This study aimed to determine the impact of various pressures on the functional composition of OCH (Odonata, Coleoptera, and Heteroptera) in streams within the northwest Rif region of Morocco. We examined how OCH traits respond to human-induced pressures in selected stream sites in Morocco’s northwestern Rif region. OCH specimens were collected from 36 sites using a Surber sampler with dimension of 20 × 20 cm and mesh size of 500 µm over the course of two years, from 2021 to 2023. We measured physico-chemical and hydraulic parameters such as temperature, pH, DO, and NO-3. Sixty-seven trait attributes from 11 trait classes were assigned to the collected OCH taxa at the family level. Following the delineation of sites along the gradient of impacts in the study area, we categorized 7 sites as slightly impacted sites (SISs), 19 sites as moderately impacted sites (MISs), and 10 sites as heavily impacted sites (HISs). We successfully identified and categorized the traits as either vulnerable or tolerant based on RLQ models. Traits such as reproductive cycles per year and tegument respiration, which were positively correlated with SISs in the RLQ model and also positively correlated with depth and chlorine, were identified as vulnerable traits. Crawling locomotion and full water swimming were identified as tolerant traits. The distribution patterns of the OCH taxa revealed a robust correlation between the taxa and the sampling sites. Notably, taxa such as Nepidae, Naucoridae, and Corixidae exhibited widespread distribution and a strong association with the SISs. On the other hand, traits related to living macroinvertebrate food sources and reproduction in vegetation, specifically clutches, exhibited a negative correlation with total dissolved solids. Incorporating OCH functional traits into biomonitoring programs allows for a more comprehensive assessment of river ecosystems. This approach provides a nuanced understanding of how different stressors impact the community composition and overall ecological health.

A new species belonging to the genus Habrophlebia Eaton, 1881 is described at the nymphal stage from the Rif Mountains of Morocco. This species presents unique features, such as the chorionic arrangement of the egg and the ornamentation of the posterior margin of abdominal tergites. It is compared to all west European Habrophlebia species and a table with discriminating characters is given. A phylogenetic reconstruction based on COI sequences fully supports the hypothesis of a new species in the Rif Mountains, with possible further distribution in southern Spain.

ABSTRACT Helicobacter pylori is a bacterium that exhibits strict host restriction to humans and non-human primates, and the bacterium is widely acknowledged as a significant etiological factor in the development of chronic gastritis, peptic ulcers, and gastric cancers. The pathogenic potential of this organism lies in its adeptness at colonizing the gastric mucosa, which is facilitated by a diverse repertoire of virulence factors, including adhesins that promote the attachment of the bacteria to the gastric epithelium. Among these adhesins, HpaA stands out due to its conserved nature and pivotal role in establishing H. pylori colonization. Moreover, this lipoprotein holds promise as an antigen for the development of effective H. pylori vaccines, thus attracting considerable attention for in-depth investigations into its molecular function and identification of binding determinants. Here, we present the elucidation of the crystallographic structure of HpaA at 2.9 Å resolution. The folding adopts an elongated protein shape, which is distinctive to the Helicobacteraceae family, and features an apical domain extension that plays a critical role in the cell-adhesion activity on gastric epithelial cells. Our study also demonstrates the ability of HpaA to induce TNF-α expression in macrophages, highlighting a novel role as an immunoregulatory effector promoting the pro-inflammatory response in vitro. These findings not only contribute to a deeper comprehension of the multifaceted role of HpaA in H. pylori pathogenesis but also establish a fundamental basis for the design and development of structure-based derivatives, aimed at enhancing the efficacy of H. pylori vaccines.IMPORTANCEHelicobacter pylori is a bacterium that can cause chronic gastritis, peptic ulcers, and gastric cancers. The bacterium adheres to the lining of the stomach using proteins called adhesins. One of these proteins, HpaA, is particularly important for H. pylori colonization and is considered a promising vaccine candidate against H. pylori infections. In this work, we determined the atomic structure of HpaA, identifying a characteristic protein fold to the Helicobacter family and delineating specific amino acids that are crucial to support the attachment to the gastric cells. Additionally, we discovered that HpaA can trigger the production of TNF-α, a proinflammatory molecule, in macrophages. These findings provide valuable insights into how H. pylori causes disease and suggest that HpaA has a dual role in both attachment and immune activation. This knowledge could contribute to the development of improved vaccine strategies for preventing H. pylori infections.

Abstract SARS-CoV-2 subunit vaccines continue to be the focus of intense clinical development worldwide. Protein antigens in these vaccines most commonly consist of the spike ectodomain fused to a heterologous trimerization sequence, designed to mimic the compact, prefusion conformation of the spike on the virus surface. Since 2020, we have produced dozens of such constructs in CHO cells, consisting of spike variants with different mutations fused to different trimerization sequences. This set of constructs displayed notable conformational heterogeneity, with two distinct trimer species consistently detected by analytical size exclusion chromatography. A recent report showed that spike ectodomain fusion constructs can adopt an alternative trimer conformation consisting of loosely associated ectodomain protomers. Here, we applied multiple biophysical and immunological techniques to demonstrate that this alternative conformation is formed to a significant extent by several SARS-CoV-2 variant spike proteins. We have also examined the influence of temperature and pH, which can induce inter-conversion of the two forms. The substantial structural differences between these trimer types may impact their performance as vaccine antigens.

Cloeon vanharteni Gattolliat & Sartori, 2008 was newly discovered in the framework of our study of Ephemeroptera in the Draa basin, located in the southern region of the High Atlas in Morocco. This discovery is rather unexpected as the species was never reported outside the Arabian Peninsula and Levant; it is thus the first record for the Maghreb. The identification was based on morphological evidence and confirmed by the mitochondrial COI barcode.

Baetis rifensis sp. nov. is the first representative of the Baetis fuscatus group to be described from the Maghreb. It was collected from streams in the Rif region of northern Morocco. All species of the B. fuscatus group are morphologically very similar, with slight differences in colour. Thus, in addition to morphological description, species delimitation based on genetic evidence was carried out. The new species was compared with other members of the B. fuscatus group from the Palaearctic region.

A new species of Alainites is described from northern of Morocco Alainites albai sp. nov. It can be separated from the other west Palearctic species by the gill number, the spination of the distal margin of tergites, the leg setation, and the paraproct shape and spination. This species is widespread in the study area but never abundant. It prefers small to medium streams with slow flow, and does not seem to be very sensitive to pollution and water logging activities.

This study aimed to evaluate the effects of the Martil River rehabilitation project and recently constructed dam infrastructures to reduce flood risks and to promote local socio-economic development on the ecological integrity of the river. The assessment focused on changes in fluvial landforms over time and the evaluation of aquatic ecosystems based on six physicochemical parameters (temperature, pH, electrical conductivity, dissolved oxygen, biochemical oxygen demand, and chemical oxygen demand), morpho-hydrological variables (stream width, water depth, and current speed), habitat indices (QBR, IHF, and MQI), and macroinvertebrate assemblages of EPT, OCH, and Chironomidae (Diptera) at five stations from autumn 2015 to spring 2018 (prior to and during the rehabilitation actions). The results showed that the river rehabilitation project led to profound changes in Martil River’s ecosystem and water quality over time. Physicochemical and habitat measurements at the rehabilitated sites revealed a major change in macroinvertebrate communities due to changes in fluvial landforms in relation to flow-sediment regimes. As a result, some typical species of lentic habitats disappeared, while alien, opportunistic, and lotic species appeared.

The involvement of trait-based approaches is crucial for understanding spatial patterns, energy flow and matter transfer in running water systems, which requires consistent knowledge of the functional structures of aquatic communities, with the advantage of combining physical properties and behavioral mechanisms of food acquisition rather than the taxonomic group. The present study indicated how functional feeding groups may be used as a proxy for classical taxonomic evaluation, as well as the potential interest in incorporating them as indicators of anthropogenic stressors. The composition and abundance of the functional feeding groups of aquatic insects were examined from September 2021 to August 2022 along the Western Rif Region.Benthic samples were collected from nine sampling points in the studied area using a Surber sampler with a mesh size of 500 µm and a diameter of 20*20 cm. The stations included in this work were chosen for their accessibility as well as their position on the hydrographic systems. The abundance of sampled aquatic organisms in the whole study area revealed 5,342 individuals belonging to 60 families and seven orders of aquatic insects, classified into five feeding functional groups. In terms of abundance, Collector-gatherers (Ephemeroptera and Diptera) were the most abundant trophic group at most of the sites, with a proportion of 38.47%. Predators (Coleoptera, Hemiptera and Odonata) were the second group at all sites, followed by Collector-filters, accounting for 39.53%, 28.14% and 22.37% respectively, while Scarpers and Shredders had the lowest representation across all sites with 4.16%. The high number of registered Collectors could be related to their ability to feed on a diverse range of food items compared to the remaining trophic guilds. According to the Canonical Correspondence Analysis results, physicochemical (i.e. T, pH, BOD5, Cl- and NO3-) and hydromorphological (i.e. current velocity and depth) variables were amongst the key predictors of shaping the functional structure of aquatic biota during this investigation. It is highly recommended to carry out suitable measures to largely attenuate anthropogenic pressures in order to preserve the integrity of freshwater bodies and their biota.

The Draa river basin is located south of the High Atlas Mountain rangee in Morocco. The Ephemeroptera (mayflies) fauna of its three sub-basins: the High, Middle, and Lower Draa are poorly known. This study contributes to the knowledge of Ephemeroptera and its distribution in relation to environmental parameters in this area. The larvae of Ephemeropteran species were collected during six field campaigns in 17 sites using a Surber sampler. A total of thirteen species belonging to ten genera and five families were identified. among which seven taxa were recorded for the first time in the southern Atlas area: Baetis maurus, Baetis rhodani, Cheleocloeon dimorphicum, Cloeon simile, Procloeon stagnicola, Labiobaetis neglectus and Oligoneuriopsis skhounate. The limits of distribution of most species can be explained by the increase in water temperature and electrical conductivity especially at lower altitudes.

Among mayflies, Baetidae are often considered as easy to recognise at the family level, but difficult to identify at lower level. In several faunistic or ecological studies, the identification remains at the family level; Baetidae are generally considered as widespread and ubiquitous, therefore as poorly informative for ecological studies or bioassessments. Here, a straightforward identification key is offered to larvae of the ten genera of Baetidae reported from Maghreb based on easily observable and understandable characters. The diversity, ecology, and distribution of each taxonomic unit (genera or subgenera) are discussed and the main difficulties for deeper identification are pointed out. Future challenges and remaining taxonomic riddles for Maghrebian Baetidae are detailed.

Based on recently collected larvae from Algeria and Morocco, the species delimitation within the genus Centroptilum Eaton, 1869 in that region is validated. Two new species are described and illustrated, one from north-eastern Algeria, and one from North Morocco, using an integrated approach with morphological and molecular evidence. A table summarising the morphological differences between the new species and Centroptilum luteolum (Müller, 1776) from Central Europe is provided. Further, molecular evidence for additional undescribed species of Centroptilum in other regions of the West Palearctic is provided and discussed.

We describe a new species of Prosopistoma collected in the High Atlas Mountains of Morocco. Prosopistoma maroccanum sp. nov. appears to be morphologically more similar to the European highly endangered P. pennigerum (Müller, 1785) than to the other Maghrebian species, P. alaini Bojkova & Soldán, 2015. A gene tree including the few available barcode sequences of Palearctic Prosopistoma specimens is provided. Possible affinities with West African species are also discussed.

Background. During the recent years, appropriate attention has been paid to the oxidative stress which damages the body’s cells, proteins, and DNA. Therefore, the need of antioxidants becomes a therapeutic and preventive priority. In addition, microbial infections also constitute a public health problem. Objective. To find efficient, reliable and safe alternatives sources to synthetic antioxidants, antibiotics and antifungals drugs. Materials and methods. Extract and essential oil of Psidium guajava were screened for their antioxidant and antimicrobial activities against gram positive bacteria (Enterococcus faecalis and Staphylococcus aureus), gram negative bacteria (Citrobacter freundii, Escherichia coli and Pseudomonas sp) and fungi (Candida albicans,Candida tropicalis and Cryptococcus neoformans), as well as to determine the functional groups of phytochemicals present in the essential oil by Fourier transform infrared spectroscopy (FTIR). Results. The results indicate that P. guajava leaves extract demonstrated very high antioxidant activity and P.guajava essential oil showed the highest polyphenols content. The antioxidant capacity showed a significant negative linear correlation to total polyphenolic content (TPC) with Pearson’s correlation coefficients. P. guajava essential oil shows high antibacterial and antifungal activity against all the studied bacteria and fungi. The FTIR analysis of P. guajava essential oil showed the presence of several functional groups (ethers, esters, ketones, terpenes, alkanes, aldehydes, aromatic hydrocarbons, alcohols, and phenols). The relationship between the chemical composition and antimicrobial activity of P.guajava essential oil suggests that the attribution of its antimicrobial activity to a particular compound or a synergistic effects between its different constituents remains difficult. Conclusions. The present study demonstrated that Psidium guajava is a valuable source of active compounds with antioxidant and antimicrobial activities. This finding suggests the new use of the fruits and the leaves extracts of this plant in the treatment of bacterial and fungal infections, as well as for the extraction of new antioxidants. Therefore, it is necessary to be carried out in another study to identify the active(s) compound(s) in P.guajava essential oil with respect to their mechanisms and synergistic actions.

La dégradation accélérée des forêts est un défi majeur pour les gestionnaires. Le reboisement constitue alors une solution importante pour l’amélioration des services écosystémiques voire la survie des écosystèmes. Comparée à d’autres essences comme l’Eucalyptus, les pins renferment une biodiversité plus élevée. Nous avons étudié la phytodiversité d’un périmètre de reboisement avec quatre espèces et deux variétés de pins (Pinus halepensis, P. pinaster var. maghrebiana et var iberica, P. pinea et P. canariensis) afin de proposer l’essence la plus convenable au reboisement. Sur les 38 relevés floristiques réalisés, 83 espèces ont été recensées appartenant à 34 familles. Les valeurs de l’indice de diversité étaient maximales dans les plantations de P. halepensis et P. pinea et minimales dans celles de P. canariensis. L’ordination multidimensionnel non métrique et la Classification Ascendante Hiérarchique ont montré trois groupes : groupe à P. halepensis et P. pinea, groupe à P. maghrebiana et Quercus suber et groupe à P. iberica et P. canariensis. P. halepensis et P. pinea ont montré un cortège floristique relativement riche mais avec un appauvrissement structurel comparé à la subéraie originale. Il ressort que le reboisement à base de P. maghrebiana et dans une deuxième place P. halepensis améliore la structure des peuplements et présente un potentiel important pour la gestion des reboisements au nord du Maroc.

Recent research in various Moroccan areas allowed an update and a revision of the Moroccan Ephemeroptera checklist. In this case, 54 species are now listed, belonging to 10 families and 26 genera. The distribution of all studied species is discussed, as well as their biogeographical affinities. Moroccan Mayflies are characterized by a clear dominance of Mediterranean elements with a strong rate of endemism (33.4%).

Procambarus clarkii, one of the most invasive aquatic species in the world, is present in the Mediterranean estuarine regions. The invasion of this species in these wetlands has a multitude of impacts, including the impact on flora. This study examines the floristic diversity and abundance of Procambarus clarkii in this wetland, as well as whether the presence of this aquatic species affects the flora. The Procambarus clarkii sampling lasted three years (2016, 2017, 2018) in the Merja zerga wetland, using fishermen’s boats. Vegetation sampling was conducted during the same Procambarus clarkii capture period to assess the abundance of Procambarus clarkii at the same time and recover the effect of this aquatic species on the flora. To strengthen our study a survey was conducted among sinners to assess the impacts of Procambarus clarkii in its living environment. The results showed that there was a significant difference in the abundance of Procambarus clarkii sampled over the years (2016, 2017, 2018). The floristic diversity of the Merja zerga lagoon is very important. The effect of Procambarus clarkii on the flora was very remarkable. Thus, the floristic species exposed to this impact are the species of genus Potamogéton. These results show that Procambarus clarkii can be considered as a factor contributing to the decline of the floristic diversity of the aquatic areas where it lives.

Protein kinases are promising drug targets for treatment of malaria. Here, starting with a medicinal chemistry approach, Baker et al. generate an imidazopyridine that selectively targets Plasmodium falciparum PKG, inhibits blood stage parasite growth in vitro and in mice and blocks transmission to mosquitoes.

In 2010 the identities of thousands of anti-Plasmodium compounds were released publicly to facilitate malaria drug development. Understanding these compounds' mechanisms of action--i.e., the specific molecular targets by which they kill the parasite--would further facilitate the drug development process. Given that kinases are promising anti-malaria targets, we screened ~14,000 cell-active compounds for activity against five different protein kinases. Collections of cell-active compounds from GlaxoSmithKline (the ~13,000-compound Tres Cantos Antimalarial Set, or TCAMS), St. Jude Children's Research Hospital (260 compounds), and the Medicines for Malaria Venture (the 400-compound Malaria Box) were screened in biochemical assays of Plasmodium falciparum calcium-dependent protein kinases 1 and 4 (CDPK1 and CDPK4), mitogen-associated protein kinase 2 (MAPK2/MAP2), protein kinase 6 (PK6), and protein kinase 7 (PK7). Novel potent inhibitors (IC50 < 1 μM) were discovered for three of the kinases: CDPK1, CDPK4, and PK6. The PK6 inhibitors are the most potent yet discovered for this enzyme and deserve further scrutiny. Additionally, kinome-wide competition assays revealed a compound that inhibits CDPK4 with few effects on ~150 human kinases, and several related compounds that inhibit CDPK1 and CDPK4 yet have limited cytotoxicity to human (HepG2) cells. Our data suggest that inhibiting multiple Plasmodium kinase targets without harming human cells is challenging but feasible.

Dominant optic neuropathies causing fiber loss in the optic nerve are among the most frequent inherited mitochondrial diseases. In most genetically resolved cases, the disease is associated to a mutation in OPA1, which encodes an inner mitochondrial dynamin involved in network fusion, cristae structure and mitochondrial genome maintenance. OPA1 cleavage is regulated by two m-AAA proteases, SPG7 and AFG3L2, which are respectively involved in Spastic Paraplegia 7 and Spino-Cerebellar Ataxia 28. Here, we identified a novel mutation c.1402C>T in AFG3L2, modifying the arginine 468 in cysteine in an evolutionary highly conserved arginine-finger motif, in a family with optic atrophy and mild intellectual disability. Ophthalmic examinations disclosed a loss of retinal nerve fibers on the temporal and nasal sides of the optic disk and a red-green dyschromatopsia. Thus, our results suggest that neuro-ophthalmological symptom as optic atrophy might be associated with AFG3L2 mutations, and should prompt the screening of this gene in patients with isolated and syndromic inherited optic neuropathies.

A 3.3 MDa macromolecular cage between two Escherichia coli proteins with seemingly incompatible symmetries–the hexameric AAA+ ATPase RavA and the decameric inducible lysine decarboxylase LdcI–is reconstructed by cryo-electron microscopy to 11 Å resolution. Combined with a 7.5 Å resolution reconstruction of the minimal complex between LdcI and the LdcI-binding domain of RavA, and the previously solved crystal structures of the individual components, this work enables to build a reliable pseudoatomic model of this unusual architecture and to identify conformational rearrangements and specific elements essential for complex formation. The design of the cage created via lateral interactions between five RavA rings is unique for the diverse AAA+ ATPase superfamily.

Background: Induction of labor (IOL) in nulliparas with premature rupture of membranes (PROM) and an unfavorable cervix at term poses challenges. Our study sought to investigate the impact of prostaglandin E2 (PGE2) compared to oxytocin on the duration of IOL in this specific group of parturients. Methods : This was retrospective matched-case study. All nulliparas with term PROM who underwent induction between January 2006 to April 2023 at Shaare Zedek Medical Center were identified. Cases induced by either PGE2 or oxytocin were matched by the following criteria: (1) time from PROM to IOL; (2) modified Bishop score prior to IOL ≤ 5; (3) newborn birthweight; and (4) vertex position. The primary outcome was time from IOL to delivery. Results: Ninety-five matched cases were identified. All had a modified Bishop score ≤ 5. Maternal age (26 ± 4.7 years old, p = 0.203) and gestational age at delivery (38.6 ± 0.6, p = 0.701) were similar between the groups. Matched factors including time from PROM to IOL (23.5 ± 19.2 versus 24.3 ± 21.4 p = 0.780), birth weight of the newborn (3111 g versus 3101 g, p = 0.842), and occiput anterior position (present on 98% in both groups p = 0.687) were similar. Time from IOL to delivery was significantly shorter by 3 h and 36 min in the group induced with oxytocin than in the group induced with PGE2 ( p = 0.025). Within 24 h, 55 (58%) of those induced with PGE2 delivered, compared to 72 (76%) of those induced with oxytocin, ( p = 0.033). The cesarean delivery rates [18 (19%) versus 17 (18%)], blood transfusion rates [2 (2%) versus 3 (3%)], and Apgar scores (8.8 versus 8.9) were similar between the groups (PGE2 versus oxytocin, respectively), p ≥ 0.387. Conclusions: Induction with oxytocin, among nulliparas with term PROM and an unfavorable cervix, was associated with a shorter time from IOL to delivery and a higher rate of vaginal delivery within 24 h, with no difference in short-term maternal or neonatal adverse outcomes.
