Fiber optic localized surface plasmon resonance (LSPR) sensors have become an effective tool in refractive index (RI) detection for biomedical applications because of their high sensitivity. However, using conventional optical fiber has caused limitations in implanting the sensor in the body. This research presents the design and construction of a new type of polymer-based LSPR sensors to address this issue. Also, finite element method (FEM) is used to design the sensor and test it theoretically. The proposed polymer optical fiber (POF) based on citrate is biocompatible, flexible, and degradable, with a rate of 22% and 27 over 12 days. The step RI structure utilizes two polymers for light transmission: poly (octamethylene maleate citrate) (POMC) as the core and poly (octamethylene citrate) (POC) as the cladding. The POF core and cladding diameters and lengths are 700 µm, 1400 µm, and 7 cm, respectively. The coupling efficiency of light to the POF was enhanced using a microsphere fiber optic tip. The obtained results show that the light coupling efficiency increased to 77.8%. Plasma surface treatment was used to immobilize gold nanoparticles (AuNPs) on the tip of the POF, as a LSPR-POF sensor. Adsorption kinetics was measured based on the pseudo-first-order model to determine the efficiency of immobilizing AuNPs, in which the adsorption rate constant (k) was obtained be 8.6 × 10 -3 min -1 . The RI sensitivity of the sensor in the range from 1.3332 to 1.3604 RIU was obtained as 7778%/RIU, and the sensitivity was enhanced ~ 5 times to the previous RI POF sensors. These results are in good agreement with theory and computer simulation. Mesenchymal stem cells (MSCs), derived from various tissues, are served as a promising source of cells in clinic and regenerative medicine. Umbilical cord-Wharton's jelly (WJ-MSCs)-derived MSCs exhibit advantages over those from adult tissues, such as no ethical concerns, shorter population doubling time, broad differentiation potential, readily available non-invasive source, prolonged maintenance of stemness properties. The aim of this study was to evaluate the effect of MRI (1.5 T, 10 min) on stemness gene expression patterns (OCT-4, SOX-2, NANOG) of WJ-MSCs. Additionally, we assessed cell viability, growth kinetics and apoptosis of WJ-MSCs after MRI treatment. The quantitative real-time reverse transcription polymerase chain reaction (qRT-PCR) data showed that transcript levels of SOX-2, NANOG in MRI-treated WJ-MSCs were increased 32- and 213-fold, respectively. MTT assay was performed at 24, 48, and 72 h post-treatment and the viability was not significantly different between the two groups. The doubling time of the MRI group was markedly higher than the control group. In addition, the colony formation ability of WJ-MSCs after MRI treatment significantly increased. Furthermore, no change in apoptosis was seen before or after MRI treatment. Our results suggest that the use of MRI can improve the quality of MSCs and enhance the efficacy of mesenchymal stem cell-based therapies., (© 2022. Aim: To assess the association serum levels of selenium (Se) and copper (Cu) with symptoms and IgG immune response to SARS-CoV-2. Patients & methods/materials: Blood samples and nasopharyngeal swabs were obtained from 126 COVID-19 patients with mild and severe symptoms. The serum levels of Cu and Se were measured by atomic-absorption spectrophotometry. Results & conclusion: Mean Se was higher in patients with mild symptoms and IgG nonresponders, whereas mean Cu was higher in patients with severe symptoms and IgG responders. The Cu/Se ratio was lower in patients with no IgG responses to infection and mild symptoms versus IgG responders with severe symptoms. These results suggest the Cu/Se ratio as a nutritional biomarker of severity and IgG immune response in COVID-19 patients.

Background: This study aimed to evaluate the expression of Liver X receptor (Lxr), Sirtuin 1 (Sirt1), apoptotic-related genes, and the protective role of N-acetylcysteine (NAC) in the liver of rats treated with Lead (Pb)., Methods: Rats were randomly divided into 5 groups, including G1 (control), G2 (single dose of Pb), G3 (continuous dose of Pb), G4 (single dose of Pb + NAC), and G5 (continuous dose of Pb + NAC). Lipid profiles and liver specific enzymes were assessed. Expression of Lxr, Sirt1, Bax and Caspase-3 genes was considered using RT-PCR., Results: Exposure to Pb caused a significant accumulation of Pb in the blood and liver tissue, increase in serum AST, ALT and ALP enzymes, as well as lipid profiles. Chronic exposure to Pb caused a significant decrease in Lxr (3.15-fold; p < 0.001) and Sirt1 (2.78-fold; p = 0.009), but significant increase in expression of Bax (4.49-fold; p < 0.001) and Caspase-3 (4.10-fold; p < 0.001) genes when compared to the control. Combined therapy with Pb + NAC in rats caused a significant decrease in AST, ALT and ALP values (28.93%, 20.80% and 28.86%, respectively) in the blood as compared to rats treated with Pb alone. Co-treated with Pb + NAC significantly increased the expression of Lxr (1.72-fold; p = 0.043) and Sirt1 (2.45-fold; p = 0.008), but decreased the expression of Bax (1.96-fold; p = 0.03) and Caspase 3 (2.22-fold; p = 0.029) genes when compared to rats treated with Pb alone., Conclusion: Chronic exposure to Pb is strongly associated with accumulation of Pb in the blood and liver, hepatic cells apoptosis, down-expression of Lxr and Sirt1 genes and consequently liver injury and abnormal lipid profiles. The aim of the "Smuovi La Salute" ("Shake Your Health") project was to implement an integrated and comprehensive model to prevent and treat overweight and obesity in low socioeconomic status (SES) and minority groups living in three different districts in the north of Italy. An app and a cookbook promoting transcultural nutrition and a healthy lifestyle were developed, and no-cost physical activities were organized. Healthy lifestyle teaching was implemented in 30 primary school classrooms. Learning was assessed through pre- and post-intervention questionnaires. At the Obesity Pediatric Clinic, overweight and obese children of migrant background or low SES were trained on transcultural nutrition and invited to participate in the project. Primary school students increased their knowledge about healthy nutrition and the importance of physical activity ( p -value < 0.001). At the Obesity Pediatric Clinic, after 6 months, pre-post-intervention variation in their consumption of vegetables and fruit was +14% ( p < 0.0001) and no variation in physical activity habits occurred ( p = 0.34). In this group, the BMI z -score was not significantly decreased (-0.17 ± 0.63, p = 0.15). This study demonstrates the feasibility and efficacy of telematic tools and targeted community approaches in improving students' knowledge with regard to healthy lifestyle, particularly in schools in suburbs with a high density of migrants and SES families. Comprehensive and integrated approaches provided to the obese patients remain mostly ineffective.

In this study, a signal-on nanobiosensor based on bivalent aptamer-Cu nanocluster was designed and optimized for specific and sensitive detection of VEGF 165 . The VEGF 165 is known as a promising biomarker in different diseases such as cancer in the angiogenic stage. Detection and quantification of VEGF 165 is a crucial step in diagnosis and monitoring the treatment plan. The represented nanostructure consists of multimerized VEGF 165 aptamer joint with ssDNA based linker in the middle and poly thymine sequences on both 3' and 5' ends as a template for Cu-nanocluster supraparticle formation. This self-assembled structure leads to accurate controlling of aggregation in the presence of VEGF 165 . This study is the first report for Cu nanocluster nucleation on ploy thymine tails of ssDNA which performed in two reduction steps to form stable CuNC supraparticle. The sensing strategy was designed based on the target-induced structure switching mode of the aptamer. In the presence of VEGF 165 , due to self-assembly induced emission and aggregation-induced emission phenomena this nanostructure depicted the visible wavelength shift and enhancement in the fluorescence emission intensity. Also, the results of the analytical performance of this nanobiosensor indicated the LOD of 12 pM which revealed high rate sensitivity. This aptasensor exhibited stability and decent response linearity range (10-800 pM, R 2 = 0.9943). The selectivity and specificity assessment showed high discriminant capability in the real serum sample. In conclusion, this signal-on nanobiosensor provides a facile, sensitive and reliable assay for clinical monitoring of the VEGF 165 concentration in serum without further sample preparation., (Copyright © 2019 Elsevier B.V. Objectives: In recent years, polypropyleneimine (PPI) dendrimers have attracted great interest as non-viral gene delivery systems because of their attractive features including highly branched architecture with number of reactive end groups. However, without being structurally modified, they are not efficient gene carriers. In the present study, generation 2 and 3 (G2 and G3) of PPI dendrimers were conjugated with alkylcarboxylate groups as linker to enhance the transfection efficiency while maintaining their low cell toxicity., Materials and Methods: First, 10-bromodecanoic acid was covalently attached to all available surface primary amines of PPI G2 and G3 to increase their lipophilicity. In the subsequent step, PPIs were conjugated to the alkylcarboxylate groups of alkylcarboxylate-PPI derivatives to increase the number of surface primary amines. Physicochemical properties of modified PPIs were determined. Transfection experiments (using both luciferase and green fluorescent protein (GFP)- expressing plasmids) and cytotoxicity assay were performed to evaluate the efficiency of the final derivatives., Results: Fabricated vectors condensed DNA effectively so that polyplexes with appropriate size (below 155 nm) and positive surface charge were constructed. Cross-linked low molecular weight PPIs (G2 or G3) with decanoate linkage increased transfection efficiency significantly while maintaining the low cytotoxicity. PPI G2 derivative exhibited increased buffering capacity which is believed to be responsible for better proton sponge mechanism leading to higher transfection efficiency., Conclusion: Our results indicated that oligomerization of low molecular weight PPI (PPI G2-alkyl-PPI G2 conjugate) could be an approach to increase the transfection efficiency and to lower the cytotoxicity of low molecular weight polycations.

Background & Aims: The presence of pathogenic microorganisms in the dust storm can cause diseases such as Asthma, Pneumonia, and respiratory infections. The aim of this study was to determine the relationship between air-borne particles with airborne microorganisms in normal and dusty days in Sanandaj, a city located in the west of Iran., Materials & Methods: Air sampling was conducted during the normal and dusty days through Andersen single-stage impactor (28.3 L/min) for 2.5 min. Air particles concentration (PM10) was measured daily and microbial sampling was also conducted on every six days and on the dusty days. Finally, the data was analyzed by SPSS-16 (ANOVA and paired T-tests)., Results: The concentration of airborne microorganisms (bacteria and fungi) was increased by an increase of the airborne particles. Particles concentration in May, June and July (twice per month) was more than of the standard value. The predominant species of bacteria and fungi during the occurrence of Dust storm was Bacillus spp. (56.2% of total bacteria) and Mycosporium spp. (28.6% of total fungi), respectively., Discussion and Conclusion: The results showed that the number of airborne microorganisms (bacteria and fungi) increased during the dust storm. Therefore, the microorganisms in the dust storm can cause biological harmful effects on human health.
