The auxin-inducible degron (AID) system has emerged as a powerful tool to conditionally deplete proteins in a range of organisms and cell types. Here, we describe a toolkit to augment the use of the AID system in Caenorhabditis elegans. We have generated a set of single-copy, tissue-specific (germline, intestine, neuron, muscle, pharynx, hypodermis, seam cell, anchor cell) and pan-somatic TIR1-expressing strains carrying a co-expressed blue fluorescent reporter to enable use of both red and green channels in experiments. These transgenes are inserted into commonly used, well-characterized genetic loci. We confirmed that our TIR1-expressing strains produce the expected depletion phenotype for several nuclear and cytoplasmic AID-tagged endogenous substrates. We have also constructed a set of plasmids for constructing repair templates to generate fluorescent protein::AID fusions through CRISPR/Cas9-mediated genome editing. These plasmids are compatible with commonly used genome editing approaches in the C. elegans community (Gibson or SapTrap assembly of plasmid repair templates or PCR-derived linear repair templates). Together these reagents will complement existing TIR1 strains and facilitate rapid and high-throughput fluorescent protein::AID tagging of genes. This battery of new TIR1-expressing strains and modular, efficient cloning vectors serves as a platform for straightforward assembly of CRISPR/Cas9 repair templates for conditional protein depletion.

La prevención de caídas, fuente principal de lesiones y mortalidad en adultos, se puede mejorar por medio de una adecuada evaluación del equilibrio. No obstante, el alto costo y complejidad de las herramientas actuales limitan su aplicación, particularmente en clínicas de atención primaria. Surge así la necesidad de metodologías más accesibles y económicas para optimizar el cuidado de salud en este segmento poblacional. En el presente artículo se introduce un estudio que explora la aplicabilidad de una versión mejorada de la Wii Balance Board (WBBm). Este dispositivo, de bajo costo y portabilidad, ha sido respaldado por diversos investigadores en términos de validez. La muestra consistió en 95 adultos, con edades comprendidas entre 40 y 85 años, quienes fueron evaluados con la WBBm con el fin de correlacionar mediciones del Centro de Presión (CoP) con parámetros clínicos y antropométricos. La investigación reveló que la WBBm proporciona señales efectivas para una evaluación cuantitativa del equilibrio y su uso es intuitivo y expedito. Sin embargo, se destaca que la interpretación del CoP está influenciada por múltiples variables clínicas y antropométricas. Esto sugiere la necesidad de cautela al generalizar la aplicabilidad de estas técnicas en una población diversa. Para concluir, aunque queda camino por recorrer en esta área de investigación, la WBBm emerge como una herramienta con un alto potencial. Su adaptabilidad y practicidad pueden facilitar una valoración más eficiente del equilibrio en entornos clínicos de atención primaria.

In sexually reproducing metazoans, spermatogenesis is the process by which uncommitted germ cells give rise to haploid sperm. Work in model systems has revealed mechanisms controlling commitment to the sperm fate, but how this fate is subsequently executed remains less clear. While studying the well-established role of the conserved nuclear hormone receptor transcription factor, NHR-23/NR1F1, in regulating C. elegans molting, we discovered that NHR-23/NR1F1 is also constitutively expressed in developing primary spermatocytes and is a critical regulator of spermatogenesis. In this novel role, NHR-23/NR1F1 functions downstream of the canonical sex-determination pathway. Degron-mediated depletion of NHR-23/NR1F1 within hermaphrodite or male germlines causes sterility due to an absence of functional sperm, as depleted animals produce arrested primary spermatocytes rather than haploid sperm. These spermatocytes arrest in prometaphase I and fail to either progress to anaphase or attempt spermatid-residual body partitioning. They make sperm-specific membranous organelles but fail to assemble their major sperm protein into fibrous bodies. NHR-23/NR1F1 appears to function independently of the known SPE-44 gene regulatory network, revealing the existence of an NHR-23/NR1F1-mediated module that regulates the spermatogenesis program.

BackgroundPublic health programs are tasked with educating the community on health topics, but it is unclear whether these programs are acceptable to learners. Currently, these programs are delivered via a variety of platforms including in-person, virtually, and over the telephone. Sickle cell trait (SCT) education for parents of children with this trait is one of many education programs provided by the Ohio Department of Health. The novel SCTaware videoconference education program was developed by a research team after central Ohio’s standard program transitioned from in-person to telephone-only education during the COVID-19 pandemic. ObjectiveOur objectives were to investigate the acceptability of the format and engagement with the SCTaware education and assess parental worry about having a child with SCT before and after receiving SCTaware. Introduction: This paper presents an initial proposal for a formal framework that, by studying the metric variables involved in information retrieval, can establish the sequence of events involved and how to perform it. Method: A systematic approach from the equations of Shannon and Weaver to establish the decidability of information retrieval systems and Conrad's equation is used to interweave the ecosystem components. Analysis: This work was developed from a detailed analysis of the scientific literature on information retrieval and through a set of inductive processes; it has been possible to build each of the components of this proposal. Results: First, we have proposed a simple way of assessing the implications of the sequence of events that occur in a search related to the overall response. Secondly, we obtain a formal equation that determines all the interrelated human and technological elements in the information retrieval processes. Conclusions: The establishment of this formal framework allows us to ascertain why we evaluate information retrieval and how one has to intervene in times of imbalance. (Contains 3 figures.)

Introduction: This paper presents an analysis of the Simple Knowledge Organization System (SKOS) compared with other alternatives for thesaurus representation in the Semantic Web. Method: Based on functional and structural changes of thesauri, provides an overview of the current context in which lexical paradigm is abandoned in favour of the conceptual paradigm. Likewise are described briefly various initiatives for the representation of thesauri using RDF vocabularies and hence for application to the Semantic Web, with particular attention to SKOS. Analysis and Results: This brief comparison will allow us to know some highlights of these proposals to raise those transcendental aspects in a model representing thesauri. SKOS includes the main proposal of this model, which organizes the concepts into diagrams and libraries. The notions of descriptors and non-descriptors terms are replaced by the association to the concepts of preferred and alternative labels and may be defined hierarchical or associative relationships between concepts. SKOS also provides for the establishment of correspondence relations between concepts belonging to different conceptual schemes. Conclusions: In accordance with the extent to which this series of requirements is fulfilled, we propose and defend the SKOS option as the best alternative for the development of multiple applications focusing on the use of thesauri in Web-based information services and systems, highlighting the advantages of this model over others proposed and in consideration of the user perspective in management, search and information query operations.
