic phase. Furthermore elastic properties show that these materials are ductile in nature and confirm that BaTaO3 is harder than the rest compounds. The calculated spin dependent magneto-electronic properties reveal the paramagnetic metallic nature of these compounds. The electrical conductivity curve demonstrates significant conductivity above room temperature. On the basis of the presented properties it is expected that these compounds could be efficient electrode materials and need experimental investigations.

Glycation of biologically important macromolecules leads to the establishment of advanced glycation end products (AGEs) having significant role in the pathophysiology of various diseases. d-Ribose, is a highly reactive pentose sugar resulting in the rapid formation of AGEs. Formation of d-ribose derived glycated DNA and LDL has been previously demonstrated; however no comparative, extensive studies have been performed to assess the immunogenicity of d-ribose glycated calf thymus DNA (CT-DNA) and LDL. In the present study, the results showed that animals immunized with d-ribose modified CT-DNA and LDL induced antibodies as detected by direct binding and competition ELISA. The modified CT-DNA and LDL were found to be highly immunogenic, eliciting high titer immunogen-specific antibodies, while the native forms of DNA was almost non-immunogenic. The induced antibodies from modified CT-DNA and LDL exhibited wide range of heterogeneity in recognizing various nucleic acid conformers, DNA bases and amino acids. Furthermore, Serum antibodies from diabetes and diabetes atherosclerosis patients were screened for their binding to native CT-DNA, LDL and glycated CT-DNA, LDL. Glycated CT-DNA showed almost equivalent binding to both diabetes and diabetic atherosclerosis group while high recognition was observed when glycated LDL was used as an antigen.

Proprotein convertase subtilisin/kexin type 9 (PCSK-9) is a serine protease of the proprotien convertase (PC) family that has profound effects on plasma low density lipoprotein cholesterol (LDL-C) levels, the major risk factor for coronary heart disease (CHD), through its ability to mediate LDL receptor (LDL-R) protein degradation and reduced recycling to the surface of hepatocytes. Thus, the current study was premeditated not only to evaluate the role of lycopene in targeting the inhibition of PCSK-9 via modulation of genes involved in cholesterol homeostasis in HFD rats but also to examine a correlation between HFD induced inflammatory cascades and subsequent regulation of PCSK-9 expression. Besides the effect of lycopene on hepatic PCSK-9 gene expression, PPI studies for PCSK-9-Lycopene complex and EGF-A of LDL-R were also performed via molecular informatics approach to assess the dual mode of action of lycopene in LDL-R recycling and increased removal of circulatory LDL-C. We for the first time deciphered that lycopene treatment significantly down-regulates the expression of hepatic PCSK-9 and HMGR, whereas, hepatic LDL-R expression was significantly up-regulated. Furthermore, lycopene ameliorated inflammation stimulated expression of PCSK-9 via suppressing the expression of inflammatory markers. The results from our molecular informatics studies confirmed that lycopene, while occupying the active site of PCSK-9 crystal structure, reduces the affinity of PCSK-9 to complex with EGF-A of LDL-R, whereas, atorvastatin makes PCSK-9-EGF-A complex formation more feasible than both of PCSK-9-EGF-A alone and Lycopene-PCSK-9-EGF-A complex. Based on above results, it can be concluded that lycopene exhibits potent hypolipidemic activities via molecular mechanisms that are either identical (HMGR inhibition) or distinct from that of statins (down-regulation of PCSK-9 mRNA synthesis). To the best of our knowledge, this is the first report that lycopene has this specific biological property. Being a natural, safer and alternative therapeutic agent, lycopene could be used as a complete regulator of cholesterol homeostasis and ASCVD.

Seismic patterns in volcanoes give an important information on volcanic activity and volcano monitoring demands accurate and robust systems to cover the volume of data registered by the monitoring sensors. This is why research on automatic volcanic pattern recognition techniques is ongoing. One of the most popular approaches is to design classifiers to discriminate patterns according to specific features, extracted from the signals. Literature presents different features and classification tools to perform these systems, reaching good results mainly in off-line experiments. This article presents the preliminary results of a proposal to design of a classifying structure where the features are selected, based on the expertise of the seismologists of the Observatorio Vulcanologico de los Andes Sur (OVDAS), the national net of volcano monitoring of Chile. The methodology implemented a multi-class classification structure based on Support Vector Machines (SVM), trained with the set of selected features. The classification structure reaches a mean error of 2.4% for all classes, achieving a better result than those obtained in previous studies where other feature selection criteria were adopted. This work verifies that the expert knowledge is relevant to select the features used in the design of the classification structures.

Japanese encephalitis (JE) is an acute viral infection of the central nervous system where the JE virus infects the lumen of the endoplasmic reticulum (ER) and rapidly accumulates substantial amount of seven different nonstructural proteins (NS). These NS proteins tend to bind on a glycoprotein receptor, ribophorin (RPN) resulting in the malfunctioning of ER in host cells, subsequently triggering an unfolded protein response. Therefore, it is of interest to predict the best possible antigenic determinants in the NS protein capable of eliciting immune response as a strategy to combat JE. Hence, it is our interest to explore the most potent NS protein among all showing the best possible molecular interaction with the RPN receptor present on ER. However, the structures of these NS protein and RPN are currently unknown. Thus, we modeled their structures using the established homology modeling techniques in the MODELLER 9v10 software. The molecular docking of NS proteins with RPN was subsequently completed using the Discovery Studio 2.5 software suite. The docked conformations of RPN with NS were further analyzed and its graphical interpretations were presented for identifying the most potential NS protein for efficient epitope activity. Further, the B cell epitopes were mapped using BCPred and the predicted epitope regions are documented. The data presented in this report provides useful insights towards the design and development of potential epitopes to generate a vaccine candidate against JEV.

Glyoxal, methylglyoxal and 3-deoxyglucosones are reactive dicarbonyl compounds, which transform free amino groups of proteins and lipoproteins macromolecule into advanced glycation end-products (AGEs). AGEs play a significant role in the pathophysiology of aging and diabetic complications because of their genotoxic effect. Glyoxal also reacts with free amino group of nucleic acids resulting in the formation of DNA–AGEs. The present study reports the genotoxicity and immunogenicity of AGEs formed by Glyoxal–Arginine–Fe 3+ (G–Arg–Fe 3+ ) system as a glycating agent. Immunogenicity of native and G–Arg–Fe 3+ –DNA was probed in female rabbits. Immunofluorescence suggests the presence of immune complex deposition in the kidney section of immunized rabbits. Spectroscopic analysis and melting temperature indicates the structural modification in the human DNA. The modified human DNA is found to be highly immunogenic, whereas unmodified form was simply non-immunogenic. This study shows the presence of auto-antibodies against G–Arg–Fe 3+ modified human DNA in the sera of diabetes type 1 and in few cases type 2 patients due to secondary complications of nephropathy. The glyco-oxidative lesions have also been detected in the lymphocyte DNA isolated from patients having type 1 and type 2 diabetes. The results show structural perturbations generating new epitopes in G–Arg–Fe 3+ –DNA rendering it pretty immunogenic.

This study presents a novel approach to synthesize glycogenic gold nanoparticles (glycogenic GNps) capped with glycated products (Schiff's base, Heyns products, fructosylamine etc.). These glycogenic GNps have been found to be active against human osteosarcoma cell line (Saos-2) with an IC 50 of 0.187 mM, while the normal human embryonic lung cell line (L-132) remained unaffected up to 1 mM concentration. The size of glycogenic GNps can also be controlled by varying the time of incubation of gold solution. Glycation reactions involving a combination of fructose and HSA (Human Serum Albumin) were found to be effective in the reduction of gold to glycogenic GNps whereas glucose in combination with HSA did not result in the reduction of gold. The progress of the reaction was followed using UV–visible spectroscopy and NBT (Nitroblue tetrazolium) assay. The glycogenic GNps were found to be spherical in shape with an average size of 24.3 nm, in a stable emulsion. These GNps were characterized using UV–visible spectroscopy, zeta potential analysis, transmission electron microscopy (TEM) and scanning electron microscopy (SEM).

Aims Advanced glycation end products (AGEs) trigger intracellular reactive oxygen species (ROS) generation, activation of receptor for AGEs (RAGE) expression/functionality and RAGE-associated signalling pathways which influence the diabetic-cum-atherosclerotic complications, whereas, the atherosclerosis progression is greatly influenced by hepatic β-Hydroxy-β-methyl-glutaryl-Co-A reductase (HMG-R) activity. The present report was premeditated to uncover the regulatory role of HMG-R inhibitors and ezetimibe (EZ) in attenuating the LDL-AGEs-induced pathogenicity via targeting cellular-ROS and RAGE-associated signalling in HEK-293 cells. Main methods The MTT assay was used to assess either the cytotoxic or cytoprotective impact of each HMG-R inhibitors, EZ, and LDL-AGEs, whereas, quantification of ROS was performed by DCFDA method. The qRT-PCR was used to detect the mRNA level of RAGE, neuropilin-1 (NRP-1) and other RAGE-associated genes like MMP-2, NF-κB, and TGFβ-1. Key findings The HMG-R inhibitors do not exert any cytotoxicity in HEK-293 cells, whereas, and LDL-AGEs negatively affected the cell viability of HEK-293 cells. However, viability of LDL-AGEs-treated HEK-293was markedly retained after simultaneous treatment with our test inhibitors. Further, DCFDA staining showed that LDL-AGEs-induced ROS was also suppressed upon treatment with our test inhibitors in HEK-293 cells. qRT-PCR analysis reflected that these inhibitors suppress the RAGE, NF-κB, TGFβ-1, and MMP-2 expression, whereas, the NRP-1 was up-regulated by these compounds in LDL-AGEs-exposed HEK-293 cells. Significance The above pharmacological effects signify that HMG-R inhibitors and EZ (alone or in combination) may implied in the treatment of AGEs-induced oxidative stress and tissue damage in diabetic complications via targeting intracellular-ROS, NRP-1 functionality and RAGE-associated genes i.e. NF-κB, TGFβ-1, and MMP-2.

The present study is initially intended to evaluate antioxidant and β-hydroxy-β-methylglutaryl-CoA reductase (HMGR) inhibitory property of Ficus virens Ait., first by in vitro analyses followed by a corroboratory molecular informatics study. Our results show that of all the sequentially extracted fraction of F. virens bark and leaves extract, F. virens bark methanol extract exhibits strong radical scavenging, antioxidant and oxidative DNA damage protective activity, which is well correlated with its total phenolic content. In addition, F. virens bark methanol extract, which is non-cytotoxic, significantly and non-covalently inhibit the HMGR activity (IC50 = 3.45 ± 0.45 µg/ml) in comparison with other extracts. The mechanistic aspect of this inhibition activity is authenticated by molecular docking study of bioactive compounds as revealed from gas chromatography–mass spectrometry data, with HMGR. The analysis for the first time indicates that quinic acid (ΔG: −8.11 kcal/mol) and paravastatin (ΔG: −8.22 kcal/mol) exhibit almost same binding energy, while other compounds also showed good binding energy, suggesting that quinic acid alone or in combination with other major bioactive compound is probably responsible for HMGR inhibitory property of the extract and plausibly can be used in in vivo system for the management, prevention, and alleviation of hypercholesterolemia as well as hypercholesterolemia-induced oxidative stress. The non-enzymatic addition of carbonyl group of sugar moiety to the amino group of DNA macromolecule leads to the formation of early glycation products (Amadori products) which undergoes rearrangement, cyclization and dehydration to form advanced glycation end products (AGEs). While the formation of glucose derived glycated DNA has been previously demonstrated, however no extensive studies have been performed to assess the glycation of DNA using d -ribose as glycating agent. d -ribose, an important monosaccharide, is a highly reactive pentose sugar which results in the rapid formation of AGEs. To the best of our knowledge, this is the first study to characterize the d -ribose-induced changes in calf thymus DNA, as well as calf thymus DNA Amadori and calf thymus DNA-AGEs. The main objective of the study is to investigate the non-enzymatic glycation of calf thymus DNA by using different concentrations of d -ribose at increasing time period. The obtained Amadori products, AGEs were characterized with respect to the extent of DNA strand break and base modifications. Additionally, their nitroblue tetrazolium (NBT) reduction assay, absorbance, agarose gel electrophoresis, fluorescence, circular dichroism (CD) and thermal denaturation ( T m ) characteristics were extensively studied. We found significant changes in the modification of DNA and in AGE-specific fluorescence, using different concentration of modifiers ( d -ribose). The results provide the mechanistic insight of d -ribose induced glycation in calf thymus DNA. The bio-availability of d -ribose makes this carbonyl species quite reactive and damaging, therefore having direct implication in diabetes. This is the preliminary study done on d -ribose glycation and warrants further study to probe the role ribose-DNA glycation in different disease state including diabetes.

Production of free radicals is associated with cigarette smoke (CS) which in turn generates oxidative stress, could be responsible for alterations in the activities of enzymatic and non-enzymatic antioxidants that links with atherosclerosis. Therefore, the putative preventive effects of F. virens extract and its bioactive compound (F18), n-Octadecanyl-O-α-D-glucopyranosyl(6’→1’’)-O-α-D-glucopyranoside were investigated on overall enzyme and non-enzymatic defense system and in oxidative stress CS-exposed rats. The enzymatic activities of hepatic and lung CAT, SOD, Gred and GST in CS exposed rats were significantly decreased, while Gpx activity in CS exposed rats was increased. Similarly, hepatic and lung GSH content was reduced when compared to value of normal control group. Simultaneous administration of FVBM extract (50 and 100 mg/rat) and F18 bioactive compound (1 mg/rat) significantly increases hepatic and lung CAT, SOD, Gred and GST activity as well GSH concentration coupled with decrease in Gpx level in CS-exposed stress rats. Moreover, our histological observations concludes the pulmonary congestion, thickening of interalveolar septa and foci of collapsed alveoli with subsequent dilation of the adjoining alveolar spaces as well as development of large irregular spaces in rats lung exposed to cigarette smoke. Similarly, the liver also showed morphological alterations with congestion in central vein, portal inflammation and necrosis in CS-exposed rats. These morphological changes reversed significantly after treatment with FVBM extract and F18 compound. Thus biochemical and histopathological studies suggested that, FVBM extract and F18 showed its protective nature against CS-exposed rats.

Genomic DNA represents the total genome of an organism and is valuable for different molecular studies. The isolation of high quality DNA is a prerequisite for these molecular techniques. The seeds of apple (Malusdomestica) contain high amount of polysaccharides, polyphenols and other secondary metabolites that can hamper DNA isolation, amplification, restriction digestion and subsequent molecular studies. Here, we report a simple, inexpensive and an effi- cient protocol for the isolation of high quality genomic DNA from seeds of different apple cultivars for random amplified polymorphic DNA (RAPD) analysis. This procedure purifies greater amounts of DNA which can be amplified via PCR or digested with endonucleases. The yield of DNA was 200-300 ng/μL (total 8-12 μg DNA from 1-2 apple seeds). The sim- plicity of the procedure makes it very practical for DNA extraction.

This study initially aimed to depict the molecular rationale evolving the role of lycopene in inhibiting the enzymatic activity of β-hydroxy-β-methylglutaryl-CoA (HMG-CoA) reductase via in vitro and in silico analysis. Our results illustrated that lycopene exhibited strong HMG-CoA reductase inhibitory activity (IC50 value of 36 ng/ml) quite better than pravastatin (IC50 = 42 ng/ml) and strong DPPH free radical scavenging activity (IC50 value = 4.57 ± 0.23 μg/ml) as compared to ascorbic acid (IC50 value = 9.82 ± 0.42 μg/ml). Moreover, the Ki value of lycopene (36 ng/ml) depicted via Dixon plot was well concurred with an IC50 value of 36 ± 1.8 ng/ml. Moreover, molecular informatics study showed that lycopene exhibited binding energy of −5.62 kcal/mol indicating high affinity for HMG-CoA reductase than HMG-CoA (ΔG: −5.34 kcal/mol). Thus, in silico data clearly demonstrate and support the in vitro results that lycopene competitively inhibit HMG-CoA reductase activity by binding at the hydrophobic portion of HMG-CoA reductase.
