14. Sharpen Your Sword for Litigation: Incumbent Strategic Reaction to the Threat of Entry.

Abstract

While patents have been shown to play a role as barriers to entry, they also reveal information about incumbent strategies and risk being invalidated. We examine the trade-off incumbents face between using their patents ex ante as entry deterrents or ex post once competitors have revealed their moves. Leveraging the unique characteristics of the pharmaceutical sector, in which it is possible to observe exactly when a competitor entry threat materializes, and exploiting exogenous variation in that timing, we show that incumbents intentionally fragment and delay the full disclosure of their intellectual property rights through continuation patents. They disproportionately reveal continuation patents after a competitor entry threat becomes concrete, tailoring their response to the threat they have received and successfully delaying competitor entry through litigation. The detected incumbents' reaction is stronger when their attacked drugs are valuable and when the patents listed at the Food and Drug Administration approval of a drug are relatively narrow in scope. 16. Micropatterns for surface potential mapping of biomolecules by Kelvin probe force microscopy

Abstract

Current methods to detect and quantify electrostatic properties of biomolecules, nowadays, still facing many challenges. For instance, they are not sensitive enough to meet the needs of more precise measurements, they require the use of external markers or antibodies, or rely on indirect calculations or models such is the case for zeta potential and electrophoretic mobility. However, Kelvin probe force microscopy (KPFM) have gained attention due to its expanding possibilities to asses electrostatic properties of biomolecules. Among the advantages that KPFM possess over other methods, stands out its high sensitivity and spatial resolution. Moreover, it does not require the use of external labels. In the interest of establishing a method for mapping surface potential of biomolecules by means of Kelvin probe force microscopy, two techniques of poly-dimethylsiloxane (PDMS) soft lithography for micro-patterning (micro channel filling and micro contact printing) are studied. Similarly, the possibility of patterning biomolecules on different substrates (mica, glass and silicon dioxide) is explored. Primarily, poly-L-lysine was used as a model biomolecule due to the exposure of amino functional groups. Different physicochemical conditions on poly-Llysine micro patterns were tested in order to assess the suitability of soft lithography combined with KPFM to measure electrostatic properties of biomolecules. In this sense, continuous KPFM scans, immersions in water, lift height and time dependence were investigated. Moreover, due to the importance of the understanding of the effects of exposure of biomolecules to elevated levels of sugars, immersions in D-ribose were performed. A concentration dependent effect was observed, affecting drastically the surface potential of polyL-lysine. Electrostatically driven patterning of colloidal gold nanoparticles was also achieved on pol-L-lysine micro patterns, resulting in a good method for marking the presence of pol-Llysine and opening the possibility to improve some properties of nanoparticle systems. Besides microchannel filling, micro contact printing of poly-L-lysine was successfully achieved on mica, glass and silicon dioxide, resulting in better quality micro patterns and understanding of the influence of using different substrates on surface potential. On this regard, continuous KPFM scans revealed a surface charge dynamic on mica, whereas on glass and silicon dioxide surface potential became more stable. Insulin, BSA and β-lactoglobulin were successfully patterned on mica by micro contact printing and imaged by KPFM. Response to pH and immersions in water was investigated showinga clear reversible shift on surface potential. Similarly, cross-patterning of different proteins on the same substrate for surface potential oneto-one comparison was successfully achieved. The classic case of avidin-biotin complex was also investigated byboth fluorescence optical microscopy and KPFM. Finally, in an attempt to stretch the applications of KPFM for surface potential mapping of biomolecules, insulin amyloid fibrils were co-fibrillated with highly charge nanoparticles. Surface potential maps of amyloid structures were achieved and the effect of continuous scanning on surface potential was assessed.

Abstract

Plastic has become part of daily life because it is used in all industrial applications. Although it is beneficial and practical, it is the material that generates the most pollution in ecosystems. Therefore, the main objective of this research is to produce plastic films capable of degrading the environment in a shorter time (bioplastics). For this purpose, different samples were made with cassava starch (pulp or commercial), using amounts in the solution of 4% w/v and 30% w/w of cassava starch and glycerol, respectively, considering variations between the percentages of chitosan. The results showed that it is possible to obtain bioplastic by the casting method with the mixture of cassava starch (pulp or commercial), glycerol, and chitosan. Likewise, thanks to the laboratory tests carried out, was possible to determine that the use of commercial starch to produce biodegradable plastic films significantly favors moisture adsorption, solubility, and biodegradability, compared to starch extracted from cassava by hand, with good statistics validity of the analyzed data., El plástico se ha convertido parte del diario vivir debido a que se utilizada en todas las aplicaciones industriales, aunque es muy útil y práctico, es el material que genera más contaminación en los ecosistemas. Por lo tanto, el objetivo principal de esta investigación es la producción de películas plásticas capaces de degradarse con el medio ambiente en un menor tiempo (bioplásticos). Para este propósito, se realizaron diferentes muestras con almidón de yuca (pulpa o comercial), utilizando cantidades en la solución de 4% p/v y 30% p/p de almidón de yuca y glicerol, respectivamente, considerando variaciones entre los porcentajes de quitosano. Los resultados demostraron que con la mezcla de almidón de yuca (pulpa o comercial), glicerol y quitosano, fue posible la elaboración de bioplástico por el método casting. Asimismo, gracias a las pruebas de laboratorio realizadas se determinó que la utilización de almidón comercial para la elaboración de películas plásticas biodegradables favorece a una mayor adsorción de humedad, solubilidad y biodegradabilidad, comparado con el almidón extraído de la yuca artesanalmente, con buena validez estadística de los datos analizados.

Abstract

Aim of study: Individual purchasing behaviour depends on economics, psychology, marketing and sensory science. Given that the list of reasons-for-buying is almost unlimited, we have defined 14 pre-set descriptors thought to entail the more important attributes when make buying decisions within a food group of selected fruits and vegetables. Area of study: We have used a United States buyer data base of over 175,000 observations. Material and methods: Each household was asked to rank their first, second, and third most important reasons for buying, within the set of descriptors. The overriding goal was to gain insight into the attributes and change over time. Using empirical models, the relative importance of the attributes is shown and forecasted for a decade beyond 2021. Main results: Price and quality were expected to be the main drivers; however, the organic attribute is one requiring significant changes in the production, inspection, distribution and marketing policies, hence considering future expectations for organics is particularly important. Research highlights: Preferences for organics have grown, but what are the expectations a decade from now? Will that interest remain so for many years to come?

Abstract

Rats exposed to a downshift in the concentration of a sucrose solution from 32% to 4% exhibit a transient suppression of consummatory behavior relative to an unshifted control always exposed to 4% sucrose. One explanation of this effect, known as consummatory successive negative contrast (cSNC), explains consummatory suppression as arising from an emotional state of frustration that redirects behavior away from the source of the devalued solution. A preliminary selective breeding protocol consisting of three experiments was performed. Experiment 1 reports results from 5 generations of selected breeding for either high (H) or low (L) recovery rates from cSNC. A control line of randomly (R) mated rats was included. cSNC was reduced in H rats, but L and R rats did not differ across generations. H rats also provided no evidence of behavioral activation in acquisition or increased persistence in extinction after partial reinforcement, rather than continuous reinforcement. L and R rats, by contrast, showed both of these effects. H rats were also significantly smaller in body size than R rats, but did not differ in terms of water intake, sucrose sensitivity, open-field activity, or responding to sucrose solutions before the downshift. In Experiment 2, H infants from the sixth selected generation showed increased bandwidth in vocalizations induced by mother-infant separation relative to L and R rats. Experiment 3 showed that H rats failed to show increased response to incentive downshift after treatment with the nonselective opioid antagonist naloxone, as done by L and R rats. The results, if replicated, may provide support for the interpretation of a significant role of frustration in cSNC.

Abstract

Introdução: O envolvimento pulmonar associado ao linfoma de Hodgkin pode ocorrer tanto de forma primária como secundária. A primária é uma entidade rara e origina-se do tecido linfóide associado a mucosa. Já a secundária, mais frequente, pode resultar da proliferação direta dos gânglios linfáticos mediastinais ou de disseminação linfática ou hematogênica de outros locais. Apresentação do caso: Paciente do sexo feminino, 42 anos, com quadro de tosse, odinofagia, dor em seios da face. Há 7 dias iniciou quadro de dispneia e febre, com piora nos últimos 3 dias. Procurou pronto atendimento diversas vezes, com diagnóstico e tratamento de sinusite. Foi realizado Rx de seios da face, sem alterações. Hemograma completo, VHS, teste para tuberculose, sem achados específicos. Discussão: O linfoma Hodgkin pulmonar primário é uma patologia incomum, com poucos casos documentados, consistindo em menos de 1% de todos os linfomas. Apresenta discreta preponderância de incidência em mulheres (1,4:1 F:M) com distribuição bimodal de idade (60 anos). Ocorre quando a proliferação linfóide clonal afeta os pulmões e não apresenta disseminação extrapulmonar no momento do diagnóstico ou nos 3 meses seguintes. Conclusão: O tratamento é variável na literatura, devido a falta de diretrizes, e é determinado de acordo com a extensão da patologia. A abordagem pode ser via cirurgia, radioterapia ou quimioterapia, sendo que muitas vezes é realizada uma associação dos métodos.

Abstract

Introdução: Craniofaringiomas Adamantinomatosos são tumores do Sistema Nervoso Central, localizados no ducto craniofaríngeo. Provenientes de células escamosas e com curso tipicamente benigno, tem prevalência na infância e incidência até, aproximadamente, os 20 anos de idade. O diagnóstico é frequentemente tardio pelo seu crescimento lentificado e clínica inespecífica, agrupada em: cefaléia, distúrbios visuais e de caráter hormonal. O controle sintomático pode ser realizado farmacologicamente, embora a localização anatômica favoreça uma abordagem cirúrgica para resolução da patologia, considerando, também, tratamento adjuvante. Apresentação do caso: Paciente do sexo feminino, 9 anos de idade, estudante e natural de Rio Verde - GO, é levado pela mãe ao pediatra, que relata que o paciente tem apresentado cefaleia holocraniana intermitente e sem fator causal específico, há aproximadamente 70 dias. Associado ao quadro, refere-se a ganho de peso sem mudanças significativas na dieta ou hábitos de vida, no entanto, não soube especificar o ganho em quilogramas. Foi solicitada Ressonância Magnética de crânio, que confirmou o diagnóstico de Craniofaringioma Adamantinomatoso. Discussão: O desenvolvimento das técnicas cirúrgicas para ressecção tumoral permite optar por uma ressecção completa ou subtotal associada à radioterapia adjuvante (RT), sendo que a segunda permitiu uma taxa maior de sobrevida livre de progressão de doença. Porém, não pode-se excluir a primeira opção, já que é preconizada para indicações específicas, sobretudo diante da localização tumoral. Portanto, a abordagem do tumor deve ser planejada de modo individualizado, já que há risco de prejuízo na qualidade de vida e funcionalidade do indivíduo, em decorrência do sítio patológico e estruturas potencialmente afetadas. Conclusão: Para estabelecer o tratamento correto deve-se observar a localização e os impactos de cada intervenção avaliando as particularidades de cada paciente.

Abstract

Based on the structural knowledge of TLR5 surface using blind docking platforms, a series of peptides derived from HMGB1 truncated acidic tail from Salmo salar was designed TLR5 agonistic. Also, a template peptide with the wild type C-terminal acidic tail sequence as reference was included (SsOri). Peptide binding poses complexed on TLR5 ectodomain model from each algorithm were filtrated based on docking scoring functions and predicted theoretical binding affinity of complex.
