International audience; The GMO90+ project was designed to identify biomarkers of exposure or health effects in Wistar Han RCC rats exposed in their diet to 2 genetically modified plants (GMP) and assess additional information with the use of metabolomic and transcriptomic techniques. Rats were fed for 6-months with 8 maize-based diets at 33% that comprised either MON810 (11% and 33%) or NK603 grains (11% and 33% with or without glyphosate treatment) or their corresponding near-isogenic controls. Extensive chemical and targeted analyses undertaken to assess each diet demonstrated that they could be used for the feeding trial. Rats were necropsied after 3 and 6 months. Based on the Organization for Economic Cooperation and Development test guideline 408, the parameters tested showed a limited number of significant differences in pairwise comparisons, very few concerning GMP versus non-GMP. In such cases, no biological relevance could be established owing to the absence of difference in biologically linked variables, dose-response effects, or clinical disorders. No alteration of the reproduction function and kidney physiology was found. Metabolomics analyses on fluids (blood, urine) were performed after 3, 4.5, and 6 months. Transcriptomics analyses on organs (liver, kidney) were performed after 3 and 6 months. Again, among the significant differences in pairwise comparisons, no GMP effect was observed in contrast to that of maize variety and culture site. Indeed, based on transcriptomic and metabolomic data, we could differentiate MON- to NK-based diets. In conclusion, using this experimental design, no biomarkers of adverse health effect could be attributed to the consumption of GMP diets in comparison with the consumption of their near-isogenic non-GMP controls.

International audience; The antifungal potency of the essential oils of Rhanterium adpressum was evaluated against four mycotoxigenic strains of the genus Fusarium. The essential oils were obtained, separately, by hydro-distillation of the aerial parts of R. adpressum (leaves and flowers). The parts were collected during the period of bloom (3 months) for 3 years. The GC-MS analysis revealed thirty-six compounds for the essential oils, divided into four classes of chemical compounds, with variable percentages according to the month of extraction. The monoterpene hydrocarbons form the main class in these oils. On the other hand, the highest percentages of the oxygenated compounds are observed in the samples collected during the month of May. The direct contact method was used to evaluate the antifungal activity of the essential oils. The activity can be attributed to their relatively high composition of oxygenated monoterpenes. Flowers extract showed strong inhibitory activity, with very interesting concentrations of IC50 and MIC for both tests on solid and liquid medium. The effect of these oils on the production of type B trichothecenes (TCTBs) was evaluated, showing a significant inhibitory effect on TCTBs production, for both extracts (leaves and flowers). The rates of inhibition were 66-97 and 76-100% of FX, 3-ADON and 15-ADON, respectively. The inhibition of fungal biomass and the production of TCTBs depended on the used concentration of the essential oils. These results suggest that the essential oils from R. adpressum are able to control the growth of the tested strains and their subsequent production of TCTB mycotoxins.

Chronic pain is associated with anxiety and depression episodes. The amygdala plays a key role in the relationship between emotional responses and chronic pain. Here, we investigated the role of Acid-Sensing Ion Channels 1a within the basolateral amygdala (BLA), in pain and associated anxiety in a rat model of monoarthritis (MoAr). Administration within the BLA of PcTx1 or mambalgin-1, two specific inhibitors of ASIC1a-containing channels significantly inhibited pain and anxiety-related behaviours in MoAr rats. The effect of PcTx1 was correlated with a reduction of c-Fos expression in the BLA. We examined the expression profile of ASICs and other genes in the amygdala in MoAr and sham animals, and found no variation of the expression of ASIC1a, which was confirmed at the protein level. However, an increase in the BLA of MoAr rats of both PI3Kinase mRNA and the phosphorylated form of Akt, along with Bdnf mRNA, suggest that the BDNF/PI3-kinase/Akt pathway might regulate ASIC1a in BLA neurons as demonstrated in spinal sensitisation phenomenon. We also observed changes in several kinase mRNAs expression (PICK1, Sgk1) that are potentially involved in ASIC1a regulation. These results show a crucial role of ASIC1a channels in the BLA in pain and anxiety-related behaviours during arthritis.

International audience; Plastic pollution represents a threat for biological communities and the ecological functions they provide in river ecosystems. In this study, we compared the microbial colonization of two plastics (biodegradable and non-biodegradable) and three natural substrata (leaves, sediment and rocks) in two study sites of an urbanized watershed differing in their plastic-contamination degree (upstream and downstream). The density and diversity of bacterial, fungal and algal communities, as well as the extracellular enzymatic activities β-glucosidase (GLU), N-acetyl-glucosaminidase (NAG) and phosphatase (PHO), were analyzed in each substrata and site over a four-week colonization experiment. Results showed higher microbial densities and enzymatic activities in leaves and sediment compared to plastics and rocks, probably due to the greater availability of organic carbon and nutrients in the former substrata. However, the microbial colonization of the two plastics was only different in the downstream site where bacterial density and enzymatic activities were higher in the biodegradable compared to the non-biodegradable plastic. Accordingly, the presence of biodegradable plastics would enhance the heterotrophic metabolism in plastic-polluted rivers.

International audience; L’expérimentation menée depuis 2016 dans l’unité Inrae à Alénya (Pyrénées-Orientales) par une équipe pluridisciplinaire a pour but de comprendre la technique d’ensilage autrement qu’au travers de l’analyse des fosses-silos abandonnées. La mise en place du protocole expérimental nous a conduits à mettre au cœur de nos préoccupations les qualités sanitaires, alimentaires et germinatives des stocks de grains expérimentaux. C’est l’expertise croisée d’agriculteurs céréaliers et de professionnels du stockage qui aide l’équipe d’archéologues à comprendre les résultats de l’ensilage expérimental. Les données anciennes et récentes en archéologie, en histoire et en ethnographie donnent quant à elles des pistes sur les différentes mises en œuvre et solutions techniques. L’objectif à terme est de produire des résultats et une méthodologie à destination des chercheurs (archéologues, historiens, agronomes) et éventuellement des solutions de stockage alternatives pour les producteurs et les transformateurs de céréales.

International audience; A series of novel 2,9-bis[(substituted-aminomethyl)]-4,7-phenyl-1,10-phenanthroline derivatives was designed, synthesized, and evaluated in vitro against three protozoan parasites (Plasmodium falciparum, Leishmania donovani and Trypanosoma brucei brucei). Pharmacological results showed antiprotozoal activity with IC50 values in the sub and μM range. In addition, the in vitro cytotoxicity of these original molecules was assessed with human HepG2 cells. The substituted diphenylphenanthroline 1l was identified as the most potent antimalarial derivative with a ratio of cytotoxic to antiparasitic activities of 505.7 against the P. falciparum CQ-resistant strain W2. Against the promastigote forms of L. donovani, the phenanthrolines 1h, 1j, 1n and 1o were the most active with IC50 from 2.52 to 4.50 μM. The phenanthroline derivative 1o was also identified as the most potent trypanosomal candidate with a selectivity index (SI) of 91 on T. brucei brucei strain. FRET melting and native mass spectrometry experiments evidenced that the nitrogen heterocyclic derivatives bind the telomeric G-quadruplexes of P. falciparum and Trypanosoma. Moreover, as the telomeres of the parasites P. falciparum and Trypanosoma could be considered to be possible targets of this kind of nitrogen heterocyclic derivatives, their potential ability to stabilize the parasitic telomeric G-quadruplexes have been determined through the FRET melting assay and by native mass spectrometry.

Human poisoning due to consumption of seafood contaminated with phycotoxins is a worldwide problem, and routine monitoring programs have been implemented in various countries to protect human consumers. Following successive episodes of unexplained shellfish toxicity since 2005 in the Arcachon Bay on the French Atlantic coast, a national research program was set up to investigate these atypical toxic events. Part of this program was devoted to fit-for-purpose cell-based assays (CBA) as complementary tools to collect toxicity data on atypical positive-mouse bioassay shellfish extracts. A collaborative study involving five laboratories was conducted. The responses of human hepatic (HepG2), human intestinal (Caco2), and mouse neuronal (Neuro2a) cell lines exposed to three known lipophilic phycotoxins-okadaic acid (OA), azaspiracid-1 (AZA1), and pectenotoxin-2 (PTX2)-were investigated. A screening strategy composed of standard operating procedures and a decision tree for dose-response modeling and assay validation were designed after a round of "trial-and-error" process. For each toxin, the shape of the concentration-response curves and the IC(50) values were determined on the three cell lines. Whereas OA induced a similar response irrespective of the cell line (complete sigmoid), PTX2 was shown to be less toxic. AZA1 induced cytotoxicity only on HepG2 and Neuro2a, but not on Caco2. Intra- and inter-laboratory coefficients of variation of cell responses were large, with mean values ranging from 35 to 54 % and from 37 to 48 %, respectively. Investigating the responses of the selected cell lines to well-known toxins is the first step supporting the use of CBA among the panel of methods for characterizing atypical shellfish toxicity. Considering these successful results, the CBA strategy will be further applied to extracts of negative, spiked, and naturally contaminated shellfish tissues.

International audience; Many plant-pathogenic often produce toxic metabolites when grow on foods and feeds. Some antioxidative components seemed to prevent fungi growth and mycotoxin formation. Recently, we had synthezised a new class of powerful antioxidative compounds, i.e. tetrahydrocurcuminoids, and structure/antioxydant activity relationships had been established. The South West of France is known to produce large amounts of corn, which can be infested by Fusarium species, and particularly F. proliferatum. In this context, the efficiency of tetrahydrocurcuminoids, which can be obtained from natural curcuminoids, was investigated to control in vitro the growth of F. proliferatum and the production of its associated mycotoxin fumonisin B1. The relation between structure and antifungal activity was particularly studied. Tetrahydrocurcumin THC1, with two guaiacyl phenolic subunits, showed the highest inhibitory activity (measured as the radial growth on agar medium) against the F. proliferatum development (67% inhibition at a concentration of 13.6 μmol.mL-1). The efficiencies of THC2 (36% at a concentration of 11.5 μmol.mL-1), which contains syringyl phenolic units and THC3 (30% at a concentration of 13.6 μmol.mL-1), which does not have any substituent on the aromatic rings were relatively close. These results indicate that the simultaneous presence of guaiacyl phenols and the enolic function of the β-diketone moiety play an important role in the inhibition mecanisms. The importance of this combination was confirmed by using n-propylguaiacol and acetylacetone as molecular models. In the same conditions, ferulic acid and eugenol, other natural phenolic antioxidants, were less efficient in inhibiting fungal growth. THC1 also reduced fumonisin B1 production in liquid medium by approximately 35%, 50%, and 75% at concentrations of 0.8, 1.3, and 1.9 μmol.mL-1, respectively. These very low inhibitory concentrations show that tetrahydrocurcuminoids could be one of the most promising biobased molecules for the control of mycotoxinogen fungal strains.

International audience; Reducing production of trichothecene B by Fusarium graminearum on cereals is necessary to avoid contamination leading to yields reduction and having harmful impacts on human and animal health. Understanding how trichothecenes biosynthesis is induced is essential. Effect of ambient pH on fungal growth, toxin biosynthesis and TRI genes expression was studied during in vitro liquid culture of F. graminearum on minimal medium. Fungal development stopped at day 3 after a sharp pH drop in the medium. At the same time, induction of TRI genes expression was observed and toxin started to accumulate one day later. Acidification seems a determinant factor for induction, as neither the toxin nor the TRI genes were detected when the pH is maintained neutral. Shifting from neutral to acidic pH by mycelium transfer induced TRI genes expression and toxin accumulation. The regulation of toxin production by the ambient pH seemed to be specific to some TRI genes since TRI5, located in the core FgTRI5 cluster, showed an immediate induction while TRI101, located elsewhere in the genome, showed a more progressive response. The regulation of trichothecenes biosynthesis by the ambient pH seems to be a general mechanism, independent from strains or chemotype as all tested strains, including F. graminearum and F. culmorum species, showed a regulation of toxin production in response to the ambient pH. We conclude that in vitro, external acidification is required for induction of TRI genes expression.

International audience; We defined reliable and sensitive PCR assays to specifically detect trichothecene-producing Fusarium spp. in milling fractions and kernel tissues of naturally infected durum wheat. The assays were based on a combination of primers derived from the trichodiene synthase and the β-tubulin genes. The occurrence of toxigenic Fusarium spp. in semolina and wheat tissues (grain ends, crease, pericarp, aleurone layer, germ and albumen) was detected, even for a weakly contaminated wheat sample. We therefore demonstrated the penetration of toxigenic Fusarium spp. into the interior of durum wheat kernel for the Nefer variety and indicated that none of the tissue structures within the wheat kernel acted as an effective barrier to fungal invasion. Moreover, after inoculation by toxigenic Fusarium strains, semolina was shown to allow high yields of trichothecenes while bran was demonstrated to contain biochemical inhibitors able to significantly reduce trichothecenes production. These results will be useful for improving breeding strategy to control trichothecenes contamination of durum wheat kernels.

International audience; The present survey examined 30 samples of rice (n=10), maize (n=10) and peanuts (n=10) from Côte d'Ivoire for aflatoxin B1, fumonisin B1 and zearalenone using immuno-assays, and ochratoxinA using a validated HPLC-florimetric method. Similarly to some other countries it appeared that in Côte d'Ivoire several mycotoxins are present in the same commodities. These mycotoxins are from different structural families: aflatoxin B1, fumonisin B1, zearalenone, and ochratoxin A. There are normally produced by fungal species from Aspergillus, Penicilium and Fusarium genera. Some samples contain four mycotoxins (86%). Four peanuts samples do not show ochratoxin A (14%) whereas they contain threatening aflatoxin B1 concentrations, above the EU regulatory limits and exceeding tolerable daily intake. The concentrations of ochratoxin A, zearalenone and fumonisin B1 are low and may not cause problem per se however fears remain for possible excess of tolerable daily intake due to eating habits and synergism with the combination of several mycotoxins. Investigations in this direction are underway together with isolation and characterization of fungal species involved.

International audience; Cereal crops are frequently affected by toxigenic Fusarium species, among which the most common and worrying in Europe are Fusarium graminearum and Fusarium culmorum. These species are the causal agents of grain contamination with type B trichothecene (TCTB) mycotoxins. To help reduce the use of synthetic fungicides while guaranteeing low mycotoxin levels, there is an urgent need to develop new, efficient and environmentally-friendly plant protection solutions. Previously, F. graminearum proteins that could serve as putative targets to block the fungal spread and toxin production were identified and a virtual screening undertaken. Here, two selected compounds, M1 and M2, predicted, respectively, as the top compounds acting on the trichodiene synthase, a key enzyme of TCTB biosynthesis, and the 24-sterol-C-methyltransferase, a protein involved in ergosterol biosynthesis, were submitted for biological tests. Corroborating in silico predictions, M1 was shown to significantly inhibit TCTB yield by a panel of strains. Results were less obvious with M2 that induced only a slight reduction in fungal biomass. To go further, seven M1 analogs were assessed, which allowed evidencing of the physicochemical properties crucial for the anti-mycotoxin activity. Altogether, our results provide the first evidence of the promising potential of computational approaches to discover new anti-mycotoxin solutions

International audience; Genomic regions determining sexual compatibility often display recombination suppression, as occurs in sex chromosomes, plant self‐incompatibility loci and fungal mating‐type loci. Regions lacking recombination can extend beyond the genes determining sexes or mating types, by several successive steps of recombination suppression. Here we review the evidence for recombination suppression around mating‐type loci in fungi, sometimes encompassing vast regions of the mating‐type chromosomes. The suppression of recombination at mating‐type loci in fungi has long been recognized and maintains the multiallelic combinations required for correct compatibility determination. We review more recent evidence for expansions of recombination suppression beyond mating‐type genes in fungi (‘evolutionary strata’), which have been little studied and may be more pervasive than commonly thought. We discuss testable hypotheses for the ultimate (evolutionary) and proximate (mechanistic) causes for such expansions of recombination suppression, including (1) antagonistic selection, (2) association of additional functions to mating‐type, such as uniparental mitochondria inheritance, (3) accumulation in the margin of nonrecombining regions of various factors, including deleterious mutations or transposable elements resulting from relaxed selection, or neutral rearrangements resulting from genetic drift. The study of recombination suppression in fungi could thus contribute to our understanding of recombination suppression expansion across a broader range of organisms.

Chromatin is a highly dynamic structure that closely relates with gene expression in eukaryotes. ATP-dependent chromatin remodelling, histone post-translational modification and DNA methylation are the main ways that mediate such plasticity. The histone variant H2A.Z is frequently encountered in eukaryotes, and can be deposited or removed from nucleosomes by chromatin remodelling complex SWR1 or INO80, respectively, leading to altered chromatin state. H2A.Z has been found to be involved in a diverse range of biological processes, including genome stability, DNA repair and transcriptional regulation. Due to their formidable production of secondary metabolites, filamentous fungi play outstanding roles in pharmaceutical production, food safety and agriculture. During the last few years, chromatin structural changes were proven to be a key factor associated with secondary metabolism in fungi. However, studies on the function of H2A.Z are scarce. Here, we summarize current knowledge of H2A.Z functions with a focus on filamentous fungi. We propose that H2A.Z is a potential target involved in the regulation of secondary metabolite biosynthesis by fungi.

Fungal biotechnology is set to play a keystone role in the emerging bioeconomy, notably to address pollution issues arising from human activities. Because they preserve biological diversity, Biological Resource Centres are considered as critical infrastructures to support the development of biotechnological solutions. Here, we report the first large-scale phenotyping of more than 1,000 fungal strains with evaluation of their growth and degradation potential towards five industrial, human-designed and recalcitrant compounds, including two synthetic dyes, two lignocellulose-derived compounds and a synthetic plastic polymer. We draw a functional map over the phylogenetic diversity of Basidiomycota and Ascomycota, to guide the selection of fungal taxa to be tested for dedicated biotechnological applications. We evidence a functional diversity at all taxonomic ranks, including between strains of a same species. Beyond demonstrating the tremendous potential of filamentous fungi, our results pave the avenue for further functional exploration to solve the ever-growing issue of ecosystems pollution., Navarro et al. present a culture-based approach to the degradation of industrial products and by-products by assessing >1,000 fungal strains. Using growth-assay-screening and a large sample of fungal phenotypes, they explore the functional differences across strains and species to demonstrate the potential of filamentous fungi in breaking down dyes and industrial by-products with lignocellulose and plastic bases.

Enniatins are mycotoxins produced by Fusarium species contaminating cereals and various agricultural commodities. The co-occurrence of these mycotoxins in large quantities with other mycotoxins such as trichothecenes and the possible synergies in toxicity could lead to serious food safety problems. Using the agar dilution method, Ammoides pusilla was selected among eight Tunisian plants for the antifungal potential of its essential oil (EO) on Fusarium avenaceum mycelial growth and its production of enniatins. Two EO batches were produced and analyzed by GC/MS-MS. Their activities were measured using both contact assays and fumigant tests (estimated IC50 were 0.1 µL·mL−1 and 7.6 µL·L−1, respectively). The A. pusilla EOs and their volatiles inhibited the germination of spores and the mycelial growth, showing a fungistatic but not fungicidal activity. The accumulation of enniatins was also significantly reduced (estimated IC50 were 0.05 µL·mL−1 for the contact assays and 4.2 µL·L−1 for the fumigation assays). The most active batch of EO was richer in thymol, the main volatile compound found. Thymol used as fumigant showed a potent fungistatic activity but not a significant antimycotoxigenic activity. Overall, our data demonstrated the bioactivity of A. pusilla EO and its high potential to control F. avenaceum and its enniatins production in agricultural commodities.

International audience; Enniatins are emerging mycotoxins produced by Fusarium species. They are frequently found as contaminants in cereal grains in recent years. The synthesis of these secondary metabolites is often a response to biotic and abiotic stresses. Therefore, the introduction of fungicides into the environment of toxigenic fungi can lead to an increase in mycotoxin accumulation. The search for molecules with both antifungal and antimycotoxigenic activities is a challenge to preserve food safety. Essential oils (EOs) have a promising potential as natural fungicides and should be used both to control fungi and/or their mycotoxin production. In the present work, we extracted the essential oil of Tunisian Thymus capitatus and tested its effect in vitro on two enniatin-producing Fusarium strains when placed in contact with the mycelium in solid medium or used as a fumigant. In agar medium, contact tests measured with two different sets of EOs for a strain of Fusarium avenaceum provided an IC 5O of 0.05 µL EO mL-1. For another strain of Fusarium sp. it was found an IC 50 of 0.08 µL EO mL-1. Fungistatic activity induced no overproduction of enniatins and in contrast, decreases of 55% were found in some samples. Volatile components diffusing in a sealed container also had fungistatic activity that was both dose (5 to 25 µL 1.5 L-1) and time (6 to 24 days) dependent. No fungal acclimation to the EOs and no persistent effect of the EOs were observed, but the decrease in protective effect with increasing incubation time was probably due to EOs alterations.

Fungal pathogens represent a serious threat towards agriculture, health, and environment. Control of fungal diseases on crops necessitates a global understanding of fungal pathogenicity determinants and their expression during infection. Genomes of phytopathogenic fungi are often compartmentalized: the core genome contains housekeeping genes whereas the fast-evolving genome mainly contains transposable elements and species-specific genes. In this study, we analysed nucleosome landscapes of four phytopathogenic fungi with contrasted genome organizations to describe and compare nucleosome repartition patterns in relation with genome structure and gene expression level. We combined MNase-seq and RNA-seq analyses to concomitantly map nucleosome-rich and transcriptionally active regions during fungal growth in axenic culture; we developed the MNase-seq Tool Suite (MSTS) to analyse and visualise data obtained from MNase-seq experiments in combination with other genomic data and notably RNA-seq expression data. We observed different characteristics of nucleosome profiles between species, as well as between genomic regions within the same species. We further linked nucleosome repartition and gene expression. Our findings support that nucleosome positioning and occupancies are subjected to evolution, in relation with underlying genome sequence modifications. Understanding genomic organization and its role in expression regulation is the next gear to understand complex cellular mechanisms and their evolution.

In heterothallic basidiomycete fungi, sexual compatibility is restricted by mating types, typically controlled by two loci: PR, encoding pheromone precursors and pheromone receptors, and HD, encoding two types of homeodomain transcription factors. We analysed the single mating-type locus of the commercial button mushroom variety, Agaricus bisporus var. bisporus, and of the related variety burnettii. We identified the location of the mating-type locus using genetic map and genome information, corresponding to the HD locus, the PR locus having lost its mating-type role. We found the mip1 and β-fg genes flanking the HD genes as in several Agaricomycetes, two copies of the β-fg gene, an additional HD2 copy in the reference genome of A. bisporus var. bisporus and an additional HD1 copy in the reference genome of A. bisporus var. burnettii. We detected a 140 kb-long inversion between mating types in an A. bisporus var. burnettii heterokaryon, trapping the HD genes, the mip1 gene and fragments of additional genes. The two varieties had islands of transposable elements at the mating-type locus, spanning 35 kb in the A. bisporus var. burnettii reference genome. Linkage analyses showed a region with low recombination in the mating-type locus region in the A. bisporus var. burnettii variety. We found high differentiation between β-fg alleles in both varieties, indicating an ancient event of recombination suppression, followed more recently by a suppression of recombination at the mip1 gene through the inversion in A. bisporus var. burnettii and a suppression of recombination across whole chromosomes in A. bisporus var. bisporus, constituting stepwise recombination suppression as in many other mating-type chromosomes and sex chromosomes.

International audience; Agaricus sinodeliciosus is an edible wild mushroom known in northwest China. It belongs to Agaricus section Bivelares that includes several popular cultivated species, such as A. bisporus, the button mushroom. The life cycle of the latter species has been described as amphithallic because both homokaryotic (n) and heterokaryotic (n+n) spores are produced that lead to heterothallic and pseudohomothallic life cycles, respectively. The type of life cycle can impact population structures and breeding strategies. The main objective of this study was to identify the different categories of spores produced by A. sinodeliciosus. Using either a morphological approach based on the number of sterigmata per basidium or a genetic approach based on the genotypes of the progeny at two loci, the proportion of heterokaryotic spores was estimated at 6% and 15%, respectively. Two codominant markers were chosen from the mitochondrial intermediate peptidase gene (MIP) and the nuc rDNA internal transcribed spacer (ITS1-5.8S-ITS2 = ITS) region. Genotypic analysis and mating tests confirmed that in A. sinodeliciosus, MIP is centromere-linked and tightly linked to the mating type locus as in A. bisporus and A. subrufescens. We conclude that A. sinodeliciosus has a unifactorial system of sexual incompatibility and an amphithallic life cycle that is predominantly heterothallic, and that its pseudohomothallism follows a nonrandom model with nonsister postmeiotic nuclei paired in the same spore, which give rise to a potentially fertile heterokaryon. This method of using two informative markers is reliable not only in selecting the homokaryotic offspring but also in classifying the homokaryons in two breeding stocks according to their mating type alleles.

In a recent revision of the genus Agaricus, A. section Xanthodermatei was split into two sections A. sect. Hondenses and A. sect. Xanthodermatei. Our objectives were to investigate the species diversity of both sections in Pakistan and to give an overview of the major clades. Phylogenetic analyses based on the combined nucLSU, ITS and TEF1 dataset from 35 specimens of both sections revealed three major clades. Analyses based on ITS dataset and 106 specimens, including 33 from Pakistan, reveal eight new species and one new record species. These nine species are described in detail. It is noteworthy that intraspecific variability as well as interspecific variability between closely related species were very low in ITS sequences in many cases. In the case of the two new species A.xanthochromaticus and A.griseovariegatus, TEF1 sequence data were much more efficient than ITS to distinguish these species from each other. The other new species are A.atroumbonatus, A.fumidicolor, A.macropeplus, A.parviniveus, A.swaticus and A.bambusetorum. The latter is the only new species of A. sect. Hondenses in which it is morphologically atypical and also the unique (sub)tropical species. Agaricusgregariomyces is recorded for the first time in Pakistan. In addition, brief descriptions are provided not only for A.bisporiticus, A.endoxanthus and A.punjabensis, which are reported again in Pakistan, but also for A.californicus, which is reported for the first time in Spain and outside North America. In total 12 species of both sections were reported in Pakistan and half of them were from subtropical climatic areas, underlining the contribution of the climatic diversity to the high species richness in this country.

International audience; Fusarium graminearum is a major fungal pathogen affecting crops of worldwide importance. F. graminearum produces type B trichothecene mycotoxins (TCTB), which are not fully eliminated during food and feed processing. Therefore, the best way to minimize TCTB contamination is to develop prevention strategies. Herein we show that treatment with the reduced form of the γ-core of the tick defensin DefMT3, referred to as TickCore3 (TC3), decreases F. graminearum growth and abrogates TCTB production. The oxidized form of TC3 loses antifungal activity, but retains anti-mycotoxin activity. Molecular dynamics show that TC3 is recruited by specific membrane phospholipids in F. graminearum and that membrane binding of the oxidized form of TC3 is unstable. Capping each of the three cysteine residues of TC3 with methyl groups reduces its inhibitory efficacy. Substitutions of the positively-charged residues lysine (Lys) 6 or arginine 7 by threonine had the highest and the lesser impact, respectively, on the anti-mycotoxin activity of TC3. We conclude that the binding of linear TC3 to F. graminearum membrane phospholipids is required for the antifungal activity of the reduced peptide. Besides, Lys6 appears essential for the anti-mycotoxin activity of the reduced peptide. Our results provide foundation for developing novel and environment-friendly strategies for controlling F. graminearum.

International audience; The genus Agaricus currently includes about 500 species in seven subgenera. Continuing an inventory of species in the Caribbean, this manuscript’s objective was to focus on those in Agaricus subg. Spissicaules. This subgenus has been recently proposed and divided into four sections by applying a new taxonomic system in which supraspecific taxa are ranked according to their divergence times. We similarly applied this system using Bayesian evolutionary analyses (BEAST). Phylogenetic analyses were performed using maximum likelihood and Bayesian inference methods, based either on multi-gene sequence data (ITS, ncLSU, and TEF1) or only on ITS, and including new collections from the Caribbean and one from Argentina. The analyses revealed seven major clades, all with divergence time estimated around 24 Ma. Among those, four were known sections of the subgenus, one remains a putative section, and we introduce the remaining two as new sections, A. sect. Fulventes and A. sect. Globoterminales. New samples were distributed among ten species-level clades. For eight of them, new species were proposed. They are distributed as follows: A. nanofulvens and A. basicingulatus are in the two new sections cited above, respectively; A. ciferrianus and A. omphalodiscus are in A. sect. Subrutilescentes; A. baronii, A. furfuripes, A. microincrustatus, and A. parvisporus are in A. sect. Rarolentes. The eight new species and the four sections to which they belong are described in detail. High levels of both species richness and phylogenetic diversity in the Caribbean region contributed to the refinement of the classification of A. subg. Spissicaules.

International audience; Agaricus subgenus Spissicaules is widely distributed in the world. In this study, 114 specimens were included in multigene phylogenetic analyses that allowed a better circumscription of the four sections in A. subg. Spissicaules. Three new species from China, A. brunneovariabilis, A. beijingensis, and A. planiceps, are described and placed in different sections. The invalidly described A. catenariocystidiosus is validated here as an additional new species. Comparisons between the sequences of the three closely related species A. thiersii, A. parasubrutilescens, and A. linzhiensis support the distinction between these three species despite the paraphyly of A. linzhiensis, which results from its high intraspecific variability with numerous heteromorphisms.

We found that nearly one sample of Peruvian corn out of six was contaminated with very high levels of Fumonisins, levels mostly ascribed to the presence of F. verticillioides. Extensive profiling of Peruvian corn kernels for fungal contaminants could provide elements to refine the potential risk associated with Fusarium toxins and help define adapted food safety standards. Fusarium graminearum is a fungal pathogen that can colonize small-grain cereals and maize and secrete type B trichothecene (TCTB) mycotoxins. The development of environmental-friendly strategies guaranteeing the safety of food and feed is a key challenge facing agriculture today. One of these strategies lies on the promising capacity of products issued from natural sources to counteract crop pests. In this work, the in vitro efficiency of sixteen extracts obtained from eight natural sources using subcritical water extraction at two temperatures was assessed against fungal growth and TCTB production by F. graminearum. Maritime pine sawdust extract was shown to be extremely efficient, leading to a significant inhibition of up to 89% of the fungal growth and up to 65% reduction of the mycotoxin production by F. graminearum. Liquid chromatography/mass spectrometry analysis of this active extract revealed the presence of three families of phenolics with a predominance of methylated compounds and suggested that the abundance of methylated structures, and therefore of hydrophobic compounds, could be a primary factor underpinning the activity of the maritime pine sawdust extract. Altogether, our data support that wood/forest by-products could be promising sources of bioactive compounds for controlling F. graminearum and its production of mycotoxins.

Mycotoxins in small grains are a significant and long-standing problem. These contaminants may be produced by members of several fungal genera, including Alternaria, Aspergillus, Fusarium, Claviceps, and Penicillium. Interventions that limit contamination can be made both pre-harvest and post-harvest. Many problems and strategies to control them and the toxins they produce are similar regardless of the location at which they are employed, while others are more common in some areas than in others. Increased knowledge of host-plant resistance, better agronomic methods, improved fungicide management, and better storage strategies all have application on a global basis. We summarize the major pre- and post-harvest control strategies currently in use. In the area of pre-harvest, these include resistant host lines, fungicides and their application guided by epidemiological models, and multiple cultural practices. In the area of post-harvest, drying, storage, cleaning and sorting, and some end-product processes were the most important at the global level. We also employed the Nominal Group discussion technique to identify and prioritize potential steps forward and to reduce problems associated with human and animal consumption of these grains. Identifying existing and potentially novel mechanisms to effectively manage mycotoxin problems in these grains is essential to ensure the safety of humans and domesticated animals that consume these grains.

International audience; Fusarium graminearum is one of the most frequent causal agents of the Fusarium Head Blight, a cereal disease spread throughout the world, reducing grain production and quality. F. graminearum isolates are genetically and phenotypically highly diverse. Notably, remarkable variations of aggressiveness between isolates have been observed, which could reflect an adaptive potential of this pathogen. In this study, we aimed to characterize the genetic basis of aggressiveness variation observed in an F1 population (n = 94), for which genome sequences of both parental strains are available. Aggressiveness was assessed by a panel of in planta and in vitro proxies during two phenotyping trials including, among others, disease severity and mycotoxin accumulation in wheat spike. One major and single QTL was mapped for all the traits measured, on chromosome I, that explained up to 90% of the variance for disease severity. The confidence interval at the QTL spanned 1.2 Mb and contained 428 genes on the reference genome. Of these, four candidates were selected based on the postulate that a non-synonymous mutation affecting protein function may be responsible for phenotypic differences. Finally, a new mutation was identified and functionally validated in the gene FgVe1, coding for a velvet protein known to be involved in pathogenicity and secondary metabolism production in several fungi.

Background Early consumption of obesogenic diets, rich in saturated fat and added sugar, is associated with a plethora of biological dysfunctions, at both peripheral and brain levels. Obesity is also linked to decreased vitamin A bioavailability, an essential molecule for brain plasticity and memory function. Methods Here we investigated in mice whether dietary vitamin A supplementation (VAS) could prevent some of the metabolic, microbiota, neuronal and cognitive alterations induced by obesogenic, high-fat and high-sugar diet (HFSD) exposure from weaning to adulthood, i.e. covering periadolescent period. Results As expected, VAS was effective in enhancing peripheral vitamin A levels as well as hippocampal retinoic acid levels, the active metabolite of vitamin A, regardless of the diet. VAS attenuated HFSD-induced excessive weight gain, without affecting metabolic changes, and prevented alterations of gut microbiota α-diversity. In HFSD-fed mice, VAS prevented recognition memory deficits but had no effect on aversive memory enhancement. Interestingly, VAS alleviated both HFSD-induced higher neuronal activation and lower glucocorticoid receptor phosphorylation in the hippocampus after training. Conclusion Dietary VAS was protective against the deleterious effects of early obesogenic diet consumption on hippocampal function, possibly through modulation of the gut-brain axis.

Fusarium avenaceum is a filamentous fungus infecting many plants and is one of the causal agents of the Fusarium Head Blight disease in wheat and barley. During plant infection, the fungus can produce secondary metabolites that are toxic for human and livestock (mycotoxins), among which the enniatins. Enniatins belong to the group of “emerging mycotoxins”: first identified as early as 1950, the increasing occurrence of enniatins has only been documented over the last twenty years. Currently, enniatins are neither routinely analysed in grain harvests nor legislatively regulated, even though they are widespread and can be encountered in high concentrations. Diversity of enniatin-producing strains as well as the mechanisms by which their biosynthesis can be modulated during plant infection and/or in response to environmental conditions are poorly known. These gaps of knowledge hinder the development of tools for controlling and mitigating their presence in crops. In the present study, a comprehensive characterisation of the phenotypic diversity within the F. avenaceum species has been carried out. One of the studied phenotypic traits was the effect of pH variations on the fungal growth and toxin production, which has led to evidence various response profiles between strains. To go further, the role of FavPac1, the homologue of pacC/RIM101 encoding a transcriptional factor involved in the response to ambient pH in fungi, has been studied. Construction and phenotyping of deleted mutants, Fav∆Pac1, for four different F. avenaceum isolates have allowed suggesting an involvement of FavPac1 in the modulation of enniatin production and of the expression of biosynthetic genes induced by pH variations. Lastly, with the aim to identify alternatives to synthetic fungicides, the capacity of plant phenolic compounds to inhibit the yield of enniatins has been investigated. The antifungal and anti-mycotoxin properties of hydroxycinnamic acids have been shown; ferulic acid has been highlighted as the most potent compound.; Fusarium avenaceum est un champignon filamenteux pathogène de nombreuses plantes et est un des acteurs majeurs de la fusariose de l’épi chez le blé et l’orge. Durant l’infection, des métabolites secondaires toxiques pour l’homme et les animaux (mycotoxines) telles que les enniatines sont produites par ce champignon. Les enniatines appartiennent au groupe de mycotoxines dîtes « émergentes » : même si connues depuis 1950, leur présence en quantités importantes et croissantes dans les aliments n’a été mise en évidence que depuis une vingtaine d’année. Actuellement la présence d’enniatines n’est pas surveillée de manière systématique dans les récoltes céréalières, les enniatines ne sont pas soumises à réglementation alors qu’elles sont omniprésentes dans les récoltes et parfois à des concentrations élevées. La diversité des souches productrices ainsi que les mécanismes par lesquels la biosynthèse d’enniatines peut être modulée au cours de l’infection de la plante et/ou en réponse aux conditions environnementales sont mal connus. Ce manque de connaissances constitue un frein à la mise au point d’outils de contrôle et de gestion de l’état sanitaire des récoltes. C’est pourquoi cette étude s’est attachée à apporter de premiers éléments de réponse sur la diversité phénotypique au sein de l’espèce F. avenaceum. Un des facteurs étudiés a concerné la réponse aux variations de pH en termes de croissance et de toxinogénèse et a mis en évidence des comportements très différents entre les isolats. Pour aller plus loin, le rôle de FavPac1, l’homologue de pacC/RIM101 codant un facteur de régulation de l’homéostasie du pH chez les champignons, a été étudié. Des mutants délétés pour ce gène, mutants Fav∆Pac1, ont été construits pour quatre souches de F. avenaceum et ont permis de suggérer l’implication de ce facteur de transcription non seulement dans la régulation de l’expression des gènes de biosynthèse, mais aussi dans la modulation de la production d’enniatines en réponse au pH. Enfin, dans le cadre de la recherche d’inhibiteurs naturels pouvant se substituer aux fongicides de synthèse, nos travaux ont démontré la capacité de certains composés phénoliques à inhiber la production d’enniatines associée à un contrôle transcriptionnel des gènes de biosynthèse et ont mis en évidence l’efficacité élevée de l’acide férulique.

Background Increased contamination of European and Asian wheat and barley crops with “emerging” mycotoxins such as enniatins or beauvericin, produced by Fusarium avenaceum and Fusarium tricinctum, suggest that these phylogenetically close species could be involved in future food-safety crises. Results The mitochondrial genomes of F. tricinctum strain INRA104 and F. avenaceum strain FaLH27 have been annotated. A comparative analysis was carried out then extended to a set of 25 wild strains. Results show that they constitute two distinct species, easily distinguished by their mitochondrial sequences. The mitochondrial genetic variability is mainly located within the intergenic regions. Marks of variations show they have evolved (i) by Single Nucleotide Polymorphisms (SNPs), (ii) by length variations mediated by insertion/deletion sequences (Indels), and (iii) by length mutations generated by DNA sliding events occurring in mononucleotide (A)n or (T)n microsatellite type sequences arranged in a peculiar palindromic organization. The optionality of these palindromes between both species argues for their mobility. The presence of Indels and SNPs in palindrome neighbouring regions suggests their involvement in these observed variations. Moreover, the intraspecific and interspecific variations in the presence/absence of group I introns suggest a high mobility, resulting from several events of gain and loss during short evolution periods. Phylogenetic analyses of intron orthologous sequences suggest that most introns could have originated from lateral transfers from phylogenetically close or distant species belonging to various Ascomycota genera and even to the Basidiomycota fungal division. Conclusions Mitochondrial genome evolution between F. tricinctum and F. avenaceum is mostly driven by two types of mobile genetic elements, implicated in genome polymorphism. The first one is represented by group I introns. Indeed, both genomes harbour optional (inter- or intra-specifically) group I introns, all carrying putatively functional hegs, arguing for a high mobility of these introns during short evolution periods. The gain events were shown to involve, for most of them, lateral transfers between phylogenetically distant species. This study has also revealed a new type of mobile genetic element constituted by a palindromic arrangement of (A) n and (T) n microsatellite sequences whose presence was related to occurrence of SNPs and Indels in the neighbouring regions.

Ochratoxin A (OTA) is one of the worldwide most important mycotoxins in terms of health and agroeconomic consequences. With the aim to promote the use of phytochemicals as alternatives to synthetic fungicides, the effect of hydroxycinnamic acids on the fungal growth and OTA yield by two major OTA-producing species was investigated. After a first step dedicated to the definition of most suitable culture conditions, the impact of 0.5 mM ferulic (FER), p-coumaric (COUM), caffeic and chlorogenic acids was evaluated on Aspergillus westerdijkiae and Penicillium verrucosum. Whereas no fungal growth reduction was observed regardless of the phenolic acid and fungal isolate, our results demonstrated the capacity of FER and COUM to inhibit OTA production. The most efficient compound was FER that led to a 70% reduction of OTA yielded by P. verrucosum and, although not statistically significant, a 35% inhibition of OTA produced by A. westerdijkiae. To further investigate the bioactivity of FER and COUM, their metabolic fate was characterized in fungal broths. The capacity of P. verrucosum to metabolize FER and COUM through a C2-clivage type degradation was demonstrated. Overall, our data support the potential use of FER to prevent OTA contamination and reduce the use of synthetic pesticides.

International audience; This study presents fungi infrequently viewed as fungal factories for secondary metabolite production resources such as mycotoxins in Ascomycota. Additionally, we demonstrated that biochemical warfare of Fusarium verticillioides factory against animal cells is not only due to mycotoxins such as fumonisins, but acute cytotoxic firing is based on different excreted secondary metabolite series, potentially leading to animal and human diseases. In this study, fumonisins, which can be followed by in situ localization, quantification, or expression of the key gene implicated in their synthesis, are used to understand secondary metabolite production by this fungus. It is known that F. verticillioides produces mycotoxins such as fumonisins on cereals, but until now, there is no evidence demonstrating a method to totally block fumonisin production on feed and food. In this paper, we explained, what was never clearly established before, that fumonisin production depends on two bottlenecks. The fumonisin synthesis and secretion in fungal articles of the mycelium are medium-independent and follow the fungal cell cycle developmental program (ontogenesis). Conversely, the fumonisin excretion into the medium depends on its composition, which also impacts fumonisin biosynthesis level. Using a high-pressure freezing method, we showed that, in non-permissive fumonisin excretion (NPFE) medium, FB1 is sequestered inside extra-vesicles and in the first third of the cell wall next to the plasmalemma, leading to the hypothesis that the fungus develops mechanisms to protect its cytosolic homeostasis against this cytotoxic. In permissive fumonisin excretion (PFE) medium, leading to very high quantities of excreted fumonisins, FB1 localized inside extra-vesicles, crosses the entire cell wall thickness, and then releases into the medium. Our results demonstrated a delayed and lower expression of Fvpks gene in mycelium developed on NPFE medium as compared to PFE medium. Conversely, higher amounts of fumonisins were accumulated in NPFE-grown mycelium than in PFE-grown mycelium. Thus, our results demonstrated for the first time that we have to take into account that the synthesis and secretion inside the article of secondary metabolites depend on the occurrence of cryptic biochemical specialized articles, differentiated in the mycelium. However, those are not morphologically different from other colonial hyphae.

Fusarium head blight is a destructive disease of grains resulting in reduced yields and contamination of grains with mycotoxins worldwide; Fusarium graminearum is its major causal agent. Chromatin structure changes play key roles in regulating mycotoxin biosynthesis in filamentous fungi. Using a split-marker approach in three F. graminearum strains INRA156, INRA349 and INRA812 (PH-1), we knocked out the gene encoding H2A.Z, a ubiquitous histone variant reported to be involved in a diverse range of biological processes in yeast, plants and animals, but rarely studied in filamentous fungi. All ΔH2A.Z mutants exhibit defects in development including radial growth, sporulation, germination and sexual reproduction, but with varying degrees of severity between them. Heterogeneity of osmotic and oxidative stress response as well as mycotoxin production was observed in ΔH2A.Z strains. Adding-back wild-type H2A.Z in INRA349ΔH2A.Z could not rescue the phenotypes. Whole genome sequencing revealed that, although H2A.Z has been removed from the genome and the deletion cassette is inserted at H2A.Z locus only, mutations occur at other loci in each mutant regardless of the genetic background. Genes affected by these mutations encode proteins involved in chromatin remodeling, such as the helicase Swr1p or an essential subunit of the histone deacetylase Rpd3S, and one protein of unknown function. These observations suggest that H2A.Z and the genes affected by such mutations are part or the same genetic interaction network. Our results underline the genetic plasticity of F. graminearum facing detrimental gene perturbation. These findings suggest that intergenic suppressions rescue deleterious phenotypes in ΔH2A.Z strains, and that H2A.Z may be essential in F. graminearum. This assumption is further supported by the fact that H2A.Z deletion failed in another Fusarium spp., i.e., the rice pathogen Fusarium fujikuroi., Author summary The function of histone variant H2A.Z has been widely studied in Saccharomyces cerevisiae and higher eukaryotes. However, there is a significant lack of knowledge of the role of H2A.Z and its regulation in filamentous fungi. Here we demonstrate for the first time that deleterious consequences of complete removal of H2A.Z from the genome of F. graminearum are mitigated by genetic suppression. The suppressors detected open new possibilities to investigate in more depth the network linking H2A.Z and other genes in F. graminearum and used to decipher unresolved questions concerning H2A.Z.

Vitamin A (retinol) and related retinoids are micronutrients provided by food. Retinol derivatives are growth factors important for development, cell differentiation and tissue homeostasis, especially in the brain. Objective: The hippocampus is a pivotal brain structure for learning and memory and hippocampal-dependent memory is highly sensitive to retinoids action. However, the underlying mechanisms are still unclear. In this study, we characterized the impact of vitamin A deficiency on memory and neuronal plasticity, focusing on the CA1 region of the hippocampus in rats. Methods: Weaned male Wistar rats were fed a control (5 UI/g) or deficient vitamin A diet (0 UI/g) for 10 weeks. The effect of vitamin A supplementation (20 UI/g) for 3 weeks was also tested. Memory performances were assessed in the Y-maze (n = 24-30/group), retinoic acid levels were measured (LC-MS/MS) in the serum and in the hippocampus (n = 5/group), CA1 neuronal architecture was analyzed with Golgi staining (n = 17-20 neurons/group) and electrophysiological patch-clamp recordings were performed on hippocampal brain slices (n = 6-11/group). Results: Vitamin A deficiency from weaning significantly lowered hippocampal levels of retinoic acid, reduced dendritic length and branching of CA1 pyramidal neurons and decreased spontaneous glutamatergic synaptic events and synaptic plasticity. When replenishment with moderate dose of dietary vitamin A for 3 weeks was done, most of the synaptic and morphological alterations were absent. Conclusion: This study provides new mechanistic insight to understand the critical role of retinoic acid in hippocampal function.

The objective of this study was to develop a structured and transparent framework to rank emerging dietary practices. The first challenge was to rank simultaneously biological and chemical hazards using the same criteria whatever the nature of the hazard. For a list of dietary practices selected based on the results of a survey, hazard identification and health effect characterization was carried out. Taking only the top five practices led to the identification of 41 triplets “emerging dietary practice – hazard – health effect”, which highlights the complexity of scoring risk in food safety. A wide variety of hazards, including microbes, parasites, mycotoxins, allergens and other chemical compounds were considered together with a range of health effects such as foodborne pathogen disease, anaphylaxis, cancer, immunosuppression, endocrine disturbance, etc. The second challenge was to develop a framework easy to populate and run. The risk-ranking framework included eight criteria: five to describe the severity, three to describe the likelihood. All of them were informed by literature data and food safety agencies' reports, plus experts’ opinion. The PROMETHEE outranking MCDA technique, available in a R package, was implemented. This risk-ranking framework applied to the results of our small-scale survey revealed that consuming nuts on a regular basis could be the emerging dietary habit presenting the highest-risk score, due to the aflatoxin B1 hazard and its associated health effect (liver cancer). This risk-ranking framework requires however to be applied furthermore in other contexts to evaluate its robustness and identify opportunities for improvement. Once consolidated, this framework will be highly relevant for food safety authorities and policy makers to move forward transparent and evidence-informed decisions.

The maize pathogen Fusarium verticillioides and their mycotoxins cause damage to plants, animals, and human health. This work aimed to evaluate the effect of crude extracts (CEs) from Agaricus subrufescens, Lentinula edodes, and Pleurotus ostreatus fruiting bodies on in vitro production of biomass and mycotoxins by two strains of F. verticillioides. Stipes and pilei were separated before extraction for A. subrufescens and L. edodes. Comparative metabolomics and dereplication of phenolic compounds were used to analyze all CEs. Mushroom CEs did not significantly inhibit the production of mycelial biomass at concentrations of 2 mg mL⁻1. CEs from A. subrufescens (stipes and pilei) and L. edodes pilei inhibited the production of fumonisins B1 + B2 + B3 by 54% to 80%, whereas CE from P. ostreatus had no effect. In contrast, CE from L. edodes stipes dramatically increased the concentration of fumonisins in culture media. Fusaric acid concentration was decreased in cultures by all CEs except L. edodes stipes. Differences in phenolic composition of the extracts may explain the different effects of the CE treatments on the production of mycotoxins. The opposing activities of stipes and pilei from L. edodes offer an opportunity to search for active compounds to control the mycotoxin production by F. verticillioides.

Cereal grains and their processed food products are frequently contaminated with mycotoxins produced by the Fusarium genus. Enniatins (ENNs), which belong to the so-called “emerging mycotoxins” family, are among the most frequently found in small grain cereals. Health hazards induced by a chronic exposure to ENNs or an association of ENNs with other major mycotoxins is a risk that cannot be excluded given the current toxicological data. Thus, efforts must be pursued to define efficient control strategies to mitigate their presence in cereal grains. A key condition for achieving this aim is to gain deep and comprehensive knowledge of the factors promoting the appearance of ENNs in crop harvests. After an update of ENN occurrence data, this review surveys the scientific literature on the Fusarium species responsible for ENN contamination and covers the recent advances concerning the abiotic determinants and the genetic regulation of ENN biosynthesis.

International audience; Fusarium head blight (FHB) is a cereal disease caused by a complex of plant pathogenic fungal species of the genera Fusarium andMicrodochium, with different epidemiological characteristics and producing or not different toxins. The FHB causes significant yieldlosses, health issues and technological quality concerns in all temperate areas worldwide. The composition of the species complexis different according to the cereal, depending on the climate of the year and the agronomic practices. These species may havedifferent susceptibilities to the fungicides used and different behaviors towards genetic resistance. Thus, investigating thesecharacteristics is required to implement integrated pest management strategies adapted to this disease. For more than 10 years,several research programs have led to significant progress in this area. Tools have been developed to more accurately characterizefungal populations and their interactions. Models have been developed to predict the risk and to adapt control measures. Themolecular mechanisms involved in the interaction between plant genes and the pathogenicity of fungal species are beginning to bedeciphered. Phenotyping methods are being developed to help in the characterization of varietal susceptibility and direct controlmethods using biocontrol products are under investigation. The goal of this paper is to review the main results obtained in thecontext of integrated management of fusarium head blight.; La fusariose de l’épi (FHB) est une maladie des céréales causée par un complexe d’espèces de champignons phytopathogènes desgenres Fusarium et Microdochium, ayant des caractéristiques épidémiologiques différentes et productrices ou non de différentestoxines. Le FHB entraîne d’importantes pertes de rendement et des problèmes de qualité sanitaire et technologique dans toutes lesrégions tempérées du globe. La composition du complexe d’espèces est différente selon les céréales, très dépendante du climat del’année et de l’itinéraire technique. Ces espèces peuvent avoir des sensibilités différentes aux fongicides utilisés et descomportements différents vis à vis des résistances présentes dans les variétés. Ainsi, il est essentiel de connaître ces caractéristiquespour mettre en œuvre les méthodes de lutte intégrée adaptées à cette maladie. Depuis plus de 10 ans, plusieurs programmes derecherche ont permis des avancées significatives sur cette thématique. Des outils ont été développés et ont permis de caractériserplus précisément les populations fongiques selon les céréales et leurs interactions. Des modèles ont été développés pour prédire lerisque et adapter les moyens de lutte. Les mécanismes moléculaires impliqués dans l’interaction entre les gènes de la plante et lapathogénicité des espèces fongiques commencent à être décryptés. Des méthodes de phénotypage se développent pour aider à lacaractérisation de la sensibilité variétale et des méthodes de lutte directe utilisant des produits de biocontrôle sont endéveloppement. L’objectif de cet article est de synthétiser les principaux résultats acquis dans l’objectif d’une protection intégréevis‐à‐vis de la fusariose des épis des céréales.

UMR BFP - Equipe Métabolisme; International audience; We analyzed a comprehensive set of contaminants in MON810 and NK603 genetically modified (GM) maize, and their non-GM counterparts, used in a rat feeding study (the GMO90 + project). Both the maize grains and the manufactured pellets were characterized. Only minor differences in contaminant levels between GM and corresponding non-GM harvests were evidenced. Fumonisin and deoxynivalenol mycotoxins were the pollutants present in the highest amounts, with concentrations that were however largely below acceptance reference values. Our data reporting slightly higher levels of fumonisin in MON810 compared to its non-GM counterpart corroborate the lower susceptibility of insect resistant Bt maize to fumonisin-producing fungi. Traces of glyphosate (0.016 mg/kg) were evidenced in grains from NK603 treated crops. Regarding the pellets, analysis of more than 650 potentially toxic substances revealed low amounts of various mycotoxins, pesticides and heavy metals. Concentrations of contaminants quantified in the pellets were however far below the maximum level of residues values set by regulatory agencies, and no substantial differences in contaminants between GM and non-GM pellets were observed. Moreover, when comparing the contamination status of grains and pellets, we demonstrate yet again that characterizing the grains is actually not sufficient to foresee the quality of the produced pellets.

An account is provided of the world’s ten most feared fungi. Within areas of interest, we have organized the entries in the order of concern. We put four human pathogens first as this is of concern to most people. This is followed by fungi producing mycotoxins that are highly harmful for humans; Aspergillus flavus, the main producer of aflatoxins, was used as an example. Problems due to indoor air fungi may also directly affect our health and we use Stachybotrys chartarum as an example. Not everyone collects and eats edible mushrooms. However, fatalities caused by mushroom intoxications often make news headlines and therefore we include one of the most poisonous of all mushrooms, Amanita phalloides, as an example. We then move on to the fungi that damage our dwellings causing serious anxiety by rotting our timber structures and flooring. Serpula lacrymans, which causes dry rot is an excellent example. The next example serves to represent all plant and forest pathogens. Here we chose Austropuccinia psidii as it is causing devastating effects in Australia and will probably do likewise in New Zealand. Finally, we chose an important amphibian pathogen which is causing serious declines in the numbers of frogs and other amphibians worldwide. Although we target the top ten most feared fungi, numerous others are causing serious concern to human health, plant production, forestry, other animals and our factories and dwellings. By highlighting ten feared fungi as an example, we aim to promote public awareness of the cost and importance of fungi.

International audience; The genus Agaricus was recently rearranged to accommodate numerous tropical taxa. Accordingly, the genus was split into six subgenera and 22 sections of which 12 are included in A. subg. Pseudochitonia. Preliminary data indicated that three putative new species belong to this subgenus. Our objectives were to describe these species, to determine to which sections they belong, and to experience the interest of some traditional traits in this new context. We morphologically described Agaricus coniferarum from France and Portugal, Agaricus iranicus from Iran, and Agaricus lusitanicus from Portugal. Multi-gene phylogenetic analyses based on ITS, LSU, and tef1 sequence data of representatives of the 12 sections clearly indicated that A. coniferarum and A. lusitanicus are placed in Agaricus sect. Bohusia, while A. iranicus is in A. sect. Sanguinolenti. Incidentally, we replaced the illegitimate name Agaricus magnivelaris by Agaricus fiardianus. In a phylogenetic tree, based on all available ITS sequence data and focussing on six related sections, we examined the phylogenetic distribution of various characters. The intensity of red discolouration when the sporocarp is rubbed or cut appeared as a phylogenetically weak informative trait. We propose a determination key leading to a group of three hardly distinguishable sections (Bohusia, Nigrobrunnescentes, and Sanguinolenti).

Nomenclatural type definitions are one of the most important concepts in biological nomenclature. Being physical objects that can be re-studied by other researchers, types permanently link taxonomy (an artificial agreement to classify biological diversity) with nomenclature (an artificial agreement to name biological diversity). Two proposals to amend the International Code of Nomenclature for algae, fungi, and plants (ICN), allowing DNA sequences alone (of any region and extent) to serve as types of taxon names for voucherless fungi (mainly putative taxa from environmental DNA sequences), have been submitted to be voted on at the 11th International Mycological Congress (Puerto Rico, July 2018). We consider various genetic processes affecting the distribution of alleles among taxa and find that alleles may not consistently and uniquely represent the species within which they are contained. Should the proposals be accepted, the meaning of nomenclatural types would change in a fundamental way from physical objects as sources of data to the data themselves. Such changes are conducive to irreproducible science, the potential typification on artefactual data, and massive creation of names with low information content, ultimately causing nomenclatural instability and unnecessary work for future researchers that would stall future explorations of fungal diversity. We conclude that the acceptance of DNA sequences alone as types of names of taxa, under the terms used in the current proposals, is unnecessary and would not solve the problem of naming putative taxa known only from DNA sequences in a scientifically defensible way. As an alternative, we highlight the use of formulas for naming putative taxa (candidate taxa) that do not require any modification of the ICN. Agaricus subrufescens Peck is a cosmopolitan species of nutritional and medicinal interest. Three types of internal transcribed spacer (ITS) rDNA sequences were recently characterized for this species. Sequences of types A, B, or both types A and B are found in American and European specimens, while sequences of type C are found in Asian (China and Thailand) and Oceanian (Hawaii) specimens. We report on a collection of this species from Iran, for the first time. Both the morphological features and ITS sequence confirm its identification as A. subrufescens. Its ITS sequence was of type C and even identical to that of specimens previously reported from Hawaii, China, and Thailand. This sequence differed from those of Europe or the Americas, where most current cultivars originate. The discovery of a specimen likely native to Iran extends the known distribution range of the ITS sequence of type C from East Asia to Iran. More collections from West Asia (and elsewhere, for example Africa) would help to better know the distribution area of the different types of ITS sequences and to increase the available germplasm resources of A. subrufescens.

Fungal spoilage of stored grains may occur when activity of water ( aw ) in cereal grain exceeds a critical limit enabling mould growth. Because it is not feasible to maintain all parts of large grain bulks below this critical moisture limit during prolonged storage time, an infection by seed-borne fungi is not rare in cereal grain stored under humid temperate or hot climates, inducing irreversible qualitative losses. Additionally, some fungal species produce harmful mycotoxins. The most harmful toxigenic species belong to the group of xerophilic species (genera Aspergillus and Penicillium ). Because mycotoxin contamination of cereal grain is a worldwide issue for public health and a permanent concern for cereal-food industries facing the challenge of a permanent monitoring mycotoxin content in their primary matters, tolerable levels of mycotoxins are severely regulated worldwide. Mycotoxin-producing species growth is closely dependent of grain moisture levels enabling biological activity in grain ecosystem. Consequently, mould growth in stored grain bulks can be anticipated through early detection of grain and mould respiration. The prevention of mycotoxigenic fungi spoilage of stored grain can be managed by a preventive strategy. The main objective of the review was to describe the different methods, material and practices combined in such an integrated preventive approach. Some solutions potentially acceptable for the decontamination of moderately contaminated grain are also discussed. Integrated management of mould spoilage risks in stored grain is based on five pillars: i/Prevention of mould development by keeping grain moisture below the critical limit of fungal growth; ii/Accurate monitoring of grain aw and temperature changes during the storage period, associated to the monitoring of early indicators of respiration activity of storage fungi; iii/Reduction of grain bulk moistening trends by physical intervention means; iv/Use of physical treatments (ozone, grain peeling or abrasion) to limit mycotoxin contamination transfer to processed cereal products; v/Possible use of bio-competitive strains of fungi or bacteria to prevent the development of mycotoxigenic fungi in grain bulks. The future research needs on this topic are also evocated.

Contamination of food with mycotoxins is a significant risk to human and animal health. Reports from the European Food Safety Authority indicate that nearly half of the food derived from cereals and unprocessed grains collected between 2007 and 2012 in 21 European countries were contaminated with Type B trichothecenes, or TCTB, and especially deoxynivalenol, or DON. These mycotoxins are produced by fungal phytopathogens on growing kernels, before harvest. On account of the global climate change, this situation may become increasingly serious. Therefore, preventing the production of mycotoxins is a task which brooks no delay. In Europe, DON is predominantly produced by Fusarium graminearum. Even though the sequential steps of the TCTB biosynthetic pathway are fairly well described, the molecular events involved in regulating this pathway are complex and remain, so far, widely misunderstood.Recent studies highlighted post-translational modifications of canonical histones as well as their variants as critical players in the regulation of mycotoxin and other secondary metabolite biosyntheses in filamentous fungi, by altering chromatin structure. In F. graminearum, it was shown that the histone deacetylase HDF1 could be involved in the activation of DON production. In contrast, H3K27me3 histone mark represses 14 % of its genome, including genes involved in secondary metabolic pathways. Histone variant H2A.Z is ubiquitous in eukaryotes and is involved in a diverse range of biological processes, including genome stability, DNA repair, transcriptional regulation and telomere silencing. However, the underlying mechanisms of these functions remain unclear. In some species, the function of H2A.Z appears to be essential. Up to now, the only one study targeted directly on the function of H2A.Z in filamentous fungi was carried out on Neurospora crassa, and identifying a role in oxidative stress response.Here, we hypothesized that H2A.Z may be involved in important biological processes of F. graminearum including those involved in the production of secondary metabolism. Therefore, this project aims to characterize the roles played by the histone variant H2A.Z in controlling development, metabolism and virulence in F. graminearum.Using a reverse genetics approach, we created six H2A.Z deleted mutants in three different F. graminearum strains. All mutants exhibit deficiency in sporulation, germination, radial growth and DON production; however, intensities in the observed effects depend on the considered genetic background. Additionally, adding back wild-type H2A.Z could not rescue mutant phenotypes. Whole-genome sequencing showed that, although H2A.Z has been totally removed from the genome, compensatory mutations occur at other sites in each mutant regardless of the genetic background, in genes involved in chromatin remodeling. Strikingly, one extra mutation was detected in the H2A.Z add-back mutants. H2A.Z overexpressed mutants have also been constructed, but no significant difference in phenotype can be observed with wild type. Considering our results as a whole, we draw the conclusion that H2A.Z is essential in F. graminearum. It is the occurrence of compensatory mutations that rescued part of the lethality caused by H2A.Z deletion. We hypothesize that profound reorganizations of gene networks allow such plasticity, with certain consequences in terms of evolution and adaptation.; La contamination des aliments par les mycotoxines représente un risque potentiel pour la santé humaine et animale. Des rapports de l'Autorité européenne de sécurité des aliments, l’EFSA, indiquent que près de la moitié des aliments dérivés des céréales et de céréales non transformées collectées entre 2007 et 2012 dans 21 pays européens étaient contaminées par des trichothécènes de type B, ou TCTB, et en particulier du déoxynivalénol, ou DON. Ces mycotoxines sont produites par des champignons phytopathogènes sur les grains en cours de remplissage, avant la récolte. Dans un contexte de changement climatique, cette situation pourrait s’aggraver. Ainsi, maitriser les contaminations en mycotoxines est une tâche urgente qui ne peut être repoussée. En Europe, le DON est principalement produit par Fusarium graminearum. Si les étapes de la voie de biosynthèse des TCTB sont assez bien décrites, les mécanismes moléculaires impliqués dans la régulation de cette voie restent, à ce jour, mal compris.Des études récentes ont mis en évidence que les modifications post-traductionnelles des histones canoniques ainsi que de leurs variantes joue un rôle critique dans les régulations des biosynthèses des mycotoxines et autres métabolites secondaires chez les champignons filamenteux, en modifiant la structure de la chromatine. Chez F. graminearum, il a été prouvé que l'histone désacétylase HDF1 est impliquée dans l'activation de la production de DON. Par contre, la marque histone H3K27me3 réprime 14 % de son génome, y compris des gènes impliqués dans les voies métaboliques secondaires. La variante d'histone H2A.Z, trouvée de façon ubiquitaire chez les eucaryotes, participe à de nombreux processus biologiques dont la stabilité génomique, la réparation de l’ADN, la régulation de transcription et la désactivation des télomères. Toutefois, les mécanismes sous-jacents de ces fonctions restent flous. Chez certaines espèces, la fonction de H2A.Z semble essentielle. À ce jour, la seule étude ciblant directement la fonction de H2A.Z chez les champignons filamenteux a été réalisée chez Neurospora crassa et a identifié son rôle dans la réponse au stress oxydatif.Nous avons ici fait l’hypothèse que H2A.Z est impliquée dans des processus biologiques importants chez F. graminearum, y compris ceux impliqués dans la production de métabolites secondaires dont les mycotoxines. Ce projet vise ainsi à caractériser les rôles joués par H2A.Z dans le contrôle du développement, du métabolisme et de la virulence chez F. graminearum.À l’aide d’une approche de génétique inverse, nous avons créé six mutants ne possédant plus le gène codant H2A.Z dans trois souches différentes de F. graminearum. Tous les mutants présentent un déficit en sporulation, germination, croissance radiale et production de DON. Cependant, l'intensité des effets observés dépend du fond génétique considéré. En outre, le rajout du gène sauvage codant H2A.Z ne restaure pas les phénotypes sauvages. Les séquençages des génomes complets des mutants ont montrés que, bien que H2A.Z ait été totalement éliminé du génome, des mutations compensatoires se produisent à d'autres loci, indépendamment du fond génétique, dans des gènes impliqués dans le remodelage de la chromatine. De manière frappante, une mutation supplémentaire a été détectée dans les mutants délétés pour H2AZ dans lesquels l’allèle sauvage a été réintroduit. Nous avons également construit les mutants surexprimés H2A.Z, mais aucune différence significative de phénotype entre les mutants et la souche sauvage n’a été observée. L’ensemble de ces résultats indique que H2A.Z est essentielle chez F. graminearum, l’occurrence de mutations compensatoires ayant compensé l’effet létal de la délétion infligée. Nous émettons l'hypothèse que les profondes réorganisations des réseaux génétique permettent cette plasticité, avec certaines conséquences en termes d'évolution et d'adaptation.
