Vegetation greening has long been acknowledged, but recent studies have pointed out that vegetation greening is possibly stalled or even reversed. However, detailed analyses about greening reversal or increased browning of vegetation remain scarce. In this study, we utilized the normalized difference vegetation index (NDVI) as an indicator of vegetation to investigate the trends of vegetation greening and browning (monotonic, interruption, and reversal) through the breaks for the additive season and trend (BFAST) method across China’s drylands from 1982 to 2022. It also reveals the impacts of ecological restoration programs (ERPs) and climate change on these vegetation trends. We find that the vegetation displays an obvious pattern of east-greening and west-browning in China’s drylands. Greening trends mainly exhibits monotonic greening (29.8 %) and greening with setback (36.8 %), whereas browning shows a greening to browning reversal (19.2 %). The increase rate of greening to browning reversal is 0.0342/yr, which is apparently greater than that of greening with setback, 0.0078/yr. This research highlights that, under the background of widespread vegetation greening, vegetation browning is progressively increasing due to the effects of climate change. Furthermore, the ERPs have significantly increased vegetation coverage, with the increase rate in 2000–2022 being twice as much as that of 1982–1999 in revegetation regions. Vegetation browning in southwestern Qingzang Plateau is primarily driven by adverse climatic factors and anthropogenic disturbances, which offset the efforts of ERPs.

Abstract Electrocatalytic alkynes semi-hydrogenation to produce alkenes with high yield and Faradaic efficiency remains technically challenging because of kinetically favorable hydrogen evolution reaction and over-hydrogenation. Here, we propose a hierarchically nanoporous Cu50Au50 alloy to improve electrocatalytic performance toward semi-hydrogenation of alkynes. Using Operando X-ray absorption spectroscopy and density functional theory calculations, we find that Au modulate the electronic structure of Cu, which could intrinsically inhibit the combination of H* to form H2 and weaken alkene adsorption, thus promoting alkyne semi-hydrogenation and hampering alkene over-hydrogenation. Finite element method simulations and experimental results unveil that hierarchically nanoporous catalysts induce a local microenvironment with abundant K+ cations by enhancing the electric field within the nanopore, accelerating water electrolysis to form more H*, thereby promoting the conversion of alkynes. As a result, the nanoporous Cu50Au50 electrocatalyst achieves highly efficient electrocatalytic semi-hydrogenation of alkynes with 94% conversion, 100% selectivity, and a 92% Faradaic efficiency over wide potential window. This work provides a general guidance of the rational design for high-performance electrocatalytic transfer semi-hydrogenation catalysts.

Abstract Background DNA methylation contributes to the epigenetic regulation of nuclear gene expression, and is associated with plant growth, development, and stress responses. Compelling evidence has emerged that long non-coding RNA (lncRNA) regulates DNA methylation. Previous genetic and physiological evidence indicates that lncRNA-CRIR1 plays a positive role in the responses of cassava plants to cold stress. However, it is unclear whether global DNA methylation changes with CRIR1-promoted cold tolerance. Results In this study, a comprehensive comparative analysis of DNA methylation and transcriptome profiles was performed to reveal the gene expression and epigenetic dynamics after CRIR1 overexpression. Compared with the wild-type plants, CRIR1-overexpressing plants present gained DNA methylation in over 37,000 genomic regions and lost DNA methylation in about 16,000 genomic regions, indicating a global decrease in DNA methylation after CRIR1 overexpression. Declining DNA methylation is not correlated with decreased/increased expression of the DNA methylase/demethylase genes, but is associated with increased transcripts of a few transcription factors, chlorophyll metabolism and photosynthesis-related genes, which could contribute to the CRIR1-promoted cold tolerance. Conclusions In summary, a first set of transcriptome and epigenome data was integrated in this study to reveal the gene expression and epigenetic dynamics after CRIR1 overexpression, with the identification of several TFs, chlorophyll metabolism and photosynthesis-related genes that may be involved in CRIR1-promoted cold tolerance. Therefore, our study has provided valuable data for the systematic study of molecular insights for plant cold stress response.

Abstract Background Problematic Internet Use (PIU), characterized by failures to control the overuse of internet, is associated with a range of functional impairments. However, there is limited research on the specific impact of PIU on inhibitory control functions, particularly in terms of differentiating between prepotent response inhibition and interference control. Therefore, the main objective of this study is to investigate these two components of inhibitory control in individuals with PIU. Methods Thirty participants who met the PIU criteria and 30 control participants were included in the present study. All participants completed the Go/No-Go and Flanker tasks, in which internet-related images and words were used as task stimuli. Results In the Go/No-Go task, all participants exhibited poorer performance in inhibiting internet-related stimuli compared to internet-unrelated stimuli, during the No-Go trials. In the Flanker task, results revealed a three-way interaction of Group, Stimulus type and Congruency. Specifically, in the incongruent condition, participants with PIU exhibited slower responses for internet-unrelated targets compared to internet-related targets, whereas no similar effect was observed among individuals with low internet use. Conclusions The findings suggest that difficulties in controlling the interference effect of internet-related information represent a key dysfunction in inhibitory control of PIU.

Learning, an important activity for both human and animals, has long been a focal point of research. During the learning process, subjects assimilate not only their own information but also information from others, a phenomenon known as social learning. While numerous studies have explored the impact of social feedback as a reward/punishment during learning, few studies have investigated whether social feedback facilitates or inhibits the learning of environmental rewards/punishments. This study aims to test the effects of social feedback on economic feedback and its cognitive processes by using the Iowa Gambling Task (IGT). One hundred ninety-two participants were recruited and categorized into one non-social feedback group and four social feedback groups. Participants in the social feedback groups were informed that after the outcome of each choice, they would also receive feedback from an online peer. This peer was a fictitious entity, with variations in identity (novice or expert) and feedback type (random or effective). The Outcome-Representation Learning model (ORL model) was used to quantify the cognitive components of learning. Behavioral results showed that both the identity of the peer and the type of feedback provided significantly influenced the deck selection, with effective social feedback increasing the ratio of chosen good decks. Results in the ORL model showed that the four social feedback groups exhibited lower learning rates for gain and loss compared to the nonsocial feedback group, which suggested, in the social feedback groups, the impact of the recent outcome on the update of value decreased. Parameters such as forgetfulness, win frequency, and deck perseverance in the expert-effective feedback group were significantly higher than those in the non-social feedback and expert-random feedback groups. These findings suggest that individuals proactively evaluate feedback providers and selectively adopt effective feedback to enhance learning.

Abstract Climate change exacerbates the threat of water scarcity over the drylands in East Asia (DEA), the world's most densely populated arid region. The water cycle continuously supplies water to support all life. Previous studies have focused on the change in individual hydrological components over DEA; however, how the projected water cycle changes under climate warming remains unclear. We demonstrate the projected response of the water cycle to global warming in different seasons utilizing the Coupled Model Intercomparison Project Phase 6. Winter in the DEA presents an intensification of the water cycle, reflected in coherent increases in evapotranspiration (E), precipitation (P), runoff, and surface soil moisture. In contrast, summer will experience a weakened water cycle in the northwestern DEA, while the southeastern part exhibits the opposite trend. From the surface and atmospheric water balance perspective, we further attribute the changes in E and P to gain a more comprehensive understanding. The increasing E is attributed to the combined effects of P and the vapor pressure deficit during summer, whereas it is dominated by P in winter. The increased P in summer is primarily attributed to the horizontal dynamic and vertical thermodynamic components associated with the strengthening and westward expansion of East Asia summer monsoon in the future. During winter, the increased P is mainly due to the vertical dynamic and horizontal thermodynamic components associated with the enhancement of vertical ascending motion and increased moisture.

Abstract Background Metabolites play critical roles in regulating nutritional qualities of plants, thereby influencing their consumption and human health. However, the genetic basis underlying the metabolite-based nutrient quality and domestication of root and tuber crops remain largely unknown. Results We report a comprehensive study combining metabolic and phenotypic genome-wide association studies to dissect the genetic basis of metabolites in the storage root (SR) of cassava. We quantify 2,980 metabolic features in 299 cultivated cassava accessions. We detect 18,218 significant marker-metabolite associations via metabolic genome-wide association mapping and identify 12 candidate genes responsible for the levels of metabolites that are of potential nutritional importance. Me3GT, MeMYB4, and UGT85K4/UGT85K5, which are involved in flavone, anthocyanin, and cyanogenic glucoside metabolism, respectively, are functionally validated through in vitro enzyme assays and in vivo gene silencing analyses. We identify a cluster of cyanogenic glucoside biosynthesis genes, among which CYP79D1, CYP71E7b, and UGT85K5 are highly co-expressed and their allelic combination contributes to low linamarin content. We find MeMYB4 is responsible for variations in cyanidin 3-O-glucoside and delphinidin 3-O-rutinoside contents, thus controlling SR endothelium color. We find human selection affects quercetin 3-O-glucoside content and SR weight per plant. The candidate gene MeFLS1 is subject to selection during cassava domestication, leading to decreased quercetin 3-O-glucoside content and thus increased SR weight per plant. Conclusions These findings reveal the genetic basis of cassava SR metabolome variation, establish a linkage between metabolites and agronomic traits, and offer useful resources for genetically improving the nutrition of cassava and other root crops.

Drought stress impairs plant growth and other physiological functions. MeHDZ14, a homeodomain-leucine zipper I transcription factor, is strongly induced by drought stress in various cassava cultivars. However, the role of MeHDZ14 in cassava growth regulation has remained unclear. Here we report that MeHDZ14 affected plant height, such that a dwarf phenotype and altered internode elongation were observed in transgenic cassava lines. MeHDZ14 was found to negatively regulate the biosynthesis of lignin. Its overexpression resulted in abaxially rolled leaves. The morphogenesis of leaf epidermal cells was inhibited by overexpression of MeHDZ14, with decreased auxin and gibberellin and increased cytokinin contents. MeHDZ14 was found to regulate many drought-responsive genes, including genes involved in cell wall synthesis and expansion. MeHDZ14 bound to the promoter of caffeic acid 3-O-methyltransferase 1 (MeCOMT1), acting as a transcriptional repressor of genes involved in cell wall development. MeHDZ14 appears to act as a negative regulator of internode elongation and epidermal cell morphogenesis during cassava leaf development.

Chilling-induced accumulation of reactive oxygen species (ROS) is harmful to plants, which usually produce anthocyanins to scavenge ROS as protection from chilling stress. As a tropical crop, cassava is hypersensitive to chilling, but the biochemical basis of this hypersensitivity remains unclear. We previously generated MeMYB2-RNAi transgenic cassava with increased chilling tolerance. Here we report that MeMYB2-RNAi transgenic cassava accumulated less ROS but more cyanidin-3-O-glucoside than the wild type under early chilling stress. Under this stress, the anthocyanin biosynthesis pathway was more active in MeMYB2-RNAi lines than in the wild type, and several genes involved in the pathway, including MeTT8, were up-regulated by MeMYB2-RNAi in the transgenic cassava. MeMYB2 bound to the MeTT8 promoter and blocked its expression under both normal and chilling conditions, thereby inhibiting anthocyanin accumulation. MeTT8 was shown to bind to the promoter of Dihydroflavonol 4-reductase (MeDFR-2) and increased MeDFR-2 expression. MeMYB2 appears to act as an inhibitor of chilling-induced anthocyanin accumulation during the rapid response of cassava to chilling stress.

Double-wall steel sheet piles (DSSPs) are widely used in large-span cofferdams for docks due to their good performance against wave action during storm surges. This paper describes a study of the dynamic behavior of a DSSP cofferdam under wave action through flume tests and a numerical simulation that combined computational fluid dynamics (CFD) and the finite element method. The influences of the water level and wave height on the DSSP cofferdam were investigated experimentally and numerically. Tall waves in shallow water broke upon and impacted the seaside pile with large dynamic wave pressure, dramatically increasing the stress and displacement of the seaside pile. The overlap of the traveling and reflected waves increased the excess pore water pressure near the seaside pile due to taller overlapped waves and higher wave frequency. The DSSP cofferdam failed under the combined actions of the dynamic wave pressure and erosion of the landside seabed. The leakage and overflow of the breaking waves resulted in significant erosion of the landside seabed and greatly weakened the support of the seabed. The dynamic wave pressure then pushed the DSSP cofferdam until it failed. The simulation with the combined methods of CFD and FEM resulted in trends that were similar to those of the test measurements. Compared to the quasi-static method and pseudo-dynamic method, the results of the simulation via the present method were much closer to the test results because the simulation included the effects of breaking waves. The reinforced measure worked well to prevent the DSSP cofferdam in a sandy seabed foundation from continuous failures of deformation–leakage–erosion–tilting. However, it failed in a clay interlayer seabed foundation due to the large settlement.

ABSTRACTDimethylsulfoniopropionate (DMSP) is one of Earth’s most abundant organosulfur molecules, which can be catabolized by marine bacteria to release climate-active gases through the cleavage and/or demethylation pathways. The marine SAR92 clade is an abundant oligotrophic group of Gammaproteobacteria in coastal seawater, but their ability to catabolize DMSP is untested. Three SAR92 clade strains isolated from coastal seawater in this study and the SAR92 representative strain HTCC2207 were all shown to catabolize DMSP as a carbon source. All the SAR92 clade strains exhibited DMSP lyase activity producing dimethylsulfide (DMS) and their genomes encoded a ratified DddD DMSP lyase. In contrast, only HTCC2207 and two isolated strains contained the DMSP demethylase dmdA gene and potentially simultaneously demethylated and cleaved DMSP to produce methanethiol (MeSH) and DMS. In SAR92 clade strains with dddD and dmdA, transcription of these genes was inducible by DMSP substrate. Bioinformatic analysis indicated that SAR92 clade bacteria containing and transcribing DddD and DmdA were widely distributed in global oceans, especially in polar regions. This study highlights the SAR92 clade of oligotrophic bacteria as potentially important catabolizers of DMSP and sources of the climate-active gases MeSH and DMS in marine environments, particularly in polar regions.IMPORTANCECatabolism of dimethylsulfoniopropionate (DMSP) by marine bacteria has important impacts on the global sulfur cycle and climate. However, whether and how members of most oligotrophic bacterial groups participate in DMSP metabolism in marine environments remains largely unknown. In this study, by characterizing culturable strains, we have revealed that bacteria of the SAR92 clade, an abundant oligotrophic group of Gammaproteobacteria in coastal seawater, can catabolize DMSP through the DMSP lyase DddD-mediated cleavage pathway and/or the DMSP demethylase DmdA-mediated demethylation pathway to produce climate-active gases dimethylsulfide and methanethiol. Additionally, we found that SAR92 clade bacteria capable of catabolizing DMSP are widely distributed in global oceans. These results indicate that SAR92 clade bacteria are potentially important DMSP degraders and sources of climate-active gases in marine environments that have been overlooked, contributing to a better understanding of the roles and mechanisms of the oligotrophic bacteria in oceanic DMSP degradation.

Abstract Bazhong City is located on stratum dominated by red-bed conditions. This type of weak geological condition with sand and mudstone interbedding is very easily affected by climatic conditions to produce rainfall-type landslides. On the basis of landslide data statistics collected in Bazhong City from 2011 to 2019, this paper uses ERA5-Land rainfall data to statistically analyze the correlation between rainfall and landslide events in Bazhong City. The landslide events in Bazhong City are greatly affected by rainfall events lasting for 10 days. Considering the influence of rainfall seepage and other processes, an effective cumulative rainfall-duration threshold curve for Bazhong City is obtained via median nonlinear fitting. Then, on the basis of the prediction interval, the rainfall threshold formula under different landslide occurrence probabilities is obtained and the critical threshold curve with a non-exceeding probability of 1% in Bazhong City is calculated and verified. Subsequently, a continuous probability distribution fitting function of landslide occurrence is established and a continuous probability distribution surface with a good fitting effect in Bazhong City is obtained. This allows a definite probability of whether future rainfall events will induce landslides to be obtained, providing an important basis for engineering disaster prevention and mitigation.

Abstract Background Emotional stimuli used as targets of working memory (WM) tasks can moderate age-related differences in WM performance, showing that aging is associated with reductions in negativity bias. This phenomenon is referred to as the positivity effect. However, there is little research on whether emotional distractors have a similar moderating effect. Moreover, the underlying neural mechanism of this effect has not been studied. In this study, we examined the behavioral and neurophysiological basis for age differences in resistance to emotional distractors within WM. Methods Older adults (n = 30, ages 60–74) and young adults (n = 35, ages 19–26) performed a 2-back task in which a digit was superimposed on a face with a happy, angry, or neutral expression as a distractor. Event-related potential (ERP) was simultaneously recorded to assess P2, N2, and later positive potential (LPP) amplitudes. Results Older adults were less accurate and slower than young adults on the WM task. Moreover, the results demonstrated a significant interaction between age and emotional valence on response accuracy, young adults' performance was worse when the distractor was neutral or positive than when it was negative, but there was no effect of the emotional valence of distractors on older adults’ WM performance. ERP analyses revealed greater P2 amplitude in older adults than young adults, regardless of the emotional valence of distractors. However, older adults and young adults did not differ on N2 or LPP amplitude, and negative distractors elicited greater N2 than positive distractors in both age groups. Conclusions The behavioral findings provided evidence of age-related reductions in negativity bias. Thus, the behavioral measures indicated a positivity effect in WM. However, the ERP results did not show this same interaction. These discrepant results raise questions about whether and to what extent older and young adults differ in controlling the effect of emotional distractors in WM.

The breaching process of landslide dams influenced by bed slopes and drainage channels are still unclear. In this study, flume tests of landslide dams including four different bed slope angles (0°, 1°, 2° and 3°) and three different drainage channel cross sections (triangle, trapezoid and compound) are carried out. The effects of bed slopes and drainage channel cross sections on the outflow discharge, breaching duration, breach development and longitudinal evolution process of landslide dams are investigated. The results show that (1) the breaching process is divided into three stages. Stage I is the breach initiation stage with a slow breach erosion. Stage II is the breach development stage with a severe breach erosion. Stage III is the attenuating and re-equilibrium stage with the formation of an armored layer and the breach erosion gradually stops. (2) With the increasing bed slope angle, the dam crest and downstream slope angle increases, which enhances the shear stress acting on the soil and the deep-cutting erosion. Thus, when the bed slope is steeper, the peak discharge is larger, the arrival time of peak is shortened and the residual dam height is smaller. (3) Drainage channels with different cross sections have different depths, widths and lateral slopes, which influence the breach development and the outflow discharge. For the triangle drainage channel, its water-soil interface area is small, both the breach deepening and widening rates are the largest, while the peak discharge is the largest and the arrival time of peak is the earliest. For the trapezoidal drainage channel, its bottom elevation is the highest and water-soil interface area is the largest, while the breach deepening rate is the lowest and the arrival time of peak is the latest. The compound drainage channel is between the two former channels. The experimental results may provide important references for emergency treatment of landslide dams.

Abstract Herein, we aimed to determine the effect of vitamin D (Vit D) and underlying mechanisms on asthma‐induced lung injury via regulation of HIF‐1α/Notch1 (hypoxia‐inducible factor 1 alpha/neurogenic locus notch homolog protein 1) signaling during autophagy. We established an asthma mouse model using respiratory syncytial virus (RSV) nasal drop combined with ovalbumin (OVA) atomization. Mice were treated with different Vit D concentrations. Pathological changes and cell apoptosis were examined using hematoxylin–eosin (HE) staining and TUNEL (terminal deoxynucleotidyl transferase‐mediated deoxyuridine triphosphate (dUTP) nick end‐labeling) assay, respectively. Additionally, periodic acid–Schiff (PAS) and Masson's trichrome staining solutions were used to examine changes in lung tissue. Immunofluorescence determined LC 3B (microtubule‐associated protein 1 light chain 3B) expression in lung tissues, whereas western blotting and immunohistochemistry were used to evaluate other proteins, including HIF‐1α and Notch1. Compared with the normal group, the asthma model group exhibited pathological lung tissue deterioration, elevated fibrosis, increased apoptosis cell numbers, and upregulated autophagy. Vitamin D supplementation ameliorated pathological changes and fibrosis in the lung tissue. Furthermore, Vit D treatment significantly suppressed apoptotic cell numbers and autophagy while enhancing the HIF‐1α/Notch1 pathway. Given the HIF‐1α/Notch1 agonistic activity, Vit D treatment inhibited apoptosis cell numbers, which were increased following asthma‐induced upregulation of autophagy. Vitamin D improved asthma‐induced lung tissue injury by suppressing autophagy via regulation of HIF‐1α/Notch1 signaling in vivo.

Abstract Marine algae and bacteria produce approximately eight billion tonnes of the organosulfur molecule dimethylsulfoniopropionate (DMSP) in Earth's surface oceans annually. DMSP is an antistress compound and, once released into the environment, a major nutrient, signaling molecule, and source of climate‐active gases. The methionine transamination pathway for DMSP synthesis is used by most known DMSP‐producing algae and bacteria. The S‐directed S‐adenosylmethionine (SAM)‐dependent 4‐methylthio‐2‐hydroxybutyrate (MTHB) S‐methyltransferase, encoded by the dsyB/DSYB gene, is the key enzyme of this pathway, generating S‐adenosylhomocysteine (SAH) and 4‐dimethylsulfonio‐2‐hydroxybutyrate (DMSHB). DsyB/DSYB, present in most haptophyte and dinoflagellate algae with the highest known intracellular DMSP concentrations, is shown to be far more abundant and transcribed in marine environments than any other known S‐methyltransferase gene in DMSP synthesis pathways. Furthermore, we demonstrate in vitro activity of the bacterial DsyB enzyme from Nisaea denitrificans and provide its crystal structure in complex with SAM and SAH‐MTHB, which together provide the first important mechanistic insights into a DMSP synthesis enzyme. Structural and mutational analyses imply that DsyB adopts a proximity and desolvation mechanism for the methyl transfer reaction. Sequence analysis suggests that this mechanism may be common to all bacterial DsyB enzymes and also, importantly, eukaryotic DSYB enzymes from e.g., algae that are the major DMSP producers in Earth's surface oceans.

S-methyl methionine (SMM) is a key molecule in production of dimethylsulfoniopropionate (DMSP), an important marine anti-stress compound, with roles in global nutrient cycling. Here, the authors determine the mechanism of SMM synthesis and uncover unexpected roles for SMM in archaea, CPR bacteria and animals.

The reactive oxygen species (ROS) signal regulates stress-induced leaf abscission in cassava. The relationship between the function of the cassava transcription factor bHLH gene and low temperature-induced leaf abscission is still unclear. Here, we report that MebHLH18, a transcription factor, involved in regulating low temperature-induced leaf abscission in cassava. The expression of the MebHLH18 gene was significantly related to low temperature-induced leaf abscission and POD level. Under low temperatures, the levels of ROS scavengers in different cassava genotypes were significantly different in the low temperature-induced leaf abscission process. Cassava gene transformation showed that MebHLH18 overexpression significantly decreased the low temperature-induced leaf abscission rate. Simultaneously, interference expression increased the rate of leaf abscission under the same conditions. ROS analysis showed a connection between the decrease in the low temperature-induced leaf abscission rate caused by MebHLH18 expression and the increase in antioxidant activity. A Genome-wide association studies analysis showed a relationship between the natural variation of the promoter region of MebHLH18 and low temperature-induced leaf abscission. Furthermore, studies showed that the change in MebHLH18 expression was caused by a single nucleotide polymorphism variation in the promoter region upstream of the gene. The high expression of MebHLH18 led to a significant increase in POD activity. The increased POD activity decreased the accumulation of ROS at low temperatures and the rate of leaf abscission. It indicates that the natural variation in the promoter region of MebHLH18 increases antioxidant levels under low temperatures and slows down low temperature-induced leaf abscission.

This study used the 400 g-ton geotechnical centrifuge model test system at the Nanjing Hydraulic Research Institute (NHRI) to investigate the breach evolution characteristics and hydrograph process of overtopping-induced breaching of landslide dams. It was achieved by taking advantage of the “time-space amplification” effect created by high-speed rotation using a centrifuge overweight force field. The effects of dam height, slope ratio, and soil gradation on the overtopping failure process of landslide dams were investigated by centrifugal model tests for the first time. In addition, a detailed physically-based dam breach model was developed to simulate the overtopping failure of landslide dams. Results show that the breach process of a landslide dam can be divided into four stages based on the measured breach morphology evolution process and the abrupt variations of breach flow discharge: initial scour on the downstream slope, retrogressive erosion to the dammed lake, erosion along the breach channel, and breach stabilization. Moreover, the peak breach flow is most sensitive to the dam height, followed by the average particle size; the time to peak is mainly affected by the slope ratio, and the relative residual dam height is primarily susceptible to the average particle size. In practice, the calculated results are consistent with the measured results. This study provides a scientific reference for the cognition of the overtopping-induced breach mechanism of landslide dams.

Cassava is an important tropical cash crop. Severe drought stresses affect cassava productivity and quality, and cause great economic losses in agricultural production. Enhancing the drought tolerance of cassava can effectively improve its yield. Long non-coding RNAs (lncRNAs) are present in a wide variety of eukaryotes. Recently, increasing evidence has shown that lncRNAs play a critical role in the responses to abiotic stresses. However, the function of cassava lncRNAs in the drought response remains largely unknown. In this study, we identified a novel lncRNA, DROUGHT-INDUCED INTERGENIC lncRNA (DIR). Gene expression analysis showed that DIR was significantly induced by drought stress treatment, but did not respond to abscisic acid (ABA) or jasmonic acid (JA) treatments. In addition, overexpression of the DIR gene enhanced proline accumulation and drought tolerance in transgenic cassava. RNA-seq analysis revealed that DIR preferentially affected drought-related genes that were linked to transcription and metabolism. Moreover, RNA pull-down mass spectrometry analysis showed that DIR interacted with 325 proteins. A protein–protein interaction (PPI) analysis found a marked enrichment in proteins associated with the mRNA export and protein quality control pathways. Collectively, these results suggest that DIR and its interacting proteins that regulate mRNA or protein metabolism are involved in mediating the drought stress response. Thus, regulating DIR expression has potential for improving cassava yield under drought conditions.

Abstract Background We previously identified six drought-inducible CC-type glutaredoxins in cassava cultivars, however, less is known about their potential role in the molecular mechanism by which cassava adapted to abiotic stress. Results Herein, we investigate one of cassava drought-responsive CC-type glutaredoxins, namely MeGRXC3, that involved in regulation of mannitol-induced inhibition on seed germination and seedling growth in transgenic Arabidopsis. MeGRXC3 overexpression up-regulates several stress-related transcription factor genes, such as PDF1.2, ERF6, ORA59, DREB2A, WRKY40, and WRKY53 in Arabidopsis. Protein interaction assays show that MeGRXC3 interacts with Arabidopsis TGA2 and TGA5 in the nucleus. Eliminated nuclear localization of MeGRXC3 failed to result mannitol-induced inhibition of seed germination and seedling growth in transgenic Arabidopsis. Mutation analysis of MeGRXC3 indicates the importance of conserved motifs for its transactivation activity in yeast. Additionally, these motifs are also indispensable for its functionality in regulating mannitol-induced inhibition of seed germination and enhancement of the stress-related transcription factors in transgenic Arabidopsis. Conclusions MeGRXC3 overexpression confers mannitol sensitivity in transgenic Arabidopsis possibly through interaction with TGA2/5 in the nucleus, and nuclear activity of MeGRXC3 is required for its function.

Abstract Background Heterozygous genomes are widespread in outcrossing and clonally propagated crops. However, the variation in heterozygosity underlying key agronomic traits and crop domestication remains largely unknown. Cassava is a staple crop in Africa and other tropical regions and has a highly heterozygous genome. Results We describe a genomic variation map from 388 resequenced genomes of cassava cultivars and wild accessions. We identify 52 loci for 23 agronomic traits through a genome-wide association study. Eighteen allelic variations in heterozygosity for nine candidate genes are significantly associated with seven key agronomic traits. We detect 81 selective sweeps with decreasing heterozygosity and nucleotide diversity, harboring 548 genes, which are enriched in multiple biological processes including growth, development, hormone metabolisms and responses, and immune-related processes. Artificial selection for decreased heterozygosity has contributed to the domestication of the large starchy storage root of cassava. Selection for homozygous GG allele in MeTIR1 during domestication contributes to increased starch content. Selection of homozygous AA allele in MeAHL17 is associated with increased storage root weight and cassava bacterial blight (CBB) susceptibility. We have verified the positive roles of MeTIR1 in increasing starch content and MeAHL17 in resistance to CBB by transient overexpression and silencing analysis. The allelic combinations in MeTIR1 and MeAHL17 may result in high starch content and resistance to CBB. Conclusions This study provides insights into allelic variation in heterozygosity associated with key agronomic traits and cassava domestication. It also offers valuable resources for the improvement of cassava and other highly heterozygous crops.

Disgust, as a part of the behavioral immune system, leads people to avoid behaviors of pathogens so as to reduce the probability of infection. Disgust also shows the source effects based on familiarity. However, these source effects have not been tested on the older population. Thus, we tested the source effects of emotional and behavioral reactions from the disgust toward older adults and the possible moderating effects of filial piety on disgust. In the first study, we employed the self-report method to test the source effects of emotional feelings of disgust amongst undergraduates. In the second study, we measured whether filial piety among community adults produced moderating effects of the disgust toward older adults. In the third study, we employed the shape discrimination task to test the source effects of behavioral avoidance to older adults among undergraduates. The first and third studies show stronger negative emotional/avoidance reactions towards unfamiliar older adults than familiar older adults, affirming the source effects of disgust towards older adults that we expected. However, we did not find moderating effects of filial piety associated with disgust. These findings can help us understand the evolutionary origin of disgust toward older adults, which is likely activated more intensely and quickly in response to unfamiliar individuals as compared with familiar individuals.

Research on the factors and mechanisms that influence outburst floods are essential for estimating outflow hydrographs and the resulting inundation. In this study, large flume tests are conducted to investigate the effects of the upstream inflow and the presence of loose erodible deposits on the breaching flow and the subsequent outburst floods. Experimental results reveal that hydrographs of the breaching flow and outburst flood can be divided into three stages where each stage is separated by inflection points and peak discharges. It is found that the larger the inflow discharge, the larger the peak discharge of the outburst flood and the shorter the time needed to reach the peak and inflection discharges of the outburst flood. The breaching flow decreases along the longitudinal direction at rates that increase with the inflow discharge. The ratio between the length of the upstream dam shoulder and the dam width is inversely related to the ratio of the outburst discharge to inflow discharge. We also show that the presence of loose deposits at the dam toe can amplify the peak discharge of outburst flood by increasing the solids content of the water flow.

Warning and evacuation are among the most effective ways for saving human lives and properties from landslide dam hazards. A new warning decision model for landslide dam break is developed using Influence Diagrams to minimize the total losses. An Influence Diagram is a simple visual representation of a decision problem. It analyzes the qualitative (causal) relationships between the variables via a logic diagram and determines the quantitative relationships via conditional probability and Bayes’ theorem. The model is applied for the warning decision-making of the 2008 Tangjiashan landslide dam. The new model unifies the dam failure probability, evacuation, life loss, and flood damage in an Influence Diagram. Besides, a warning criterion is proposed for efficient decision-making. The model is more advanced than the decision tree since the inter-relationships of influence factors are qualitatively analyzed with causality connections and quantitatively analyzed with conditional probabilities. It is more efficient than a dynamic decision-making model (DYDEM) as it can directly calculate the three types of flood loss (i.e., evacuation cost, flood damage, and monetized life loss) and the expected total loss. Moreover, the probabilities of the influence factors leading to known results can be obtained through inversion analysis based on Bayesian theory. The new warning decision model offers an efficient way to save lives from landslide dam breaking and avoid unnecessary expenses from premature warning and evacuation.

The design of a gas diffusion layer (GDL) is an effective way to manage water transport, thus improving the performance of air-cooled fuel cells. In the present study, three group designs of GDL with polytetrafluoroethylene (PTFE)—uniformly doped, in-planed sandwich doped and through-plane gradient doped—are proposed, and their effects on the performance of air-cooled fuel cells are explored by numerical simulation. The distribution of key physical quantities in the cathode catalyst layer (CCL), current density and the uniformity of current density distribution in the CCL were analyzed in detail. The results show that properly reducing the amount of PTFE in GDL is beneficial to promoting the water retaining capacity of air-cooled fuel cells, and then improving the performance of fuel cells. The performance of the in-plane sandwich GDL design cannot exceed the design with 10% PTFE uniformly doped, and this design will aggravate the uneven distribution of current density in CCL. Compared with the design of GDL with 40% PTFE uniformly doped, the current density can be improved by 22% when operating at 0.6 V by gradient increasing the PTFE content in GDL from the GDL/MPL interface to the gas channel. Furthermore, this design can maintain as good a current density uniformity as uniformly doping schemes.

The outbreak of coronavirus disease 2019 (COVID-19) has been a global emergency, affecting millions of individuals both physically and psychologically. The present research investigated the associations between social media exposure and depression during the COVID-19 outbreak by examining the mediating role of psychological distress and the moderating role of emotion regulation among members of the general public in China. Participants (N = 485) completed a set of questionnaires online, including demographic information, self-rated physical health, and social media exposure to topics related to COVID-19. The Impact of Event Scale-Revised (IES-R), the Beck Depression Inventory-II (BDI-II), and the Emotion Regulation Questionnaire (ERQ) were utilized to measure psychological distress about COVID-19, depression, and emotion regulation strategies, respectively. Results found that older age and greater levels of social media exposure were associated with more psychological distress about the virus (r = 0.14, p = 0.003; r = 0.22, p < 0.001). Results of the moderated mediation model suggest that psychological distress mediated the relationship between social media exposure and depression (β = 0.10; Boot 95% CI = 0.07, 0.15). Furthermore, expressive suppression moderated the relationship between psychological distress and depression (β = 0.10, p = 0.017). The findings are discussed in terms of the need for mental health assistance for individuals at high risk of depression, including the elderly and individuals who reported greater psychological distress and those who showed preference usage of suppression, during the COVID-19 crisis.

Mega earthquakes or serious rainfall storms often cause crowded landslides in mountainous areas. A large part of these landslides are very likely blocking rivers and forming landslide dams in series along rivers. The risks of cascading failure of landslide dams are significantly different from that of a single dam. This paper presented the work on risk-based warning decision making on cascading breaching of the 2008 Tangjiashan landslide dam and two small downstream landslide dams in a series along Tongkou River. The optimal decision was made by achieving minimal expected total loss. Cascade breaching of a series of landslide dams is more likely to produce a multi-peak flood. When the coming of the breaching flood from the upstream dam perfectly overlaps with the dam breaching flood of the downstream dam, a higher overlapped peak flood would occur. When overlapped peak flood occurs, the flood risk would be larger and evacuation warning needs to be issued earlier to avoid serious life loss and flood damages. When multi-peak flood occurs, people may be misled by the warning of the previous peak flood and suddenly attacked by the peak flood thereafter, incurring catastrophic loss. Systematical decision making needs to be conducted to sufficiently concern the risk caused by each peak of the breaching flood. The dam failure probability Pf linearly influences the expected life loss and flood damage but does not influence the evacuation cost. The expected total loss significantly decreases with Pf when the warning time was insufficient. However, it would not change much with Pf when warning time is sufficient.

Abstract Background Banana wilt disease, caused by Fusarium oxysporum f. sp. cubense Tropical Race 4 (Foc TR4), is one of the most devastating diseases in banana (Musa spp.). Foc is a soil borne pathogen that causes rot of the roots or wilt of leaves by colonizing the xylem vessels. The dual RNA sequencing is used to simultaneously assess the transcriptomes of pathogen and host. This method greatly helps to understand the responses of pathogen and host to each other and discover the potential pathogenic mechanism. Results Plantlets of two economically important banana cultivars, Foc TR4 less susceptible cultivar NK and susceptible cultivar BX, were used to research the Foc-banana interaction mechanism. Notably, the infected NK had more significantly up-regulated genes on the respiration machinery including TCA cycle, glyoxylate, glycerol, and glycolysis compared to BX at 27 h post inoculation (hpi). In addition, genes involved in plant-pathogen interaction, starch, sucrose, linolenic acid and sphingolipid metabolisms were uniquely more greatly induced in BX than those in NK during the whole infection. Genes related to the biosynthesis and metabolism of SA and JA were greatly induced in the infected NK; while auxin and abscisic acid metabolisms related genes were strongly stimulated in the infected BX at 27 hpi. Furthermore, most of fungal genes were more highly expressed in the roots of BX than in those of NK. The fungal genes related to pathogenicity, pectin and chitin metabolism, reactive oxygen scavenging played the important roles during the infection of Foc. CCP1 (cytochrome c peroxidase 1) was verified to involve in cellulose utilization, oxidative stress response and pathogenicity of fungus. Conclusion The transcriptome indicated that NK had much faster defense response against Foc TR4 than BX and the expression levels of fungal genes were higher in BX than those in NK. The metabolisms of carbon, nitrogen, and signal transduction molecular were differentially involved in pathogen infection in BX and NK. Additionally, the putative virulence associated fungal genes involved in colonization, nutrition acquirement and transport provided more insights into the infection process of Foc TR4 in banana roots.

Abstract Background CC-type glutaredoxins (GRXs) are plant-specific glutaredoxin, play regulatory roles in response of biotic and abiotic stress. However, it is not clear whether the CC-type GRXs are involve in drought response in cassava (Manihot esculenta), an important tropical tuber root crop. Results Herein, genome-wide analysis identified 18 CC-type GRXs in the cassava genome, of which six (namely MeGRXC3, C4, C7, C14, C15, and C18) were induced by drought stress in leaves of two cassava cultivars Argentina 7 (Arg7) and South China 124 (SC124). Exogenous abscisic acid (ABA) application induced the expression of all the six CC-type GRXs in leaves of both Arg7 and SC124 plants. Overexpression of MeGRXC15 in Arabidopsis (Col-0) increases tolerance of ABA on the sealed agar plates, but results in drought hypersensitivity in soil-grown plants. The results of microarray assays show that MeGRXC15 overexpression affected the expression of a set of transcription factors which involve in stress response, ABA, and JA/ET signalling pathway. The results of protein interaction analysis show that MeGRXC15 can interact with TGA5 from Arabidopsis and MeTGA074 from cassava. Conclusions CC-type glutaredoxins play regulatory roles in cassava response to drought possibly through ABA signalling pathway.

When people express a moral judgment, others make inferences about their personality, such as whether they are warm or competent. People may use this interpersonal process to present themselves in a way that is socially acceptable in the current circumstances. Across four studies, we investigated this hypothesis in Chinese culture and showed that college student participants tended to associate others’ deontological moral judgments with warmth and utilitarian moral judgments with competence (Study 1, Mage = 21.1, SD = 2.45; Study 2, Mage = 20.53, SD = 1.87). In addition, participants made more deontological judgments after preparing to be interviewed for a job requiring them to be in a warm social role, and more utilitarian judgments after preparing for a job requiring them to be in a competent social role (Study 3, Mage = 19.5, SD = 1.63). This effect held true in moral dilemmas involving different degrees of hypothetical personal involvement, and appeared to be mediated by the perception of others’ expectations (Study 4, Mage = 19.92, SD = 1.97). The results suggest an important role for social cognition as an influence on moral judgments in Chinese culture.

Numerous landslide dams have been induced in recent years as a result of frequent earthquakes and extreme climate hazards. Landslide dams present serious threats to lives and properties downstream due to potentially breaching floods from the impounded lakes. To investigate the factors influencing the stability of landslide dams, a large database has been established based on an in-depth investigation of 1,737 landslide dam cases. The effects of triggers, dam materials, and geomorphic characteristics of landslide dams on dam stability are comprehensively analyzed. Various evaluation indexes of landslide dam stability are assessed based on this database, and stability evaluation can be further improved by considering the dam materials. Stability analyses of aftershocks, surges, and artificial engineering measures on landslide dams are summarized. Overtopping and seepage failures are the most common failure modes of landslide dams. The failure processes and mechanisms of landslide dams caused by overtopping and seepage are reviewed from the perspective of model experiments and numerical analyses. Finally, the research gaps are highlighted, and pathways to achieve a more complete understanding of landslide dam stability are suggested. This comprehensive review of the recent advances in stability and failure mechanisms of landslide dams can serve as a key reference for stability prediction and emergency risk mitigation.

Excessive nitrogen (N) application is widespread in Southern China. The effects of N fertilization on soil properties and crop physiology are poorly understood in tropical red loam soil. We conducted a field experiment to evaluate the effect of nitrogen fertilization rates on physiological attributes (chlorophyll, plant metabolic enzymes, soluble matters) on banana leaves, soil properties (soil enzymes, soil organic matter (SOM), soil available nutrients) as well as banana crop yield in a subtropical region of southern China. The N rates tested were 0 (N0), 145 (N145), 248 (N248), 352 (N352), 414 (NFT), and 455 (N455) g N per plant. The correlations among soil factors, leaf physiological factors and crop yield were evaluated. The results indiated that the high rates of N fertilization (NFT and N455) significantly decreased soil available potassium (K) content, available phosphorus (P) content, glutamine synthetase (GS) activity, and soluble protein and sugar contents compared with lower N rates. The N352 treatment had the highest crop yields compared with higher N rates treatments, followed by the N455 treatment. However, there were no significant differences in crop yields among N fertilization treatments. Factor analysis showed that the N352 treatment had the highest integrated score for soil and leaf physiological factors among all treatments. Moreover, the N352 treatment was the most effective in improving carbon and nitrogen metabolism in banana. Crop yield was significantly and positively linearly correlated with the integrated score (r = 0.823, p < 0.05). Path analysis revealed that invertase, SOM and sucrose synthase (SS) had a strong positive effect on banana yield. Canonical correspondence analysis (CCA) suggested that available K, invertase, acid phosphatase and available P were the most important factors impacting leaf physiological attributes. Cluster analysis demonstrated distinct differences in N application treatment related to variations in soil and leaf factors. This study suggested that excessive N fertilization had a negative effect on soil fertility, crop physiology and yield. The lower N rates were more effective in improving crop yield than higher rates of N fertilization. The N rate of 352 g N per plant (N352) was recommended to reduce excess N input while maintaining the higher yield for local farmers’ banana planting.

Landslide dams are dangerous because the outburst floods produced by dam failures seriously threaten life and property downstream. In this study, a series of physical flume tests were conducted to investigate the breaching process of landslide dams with fine-grained, well graded, and coarse-grained material under different inflow conditions. The effects of dam material and inflow discharge on the breach development, outflow discharge and erosion characteristics were studied. The erosion resistance of materials and lateral collapses were also discussed. Experimental results reveal that the whole breaching process is determined by the water-sediment interaction. For the fine-grained dams, a general constant downstream slope angle is maintained during the breaching process. For the well-graded dams, a step-pool structure is generated due to the scarp erosion. For the coarse-grained dams, they can remain stable under normal circumstances but fail by overtopping in a short duration under the extreme inflow condition. The final breach of the dam with higher fine content or larger inflow discharge is deeper and narrower. In addition, many fluctuations are observed in the changing curve of the erosion rates along the flow direction for the well-graded and coarse-grained dams. The erosion resistance of materials increases along the flow direction, which needs to be further considered in physically based breach models. Furthermore, the lateral collapse is affected by the dam material instead of inflow discharge. The lower fine content causes more lateral collapses with smaller volumes.

It has been widely accepted that moral violations that involve impurity (such as spitting in public) induce the emotion of disgust, but there has been a debate about whether moral violations that do not involve impurity (such as swearing in public) also induce the same emotion. The answer to this question may have implication for understanding where morality comes from and how people make moral judgments. This study aimed to compared the neural mechanisms underlying two kinds of moral violation by using an affective priming task to test the effect of sentences depicting moral violation behaviors with and without physical impurity on subsequent detection of disgusted faces in a visual search task. After reading each sentence, participants completed the face search task. Behavioral and electrophysiological (event-related potential, or ERP) indices of affective priming (P2, N400, LPP) and attention allocation (N2pc) were analyzed. Results of behavioral data and ERP data showed that moral violations both with and without impurity promoted the detection of disgusted faces (RT, N2pc); moral violations without impurity impeded the detection of neutral faces (N400). No priming effect was found on P2 and LPP. The results suggest both types of moral violation influenced the processing of disgusted faces and neutral faces, but the neural activity with temporal characteristics was different.

Abstract Background Glutathione S-transferases (GSTs) have been reported to regulate the plant tolerance to environmental stresses. Many plant GSTs exhibited the roles on promoting tolerance to drought stress, oxidative stress and plant hormones. The biological function of GSTs has been well characterized in Arabidopsis thaliana in response to exogenous environmental stresses. However, their regulation function under exogenous environmental stresses regulating leaf abscission in cassava (Manihot esculenta Crantz) remained unknown. Results Here, 83 GSTs were identified from tropical plant cassava. The amino acid motifs and phylogenetic analyses indicated that MeGSTs were divided into 9 classes. The global expression analyses were carried out to analyze the gene expression patterns of MeGST in cassava abscission zones by comparing the MeGST genes expression patterns in both ethylene and drought induced cassava leaf abscission. Totally, 34 GSTs were detected to express in both ethylene and drought induced leaf abscission in cassava abscission zones. Comparison of GST expression profiling between ethylene and drought induced leaf abscission suggested that Tau GST genes showed with the similar expression in both treatments induced leaf abscission in cassava abscission zone. GO annotation indicated that all 17 Tau GST genes participated in the pathway of toxin catabolism (GO: 0009407). The expression levels of 17 Tau MeGST genes were analyzed in two cassava cultivars, ‘SC124’ and ‘Arg7’, the two cultivars exhibit different levels of leaf abscission when suffered from the same environmental stress. Higher expression levels of Tau MeGSTs were detected in the precocious abscission Arg7 cultivar, while lower expression levels in delayed abscission SC124 cultivar. All the results indicated that Tau MeGSTs have the function in regulation the cassava leaf abscission under environmental stresses. Conclusion Analysis of the expression patterns of GSTs in various abscission-promoting treatments in cassava abscission zones helps us to understand the possible roles of GSTs in cassava leaf abscission.

Erecting underground structures in regions with unidentified weak layers, cavities, and faults is highly dangerous and potentially disastrous. An efficient and accurate near-surface exploration method is thus of great significance for guiding construction. In near-surface detection, imaging methods suffer from artifacts that the complex structure caused and a lack of efficiency. In order to realize a rapid, accurate, robust near-surface seismic imaging, a minimum variance spatial smoothing (MVSS) beamforming method is proposed for the seismic detection and imaging of underground geological structures under a homogeneous assumption. Algorithms such as minimum variance (MV) and spatial smoothing (SS), the coherence factor (CF) matrix, and the diagonal loading (DL) methods were used to improve imaging quality. Furthermore, it was found that a signal advance correction helped improve the focusing effect in near-surface situations. The feasibility and imaging quality of MVSS beamforming are verified in cave models, layer models, and cave-layer models by numerical simulations, confirming that the MVSS beamforming method can be adapted for seismic imaging. The performance of MVSS beamforming is evaluated in the comparison with Kirchhoff migration, the DAS beamforming method, and reverse time migration. MVSS beamforming has a high computational efficiency and a higher imaging resolution. MVSS beamforming also significantly suppresses the unnecessary components in seismic signals such as S-waves, surface waves, and white noise. Moreover, compared with basic delay and sum (DAS) beamforming, MVSS beamforming has a higher vertical resolution and adaptively suppresses interferences. The results show that the MVSS beamforming imaging method might be helpful for detecting near-surface underground structures and for guiding engineering construction.

Injecting grout into the gaps between tunnel shield segments and surrounding rocks can reduce ground subsidence and prevent ground water penetration. However, insufficient grouting and grouting defects may cause serious geological disasters. Ground penetrating radar (GPR) is widely used as a nondestructive testing (NDT) method to evaluate grouting quality and determine the existence of defects. This paper provides an overview of GPR applications for grouting defect detection behind tunnel shield segments. State-of-the-art methodologies, field cases, experimental tests and signal processing methods are discussed. The reported field cases and model test results show that GPR can detect grouting defects behind shield tunnel segments by identifying reflected waves. However, some subsequent problems still exist, including the interference of steel bars and small differences in the dielectric constants among media. Recent studies have focused on enhancing the signal-to-noise ratio and imaging methods. Advanced GPR signal processing methods, including full waveform inversion and machine learning methods, are promising for detecting imaging defects. Additionally, we conduct a preliminary experiment to investigate environmental noise, antenna configuration and coupling condition influences. Some promising topics, including multichannel configuration, rapid evaluation methods, elastic wave method scanning equipment for evaluating grout quality and comprehensive NDT methods, are recommended for future studies.

The enhancement of the detection of weak signals against a strong noise background is a key problem in local gear fault diagnosis. Because the periodic impact signal generated by local gear damage is often modulated by high-frequency components, fault information is submerged in its envelope signal when demodulating the fault signal. However, the traditional bistable stochastic resonance (BSR) system cannot accurately match the asymmetric characteristics of the envelope signal because of its symmetrical potential well, which weakens the detection performance for weak faults. In order to overcome this problem, a novel method based on underdamped asymmetric periodic potential stochastic resonance (UAPPSR) is proposed to enhance the weak feature extraction of the local gear damage. The main advantage of this method is that it can better match the characteristics of the envelope signal by using the asymmetry of its potential well in the UAPPSR system and it can effectively enhance the extraction effect of periodic impact signals. Furthermore, the proposed method enjoys a good anti-noise capability and robustness and can strengthen weak fault characteristics under different noise levels. Thirdly, by reasonably adjusting the system parameters of the UAPPSR, the effective detection of input signals with different frequencies can be realized. Numerical simulations and experimental tests are performed on a gear with a local root crack, and the vibration signals are analyzed to validate the effectiveness of the proposed method. The comparison results show that the proposed method possesses a better resonance output effect and is more suitable for weak fault feature extraction under a strong noise background.

Abstract The mitogen-activated protein kinase (MAPK) cascade, which is a major signal transduction pathway widely distributed in eukaryotes, has an important function in plant development and stress responses. However, less information is known regarding the MAPKKK and MAPKK gene families in the important fruit crop banana. In this study, 10 MAPKK and 77 MAPKKK genes were identified in the banana genome, and were classified into 4 and 3 subfamilies respectively based on phylogenetic analysis. Majority of MAPKKK and MAPKK genes in the same subfamily shared similar gene structures and conserved motifs. The comprehensive transcriptome analysis indicated that MAPKKK-MAPKK genes is involved in tissue development, fruit development and ripening, and response to abiotic stress of drought, cold and salt in two banana genotypes. Interaction networks and co-expression assays demonstrated that MAPK signaling cascade mediated network participates in multiple stress signaling, which was strongly activated in Fen Jiao (FJ). The findings of this study advance understanding of the intricately transcriptional control of MAPKKK-MAPKK genes and provide robust candidate genes for further genetic improvement of banana.

Expansive soil is a kind of clay soil that expands rapidly after being immersed in water and shrinks significantly after losing water. The expansive soil slopes exposed to the atmosphere are in the process of continuous dry and wet cycles and are extremely unstable under the conditions of rainfall. Based on saturated-unsaturated seepage theory, the unsaturated seepage process and humidification process of the expansive soil slope under rainfall were simulated, and a related FORTRAN language program was developed. The effects of matrix suction changes, seepage softening, and moistening expansion during the seepage process were considered. The effects of strength attenuation, seepage softening, and moistening expansion on the overall stability of the expansive soil slope were analyzed. The results show that under the action of rainfall, the failure mode of the expansive soil slope after multiple dry-wet cycles is shallow collapse, and the failure surface is located in the weathered area, which is quite different from the traction landslide of the homogeneous expansive soil slope. After considering the moistening expansion and softening effects, the maximum displacement of the slope increased by an order of magnitude, and the safety factor decreased significantly. The significant increase in the horizontal stress of the lower half slope is the main reason for the landslide. The research results well explain the special tractive and shallow properties of typical expansive soil landslides.

Karst geological anomalies at pile locations significantly affect the bearing capacity and construction safety of the piles, posing a significant challenge for urbanization. Borehole geophysical methods are required to extend the detection range and identify karst voids that are at pole locations and near drilled boreholes. In this paper, we developed a near offset and small diameter single borehole ground penetration radar (GPR) prototype. A signal processing method combining complex signal analysis by Hilbert transform (HT) and medium filtering was suggested to differentiate the weak backscattered wave from borehole background noise. A controlled horizontal borehole experiment was used to demonstrate the applicability of the prototype and the advantages of the signal analysis method prior to application in a real project. The controlled test presented three typical wave events corresponding to a soil–rock interface, rock fractures, and karst voids. Field tests were conducted at a freeway bridge extension project in an urban karst area. Multiple karst voids, sinkholes, rock fractures, and integrated bedrock were identified by analysis of four typical detection scenarios. The remediation of the karst voids and a rotary bored piling with real-time steel casing construction strategy were designed based on the investigation results. The construction feedback demonstrates that single borehole radar detection is effective for the investigation of anomalies at pile locations in urban karst areas.

Panax notoginseng (Sanqi), a traditional Chinese medical drug which has been applied to medical use for over four centuries, contains high content of dammarane-type tetracyclic triterpenoid saponins. A number of stereoisomeric dammarane-type saponins exist in this precious herb, and some are particularly regarded as “biomarkers” in processed notoginseng. Contemporary researches have indicated that some saponin stereoisomers may show stereospecific pharmacological activities, such as anti-tumor, antioxidative, anti-photoaging, anti-inflammatory, antidiabetic, and neuro-protective activities, as well as stereoselective effects on ion channel current regulation, cardiovascular system, and immune system. The current review provides a comprehensive overview of chemical compositions of raw and processed P. notoginseng with a particular emphasis on saponin stereoisomers. Besides, the pharmacological and pharmacokinetic researches, as well as determination and biotechnological preparation methods of stereoisomeric saponins in notoginseng are discussed extensively.

As a crucial living environment, the Internet shapes cognition. The Internet provides massive information that can be accessed quickly via hyperlinks, but the information is typically fragmentary and concrete rather than integrative. According to construal level theory, the processing of this concrete and fragmentary information, should reduce attentional scope. Two experiments were conducted to test this hypothesis. In Experiment 1, three groups of participants were asked to shop online, read magazines or have a rest respectively, and a divided attention Navon-letter task was employed to measure the attentional scope before and after the assigned activity. It was found that the difference between reaction times in response to local vs. global targets was decreased only after Internet use, while there was no decrease in either the reading or resting group. In Experiment 2, the same procedure was used, and EEG/ERP methods were used to record both behavioral response and neural activity. Results showed that before the assigned activity, there was no significant difference in N2 amplitude in response to local vs. global targets in any of the three groups; during the activity, the lower-alpha activity induced by Internet use was significantly lower than that induced by reading or resting; after the activity, correspondingly, a more negative N2 wave was induced by the global than local targets only in the Internet group, while there were no significant differences in the other groups. Consistent with construal level theory, the results suggest that when surfing the Internet, attentional scope is reduced, and this effect might continue after Internet activity.
