Objective. To explore the ability of asiatic acid to interfere with the invasion and proliferation of breast cancer cells by inhibiting WAVE3 expression and activation through the PI3K/AKT signaling pathway. Methods. The MDA-MB-231 cells with strong invasiveness were screened by transwell assay, and plasmids with high expression of WAVE3 were constructed for transfection. The transfection effect and protein expression level of plasmids were verified by PCR and WB. The effects of asiatic acid on cell proliferation and invasion were investigated by flow cytometry. The xenografted tumor models in nude mice were established to study the antitumor activity of asiatic acid. Results. Asiatic acid significantly inhibited the activity of MDA-MB-231 cells, and the expression level of WAVE3 increased significantly in the tissue of ductal carcinoma in situ and was lower than that in the metastasis group. After plasmid transfection, the mRNA and protein expression of WAVE3 increased significantly in the cells. Asiatic acid at different concentrations had an impact on cell apoptosis and invasion and could significantly inhibit the expression of WAVE3, P53, p-PI3K, p-AKT, and other proteins. The T/C(%) of asiatic acid (50 mg/kg) for MDA-MB-231(F10) xenografted tumor in nude mice was 46.33%, with a tumor inhibition rate of 59.55%. Asiatic acid could significantly inhibit the growth of MDA-MB-231 (F10) xenografted tumors in nude mice (p<0.05). Conclusions. Asiatic acid interferes with the ability of breast cancer cells to invade and proliferate by inhibiting WAVE3 expression and activation and the mechanism of action may be related to the PI3K/AKT signaling pathway.

Two new ent-kaurene diterpenoids, 15α,20-dihydroxy-6,7-seco-entkaur-16-ene-7,1α(6,11α)-diolide (1), 6β-butyroxy-3β-hydroxy-6,7-seco-6,20-epoxy-7,1α-olide-entkaur-16-en-15-one (2), together with 25 known compounds, 3–27, were isolated from the leaves of Isodon japonicus. Their structures were established by spectroscopic methods, including 2D-NMR techniques.

On the basses of analyzing the defects of variable blank holder force hydraulic press on construction and its rigid being not enough,the realization of designing method,working principle,hydraulic and system for ne controlling w type upper multi-point deep drawing with variable blank—holding force controlled by computer was introduced.In order to verify the effect of application of this set up,the testing results of deep drawing a rectangular box and a cylindrical cup formed component was presented. The results show that the equipment has low cost, general in using and optimal efficient of controlling.

6. Study on Cold Precision Forging of Non Straight Wall Cavity Piston

Brake piston is a huge demand non straight wall cavity part for the typical automotive industry; the traditional processing method is machine processing, or preforming by cold extrusion, and then machining. In this paper, the combined cold precision forging method of cold extrusion and spinning was proposed, which might improve the overall performance of parts and reduce costs. The rigid plastic finite element model of cold extrusion and spinning was established to simulate the forming process. The results showed that the combined cold precision forging method was available to manufacture non-straight wall cavity piston.

Two new ent -kaurene diterpenoids, 15α-acetoxyl-6,11α-epoxy-6α-hydroxy-20-oxo-6,7-seco- ent -kaur-16-en-1,7-olide ( 1 ), 15α-hydroxy-20-oxo-6,7-seco- ent -kaur-16-en-1,7α(6,11α)-diolide ( 2 ), together with ten known compounds ( 5 – 14 ) were isolated from the leaves of Isodon rubescens . Their structures were elucidated mainly by various spectroscopic techniques and finally confirmed by single-crystal X-ray diffraction. Compounds 1 , 2 , 8 and 12 were evaluated for their cytotoxicities against EC-1, U87, A549, MCF-7 and Hela cell lines.

8. Analysis on SNPs linked with wood properties of Populus nigraL. gene resources

In this study, 115 Populus nigra L. collected from Europe were used as materials. They were detected for SNPs of genes linked with lignin and holocellulose (4CL, PAL and CesA2) by TaqMan technology and correlations between SNPs and wood properties (the wood speciely gravity, fiber length, fiber width, microfibre angle, lignin, holocellulose content and a-Cellulose content) were also studied. results showed that (1) 27 SNPs were obtained in the genes, such as 4CL, PAL and CesA2 associated with lignin synthesis. Among them, 17 SNPs were transition (A-G, C-T), 10 SNPs were transversion (A-C, G-C, G-T, and A-T). (2) Three of the SNPs were discriminated, a significant negative correlation between holocellulose content of four-year old P. nigra and SNP1 was detected, and contribution ratio was 11.11%. SNP2 and SNP3 had no significant correlation with wood properties. (3) Wood properties of various genotypes of SNP1 were significantly different. CC and CT genotype relative to TT genotype had larger holocellulose content. SNP1 would be an efficient marker to choose P. nigra gene resources that have larger holocellulose content.

It is difficult in the description of rock cuttings' lithology category with a traditional manual method since PDC bits have been widely used in the process of drilling. To recognize the tiny cuttings' lithology category effectively, a digital compact system based on high-resolution color linear CCD and microphotography is established and introduced in this paper. The system is mainly constructed by high resolution color linear CCD, micro lens, white LED light sources and conveyor. The Color linear CCD with up to 4080 pixels per line combined with micro lens leads to a resultant reproduction ratio of 1:1. It ensures that tiny cuttings' images can be captured clearly, and hence the detailed information of cuttings' color and texture can be obtained. With the linear scanning, we can get images with both high-resolution and larger field of view than array CCD. Optimal parameter configuration is determined by the experiments of hardware control program and optical parameter adjustment. The software to recognize cuttings' lithology category is developed by extracting multi-character values from the image information and by using the LIBSVM classifier. With the optimal parameter configuration and the developed recognition software, the cuttings' lithology category has been well described with a average recognition rate about 95% for mudstones and about 90% for sandstones, which proves that the cuttings' images captured by the high-resolution linear CCD meet the requirement of the subsequent recognition algorithm. The obtained results show that the digital compact system is greatly potential to be used in oil field logging.

12. A new guaianolide from the roots of Pterocypsela elata

In order to find more biologically active substances, we have carefully investigated the chemical constituents of Pterocypsela elata. One new guaianolide, (4 S )-11-methoxycarbonyl-guaiane-1(10),5(6),7(11),8(9)-tetraen-6,12-olide was isolated from the EtOAc extracts of the roots of P. elata. Its structure was elucidated by various spectroscopic methods and confirmed by X-ray crystallographic analysis.

The fractionated crystallization behavior of the minor dispersed HDPE phase in PS/POE/HDPE/SBS quaternary blends was investigated by differential scanning calorimetry (DSC). Interestingly, we found that the fractionated crystallization behavior of HDPE was molecular weight dependent. At a fixed composition, HDPE with lower molecular weight showed more obvious fractionated crystallization behavior than HDPE with higher molecular weight. This was ascribed to a finer dispersion of HDPE with lower molecular weight, as evidenced by SEM observations. The fractionated crystallization behavior of HDPE in the blends became less obvious with increasing of its content, due to a change of phase morphology from droplet to co-continuous structure. Correspondingly, a change of tensile toughness of the blends from brittle to ductile mode was observed. [ABSTRACT FROM AUTHOR]
